通常可通过植物叶片的形态来区分不同植物的种类。叶片由茎顶端分生组织侧翼发育而成,为多种多样大小和形状的扁平结构。叶片的结构看似简单,但调控叶片形态和结构发育的分子机理错综复杂,叶片的发育受植物激素、转录因子、一系列蛋白因子及环境的共同调控。本文回顾了叶片边缘形态和叶脉发育研究的最新进展。在叶边缘形态方面,Aux/IAA生长素响应抑制家族蛋白通过调节生长素浓度最大点的离散分布影响小叶的起始和生长以及叶边缘结构;NAM/CUC转录因子促进叶边缘锯齿的分离以及复叶中小叶的分离和分化,NAM/CUC和Aux/IAA通过不同通路实现对生长素的调控;拟南芥RAX1基因/番茄Potato-leaf基因和拟南芥JAG基因/番茄LYR基因促进叶边缘锯齿发育;RCO调控复叶小叶的发育不通过改变生长素的分布来实现;在番茄中反式小干扰RNA途径中的因子参与叶边缘形态发育;另外,在拟南芥中,mir164A、CUC2、PIN1、DPA4、SVR9-1及SVR9L-1构成复杂的调控网络影响叶边缘锯齿的发育。在叶脉发育方面,PIN1能否正确的定位会影响叶脉发育;AS1和AS2共同参与叶片远近轴极性的分化;另外AXR6、MP、BDL、CVP因子功能的缺失影响叶脉发育;生长素、PIN1、Aux/IAA、MP、ATHB8构成反馈循环调控子叶叶脉的形成。  相似文献

The shoot stem cell niche, contained within the shoot apical meristem (SAM) is maintained in Arabidopsis by the homeodomain protein SHOOT MERISTEMLESS (STM). STM is a mobile protein that traffics cell‐to‐cell, presumably through plasmodesmata. In maize, the STM homolog KNOTTED1 shows clear differences between mRNA and protein localization domains in the SAM. However, the STM mRNA and protein localization domains are not obviously different in Arabidopsis, and the functional relevance of STM mobility is unknown. Using a non‐mobile version of STM (2xNLS‐YFP‐STM), we show that STM mobility is required to suppress axillary meristem formation during embryogenesis, to maintain meristem size, and to precisely specify organ boundaries throughout development. STM and organ boundary genes CUP SHAPED COTYLEDON1 (CUC1), CUC2 and CUC3 regulate each other during embryogenesis to establish the embryonic SAM and to specify cotyledon boundaries, and STM controls CUC expression post‐embryonically at organ boundary domains. We show that organ boundary specification by correct spatial expression of CUC genes requires STM mobility in the meristem. Our data suggest that STM mobility is critical for its normal function in shoot stem cell control.  相似文献

生物多样性研究的关键问题之一是表型多样性的形成和演化机制, 因为表型多样性与物种多样性密切相关, 同时又承载着遗传和环境的变异信息。植物的叶具有丰富的形态多样性, 而叶形多样性很大程度上体现在叶边缘形态的变异。叶边缘的形态可从全缘、锯齿状到具有不同程度(深浅)和不同式样(羽状或掌状、回数等)的裂片(在发育研究中复叶的小叶也描述为裂片)。关于叶缘齿/裂的发育调控机制, 在拟南芥(Arabidopsis thaliana)、碎米荠(Cardamine hirsuta)、番茄(Solanum lycopersicum)等模式植物中已有较深入的探讨。研究发现, 很多转录因子、小分子RNA及植物激素对叶齿/裂或小叶的形成具有调控作用, 其中生长素输出途径中的转录因子NAM/CUC、miR164以及高浓度生长素的反馈调控可能起到核心作用, 而且该调控模块在真双子叶植物中较为保守; TCP类、SPL类转录因子和其他一些miRNA也在生长素输出途径中发挥作用; 关于KNOX家族转录因子的作用, 虽然多数研究是围绕复叶的形态建成, 但也有数据显示其在叶裂发育中发挥作用。此外, 对拟南芥和碎米荠等十字花科植物的研究还发现, 调控基因RCO通过抑制小叶/裂片之间的细胞增殖而对小叶/叶裂的发育发挥作用。本文综述上述多角度的研究进展, 并尝试概括叶边缘形态的发育调控网络, 为关于叶缘形态多样性形成机制的研究提供可参考的切入点。  相似文献

