锥栗种群在鼎湖山三个群落中的遗传分化研究   总被引:14,自引:1,他引:14  
采用扩增片段长度多态性(AFLP,Amplified fragmen length polymorphism)方法对锥栗(Castanopsis chinensis)在3个不同群落亚种群的遗传分化进行了研究。研究地点位于广东省鼎湖山,3个群落分别是:针叶林群落,针阔叶混交林群落,常绿阔叶林群落。采集的叶要进行DNA提取,酶切、连接、PCR扩增后进行变异性聚丙烯酰胺的凝脉电泳,银染后把所得带进行1/0标记。结果表明,四组引物对分别扩增出27,20,33,39条带,其中分别有15,15,18,26条是多态性带,说明其多样性较高。用AMOVA(Analysis of molecular variance) 分析表明75.36%的遗传变异表现在种群内,24.64%的遗传变异表现在种群间,极显著(P<0.001)。使用Nei遗传距离进行了算术平均数的非加权成组配对法(UPGMA)。其聚类结果表明常绿阔叶林群落亚种群和针阔叶混交林群落亚种群的种群聚合在一起。上述结果说明锥栗不同亚种群有不同的遗传多样性,种群间遗传分化较大。这是种群本身遗传特性的反映,并与群落演替密切相关,即不同群落微环境对种群遗传的影响。  相似文献

As a commercially important species in East Asia, the natural resources of Scapharca broughtonii have been suffering from severe population decline across its main habitats. In China, recovery efforts for S. broughtonii are in progress. To provide scientific bases for fisheries management and conservation program, genetic diversity and population structure of seven wild populations of S. broughtonii from the northern China coast was assessed using seven microsatellite loci in this study. High genetic diversity was present in all the seven populations, as observed in mean allelic richness per locus (11.3–12.5), and average expected heterozygosity (0.835–0.867). No significant difference in allelic richness or expected heterozygosity was observed among the seven populations. Pairwise FST estimates and NJ tree topologies based on DC distances indicated that the seven populations fell into two groups, showing a clear division between the populations from the south and north of the Shandong Peninsula. Genetic differentiation was further analyzed using AMOVA and assignation tests. Genetic barrier analysis using Monmonier algorithm also confirmed that the Shandong Peninsula was the putative barrier separating the northern and southern populations. In addition, marine currents probably play an important role in high gene flow among three populations from the same marine gyre.  相似文献

Eleutheronema tetradactylum is an economically important fish species in China water. To investigate the genetic diversity and describe population structure of it, an 1151 base pair (bp) fragment of the mitochondrial DNA Cytb sequence was analyzed in 120 individuals from four populations in the East China Sea and the South China Sea. A total of 16 haplotypes were defined by 24 variable nucleotide sites. High level of haplotype diversity and low nucleotide diversity were observed in all populations. The results of AMOVA detected that 89.44% of the genetic variation occurred within populations. Significant genetic differentiations were detected among populations (0.05097, P < 0.05), but no large-scale regional differences were detected. Analysis of neutral evolution and mismatch distribution suggested no recent population expansion happened. The present results provided new information for genetic assessment, fishery management and conservation of this species.  相似文献

We analysed the historical genetic diversity of human populations in Europe at the mtDNA control region for 48 ancient Britons who lived between ca AD 300 and 1000, and compared these with 6320 modern mtDNA genotypes from England and across Europe and the Middle East. We found that the historical sample shows greater genetic diversity than for modern England and other modern populations, indicating the loss of diversity over the last millennium. The pattern of haplotypic diversity was clearly European in the ancient sample, representing each of the modern haplogroups. There was also increased representation of one of the ancient haplotypes in modern populations. We consider these results in the context of possible selection or stochastic processes.  相似文献

为了明确我国北方不同地理种群甜菜夜蛾Spodoptera exigua遗传多样性与种群遗传结构,阐明该种害虫的种群历史动态,首次对采自我国北方8省17县(市)304头甜菜夜蛾样品进行mt DNA Cytb基因序列测定与分析,利用Dna SP 5.0和Arlequin 3.0软件分析种群遗传多样性、遗传结构、遗传分化与分子变异,基于MP、ML与贝叶斯法构建单倍型系统发育树,与此同时,基于Median-joining法对所有个体构建单倍型网络关系图。结果表明,在所分析的304个序列样本中,共检测出19个单倍型,其中,包括9个共享单倍型,单倍型Hap6为所有种群所共享。总群体具有较低的遗传多样性(Hd=0.422±0.035,π=0.00119±0.00011)与较小的遗传分化(F_(ST)=0.108,P0.001)。单倍型系统发育分析与网络关系图结果表明,虽然19个单倍型被分为2个分支,但各单倍型相互散布在不同种群中,未形成明显谱系地理格局。AMOVA分析表明,甜菜夜蛾遗传变异主要来自种群内(89.18%),种群间变异水平较低(10.82%)。中性检验(Tajima's D=-1.897,P0.05;Fu's FS=-4.424,P0.05)与错配分布分析表明,我国北方地区甜菜夜蛾种群曾经历过种群的近期扩张。  相似文献

