Phosphohexose isomerase polymorphism in horse erythrocytes   总被引:2,自引:0,他引:2  
Four individual patterns of phosphohexose isomerase (PHI) from horse red cells were found by starch gel electrophoresis. Population and family data of two Swedish horse breeds were consistent with the theory that the PHI types were controlled by three codominant, autosomal alleles designated PHIF, PHII and PHIS( F , I and S ).  相似文献

Abstract Three calmodulin antagonists, trifluoperazin, compound 48/ 80 and calmidazolium provoke K+ efflux from metabolizing yeast cells. This K+ efflux is accompanied by a shrinkage of the cells. Part of the cells show a gradual shrinkage whilst the remainder of the cells shrink via an all-or-none process. Cells shrunken via an all-or-none process have the same size as cells which have been completely permeabilized by a cationic detergent with loss of all their K+.  相似文献

A polymorphism of S-globulin was detected in serum of Silurus glanis L. by starch gel electrophoresis. Three phenotypes were observed which are apparently controlled by two codominant alleles, SgA and SgB , of an autosomal locus, Sg . Although on electrophoresis S-globulin and transferrin have similar mobilities, the properties of the two proteins differ.  相似文献

Cultures of 19 saprophytic basidiomycete species were examined for aluminium resistance using a micro-well technique. The following aluminium concentrations have been employed: 0, 1, e1, e2, e3, and e4 mM given as sulphate. Three of the saprophytic species were also tested at the pH values generated by the same A1 concentrations. The following species were considered resistant: Mycena galopus, M. metata, M. inclinata, M. viridimarginata, Marasmius androsaceus, Galerina calyptrata , and Hypholoma capnoides . The following were sensitive: Micromphale perforans, Mycena cinerella, Clitocybe fragrans , and Clitocybe metachroa . The remaining species showed intermediate responses. The autecology of species exhibiting either a high resistance or a high sensitivity is discussed. Mycena galopus , found to be particularly resistant, may be specially tolerant to acidification since it is reported to increase or persist in polluted environments by several authors.  相似文献

So far, at least eight alleles in the goat CSN2 locus have been associated with the level of β -casein expression in milk. Alleles CSN2 A , CSN2 A 1, CSN2 B , CSN2 C , CSN2 D and CSN2 E have been associated with normal content (allele effects of about 5 g of β -casein per litre), whereas the CSN2 0 and CSN2 01 alleles have been associated with non-detectable levels of β -casein. Most of these alleles have been characterized genetically. Herein, we report the identification of a previously unreported SNP in the goat CSN2 promoter region ( AJ011018 :g.1311T>C), which is associated with the absence of β -casein in the milk. Furthermore, we developed a PCR-based method that allows detection of this mutation.  相似文献

Studies were carried out on the polymorphism of alkaline ribonuclease in Black-and-White cattle in the north-eastern region of Poland. Leucocytes isolated from peripheral blood were used. Three phenotypes were defined, consisting of the combination of two alleles. The gene frequency was: q4= 0,7082, qB= 0,2918. In calves the frequency of allele RNA-aseB and number of RNA-aseA/ RNA -aseB heterozygotes were higher than in adult animals.  相似文献

Three strains of aerobic, Gram-negative, rod-shaped, non-spore-forming, yellow-pigmented bacteria (OD1T, YOxT and NS13), which were isolated in previous studies by enrichment in a mineral medium with potassium oxalate as the sole carbon source, were characterized. On the basis of 16S rRNA gene sequence similarity, strains OD1T, YOxT and NS13 belong to the Betaproteobacteria , most closely related to Oxalicibacterium flavum TA17T (97.2–99.7% sequence similarity). The major whole-cell fatty acids were C16:0, C16:1ω7c and C17:0 cyclo. The results of DNA–DNA hybridization and physiological and biochemical tests allowed genotypic and phenotypic differentiation of strains OD1T and YOxT from O. flavum TA17T and from each other. Therefore, it is concluded that the strains OD1T and YOxT represent novel species within the genus Oxalicibacterium , for which the names Oxalicibacterium horti sp. nov. (type strain OD1T=DSM 21640T=NBRC 13594T) and Oxalicibacterium faecigallinarum sp. nov. (type strain YOxT=DSM 21641T=CCM 2767T) are proposed.  相似文献

In the January 1999 issue of New Phytologist , we published the research paper entitled 'The effect of light on the growth and reproduction of Floerkea proserpinacoides ' by Margaret F. McKenna and Gilles Houle ( New Phytol . (1999) 141 , 99–108). Since its publication, the authors have identified two important errors in the text. First, values given for leaf area ratio (LAR) and specific leaf area (SLA) have been given erroneously in units of cm2 g−1; the correct units are cm2 mg−1. Second, the values for unit leaf rate (ULR) have been given incorrectly as tenfold lower than they should be; these should be multiplied by ten to give the correct values in mg m−2 d−1.

