We present the results of a series of computer simulations thatexamined the impact of winner, loser, and bystander effectson hierarchy formation in fused groups. These effects and theirimplications for hierarchy structure and aggressive interactionswere first examined in small four-member groups. Subsequentto this, the two small groups were fused into a single largergroup. Further interactions took place in this fused group,generating a new hierarchy. Our models demonstrate clearly thatwinner, loser, and bystander effects strongly influence boththe structure and types of interactions that emerge from thefusion of smaller groups. Four conditions produced results inwhich the same general patterns were uncovered in pre- and postfusiongroups: (1) winner effects alone, (2) bystander loser effectsalone, (3) winner and bystander winner effects operating simultaneously,and (4) all four effects in play simultaneously. Outside thisparameter space, hierarchy structure and the nature of aggressiveinteractions differed in pre- and postfusion groups. When onlyloser effects were in play, one of the two clear alphas fromthe prefused groups dropped in rank in the eight-member fusedgroup. When bystander winner effects were in play, it was difficultto rank any of the eight individuals in the fused group, andplayers interacted almost exclusively with those that were notin their original four-member group. When loser and bystanderloser effects operated simultaneously, two top-ranking individualsemerged in the fused groups, but the relative rank of the otherplayers was difficult to assign.  相似文献

We studied whether in utero exposure to economic hardship during a grandmother’s pregnancy has a transgenerational effect on her grandchildren's health condition. We used an individual-level three-generation data set covering people born between 1734 and 1840 in the municipality of Rendalen in Norway. We found a culling effect in which grandchildren whose grandmothers gave birth in years of economic hardship lived approximately ten years longer than grandchildren whose mothers were born in years of economic well-being. This impact was only observed among the grandmothers who belong to the lowest social classes. Our results also showed that in higher social classes, economic hardship during a grandmother’s pregnancy deteriorated her grandchildren’s health by “scarring” the mother’s health.  相似文献

The well-known mitogenic effects of TSH observed in vivo on the thyroid are not always reproducible of human thyroid cells in vitro where conflicting results have been obtained. In order to clarify this issue, we have used primary cultures of human thyroid cells obtained from normal tissue and maintained in serum-free medium for several days. In this in vitro model we have studied the effect of TSH on growth by measuring three different parameters: [3H]-thymidine incorporation, cell counts, and DNA measurement. Monolayer cultures were plated at both low and high cell density (2 x 10(4) and 8 x 10(4) cells/25 mm well, respectively). Although at either cell density cultures were equally able to functionally respond to TSH in terms of cAMP accumulation a significant growth response to TSH was observed only in low density cultures. In high density cultures TSH had an antimitogenic effect. Moreover, TSH potentiated the mitogenic effect of insulin only in low density cultures. In contrast to TSH, FCS induced a similar proliferative response at both high and low cell density. Following TSH stimulation, cAMP content was always increased, paralleling the effect of growth in low density but not in high density cultures. The cAMP analogues dibutyryl-cAMP and 8-bromo-cAMP, as well as cholera toxin and forskolin, did not mimic the mitogenic effect of TSH but had an antiproliferative effect. In addition, these agents blunted the proliferative effect of insulin. These data suggest that in thyroid cells TSH is able to elicit both a mitogenic and an antimitogenic effect depending on the environmental conditions such as cell density.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Polarization effects in light scattering are sensitive indicators of cell structure and structural changes in time. In the spectral regions where the optical properties of the scatterers are relatively constant, the scattering pattern scales, it contracts or expands in a predictable manner as a function of the wavelength. In the spectral regions where the optical properties are strongly wavelength dependent (near absorption bands, etc.) the scattering curves do not scale, but change drastically in phase and amplitude as the wavelength is varied. Reported here is an empirical study of the magnitude of the influence of absorption on the polarization effects in light scattering. Scattering curves have been obtained for human red blood cells in the absorption band (blue light) and far from the absorption band (red light). The scattering at these wavelengths shows very strong nonscaling differences. These observations suggest the use of polarization effects in light scattering and their wavelength dependence for the studies of structural changes in cell nuclei. Nucleoproteins have strong absorption, optical rotatory dispersion and circular dichroism bands in the ultraviolet region of the spectrum, whereas there is little ψ-dependence in the visible range. There is also the possibility of binding specific chromophoric dyes to cell components, thus introducing absorption bands in the visible range, where scattering instrumentation and laser light sources are more readily available.  相似文献

