The possible involvement of IAA in the effect thatAzospirillum brasilense has on the elongation and morphology ofPanicum miliaceum roots was examined by comparing in a Petri dish system the effects of inoculation with a wild strain (Cd) with those of an IAA-overproducing mutant (FT-326). Both bacterial strains produced IAA in culture in the absence of tryptophan. At the stationary growth phase, production of IAA by FT-326 wasca. 12 times greater than that of Cd. When inoculation was made with bacterial concentrations higher than, 106 colony forming units ml–1 (CFU ml–1), both strains inhibited root elongation to the same extent. At lower concentrations Cd enhanced elongation, by 15–20%, while FT-326 was ineffective. Both strains promoted root-hair development, and root-hairs were produced nearer the root tip the higher the bacterial concentration (e. g. root elongation region was reduced). Effects of FT-326 on root-hair development were greater than those of Cd. Acidified ether extracts of Cd and FT-326 cultures had inhibitory or promoting effects on root elongation depending on the dilution applied. At low dilutions, extracts from FT-326 were more inhibitory for elongation than those from Cd. At higher dilutions root elongation was promoted, but FT-326 extracts had to be more diluted than those from Cd. Dilutions that promoted root elongation contained supra-optimal concentrations of IAA, 1–3 orders of magnitude higher than those required for optimal enhancement by synthetic IAA. It is suggested that the bacteria produce in culture an IAA-antagonist or growth inhibitor that decreases the effectiveness of IAA action. The large variability reported for the effects ofAzospirillum on root elongation could be the result of the opposite effects on root elongation of IAA and other compounds, produced by the bacteria.  相似文献

The influence of endogenous root nodules phenolic acids on indoleacetic acid (IAA) production by its symbiont (Rhizobium) was examined. The root nodules contain higher amount of IAA and phenolic acids than non-nodulated roots. Presence of IAA metabolizing enzymes, IAA oxidase, peroxidase, and polyphenol oxidase indicate the metabolism of IAA in the nodules and roots. Three most abundant endogenous root nodule phenolic acids (protocatechuic acid, 4-hydroxybenzaldehyde and p-coumaric acid) have been identified and their effects on IAA production by the symbiont have been studied in l-tryptophan supplemented yeast extract basal medium. Protocatechuic acid (1.5 μg ml−1) showed maximum stimulation (2.15-fold over control) of IAA production in rhizobial culture. These results indicate that the phenolic acids present in the nodule might serve as a stimulator for IAA production by the symbiont (Rhizobium). Mycorrhization helper bacteria (MHB), isolated from phylogenetically distinct ectomycorrhizal symbioses involving Lactarius rufus, Laccaria bicolor or Suillus luteus, were tested for fungus specificity to enhance L. rufus–Pinus sylvestris or L. bicolor–P. sylvestris mycorrhiza formation. As MHB isolated from the L. rufus and S. luteus mycorrhiza were originally characterised using a microcosm system, we assessed their ability to enhance mycorrhiza formation in a glasshouse system in order to determine the extent to which MHB are system-specific. Paenibacillus sp. EJP73, an MHB for L. rufus in the microcosm, significantly enhanced L. bicolor mycorrhiza formation in the glasshouse, demonstrating that the MHB effect of this bacterium is neither fungus-specific nor limited to the original experimental system. Although the five MHB strains studied were unable to significantly enhance L. rufus mycorrhiza formation, two of them did have a significant effect on dichotomous short root branching by L. rufus. The effect was specific to Paenibacillus sp. EJP73 and Burkholderia sp. EJP67, the two strains isolated from L. rufus mycorrhiza, and was not associated with auxin production. Altered mycorrhiza architecture rather than absolute number of mycorrhizal roots may be an important previously overlooked parameter for defining MHB effects.  相似文献