In Arabidopsis, SEUSS (SEU) and SEUSS‐LIKE 2 (SLK2) are components of the LEUNIG (LUG) repressor complex that coordinates various aspects of post‐embryonic development. The complex also plays a critical role during embryogenesis, as seu slk2 double mutants have small, narrow cotyledons and lack a shoot apical meristem (SAM). Here we show that seu slk2 double mutant embryos exhibit delayed cotyledon outgrowth and that this is associated with altered PIN‐FORMED1 (PIN1) expression and localisation during the early stages of embryogenesis. These observations suggest that SEU and SLK2 promote the transition to bilateral symmetry by modulating auxin distribution in the embryonic shoot. This study also shows that loss of SAM formation in seu slk2 mutants is associated with reduced expression of the class I KNOX (KNOXI) genes SHOOTMERISTEMLESS (STM), BREVIPEDICELLUS and KNAT2. Furthermore, elevating STM expression in seu slk2 mutant embryos was sufficient to restore SAM formation but not post‐embryonic activity, while both SAM formation and activity were rescued when SLK2 expression was restored in either the cotyledons or boundary regions. These results demonstrate that SEU and SLK2 function redundantly to promote embryonic shoot development and likely act through a non‐cell autonomous pathway to promote KNOXI activity.  相似文献

Abstract Two methods of analyses were used to investigate tooth development in serrate (se) mutant and wild-type Columbia-1 (Col-1) Arabidopsis thaliana leaves. There were almost twice as many teeth with deeper sinuses and two orders of toothing on the margins of serrate compared with Columbia-1 leaves. The main objective of this study was to test three hypotheses relative to the source of polymorphism in tooth development: (i) Teeth share similar growth rates and initial sizes, but the deeper teeth are initiated earlier in leaf development. (ii) Teeth share similar timing of initiation and growth rates, but the deeper teeth have a larger initial size. (iii) Teeth share similar timing of initiation and initial sizes, but the deeper teeth have a faster growth rate. Leaf plastochron index (LPI) was used as the time variable for leaf development. Results showed teeth in se were initiated at −27 LPI, 15 plastochrons earlier than those of Col-1. Serrate leaf expansion was biphasic, with the early phase expanding at half the relative plastochron rate of the later phase, which equaled the constant relative expansion rate of Col-1 leaves. Allometric analyses of tooth development obscured the interactions between time of tooth and leaf initiation and the early phase of leaf expansion characteristic of serrate leaves and teeth. Timing of developmental events that allometric analysis obscured can be readily detected with the LPI as a developmental index. Received 25 January 2000; accepted 17 March 2000  相似文献

Abstract

Leaves at the apex of a mature Aphanoregma patens (Hedw.) Lindb. (Physcomitrella patens (Hedw.) Bruch Schimp. in B.S.G.) gametophore differ markedly in size and form from those at its base. To determine how these differences are produced during development, we first examined qualitative and quantitative differences between successive leaves along the stem and among leaves at different developmental stages. Differences between successive leaves were slight and cumulative. Local changes in cell number and size combined to produce a regularly shaped and approximately bilaterally symmetrical leaf suggesting that cell division and cell expansion are regionally regulated and coordinated at the organ level. The midrib and marginal teeth are discrete characters, which were prefigured by changes in cell shape in leaves that lacked these characters. In leaf primordia, cell proliferation was responsible for most of the changes in leaf form and size early in development and may have continued as cell expansion took over as the primary contributor to leaf growth and morphogenesis. Thus, leaf heteroblasty in Physcomitrella probably results from modulation of a single developmental programme by external and/or internal forces, which alter progressively in intensity as a gametophore grows. We applied exogenous cytokinin and auxin separately to growing cultures to explore their effects on leaf growth. Cytokinin and auxin stimulated leaf cell division and leaf cell elongation, respectively. Also, young upper leaves of gametophores exposed to exogenous auxin closely resembled basal leaves of untreated plants. Therefore, endogenous cytokinins and auxins may be among the modulating internal forces involved in leaf morphogenesis and the establishment of leaf heteroblasty.  相似文献

A leaf undergoes determinate growth from a primordium on flank of the shoot apical meristem. Several intrinsic pathways restrict meristematic activity in the leaf of Arabidopsis; however, other factors remain to be defined. We report here that the overexpression of MINI ZINC FINGER1 (MIF1) or MIF3 disrupted the leaf determinate growth by inducing ectopic shoot meristems on leaf margins. These ectopic meristems occurred along margins of late rosette leaves at serration sinuses in an ERECTA-dependent manner. Expression of STM was activated in these ectopic meristems but not other leaf regions. The formation of ectopic meristems was independent of the BP gene but suppressed by exogenous gibberellic acid. In addition, reduced auxin response along leaf margins and subsequent response peak in the sinus were correlated with the occurrence of ectopic meristems. Our results suggest that the sinus of leaf serration is a quiescent domain possessing the potential for meristem formation. MIF1- or MIF3-overexpressing transgenic plants may provide a new genetic system for dissecting the molecular mechanism that maintains leaf determinate growth, and for understanding the interactions between hormone actions and meristematic activity.  相似文献

