An axenic culture of a cyanobacterium (strain HPS) was raised from a field specimen of greenish planktonic floccose mass from a local lake, Ganga Sagar. Intense morphological examination and comparative morphological assessment with the genera Fischerella and Hapalosiphon and all the known strains of the genus Westiellopsis indicated that the strain HPS differed in morphology with the closely related strains in trichome arrangement, size of vegetative cells, heterocytes, monocytes, and number of rows of main filaments. There were also differences in the habitat preference, being aquatic rather than terrestrial or subaerial. Intense ecological characterization of the habitat was performed through measurements of important physicochemical characteristics. 16S rRNA gene-based identification and phylogenetic placement indicated conclusively that the strain HPS was different from the most closely related strain Westiellopsis prolifica SAG 16.93. Phylogenetic inferences drawn in between all the branched heterocytous forms and subsequent 16S-23S ITS analyses and folding of the secondary structures revealed an entirely new form that is unknown till now. Subsequent nifD and rbcL gene-based phylogenetic assessments indicated that strain HPS is phylogenetically different from all the other previously known species of true branching cyanobacteria, along with also pointing toward the huge database inconsistencies in case of true branched cyanobacteria. Assessment of morphological and ecological differences along with comprehensive phylogenetic evaluation indicated that the strain HPS is a new species of the genus Westiellopsis and the name being proposed is Westiellopsis ramosa sp. nov.  相似文献

The similar geographic distributions of an extinct (Dasypus bellus) and an extant (D. novemcinctus) armadillo species have long been of interest to scholars because of the unresolved phylogeny. The relationship between the two species has been investigated through morphological and phylogenetic studies, whereas the ecological perspective has been overlooked, the importance of which is more and more acknowledged in speciation events. Here, we used ecological niche models to study the climatic niche similarity of three species of Dasypus (D. bellus, D. novemcinctus, and D. kappleri) and provide new insights on the relationship among them. The climatic niche similarity was compared in two ways: hindcast of ecological niche models based on occurrences and climatic layers, and direct niche boundary comparison along bioclimatic axes. The fossil records of D. bellus were not predicted suitable by the ecological niche models of the two extant armadillos. The direct comparison of niche boundary showed that D. bellus lived in colder and relative dryer climates, with high temperature variation and low precipitation variation. Our results did not support the previously assumed ecological similarity of D. bellus and D. novemcinctus based on their geographic distributions and emphasized the possibility of a cold adapted characteristic of the life history of D. bellus.  相似文献

