Graft compatibility has been studied in vitro using callus tissues of apricot ( Prunus armeniaca) and different Prunus rootstocks to form scion/rootstock combinations with different degrees of graft compatibility. In these species, incompatibility is manifested by a breakdown of the trees at the union area that can occur some years after grafting. Here, the possibility of obtaining an early detection method to determine graft incompatibility is explored by callus fusion in vitro. The adhesion of the two callus partners, the development of the cells at the contact surface (cell arrangement, intensity of cell-wall staining), and the presence of lipid and phenolic compounds have been studied during the first 3 weeks after grafting in both compatible and incompatible combinations. Differences were observed at the second and the third week of callus co-culture in most of the characters determined, although these differences were present as early as the first week in the case of phenolic compounds. The behaviour of the grafts grown in vitro was correlated to that of the same combinations in the field, suggesting that callus fusion in vitro could be a possible and reliable method for an early detection of graft incompatibility in different Prunus combinations.  相似文献

The relationship between formation of CVB, which connects vascularcut ends of stock and scion, and the condition of tissue whereCVB is expected to be induced was studied using a cactus graftcombination of Notocactus submammulosus var. pampeanus youngseedlings on Hylocereus trigonus plants. When a scion with its basal part removed by a transverse cutwas put onto a transverse cut surface of a stock with a distanceof 1 mm between the vascular cut ends of both graft components,CVB was formed in only 13% of the grafts in the callus massmade by adhesion between calluses of stock and scion. The percentagearose to ca. 100% when 100 ppm NAA was applied to the scionapices. This promotive effect of NAA decreased with increasingtime from grafting to NAA application. When wound calluses of stock and scion were prevented from contactonly at the portion between the vascular cut ends of stock andscion by introducing a piece of aluminium foil into the graftunion, CVB was formed in 80% of the grafts in the callus ofthe stock without NAA application. In the case where the scionwas grafted at right angle onto an additional longitudinal cutsurface made 1 mm from the vascular bundle of the stock withremaining intact parenchyma between this cut surface and thevascular bundle of the stock, CVB was not formed even when NAAwas applied. These results suggested that wound response wasa prerequisite for CVB differentiation and that surface-to-surfaceadhesion of calluses inhibited CVB formation by making calluscells differentiate into quiescent parenchyma cells rapidly.The mechanism of this phenomenon is discussed. In order to determine the importance of direct cellular contactfor the formation of a compatible graft, I have monitored thedevelopment of compatible autografts in Sedum telephoides inwhich porous membrane filters were inserted between the graftingsurfaces. The tensile strengths of these grafts levelled offat approximately 10 g breaking weight per square millimetregraft area baween 2 and 5 d after grafting. Callus proliferationoccurd at both grafting surface by 5 d after grafting. By 21d after grafting, vascular redifferentiation across the graftinterface was complete. These results indicate that direct cellularcontact is not necessary for the (I) cohesion of the stock andscion, (2) callus proliferation or (3) vascular redifferentiationthat occur during the formation of a compatible graft. The shoot growth of compatible ( Prunus persica L. Batsch cv. Springtime grafted on Prunus cerasifera L. Ehrh cv. Myrobolan P2032) and incompatible ( Prunus persica L. Batsch cv. Springtime grafted on Prunus cerasifera L. Ehrh cv. Myrobolan P18) peach/plum grafts was observed over a period of 100 days after grafting under controlled conditions. Leaf and root activities were determined by studying carbon assimilation and partitioning, leaf mineral contents and water relations. Shoot length and leaf number were not significantly affected in the incompatible combination during the first 55 days after grafting, but then, shoot growth rate was significantly reduced. Final total dry weights of the shoot were similar in both graft combinations. The incompatible combination did not show any water stress. Soluble sugar and starch contents increased in the leaves of the incompatible combination, accounting for about 36% of the increase of leaf dry weight per unit area. Photosynthesis was affected by the compatibility of the grafts. Leaf nitrogen content (% dry weight) fell in the incompatible graft combination 65 days after grafting. However, nitrogen content on an area basis was not affected. The possibility of nitrogen stress is discussed.  相似文献