Pueraria lobata root (PLR), well known as Kudzu root, has recently become commercially available in Western dietary supplements for menopausal symptoms. The scientific basis for its action has been attributed to the action of phytoestrogens. This study aimed to investigate the estrogen-like activity of isoflavonoids isolated from P. lobata root and their safety with respect to their effect on breast cancer cell proliferation. In an E-screen assay, crude MeOH extract of PLR significantly increased the proliferation of MCF-7 cells in a concentration-dependent manner. Among the four fractions obtained by solvent fractionation of MeOH extract, the n-BuOH fraction had significant estrogen-like activities at all concentrations tested. Phytochemical analysis of the n-BuOH fraction led to the isolation of 10 isoflavones (110), among which genistein (10) had significant estrogen-like activities at all concentrations tested. These activities were significantly enhanced by treatment with genistein and 17β-estradiol compared with 17β-estradiol alone, and this effect was mediated by decreased expression of estrogen receptor (ER)α and phospho-ERα in MCF-7 cells. In a cell cytotoxicity assay, genistein (10) exhibited significant cytotoxicity in both ER-positive MCF-7 and ER-negative MDA-MB-231 breast cancer cells. This cytotoxicity was characterized by the induction of apoptotic cells stained with annexin V conjugated with Alexa Fluor 488 and involved activation of mitochondria-independent and -dependent apoptosis pathways in MCF-7 cells. Our results demonstrated that genistein (10) has estrogen-like effects dependent on ER pathway activation and anti-proliferative effects mediated by the apoptosis pathway rather than the ER pathway in MCF-7 breast cancer cells.  相似文献

Cultures of MDCK II and human fibroblast cells were fed radioactive sphingosine and a radioactive GM3 ganglioside derivative containing a photoactivable group. The derived cell homogenates were treated with Triton X-100 and fractionated by sucrose-gradient centrifugation to prepare a detergent-insoluble membrane fraction known to be enriched in sphingolipid and caveolin-1, i.e. of caveolae. The detergent-insoluble membrane fraction prepared after feeding [1-3H]sphingosine to cells, was found to be highly enriched, with respect to protein content, in metabolically radiolabeled sphingomyelin and glycosphingolipids (about 18-fold). By feeding cells photoactivable radioactive GM3, after 2 h-chase, caveolin-1, CAV1, and proteins of high molecular mass became cross-linked to GM3, the cross-linking complexes being highly concentrated in the detergent-insoluble membrane fraction. The interaction between the ganglioside derivative and CAV1 was a time-dependent, transient process so that CAV1 cross-linking to GM3 was hardly detectable after a 24-h chase followed the pulse time. After a 24-h chase, only the high molecular mass proteins cross-linked to GM3 could be clearly observed. These results suggest that a portion of the GM3 administered to cells enters caveolae and moves to the glycosphingolipid domains, or enters caveolae that are then rapidly catabolized. Electron microscopy of cells in a culture immunostained with a monoclonal antibody to GM3 and a secondary gold-conjugated antibody detected several clusters of gangliosides on the plasma membranes separate from caveolae; gangliosides located inside the caveolae could not be detected. Scanning confocal microscopy of cells immunostained with anti-GM3 and anti-CAV1 Ig showed only a very small overlap with the CAV1 and GM3 signals. Thus, the biochemical and microscopic studies suggest that caveolae contain at most a low level of gangliosides and are separate from the GM3 ganglioside enriched domains.  相似文献

A case of chronic lymphatic leukemia (CLL) is described in which IgD and IgA are the copredominant membrane immunoglobulins. Since CLL represents malignant proliferations of B lymphocytes arrested at discrete points during maturation, the findings in this case suggest that at least some of the developing cells destined to synthesize IgA for secretion pass through a stage in which immunoglobulins D and A are present together on the cell membrane.  相似文献