Three NADH diaphorase phenotypes were observed in the red cells of sheep. Breeding data indicated that this polymorphism was under the control of two autosomal co-dominant alleles, designated DiaF and Dias. Phenotype Dia F had significantly lower NADH diaphorase activity than phenotype Dia S. The frzquency of DiaF and DiaS was determined in 9 different breeds.  相似文献

Abstract: Three serine residues (Ser193, Ser194, Ser197) in the fifth transmembrane-spanning region of the D2 dopamine receptor have been mutated separately to alanine and the effects of the mutations determined in ligand-binding experiments with [3H]spiperone. For many antagonists the mutations had little effect, showing that the overall conformation of the mutant receptors was similar to that of the native, although there were effects on the binding of certain antagonists. The effect of the mutations on agonist binding to the free receptor (uncoupled from G proteins) was determined in the presence of GTP (100 µ M ). This showed that there was no single mode of binding of catecholamine agonists to the receptor and that all three serine residues can participate in the binding of some agonists, possibly through hydrogen bonds to the catechol hydroxyl groups. Coupling of the mutant receptors to G proteins was assessed from agonist-binding curves in the absence of GTP, when higher and lower affinity agonist-binding sites were seen. Receptor/G protein coupling was generally unaffected by the Ala193 and Ala194 mutations, but the Ala197 mutation eliminated receptor/G protein coupling for some agonists. These data show that the interactions of agonists with the free and coupled forms of the receptor are different.  相似文献

Abstract β-xylosidase (EC 3.2.1.37) has been purified from Aspergillus nidulans mycelium grown on oat-spelt xylan as sole carbon source. Its pH optimum for activity was found to be 5.0 and the optimum temperature was 50 °C. Its molecular mass was estimated by gel filtration to be 180000. Using p-nitrophenyl-β-d-xylopyranoside as substrate, the K m and V max values have been found to be 1.1 mM and 25.6 μmol min−1(mg protein)−1, respectively. Enzyme activity was inhibited by Hg2+, Ag2+, and Cu2+ at a concentration of 1 × 10−3 M. The synthesis of β-xylosidase in A. nidulans is strongly induced by arabinose and xylose and is subject to carbon catabolite repression mediated by the cre A gene product.  相似文献

Two new erythrocyte antigens have been detected by isoimmune antibodies obtained from Gottingen Miniature pigs. By family studies they proved to belong to the E system. The factor designated En is genetically controlled by alleles E 2= E edghkmn, E 3= E aegln, E 4= E edfhkmn, E 6= E aefln, E 9= E edghjmn, E 12= E aegmno and E edgkln, and the factor designated Eo by the allele E 12= E aegmno.
The En blood factor occurs in all the breeds studied and in frequency it comes close to a related Ee factor. It stands in opposition not only to Eb but also to Ei factor. The Eo factor can be characterized as a rarely occurring Ea subgroup. Besides being antithetic to Ed it is also antithetic to El and Ei factors. For the present, it was found only in Miniature pigs.  相似文献

The generally rhizotoxic ion Al3+ often enhances root growth at low concentrations. The hypothesis that Al3+ enhances growth by relieving H+ toxicity was tested with wheat seedlings ( Triticum aestivum L.). Growth enhancement by Al3+ only occurred under acidic conditions that reduced root elongation. Al3+ increased cell membrane electrical polarity and stimulated H+ extrusion. Previous investigations have shown that Al3+ decreases solute leakage at low pH and that the alleviation of H+ toxicity by cations appears to be a general phenomenon with effectiveness dependent upon charge (C3+>C2+>Cl+). Alleviation of one cation toxicity by another toxic cation appears to be reciprocal so that Al3+ toxicity is relieved by H+. It has been argued previously that this latter phenomenon accounts for the apparent toxicity of ALOH2+ and Al(OH)+2. Reduction of cell-surface electrical potential by the ameliorative cation may reduce the cell-surface activity of the toxic cation.  相似文献

SYNOPSIS. Cell lines of Acanthamoeba castellanii resistant to erythromycin (EryR), chloramphenicol (CapR), and oligomycin (OliR) have been isolated. These may be the first such mutants for A. castellanii. These mutants have been phenotypically stable for 2 years, surviving storage and vegetative multiplication in the absence of drugs. Resistance was specific for each drug, but double mutants (e.g. EryRCapR) were obtained by stepwise selection. Mutant frequencies were determined in multiwell plates; <10 colony forming units (CFU/105 amebas were observed in wild-type populations 12 days after incubation in 500 μg Ery/ml, 2.5 mg Cap/ml, or 15 μg Oli/ml. After 30 days, averages of 100 CFU/105 amebas were observed in Ery and Cap, whereas, frequencies for Oli remained unchanged. Frequencies for EryR and CapR were consistent with rates of recovery from these drugs in batch cultures. We were unable to obtain spontaneous mutants resistant to cycloheximide, emetine, 5-fluorodeoxyuridine, or ethidium bromide. EryR, CapR and OliR could be mitochondrial mutants.  相似文献