Initial vascular differentiation is generally considered tooccur in procambium. In ferns, however, a provascular tissueimmediately subjacent to the promeristem has been suggestedas an initial stage within which the procambium is subsequentlyformed. In contrast to this interpretation, a zonation conceptapplied in ferns recognizes a promeristem consisting of severallayers of cells in which no differentiation takes place. Thisstudy demonstrates that the shoot apex of Matteuccia struthiopterishas one cell layer of promeristem. Immediately subjacent tothe promeristem is the provascular tissue surrounding a centralgroup of pith mother cells. The developmental continuity betweenthe provascular tissue and the mature vascular tissue, and betweenthe pith mother cells and the pith, through transitional stages,indicates that the initial differentiation of vascular tissueand pith takes place in this prestelar tissue. The continuityof vascular differentiation in the area confronting young leavesor incipient leaf positions is interrupted by the formationof leaf gap initials. Developing leaves thus begin to exertinfluence on the vascular system at the prestelar stage. Homografting of Arabidopsis thaliana scions on stocks of A. thaliana and heterografting on other species were used to study the compatibility and the ontogeny of graft union formation. Highly compatible homografting with scions of young leafy inflorescence stems was obtained on stocks of inflorescence stems growing from large 3-month-old A. thaliana plants. Histologic analysis revealed four developmental stages of graft union formation in Arabidopsis homografting: (1) development of a necrotic layer, (2) callus proliferation in the grafted scion, (3) differentiation of new vascular tissues within the scion, and (4) a full vascular graft union formation between the scion and the stock. Vascular connections were formed within the callus bridge between rootstocks and scions 15 days after grafting. Heterografts of Arabidopsis on two members of Brassicaceae, cabbage (Brassica) and radish (Raphanus), showed partial incompatible interaction with a lower level of vascular differentiation. Arabidopsis grafting on tomato (Solanaceae) rootstock showed complete incompatibility and limited noncontinuous differentiation of new vascular tissues that did not cross the scion/stock boundary. Although lacking scion/stock vascular connections, Arabidopsis scions grafted onto tomato rootstock flowered and produced seeds. This may indicate some nonvascular functional connections between the two plants, probably of parenchyma cells, further emphasizing the usefulness of Arabidopsis as a model plant for studying various levels of the complicated scion/stock relationships expressed in grafting biology. Experiments with dye transport in the xylem showed that although in general there was an agreement between the histologic study and dye transport, in Arabidopsis homografts water transport frequency was lower than functional and histologic compatability. We conclude that homografting and heterografting of Arabidopsis inflorescence stems is a convenient and reproducible method for studying the fundamental cellular genetic and molecular aspects of grafting biology.  相似文献

The origin of callus from the exposed surface of the cambialzone in Trema orientalis Bl. and Julbernardia globiflora (Benth.)Troupin is described and discussed in relation to what has beenobserved in other species. It is suggested that in differentspecies callus may develop from any of the undifferentiatedcentripetal products of the vascular cambium, but that the kindof tissue contributing to callus initiation depends upon thespecies and on the histology of the cambial zone. The establishment of a new vascular cambium and phellogen isalso described. It is found that cambium formation is not dependenton the presence of a preexisting cambium in the surroundingtissues and that the first-formed xylem is abnormal in structure.  相似文献