Human fibroblast cell lines were pulse-treated for 1 h with either methylnitrosourea (MNU) or ethylnitrosourea (ENU) at various time intervals before harvesting for chromosome analysis. Cells treated with 1 X 10(-3) M, 5 X 10(-4) M, and 1 X 10(-4) M final concentrations of MNU and ENU during the G2 or M phases of the cell cycle showed a significant increase in chromatid-type abnormalities over controls. Cells exposed to MNU or ENU 23 h before harvest showed some chromosome-type abnormalities, reflecting probable damage induced during the G1 phase of the cell cycle or derived from chromatid damage induced during the previous cell cycle. The mitotic indices and incidences of abnormalities suggested a dose response effect when cells were treated with the two higher concentrations and the three concentrations, respectively, of MNU or ENU. Chromatid abnormalities were observed in MUN and ENU-treated cells from each of four cell lines. From this investigation, it was concluded that MNU and ENU treatment of human diploid cell lines in vitro induced both chromatid and chromosome aberrations. MNU and ENU, both of which had previously been shown to be mutagenic in experimental animals, are, therefore, also considered to be mutagenic at the chromosome level in human fibroblasts grown and treated in cell culture.  相似文献

Using a bioassay-directed fractionation of Pycnocycla spinosa Decne. Ex Boiss. var. spinosa, a new polycyclic diterpenoid, 3,7,10,14,15-pentaacetyl-5-butanoyl-13,17-epoxy-8-myrsinene (1), two known compounds; vanillin (2) and isoacetovanilon (3), and a new phenolic compound, 6-(4-hydroxy-3-methoxyphenyl)-hexanoic acid (4) with inhibitory effects on KCl-induced smooth muscle contractions on the rat isolated ileum were obtained. Compound 1, the most active of the series, which exerted potent antispasmodic activity with IC50 value of IC50 = 0.062 ± 0.011 μM, is likely the main active ingredient of the extract. The structures of the isolated compounds were established based on 13C and 1H NMR as well as 2D NMR, IR, HR-MS, and X-ray crystallographic methods.  相似文献

The rate of electron transfer through Photosystem I (reduced 2,6-dichlorophenol indophenol (DCIPH2 → methylviologen) in a low-salt thylakoid suspension is inhibited by Mg2+ both under light-limited and the light-saturated conditions, the magnitude of inhibition being the same. The 2,6-dichlorophenol indophenol (DCIP) concentration dependence of the light-saturated rate in the presence and in the absence of Mg2+ shows that the overall rate constant of the photoreaction is not altered by Mg2+. With N,N,N′,N′-tetramethyl-p-phenylenediamine or 2,3,5,6-tetramethylphenylenediamine as electron donor only the light-limited rate, not the light-saturated rate, is inhibited by Mg2+ and the magnitude of inhibition is the same as with DCIP as donor. The results are interpreted in terms of heterogeneous Photosystem I, consisting of two types, PS I-A and PS I-B, where PS I-A is involved in cation-regulation of excitation energy distribution and becomes unavailable for DCIPH2 → methyl viologen photoelectron transfer in the presence of Mg2+.  相似文献

Eleven years have passed since the start of the first trial of dendritic cell (DC) vaccination for melanoma. A review of 54 trials was performed to evaluate the relationship between clinical effects and vaccine parameters. Significant differences were found between use of immature and mature DCs with regard to progressive disease (PD), between stage III and IV for clinical response, between use and non-use of adjuvants with regard to stable disease (SD) in treatment with tumor/tumor lysate-pulsed DCs, between positive and negative delayed-type hypersensitivity (DTH) for PD, and between increased and unchanged interferon (IFN)-γ-secreting T cells for clinical response. These results are consistent with the partial efficacy of vaccination with mature DCs in early stage melanoma and the partial correlation of efficacy with positive DTH and increased IFN-γ-secreting T cells. DC vaccination alone had a limited clinical effect and a modified regimen is needed to enhance antigen-specific cytotoxic T cells and decrease immunosuppression.  相似文献