Hairy roots of Cichorium intybus obtained by infecting with different Agrobacterium rhizogenes strains (LMG-150 and A20/83) were studied for total endogenous indole-3-acetic acid (IAA) levels and indole-3-acetic acid oxidase (IAAO) activity. The roots initiated by LMG- 150 showed higher endogenous IAA levels as well as IAAO activity as compared to the roots from A20/83. Coumarin production in roots obtained by both of these strains strictly correlated with growth, with higher content in the roots obtained by LMG- 150. Moreover roots from LMG-150 showed increased growth index, length of primary roots and number of secondary and tertiary roots. The roots derived from LMG-150 were studied for total endogenous IAA and IAAO activity under the exogenous administration of polyamines and fungal elicitors. The treatment with putrescine (Put) at 1.5 mM level showed maximum endogenous IAA levels and IAAO activity as compared to the control and other polyamine administration, it also supported faster growth in terms of biomass accumulation, and total coumarin production. Of the various treatments, mycelial extract (ME) and culture media filtrate (CMF) of Pythium aphanidermatum and Phytopthora parasitica var. nicotiana, the treatment with 1 % CMF of P. parasitica var. nicotiana, resulted in maximum IAA levels and IAAO activity, which was supported by maximum biomass, coumarin production as compared to the control and other elicitor treatments. Two different regenerants of chicory obtained through A. rhizogenes LMG-150 designated as T-I and T-II, were studied for total endogenous IAA levels and IAAO activity. T-II showed higher titers of IAA with higher activity of IAAO as compared to T-I. Endogenous titer of IAA and IAAO activity was found to be maximum in transformed roots as compared to T-I, T-II, normal roots and normal plants. Our work showed a variation in endogenous auxin levels in these transformed plants. There exists a synergistic effect of endogenous IAA titers and polyamines in regulating root morphogenesis. Fungal elicitors influenced growth and coumarin production and an elicitor preparation of 1 % CMF of P. parasitica var. nicotiana gave spontaneous regeneration of shoots. The implications of these results are discussed.  相似文献

We have studied the biomass and alkaloid production of geneticallytransformed hairy root cultures of Lobelia inflata L. Thehairy root clone 8009/h7 transformed with Agrobacteriumrhizogenes strain R 1601 was cultivated on B5 solid media containingdifferent amounts of the growth regulators KIN, IAA or NAA. KIN significantlydecreased growth and lobeline production and strongly inhibited biomassformation at 5 mg/l. IAA and NAA had characteristic morphologicaleffects on growth, in increasing the number of the lateral roots. However theyrestricted linear growth. Addition of IAA or NAA into the culture mediumincreased the biomass formation and lobeline production of hairy roots. It wasfound that the greatest amount of lobeline was obtained at the 0.2mg/l IAA concentration, similar to the effect of NAA.  相似文献

In order to study the influence of Arbuscular Mycorrhiza (AM) on the development of root rot infection, tomato plants were raised with or withoutGlomus mosseae and/orPhytophthora nicotianae var.parasitica in a sand culture system. All plants were fed with a nutrient solution containing one of two phosphorus (P) levels, 32µM (I P) or 96µM (II P), to test the consequence of enhanced P nutrition by the AM fungus on disease dynamics. Mycorrhizal plants had a similar development to that of control plants. Treatment withPhytophthora nicotianae var.parasitica resulted in a visible reduction in plant weight and in a widespread root necrosis in plants without mycorrhiza. The presence of the AM fungus decreased both weight reduction and root necrosis. The percentage reduction of adventitious root necrosis and of necrotic root apices ranged between 63 and 89% The enhancement of P nutrition increased plant development, but did not appreciably decrease disease spread. In our system, mycorrhiza increased plant resistance toP. nicotianae var.parasitica infection. Although a contribution of P nutrition by mycorrhiza cannot be excluded, other mechanisms appear to play a crucial role.  相似文献

Galactoglucomannan oligosaccharides (GGMOs) activity in K. humboldtiana root culture has been determined. GGMOs inhibited adventitious root growth and lateral root induction in contrast to IAA, IBA, and NAA stimulating effect in these processes. Similarly, the combination of GGMOs with natural auxins (IAA, IBA) evoked an inhibition of adventitious root growth and lateral root induction that depended on the oligosaccharides concentration and the type of auxin. The growth stimulating effect of the synthetic auxin, NAA, in adventitious roots was negatively affected by GGMOs, but they were without influence on lateral root induction. The presence of oligosaccharides triggered lateral root position on adventitious roots and the anatomy of adventitious roots (diameter, proportion of primary cortex to the central cylinder, number and size of primary cortical cells, intercellular spaces, and the number of starch grains in cells of primary cortex) in dependence on their coactions with auxin.  相似文献