To study the influence of morphogenetic gradients on vasculardifferentiation patterns, a new technique was developed whichallows different substances to be applied at opposite ends ofa tissue block. It yielded information on the mobility of particularmorphogens and on the dependence of callus formation and trachearyelement differentiation on their presence. Application of indol-3ylacetic acid (1AA) (10 mg l–1), zeatin (0.1 mg l–1)and sucrose (3 per cent, w/v) in various combinations to theends of cylindrical explants of lettuce pith (Lactuca sativaL.) showed that (a) callus formation was stimulated by IAA,whereas induction of tracheary elements required both IAA andzeatin; (b) callus was confined to a few millimetres at theends of the explants, and tracheary elements occurred mainlywithin the callus; (c) sucrose or its metabolic products diffusedthe 10 mm length of the explants, while IAA and zeatin wereeffective only close to the application site; and (d) some callusand tracheary elements formed when no sucrose was applied, butboth increased with sucrose application, though inhibition oftracheary elements formation occurred with high sucrose concentrations. differentiation, pith explant, tissue culture, xylogenesis, indol-3yl acetic acid, sucrose, zeatin, lettuce, Lactuca sativa  相似文献

Although an initial stage of vascular differentiation precedingprocambium has been demonstrated in ferns, its presence in seedplants has not been accepted generally. In the shoot apex ofcarrot, a short cylinder of provascular tissue is recognizedas the initial stage of vascular differentiation. This firstbecomes apparent through the enlargement and vacuolation ofpith and cortical tissue rather than as a result of specificchanges in the provascular tissue itself. Procambium in discretestrands differentiates acropetally in the provascular tissuein relation to developing leaf primordia. Provascular tissueis not recognized above the axil of the youngest leaf primordiumbut it is distinct at or above the level at which the traceof the youngest primordium diverges. Support for the recognitionof provascular tissue is provided by a positive reaction tohistochemical tests for carboxylesterases in this tissue aswell as in procambium and later stages of vascular differentiation.Copyright1999 Annals of Botany Company. Summary The occurrence of plasmodesmata in the graft interfaces of two heteroplastic grafts (Impatiens walleriana onImpatiens olivieri andHelianthus annum onVicia faba) has been studied. For both systems two types of intercellular strand are described: 1. Continuous plasmodesmata interconnecting the cells of stock and scion and 2. half plasmodesmata traversing the wall part of one partner cell without connection to the abutting cell. Single strands or branched forms occur in both types of plasmodesma. In the case of half plasmodesmata, branchings with extended median nodules predominate. The distribution of half and continuous plasmodesmata varies with the different areas of a graft interface: in the region of bridging vascular tissues most cell connections are continuous. In areas where cortex or pith-derived callus cells and those of misaligned tissues (cortex/vascular tissue; cortex/pith; pith/vascular tissue) match, discontinuous strands predominate.Branched half plasmodesmata also occur in presumably fused walls between related callus cells; they are typical structures secondarily formed in non-division walls.The results are discussed with regard to compatibility/incompatibility phenomena in heterografts and the development and function of interspecific cell bridges.  相似文献

The effect of application of heat on the development of graftunions in dormant Picea sitchensis (Bong.) Carr (Sitka spruce)was observed at monthly intervals from October to March. Noadvantage over controls was gained in terms of final graft successrates. It is suggested that this may have been a result of themild winter conditions during the course of the experiment,which enabled callus formation to occur in unheated controls.On the other hand, scions of grafts which had been heated for3 weeks, and then returned to ambient conditions showed considerablygreater growth than unheated controls during the following springand summer. The amount of extension depended on how late inthe winter the graft had been prepared and heated, with leastgrowth by grafts made in October, and most by grafts made inMarch. Microscopical examination showed that callus formationwas more rapid in heated grafts than in controls, although callusformation occurred in all control grafts examined. Heat alsoinduced cambial reactivation and tracheid formation, particularlyin the vicinity of needle or bud traces in the scion. In keepingwith this, it was found that the presence of needles and budswas essential for cambial reactivation in normal dormant stemsand that reactivation was restricted to the heated region. Graft union development in plants has been studied mainly by destructive methods such as histological studies. The aim of this work was to evaluate whether the chlorophyll fluorescence imaging (CFI) technique is sensitive enough to reflect changes at the cellular level in different Solanaceae grafted plants 30 d after grafting, when both grafted partners were well fused and strong enough in all plant combinations. The pepper cultivar ‘Adige’ was grafted onto different Capsicum spp. accessions typified with different compatibility degrees; eggplant was grafted on Solanum torvum and pepper homografts as compatible unions; pepper was grafted on S. torvum and on tomato as incompatible unions. ‘Adige’/’Adige’ and ‘Adige’/pepper A25 showed a higher maximum quantum efficiency of PSII associated with higher values of actual quantum efficiency of PSII and photochemical quenching as well as with vascular regeneration across the graft interface. Our results highlighted that CFI changes reflected histological observations in grafted Solanaceae plants.  相似文献

