Our main goals were to identify diagnostic characters at the species, genus, and subfamily levels, find anatomical features with potential for future morphological and molecular (combined) phylogenetic analyses, and to reconstruct the evolution of wood anatomical characters in two subfamilies of Primulaceae in a molecular phylogenetic framework. We investigated twenty-seven species from the woody Myrsinoideae (4 genera) and Theophrastoideae (2 genera) using scanning electron, light, and epifluorescence microscopy. Samples were prepared using standard protocols. Based on the wood anatomical characters, we were able to identify synapomorphies and to detect evolutionary trends of interest for the genera and subfamilies. Both subfamilies share the presence of diffuse porosity, simple perforation plates, septate fibres, and scanty paratracheal axial parenchyma. Theophrastoideae species have rays?>?10 cells wide and short (<?350 µm) vessel elements, and Myrsinoideae have breakdown areas in rays and longer vessel elements. Ardisia and Stylogyne have scalariform intervessel pits, Myrsine exhibit breakdown areas in rays, and two Cybianthus species from subgenus Weilgetia have distinguishing features (e.g., scalariform perforation plate in C. nemoralis and the absence of rays in C. densiflorus). Overall, when combining characters, we were able to segregate the Neotropical Primulaceae subfamilies and genera from each other and from the subfamily Maesoideae based on wood anatomy.  相似文献

A new species, Ulmus priamurica sp. nov. (Ulmaceae), from the deposits of the Sazanka Formation (upper Middle–Upper Miocene) of the Erkovetskii Brown Coal Field (Amur Region, Russia) is described based on anatomical features of fossil wood. The new species shows some wood anatomical characters of the extant elms U. japonica and U. americana. Fossil wood of Ulmus was found in the Amur Region for the first time.  相似文献

New species of the pine fossil wood, Pinus priamurensis sp. nov. (Pinaceae) from the Sazanka Formation (upper Middle Miocene–Upper Miocene) of the Erkovetskii Brown Coal Field (Amur Region) is described. The new species shares some wood anatomical features with modern species of the subsection Pinus (section Pinus, subgenus Pinus). A new species, Betula erkovetskiensis Blokhina et O.V. Bondarenko (Betulaceae), from the deposits of the Sazanka Formation (upper Middle?Upper Miocene) of the Erkovetskii Brown Coal Field (Amur Region, Russia) is described based on anatomical features of fossil wood. The new species shows some wood anatomical characters of the extant birch subgenus Betula, B. davurica, B. nigra (section Dahuricae), and B. papyrifera (section Betula). Chusquea is a diverse but monophyletic genus of Neotropical woody bamboos from primarily montane forests that comprises four well-supported lineages: subg. Magnifoliae, subg. Platonia, subg. Rettbergia, and the Euchusquea clade (comprising subg. Swallenochloa and subg. Chusquea). However, the relationships among clades or taxa within the Euchusquea clade inferred from molecular data are mostly not congruent with those inferred from morphological evidence, consequently limiting our ability to understand species relationships. In this study we generated foliar micromorphological and anatomical data for the Chusquea ramosissima informal group (Chusquea ramosissima, C. tenella, and C. tenuiglumis), and for the putative new species from Bolivia in this group, in order to test the value of these types of data for defining species and to seek potential synapomorphies for this group. Our results demonstrate that epidermal features, mainly with regard to the stomatal apparatus, proved to be more valuable in distinguishing species than anatomical characters. The presence of horizontally elongated silica cells over the veins and adaxial arm cells with invaginations from the abaxial side was shared by all the studied species but is not unique to this group. The type of trichomes, shape of silica bodies, type of arm cells, and midrib structure may be useful to lesser degree. All four species exhibited intraspecific variation in development of the papillae on the long cells. Support for the recognition of the new species from Bolivia is provided by micromorphological characters. An identification key based on leaf blade features is provided for the four studied species.  相似文献

The aim of this study was to investigate aerobiological dynamics of pollen in the Mediterranean part of Croatia in the air of the city of Split. Pollen monitoring during the period from 2005 to 2013 was performed using a Hirst volumetric trap. Among the identified pollen of 50 taxa, 21 were allergenic. The average annual pollen index was 33,513. Three pollination seasons were established: early winter season dominated by tree pollen, spring–summer season dominated by herbaceous plants and summer–autumn season with lower amounts of Parietaria and Cupressaceae pollen. According to the abundance, the main taxa were: Cupressaceae, Parietaria/Urtica, Pinus, Quercus, Olea, Carpinus/Ostrya, Poaceae, Platanus and Ambrosia. The annual pollen index together with the daily maximum concentrations showed an upward trend for selected taxa during the study period. The highest monthly pollen index and the highest biodiversity were recorded in April and the lowest during the late autumn and winter months. The pollen calendar created for the city of Split confirmed Mediterranean features of the pollen spectrum. The longest pollen seasons were recorded for Cupressaceae, Parietaria/Urtica and Poaceae pollen types. The correlations between pollen concentrations and meteorological parameters were analyzed. The correlations between pollen concentrations and temperature were positive, while the humidity and the precipitation mostly showed negative influence.  相似文献