Fungi were isolated from young, serial-washed roots of Calluna sampled from a Danish heathland, Hjelm Hede. Of the 626 isolates, those that were dark, sterile and septate were divided into 13 morphological groups based on their appearance in culture on malt agar. Mycorrhizal synthesis in vitro showed that several groups formed typical ericoid mycorrhiza with seedlings of Calluna; these ericoid mycorrhizal fungi were morphologically similar to Hymenoscyphus ericae. The identities of the other dark, septate fungi are uncertain. Oidiodendron spp. were isolated in a very low frequency; these fungi also formed typical ericoid mycorrhiza. The Calluna root system on Hjelm Hede demonstrated a high morphological diversity among the associated dark, septate fungi suggesting that more than one fungus could coexist in the same host root system.  相似文献

Corn smut caused by Ustilago maydis (DC) Corda on maize (Zea mays L.) is characterized by gall (tumour) formation on aerial parts of the plant. Young galls on immature corn ears are called huitlacoche and are highly appreciated as a food delicacy. Several reports have suggested that production of the auxin indole-3-acetic acid (IAA) by U. maydis might be involved in tumour formation. Because strains showing defects in IAA biosynthesis (Iaa phenotype) would be valuable in elucidating the role of IAA in pathogenicity, in a previous study we isolated and analysed several such mutants. In the present work, we have crossed a null Iaa auxotrophic mutant with compatible wild-type strains. The main objective of the present work was to evaluate the pathogenicity of crosses involving wild-type and Iaa strains, in order to examine its relation to IAA production. Significant differences were found in growth, IAA production and ability to survive in water suspension among wild-type and mutant progeny strains. In general, high levels of pathogenicity were associated to high levels of IAA production by the strains, which supports the hypothesis that U. maydis strains require the ability to produce IAA in order to induce tumours in the host.  相似文献

Indole-3-acetic acid (IAA) has been unequivocally identified in culture supernatants of Rhizobium strains by gas chromatography-mass spectrometry. A method for accurately quantitating IAA in bacterial culture supernatants, employing deuterium-labeled IAA as an internal standard, has been developed. Similar IAA concentrations were found in culture supernatants of chosen Rhizobium mutants (defective in nodule formation) and their corresponding parent strains. Since some of the mutants are known to adhere to root hairs, it can be concluded that root hair curling is not simply a consequence of IAA production by rhizobia.  相似文献

Effects of auxins (IAA, IBA and NAA) on K. humboldtiana root culture cultivated in 16-h photoperiod or in dark have been observed. Light affected positively the production of biomass when cultivated on medium supplemented with NAA in 10 and 25 mol –1 concentrations. In the presence of IAA and IBA these values were significantly lower. The growth dynamics of root cultures depended on the auxin used. The best adventitious roots elongation and lateral roots induction on media supplemented with IBA has been ascertained. Morphological and anatomical differences in dependence on auxin used were observed. NAA supported the formation of huge callus-like mass besides mostly very short roots, especially under the light. Similarly IAA induced short roots, and IBA seems to be the most effective substance for the root elongation in this model system. NAA induced roots with larger diameter under the light compared with the other two auxins used. The reason is in the different anatomical structure of roots which was characterized by higher number of cell layers and large intercellulars in the cortex. The shape of cortical cells in the presence of IBA depended on the light conditions. Isodiametric cortical cells were present in roots cultivated in 16-h photoperiod, irregularly-shaped cells in the dark. The effect of light conditions was the smallest in the case of roots grown on IAA enriched media.  相似文献

Summary The influence of a morphactin, chlorflurenol-methylester (CFM), on the growth, the morphogenesis and the isoelectric peroxidase pattern was investigated in both callus cultures (two different tissue culture strains) and multiple bud cultures ofNicotiana tabacum var.Wisconsin. CFM (range of concentration between 10–6g/ml and 10–4g/ml) was applied singly, or in combination with a cytokinin, benzylaminopurine (BAP), or with an auxin, indoleacetic acid (IAA), or with IAA plus BAP.In general, the callus growth was inhibited under the influence of CFM. In some of the experiments carried out in hormone-free media, growth stimulation was observed. Even minimal inhibition or stimulation of the callus growth was always accompanied by characteristic changes in the peroxidase patterns.The following results show the influence of the morphactin CFM on cytokinin effects (endogenous cytokinin or equally the exogenously applied cytokinin, BAP). (1) In the multiple bud cultures, BAP and CFM (both substances combined with IAA) similarly caused inhibition of root formation and stimulation of bud formation. The bands in the peroxidase patterns, characteristic of cytokinin action, were accentuated also of those bud cultures which had been treated with BAP or with CFM. (2) In the callus cultures, the cytokinin characteristics appeared under CFM influence in the peroxidase patterns of one of the tissue culture strains only when CFM was applied in combination with BAP and not in combinations of CFM with IAA.The observed morphactin-induced increase in the cytokinin effects could occur via changes in the hormone level of the tissue.  相似文献