Vitamin E and carotenoids are known to act as antioxidants both in vitro and in vivo. In this review we present a series of studies in healthy subjects and in patients who exhibit either acute or chronic oxidative stress. In the EU-Commission funded VITAGE project we investigated the status and effects of vitamin E and carotenoids on oxidative stress in 300 healthy volunteers. Depletion studies limiting dietary vitamin E or carotenoid intake to 25% of the dietary reference intakes and subsequent repletion by supplementation with either large doses of vitamin E or intermediate doses of carotenoids showed significant changes in ex vivo LDL oxidizability, total plasma peroxide concentrations and urinary 8-oxo-7,8-dihydro-2-deoxyguanosine excretion. Patients on chronic hemodialysis present with oxidative stress in the presence of normal vitamin E but impaired vitamin C status and, due to anemia, need to be treated with parenteral iron. We studied the effects of a single oral dose of vitamin E taken 6 h prior to intravenous infusion of 100 mg iron, which exceeded the iron-binding capacity of transferrin. Vitamin E significantly reduced and in combination with a single dose of vitamin C completely abrogated acute oxidative stress induced by the iron load. Patients with cystic fibrosis are exposed to chronic oxidative stress due to an overproduction of reactive oxygen species as a result of neutrophil-dominated lung inflammation and impaired antioxidant status. Biochemical vitamin E and carotenoid deficiencies could be fully corrected even in the presence of fat malabsorption using intermediate doses of either RRR -tocopherol or all-rac -tocopheryl acetate and water-miscible all-trans β-carotene. Long-term supplementation reduced ex vivo LDL oxidizability, in vivo lipid peroxidation and lung inflammation.  相似文献

The binding of human chorionic gonadotropin and human luteinizing hormone to particulate receptors of rat testes has generally been assumed to follow an equilibrium model similar to that proposed for many enzyme systems. Our work shows that equilibrium dissociation constant (Kd) and number of hormone binding sites (Bmax) are highly sensitive to changes in hormone and/ or receptor concentration and to treatment received by tissue or receptor preparation prior to the assay. The results of binding assays obtained using receptor preparation pretreated with hormone (labeled as well as unlabeled) indicated that the binding reaction between hormone and receptor was irreversible and that pretreatment of the tissue with hormone greatly alters the number of high affinity gonadotropin binding sites in the testicular homogenate. Data from studies involving increasing receptor concentrations revealed that increasing the mass of particulate receptors in the binding assays leads to higher Kd as well as Bmax values. These findings are incompatible with a binding model based upon occupancy of receptor sites and the state of equilibrium implied. 为明确香蕉根系分泌物对枯萎病菌及其生防枯草芽孢杆菌的生物效应,采用离位溶液培养法收集抗枯萎病香蕉品种(南天黄)和感枯萎病香蕉品种(桂蕉6号)的根系分泌物,研究根系分泌物对土壤微生物种群数量、香蕉枯萎病菌和枯草芽孢杆菌生长的影响。结果表明: 抗病品种根系分泌物能显著减少土壤真菌的数量,抑制枯萎病菌孢子的萌发;而感病品种根系分泌物能显著促进病菌菌丝生长和孢子的萌发,两个品种根系分泌物均能显著促进枯草芽孢杆菌的生长和生物膜形成。经抗(感)病香蕉品种根系分泌物处理,病菌菌丝生长速率分别为11.28和12.28 mm·d-1,病菌孢子的萌发率分别为34.6%和79.5%;枯草芽孢杆菌培养12 h后菌体生长量的OD600分别为1.27和1.14,生物膜形成量在静置培养72 h后OD570分别达1.11和1.30,两个品种处理间的差异均达显著水平。枯草芽孢杆菌在香蕉感病品种根际中定殖的菌量显著高于抗病品种。通过对两个品种根系分泌物中可溶性总糖、游离氨基酸和有机酸的含量和组成分析,发现抗病品种根系分泌物中有机酸和游离氨基酸的含量明显高于感病品种,在各组成成分中,以乙酸和脯氨酸在抗(感)病香蕉品种根系分泌物中含量比值较高,分别达3.7和2.4倍。综上所述,抗病品种根系分泌物能抑制病菌生长,感病品种根系分泌物则会显著促进病菌生长,而两个品种根系分泌物均能显著促进枯草芽孢杆菌的生长和生物膜的形成。  相似文献

