It has long been recognized that inhibition of plant water transport by either osmotic stress or salinity is mediated by aquaporins (AQPs), but the function and regulation of AQPs are highly variable among distinct isoforms and across different species. In this study, cucumber seedlings were subjected to polyethylene glycol (PEG) or NaCl stress for duration of 2 h or 24 h. The 2 h treatment with PEG or NaCl had non‐significant effect on the expression of plasma membrane AQP (CsPIPs) in roots, indicating the decrease in hydraulic conductivity of roots (Lpr) and root cells (Lprc) measured in these conditions were due to changes in AQP activity. After both 2 h and 24 h PEG or NaCl exposure, the decrease in hydraulic conductivity of leaves (Kleaf) and leaf cells (Lplc) could be attributed to a down‐regulation of the two most highly expressed isoforms, CsPIP1;2 and CsPIP2;4. In roots, both Lpr and Lprc were further reduced after 24 h PEG exposure, but partially recovered after 24 h NaCl treatment, which were consistent with changes in the expression of CsPIP genes. Overall, the results demonstrated differential responses of CsPIPs in mediating water transport of cucumber seedlings, and the regulatory mechanisms differed according to applied stresses, stress durations and specific organs.

Studies have suggested that increased root hydraulic conductivity in mycorrhizal roots could be the result of increased cell‐to‐cell water flux via aquaporins. This study aimed to elucidate if the key effect of the regulation of maize aquaporins by the arbuscular mycorrhizal (AM) symbiosis is the enhancement of root cell water transport capacity. Thus, water permeability coefficient (Pf) and cell hydraulic conductivity (Lpc) were measured in root protoplast and intact cortex cells of AM and non‐AM plants subjected or not to water stress. Results showed that cells from droughted‐AM roots maintained Pf and Lpc values of nonstressed plants, whereas in non‐AM roots, these values declined drastically as a consequence of water deficit. Interestingly, the phosphorylation status of PIP2 aquaporins increased in AM plants subjected to water deficit, and Pf values higher than 12 μm s?1 were found only in protoplasts from AM roots, revealing the higher water permeability of AM root cells. In parallel, the AM symbiosis increased stomatal conductance, net photosynthesis, and related parameters, showing a higher photosynthetic capacity in these plants. This study demonstrates a better performance of AM root cells in water transport under water deficit, which is connected to the shoot physiological performance in terms of photosynthetic capacity.

We investigated the hydraulic consequences of a major decrease in root‐to‐leaf area ratio (AR:AL) caused by nutrient amendments to 15‐year‐old Pinus taeda L. stands on sandy soil. In theory, such a reduction in AR:AL should compromise the trees’ ability to extract water from drying sand. Under equally high soil moisture, canopy stomatal conductance (GS) of fertilized trees (F) was 50% that of irrigated/fertilized trees (IF), irrigated trees (I), and untreated control trees (C). As predicted from theory, F trees also decreased their stomatal sensitivity to vapour pressure deficit by 50%. The lower GS in F was associated with 50% reduction in leaf‐specific hydraulic conductance (KL) compared with other treatments. The lower KL in F was in turn a result of a higher leaf area per sapwood area and a lower specific conductivity (conducting efficiency) of the plant and its root xylem. The root xylem of F trees was also 50% more resistant to cavitation than the other treatments. A transport model predicted that the lower AR:AL in IF trees resulted in a considerably restricted ability to extract water during drought. However, this deficiency was not exposed because irrigation minimized drought. In contrast, the lower AR:AL in F trees caused only a limited restriction in water extraction during drought owing to the more cavitation resistant root xylem in this treatment. In both fertilized treatments, approximate safety margins from predicted hydraulic failure were minimal suggesting increased vulnerability to drought‐induced dieback compared with non‐fertilized trees. However, IF trees are likely to be so affected even under a mild drought if irrigation is withheld.

Hydraulic conductance was measured on leaf and stem segments excised from sugarcane plants at different stages of development. Maximum transpiration rates and leaf water potential (ΨL) associated with maximum transpiration were also measured in intact plants as a function of plant size. Leaf specific hydraulic conductivity (Lsc) and transpiration on a unit leaf area basis (E) were maximal in plants with approximately 0.2 m2 leaf area and decreased with increasing plant size. These changes in Fand Lsc were nearly parallel, which prevented φL in larger plants from decreasing to levels associated with substantial loss in xylem conductivity caused by embolism formation. Coordination of changes in E and leaf hydraulic properties was not mediated by declining leaf water status, since φL increased with plant size. Hydraulic constrictions were present at nodes and in the node-leaf sheath-leaf blade pathway. This pattern of constrictions is in accord with the idea of plant segmentation into regions differing in water transport efficiency and would tend to confine embolisms to the relatively expendable leaves at terminal positions in the pathway, thereby preserving water transport through the stem.