Modification of the Pollen-Stigma Interaction in Brassica oleracea by Water   总被引:2,自引:0,他引:2  
The presence of a film of distilled water on the stigma surfaceof freshly opened flowers results in complete inhibition ofpollen following both incompatible and compatible pollinationsin self-incompatible (SI) genotypes of Brassica oleracea, SIgenotypes of B. campestris and one self-compatible (SC) genotypeof B. campestris. The application of water to the stigmas afterpollination also resulted in a marked reduction in pollen germinationand tube penetration. An increase in the time intervals betweenthe application of pollen onto the stigma and the water treatmentprogressively reduced this inhibition. Pollen germination wasalso completely inhibited when stigmas from freshly-opened flowersof SI B. campestris and B. oleracea genotypes were washed inwater, dried and pollinated with pollen grains of either compatibility.The ability of stigmas to induce pollen germination and tubegrowth was restored over a period, the length of which was dependenton the incompatibility (S) genotype. Stigmas of B. napus (SC)and SC mutants of SI B. campestris were found to be affectedby washing, but stigmas of a SC variety of B. campestris andthe immature stigmas from buds of B. oleracea were found tobe considerably less affected. Microscopic examination of pollenplaced on washed stigmas reveals that grains, irrespective oftheir compatibility, fail to hydrate normally. When inducedto hydrate by raising atmospheric humidity, pollen grains onwashed stigmas did germinate, but most of the tubes failed topenetrate the papillar wall and very few entered the style.It is proposed that the water treatment mobilises componentsof the pellicle which reorganize to block the activity of molecules,present in both SC and SI individuals, responsible for establishingfull contact between the pellicle and pollen grain coating. Grafting has been adopted for a wide range of crops to enhance productivity and resilience; for example, grafting of Solanaceous crops couples disease-resistant rootstocks with scions that produce high-quality fruit. However, incompatibility severely limits the application of grafting and graft incompatibility remains poorly understood. In grafts, immediate incompatibility results in rapid death, but delayed incompatibility can take months or even years to manifest, creating a significant economic burden for perennial crop production. To gain insight into the genetic mechanisms underlying this phenomenon, we developed a model system using heterografting of tomato (Solanum lycopersicum) and pepper (Capsicum annuum). These grafted plants express signs of anatomical junction failure within the first week of grafting. By generating a detailed timeline for junction formation, we were able to pinpoint the cellular basis for this delayed incompatibility. Furthermore, we inferred gene regulatory networks for compatible self-grafts and incompatible heterografts based on these key anatomical events, which predict core regulators for grafting. Finally, we examined the role of vascular development in graft formation and uncovered SlWOX4 as a potential regulator of graft compatibility. Following this predicted regulator up with functional analysis, we show that Slwox4 homografts fail to form xylem bridges across the junction, demonstrating that indeed, SlWOX4 is essential for vascular reconnection during grafting, and may function as an early indicator of graft failure.

