Sexual dimorphisms in the dermal structures of two elasmobranch species have previously been reported and it has been linked to the use of the mouth by males during copulation. Until relatively recently, the fact, that male Scyliorhinus canicula use their mouths for grasping and biting females during copulation was unknown. This study reveals that not only do adult (M ≥ 525 mm, F ≥ 550 mm) S. canicula show a sexual dimorphism in the epidermis and dermis, but that hatchling S. canicula are born with a sexually dimorphic epidermal layer and this persists into the juvenile stage (M < 525 mm, F < 550 mm). A sexual dimorphism was found in all size classes with both hatchling and juvenile female S. canicula having significantly thicker epidermal layers than hatchling and juvenile male S. canicula. Adult female S. canicula were found to possess both a significantly thicker epidermal and dermal layer than adult male S. canicula. The presence of a sexual dimorphism in the epidermal and dermal layers of adult S. canicula could be directly related to reproductive behaviour in response to the male biting the female prior to copulation.  相似文献

The use of archival depth telemetry as a means of remotely assessing the reproductive rates of free‐ranging fishes is explored. This is achieved by electronically tracking the vertical movements of individual female small spotted catsharks Scyliorhinus canicula in the natural environment, whilst simultaneously evaluating the temporal and vertical distributions of egg‐laying in this species. Distinctive patterns of short‐term (0·3–3·7 h), shallow‐water activity are documented in the time–depth profiles of female S. canicula that occur at an appropriate depth (1·0–2·3 m) and periodicity (every 10–12 days) to represent egg‐laying behaviour. Putative egg‐laying behaviour was exhibited simultaneously by two individually tracked female S. canicula during late‐spring and early‐summer. The results highlight that, provided species behaviour is suitable and complementary methods such as previous data, laboratory experiments and field surveys can be used to validate the patterns observed, archival depth telemetry offers an unobtrusive means by which egg production and egg‐laying behaviour of free‐living fishes can be estimated. As precise information regarding life‐history parameters is difficult to obtain for free‐ranging fish species, this technique could be used to improve the parameterization of species demographic models that are relevant to the management of wild fish populations.  相似文献

Photo‐tagging, i.e. using a specific software to match colour patterns on photographs, was tested as a means to identify individual Indo‐Pacific Pterois volitans to assist with population and movement studies of this invasive species. The stripe pattern on the flank of adult P. volitans (n = 48) was the most individually distinctive of three body regions tested, leading to correct individual identification on 68 and 82% of tests with a single and two images of the reference individual, respectively. Photo‐tagging is inexpensive, logistically simple and can involve citizen scientists, making it a viable alternative to traditional tagging to provide information on P. volitans distribution, movement patterns and recolonization rates after removals.  相似文献

A dorsal‐fin photo‐identification technique paired with a non‐invasive parallel laser photogrammetry technique was used to non‐invasively identify individual Sphyrna mokarran over time. Based on the data collected over a duration of 59 days, 16 different S. mokarran (mean ± s.d . pre‐caudal length: 220·82 ± 13·66 cm; mean ± s.d . cephalofoil width: 71·38 ± 7·94 cm) were identified using dorsal‐fin photo‐identification, with a mean ± s.d . shark re‐sighting frequency of 4·05 ± 3·06 at‐sea days. The results illustrate a high S. mokarran sighting rate and therefore, the utilization of parallel laser photogrammetry and dorsal‐fin photo‐identification may be a plausible multi‐year approach to aid in non‐invasively determining the growth rate and inter‐annual site fidelity of these animals.  相似文献