老挝纹胸鮡是分布于澜沧江流域的一种特有鱼类,喜激流底栖生活。为了解其各地理种群间的遗传结构与遗传变异,文章分析了129尾采自澜沧江8条支流水系的老挝纹胸鮡mtDNA细胞色素b基因1 138 bp的序列变异,共发现16个多态性位点,定义了15个单倍型。澜沧江老挝纹胸鮡遗传变异较低,平均单倍型多样性指数和核苷酸多样性指数分别是h=0.299,π=0.299,有3个种群甚至没有变异,分子变异方差分析结果表明各种群内变异大于各种群间变异,种群间没有差异。对老挝纹胸鮡所有种群进行的单倍型错配分布呈现单峰型,中性检验结果均为负值(Tajima’s D=?2.36965,P<0.02;Fu’s Fs=?20.975,P<0.05)表明,澜沧江老挝纹胸鮡自然种群可能经历过近期的种群扩张事件。  相似文献

In this study, the mitochondrial DNA (mtDNA) control region and the mitochondrial cytochrome b gene of stranded Indo-Pacific humpback dolphin (Sousa chinensis) samples from the Pearl River Estuary and Xiamen waters were sequenced and analyzed. The result of mtDNA control region revealed 34 variable sites and four unique haplotypes (named as A, B, C and D) identified among the total samples from these two water areas, and the most common haplotype (A) was shared by 75% of the dolphins sampled from the two water areas. The haplotypic diversity (h) was 0.455 and the nucleotide diversity (π) was 0.0088. The phylogenetic analysis showed that the haplotype A, C, and D were closely related, but the haplotype B (unique for XM01 from Xiamen) was far from the other three. By scanning cytochrome b fragments, two haplotypes (A and B) were identified in these two water areas, and the most common haplotype (A) was shared by 91.67% individuals, while XM01 from Xiamen as the only exception. The date suggest that there is a possibility of gene exchange between the two populations in the Pearl River Estuary and Xiamen waters, and there possibly exists a unique maternal lineage in Xiamen waters.  相似文献

Eleven polymorphic microsatellites were isolated from an important foodfish species, silver pomfret (Pampus argenteus). The loci showed different variation patterns in 24 unrelated P. argenteus individuals, with a mean number of alleles of 11.5 and a mean expected heterozygosity of 0.85. Eight of the 11 loci conformed to Hardy–Weinberg equilibrium. All and six of the 11 markers amplified specific and polymorphic products in Chinese pomfret (Pampus sinesis) and dark pomfret (Pamus cinereus), respectively. These markers should be useful for population genetic analysis and biodiversity studies of P. argenteus and other pomfret species.  相似文献

怒江扎那纹胸鮡的遗传多样性和遗传分化   总被引:1,自引:1,他引:1  
怒江水电开发将对扎那纹胸鮡产生不利的影响。为了解扎那纹胸鮡遗传多样性和遗传分化情况, 文章测定了采自怒江中下游怒江州地区的贡山、古登和泸水及保山市地区的道街、勐糯和木城6个扎那纹胸鮡群体共102个个体的线粒体Cyt b基因序列。结果显示, 在1 137 bp序列中共检测到87个变异位点, 定义了36个单元型。总样品的单元型多样性(h)和核苷酸多样性(π)分别为0.851±0.028和0.01356±0.0008。扎那纹胸鮡的遗传多样性相对较低, 但怒江州种群遗传多样性显著高于保山市种群。群体间分化指数(FST)(0.475~0.846)明显高于群体内分化指数(0.002~0.108), 且各群体间分化指数和地理距离呈线性正相关。利用AMOVA(Analysis of molecular variance )对遗传分化进行分割, 群体间和群体内分别占53.65%和 46.35%, 群体间遗传分化指数(FST)为0.5365 (P<0.01), 扎那纹胸鮡在怒江州和保山种群分化显著。单元型分子系统树和简约网络图显示, 扎那纹胸鮡单元型聚为两个独立的支系: 怒江州支系和保山市支系。这些鱼类至少代表一个管理单位, 但也可能是一个进化显著单位。因此, 建议保护扎那纹胸鮡种群, 在水电工程建设时应充分考虑扎那纹胸鮡种群结构现状, 避免不同区域的种群之间发生基因交流。  相似文献