A Gram-stain-negative, facultative aerobic, non-flagellated, and rod-shaped bacterium, designated AR-13T, was isolated from a seawater on the East Sea in South Korea, and subjected to a polyphasic taxonomic study. Strain AR-13T grew optimally at 30°C, at pH 7.0–8.0 and in the presence of 0–0.5% (w/v) NaCl. The phylogenetic trees based on 16S rRNA gene sequences showed that strain AR-13T fell within the clade comprising the type strains of Arcobacter species, clustering coherently with the type strain of Arcobacter venerupis. Strain AR-13T exhibited 16S rRNA gene sequence similarity values of 98.1% to the type strain of A. venerupis and of 93.2–96.9% to the type strains of the other Arcobacter species. Strain AR-13T contained MK-6 as the only menaquinone and summed feature 3 (C16:1ω7c and/or C16:1ω6c), C16:0, C18:1ω7c, and summed feature 2 (iso-C16:1 I and/or C14:0 3-OH) as the major fatty acids. The major polar lipids detected in strain AR-13T were phosphatidylethanolamine, phosphatidylglycerol, and one unidentified aminophospholipid. The DNA G+C content was 28.3 mol% and its mean DNA-DNA relatedness value with the type strain of A. venerupis was 21%. Differential phenotypic properties, together with its phylogenetic and genetic distinctiveness, revealed that strain AR-13T is separated from recognized Arcobacter species. On the basis of the data presented, strain AR-13T is considered to represent a novel species of the genus Arcobacter, for which the name Arcobacter acticola sp. nov. is proposed. Strain ZZ-8T, a Gram-negative, aerobic, non-spore-forming, non-motile, yellow-pigmented, rod-shaped bacterium, was isolated from metolachlor-contaminated soil in China. The taxonomic position was investigated using a polyphasic approach. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain ZZ-8T is a member of the genus Flavobacterium and shows high sequence similarity to Flavobacterium humicola UCM-46T (97.2%) and Flavobacterium pedocola UCM-R36T (97.1%), and lower (<?97%) sequence similarity to other known Flavobacterium species. Chemotaxonomic analysis revealed that strain ZZ-8T possessed MK-6 as the major respiratory quinone; and iso-C15:0 (28.5%), summed feature 9 (iso-C17:1 w9c/C16:0 10-methyl, 22.9%), iso-C17:0 3-OH (17.0%), iso-C15:0 3-OH (8.9%), iso-C15:1 G (8.6%) and summed feature 3 (C16:1 w7c/C16:1 w6c, 5.7%) as the predominant fatty acids. The polar lipids of strain ZZ-8T were determined to be lipids, a glycolipid, aminolipids and phosphatidylethanolamine. Strain ZZ-8T showed low DNA–DNA relatedness with F. pedocola UCM-R36T (43.23?±?4.1%) and F. humicola UCM-46T (29.17?±?3.8%). The DNA G+C content was 43.3 mol%. Based on the phylogenetic and phenotypic characteristics, chemotaxonomic data and DNA–DNA hybridization, strain ZZ-8T is considered a novel species of the genus Flavobacterium, for which the name Flavobacterium zaozhuangense sp. nov. Using yeast genome databases and literature data, phylogenetic analysis of pectinase PGU genes from 112 Saccharomyces strains assigned to the biological species S. arboricola, S. bayanus (var. uvarum), S. cariocanus, S. cerevisiae, S. kudriavzevii, S. mikatae, S. paradoxus, and the hybrid taxon S. pastorianus (syn. S. carlsbergensis) was carried out. A superfamily of divergent PGU genes was found. Natural interspecies transfer of the PGU gene both from S. cerevisiae to S. bayanus and from S. paradoxus to S. cerevisiae may, however, occur. Within the Saccharomyces species, identity of the PGU nucleotide sequences was 98.8–100% for S. cerevisiae, 86.1–95.7% for S. bayanus (var. uvarum), 94–98.3% for S. kudriavzevii, and 96.8–100% for S. paradoxus/S. cariocanus. For the first time, a family of polymeric PGU1b, PGU2b, PGU3b and PGU4b genes is documented for the yeast S. bayanus var. uvarum, a variety important for winemaking.  相似文献

A heterotrophic bacterial strain AGD 8-3 capable of denitrification under extreme haloalkaline conditions was isolated from soda solonchak soils of the Kulunda steppe (Russia). The strain was classified within the genus Halomonas. According to the results of 16S rRNA gene sequencing, Halomonas axialensis, H. meridiana, and H. aquamarina are most closely related to strain AGD 8-3 (96.6% similarity). Similar to other members of the genus, the strain can grow within a wide range of salinity and pH. The strain was found to be capable of aerobic reduction of chromate and selenite on mineral media at 160 g/l salinity and pH 9.5–10. The relatively low level of phylogenetic similarity and the phenotypic characteristics supported classification of strain AGD 8-3 as a new species Halomonas chromatireducens.  相似文献

The taxonomic position of a new Saccharothrix strain, designated MB46T, isolated from a Saharan soil sample collected in Mzab region (Ghardaïa province, South Algeria) was established following a polyphasic approach. The novel microorganism has morphological and chemical characteristics typical of the members of the genus Saccharothrix and formed a phyletic line at the periphery of the Saccharothrix espanaensis subcluster in the 16S rRNA gene dendrograms. Results of the 16S rRNA gene sequence comparisons revealed that strain MB46T shares high degrees of similarity with S. espanaensis DSM 44229T (99.2%), Saccharothrix variisporea DSM 43911T (98.7%) and Saccharothrix texasensis NRRL B-16134T (98.6%). However, the new strain exhibited only 12.5–17.5% DNA relatedness to the neighbouring Saccharothrix spp. On the basis of phenotypic characteristics, 16S rRNA gene sequence comparisons and DNA-DNA hybridizations, strain MB46T is concluded to represent a novel species of the genus Saccharothrix, for which the name Saccharothrix ghardaiensis sp. nov. (type strain MB46T = DSM 46886T = CECT 9046T) is proposed.  相似文献