Functions of α‐ and β‐branch carotenoids in whole‐plant acclimation to photo‐oxidative stress were studied in Arabidopsis thaliana wild‐type (wt) and carotenoid mutants, lut ein deficient (lut2, lut5), n on‐p hotochemical q uenching1 (npq1) and s uppressor of z eaxanthin‐l ess1 (szl1) npq1 double mutant. Photo‐oxidative stress was applied by exposing plants to sunflecks. The sunflecks caused reduction of chlorophyll content in all plants, but more severely in those having high α‐ to β‐branch carotenoid composition (α/β‐ratio) (lut5, szl1npq1). While this did not alter carotenoid composition in wt or lut2, which accumulates only β‐branch carotenoids, increased xanthophyll levels were found in the mutants with high α/β‐ratios (lut5, szl1npq1) or without xanthophyll‐cycle operation (npq1, szl1npq1). The PsbS protein content increased in all sunfleck plants but lut2. These changes were accompanied by no change (npq1, szl1npq1) or enhanced capacity (wt, lut5) of NPQ. Leaf mass per area increased in lut2, but decreased in wt and lut5 that showed increased NPQ. The sunflecks decelerated primary root growth in wt and npq1 having normal α/β‐ratios, but suppressed lateral root formation in lut5 and szl1npq1 having high α/β‐ratios. The results highlight the importance of proper regulation of the α‐ and β‐branch carotenoid pathways for whole‐plant acclimation, not only leaf photoprotection, under photo‐oxidative stress.  相似文献

Photo‐electrochemical (PEC) solar energy conversion offers the promise of low‐cost renewable fuel generation from abundant sunlight and water. In this Review, recent developments in photo‐electrochemical water splitting are discussed with respect to this promise. State‐of‐the‐art photo‐electrochemical device performance is put in context with the current understanding of the necessary requirements for cost‐effective solar hydrogen generation (in terms of solar‐to‐hydrogen conversion efficiency and system durability, in particular). Several important studies of photo‐electrochemical hydrogen generation at p‐type photocathodes are highlighted, mostly with protection layers (for enhanced durability), but also a few recent examples where protective layers are not needed. Recent work with the widely studied n‐type BiVO4 photoanode is detailed, which highlights the needs and necessities for the next big photoanode material yet to be discovered. The emerging new research direction of photo‐electrocatalytic upgrading of biomass substrates toward value‐added chemicals is then discussed, before closing with a commentary on how research on PEC materials remains a worthwhile endeavor.  相似文献

Photo‐current loss in donor‐acceptor (DA) polymer‐fullerene bulk heterojunction solar cells was studied via carrier transport and recombination measurements. Focusing on the DA polymer poly((4,4‐dioctyldithieno (3,2‐b:2',3'‐d) silole)‐2,6‐diyl‐alt‐(2,1,3‐benzothiadiazole)‐4,7‐diyl) (DTS‐BTD) we found that the carrier transport is well‐balanced and attribute the loss mechanism in DTS‐BTD solar cells to carrier recombination. Using carrier extraction with linear increasing voltage (photo‐CELIV) and transient photo‐voltage (TPV), we show that carrier recombination plays an important role in photo‐current extraction at open circuit conditions due to increase in photo‐excited carrier concentration. Delay time dependent photo‐CELIV and temperature dependent transport studies suggest that the recombination rate is related to the degree of energetic disorder in the polymer: fullerene blends.  相似文献

Brain arteriovenous malformations (AVMs) which associate with angiogenesis due to local hypertension, chronic cerebral ischaemia and tissue hypoxia usually lead to haemorrhage, however, the therapeutic medicine for the disease is still lacking. 2‐methoxyestradiol (2‐ME) has been shown effective in the anti‐angiogenic treatment. This study was conducted to examine whether and how 2‐ME could improve the vascular malformations. Intracranial venous hypertension (VH) model produced in adult male Sprague‐Dawley rats and culture of human umbilical vein endothelial cells (HUVECs) at the anoxia condition were used to induce in vivo and in vitro angiogenesis, respectively. Lentiviral vectors of ID‐1 and p53 genes and of their siRNA were intracranially injected into rats and transfected into HUVECs to overexpress and down‐regulate these molecules. 2‐ME treatment not only reduced the in vivo progression of brain tissue angiogenesis in the intracranial VH rats and the in vitro increases in microvasculature formation, cellular migration and HIF‐1α expression induced by anoxia in HUVECs but also reversed the up‐regulation of ID‐1 and down‐regulation of p53 in both the in vivo and in vitro angiogenesis models. All of the anti‐angiogenesis effects of 2‐ME observed in VH rats and anoxic HUVECs were abrogated by ID‐1 overexpression and p53 knockdown. Our data collectively suggest that 2‐ME treatment inhibits hypoxia/anoxia‐induced angiogenesis dependently on ID‐1 down‐regulation and p53 up‐regulation, providing a potential alternative medical treatment for un‐ruptured AVM patients.  相似文献

