Three fossil coral samples (Pocillopora elegans and Pocillopora damicornis) were collected from a beachrock in Puerto Escondido, central Pacific coast of Costa Rica. The age of the samples ranged from 1272+/-60 to 2010+/-60 years BP. They represent some of the few Holocene fossil corals known from the Pacific coast of the Americas. The uplift rate for the central Pacific coast of Costa Rica was calculated in 0.4 to 2.1 mm year(-1).  相似文献

Coral-associated microbial communities from three coral species (Pocillopora damicornis, Acropora tenuis and Favites abdita) were examined every 3 months (January, March, June, October) over a period of 1 year on Ningaloo Reef, Western Australia. Tissue from corals was collected throughout the year and additional sampling of coral mucus and seawater samples was performed in January. Tissue samples were also obtained in October from P. damicornis coral colonies on Rottnest Island off Perth, 1200 km south of Ningaloo Reef, to provide comparisons between coral-microbial associates in different locations. The community structures of the coral-associated microorganisms were analysed using phylogenetic analysis of 16S rRNA gene clone libraries, which demonstrated highly diverse microbial profiles among all the coral species sampled. Principal component analysis revealed that samples grouped according to time and not species, indicating that coral-microbial associations may be a result of environmental drivers such as oceanographic characteristics, benthic community structure and temperature. Tissue samples from P. damicornis at Rottnest Island revealed similarities in bacteria to the samples at Ningaloo Reef. This study highlights that coral-associated microbial communities are highly diverse; however, the complex interactions that determine the stability of these associations are not necessarily dependent on coral host specificity.  相似文献

To examine the response to exposure to a thermal gradient in coral, we assessed the effect of a gradual 10 degrees C temperature increase (22 to 32 degrees C over 10 h) on normal (N), partially bleached (P) and control (C) samples collected from different branches of the same coral (Pocillopora capitata). We examined markers of oxidative stress, including lipid peroxidation (MDA) and superoxide dismutase (SOD) activity, indicators of bleaching, including chlorophyll a (Chl a) and carotenoid pigment (PC) levels, as well as zooxanthellae density. Our results revealed that N, P and C coral samples all contained higher levels of PC versus Chl a. The levels of both pigments increased as the temperature increased from 22 to 28 degrees C only in N and C samples, whereas P samples showed less cellular damage than N and C samples at temperatures between 26 and 28 degrees C, and had greater antioxidant activities at temperatures between 26 and 30 degrees C. The rate of zooxanthellar expulsion consistently increased with temperature in all three coral types across the entire temperature range. Collectively, these results indicate that temperature has a direct effect on the antagonistic relationship between temperature-induced damage and protective antioxidant mechanisms in this type of coral.  相似文献

Coral bleaching is the disruption of symbioses between coral animals and their photosynthetic microalgal endosymbionts (zooxanthellae). It has been suggested that large-scale bleaching episodes are linked to global warming. The data presented here demonstrate that Vibrio coralliilyticus is an etiological agent of bleaching of the coral Pocillopora damicornis. This bacterium was present at high levels in bleached P. damicornis but absent from healthy corals. The bacterium was isolated in pure culture, characterized microbiologically, and shown to cause bleaching when it was inoculated onto healthy corals at 25 degrees C. The pathogen was reisolated from the diseased tissues of the infected corals. The zooxanthella concentration in the bacterium-bleached corals was less than 12% of the zooxanthella concentration in healthy corals. When P. damicornis was infected with V. coralliilyticus at higher temperatures (27 and 29 degrees C), the corals lysed within 2 weeks, indicating that the seawater temperature is a critical environmental parameter in determining the outcome of infection. A large increase in the level of the extracellular protease activity of V. coralliilyticus occurred at the same temperature range (24 to 28 degrees C) as the transition from bleaching to lysis of the corals. We suggest that bleaching of P. damicornis results from an attack on the algae, whereas bacterium-induced lysis and death are promoted by bacterial extracellular proteases. The data presented here support the bacterial hypothesis of coral bleaching.  相似文献

The effect of different photon flux densities (PFD) and temperatures on the relative growth rate (RGR) and the concentration of three halogenated monoterpenes in samples of Plocamium cartilagineum L.( Dixon), a marine alga (Rhodophyceae), were studied. The highest RGR (22.8 +/- 0.04 d(-1)) was obtained at 15 degreebC and 41 ,mol m(-2) s)(-1) of PFD and the lowest (18.0 +/- 0.12 d(-1)) was obtained at 18 degrees C and 120 micromol m(-2) s(-1). The different temperatures and light used in assays did not affect significantly the production of organic compounds. The production of mertensene and violacene was not affected significantly. However, compound 1 reached the highest concentration at 15 degrees C and 65 micromol m(-2) s(-1)). The relationship between growth and production of monoterpenes of P. cartilagineum and the effect of temperature and the PFD were analyzed.  相似文献