This paper describes and illustrates Heteropterys serrata, a new species endemic to semideciduous forests associated with inselbergs in the state of Espírito Santo, Brazil. Morphological and wood anatomical traits of the new species are compared to those of other species of the Heteropterys Metallophyllis informal group. Based on wood anatomy, H. serrata and H. nitida (the most morphologically similar species) have different axial parenchyma, which is scarce paratracheal in H. nitida (and some other species of the Metallophyllis informal group) and aliform confluent in H. serrata. The most notable morphological and anatomical characters that distinguish the new species are the young hexagonal stems, the unusual widely serrate leaf margins and the aliform confluent axial parenchyma in the wood.  相似文献

Garden history can be investigated through archaeobotanical research. This paper discusses the plant remains which were obtained from the soil of the historical garden of the manor of Kumpula in Helsinki, Finland. This study was an experiment to enable macrofossil analysis without archaeological excavations. The aim was to develop an alternative method for sampling for macrofossils also including radiocarbon dates, and to evaluate the usability, cost and functionality of this method. The character of the garden was also considered. The soil samples for macrofossil analysis were collected from the garden from three to eight different levels using an end-filling open-ended sampler. A total of 38 one litre soil samples from eight different pits yielded 2,036 identified macrofossils, mostly seeds. These comprised 63 different taxa, of which 26 were identified to species level with certainty. Taxa with more than 25 seeds found were Chelidonium majus, Chenopodium spp., Juncus spp., Rubus idaeus, Sambucus racemosa and Urtica dioica. Important species were Secale cereale and Hordeum vulgare. Nine AMS radiocarbon dates were obtained from macrofossil material from four different pits, giving results ranging from 1120–920 cal bc to cal ad 1680–1930 for charred wood, and from cal ad 1450–1640 to 1640–1930 for charred grains of Secale cereale and seeds of Chenopodium album. The sampling method proved to work reasonably well, considering the limitations of the sample size.  相似文献

Rinorea, the second most species-rich genus in the Violaceae, has been shown to be polyphyletic with four separate clades recovered in phylogenetic studies. Among these clades is the Rinorea crenata group, which is composed of three Neotropical species. This group has been shown in family- and genus-level molecular phylogenies to be resolved outside of a large clade representing Rinorea s.str. Based on phylogenetic, morphological, and anatomical evidence, Bribria, a new genus, is segregated from Rinorea s.str. and described, with new combinations made for its three species: Bribria apiculata, Bribria crenata, and Bribria oraria. In addition, two new sections in Rinorea s.str. are described to accommodate the remaining Neotropical species: Rinorea sect. Rinorea and Rinorea sect. Pubiflora, which correspond to Group IIa Rinorea and Group IIc Pubiflora, respectively, in W. H. A. Hekking’s monograph of Neotropical Rinorea.  相似文献

The phylum Neocallimastigomycota contains eight genera (about 20 species) of strictly anaerobic fungi. The evolutionary relationships of these genera are uncertain due to insufficient sequence data to infer their phylogenies. Based on morphology and molecular phylogeny, thirteen isolates obtained from yak faeces and rumen digesta in China were assigned to Neocallimastix frontalis (nine isolates), Orpinomyces joyonii (two isolates) and Caecomyces sp. (two isolates), respectively. The phylogenetic relationships of the eight genera were evaluated using complete ITS and partial LSU sequences, compared to the ITS1 region which has been widely used in this phylum in the past. Five monophyletic lineages corresponding to six of the eight genera were statistically supported. Isolates of Caecomyces and Cyllamyces were present in a single lineage and could not be separated properly. Members of Neocallimastigomycota with uniflagellate zoospores represented by Piromyces were polyphyletic. The Piromyces-like genus Oontomyces was consistently closely related to the traditional Anaeromyces, and separated the latter genus into two clades. The phylogenetic position of the Piromyces-like genus Buwchfawromyces remained unresolved. Orpinomyces and Neocallimastix, sharing polyflagellate zoospores, were supported as sister genera in the LSU phylogeny. Apparently ITS, specifically ITS1 alone, is not a good marker to resolve the generic affinities of the studied fungi. The LSU sequences are easier to align and appear to work well to resolve generic relationships. This study provides a comparative phylogenetic revision of Neocallimastigomycota isolates known from culture and sequence data.  相似文献