Aerobic methane oxidation is a key process in the global carbon cycle that acts as a major sink of methane. In this study, we describe a novel methanotroph designated EMGL16-1 that was isolated from a freshwater lake using the floating filter culture technique. Based on a phylogenetic analysis of 16S rRNA gene sequences, the isolate was found to be closely related to the genus Methylomonas in the family Methylococcaceae of the class Gammaproteobacteria with 94.2–97.4% 16S rRNA gene similarity to Methylomonas type strains. Comparison of chemotaxonomic and physiological properties further suggested that strain EMGL16-1 was taxonomically distinct from other species in the genus Methylomonas. The isolate was versatile in utilizing nitrogen sources such as molecular nitrogen, nitrate, nitrite, urea, and ammonium. The genes coding for subunit of the particulate form methane monooxygenase (pmoA), soluble methane monooxygenase (mmoX), and methanol dehydrogenase (mxaF) were detected in strain EMGL16-1. Phylogenetic analysis of mmoX indicated that mmoX of strain EMGL16-1 is distinct from those of other strains in the genus Methylomonas. This isolate probably represents a novel species in the genus. Our study provides new insights into the diversity of species in the genus Methylomonas and their environmental adaptations.  相似文献

A novel strain, alga-05, of alkaliphilic purple nonsulfur bacteria was isolated from sediments of a small saline (60 g/l) soda lake near Lake Algin (Barguzin Valley, Buryat Republic, Russia). These bacteria contain bacteriochlorophyll a and carotenoids of the alternative spirilloxanthin group with predominating demethylspheroidenone. They are facultative anaerobes; their photosynthetic structures are of the vesicular type and arranged along the cell periphery. Growth of this strain is possible in a salinity range of 5–80 g/l NaCl, with an optimum at 20 g/l NaCl. Best growth occurred at 20–35°C. Analysis of the 16S rRNA gene sequences demonstrated that the studied isolate is closely related to the alkaliphilic purple nonsulfur bacterium Rhodobaca bogoriensis (99% similarity) isolated from soda lakes of the African Rift Zone. According to the results of DNA-DNA hybridization, strain alga-05 has a 52% similarity with the type species of the genus Rhodobaca. On the basis of the obtained genotypic data and some phenotypic properties (dwelling in a hypersaline soda lake of Siberia, moderate halophily, ability to grow at relatively low temperatures, etc.), the isolated strain of purple bacteria was described as a new species of the genus Rhodobaca, Rca. barguzinensis sp. nov.  相似文献

Agarum clathratum, brown algae, play important ecological roles in marine ecosystem, but can cause secondary environment pollution when they pile up on the beach. In order to resolve the environment problem by A. clathratum, we focus to isolate and identify Penicillium because many species are well known to produce extracellular enzymes. A total of 32 Penicillium strains were isolated from A. clathratum samples that collected from 13 sites along the mid-east coast of Korea in summer. They were identified based on morphological characters and phylogenetic analysis using β-tubulin DNA sequences as well as a combined dataset of β-tubulin and calmodulin. A total of 32 strains were isolated and they were identified to 13 Penicillium species. The commonly isolated species were Penicillium citrinum, P. roseomaculatum, and Penicillium sp. Among 13 Penicillium species, four species – P. bilaiae, P. cremeogriseum, P. madriti, and P. roseomaculatum – have not been previously recorded in Korea. For these four new species records to Korea, we provide morphological characteristics of each strain.  相似文献

