Maize is a major staple food for over 300 million people in sub‐Saharan Africa. Sustainable productivity of this primary crop has been recently threatened by Fall armyworm (FAW), Spodoptera frugiperda invasion. Due to lack of environmentally safe management strategies, immediate responses by growers and governments to tackle FAW are based on rampant use of pesticides. Looking for efficient biopesticides, twenty entomopathogenic fungal isolates (14 Metarhizium anisopliae and 6 Beauveria bassiana) were screened for their efficacy against eggs and second instar larvae of FAW. A single discriminating concentration of 1 × 108 conidia ml?1 and four replicates per treatment were used in all experiments. Isolates were assessed for their ability to cause mortality of FAW second instar larvae, eggs and the neonate larvae that emerged from treated eggs. Among the isolates tested, only B. bassiana ICIPE 676 caused moderate mortality of 30% to second instar larvae. Metarhizium anisopliae ICIPE 78, ICIPE 40 and ICIPE 20 caused egg mortalities of 87.0%, 83.0% and 79.5%, respectively, and M. anisopliae ICIPE 41 and ICIPE 7 outperformed all the others by causing 96.5% and 93.7% mortality to the neonate larvae, respectively. The cumulated mortality of eggs and neonates was highest with M. anisopliae ICIPE 41 (97.5%), followed by M. anisopliae ICIPE 7, 655, 40, 20 and 78 with total mortality of 96.0%, 95.0%, 93.5%, 93.0% and 92.0%, respectively. These isolates with high cumulated mortality (≥92%), especially ICIPE 78 and 7, which are already commercialized for spider mites and ticks control respectively, would be good candidates for development as biopesticides for management of FAW in Africa if further evidence of their efficacy is obtained in the field.  相似文献

The influence of 15 commercially available fungicides on the germination, growth and virulence of Metarhizium anisopliae, Beauveria bassiana, Isaria fumosorosea, and Lecanillium longisporum was evaluated. The influence of the fungicides on conidial germination was dependant on the fungicide type and dose. Most fungicides retarded conidial germination of all the fungi tested at 10× and at the recommended rate of application, however, their toxicity declined at lower concentrations. Most of the fungicides inhibited mycelial growth of B. bassiana, whereas L. longisporum growth was unaffected. Only two and eight fungicides influenced mycelial growth of I. fumosorosea and M. anisopliae, respectively. None of the fungicides influenced the virulence of B. bassiana and L. longisporum, however, tolylfluanid and azoxystrobin reduced the virulence of M. anisopliae and I. fumosorosea, respectively. These studies clearly show that certain fungicides have the potential to inhibit germination of entomopathogenic fungi in vitro but appear to have little or no effect on their virulence against target insects.  相似文献

Termites adjust their response to entomopathogenic fungi according to the profile of fungal volatile organic compounds (VOCs). This study demonstrates the pathogenicity of Metarhizium anisopliae, Beauveria bassiana and Isaria fumosorosea (=Paecilomyces fumosoroseus) towards the Formosan subterranean termite, Coptotermes formosanus Shiraki (Isoptera: Rhinotermitidae). Using no-choice assays, M. anisopliae was found to be highly virulent (LT50 3.10 d) when compared to B. bassiana (LT50 6.62 d) and I. fumosorosea (LT50 12.39 d). Also using choice assays, the foraging behavior of C. formosanus was determined in the presence of pathogenic fungi. The highly pathogenic fungi (M. anisopliae) elicited a repellent response, causing most of the termites to forage in a safe zone farthest from the fungal source. This repellency resulted in relatively low mortality similar to the controls. The repellency of M. anisopliae conidia can be used to protect human belongings and timber from termites. While I. fumosorosea cultures were not repellent to C. formosanus workers, the termites were highly susceptible to infection. Electroantennographic responses of workers showed approximately 47% and 78% lower level of response to conidia of B. bassiana and I. fumosorosea, respectively, as compared to M. anisopliae. The VOC profile of repellent cultures of M. anisopliae mainly consisted of paraffins (60.97%), while the major proportion of the I. fumosorosea profile consisted of branched and cyclic alkanes (84.41%). From the above findings, we conclude that the incorporation of I. fumosorosea may increase the control potential of bait.  相似文献