The aims of this work were to (a) evaluate the susceptibility of endospores of Bacillus cereus, B. licheniformis, B. sphaericus and B. subtilis to photodynamic inactivation using a tricationic porphyrin as photosensitizer, (b) assess the efficiency of adsorption of the photosensitizer in endospore material as a determinant of the susceptibility of endospores of different Bacillus species to photo‐inactivation, (c) determine the value of B. cereus as a model organism for studies of antimicrobial photodynamic inactivation of bacterial endospores. The results of irradiation experiments with endospores of four species of Bacillus showed that B. cereus was the only species for which efficient endospore photo‐inactivation (> 3 log reduction) could be achieved. Endospores of B. licheniformis, B. sphaericus and B. subtilis were virtually resistant to photo‐inactivation with tricationic porphyrin. The amount of porphyrin bound to endospore material was not significantly different between species, regardless of the presence of an exosporium or exosporium‐like outer layer. The sensitivity of endospores to photodynamic inactivation with a tricationic porphyrin is highly variable among different species of the genus Bacillus. The presence of an exosporium in endospores of B. cereus and B. sphaericus, or an exosporium‐like glycoprotein layer in endospores of B. subtilis, did not affect the amount of bound photosensitizer and did not explain the inter‐species variability in susceptibility to photodynamic inactivation. The results imply that the use of B. cereus as a more amenable surrogate of the exosporium‐producing B. anthracis must be carefully considered when testing new photosensitizers for their antimicrobial photo‐inactivation properties.  相似文献

We evaluated sex, age, nutritional status, and infectious disease (ID) as predictors of two biomarkers of cell‐mediated immunity (CMI), delayed‐type hypersensitivity to Candida albicans (DTH‐Candida), and anti‐Epstein‐Barr virus antibody (EBV Ab), among 200 children in Kilimanjaro, Tanzania. DTH‐Candida, which decreases with compromised CMI, was positively associated with age (OR: 1.27; 95% CI: 1.02, 1.57) and triceps skinfold (TSF; OR: 1.16; 95% CI: 1.02, 1.26), and inversely associated with height‐for‐age Z score (HAZ; OR: 0.86; 95% CI: 0.68, 1.08) and diagnosed ID (OR: 0.48; 95% CI: 0.22, 1.08). There was significant interaction between TSF and ID: DTH‐Candida exhibited a strong inverse association with ID among children with low TSF (OR: 0.16; 95% CI: 0.05, 0.50) and a strong positive association with TSF among children with ID (OR: 2.64; 95% CI: 1.29, 5.42). EBV Ab, which increases with compromised CMI, was inversely associated with male sex (β: ?0.47; 95% CI: ?0.70, ?0.24) and TSF (β: ?0.04; 95% CI: ?0.08, 0.00), and positively associated with HAZ (β: 0.06; 95% CI: ?0.03, 0.15). Among males, EBV Ab was positively associated with anemia. Among normal HAZ children, EBV Ab was inversely associated with TSF. There was no association between DTH‐Candida and EBV Ab. While DTH‐Candida provides a direct measure of CMI, our results suggest that interpretation of EBV‐Ab among Kilimanjaro children was complicated by its indirect relationship with CMI. Among our sample, CMI increased with age and adequate nutrition and was compromised during acute ID. The suggestive CMI‐compromising effect of increasing height‐for‐age may bear further exploration. The cosmopolitan, bloom‐forming diatom, Skeletonema costatum, is a prominent primary producer in coastal oceans, fixing CO2 with ribulose 1,5‐bisphosphate carboxylase/oxygenase (RubisCO) that is phylogenetically distinct from terrestrial plant RubisCO. RubisCOs are subdivided into groups based on sequence similarity of their large subunits (IA–ID, II, and III). ID is present in several major oceanic primary producers, including diatoms such as S. costatum, coccolithophores, and some dinoflagellates, and differs substantially in amino acid sequence from the well‐studied IB enzymes present in most cyanobacteria and in green algae and plants. Despite this sequence divergence, and differences in isotopic discrimination apparent in other RubisCO enzymes, stable carbon isotope compositions of diatoms and other marine phytoplankton are generally interpreted assuming enzymatic isotopic discrimination similar to spinach RubisCO (IB). To interpret phytoplankton δ13C values, S. costatum RubisCO was characterized via sequence analysis, and measurement of its KCO2 and Vmax, and degree of isotopic discrimination. The sequence of this enzyme placed it among other diatom ID RubisCOs. Michaelis‐Menten parameters were similar to other ID enzymes (KCO2 = 48.9 ± 2.8 μm ; Vmax = 165.1 ± 6.3 nmol min?1 mg?1). However, isotopic discrimination (ε = [12k/13k ? 1] × 1000) was low (18.5‰; 17.0–19.9, 95% CI) when compared to IA and IB RubisCOs (22–29‰), though not as low as ID from coccolithophore, Emiliania huxleyi (11.1‰). Variability in εvalues among RubisCOs from primary producers is likely reflected in δ13C values of oceanic biomass. Currently, δ13C variability is ascribed to physical or chemical factors (e.g. illumination, nutrient availability) and physiological responses to these factors (e.g. carbon‐concentrating mechanisms). Estimating the importance of these factors from δ13C measurements requires an accurate εvalue, and a mass‐balance model using the εvalue for S. costatum RubisCO is presented. Clearly, appropriate εvalues must be included in interpreting δ13C values of environmental samples.  相似文献