A bacterial isolate was recovered from a soil sample collected in Jeollabuk-do Province, South Korea, and subjected to polyphasic taxonomic assessment. Cells of the isolate, designated strain S1-2-1-2-1T, were observed to be rod-shaped, pink in color, and Gram-stain negative. The strain was able to grow at temperature range from 10 to 30 °C, with an optimum of 25 °C, and growth occurred at pH 6–8. Comparative 16S rRNA gene sequence analysis showed that strain S1-2-1-2-1T belongs to the genus Hymenobacter, with closely related type strains being Hymenobacter daeguensis 16F3Y-2T (95.8% similarity), Hymenobacter rubidus DG7BT (95.8%), Hymenobacter soli PBT (95.7%), Hymenobacter terrenus MIMtkLc17T (95.6%), Hymenobacter terrae DG7AT (95.3%), and Hymenobacter saemangeumensis GSR0100T (95.2%). The genomic DNA G+C content of strain S1-2-1-2-1T was 63.0 mol%. The main polar lipid of this strain was phosphatidylethanolamine, the predominant respiratory quinone was menaquinone-7, and the major fatty acids were C15:0 iso (27.3%), summed feature 3 (C16:1 ω7c/C16:1 ω6c) (16.5%), C15:0 anteiso (15.3%), and C16:0 (14.7%), supporting the affiliation of this strain with the genus Hymenobacter. The results of this polyphasic analysis allowed for the genotypic and phenotypic differentiation of strain S1-2-1-2-1T from recognized Hymenobacter species. On the basis of its phenotypic properties, genotypic distinctiveness, and chemotaxonomic features, strain S1-2-1-2-1T is considered to represent a novel species of the genus Hymenobacter, for which the name Hymenobacter agri sp. nov. is proposed. The geographic ranges, karyotypes, ecological preferences, and mitochondrial DNA polymorphism were analyzed to reveal the origin and taxonomic position of Agrodiaetus pljushtchi and related species of A. damone species group. According to the data obtained, the common ancestor of these species originated in Asia Minor in Late Pliocene, 1.2–1.8 million years ago. In Early Pleistocene (0.8–1.2 million years ago), a representative of this group migrated into the Crimea and gave rise to A. pljushtchi. Another branch originated from the common ancestor, which migrated into Central Asia and Siberia and gave rise to a variety of partly sympatric taxa. Recently, representatives of the Central Asian — Siberian lineage occupied Eastern Europe and approached the area of A. pljushtchi but did not overlap with it. The karyotype n = 67 is an apomorphy of the A. damone species complex. However, this karyotype is not species-specific and cannot be used to differentiate taxa within the complex. The similarity of karyotypes is thus not sufficient to consider A. pljushtchi and A. damone as conspecific taxa. The natural history data support the isolated position of A. pljushtchi, and phylogenetic analysis shows that A. pljushtchi and A. damone are not closely related.  相似文献

Nicotiana sect. Repandae is a group of four allotetraploid species originating from a single allopolyploidisation event approximately 5 million years ago. Previous phylogenetic analyses support the hypothesis of N. nudicaulis as sister to the other three species. This is concordant with changes in genome size, separating those with genome downsizing (N. nudicaulis) from those with genome upsizing (N. repanda, N. nesophila, N. stocktonii). However, a recent analysis reflecting genome dynamics of different transposable element families reconstructed greater similarity between N. nudicaulis and the Revillagigedo Island taxa (N. nesophila and N. stocktonii), thereby placing N. repanda as sister to the rest of the group. This could reflect a different phylogenetic hypothesis or the unique evolutionary history of these particular elements. Here we re-examine relationships in this group and investigate genome-wide patterns in repetitive DNA, utilising high-throughput sequencing and a genome skimming approach. Repetitive DNA clusters provide support for N. nudicaulis as sister to the rest of the section, with N. repanda sister to the two Revillagigedo Island species. Clade-specific patterns in the occurrence and abundance of particular repeats confirm the original (N. nudicaulis (N. repanda (N. nesophila + N. stocktonii))) hypothesis. Furthermore, overall repeat dynamics in the island species N. nesophila and N. stocktonii confirm their similarity to N. repanda and the distinctive patterns between these three species and N. nudicaulis. Together these results suggest that broad-scale repeat dynamics do in fact reflect evolutionary history and could be predicted based on phylogenetic distance.  相似文献