新疆焉耆盆地人类活动与气候变化的效应机制   总被引:2,自引:0,他引:2  
通过对新疆焉耆盆地及其周边近40a(1973—2014)的气候变化趋势检测、LUCC和生物量估算,探讨气候变化和人类活动的生态效应机制,研究区域陆地生态系统演变及其归因。分析结果表明:(1)焉耆盆地山区和平原区降水变化都有明显的突变点,并呈现增加趋势,蒸发量在山区减少,在平原区波动性减少趋势;(2)LUCC分析表明,山区裸地面积减少5.40%,冰川面积减少3.36%,高地植被面积增加8.76%;同时平原区天然绿洲面积增加1.96%,沙漠面积减少1.62%,水域面积减少1.30%,人工绿洲面积增加15.41%,湿地面积增加1.27%;(3)山区陆地生态系统对区域气候变化非常敏感,其中降水变化是决定山区地表植被生存状态和分布的重要因素;(4)人类活动的推动作用和有益气候变化的支撑是绿洲平原区生态系统好转的原因,其中人口急剧增加和社会经济快速发展,导致绿洲平原区生态系统结构及其时空分布的主要因素。焉耆盆地及其周围区域陆地生态系统的演变对气候变化和人类活动有明显的时空尺度效应,其反应程度各不相同。  相似文献

When the objective is to administer the best of two treatments to an individual, it is necessary to know his or her individual treatment effects (ITEs) and the correlation between the potential responses (PRs) and under treatments 1 and 0. Data that are generated in a parallel-group design RCT does not allow the ITE to be determined because only two samples from the marginal distributions of these PRs are observed and not the corresponding joint distribution. This is due to the “fundamental problem of causal inference.” Here, we present a counterfactual approach for estimating the joint distribution of two normally distributed responses to two treatments. This joint distribution of the PRs and can be estimated by assuming a bivariate normal distribution for the PRs and by using a normally distributed baseline biomarker functionally related to the sum . Such a functional relationship is plausible since a biomarker and the sum encode for the same information in an RCT, namely the variation between subjects. The estimation of the joint trivariate distribution is subjected to some constraints. These constraints can be framed in the context of linear regressions with regard to the proportions of variances in the responses explained and with regard to the residual variation. This presents new insights on the presence of treatment–biomarker interactions. We applied our approach to example data on exercise and heart rate and extended the approach to survival data.  相似文献

森林碳汇空间相关性与溢出效应对林业产业的区域统筹发展具有重要作用,科学核算各地区的森林碳汇量并分析其空间关联特征是制定差异化碳汇发展政策的重要基础。以森林蓄积量扩展法核算我国31个省(市、自治区)1993—2018年6次森林资源清查期间的森林碳汇量,探究省域间森林碳汇量的相关性特征,并利用空间计量模型分析森林碳汇的外溢效应和影响因素。结果表明:(1)我国整体森林碳汇量不断增加,不同地区的森林碳汇量差别较大。西南省份和东北林区森林碳汇量处于第一梯队,上海和北京碳汇增速较快。(2)研究期间内的Moran′s I指数先呈现倒“V”形的变化特征,之后又以较为稳定的趋势上升,我国各地区的森林碳汇分布存在显著的空间关联性。(3)森林碳汇的空间外溢效应显著,根据空间杜宾模型将外溢效应分解为直接效应、间接效应和总效应。林业管理水平和森林蓄积水平对本地和相邻地区森林碳汇量有正向影响,林业产业发展水平对本地区的森林碳汇量有负向影响。综上,我国各地方政府对差异化林业碳汇政策的制定和执行应兼顾区域因素,以我国林业政策的总体空间规划来综合统筹各区域森林政策,在“山水林田湖草沙”的命运共同体理念引领下,实现林业的...  相似文献