The red imported fire ant, Solenopsis invicta Buren, is an important medical, agricultural, and ecological pest with a global distribution. The main food source of S. invicta is liquid carbohydrates (e.g., plant exudates and honeydew excreted by Hemipterans). This species evolved many strategies to forage droplets of liquid food. The photo shows that S. invicta workers surrounded and fed along the edge of the sucrose water droplet in the field. The diagrammatic sketch (at the bottom of the figure) shows how ants stacked to feed the droplet in the laboratory. After the edge of the droplet was ‘saturated’ by surrounding ants, the stacking ants stepped on other individuals and climbed onto the droplet (see pages 499–507). An automated multi‐pumping flow system is proposed for the chemiluminometric determination of ascorbic acid in pharmaceutical formulations, relying on the ability of semiconductor nanocrystals to generate short‐lived reactive species upon photo‐irradiation. A photo‐unit based on visible‐light‐emitting diodes is used to photo‐excite cadmium telluride (CdTe) quantum dots capped with glutathione, leading to the generation of radicals that react with luminol under alkaline conditions, yielding the chemiluminescence. Ascorbic acid acts as a radical scavenger, preventing the oxidation of luminol, thus ensuring a concentration‐dependent chemiluminescence quenching. After system optimization, a linear working range of 5.0 × 10‐7 to 5.0 × 10‐6 mol/L ascorbic acid (r = 0.9967, n = 5) was attained, with a detection limit of 3.05 × 10‐7 mol/L and a sampling rate of 200/h. The flow system was applied to the analysis of pharmaceutical formulations and the results were in good agreement with those obtained by the reference titrimetric procedure (RD < ± 4.3%, n = 7). Recognition of individuals within an animal population is central to a range of estimates about population structure and dynamics. However, traditional methods of distinguishing individuals, by some form of physical marking, often rely on capture and handling which may affect aspects of normal behavior. Photographic identification has been used as a less‐invasive alternative, but limitations in both manual and computer‐automated recognition of individuals are particularly problematic for smaller taxa (<500 g). In this study, we explored the use of photographic identification for individuals of a free‐ranging, small terrestrial reptile using (a) independent observers, and (b) automated matching with the Interactive Individual Identification System (I3S Pattern) computer algorithm. We tested the technique on individuals of an Australian skink in the Egernia group, Slater's skink Liopholis slateri, whose natural history and varied scale markings make it a potentially suitable candidate for photo‐identification. From ‘photographic captures’ of skink head profiles, we designed a multi‐choice key based on alternate character states and tested the abilities of observers — with or without experience in wildlife survey — to identify individuals using categorized test photos. We also used the I3S Pattern algorithm to match the same set of test photos against a database of 30 individuals. Experienced observers identified a significantly higher proportion of photos correctly (74%) than those with no experience (63%) while the I3S software correctly matched 67% as the first ranked match and 83% of images in the top five ranks. This study is one of the first to investigate photo identification with a free‐ranging small vertebrate. The method demonstrated here has the potential to be applied to the developing field of camera‐traps for wildlife survey and thus a wide range of survey and monitoring applications.  相似文献