Domestication is an excellent model to study diversification and this evolutionary process can be different in perennial plants, such as fruit trees, compared to annual crops. Here, we inferred the history of wild apricot species divergence and of apricot domestication history across Eurasia, with a special focus on Central and Eastern Asia, based on microsatellite markers and approximate Bayesian computation. We significantly extended our previous sampling of apricots in Europe and Central Asia towards Eastern Asia, resulting in a total sample of 271 cultivated samples and 306 wild apricots across Eurasia, mainly Prunus armeniaca and Prunus sibirica, with some Prunus mume and Prunus mandshurica. We recovered wild Chinese species as genetically differentiated clusters, with P. sibirica being divided into two clusters, one possibly resulting from hybridization with P. armeniaca. Central Asia also appeared as a diversification centre of wild apricots. We further revealed at least three domestication events, without bottlenecks, that gave rise to European, Southern Central Asian and Chinese cultivated apricots, with ancient gene flow among them. The domestication event in China possibly resulted from ancient hybridization between wild populations from Central and Eastern Asia. We also detected extensive footprints of recent admixture in all groups of cultivated apricots. Our results thus show that apricot is an excellent model for studying speciation and domestication in long‐lived perennial fruit trees.  相似文献

Inoculation of the potato cut surfaces with an incompatiblerace of Phytophthora infestans induced an accumulation of rishitin,but only a trace occurred when infected by a compatible race.When the tuber was cut, a large amount of steroid-glycoalkaloids(solanine) accumulated in the cut surface tissue, although onlya trace was found in intact tissue. Only a small amount of solanine,if any, was contained in the wound periderm tissue. Most ofthe solanine seemed to be distributed in tissue neighbouringthe newly formed meristematic tissue zone. Distribution of solanineas a function of distance from the cut surface was exponential.Infection of the cut surface by an incompatible race of Phytophthorainfestans reduced the accumulation of solanine. The higher theconcentration of zoospore used, the less the solanine content.It has been reported that the higher the concentration of zoosporesused for inoculation of the cut surface, the less the numberof renewed meristematic cells in the wound tissue. In experimentsusing fresh and aged tubers, a good correlation between thenumber of renewed meristematic cells and solanine content wasfound. The accumulation of solanine in the wound tissue andits reduction due to infection by an incompatible race may berelated to renewed meristematic cells formation and its reductioncaused by the infection. Pith callus tissues were grown under continuous blue (450 mµ),green (545 mµ), red (650 mµ), and ‘white’(full-spectrum) light, and in the dark for 22 days at 27±2°C at energy levels of 15,000 ergs cm–2 sec–1. Mean increases in fresh weight of tissues grown under ‘white’and blue light were significantly greater than those of tissuesgrown in green and red light and in the dark. Tissues grownin the dark yielded mean fresh weight increases significantlylower than tissues grown under blue, red, and ‘white’light. No significant differences were shown between blue and‘white’, red and green, and green and dark treatmentsrespectively. Cell differentiation occurred in all treatmentsonly to the extent of vessel element formation. There were nodifferences in degree of differentiation between treatments. It was proposed that the high-energy reaction of photomorphogenesiswas in operation in the Pelargonium callus tissue. The resultsindicated the presence in the tissue of high-energy photoreceptor(s).The use of high-intensity, incandescent illumination for experimentalprocedures approximating natural conditions of irradiation wasindicated as desirable for pith callus tissues of Pelargoniumzonale var. Surgical techniques were applied to the shoot apex of carrot(Daucus carota L.) to test the interpretation that provasculartissue is the initial stage of vascular differentiation andto localize the sources of the influences that control its differentiation.If the apex is isolated laterally by vertical incisions leavingit at the summit of a plug of pith tissue, vascular differentiationproceeds normally and an independent vascular system is formedin the pith plug. If all leaf primordia are systematically suppressed,provascular tissue continues to differentiate as an acropetalextension of the pre-existing vascular system but no furtherdifferentiation occurs. When the apex is isolated laterallyand all leaf primordia are suppressed, provascular tissue continuesto be formed acropetally and is extended basipetally into thepith plug by redifferentiation of pith cells, but no furtherdifferentiation occurs. This tissue reacts positively to histochemicaltests for esterase indicating its vascular nature. If only oneleaf primordium is allowed to develop on an isolated shoot apex,its vascular system develops normally and extends basipetallyinto the pith plug, but there is no extension of provasculartissue into the pith plug.