Seventeen isolates of Metarhizium anisopliae (Metschnikoff) Sorokin and two isolates of Beauveria bassiana (Balsamo) Vuillemin were evaluated for their pathogenicity against the tobacco spider mite, Tetranychus evansi Baker & Pritchard. In the laboratory all the fungal isolates were pathogenic to the adult female mites, causing mortality between 22.1 and 82.6%. Isolates causing more than 70% mortality were subjected to dose–response mortality bioassays. The lethal concentration causing 50% mortality (LC50) values ranged between 0.7×107 and 2.5×107 conidia ml−1. The lethal time to 50% mortality (LT50) values of the most active isolates of B. bassiana and M. anisopliae strains varied between 4.6 and 5.8 days. Potted tomato plants were artificially infested with T. evansi and treated with B. bassiana isolate GPK and M. anisopliae isolate ICIPE78. Both fungal isolates reduced the population density of mites as compared to untreated controls. However, conidia formulated in oil outperformed the ones formulated in water. This study demonstrates the prospects of pathogenic fungi for the management of T. evansi.  相似文献

Tomato, Solanum lycopersicum, is a widely consumed and economically important vegetable in the tropics. However, its production and productivity have been recently hampered by the invasive Tuta absoluta (Meyrick), a key pest of tomato causing extensive economic damage in Africa. Currently, the commonly used management strategy is synthetic insecticides with hazardous effects on humans and environment. The application of entomopathogenic fungal and viral-based biopesticides is promising alternatives. To identify potent strains, pathogenicity of dry conidia of 12 Metarhizium anisopliae isolates was evaluated against adult T. absoluta. Further effects of the most potent isolates on pupation through the late 4th instar larval infection, adult emergence and horizontal transmission of inoculum were assessed. Metarhizium anisopliae ICIPE 18, ICIPE 20 and ICIPE 665 outperformed the other isolates causing adult mortality of 95.0%, 87.5% and 86.25%, respectively. The three potent isolates differed significantly for LT50 values of 5.13, 3.17 and 2.38 days for ICIPE 18, ICIPE 20 and ICIPE 665, respectively. Pupation and adult emergence were significantly reduced by these potent isolates, which were also compatible with Tuta pheromone (TUA-Optima®) with ≥90% conidial germination 24 hr post-exposure. These potent isolates could be developed as effective biopesticides and used in combination with TUA-Optima® for mass trapping and autodissemination for T. absoluta management in solanaceous crop production systems.  相似文献

Spoladea recurvalis (Fbr.) (Lepidoptera: Crambidae) larvae can cause up to 100% foliage loss on amaranths during severe outbreaks. The Bacillus thuringiensis Subsp. kurstaki product Halt® is a biologically safe biopesticide recommended for the management of Spodoptera exigua (Hübner) and Plutella xylostella (Linnaeus). Spoladea recurvalis larvae are less susceptible to the product. Thirteen chemical additives to improve the efficacy of a Bt spray for control of S. recurvalis were evaluated in laboratory bioassays against second-instar larvae. All the additives except calcium chloride caused an increase in mortality when applied in mixtures with Bt. Among the seven inorganic salts, boric acid was the only inorganic acid that caused more than 50% larval mortality. Boric acid at a concentration of 0.05% increased the activity of B. thuringiensis by 2.9-fold. Boric acid had the shortest LT50 values of 5.3 days compared with all other combinations. Two nitrogenous compounds, peptone and sodium nitrate, caused 54 and 51% larval mortalities; however, this increase in efficacy was not significantly different from the mortalities caused by Bt applied without any additive. Urea increased larval mortality from 40% to 51%, although the increase was not significant from a Bt spray application only. Citric acid had no significant effect on the efficacy of Bt spray against S. recurvalis larvae. Overall, among the additives evaluated, the efficacy of Bt spray was most enhanced by boric acid and could be further evaluated under field conditions for validation, and integration into an Integrated Pest Management (IPM) strategy for S. recurvalis management.  相似文献