In the present study, we investigated the effects of pH treatments on NaCl tolerance in mycorrhizal and non-mycorrhizal American elm. American elm (Ulmus americana) seedlings were inoculated with Hebeloma crustuliniforme, Laccaria bicolor or with both mycorrhizal fungi and subsequently subjected to different pH solutions (pH 3, 6 and 9) containing 0 mM (control) and 60 mM NaCl for 4 weeks. Inoculation with the mycorrhizal fungi did not have a large effect on seedling dry weights when the pH and NaCl treatments were considered independently. However, when the inoculated seedlings were treated with 60 mM NaCl at pH 3 or 6, shoot to root ratios and root hydraulic conductivity were higher compared with non-inoculated plants, likely reflecting changes in seedling water flow properties. At pH 6, transpiration rates were about twofold lower in non-inoculated plants treated with NaCl compared with non-treated controls. For NaCl-treated H. crustuliniforme- and L. bicolor-inoculated plants, the greatest reduction of transpiration rates was at pH 9. Treatment with 60 mM NaCl reduced leaf chlorophyll concentrations more in non-inoculated compared with inoculated plants, with the greatest, twofold, decrease occurring at pH 6. At pH 3, root Na concentrations were higher in inoculated than non-inoculated seedlings; however, there was no effect of inoculation on root Na concentrations at pH 6 and 9. Contrary to the roots, the leaves of inoculated plants had lower Na concentrations at pH 6 and 9, but not at pH 3. The results point to an interaction between ECM fungi and root zone pH for salt tolerance of American elm.

Broccoli (Brassica oleracea L. var. Italica) is a recognised health-promoting vegetable, which is moderately sensitive to salinity. In this study, the primary response of broccoli plants (cv. Marathon) to salinity has been characterised. For this, leaf water relations, nutrient composition, root hydraulic conductivity (L 0) and the effect of mercury (an aquaporin blocker) on L 0 were determined for plants grown with 0, 20, 40, 60, 80 or 100 mM NaCl for 2 weeks. During the 2 weeks of treatment, the plants showed a two-phase growth response to salinity. During the first phase (1 week), growth reduction was high, probably related to water stress as no osmotic adjustment occurred and reductions of L 0, the mercury effect and Gs were observed. After 2 weeks, the growth reduction could have resulted from internal injury caused by Na+ or Cl, since osmotic adjustment was achieved and water relations plus the mercury effect were re-established to a high degree, indicating high aquaporin functionality. The fact that aquaporin functionality fits well with the overall water relations response is very relevant, since the two-phase adaptation to salinity may imply two types of aquaporin regulation.

While investigations into shoot responses to elevated atmospheric CO2 are extensive, few studies have focused on how an elevated atmospheric CO2 environment might impact root functions such as water uptake and transport. Knowledge of functional root responses may be particularly important in ecosystems where water is limiting if predictions about global climate change are true. In this study we investigated the effect of elevated CO2 on the root hydraulic conductivity (Lp) of a C3 perennial, Larrea tridentata, and a C3 annual, Helianthus annuus. The plants were grown in a glasshouse under ambient (360 μmol mol–1) and elevated (700 μmol mol–1) CO2. The Lp through intact root systems was measured using a hydrostatic pressure-induced flow system. Leaf gas exchange was also determined for both species and leaf water potential (ψleaf) was determined in L. tridentata. The Lp of L. tridentata roots was unchanged by an elevated CO2 growth environment. Stomatal conductance (gs) and transpiration (E) decreased and photosynthetic rate (Anet) and Ψleaf increased in L. tridentata. There were no changes in biomass, leaf area, stem diameter or root : shoot (R : S) ratio for L. tridentata. In H. annuus, elevated CO2 induced a nearly two-fold decrease in root Lp. There was no effect of growth under elevated CO2 on Anet, gs, E, above- and below-ground dry mass, R : S ratio, leaf area, root length or stem diameter in this species. The results demonstrate that rising atmospheric CO2 can impact water uptake and transport in roots in a species-specific manner. Possible mechanisms for the observed decrease in root Lp in H. annuus under elevated CO2 are currently under investigation and may relate to either axial or radial components of root Lp.