Reef-building scleractinian corals widely engage in symbiotic relationships with Symbiodinium dinoflagellates (zooxanthellae), which reside inside cells of the gastrodermis. In most cases, sexually produced larvae acquire their symbionts from the environment in the early developmental stages preceding settlement; however, some scleractinian corals maternally "seed" their oocytes with symbionts, and these symbionts are reported to be restricted to the gastrodermis at the time of its formation (gastrulation). A precise mechanism for how Symbiodinium are translocated to endoderm in these seeded species was previously unknown. In order to examine the process of endoderm formation and Symbiodinium localization during gastrulation, we have examined two species of "robust" clade scleractinians: Fungia scutaria (nonseeded) and Pocillopora meandrina (maternally seeded). We determined that both species, independent of whether or not they are seeded, undergo a "nutritive" stage before gastrulation, wherein lipid-rich cells (F. scutaria) or membrane-bound cellular fragments (P. meandrina) are passed to the blastocoel where they are subsequently taken up by the definitive endoderm. This emergent property of anthozoan development has been co-opted to facilitate the movement of Symbiodinium to the blastocoel (future site of endoderm), in the seeded species, where they are later phagocytosed by the newly formed definitive endoderm. Additionally, both species of robust clade scleractinians examined gastrulate by way of invagination, as do the majority of anthozoans. This invagination differs from the prawn chip-type gastrulation seen in the complex clade corals and provides evidence for a possible linkage between gastrulation type and phylogenetic history.  相似文献

O(2) evolution from single turnover flashes of up to 96 micromol absorbed quanta m(-2) and from multiple turnover pulses of 8.6 and 38.6 ms duration and 12800 and 850 micromol absorbed quanta m(-2) s(-1) intensity, respectively, was measured in sunflower leaves with the help of zirconium O(2) analyser. O(2) evolution from one flash could be measured with 1% accuracy on the background of 10-50 micromol O(2) mol(-1). Before the measurements leaves were pre-adapted either at 30-60 or 1700 micromol quanta m(-2) s(-1) to induce different non-photochemical excitation quenching (q(N)). Short (1 min) exposures at the high light that created only energy-dependent, q(E) type quenching, caused no changes in the O(2) yield from saturating flashes or pulses that could be related to the q(E) quenching, but the yield from low intensity flashes and pulses decreased considerably. Long 30-60-min exposures at the high light induced a reversible inhibitory, q(I) type quenching that decreased the O(2) yield from both, saturating and limiting flashes and pulses (but more from the limiting ones), which reversed within 15 min under the low light. The results are in agreement with the notion that q(E) is caused by a quenching process in the PSII antenna and no changes occur in the PSII centres, but the reversible (15-30 min) q(I) quenching is accompanied by inactivation of a part of PSII centres.  相似文献

The morphology (mainly prosthecae length), ultrastructure, and antenna pigment composition of the green sulfur bacterium Prosthecochloris aestuarii changed when grown under different light intensities. At light intensities of 0.5 and 5 micromol quanta m(-2) s(-1), the cells had a star-like morphology. Prosthecae, the characteristic appendages of the genus Prosthecochloris, were 232 nm and 194 nm long, respectively. In contrast, when grown at 100 micromol quanta m(-2) s(-1), these appendages were shorter (98 nm) and the cells appeared more rod-shaped. Transmission electron microscopy revealed a significant decrease in the cell perimeter to area ratio and in the number of chlorosomes per linear microm of membrane as light intensity increased. In addition to these morphological and ultrastructural responses, Prosthecochloris aestuarii exhibited changes in its pigment composition as a function of light regime. Lower specific pigment content and synthesis rates were found in cultures grown at light intensities above 5 micromol quanta m(-2) s(-1). A blue shift in the bacteriochlorophyll (BChl) c Q(y) absorption maximum of up to 17.5 nm was observed under saturating light conditions (100 micromol quanta m(-2) s(-1)). This displacement was accompanied by changes in the composition of BChl c homologs and by a very low carotenoid content. The morphological, ultrastructural and functional changes exhibited by Prosthecochloris aestuarii revealed the strong light-response capacity of this bacterium to both high and low photon-flux densities.  相似文献

