The small Japanese "firefly squid," Watasenia scintillans, emits a bluish luminescence from dermal photogenic organs distributed along the ventral aspects of the head, mantle, funnel, arms and eyes. The brightest light is emitted by a cluster of three tiny organs located at the tip of each of the fourth pair of arms. Studies of extracts of the arm organs show that the light is due to a luciferin-luciferase reaction in which the luciferase is membrane-bound. The other components of the reaction are coelenterazine disulfate (luciferin), ATP, Mg(2+), and molecular oxygen. Based on the results, a reaction scheme is proposed which involves a rapid base/luciferase-catalyzed enolization of the keto group of the C-3 carbon of luciferin, followed by an adenylation of the enol group by ATP. The AMP serves as a recognition moiety for docking the substrate molecule to a luciferase bound to membrane, after which AMP is cleaved and a four-membered dioxetanone intermediate is formed by the addition of molecular oxygen. The intermediate then spontaneously decomposes to yield CO(2) and coelenteramide disulfate (oxyluciferin) in the excited state, which serves as the light emitter in the reaction.  相似文献

The mature sperm of Ptinella aptera is a helically coiled, flagellate gamete ca. 1.4 mm long—twice the length of the beetle itself. The rod-like acrosome, comprising the anterior part of the sperm, is 0.4 μm thick but is expanded as a flange around the nucleus and the base of the tail, increasing the diameter of the sperm to 2 μm. The bulk of the tail consists of a pair of bodies with a characteristic ultrastructure of longitudinal tubules beneath a lamellar cortical layer. These bodies are probably homologous with the mitochondrial derivatives of other insect sperm. The axoneme is helically coiled and is flanked by a single accessory body. One of the ‘structured bodies’ is connected to the acrosome and the other to the accessory body. The sperm move actively in the female reproductive tract. The functional significance of this behaviour and of the evolution of the large gamete itself is discussed in relation to existing hypotheses.  相似文献

本文记述了寄生在鲫鱼(Carassius auratus)体表和鳃上一种新的车轮虫(Trichodina carassii sp.nov.),虫体直径70(54.6—80.4)μm,附着盘直径58(51.6—67.2)μm,齿环直径32(27.0—36.2)μm,齿体25—30个,齿环中央有7—14个旧齿体残余物形成的颗粒结构。大核马蹄形。小核短杆状,在大核一端的外侧,其位置略有变动。口沟390—400°。齿钩镰刀状,齿棘较直,侧面具有若干个缺刻,末端钝圆或平截,近锥体的后方有一个不太明显的小突起。    相似文献

In this study, we describe 2 new species of Eimeria associated with the yellow-crowned Amazon Amazona ochrocephala. Eimeria amazonae n. sp. has bilayered, ellipsoidal, and smooth oocysts that measure 48.9 × 36.2 μm; the length/width ratio is 1.35. The micropyle and oocyst residuum are both absent, but the polar granule is present. Ovoidal sporocysts are 22.2 × 11.9 μm. Stieda and sub-Stieda bodies and sporocyst residuum are present. The 2 elongate sporozoites are curved and measure 18.1 × 3.4 μm; both have 2 refractile bodies. Eimeria ochrocephalae n. sp. has bilayered, ellipsoidal, and smooth oocysts that measure 43.8 × 27.7 μm; the length/width ratio is 1.58. The micropyle and oocyst residuum are absent, but the polar granule is present; ovoidal sporocysts are 20.6 × 10.1 μm. Stieda and sub-Stieda bodies and sporocyst residuum are present; 2 elongate and curved sporozoites are 15.8 × 3.4 μm, each of which has 2 refractile bodies.  相似文献