The effects of an E-strain fungus (Wilcoxina mikolae var. mikolae) and an ectomycorrhizal fungus (Hebeloma crustuliniforme) on growth and water relations of balsam poplar were examined and compared in the present study. Balsam poplar roots inoculated with W. mikolae var. mikolae (Wm) exhibited structures consistent with ectendomycorrhizal (EEM) associations, including a mantle surrounding the outside of the root and an extensive Hartig net that was located between cortical cells and extended to the vascular cylinder. Roots colonized with H. crustuliniforme (Hc) developed a mantle layer, indicative of an ectomycorrhizal (ECM) association, around the outer part of the root, but no distinct Hartig net was present. Wm-colonized balsam poplar also showed increased shoot growth, stomatal conductance (g s), and root volumes compared with non-inoculated and Hc-inoculated plants. However, Hc-inoculated plants had higher root hydraulic conductivity (L pr) compared with non-inoculated plants and Wm-inoculated plants. These results suggest that L pr was not a growth-limiting factor in balsam poplar and that hyphal penetration of the root cortex in itself may have little influence on root hydraulic properties.

The importance of aquaporins for root hydraulic conductance (LP) was investigated along roots of the desert succulent Agave deserti in wet, dry and rewetted soil. Water channel activity was inferred from HgCl2‐induced reductions of LP that were reversible by 2‐mercaptoethanol. Under wet conditions, HgCl2 reduced LP for the distal root region by 50% and for the root region near the shoot base by 36% but did not affect LP for the mid‐root region. For all root regions, LP decreased by 30–60% during 10 d in drying soil and was not further reduced by HgCl2. After soil rewetting, LP increased to pre‐drying values and was again reduced by HgCl2 for the distal and the basal root regions but not the mid‐root region. For the distal region, water channels in the epidermis/exodermis made a disproportionately large contribution to radial hydraulic conductance of the intact segment; for the basal region, water channel activity was highest in the cortex and endodermis. The role of water channels was greatest in tissues in which cells were metabolically active both in the distal root region, where new apical growth occurs in wet soil, and in the basal region, which is the most likely root region to intercept light rainfall.

Understanding how root system architecture (RSA) adapts to changing nitrogen and water availability is important for improving acquisition. A sand rhizotron system was developed to study RSA in a porous substrate under tightly regulated nutrient supply. The RSA of Arabidopsis seedlings under differing nitrate (NO3) and water supplies in agar and sand was described. The hydraulic conductivity of the root environment was manipulated by using altered sand particle size and matric potentials. Ion‐selective microelectrodes were used to quantify NO3 at the surface of growing primary roots in sands of different particle sizes. Differences in RSA were observed between seedlings grown on agar and sand, and the influence of NO3 (0.1–10.0 mm ) and water on RSA was determined. Primary root length (PRL) was a function of water flux and independent of NO3. The percentage of roots with laterals correlated with water flux, whereas NO3 supply was important for basal root (BR) growth. In agar and sand, the NO3 activities at the root surface were higher than those supplied in the nutrient solution. The sand rhizotron system is a useful tool for the study of RSA, providing a porous growth environment that can be used to simulate the effects of hydraulic conductivity on growth.

Main conclusion

Changes in root and leaf hydraulic properties and stimulation of transpiration rates that were initially triggered by defoliation were accompanied by corresponding changes in leaf and root aquaporin expression. Aspen (Populus tremuloides) seedlings were subjected to defoliation treatments by removing 50, 75 % or all of the leaves. Root hydraulic conductivity (Lpr) was sharply reduced in plants defoliated for 1 day and 1 week. The decrease in L pr could not be prevented by stem girdling and it was accompanied in one-day-defoliated plants by a large decrease in the root expression of PIP1,2 aquaporin and an over twofold decrease in hydraulic conductivity of root cortical cells (L pc). Contrary to L pr and L pc, 50 and 75 % defoliation treatments profoundly increased leaf lamina conductance (K lam) after 1 day and this increase was similar in magnitude for both defoliation treatments. Transpiration rates (E) rapidly declined after the removal of 75 % of leaves. However, E increased by over twofold in defoliated plants after 1 day and the increases in E and K lam were accompanied by five- and tenfold increases in the leaf expression of PIP2;4 in 50 and 75 % defoliation treatments, respectively. Defoliation treatments also stimulated net photosynthesis after 1 day and 3 weeks, although the increase was not as high as E. Leaf water potentials remained relatively stable following defoliation with the exception of a small decrease 1 day after defoliation which suggests that root water transport did not initially keep pace with the increased transpirational water loss. The results demonstrate the importance of root and leaf hydraulic properties in plant responses to defoliation and point to the involvement of PIP aquaporins in the early events following the loss of leaves.