Bacteria associated with three coral species, Acropora tenuis, Pocillopora damicornis and Tubastrea faulkneri, were assessed before and after coral mass spawning on Ningaloo Reef in Western Australia. Two colonies of each species were sampled before and after the mass spawning event and two additional samples were collected for P. damicornis after planulation. A variable 470 bp region of the 16 S rRNA gene was selected for pyrosequencing to provide an understanding of potential variations in coral-associated bacterial diversity and community structure. Bacterial diversity increased for all coral species after spawning as assessed by Chao1 diversity indicators. Minimal changes in community structure were observed at the class level and data at the taxonomical level of genus incorporated into a PCA analysis indicated that despite bacterial diversity increasing after spawning, coral-associated community structure did not shift greatly with samples grouped according to species. However, interesting changes could be detected from the dataset; for example, α-Proteobacteria increased in relative abundance after coral spawning and particularly the Roseobacter clade was found to be prominent in all coral species, indicating that this group may be important in coral reproduction.  相似文献

The adults of many coral reef fish species are site-attached, and their habitat is selected at the time of settlement by their larvae. The length of the planktonic larval period varies both intra- and interspecifically, and it is unknown how the age and size of larvae may affect their selection of habitat. To investigate the influence of age and size on habitat selection, I collected newly settled Hawaiian domino damselfish, Dascyllus albisella, daily from grids containing three coral species at four locations in Kaneohe Bay, Oahu, Hawaii. I recorded the coral species each fish was collected on, and measured and aged (by otoliths) the collected fish. The results indicate that the coral Pocillopora meandrina was selected by settling fish significantly more than the other two coral species. Younger and smaller larvae selected this coral species more frequently than older/larger larvae. In addition, younger/smaller individuals were found more commonly inside the bay than older/larger settling larvae. Differences in the choice of coral species and location of settlement may be partly due to ontogenetic differences in the sensory capacities of larvae to detect corals, conspecifics, and predators, or to a larval competency period. The genotypic composition of populations of the asexually viviparous coral Pocillopora damicornis varies in a manner that challenges classical models of the roles of sexual and asexual reproduction. On the geographically isolated Hawaiian reefs and high latitude reefs in Western Australia, P. damicornis populations are highly clonal although it has been argued that sexual reproduction via broadcast spawning generates widely dispersed colonists. In contrast, on eastern Australia's tropical Great Barrier Reef populations show little evidence of clonality. Here, we compare the genotypic diversity of adult and juvenile colonies of P. damicornis at seven sites on eastern Australia's high latitude Lord Howe Island reefs to determine if levels of clonality vary with habitat heterogeneity and age of colonies (as predicted by theory) or alternatively if clonality is again always high as for other isolated reef systems. We found 55-100% of the genotypic diversity expected for random mating at all seven sites and little evidence of asexual recruitment irrespective of habitat heterogeneity (sheltered versus wave exposed) or colony age. We found reduced levels of genetic diversity compared with tropical reefs (2.75 versus 4 alleles/locus), which supports earlier findings that Lord Howe Island is an isolated reef system. Furthermore, heterozygote deficits coupled with significant genetic subdivision among sites (FST=0.102+/-0.03) is typical of populations that have limited larval connections and are inbred. We conclude that the genetic structure of P. damicornis at Lord Howe Island reflects populations that are maintained through localised recruitment of sexually produced larvae.  相似文献

Coral reefs in the Tropical Eastern Pacific (TEP) are among the most isolated in the world. This isolation has resulted in relatively low species diversity but comparatively high endemism. The dominant reef-building corals of the TEP are the Pocillopora corals, a ubiquitous Indo-Pacific genus commonly regarded as inferior reef-builder. In addition to being the dominant reef-builders in the TEP, the Pocilloporids have undergone a reproductive shift from internally brooding larvae through most of their Indo-Pacific range to free-spawning in the TEP. Using genetic data from the internally transcribed spacer (ITS) regions of the nuclear ribosomal DNA gene cluster, we show here that this apparent reproductive shift coincides with interspecific hybridization among TEP Pocillopora species. We document a pattern of one-way gene flow into the main TEP reef builder P. damicornis from one or both of its TEP congeners — P. eydouxi and P. elegans . Our data provide preliminary evidence that trans-Pacific gene flow in P. damicornis between the Central and Eastern Pacific is restricted as well (ΦST = 0.419, P  < 0.0001). In combination, these results suggest that Eastern Pacific corals exist in relative isolation from their Central Pacific counterparts and interact with each other differently via hybridization.  相似文献