【背景】植物内生菌长期与宿主共生,对宿主生长发育产生影响。葛根作为重要的药食两用作物,葛根内生菌的研究具有重要实践意义。【目的】对广西葛根根部内生细菌进行分离、鉴定及促植物生长特性分析,旨在了解该药食同源植物内生细菌种群结构及其促生特性,为分析内生菌群体在药食同源植物产量和品质形成的作用及其内生细菌资源的开发利用提供参考。【方法】采用6种不同的培养基从广西葛根的根瘤、根系和根愈伤组织分离内生细菌,16S rRNA基因测序和系统发育分析内生细菌的分布特征和遗传多样性,采用生理生化方法测定分离菌株的固氮活性、溶磷特性、产生嗜铁素、分泌吲哚乙酸(indole-3-aceticacid,IAA)等促生特性。【结果】从葛根根瘤、根系和根部愈伤组织中共分离得到223个菌株,16S rRNA基因测序鉴定这些菌株隶属于2门4纲10科19属,其中芽孢杆菌属、假单胞菌属、土壤杆菌属、肠杆菌属为葛根优势菌群;内生细菌数量和群落组成存在明显的组织特异性,其数量表现为根瘤>根系>根愈伤组织,但其种群多样性表现为根愈伤组织>根系>根瘤。不同培养基分离出的细菌种群丰富度有差异。从供试菌株中筛...  相似文献

The interaction between mycorrhiza and leaf endophytes (Neotyphodium sp.) was studied in three Poa bonariensis populations, a native grass, differing significantly in endophyte infection. The association between endophytes and mycorrhizal fungi colonisation was assessed by analysing plant roots collected from the field. We found that roots from endophyte-infected populations showed a significantly higher frequency of colonisation by mycorrhizal fungi and that soil parameters were not related to endophyte infection or mycorrhiza colonization. In addition, we did not observe significant differences in the number of AM propagules in soils of the three populations sites. We also report the simultaneous development of Paris-type and Arum-type mycorrhiza morphology within the same root systems of P. bonariensis. The co-occurrence of both colonisation types in one and the same root system found in the three populations, which differed in Neotyphodium infection, suggests that foliar endophytes do not determine AM morphology. The percentage of root length colonised by different types of fungal structures (coils, arbuscules, longitudinal hyphae and vesicles) showed significant and positive differences in arbuscular frequency associated with endophyte infection, whereas the much smaller amounts of vesicles and hyphal coils did not differ significantly.  相似文献

[背景] 多种甲基杆菌属细菌对寄主植物有促生作用,其分布区域较广。筛选具有耐盐与促生特性的甲基杆菌属菌株可为微生物菌肥的开发提供依据。[目的] 从新疆乌尔禾地区盐渍土壤中筛选耐盐促生菌,对其培养基成分进行优化及促生能力进行研究,为微生物菌肥的开发提供依据。[方法] 采用阿须贝无氮培养基筛选耐盐菌株,对菌株进行基因序列分析及生理生化测定,采用平板试验法初步研究该菌对拟南芥的生长影响。[结果] 筛选出中度耐盐菌株W-1,经鉴定为甲基杆菌属(Methylobacteriumsp.)。菌株生长最佳无机盐为NaCl,最适浓度为1%–3%,最高耐受浓度达7%。最佳氮源为酸水解酪蛋白,产生长素最高达33.53 mg/L。溶磷能力达28.71 mg/L。菌株W-1接种拟南芥幼苗后叶绿素a和叶绿素b含量均高于对照组,同时对其根系发育有显著的促进作用。[结论] 菌株W-1促生性能显著,可为生物肥料制备提供菌种资源。  相似文献