Salvia subg. Calosphace (Lamiaceae, Lamiales) is a highly diverse clade endemic to the New World. The phylogenetic relationships of Calosphace have been previously investigated using DNA sequences of nuclear ITS region and plastid psbA–trnH intergenic spacer, but the resulting trees lack resolution and support for many clades. The present paper reassesses the phylogenetic relationships of subgenus Calosphace, including a broader taxon sampling, with a special focus on representing previously unsampled sections, and using an additional plastid marker (trnL–trnF region). Our results show increased resolution and overall patterns of support, recovering ten main clades. Within core Calosphace, the most inclusive of these main clades, 17 new subclades were identified. Of the 42 sections for which more than one species was analysed, only 12 are monophyletic. Our biogeographical analysis identified at least twelve migrations to South America from Mexican and Central American lineages, in agreement with previous suggestions of multiple origins of South American Calosphace diversity. This analysis also confirmed a colonization of the Antilles by Andean lineages. The reconstruction of ancestral states of pollination syndromes showed multiple shifts to ornithophily from melittophily and one reversal to the latter.  相似文献

Studies of phylogenetic relationships among cypresses of the Old World (Cupressus; Cupressaceae) have been plagued by unresolved relationships, poor branch support, and conflict between data sets and methods of analysis. In this study, we combined 5.4 kb of aligned DNA sequence and 157 binary characters with previously published data in examining phylogenetic relationships among Cupressus species. Bayesian and parsimony analysis of the combined data or of the nuclear data alone always recovered three principal clades of Cupressus; however, tests of phylogenetic incongruence could not distinguish between competing relationships among the three principal Cupressus lineages. In contrast, incongruence tests often found statistically significant conflict between the nuclear and plastid data, particularly with respect to the placement of C. chengiana. Consistent with previous studies and prevailing taxonomic opinion, we find C. darjeelingensis more closely related to cypresses of the New World (Hesperocyparis). In contrast, we placed accessions of C. assamica and C. tonkinensis, two putatively Old World species suggested to be misidentified New World taxa by some authors, within well-supported Old World clades. Statistical analysis of genetic distances suggests instances in which taxa recognized as distinct species by some authors are identical or nearly so and may best be considered a single taxon. Conversely, we identify instances in which infraspecific taxa are more distantly related to one another than those traditionally recognized as distinct species. Factors confounding cypress taxonomies, including poor morphological differentiation, misidentification, and the use of accessions of questionable provenance, are discussed.  相似文献

Plastid DNA markers sequencing and DNA fingerprinting approaches were used and compared for resolving molecular phylogeny of closely related, previously unexplored Amorphophallus species of India. The utility of individual plastid markers namely rbcL, matK, trnHpsbA, trnLCtrnLD, their combined dataset and two fingerprinting techniques viz. RAPD and ISSR were tested for their efficacy to resolves Amorphophallus species into three sections specific clades namely Rhaphiophallus, Conophallus and Amorphophallus. In the present study, sequences of these four plastid DNA regions as well as RAPD and ISSR profiles of 16 Amorphophallus species together with six varieties of two species were generated and analyzed. Maximum likelihood and Bayesian Inference based construction of phylogenetic trees indicated that among the four plastid DNA regions tested individually and their combined dataset, rbcL was found best suited for resolving closely related Amorphophallus species into section specific clades. When analyzed individually, rbcL exhibited better discrimination ability than matK, trnHpsbA, trnLCtrnLD and combination of all four tested plastid markers. Among two fingerprinting techniques used, the resolution of Amorphophallus species using RAPD was better than ISSR and combination of RAPD +ISSR and in congruence with resolution based on rbcL.  相似文献

The phylogeny of phagotrophic euglenids is widely based on nuclear small subunit ribosomal DNA (18S rDNA) sequence data, but most analyses suffer from weakness in statistical support regarding the “connecting backbone” between monophyletic clades. Moreover, the position of Entosiphon has remained unclear. Testing the 18S rDNA capability for a phylogeny of phagotrophic euglenids, we isolated sequences of Peranema sp. and Ploeotia edaphica and utilized secondary structure data as a prerequisite for recognition of homologous positions. We found a unique, clade-specific nucleotide substitution in the deduced 18S rDNA helix 44. Since our 18S rDNA phylogenies could only in part resolve positions of phagotrophic lineages, but did not verify that of Entosiphon, we investigated the phagotrophic key taxa Peranema trichophorum, Petalomonas cantuscygni, Ploeotia costata, and Entosiphon sulcatum ultrastructurally. Additionally, we explored the presence or absence of the euglenid reserve carbohydrate paramylon by specific staining with monoclonal anti-β-1,3-glucan antibodies. Paramylon was found to be clearly present in P. trichophorum and E. sulcatum, but was absent in Pt. cantuscygni and Pl. costata. Combined results of our molecular, ultrastructural, and immunocytochemical investigations suggest that Entosiphon sulcatum is the sister taxon of a monophyletic euglenid crown clade, characterized by a helical pellicle, which we propose to rename. This phylogenetic affiliation is confirmed by a clade-specific primary absence of the unique nucleotide substitution in helix 44 and by the common presence of paramylon.  相似文献