Large membrane-bound inclusions were clearly visible within the gastrodermis and lipid-containing cells of planulae and settled larvae of the zooxanthellate coral, Pocillopora damicornis after fixation or freeze-substitution. We suggest that these inclusions may be a novel potassium (kalium) chloride concentrating organelle, for which we propose the name kalisome. The inclusions were more abundant in settled larvae than in planulae and were not present in mature polyps. In planulae of the azooxanthellate coral, Dendrophyllia sp. these inclusions were extremely rare. Quantitative X-ray microanalysis of freeze-substituted preparations showed that the inclusions in P. damicornis settled larvae contained very high, positively correlated, concentrations of K (2.5 mol x kg(-1)) and Cl (2.5 mol x kg(-1)). Lower concentrations of both K (1.2 mol x kg(-1)) and Cl (1.3 mol x kg(-1)) were detected in P. damicornis planulae, yet higher concentrations were measured in Dendrophyllia planulae (K=6.0 mol x kg(-1); Cl=5.1 mol x kg(-1)). No significant (P>0.05) differences in concentration were observed between inclusions in freeze-substituted and freeze-dried sections of planulae. Symbiotic algae (zooxanthellae) in P. damicornis planulae and settled larvae also contained deposits with high levels of K and Cl, but these were not positively correlated and no structures associated with them were retained by fixation. Significant (P<0.05) concentration differences were also observed between deposits in freeze-substituted and freeze-dried sections. However, similar to 'kalisomes,' zooxanthellae deposits were more abundant in settled larvae than planulae and absent in mature polyps. Higher concentrations of K and Cl were also detected in settled larvae (K=0.7 mol x kg(-1); Cl=1.1 mol x kg(-1)) in comparison to planulae (K=0.4 mol x kg(-1); Cl=0.5 mol x kg(-1)).  相似文献

Biochemical and molecular biomarkers tools are utilized as early warning signatures of contaminant exposure to target and non-target organisms. The objective of this study was to investigate the sublethal effects of iron chloride to the larvae of the lace coral Pocillopora damicornis by measuring a suit of oxidative-stress biomarkers. The larvae were exposed to a range of sublethal concentrations of iron chloride (0.01, 0.1, 1, 10, and 100 ppm) for seven days. With reference to oxidative stress biomarkers, the no-observed effect concentration (NOEC) and the lowest observed effect concentration (LOEC) of iron chloride were observed to be 0.01 and 100 ppm respectively. At the end of the seventh day the antioxidant status of the larvae was evaluated by the levels of glutathione (GSH), glutathione peroxidase (GPX), glutathione reductase (GR), and glutathione-S-transferase (GST), in both experimental and control groups. For the quantification of cellular oxidative damage, lipid peroxidation (LPO) activity was determined in the same and the extent of DNA damage was assessed by the expression of DNA apurinic/apyrimidinic (AP) sites. Iron chloride exhibited a concentration-dependent inhibition of GSH and GPX and induction of GR, GST, LPO, and DNA-AP sites in the P. damicornis larvae when compared to the control group. The oxidative stress biomarkers of the larvae exposed to 0.1, 1, and 10 ppm of iron chloride did not show any significant overall differences when compared to the control group. However the activities of LPO, GSH, GPX, GR, GST and DNA-AP in the larval group exposed to 100 ppm of iron chloride exhibited statistically significant (P=0.002, 0.003, 0.002, 0.002, 0.005 and 0.007) differences when compared to the control group. The research results indicated that iron chloride in concentrations at the 100 ppm level caused oxidative stress in the P. damicornis larvae.  相似文献

Limitations of carbon fixation within spinach leaves due to light and CO2 were investigated. Under equivalent photon fluxes, carbon fixation was higher when leaves were irradiated adaxially compared to abaxially. Maximal carbon fixation occurred in the middle of the palisade mesophyll under adaxial illumination, whereas, maximal carbon fixation occurred in the spongy mesophyll under abaxial illumination. Total carbon fixation and the pattern of carbon fixation across leaves were similar, when leaves were irradiated with 800 micromol quanta m(-2) s(-1) either adaxially alone or adaxially plus abaxially (1,600 micromol quanta m(-2) s(-1)). In contrast, when both leaf surfaces were irradiated simultaneously with 200 micromol quanta m(-2) s(-1), total carbon fixation increased and carbon fixation in the middle of the leaf was higher compared to leaves irradiated unilaterally with the low light. Feeding 14CO2 through either the adaxial or abaxial leaf surface did not change the pattern of carbon fixation across the leaf. Increasing 14CO2 pulse-feeding times from 5 s to 60 s allowed more 14CO2 to be fixed but did not change the pattern of 14CO2 fixation across the leaf. We concluded that in spinach, the distribution of both light and Rubisco activity within leaves has significant effects on the patterns of carbon fixation across leaves; whereas CO2 diffusion does not appear to affect the carbon fixation pattern within spinach leaves.  相似文献