南亚热带森林片断化对厚壳桂种群遗传结构的影响   总被引:4,自引:0,他引:4  
森林片断化是造成全球生物多样性丧失的主要原因,但从种群遗传学角度开展的研究还远远不够。因此,本文采用ISSR(inter-simplesequencerepeat)方法对广东省鼎湖山、黑石顶、大雾岭、古田、萝岗和饶平6个地点的南亚热带片断化森林中的厚壳桂(Cryptocaryachinensis)种群的遗传多样性进行了研究。每个种群分成体、小树和幼苗3个年龄级采样,共采集厚壳桂样品448个。7个ISSR引物共扩增出209条多态性带。研究结果表明,6个厚壳桂种群的总遗传多样性为0.1529,种群平均遗传多样性为0.0537。和保护较好的厚壳桂种群相比,萝岗和饶平的两个村边林种群的遗传多样性较低。6个厚壳桂种群间的遗传分化系数(GST)为0.6489。AMOVA分析结果表明,总的遗传变异中,67.97%(P<0.0002)的遗传变异存在于种群间,2.06%(P<0.01)的遗传变异存在于不同年龄级之间,29.96%(P<0.0002)的遗传变异存在于种群内。基于遗传距离的UPGMA分析结果表明,6个厚壳桂种群大致分成两支:一支包括大雾岭和黑石顶2个种群,另一支包括其他4个种群。鼎湖山、古田、大雾岭和黑石顶不同年龄级均各自聚合在一起。可以看出,森林的破碎化导致了南亚热带厚壳桂种群遗传多样性降低,种群间基因流受阻,提示我们对于同一地区的易危物种,这种威胁将很大,急需开展相关的保护研究。  相似文献

海草是生长在潮间带和潮下带的单子叶植物,由海草植物组成的海草床是生态系统服务价值最高的生态系统之一.然而,近几十年人类活动干扰、全球气候变化等因素导致海草床衰退严重.海菖蒲是分布于热带、体型最大的雌雄异株海草,我国位于该物种的分布北缘,本文对其克隆多样性和遗传结构进行研究,以期为该海草的保护提供参考.采用4对多态微卫星标记对采自海南岛4个地点的现存海菖蒲种群的样品进行基因型分型.结果表明:海菖蒲种群克隆多样性和遗传多样性较低,这与所研究种群处于分布区北缘有关;种群间遗传分化值范围较大(0.073~0.309),这可能是由于分布于不同港湾的种群间距离范围较大以及局域绝灭/再拓殖的遗传漂变效应所致;各种群未发现近期经历种群瓶颈的信号,很可能是由于种群内遗传多样性已经很低,种群减小未能导致遗传多样性明显降低.根据种群遗传特征,提出了重点保护种群的建议,鉴于目前我国海菖蒲等海草快速衰退的局面,应强化海草保护并实施海草床生态恢复.  相似文献

1. Nucleotide sequences of a 280 base pair region of the cytochrome b gene were used to assess genetic diversity and to infer population histories in the New Zealand mayfly Acanthophlebia cruentata. 2. A hierarchial examination of populations from 19 streams at different spatial scales in the central and northern North Island of New Zealand found 34 haplotypes. A common haplotype was found in all central region streams and unique haplotypes in northern streams. Several central streams had region specific haplotypes with genetically differentiated populations at the 70–100 km scale. 3. Haplotype diversity was high (0.53–0.8) at most sites, but low (0–0.22) in some central sites. amova analyses found significant genetic diversity among regions (69%) and among catchments (58%). Most population pairwise FST tests were significant, with non‐significant pairwise tests among sites in the central region and pairs of sites between neighbouring streams. 4. The levels of sequence divergence are interpreted as the result of Pleistocene divergence in multiple refugia, leading to the evolution of regionally unique haplotypes. The low diversity in some central region populations may result from recent colonisation following local extinctions, associated with volcanic events.  相似文献