We have identified a novel 399 bp repetitive DNA element (which we designate beta  ) 9 bp upstream of a seryl-tRNACAG gene in the genome of Candida albicans . There are two copies of the seryl-tRNACAG gene, one on each homologue of chromosome VI, and the beta element is found upstream of one copy of the gene in C. albicans strain 2005E. The beta element is not present upstream of either copy of the seryl-tRNACAG gene in eight other laboratory strains of C. albicans tested, but was detected in this location in several fresh clinical isolates. Southern blot analysis indicated that there are approximately eight copies of the beta element per diploid C. albicans genome and that it is a mobile element, being present on at least two different chromosomes. Three unique genomic DNA clones containing the beta element were isolated from strain 2005E; in each case, a different tRNA gene was found immediately adjacent to the beta element. Three new tRNA genes from C. albicans have thus been identified: tRNAAsp, tRNAAla and tRNAIle. The beta element shows no significant sequence homology to other known prokaryotic or eukaryotic repetitive elements, although an 8 bp repeat at the 3' end of the element is identical to that of the Ty3 retrotransposable element of Saccharomyces cerevisiae . We propose that the beta element is a solo long terminal repeat (LTR) sequence of a Ty3/gypsy-like transposable element in C. albicans that is closely associated with tRNA genes.  相似文献

Electrophoretic variants of serum esterases and β-globulins in two subspecies of paradise fish ( Macropodus opercularis ) were studied. Four esterase loci ( Est-1, Est-2, Est-3 and Est-4 ), a single transferin ( Tf ) and another major β-globulin locus ( Bg ) were identified by segregational analysis. Est-3 seems to be a monomorphic. locus. Three alleles of Est-1 , two of Est-2 , two of Est-4 , four of Tf and two alleles of Bg were found in the laboratory population. None of these loci were closely linked. Electrophoretic patterns of F1 hybrids confirmed the monomeric structures of each of the studied proteins. Allelic segregation at the Tf and Bg loci was normal in F2 and backcross populations. In crosses of the two Macropodus subspecies there were deviations from Mendelian ratios because of missing recombinant esterase phenotypes. Each of these would have been homozygous Est-2f/f . We suppose that Est-2f/f causes lethality in the early phase of development, except in the Est-1c/c, Est-2f/f combination characteristic of the parental subspecies M.o. concolor .  相似文献

Mechanism of copper-enhanced photoinhibition in thylakoid membranes   总被引:2,自引:0,他引:2  
The effect of copper on photoinhibition of photosystem II (PSII) in vitro was studied in bean ( Phaseolus vulgaris L. cv. Dufrix) and pumpkin ( Cucurbita pepo L.) thylakoids. The thylakoids were illuminated at 200–2 000 μmol photons m−2 s−1 in the presence of 70–1 830 added Cu2+ ions per PSII. Three lines of evidence show that the irreversible damage of PSII caused by illumination of thylakoids in the presence of Cu2+ was mainly due to donor-side photoinhibition resulting from inhibition of the PSII donor side by Cu2+. First, addition of an artificial electron donor partially restored PSII activity of thylakoids that had been illuminated in the presence of Cu2+. Second, already moderate light was enough to cause rapid inhibition of PSII, and the inhibition could be saturated by light. Third, the extrinsic polypeptides of the oxygen-evolving complex were found to become oxidized by the combined effect of Cu2+ and light. The presence of oxygen was not necessary for the copper-induced enhancement of photoinhibition of PSII. When the illumination was prolonged, copper caused a gradual collapse of the thylakoid structure by increasing degradation of thylakoid proteins.  相似文献

The concentrations of the main plasma inorganic electrolytes Na+, K+, Ca2+, Mg2+, Cl- and and PO43- have been determined for different orders of marine fishes. For Na+ and Cl- a typical decrease was found when passing from cyclostomes, holocephalans and elasmobranchs to teleosts. The concentrations of K+, Ca2+ and Mg2+ showed a similar trend except that there was a rise in the teleost group, which showed a large range of variation for these three ions. In the case of PO43- no significant differences between groups were found.  相似文献

Genetic polymorphism of the pig plasma vitamin D binding protein Gc was demonstrated by agarose isoelectrofocusing followed by either autoradiography or immunofixation with specific human Gc antiserum. Three different types F, FS and S were observed. Family data supported the genetic theory that the Gc types are controlled by two autosomal codominant alleles GcF and Gcs . Both alleles are present in Yorkshire and Duroc. In Danish Landrace and Hampshire only the GcF allele was observed.  相似文献

Three new rapid methods for detection of Salmonella spp. have been studied. The fluorogenic MUCAP test (Biolife, Italy), the MicroScreenR latex slide agglutination test (Mercia, UK) and the Rambach agar test (Technogram, France) were compared for their sensitivity and their specificity. Some 175 strains, incuding 74 Salmonella strains and 101 non- Salmonella strains were included in the study. The sensitivities of the MUCAP, the MicroScreenR and Rambach agar tests were 100%, 96% and 91%, respectively, and their specificities 80%, 96% and 100%, respectively.  相似文献