The ultrastructure of the upper lip, which is the location of the light organ in luminescent ostracods, is described for two species of marine ostracods (Vargula graminicola, and V. tsujii). The light organ contains four primary cell types (here designated A–D) readily identified both by the types of vesicles that they contain and their location. Cell type A, restricted to the center of the light organ, has vesicles that are homogeneous throughout. Cell type B has vesicles with a single electron-dense central area. This cell is most common in the lateral regions but is also found in the tusks. Immunocytochemical experiments revealed that luciferase is found in B cells, but not in any other cells or in control specimens. Cell type C has vesicles containing many electrondense microstructures; this cell type is the least common and is found along with B cells within the lateral margins of the light organ. Cell type D has granulated vesicles and is restricted to the tusks. Metachromatic staining with toluidine blue suggests that the vesicles contain acidic mucosubstances. The cells of the light organ are very long (360–460 μm) and extend through its entire length. The cells found exclusively in the tusks are shorter (ea. 150 μm). All cells possess similar polarity: proximal synthesis, middle transitional, and distal secretory regions. These cells terminate ventrally at nozzles on the anterior portion of the upper lip, and on the tusks. The upper lip and light organ possess two large longitudinal muscles, a central muscle, and several smaller “bridle” muscles that attach onto sclerotized ridges. 利用光学显微镜、荧光显微镜、扫描电镜及分子生物学等方法, 对分布于我国东海海域的剧毒卡尔藻(Karlodinium veneficum)藻株(LAMB090611)的形态特征和显微结构进行了描述, 并探讨了其分子系统进化关系。该藻株细胞长11.1-18.7 μm, 平均值为(14.2±1.8) μm, 宽8.2-14.7 μm, 平均值为(10.8±1.5) μm。细胞形态结构特征为: 上下锥体积基本相同; 顶沟短而直; 腹孔明显; 纵沟延伸至上锥; 横沟错位距离约占细胞总长的28%-38%; 含有2或4个不规则形态的叶绿体; 细胞核位于中部或下锥。此藻种的暴发可引发有害赤潮(harmful algal bloom)。当前加强有害赤潮的预防和监测工作是减少危害的有效途径, 而对引发赤潮原因种的准确识别和鉴定则是基础和关键。  相似文献

The emission of light in the marine ostracod Cypridina hilgendorfii (presently Vargula hilgendorfii) is produced by the Cypridina luciferin-luciferase reaction in the presence of molecular oxygen. Cypridina luciferin has an asymmetric carbon derived from isoleucine, and the absolute configuration is identical to the C-3 position in L-isoleucine or D-alloisoleucine. To determine the stereoselective incorporation of the isoleucine isomers (L-isoleucine, D-isoleucine, L-alloisoleucine, and D-alloisoleucine), we synthesized four (2)H-labeled isoleucine isomers and examined their incorporation into Cypridina luciferin by feeding experiments. Judging by these results, L-isoleucine is predominantly incorporated into Cypridina luciferin. This suggests that the isoleucine unit of Cypridina luciferin is derived from L-isoleucine, but not from D-alloisoleucine.  相似文献

The luminescence analysis of ATP requires the rapid and adequate mixing of 10 μliters of low viscosity ATP extract with 100 μliters of viscous luciferin-luciferase preparation. The enzyme-substrate mixture is itself sensitive and will luminesce if excessively agitated. Therefore, mixing must be rapid enough to maximize ATP dependent light emission, but not so vigorous as to cause excitation of the luciferin-luciferase in the absence of ATP. To satisfy this requirement, an automated, pneumatic sample injector was developed and its performance compared to manual injection and to two commercially available mechanical systems. Over a wide range of ATP concentrations (1 × 10?7 to 1 × 10?5 μg/μliters) the pneumatic injector demonstrated precision and sensitivity comparable to one of the mechanical injectors and superior to either manual injection or the other mechanical system. In addition, the automated system offered automatic refill, variable injection velocity, volume control, reliability, and ease of operation.  相似文献

The macula lagenae of the anabantide fish Colisa labiosa was studied with light and transmission electron microscopy. (1) The sensory area is naturally divided in a central area (A) surrounded by a peripheral part (B). (2) Generally the central hair cells are separated by supporting cells, while the peripheral hair cells are found in groups. The cells of a group are not separated by supporting cells. (3) Tubuli-like structures, hexagonal in cross section, are found in all cells. In peripheral hair cells the longitudinally oriented tubuli-like structures are aggregated in thick bundles. (4) Variation in shape, electron density, stereocilia arrangement and size of mitochondria was found in different hair cells. (5) The central hair cells contain large accumulations of presynaptic bodies (10–44). Contrarily, the peripheral hair cells contain only a few pre-synaptic bodies (1–3). (6) The central hair cells are innervated by thick afferent (6–15 μm) and fine presumed efferent (less than 1 μm nerve fibres, while the peripheral hair cells are innervated by thin (1–6 μm) afferent nerve fibres only.  相似文献