1. Complete sequences of 1140 base pair of the cytochrome b gene from 133 specimens were obtained from nine localities including the inflow drainage system, isolated lakes and outflow drainage system in Qinghai–Tibetan Plateau to assess genetic diversity and to infer population histories of the freshwater fish Schizopygopsis pylzovi. 2. Nucleotide diversities (π) were moderate (0.0024–0.0045) in populations from the outflow drainage system and Tuosuo Lake, but low (0.0018–0.0021) in populations from Qiadam Basin. It is probable that the low intra‐population variability is related with the paleoenvironmental fluctuation in Qiadam Basin, suggesting that the populations from Qiadam Basin have experienced severe bottleneck events in history. 3. Phylogenetic tree topologies indicate that the individuals from different populations did not form reciprocal monophyly, but the populations from the adjacent drainages cluster geographically. Most population pairwise FST tests were significant, with non‐significant pairwise tests between Tuosu Lake and Tuosuo Lake in the north‐west of the Qinghai–Tibetan Plateau. Analysis of molecular variance (amova ) indicates that the significant genetic variation was explained at the levels of catchments within and among, not among specific boundaries or inflow and outflow drainage systems. 4. The nested clade phylogeographical analysis indicates that historical processes are very important in the observed geographical structuring of S. pylzovi, and the contemporary population structure and differentiation of S. pylzovi may be consistent with the historical tectonic events occurred in the course of uplifts of the Qinghai–Tibetan Plateau. Fluctuations of the ecogeographical environment and major hydrographic formation might have promoted contiguous range expansion of freshwater fish populations, whereas the geological barriers among drainages have resulted in the fragmentation of population and restricted the gene flow among populations. 5. The significantly large negative Fs‐value (−24.91, P < 0.01) of Fu's Fs‐test and the unimodal mismatch distribution indicate that the species S. pylzovi underwent a sudden population expansion after the historical tectonic event of the Gonghe Movement. 6. The results of this study indicate that each population from the Qinghai–Tibetan Plateau should be managed and conserved separately and that efforts should be directed towards preserving the genetic integrity of each group.  相似文献

The origin and demographic history of the ethnic populations of China have not been clearly resolved. In this study, we examined the hypervariable segment I sequences (HVSI) of the mitochondrial DNA control region in 372 individuals from nine Chinese populations and one northern Thai population. A relatively high percentage of individuals was found to share sequences with those from other populations of the same ethnogenesis. In general, the populations of southern or Pai-Yuei tribal origin showed high haplotype diversity and nucleotide diversity compared with the populations of northern or Di-Qiang tribal origin. Mismatch distributions from these populations showed concordant features. All except the northern groups Nu, Lisu, Tibetan, and Mongolian showed typical signatures of ancient population expansions in the mismatch distributions and neutrality tests. Episodes of extreme size reduction in the past are one of the likely explanations for the absence of evidence of expansion in northern populations. Small sample sizes as well as samples from isolated subpopulations contributed to the bumpy mismatch distributions observed. Phylogenetic analysis and haplotype sharing among populations suggest that current mtDNA variation in these ethnic populations could reveal their ethnohistory to some extent, but in general, linguistic and geographic classifications of the populations did not agree well with classification by mtDNA variation.  相似文献

Silver pomfret (Pampus argenteus) is an important fisheries resource in China, but harvest management lacks identification of management units (genetic stocks or populations). Using amplified fragment length polymorphism (AFLP), we investigated the population genetic structure of silver pomfret from the Yellow and East China Seas. In 143 individuals examined from six locations (three locations in the Yellow Sea, three in the East China Sea), a total of 194 loci were detected by six primer combinations. In all loci, 149 loci (76.8%) were polymorphic, and the percentage of polymorphic loci varied within samples from 62.9% to 67.0%. Among the six samples, Nei’s genetic diversity was 0.23 ± 0.20–0.25 ± 0.21, and Shannon’s index was 0.34 ± 0.28–0.36 ± 0.29. Analyses revealed significant genetic differentiation among the six samples, indicating at least six separate populations of silver pomfret in the Southern Yellow and East China Seas.  相似文献