A series of novel alkyl substituted purines were synthesized. 6‐[4‐(4‐Propoxyphenyl)piperazin‐1‐yl]‐9H‐purine was used as the key starting material, which was synthesized via a multistep protocol and finally subjected for N‐alkylation with various alkyl halides with an aim to get prospective antimicrobial agents. The structures of the novel compounds were established by substantiating them through spectral techniques like 1H‐NMR, 13C‐NMR, FT‐IR and EI‐MS. They were explored for antitubercular activity against Mycobacterium tuberculosis H37RV. Furthermore, they were checked for their antimicrobial activity concerning bacterial and fungal strains. The title compounds exhibited considerable antimicrobial activity without any significant toxicity. In silico studies depicted their good binding profile against Mycobacterium tuberculosis enoyl reductase (InhA; PDB ID: 4TZK) and Candida albicans dihydrofolate reductase (PDB ID: 1AI9). The title compounds obeyed Lipinski's parameters and have exhibited good drug‐like properties.  相似文献

For the enantiopure synthesis of novel chiral GABA uptake inhibitors, nipecotic acid ( 1 ) is an important key precursor. To characterize accurately the pharmacological activity of these interesting target compounds, the determination of the correct enantiomeric purity of nipecotic acid as the starting material is indispensable. In this report, a sensitive high‐performance liquid chromatography (HPLC) based method for the separation and quantitation of both enantiomers of nipecotic acid as 1‐(7‐nitrobenzo[c ][1,2,5]oxadiazol‐4‐yl) derivatives ( 5 ) on a Chiralpak ID‐3 column (Daicel, Illkirch, France) was established. UV/Vis‐detection at 490 nm was chosen to ensure a selective determination of even highly enantioenriched samples. Reliability was demonstrated by validation of specificity, linearity, lower limit of quantification (LLOQ), accuracy, and precision. By spiking highly enantiopure samples with small amounts of racemic rac ‐ 5 , it was proven that the established HPLC method is able to detect even slight changes in enantiomeric excess (ee) values. Thus, accurate determination of ee values up to 99.87% ee for (R )‐ 5 and 99.86% ee for (S )‐ 5 over a linear concentration range of 11500 μM for (R )‐ 5 and of 11455 μM for (S )‐ 5 could be demonstrated.  相似文献

The spotted eagle ray Aetobatus narinari is characterized by pigmentation patterns that are retained for up to 3·5 years. These pigmentations can be used to identify individuals through photo‐identification. Only one study has validated this technique, but no study has estimated the percentage of correct identification of the rays using this technique. In order to carry out demographic research, a reliable photographic identification technique is needed. To achieve this validation for A. narinari, a double‐mark system was established over 11 months and photographs of the dorsal surface of 191 rays were taken. Three body parts with distinctive natural patterns were analysed (dorsal surface of the cephalic region, dorsal surface of the pectoral fins and dorsal surface of the pelvic fins) in order to determine the body part that could be used to give the highest percentage of correct identification. The dorsal surface of the pectoral fins of A. narinari provides the most accurate photo‐identification to distinguish individuals (88·2%).  相似文献