Aims: Larval stages of Frankliniella occidentalis are known to be refractory to fungal infection compared with the adult stage. The objective of this study was to identify promising fungal isolate(s) for the control of larval stages of F. occidentalis. Methods and Results: Ten isolates of Metarhizium anisopliae and eight of Beauveria bassiana were screened for virulence against second‐instar larvae of F. occidentalis. Conidial production and genetic polymorphism were also investigated. Metarhizium anisopliae isolates ICIPE 7, ICIPE 20, ICIPE 69 and ICIPE 665 had the shortest LT50 values of 8·0–8·9 days. ICIPE 69, ICIPE 7 and ICIPE 20 had the lowest LC50 values of 1·1 × 107, 2·0 × 107 and 3·0 × 107 conidia ml?1, respectively. Metarhizium anisopliae isolate ICIPE 69 produced significantly more conidia than M. anisopliae isolates ICIPE 7 and ICIPE 20. Internally transcribed spacers sequences alignment showed differences in nucleotides composition, which can partly explain differences in virulence. Conclusion: These results coupled with the previous ones on virulence and field efficacy against other species of thrips make M. anisopliae isolate ICIPE 69 a good candidate. Significance and Impact of the Study: Metarhizium anisopliae isolate ICIPE 69 can be suggested for development as fungus‐based biopesticide for thrips management.  相似文献

An approach to ensure effective pest biocontrol would be to select native isolates of biological control agents (BCAs). A survey to isolate entomopathogenic fungi (EPF) from a hazelnut growing area has been carried out. EPF were recovered from 133 of the 295 soil samples. The main species isolated were Metarhizium anisopliae sensu lato (36%) and Beauveria bassiana sensu lato (42.8%). With regard to controlling abiotic factors, altitude had an effect on the distribution of B. bassiana, but not on M. anisopliae. Cropping system did not have an effect on the occurrence of these EPF, while pH appeared as a predictive variable for both. In addition, we tested the virulence of six of these isolates: three M. anisopliae and three of B. bassiana against larvae of Curculio nucum L. The highest larval mortality (reaching 80%) was due to M. anisopliae (strain 34) when applied in simultaneous combination with four entomopathogenic nematode species: Steinernema carpocapsae (strain B14), Steinernema feltiae (strain D114), Steinernema. sp. (strain D122) and Heterorhabditis bacteriophora (strain DG46). The effect of nematodes was greater and no antagonistic or synergistic effects were observed.  相似文献

Performance of soil application of azadirachtin products with entomopathogens was evaluated as single treatments, in multiple combinations and in different concentrations against the soil stages of western flower thrips, Frankliniella occidentalis (Pergande), in French beans Phaseolus vulgaris L. Treatments consisted of NeemAzal‐T solution, neem pellets, Steinernema carpocapsae (Weiser) Nemastar®, isolates of Metarhizium anisopliae (Metschnikoff) Sorokin (IPP 2539 and ICIPE‐69) and Beauveria bassiana (Balsamo) – Naturalis®. All treatments were analysed for the number of emerging adults, while emerged adults in fungi‐based treatments were analysed additionally for the retarded development of mycosis as a possible cause of secondary mortality. Possible interactive effects in combined treatments were analysed using a generalized linear model (GLM) approach, and three levels of dose–response combinations of the selected treatments were further tested. Bioassay results of the single treatments indicated between 43% and 60% reduction in adult emergence with NeemAzal‐T solution thus proving to be the most efficient. However, most cadavers with entomopathogenic fungi (EPF) treatments showed the development of mycosis. Therefore, the reduction in adult emergence attributed to the EPF was altogether >87%. Combined treatments with Steinernema, Metarhizium (ICIPE‐69), NeemAzal‐T and neem pellets resulted in total reduction in adult emergence of 95–97% when late mortality by mycosis was considered. Of the treatment combinations, two showed synergistic, four additive and one an antagonistic response. Combining low concentration of entomopathogenic nematode (EPN) (100 IJ/cm2) with NeemAzal‐T resulted in satisfactory control compared to the operational dose of EPN, while the highest concentration of Manisopliae (108 conidia) combined with Steinernema showed the best performance with 74% reduction in adult emergence.  相似文献