Summary Success has been achieved in developing a complete protocol for mass propagation of Anogeissus pendula and A. latifolia, two important forest species found in India. Seeds cultured on plant growth regulator-free, semisolid Murashige and Skoog (MS) medium germinated within 5–6 wk and formed 4–6-cm long shoots. The shoots multiplied on MS+4.4 μM benzyladenine (BA)+5.7 μM indoleacetic acid (IAA) + casein hydrolysate (100 mgl−1) + ascorbic acid (50 mgl−1) + sucrose (3%) + agar (0.8%). A majority of the genotypes rooted with more than 90% efficiency when 5–6 cm individual shoots were cultured on 1/2MS (only major salts reduced to half strength)+2.3 μM IAA+2.5 μM indolebutyric acid (IBA) + sucrose (3%)+agar (0.8%) for 15 d. Those 10% (approx.) genotypes that did not root well on the above medium could be rooted with ease by increasing the concentration of IAA in the rooting media from 2.3 to 5.7 μM. The in vitro-raised plants were successfully transferred to the soil with a success rate of over 85%. Using this protocol, over 560 000 tissue-cultured plants of these two species have been produced and dispatched to various state forest departments for field trials and routine plantations.  相似文献

Propagation by softwood canes and cuttings is preferred as a practical system for vegetative reproduction of many ornamental plant species, despite the advances in tissue culture techniques. Dracaena purplecompacta L. is a species that has a high demand for exports. Conversely, coconut water (CW) is a rich supplement that naturally contains plant growth regulators such as indole acetic acid (IAA). The objective of this work was to evaluate the potential of CW extracts containing natural IAA, on adventitious root development in vegetative propagation of ornamental plant canes of D. purplecompacta L. Five different concentrations (28, 57, 143, 286, 571 μM of natural IAA) of CW extracts were tested. Another set of treatment was carried out with the same concentrations of authentic IAA hormone for comparison purpose. The 143-μM IAA CW extract recorded the best root induction and development. It was found that the root expression was faster (5 weeks) with the use of the novel method. In the conventional method, the canes are propagated by quick dip application of commercial product containing artificial hormone IAA and placing them on coir fiber dust beds. It takes up to 6 weeks for the canes to develop adventitious roots to the desired level. Steeping canes in 143-μM IAA CW extract improved rooting in D. purplecompacta L., and it was comparable to the application of 143-μM authentic IAA. The study indicates that adventitious root development, shoot development, and leaf emergence of D. purplecompacta L. is promoted by IAA CW extracts.  相似文献

This study examined the influence of three inoculant strains of Bradyrhizobium japonicum (Thal-8, Tal 620, Dulawala) on the ability of chickpea (Cicer arietinum (L.) to adapt to drought-stress. Strain Thal-8 was most effective in the root-nodule symbiosis and also partially alleviated decreased growth and yield imposed by drought stress. Strain Thal-8, in pure culture, also produced higher amounts of gibberellic acid (GA) and indole-3-acetic acid (IAA) and lower amounts of abscisic acid (ABA) than the other two test strains. Thal-8 increased the root biomass, GA and IAA contents of leaves of chickpea plants, including ICC 4948NN, a non-nodulating line. These results are consistent with the hypothesis that GA and IAA is produced by the Thal-8 strain and/or elevates levels of these phytohormones in chickpeas. This contributes to its high performance as a nitrogen-fixing microsymbiont. The growth-promoting response evoked by different strains of Bradyrhizobium correlated with higher ratios of GA and IAA relative to ABA phytohormones in the plants.  相似文献

A total of 216 bacterial strains were isolated from rice rhizospheric soils in Northern Thailand. The bacterial strains were initially tested for solubilization of inorganic phosphate, indole acetic acid (IAA) production, selected strains were then tested for optimized conditions for IAA production and whether these caused stimulatory effects on bean and maize seedling growth. It was found that all strains had solubilized inorganic phosphate (P), but only 18.05% produced IAA. The best IAA producer was identified by biochemical testing and 16S rDNA sequence analysis as Klebsiella SN 1.1. In addition to being the best IAA producer, this strain was a high P-solubilizer and produced the highest amount of IAA (291.97 ± 0.19 ppm) in culture media supplemented with l-tryptophan. The maximum production of IAA was achieved after 9 days of incubation. The culture requirements were optimized for maximum IAA production. The tested of IAA production by selected isolates was studied in a medium with 0, 0.1, 0.2, 0.5, 0.7, and 0.9% (v/v) l-tryptophan. Low levels (12.6 ppm) of IAA production was recorded without tryptophan addition. Production of IAA in Klebsiella SN 1.1 increased with an increase to 0.2% (v/v) tryptophan concentration. The production of IAA was further confirmed by extraction of crude IAA from this isolate and subsequent Thin Layer Chromatography (TLC) analysis. A specific spot from the extracted IAA production was found to correspond with a standard spot of IAA with the same R f value. The Klebsiella strain SN 1.1 also demonstrated stimulatory effects on bean seedlings in vivo.  相似文献