The genetic population structure of a large, wide-ranging marsupial, the red kangaroo ( Macropus rufus ) was assessed using sequence and haplotype frequency data of mitochondrial DNA (mtDNA) from locations across the species range in Australia. Results from sequence data revealed extensive haplotype diversity within the red kangaroo (32/34 sequences were unique). Sequence diversity was distributed within rather than between geographical regions across the species range. Genetic connectivity across the range of the species has therefore been maintained over the long term. On a smaller within-region scale, significant genetic structuring was evident from heterogeneity of haplotype frequencies amongst sampling sites. The geographical scale of panmictic populations differed across the continent with more restricted genetic populations occurring in areas with greater topographic and habitat complexity. We propose that these differences in area of genetic populations are the result of population responses to limiting ecological factors during drought.  相似文献

Theory predicts that genetic diversity and genetic differentiation may strongly vary among populations of the same species depending on population turnover and local population sizes. Yet, despite the importance of these predictions for evolutionary and conservation issues, empirical studies comparing high‐turnover and low‐turnover populations of the same species are scarce. In this study, we used Daphnia magna, a freshwater crustacean, as a model organism for such a comparison. In the southern/central part of its range, D. magna inhabits medium‐sized, stable ponds, whereas in the north, it occurs in small rock pools with strong population turnover. We found that these northern populations have a significantly lower genetic diversity and higher genetic differentiation compared to the southern/central populations. Total genetic diversity across populations was only about half and average within‐population diversity only about a third of that in southern/central populations. Moreover, an average southern population contains more genetic diversity than the whole metapopulation system in the north. We based our analyses both on silent sites and microsatellites. The similarity of our results despite the contrasting mutation rates of these markers suggests that the differences are caused by contemporary rather than by historical processes. Our findings show that variation in population turnover and population size may have a major impact on the genetic diversity and differentiation of populations, and hence may lead to differences in evolutionary processes like local adaptation, hybrid vigour and breeding system evolution in different parts of a species range.  相似文献

DNA barcoding provides an efficient method for species-level identifications. In this study, we have amplified partial sequences of mitochondrial cytochrome c oxidase I (COI) gene from 110 specimens of 45 species of Caenogastropoda collected from the coast along China to evaluate whether DNA barcodes can distinguish these species accurately. The average Kimura 2-parameter (K2P) distances within species, genera and families were 0.44%, 13.96% and 22.27%, respectively. Both the neighbour-joining tree and the Bayesian tree showed a clear discrimination of all the species in our study with highly supported clades. These results proved that the species of Caenogastropoda can be efficiently and accurately identified by DNA barcoding based on the COI gene.  相似文献

Population genetic structure is determined both by current processes and historical events. Current processes include gene flow, which is largely influenced by the migration capacity of a species. Historical events are, for example, glaciation periods, which have had a major impact on the distribution of many species. Species with a low capacity or tendency to move about or disperse often exhibit clear spatial genetic structures, whereas mobile species mostly show less spatial genetic differentiation. In this paper we report on the genetic structure of a small, wingless arthropod species (Orchesella cincta: Collembola) in Europe. For this purpose we used mtDNA COII sequences and AFLP markers. We show that large genetic differences exist between populations of O. cincta, as expected from O. cincta's winglessness and sedentary lifestyle. Despite the fact that most variability was observed within populations (59%), a highly significant amount of AFLP variation (25%) was observed between populations from northwestern Europe, central Europe and Italy. This suggests that gene flow among regions is extremely low, which is additionally supported by the lack of shared mtDNA alleles between regions. Based on the genetic variation and sequence differences observed we conclude that the subdivision occurred long before the last glaciation periods. Although the populations still interbreed in the lab, we assume that in the long term the genetic isolation of these regions may lead to speciation processes.  相似文献

测定了9个不同地理区域的小稻蝗Oxya intricata (Stal)共33个个体的线粒体DNA (mtDNA)细胞色素b基因部分序列,比较其同源性,计算核苷酸组成,并用瘤锥蝗科的曲尾似橄蝗Pseudomorphacris hollisi和斑腿蝗科的芋蝗Gesonula punctifrons作外群构建NJ分子系统树。在获得的小稻蝗432 bp的序列中, A+T约占70.3%,其中30个核苷酸位点存在变异(约占所测核苷酸的6.9%),有5个位点引起了氨基酸的变异。就每个氨基酸密码子来看,第3位点的A+T含量较高,达90.4%。由NJ树显示,小稻蝗mtDNA 细胞色素b序列不同单倍型之间有一定的分歧,形成不同的簇类关系,其分枝与地理分布没有直接的对应关系,但总体上看,这种簇类关系基本上呈平行分布,没有明显的地域性差别。  相似文献