Nineteen different isolates of the entomopathogenic fungi Beauveria bassiana sensu lato (s.l.) and Metarhizium anisopliae s.l. (Ascomycota: Hypocreales), recovered from different soil samples (field crops, fruit orchards, vegetable fields and forests) and insect cadavers were tested against Rhynchophorus ferrugineus (Olivier) (Coleoptera: Curculionidae) at two different spore concentrations (1 × 107 and 1 × 108 conidia mL?1). Three isolates of B. bassiana and two of M. anisopliae gave >88 % larval and >75 % adult mortality of R. ferrugineus on their highest dose rate respectively. More sporulating cadavers (mycosis) resulted from a high dose rate compared to low dose on both life stages of R. ferrugineus. The current study confirmed the lethal action of B. bassiana and M. anisopliae isolates with mortality levels usually directly proportional to the conidial concentration. This study further confirmed that the isolates recovered from R. ferrugineus dead cadavers gave more mortality compared to the other sources. In the virulence assay two isolates of B. bassiana caused the highest percentage of both larval and adult mortality at all exposure intervals which suggest that they may be the most promising for use in sustainable management programs aimed at microbial control in date palm orchards.  相似文献

Although Turkey is the first among all hazelnut-producing countries, yield per unit area of this crop is low in comparison to other countries, mainly because many insect species seriously damage hazelnut trees and their fruit. To find effective and safe biocontrol agents, we conducted a survey study to isolate entomopathogenic fungi from the hazelnut-growing region of Turkey and characterized the isolated strains in detail. In addition, we determined the effectiveness of seven selected strains from this region against Melolontha melolontha (Coleoptera: Scarabaeidae) which is one of the most serious pests of hazelnut. In 2006 and 2007, 301 soil samples were collected randomly and analyzed for presence of entomopathogenic fungi using the Galleria bait method. Entomopathogenic fungi were found to occur in 20.59% of the soil samples studied. Based on morphology, ITS sequence and partial sequencing of the 18S (SSU rDNA) and EF1-α genes, the isolates were identified as Metarhizium anisopliae var. anisopliae, Metarhizium sp., Beauveria bassiana, Beauveria cf. bassiana, Isaria fumosorosea and Evlachovaea sp. Metarhizium anisopliae var. anisopliae was isolated from 34 sites and was the most frequent and abundant entomopathogenic species recovered. All the isolates tested were pathogenic to M. melolontha. M. anisopliae var. anisopliae KTU-27 and Evlachovaea sp. KTU-36 produced the highest insecticidal activity (86.6%) within 15 days after inoculation. Our results suggest that entomopathogenic fungi could be good biocontrol agents against M. melolontha, and are discussed with respect to ecology of fungi in relation to habitat in order to evaluate biocontrol potential of these isolates. This is the first study of the distribution of entomopathogenic fungi in the hazelnut-growing region of Turkey and of their pathogenicities against M. melolontha.  相似文献

Various endophytic fungi of native plants and crops are important entomopathogens. The objective of this study was to investigate the entomopathogenic action of Beauveria bassiana (Balsamo) Vuillemin, Metarhizium robertsii (Metchnikoff) Sorokin, and Isaria fumosorosea (Wize) Brown & Smith (all Ascomycota: Hypocreales) against larvae of Sesamia nonagrioides (Lefebvre) (Lepidoptera: Noctuidae) artificially introduced into Sorghum bicolor L. (Moench) (Poaceae) plants under natural environmental conditions. Sorghum bicolor is an economically important crop cultivated for grain, fiber, forage, and lately for biofuel, and S. nonagrioides is its main pest in Mediterranean areas. Young sorghum plants were inoculated with the entomopathogens by spraying in the field. Plant water status, chlorophyll concentration, photosynthesis, and transpiration were not affected. Thirty days after endophyte establishment, plants were infested with fourth instars of S. nonagrioides. The endophytes prevented 50–70% of larvae from entering stalks. Larval mortality was 70–100% and tunnel lengths were reduced by 60–87%. Larval infestation resulted in reduced electron transport capacity and net photosynthetic rate, which was ameliorated in the presence of I. fumosorosea and reversed by B. bassiana and M. robertsii. The growth of sorghum was unaffected in all treatments during the experimental period. Beauveria bassiana and M. robertsii can protect sweet sorghum from damage induced by S. nonagrioides under natural environmental conditions without affecting plant physiology and growth.  相似文献