During an investigation exploring potential sources of novel species and natural products, a novel actinomycete with antifungal activity, designated strain NEAU-Gz11T, was isolated from a soil sample, which was collected from Gama, Chad. The isolate was found to have morphological and chemotaxonomic characteristics typical of members of the genus Streptomyces. 16S rRNA gene sequence similarity studies showed that strain NEAU-Gz11T belongs to the genus Streptomyces with high sequence similarity to Streptomyces hiroshimensis JCM 4098T (98.0 %). Similarities to other type strains of the genus Streptomyces were lower than 98.0 %. However, the physiological and biochemical characteristics and low levels of DNA–DNA relatedness could differentiate the isolate genotypically and phenotypically from S. hiroshimensis JCM 4098T. Therefore, the strain is concluded to represent a novel species of the genus Streptomyces, for which the name Streptomyces gamaensis sp. nov. is proposed. The type strain is NEAU-Gz11T (=CGMCC 4.7304T=DSM 101531T).  相似文献

During a survey on corticolous Dothideomycetes in Mallorca, several collections with ascomata, asci, and ascospores matching the genus Stigmatodiscus (Stigmatodiscales, Dothideomycetes) were revealed, which did not fit any described species. Therefore, these collections were cultured and sequenced, and a multigene matrix of four loci (nuc18S-ITS-28S rDNA, rpb2, tef1 and tub2) was produced. Based on the results of the phylogenetic analyses of this matrix and of morphological investigations, three new species (Stigmatodiscus labiatus, S. oculatus, and S. pinicola) are described and illustrated, Asterodiscus is synonymised with Stigmatodiscus and the new combination S. tamaricis is proposed. A key to all currently known species of Stigmatodiscus is provided.  相似文献

The study of cryptic species allows to describe and to understand biodiversity, and the evolutionary processes shaping it. Mites of the family Rhinonyssidae are permanent parasites of the nasal cavities of birds, currently including about 500 described species and 12 genera. Here, we tested the hypothesis that mites from five populations of the genus Tinaminyssus—three isolated from European turtle doves (Streptopelia turtur), and two from Eurasian collared doves (Streptopelia decaocto; Aves: Columbiformes)—are, in fact, two cryptic species inhabiting different hosts. First, we performed a morphometrical study on 16 traits. Then, we used the ITS1-5.8S rDNA-ITS2 nuclear region (ITS region), and a fragment of the mitochondrial cytochrome c-oxidase 1 (COI) to carry out phylogenetic and species delimitation analyses on Tinaminyssus species. Morphological analyses revealed a lack of biometric differentiation among Tinaminyssus populations from the two host species. However, molecular analyses indicated a high degree of genetic differentiation between populations of Tinaminyssus sp. from S. turtur and S. decaocto. Overall, results show that they can be considered as different cryptic species, suggesting a case of evolutionary stasis, likely because of the anatomical similarity between closely-related bird host species.  相似文献