The small spotted dogfish Scyliorhinus canicula and the blackmouth dogfish Galeus melastomus, whose depth distributions overlap in the upper part of the slope (c. 500 m depth), where they have access to the same prey community, have well‐developed eyes and a pure‐rod retina with a single layer of photoreceptors. Interspecific differences in rod outer segment length (LROS) within retinal regions were found. In the periphery and the retinal centre G. melastomus showed a LROS 24 and 30% longer, respectively, than S. canicula and, therefore, a potential for increased sensitivity. In both species longer LROS were always found in correspondence with the retinal centre where the ganglion cell topography formed a horizontal meridian that allowed for better discrimination of the horizon in the visual field. In this area LROS reached 53·4±4·1μm in S. canicula and 77·1±10·5μm in G. melastomus against 46·3±4·2μm and 61·1±10·1μm in the retinal periphery. No significant differences were recorded in LROS and rod density during growth. In both species, a rapid increase of theoretical visual acuity was found to be related to an increase in fish LT and lens size. Visual acuity ranged between 1·7 and 3 cycles degree‐1 in S. canicula and 2·4 and 4·2 in G. melastomus. The G. melastomus rod visual pigment showed the characteristic spectral adaptation to vision in deep‐water (λmax of 481 nm), but was also well placed to detect the bioluminescence of some of its main prey species. In S. canicula the visual pigment absorption (λmax of 496 nm) was more typical of shallow water living fishes. The opsin sequences of the two visual pigments are discussed and key amino acid sites were identified where sequence changes could be responsible for the spectral absorption differences between the two species. The possible relationship between LROS, visual acuity, visual pigment absorption, depth distribution and feeding behaviour are discussed.  相似文献

Many flowering plants show self‐incompatibility, an intra‐specific reproductive barrier by which pistils reject self‐pollen to prevent inbreeding and accept non‐self pollen to promote out‐crossing. In Petunia, the polymorphic S–locus determines self/non‐self recognition. The locus contains a gene encoding an S–RNase, which controls pistil specificity, and multiple S‐locus F‐box (SLF) genes that collectively control pollen specificity. Each SLF is a component of an SCF (Skp1/Cullin/F‐box) complex that is responsible for mediating degradation of non‐self S‐RNase(s), with which the SLF interacts, via the ubiquitin–26S proteasome pathway. A complete set of SLFs is required to detoxify all non‐self S‐RNases to allow cross‐compatible pollination. Here, we show that SLF1 of Petunia inflata is itself subject to degradation via the ubiquitin–26S proteasome pathway, and identify an 18 amino acid sequence in the C‐terminal region of S2‐SLF1 (SLF1 of S2 haplotype) that contains a degradation motif. Seven of the 18 amino acids are conserved among all 17 SLF proteins of S2 haplotype and S3 haplotype involved in pollen specificity, suggesting that all SLF proteins are probably subject to similar degradation. Deleting the 18 amino acid sequence from S2‐SLF1 stabilized the protein but abolished its function in self‐incompatibility, suggesting that dynamic cycling of SLF proteins is an integral part of their function in self‐incompatibility.  相似文献

Encephalitozoon cuniculi is a unicellular, obligate intracellular eukaryotic parasite in the Microsporidia family and one of the agents responsible for microsporidosis infections in humans. Like most Microsporidia, the genome of E. cuniculi is markedly reduced and the organism contains mitochondria‐like organelles called mitosomes instead of mitochondria. Here we report the solution NMR structure for a protein physically associated with mitosome‐like organelles in E. cuniculi, the 128‐residue, adrenodoxin‐like protein Ec‐Adx (UniProt ID Q8SV19) in the [2Fe‐2S] ferredoxin superfamily. Oxidized Ec‐Adx contains a mixed four‐strand β‐sheet, β2‐β1‐β4‐β3 (↓↑↑↓), loosely encircled by three α‐helices and two 310‐helices. This fold is similar to the structure observed in other adrenodoxin and adrenodoxin‐like proteins except for the absence of a fifth anti‐parallel β‐strand next to β3 and the position of α3. Cross peaks are missing or cannot be unambiguously assigned for 20 amide resonances in the 1H‐15N HSQC spectrum of Ec‐Adx. These missing residues are clustered primarily in two regions, G48‐V61 and L94‐L98, containing the four cysteine residues predicted to ligate the paramagnetic [2Fe‐2S] cluster. Missing amide resonances in 1H‐15N HSQC spectra are detrimental to NMR‐based solution structure calculations because 1H‐1H NOE restraints are absent (glass half‐empty) and this may account for the absent β‐strand (β5) and the position of α3 in oxidized Ec‐Adx.