Dehydration‐responsive element binding factors (DREBs) play important roles in plant growth, development, and stress signaling pathways in model plants. However, little is known about the function of DREBs in apple (Malus × domestica), a widely cultivated crop that is frequently threatened by drought. We isolated a DREB gene from Malus sieversii (Ledeb.) Roem., MsDREB6.2, and investigated its functions using overexpression analysis and c himeric re pressor gene‐s ilencing t echnology (CRES‐T). We identified possible target genes of the protein encoded by MsDREB6.2 using electrophoretic mobility shift assays (EMSAs) and chromatin immunoprecipitation (ChIP). Overexpression of MsDREB6.2 increased the expression of a key cytokinin (CK) catabolism gene, MdCKX4a, which led to a significant reduction in endogenous CK levels, and caused a decrease in shoot:root ratio in transgenic apple plants. Overexpression of MsDREB6.2 resulted in a decrease in stomatal aperture and density and an increase in root hydraulic conductance (L0), and thereby enhanced drought tolerance in transgenic plants. Furthermore, manipulating the level of MsDREB6.2 expression altered the expression of two aquaporin (AQP) genes. The effect of the two AQP genes on L0 was further characterized using the AQP inhibitor HgCl2. Based on these observations, we conclude that MsDREB6.2 enhances drought tolerance and that its function may be due, at least in part, to its influence on stomatal opening, root growth, and AQP expression. These results may have applications in apple rootstock breeding programs aimed at developing drought‐resistant apple varieties.

  • Soil pH is a major factor affecting plant growth. Plant responses to pH conditions widely vary between different species of plants. However, the exact mechanisms of high pH tolerance of plants are largely unknown. In the present study, we compared the pH responses of paper birch (Betula papyrifera) seedlings, a relatively sensitive species to high soil pH, with red‐osier dogwood (Cornus sericea), reported to be relatively tolerant of high pH conditions. We examined the hypotheses that tolerance of plants to high root zone pH is linked to effective control of root apoplastic pH to facilitate nutrient and water transport processes
  • In the study, we exposed paper birch and red‐osier dogwood seedlings for six weeks to pH 5, 7 and 9 under controlled‐environment conditions in hydroponic culture. Then, we measured biomass, gas exchange, root hydraulic conductivity, ferric chelate reductase (FCR) activity, xylem sap pH and the relative abundance of major elements in leaf protoplasts and apoplasts.
  • The study sheds new light on the rarely studied high pH tolerance mechanisms in plants. We found that compared with paper birch, red‐osier dogwood showed greater growth, higher gas exchange, and maintained higher root hydraulic conductivity as well as lower xylem sap pH under high pH conditions.
  • The results suggest that the relatively high pH tolerance of dogwood is associated with greater water uptake ability and maintenance of low apoplastic pH. These traits may have a significant impact on the uptake of Fe and Mn by leaf cells.

Recent work has shown that stomatal conductance (gs) and assimilation (A) are responsive to changes in the hydraulic conductance of the soil to leaf pathway (KL), but no study has quantitatively described this relationship under controlled conditions where steady‐state flow is promoted. Under steady‐state conditions, the relationship between gs, water potential (Ψ) and KL can be assumed to follow the Ohm's law analogy for fluid flow. When boundary layer conductance is large relative to gs, the Ohm's law analogy leads to gs = KLsoilleaf)/D, where D is the vapour pressure deficit. Consequently, if stomata regulate Ψleaf and limit A, a reduction in KL will cause gs and A to decline. We evaluated the regulation of Ψleaf and A in response to changes in KL in well‐watered ponderosa pine seedlings (Pinus ponderosa). To vary KL, we systematically reduced stem hydraulic conductivity (k) using an air injection technique to induce cavitation while simultaneously measuring Ψleaf and canopy gas exchange in the laboratory under constant light and D. Short‐statured seedlings (< 1 m tall) and hour‐long equilibration times promoted steady‐state flow conditions. We found that Ψleaf remained constant near ? 1·5 MPa except at the extreme 99% reduction of k when Ψleaf fell to ? 2·1 MPa. Transpiration, gs, A and KL all declined with decreasing k (P < 0·001). As a result of the near homeostasis in bulk Ψleaf, gs and A were directly proportional to KL (R2 > 0·90), indicating that changes in KL may affect plant carbon gain.