利用浮游生物Ⅰ(孔径505 μm)、Ⅱ(160 μm)、Ⅲ(77 μm)型拖网所得的10个站位、4个季节丰度数据,以浮游动物生物体积为单位划分粒级,探讨了国华电厂排水口附近海域浮游动物Sheldon型粒径谱和标准化粒径谱的时空变化特征,以期探明海域增温对浮游动物粒径谱影响.结果表明: 浮游动物个体体积范围为0.00012~127.0 mm3·ind-1,可划分为21个对数粒级组,对数范围为-13.06~6.99.据Sheldon型粒径谱结果,构成不同月份粒径谱主谱峰的主要种类有桡足幼体、墨氏胸刺水蚤、中华哲水蚤、仔鱼、百陶箭虫、拿卡箭虫和球型侧腕水母,小谱峰大多由个体较小的幼体类、剑水蚤类、针刺拟哲水蚤构成.在不同增温区断面中,桡足幼体、鱼卵和剑水蚤类等基本不受增温影响,而大型浮游动物,如百陶箭虫、拿卡箭虫、球型侧腕水母、中华哲水蚤和瓜水母等明显倾向于迁离排水口.从标准化粒径谱参数变化看,截距从低到高分别发生在11、2、5和8月;斜率变化以2月最小,5月与8月斜率相似且较大,表明2月小型浮游动物在群落中所占比例最高,而中大型浮游动物以5月和8月较高.在不同断面中,斜率以距离排水口0.2 km断面最低,且随着断面距离增加而增加,说明距离排水口越近,浮游动物小型化越明显.象山港标准化粒径谱年平均截距为4.68,斜率为-0.655.  相似文献

There are two sewage outfalls along the Jordanian coastline in the Gulf of Aqaba. During 1982 and 1983 a total of 328 core samples (0.01 m2 to a depth of 15 cm) were collected from the two outfalls, two control stations which resemble the outfalls in depth and sediment texture, and from two stations 100 m on both sides of each outfall. Faunal analysis revealed that the total number of individuals, number of species, species richness, and faunal similarities of macrobenthic invertebrates were lower at the sewage outfall near the phosphate loading port than the control station during both collections. At the 100 m stations, the numbers of individuals were generally higher than the sewage and control stations. The number of species, however, was highest at the control station and lowest at the sewage outfall. At the other sewage outfall, where the sewage effluent is discharged sporadically, no measurable effects on macrobenthic invertebrates were found.  相似文献

The aim of this work was to analyse the effect of temperature in three chestnut cultivars, Aveleira, Judia and Longal. For this purpose, gas exchange, thylakoid membrane potential, photosynthetic pigment and lipid content data in July, September and October under different temperatures (31, 26 and 18 degrees C) were determined. With respect to gas exchanges, significant changes in photosynthesis rate of Aveleira were observed between July and September (7mumol CO2m(-2)s(-1)). In contrast, Judia and Longal showed a strong increase in this period, 6.1-8.5 and 4.9-6.7 micromol CO2m(-2)s(-1), for Judia and Longal, which represent an increase of about 15% and 43%, respectively. Similar patterns were detected in daylight photosynthesis measurements for Judia and Longal, in which an almost 60% decrease was observed, in contrast to 40% for Aveleira, from morning to midday, when temperatures increased from 27 to 34 degrees C. In addition to high photosynthetic rates in the hottest month, Aveleira was also the sunniest cultivar according its highest value on chlorophyll a/b ratio (3.65). Cultivars also presented maximal thylakoid membrane potential at different temperatures, with their values being 20.8, 17.8 and 17.2 degrees C for Aveleira, Longal and Judia, respectively. These results were also supported by thylakoid fatty acid composition which indicated that the unsaturation index of Aveleira (158) was the lowest in comparison with other two cultivars, 168 and 175, for Longal and Judia, respectively.  相似文献