A novel actinomycete, designated strain YIM CF22T, was isolated from a desert soil sample collected from Turpan in Xinjiang Uyghur Autonomous Region, north-western China. The taxonomic position of the strain YIM CF22T is described based on a polyphasic approach. Strain YIM CF22T was found to form irregular sporangia on agar media. It contains meso-diaminopimelic acid in the cell wall peptidoglycan. The major menaquinone was identified as MK-9(H4); the polar lipids were identified as diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol, two unidentified phospholipids and two unidentified glycolipids. The whole cell sugars were found to be ribose, mannose, galactose, glucose and xylose. The major cellular fatty acids were found to be (>?5%) iso-C16:0 (43.5%), anteiso-C17:0 (10.2%), iso-C15:0 (7.1%), C17:1 ω8c (6.3%) and iso H-C16:1 (5.9%). The G+C content was determined to be 70.8%. 16S rRNA gene sequence analysis of strain YIM CF22T showed high similarity (97.0%) to Actinoplanes rishiriensis NBRC 108556T. The strain also showed high 16S rRNA gene sequence similarities to Verrucosispora sediminis CGMCC 4.3550T (96.9%) and Micromonospora tulbaghiae DSM 45142T (96.8%). Phylogenetic analysis based on the 16S rRNA gene sequence indicated that strain YIM CF22T clusters with A. rishiriensis NBRC 108556T, Actinoplanes globisporus JCM 3186T and Actinoplanes rhizophilus NEAU-A-2T. Based on the differential phenotypic characteristics and the results of DNA–DNA relatedness and phylogenetic analysis, it is proposed that strain YIM CF22T represents a novel species of the genus Actinoplanes, for which the name Actinoplanes deserti sp. nov. is proposed. A novel yellow pigmented, Gram-positive, aerobic and heavy metal biosorptive bacterium designated SYP-B2667T was isolated from rhizosphere soil of Epilobium hirsutum L. in Tongren, Guizhou province, China. Based on 16S rRNA gene sequence analyses, it was shown that strain SYP-B2667T represents a novel species in the genus Leucobacter, with Leucobacter chromiireducens subsp. solipictus JCM 15573T as a close phylogenetic neighbour (sequence similarity of 98.2%). Chemotaxonomic characteristics also supported the affiliation to the genus Leucobacter. Strain SYP-B2667T was determined to have a DNA G+C content of 66.6 mol%; 2,4-diaminobutyric acid in the cell wall peptidoglycan amino acids; MK-11 as predominant menaquinone; an abundance of anteiso-C15:0 and anteiso-C17:0 fatty acids; and polar lipids including diphosphatidylglycerol, phosphatidylglycerol, glycolipids and unidentified phospholipids. The DNA–DNA hybridization value between strain SYP-B2667T and L. chromiireducens subsp. solipictus JCM 15573T was 19.7?±?2.8%. Based on these phylogenetic and phenotypic results, it can be concluded that strain SYP-B2667T represents a novel species, for which the name Leucobacter epilobiisoli sp. nov. is proposed. The type strain is SYP-B2667T (=DSM 105145T=CPCC 204976T). This strain can tolerate and adsorb five heavy metals and so may have potential to facilitate heavy metal removal and bioremediation.  相似文献

The present study focused on identification and genotypic characterization of Lactic acid bacteria (LAB) in the intestine of freshwater fish. 76 strains of LAB were isolated and identified by 16S rRNA gene sequences and hsp60 gene sequences as different strains of Lactobacillus plantarum, Lactobacillus pentosus, Lactobacillus fermentum, Lactobacillus delbrueckii subsp. bulgaricus, Lactobacillus brevis, Lactobacillus reuteri, Lactobacillus salivarius, Pediococcus pentosaceus, Pediococcus acidilactici, Weissella paramesenteroides, Weissella cibaria, Enterococcus faecium, and Enterococcus durans. The hsp60 gene showed a higher level of sequence variation among the isolates examined, with lower interspecies sequence similarity providing more resolutions at the species level than the 16S rRNA gene. Phylogenetic tree derived from hsp60 gene sequences with higher bootstrap values at the nodal branches was more consistent as compared to phylogenetic tree constructed from 16S rRNA gene sequences. Closely related species L. plantarum and L. pentosus as well as species L. delbrueckii subsp. bulgaricus and L. fermentum were segregated in different cluster in hsp60 phylogenetic tree whereas such a distribution was not apparent in 16S rRNA phylogenetic tree. In silico restriction analysis revealed a high level of polymorphism within hsp60 gene sequences. Restriction pattern with enzymes AgsI and MseI in hsp60 gene sequences allowed differentiation of all the species including closely related species L. plantarum and L. pentosus, E. faecium and E. durans. In general, hsp60 gene with higher evolutionary divergence proved to be a better phylogenetic marker for the group LAB.  相似文献

An analysis of indicator values of four Saussurea DC. species, S. baicalensis (Adams) B. L. Rob., S. frolowii Ledeb., S. salicifolia (L.) DC., and S. schanginiana (Wydl.) Fisch. ex Serg., have been carried out by the method of indicator scales of soil moisture and nutrient availability and salinity in more than 1600 relevés. The size of their ecological ranges and the values of their competitiveness in phytocenoses are determined. As a result of a comparison of the locality of local populations within the ecological range and their occurrence, the vulnerability of S. baicalensis, S. frolowii, S. salicifolia, and S. schanginiana in the Kuznetsk Alatau is evaluated. The types of rarity for these species in the Kuznetsk Alatau are determined.  相似文献