Water movement between a root and the soil depends on the hydraulic conductances of the soil, the root, and the intervening root-soil air gap (Lgap) created as roots shrink during soil drying. To measure Lgap, segments of young cylindrical roots of Agave deserti about 3 mm in diameter were placed concentrically or eccentrically within tubes of moistened filter paper at a known water potential. As the width of the air gap between the filter paper and a concentrically located root was made smaller, measured Lgap increased less than did predicted Lgap based on isothermal conditions. For gaps of the size expected in the soil during water loss from roots (e.g., 10% of the root radius), the underprediction was about 70% and was primarily caused by a lowering of the root surface temperature accompanying water evaporation. As a root segment was eccentrically moved toward the filter paper, the measured Lgap increased. For the most eccentric case of touching the filter paper, the measured Lgap was 2.4-fold greater than for the concentric case, compared with an infinite Lgap predicted if the water potential were constant around the root surface. When a root touched soil with a water potential of -1.0MPa, Lgap estimated using a graphical method increased about 2.3-fold and the overall conductance of the root-soil system increased by 31% compared with the concentric case. For markedly eccentric locations of roots in air gaps, Lgap, which can be the principal conductance initially limiting water loss from roots to a drying soil, can be about 60% of the value predicted for the concentric isothermal case.

We investigated how root hydraulic conductance (normalized to root dry weight, Lo) is regulated by the shoot. Shoot topping (about 30% reduction in leaf area) reduced Lo of grapevine (Vitis vinifera L.), soybean (Glycine max L.) and maize (Zea mays L.) by 50 to 60%. More detailed investigations with soybean and grapevine showed that the reduction in Lo was not correlated with the reduction in leaf area, and shading or cutting single leaves had a similar effect. Percentage reduction in Lo was largest when initial Lo was high in soybean. Inhibition of Lo by weak acid (low pH) was smaller after shoot damage or leaf shading. The half time of reduction in Lo was approximately 5 min after total shoot decapitation. These characteristics indicate involvement of aquaporins. We excluded phloem‐borne signals and auxin‐mediated signals. Xylem‐mediated hydraulic signals are possible since turgor rapidly decreased within root cortex cells after shoot topping. There was a significant reduction in the expression of several aquaporins in the plasma membrane intrinsic protein (PIP) family of both grapevine and soybean. In soybean, there was a five‐ to 10‐fold reduction in GmPIP1;6 expression over 0.5–1 h which was sustained over the period of reduced Lo.

Water relations, mineral composition, growth and root morphology were studied in pepper plants (Capsicum annuum L. cv California Wonder). Two NaCl concentrations (30 and 60 mM) and two nutrient solutions in which the concentrations of macronutrients were increased were used to assess the ionic and osmotic effects of NaCl in these plants. The hydraulic conductivity (Lo), stomatal conductance (gs), percentage of open stomata and pressure potential (Ψp) decreased with all treatments, in a similar way for 30 mM NaCl and for its iso-osmotic solution of macronutrients, however, the decrease was higher for 60 mM NaCl than for its iso-osmotic solution. Ion analyses also revealed that nutrient concentrations were altered greatly at 60 mM NaCl. Also, changes in morphology, such as increases in cortex cell size and in intercellular spaces, were detected. Therefore, at low salinity, the effect of NaCl was mainly osmotic, however, under higher salinity also the toxicity of Na+ and Cl participate.

The adaptation capacity of olive trees to different environments is well recognized. However, the presence of microorganisms in the soil is also a key factor in the response of these trees to drought. The objective of the present study was to elucidate the effects of different arbuscular mycorrhizal (AM) fungi coming from diverse soils on olive plant growth and water relations. Olive plants were inoculated with native AM fungal populations from two contrasting environments, that is, semi‐arid – Freila (FL) and humid – Grazalema (GZ) regions, and subjected to drought stress. Results showed that plants grew better on GZ soil inoculated with GZ fungi, indicating a preference of AM fungi for their corresponding soil. Furthermore, under these conditions, the highest AM fungal diversity was found. However, the highest root hydraulic conductivity (Lpr) value was achieved by plants inoculated with GZ fungi and growing in FL soil under drought conditions. So, this AM inoculum also functioned in soils from different origins. Nine novel aquaporin genes were also cloned from olive roots. Diverse correlation and association values were found among different aquaporin expressions and abundances and Lpr, indicating how the interaction of different aquaporins may render diverse Lpr values.