Photosynthesis versus irradiance curves and their associated photosynthetic parameters from different phototrophic biofilms isolated from an extreme acidic environment (Río Tinto, SW, Spain) were studied in order to relate them to their species composition and the physicochemical characteristics of their respective sampling locations. The results indicated that the biofilms are low light acclimated showing a photoinhibition model; only floating communities of filamentous algae showed a light saturation model. Thus, all the biofilms analysed showed photoinhibition over 60 μmol photon m(-2) s(-1) except in the case of Zygnemopsis sp. sample, which showed a light-saturated photosynthesis model under irradiations higher that 200 μmol photon m(-2) s(-1). The highest values of compensation light intensity (I(c)) were showed also by Zygnemosis sp. biofilm (c. 40 μmol photon m(-2) s(-1)), followed by Euglena mutabilis and Chlorella sp. samples (c. 20 μmol photon m(-2) s(-1)). The diatom sample showed the lowest I(c) values (c. 5 μmol photon m(-2) s(-1)). As far as we know this is the first attempt to determine the photosynthetic activity of low pH and heavy metal tolerant phototrophic biofilms, which may give light in the understanding of the ecological importance of these biofilms for the maintenance of the primary production of these extreme and unique ecosystems.  相似文献

ABSTRACT. Four isosporan species are described from the small tree finch, Camarhynchus parvulus from Isabela Island on the Galapagos Archipelago. Isospora exigua n. sp. oocysts subspheroidal, one-layered, smooth, yellow-brown color, 20.4 × 20.1 (20-23 × 18-23) μm, with no micropyle, residuum, or polar body. Sporocysts ovoidal, 14 × 9.5 (13-15 × 8-10) μm, with small Stieda and substieda bodies and irregular-shaped residuum. Isospora rotunda n. sp. oocysts subspheroidal, single-layered, smooth, yellow-brown wall with large polar body and no micropyle or residuum, 20.9 × 20.8 (20-24 × 19-23) μm. Sporocysts ovoidal, 15 × 9.7 (13-16 × 9-10) μm with knob-like Stieda bodies, prominent substieda bodies and round residuum. Isospora fragmenta n. sp. oocysts subspheroidal with no micropyle or residuum but with many splinter-like polar granules and a smooth, colorless, single-layered wall, 25.3 × 24.2 (24-27 × 23-25) μm. Sporocysts piriform 15.4 × 11.5 (14-17 × 11-12) μm with knob-like Stieda bodies, prominent substieda bodies, and irregular-shaped residuum. Isospora temeraria n. sp. oocysts ellipsoidal with one polar body, no micropyle or residuum, and wall of a single layer, smooth, yellow-brown color, 25.4 × 21.1 (21-30 × 17-23) μm. Sporocysts piriform, 15 × 10 (14-15 × 9-11) μm with knob-like Stieda bodies, prominent substieda bodies, and a round residuum. One woodpecker finch, Cactospiza pallida, was found to be infected with I. exigua, and a warbler finch, Certhidea olivacea was infected with I. fragmenta.  相似文献

We studied the interactions of the CO(2)-concentrating mechanism and variable light in the filamentous cyanobacterium Leptolyngbya sp. CPCC 696 acclimated to low light (15 μmol m(-2) s(-1) PPFD) and low inorganic carbon (50 μM Ci). Mass spectrometric and polarographic analysis revealed that mediated CO(2) uptake along with both active Na(+)-independent and Na(+)-dependent HCO(3)(-) transport, likely through Na(+)/HCO(3)(-) symport, were employed to concentrate Ci internally. Combined transport of CO(2) and HCO(3)(-) required about 30 kJ mol(-1) of energy from photosynthetic electron transport to support an intracellular Ci accumulation 550-fold greater than the external Ci. Initially, Leptolyngbya rapidly induced oxygen evolution and Ci transport to reach 40-50% of maximum values by 50 μmol m(-2) s(-1) PPFD. Thereafter, photosynthesis and Ci transport increased gradually to saturation around 1,800 μmol m(-2) s(-1) PPFD. Leptolyngbya showed a low intrinsic susceptibility to photoinhibition of oxygen evolution up to PPFD of 3,000 μmol m(-2) s(-1). Intracellular Ci accumulation showed a lag under low light but then peaked at about 500 μmol photons m(-2) s(-1) and remained high thereafter. Ci influx was accompanied by a simultaneous, light-dependent, outward flux of CO(2) and by internal CO(2)/HCO(3)(-) cycling. The high-affinity and high-capacity CCM of Leptolyngbya responded dynamically to fluctuating PPFD and used excitation energy in excess of the needs of CO(2) fixation by increasing Ci transport, accumulation and Ci cycling. This capacity may allow Leptolyngbya to tolerate periodic exposure to excess high light by consuming electron equivalents and keeping PSII open.  相似文献