The balance between cell proliferation and cell differentiation is essential for leaf patterning. However, identification of the factors coordinating leaf patterning and cell growth behavior is challenging. Here, we characterized a temperature‐sensitive Arabidopsis mutant with leaf blade and venation defects. We mapped the mutation to the sub‐2 allele of the SCRAMBLED/STRUBBELIG (SCM/SUB) receptor‐like kinase gene whose functions in leaf development have not been demonstrated. The sub‐2 mutant displayed impaired blade development, asymmetric leaf shape and altered venation patterning under high ambient temperature (30°C), but these defects were less pronounced at normal growth temperature (22°C). Loss of SCM/SUB function results in reduced cell proliferation and abnormal cell expansion, as well as altered auxin patterning. SCM/SUB is initially expressed throughout leaf primordia and becomes restricted to the vascular cells, coinciding with its roles in early leaf patterning and venation formation. Furthermore, constitutive expression of the SCM/SUB gene also restricts organ growth by inhibiting the transition from cell proliferation to expansion. We propose the existence of a SCM/SUB‐mediated developmental stage‐specific signal for leaf patterning, and highlight the importance of the balance between cell proliferation and differentiation for leaf morphogenesis.  相似文献

The phytohormone auxin plays a critical role in plant development, including embryogenesis, organogenesis, tropism, apical dominance and in cell growth, division, and expansion. In these processes, the concentration gradient of auxin, which is established by polar auxin transport mediated by PIN-FORMED (PIN) proteins and several ATP-binding cassette/multi-drug resistance/P-glycoprotein (ABCB/MDR/PGP) transporters, is a crucial signal. Here, we characterized the function of ABCB19 in the control of Arabidopsis organ boundary development. We identified a new abcb19 allele, abcb19-5, which showed stem-cauline leaf and stem-pedicel fusion defects. By virtue of the DII-VENUS marker, the auxin level was found to be increased at the organ boundary region in the inflorescence apex. The expression of CUP-SHAPED COTYLEDON2 (CUC2) was decreased, while no obvious change in the expression of CUC3 was observed, in abcb19. In addition, the fusion defects were greatly enhanced in cuc3 abcb19-5, which was reminiscent of cuc2 cuc3. We also found that some other organ boundary genes, such as LOF1/2 were down-regulated in abcb19. Together, these results reveal a new aspect of auxin transporter ABCB19 function, which is largely dependent on the positive regulation of organ boundary genes CUC2 and LOFs at the postembryonic organ boundary.  相似文献

In Arabidopsis, INDOLE-3-BUTYRIC ACID RESPONSE5 (IBR5), a putative dual-specificity protein phosphatase, is a positive regulator of auxin response. Mutations in IBR5 result in decreased plant height, defective vascular development, increased leaf serration, fewer lateral roots, and resistance to the phytohormones auxin and abscisic acid. Abstract: Radular teeth occur between the jaws in two specimens of the Late Cretaceous scaphitid ammonite Rhaeboceras halli (Meek and Hayden, 1856) from the Western Interior of the United States. The detailed morphology of the teeth has been revealed by propagation phase contrast X‐ray synchrotron microtomography. Each row of the radula of R. halli consists of a total of seven teeth (a central rachidian, two pairs of lateral and one pair of marginal teeth), as in other known ammonoid radulae, although the central tooth could not be confirmed in the specimens examined. The lateral teeth are multicuspid and robust, and the marginal teeth are long (4.6 mm) and slender. In overall morphology, the heterodont and ctenoglossan radula of R. halli is similar that of Jurassic and Cretaceous ammonites with the same aptychus‐type lower jaw, that is, the Aptychophora. This discovery reveals the range of variation in radular morphology, which could be related to ecological or phylogenetic factors. It also invalidates the hypothesis that the hook‐like structures in R. halli previously described are radular elements.  相似文献

Compound leaf development requires highly regulated cell proliferation, differentiation, and expansion patterns. We identified loss-of-function alleles at the SMOOTH LEAF MARGIN1 (SLM1) locus in Medicago truncatula, a model legume species with trifoliate adult leaves. SLM1 encodes an auxin efflux carrier protein and is the ortholog of Arabidopsis thaliana PIN-FORMED1 (PIN1). Auxin distribution is impaired in the slm1 mutant, resulting in pleiotropic phenotypes in different organs. The most striking change in slm1 is the increase in the number of terminal leaflets and a simultaneous reduction in the number of lateral leaflets, accompanied by reduced expression of SINGLE LEAFLET1 (SGL1), an ortholog of LEAFY. Characterization of the mutant indicates that distinct developmental domains exist in the formation of terminal and lateral leaflets. In contrast with the pinnate compound leaves in the wild type, the slm1 sgl1 double mutant shows nonpeltately palmate leaves, suggesting that the terminal leaflet primordium in M. truncatula has a unique developmental mechanism. Further investigations on the development of leaf serrations reveal different ontogenies between distal serration and marginal serration formation as well as between serration and leaflet formation. These data suggest that regulation of the elaboration of compound leaves and serrations is context dependent and tightly correlated with the auxin/SLM1 module in M. truncatula.  相似文献