Nothofagus obliqua, N. dombeyi, N. alpina and N. antarctica are characteristic tree species of the temperate forests on the western slopes of the Andes with centres of distribution that differ in their temperature and moisture regimes. We tested branch wood from co-occurring specimens of these species for the inherent differences in xylem anatomy and theoretical hydraulic conductance to evaluate their resistance to drought or frost. The hydraulic conductivity of the xylem was calculated using a modified Hagen–Poiseuille equation and related to wood density. Conduit dimensions were used to predict the water potential that would cause 50 % loss of hydraulic conductivity (Ψ 50). Nothofagus alpina, which mainly grows at sites with low frost frequency, exhibited the largest conduits and the highest mean values for conduit area, fraction of conduit area in the cross-section and hydraulic conductivity, but the lowest wood density. Opposite relationships were found in the plastic N. antarctica, whose xylem seems to be least vulnerable to freezing-induced, but also to drought-induced embolism. Calculated Ψ 50 was highest (least negative) in N. alpina, indicating a relatively high susceptibility to cavitation. The xylem of the thermophilic N. obliqua and of N. dombeyi, which mainly occurs under oceanic climate, but can also survive at sporadically dry and warm sites, is not particularly adapted to periods of drought stress. Across all species, wood density was negatively correlated with the calculated hydraulic conductance. The xylem traits of N. alpina might contribute to its relatively high growth rate and facilitate its spread into forest gaps.  相似文献

We did a phylogenetic analysis on 21 of the 23 species of Stigmatopteris, a neotropical montane genus of wet forest floors. The analysis was based on four plastid markers: rbcL, rps4-trnS, trnG-trnR, and trnL-trnF. Two of the most closely related families to the Dryopteridaceae (Didymochlaenaceae and Hypodematiaceae) and 12 dryopteroid genera were used as outgroups. Eight morphological characters were mapped on the resulting tree. Stigmatopteris was recovered as monophyletic and sister to a clade formed by polystichoid ferns such as Arachniodes, Dryopteris, and Polystichum. Synapomorphies that distinguish Stigmatopteris are 1-pinnate-pinnatifid laminae, visible punctate glands in the mesophyll, the presence of hydathodes, uniseriate cilia on the scale margins, and loss of indusia. Within the genus are two main clades. The first consists of four species characterized by pinnae long-decurrent in at least the distal third of the lamina. The second consists of the remaining 17 species in the analysis and has no known morphological synapomorphies. Nested within this second clade is a subclade of six species endemic to southeastern Brazil. Anastomosing veins, a character often used in keys to distinguish the species, evolved three times within the genus.  相似文献

The neotropical plant genus Drymonia displays a remarkable variety of floral shapes and colors. One feature that is particularly important to coevolution with pollinators involves the variable shapes and widths of corolla tubes. To evaluate the evolutionary context for changes in corolla shape, we constructed a phylogeny of 50 of the 75 species of Drymonia using molecular markers from plastid (trnK-matK) and nuclear regions (ITS and ETS). Mapping tube shapes on the phylogeny supports open, bell-shaped (campanulate) corolla shape as the ancestral character state for Drymonia, with multiple independent origins of constriction in the corolla tube. Corollas with constrictions take one of three tube shapes: a constricted flower opening and throat with a large, expanded pouch on the lower surface (hypocyrtoid); a constricted flower opening and throat lacking an expanded pouch on the lower surface (urceolate); or a constricted opening and throat where the sides of the corolla appear laterally compressed. Fieldwork demonstrates euglossine bees (mostly Euglossa spp. and Epicharis spp.) visit campanulate corollas while hummingbirds visit corollas that are constricted. Results support eight independent origins of constricted corolla tubes from ancestors with campanulate corolla tubes: 3 hypocyrtoid clades, 3 laterally compressed clades, and 3 urceolate clades (one of which represents a shift from a hypocyrtoid ancestor). Constricted corollas are associated with shifts from the ancestral condition of poricidal anther dehiscence, which presents pollen to pollinators in multiple small doses, to the derived condition of longitudinal anther dehiscence, which presents all pollen to pollinators simultaneously. The association of hummingbird pollination with constricted corolla tubes suggests that narrowing evolved as a barrier mechanism that prohibits the visitation of flowers by bees.  相似文献