Marinesco bodies were discovered in substantia nigra neurons of human brain in 1902. The relationships between these intranuclear inclusions and the other structures of the cellular nucleus are still obscure. The aim of this study is to elucidate the morphological and cytochemical peculiarities of intranuclear ubiquitin-immunopositive bodies in the substantia nigra neurons of human brain and to evaluate the interconnections of these peculiarities with nucleolus by means of light microscopy, immunocytochemistry, and confocal laser microscopy. It is found that up to 20% of neurons in substantia nigra of human brain contain ubiquitin-immunopositive Marinesco bodies. These rounded structures are 1–8 μm—more often 2–4 μm—in diameter. Only one-third of them are tightly adjacent to the nucleolus. By a method of silver impregnation of argentophilic proteins associated with nucleolar organizer, the absence was shown of argentophilic proteins, which are characteristic for the nucleolus, in Marinesco bodies. Special ubiquitin-positive substantially smaller structures (less than 1 μm) are revealed in the neurons’ nuclei along with Marinesco bodies. These structures are probably the initial forms in the formation of Marinesco bodies. The existence of two types of ubiquitin-immunopositive intranuclear bodies is revealed by means of confocal microscopy: one has high intensity of immunofluorescence, and the other has low intensity. Heterogeneous distribution of immunopositive product is characteristic of the former. The presence of DNA in Marinesco bodies is detected by using SYTOX Green fluorescent dye. The absence of peripheral heterochromatin zone and weak susceptibility to toluidine blue together with the presence of DNA and the absence of argentophilic proteins suggests substantial structural and chemical differences between Marinesco bodies and nucleoli, which argues against the idea that the detected bodies are modified nucleoli.  相似文献

Two components can be obtained from the luminescent mollusc Pholas dactylus which, when mixed aerobically, produce light. Purification of the substrate luciferin is described. This component is a protein (mol. wt. 50000); absorption and fluorescence spectra are given.

The kinetics of the reaction leading to light emission were studied. A biphasic reaction could be induced by incubation of luciferin-luciferase in the absence of O2 or by incubation at pH 4.8 at o°. Injection of reduced flavin mononucleotide resulted in a flash if injected during the reaction but not after. Among metal ions, only Fe2+ showed a strong stimulation.  相似文献

岩黄连光合与蒸腾特性及其对光照强度和CO2浓度的响应   总被引:15,自引:2,他引:15  
采用LI-6400便携式光合测定系统(Li-CorInc.,USA)对岩黄连叶片的气体交换进行了测定。结果表明(1)岩黄连叶片的光饱和点(LSP)为329.18μmol.m-2.s-1左右,光补偿点(LCP)为12.76μmol.m-2.s-1,最大净光合速率为2.96μmol.m-2.s-1,暗呼吸速率(Rd)为0.17μmol.m-2.s-1。光饱和点和光补偿点都比效低,表明岩黄连对光照的要求不高,属于阴生植物。(2)4月份,岩黄连Pn随CO2浓度升高而逐渐增大。当CO2浓度由50μmol.mol-1增加到600μmol.mol-1,Pn几乎呈直线上升,600~1000μmol.mol-1范围内逐渐缓和,到1000μmol.mol-1以后Pn变化平稳。由曲线估算CO2饱和点(CSP)大约在1000μmol.mol-1左右。CO2的补偿点为68.80μmol.mol-1。羧化效率为0.0308μmol.m-2.s-1。(3)岩黄连叶片水分利用率(WUE)随有效光辐射强度(PAR)的增强呈抛物线状变化,PAR在200μmol.m-2.s-1内呈直线上升,到200μmol.m-2.s-1时WUE达最大值,大于200μmol.m-2.s-1后WUE呈逐渐下降趋势。  相似文献

The cortical region of the 2-cell stage egg of the gastropod Lymnaea palustris was studied by light and electron microscopy. This region includes (1) a vitelline membrane and perivitelline space which contain membrane-limited dense bodies derived from the cell surface, (2) oolemma with surface coat material and microvilli, and (3) a peripheral zone of cytoplasm (0.5-5.0 μm wide) composed of irregular vesicles, electron dense granules, and cytoplasmic microtubules. Microtubules are most abundant in the equatorial region of the egg, where they form arrays that are parallel and oblique to the egg's surface. Microtubular profiles also occur in the cortical region at the animal and vegetal poles of the egg and in the endoplasm. They may play a role in cortical segregation.  相似文献