Cowpea (Vigna unguiculata) production is constrained by biotic and abiotic factors, among which Cowpea aphid (Aphis craccivora) is ranked a key insect pest that severely limits its potential for provision of food and nutritional security to millions of people in sub‐Saharan Africa. The use of entomopathogenic fungi for A. craccivora management has been recently demonstrated at laboratory and field levels as alternative to synthetic insecticides, but with low adoption in Africa. This study assessed the efficacy of aqueous and oil formulations of Metarhizium anisopliae ICIPE 62 against A. craccivora under field conditions. Metarhizium anisopliae formulations and a commonly used insecticide Duduthrin® were applied using knapsack sprayers with target output of 350 L/ha. Data on aphid infestation levels were collected weekly. ICIPE 62 efficacy in inducing mortality was also assessed 24 hr post‐treatment coupled with mycosis test. Further, leaf and grain yields were determined. After six weeks post‐treatment in the wet season, there was no significant reduction in aphid density in fungus‐treated plots compared to control and Duduthrin®‐treated plots. However, in the dry season six weeks after applying the treatments, oil formulation spray resulted low aphid density compared to control and Duduthrin®‐treated plots. ICIPE 62 formulations did not negatively affect the natural enemies’ population. Leaf yield from the various treatments did not differ significantly in the wet season, but the two fungal formulations recorded higher yields in dry season compared to other treatments. Grain yields in wet and dry seasons were lower in control and Duduthrin®‐treated plots compared to both ICIPE 62 formulations. This study showed that both M. anisopliae ICIPE 62 formulations are effective in suppressing A. craccivora population under field conditions without adverse effects on its beneficial insects. The study also revealed that efficacy of fungal‐based biopesticides is highly dependent on environmental conditions.  相似文献

Abstract Feeding experiments using three strains of entomopathogenic fungi, Beauveria bassiana, Metarhizium anisopliae and Isaria fumosorosea were conducted with newly moulted 3rd–5th instar Ocinara varians Walker larvae in the laboratory. The mortality of larvae immersed individually in spore suspension (1 × 107 spores/mL) of all the strains was ≥ 80% except 5th instar larvae treated with M. anisopliae which transformed into pupae, but did not result in adult emergence. The growth (total body mass), consumption, relative consumption rate and relative growth rate, were reduced at all three larval stages, while developmental time was extended in infected larvae with concurrent significant increase in approximate digestibility in infected larvae. Conversion of digested food (ECD) and ingested food (ECI) values declined in infected larvae as compared to the healthy larvae (control). The 5th instar larvae treated with M. anisopliae showed higher ECD and ECI values than control. Based on mortality and growth inhibition it can be suggested that all the studied fungal strains have a high potential for biocontrol and could be developed into biocontrol agents against O. varians.  相似文献

The American cockroach Periplaneta americana, one of the worlds' most important urban insect pests was tested with entomopathogenic fungi. Most promising Metarhizium anisopliae, Metarhizium robertsii and Beauveria bassiana killed nymphs (≥ 81.7% mortality, 25 days after treatment), and these fungi developed on all dead insects. Other fungi tested were less virulent (Metarhizium frigidum and Purpureocillium lilacinum) or avirulent (Isaria cateniobliqua, Isaria farinosa, Simplicillium lanosoniveum, Sporothrix insectorum and Tolypocladium cylindrosporum). Intrageneric and intraspecific variability of fungal activity was detected. Adults were highly susceptible, and oothecae proved to be more resistant than nymphs and adults to infection with M. anisopliae IP 46. Findings of the study underscore the potential of fungi as biocontrol agents against this pest.  相似文献

AdultGlossina morsitans morsitans exposed to wet conidia ofBeauveria bassiana andMetarhizium anisopliae suffered high mortalities ranging from 90 to 100% by 2 weeks post-exposure. Infected ♂ ♂ maintained in the same cages with non-infected ♀♀ throughout the experimental period transmitted the fungal infection to the ♀♀ resulting in mortalities of 65% withB. bassiana and 55% withM. anisopliae. Likewise, infected ♀♀ maintained together with non-infected ♂♂ transmitted the infection to the ♂♂ resulting in mortalities of 75% withB. bassiana and 45% withM. anisopliae. Female tsetse flies infected withB. bassiana andM. anisopliae and maintained in the same cages with non-infected ♀♀ also transmitted infection to the non-infected tsetse resulting in mortalities of 62% and 48% withB. bassiana andM. anisopliae respectively. Infected tsetse exposed to non-infected tsetse of the opposite sex for only 30 min were also able to transmit the fungal infection. Pupae produced by female tsetse infected withB. bassiana andM. anisopliae exhibited higher pupal mortality than those produced by non-infected ♀♀. However, pupae exposed directly to dry spores ofB. bassiana andM. anisopliae had no increase in pupal mortality but adults emerging from theB. bassiana-exposed pupae had markedly reduced longevity.   相似文献

The interactions between the entomopathogenic bacterium Bacillus thuringiensis ssp. kurstaki and two entomopathogenic fungi Beauveria bassiana Balsamo (Vuillemin) (Hypocreales: Cordycipitaceae) and Metarhizium robertsii (Metchnikoff) Sorokin (Hypocreales: Clavicipitaceae) were examined on larvae of Sesamia nonagrioides (Lefebvre) (Lepidoptera: Noctuidae) in 8, 13 and 16 days post-treatment intervals. An overall positive interaction between the pathogens was observed and the larval mortality at 16 days was 56–100 % exposed to M. robertsii combined with B. thuringiensis subsp. kurstaki, whereas B. bassiana combined with B. thuringiensis ssp. kurstaki killed 54–100 % of exposed larvae. After 8 days, in 6 of the combinations, we found an additive relationship between the pathogens, whereas, a negative interaction was observed in 10 of them. In contrast, after 13 days, in 2 of the combinations the positive interaction could be considered as synergistic between pathogens, in 10 as additive, and in only 4 as negative. Finally, after 16 days, in 11 of the combinations we found an additive connection between the pathogens, wheras a negative interaction was seen in 5. Applying both pathogens simultaneously offers a method of Sesamia nonagrioides control that could be more effective than using each pathogen separately.  相似文献

Leaf extracts of Capurnia aurea (Fabaceae) have been reported to attract ticks and can be considered for use in combination with entomopathogenic fungus in an autodissemination approach, whereby ticks that are attracted to baited traps are infected with the inoculum. Bioassays were undertaken to assess the compatibility of aqueous, methanol and acetone extracts of C. aurea with conidia of Metarhizium anisopliae sensu stricto (ss) (Hypocreales: Clavicipitaceae) isolate ICIPE 7 in terms of mycelial growth, conidial production and spore viability. Aqueous extract of C. aurea was compatible with the fungus at all the concentrations tested whereas methanol and acetone extracts inhibited all the fungal growth parameters. The virulence of M. anisopliae formulated in different extracts of C. aurea was also tested against different developmental stages of Rhipicephalus pulchellus in laboratory bioassays. No significant differences in virulence were observed between M. anisopliae applied alone and M. anisopliae formulated in different concentrations of extracts of C. aurea among the larvae, nymphs and adults. However, mortality varied according to the developmental stage, with larval stage being the most susceptible. Aqueous extract was compatible with M. anisopliae and could be therefore mixed together for ‘spot‐spray’ treatments as low cost and environmentally friendly technology for tick control in grazing pasture.  相似文献

Dermestes maculatus De Geer (Coleoptera: Dermestidae) is a carrion feeder that is also a pest of poultry houses, museums, silkworm culture, and many stored foods. The Hypocreales, Beauveria bassiana (Balsamo) Vuillemin, Metarhizium anisopliae (Metschnikoff), and Isaria fumosorosea Wize, were tested for efficacy against D. maculatus larvae on concrete, plastic, leather, and wood surfaces. Only wood had a significant negative effect on efficacy, and B. bassiana was the most efficacious species. The conidia of all three species lost viability quickly on wood with various responses on the other surfaces. When beetle larvae were exposed to deposited B. bassiana and incubated at 43, 56, 75, or 82% relative humidity, mortality was greatest at the lowest humidity suggesting enhancement of fungal infection by desiccation stress. The results indicate that entomopathogenic Hypocreales, especially B. bassiana, may be useful for control of hide beetles when applied at a low dose to surfaces that do not impact the viability of conidia.  相似文献
