Effect of Streptomycin on Some Enzyme Systems of Bacillus subtilis   总被引:1,自引:0,他引:1       下载免费PDF全文
Streptomycin slightly inhibited lactic and malic dehydrogenases of Bacillus subtilis, and inhibited isocitric dehydrogenase to about 60%. The formation of lactic dehydrogenase, glutamic dehydrogenase, alpha-alanine dehydrogenase, and succinic dehydrogenase was stimulated by the antibiotic at a concentration causing 50% inhibition of bacterial growth. Streptomycin had practically no influence on the formation of malic dehydrogenase, but the antibiotic produced 48% inhibition of the synthesis of isocitric dehydrogenase.  相似文献

Enzyme patterns were obtained by starch-gel electrophoresis of cell-free extracts from organisms in the family Bacteroidaceae. Esterases, phosphatases, and lactic and succinic dehydrogenases were detected but offered no possibility for classification purposes. Alpha-glycerophosphate dehydrogenase and 6-phosphogluconate dehydrogenase were not detected. Zymograms of malic, glutamic, and glucose-6-phosphate dehydrogenases separated Bacteroides species from Sphaerophorus and Fusobacterium species. Malic dehydrogenase and glucose-6-phosphate dehydrogenase were found in Bacteroides but not Sphaerophorus and Fusobacterium. These dehydrogenase zymograms placed three gram-negative, nonsporeforming anaerobic rod isolates with the Bacteroides species. A close correlation was found between the classification of Bacteroidaceae by zymogram analysis and a numerical taxonomy scheme previously published from this laboratory.  相似文献

A histochemical study on the distribution of mitochondrial and oxidative enzymes viz., succinic, NAD and NADP-linked isocitrate and malic dehydrogenases and cytochrome oxidase in the hepatopancreas of Scylla serrata (Forskål) shows that the R cells possess the highest concentration of them at the apical parts; F and B cells showed poor staining reactions whereas E cells and connective tissue exhibited trace staining reactions. A moderate staining for these was obtained in the cells lining the main ducts.

The bilateral removal of eyestalks resulted in the stimulation of succinic and NAD-linked malic dehydrogenases and cytochrome oxidase, observed 4 h after the operation; after 24 h, however, these enzymes showed a slight reduction in the activity. A significant increase in the activity of NADP-linked isocitrate and malic dehydrogenases was noted 24 h after eyestalk ablation. The major alterations were noticed in the R cells.

After injecting eyestalk extract into destalked or intact crabs, a fall in the staining intensity of succinic, NAD-linked malic and isocitrate dehydrogenases in the R cells was apparent 2–4 h after the treatment but was subsequently re-established after 24 h.

It seems from the present results that there may be a factor(s) in the eyestalk of S. serrata which regulates the oxidative metabolism in the hepatopancreas. The physiological significance of oxidative enzymes in various cytologic elements is discussed.  相似文献

Summary Histochemical observations were made of oxidative and reducing enzymes in relation to several metabolic pathways in the developing tooth. The localization and activity of dehydrogenases and oxidases could be divided roughly into two types.Type one refers to enzymatic activities of lactic, succinic, malic and glucose-6-phosphate dehydrogenase and aconitase and cytochrome oxidase, which were low in undifferentiated dental epithelium and increased in proportion to cell differentiation. Comparatively high enzymatic activities were reflected in high cell function.Type two did not increase in the enzymatic activity during differentiation. Glyceraldehyde-3-phosphate, TPN isocitric, 6-phosphogluconate, glutamic, -glycerophosphate and -hydroxybutyric dehydrogenase and monoamine oxidase belong to this type.With 12 Figures in the TextPresented in Parts at the 3rd Annual Meeting in September, 1962 of the Japanese Histochemical Association.  相似文献

We have investigated the circular dichroism induced in Cibacron Blue and Congo Red upon binding to several dehydrogenases to probe the conformation of the bound dyes. The circular dichroism spectra of Congo Red are quite similar when the dye is bound to lactic dehydrogenase, glyceraldehyde-3-phosphate dehydrogenase, and alcohol dehydrogenase but has bands of opposite sign when bound to cytoplasmic malic dehydrogenase. The circular dichroism spectra of Cibacron Blue bound to these same dehydrogenases are quite different from one another. Since circular dichroism is sensitive to the conformation of bound dye, these differences argue for at least local changes in dye conformation or environment when bound to different dehydrogenases. Congo Red appears to be less sensitive to these effects than Cibacron Blue.  相似文献

Effects of low-molecular-weight organic acids on the accumulation and transportation of gadolinium (Gd) in tomato plants were studied under hydroponic condition. The results indicated that changes of organic acids occurred in the processes of Gd accumulation and transportation in tomato plants which were treated with extraneous Gd solutions. Malic, citric, and succinic acids contributed to both Gd accumulation in roots and transportation in xylem vessels. When Gd was unloaded from the xylem to the leaf cells, formic, lactic, citric, and succinic acids played important roles in Gd accumulation in leaves. When tomato plants were cultured in the uptake solution of Gd-containing malic, citric, or succinic acid for 48 h, the succinic acid in roots and leaves and the malic acid in xylem saps both increased obviously. From the results above, we can conclude that succinic acid had the most important role in Gd accumulation in tomato roots and leaves, while malic acid transported Gd via xylem vessels more effectively.  相似文献

SYNOPSIS. Transection and repair in Blepharisma intermedium modify the distribution of several enzymes. The number of active phagosomes is reduced and there are increased staining reactions for acid phosphatase, the nucleases and type-B esterases in the ground cytoplasm. 5'-Nucleotidase activity is localized in the cytoplasmic matrix. Except for ribonuclease, the enzymes may have a perimacronuclear deposition at some period during regeneration. Acid phosphatase may be involved in development of the oral membranelles. Adenosine triphosphatase and lactic dehydrogenase have a moderate staining intensity whereas malic and succinic dehydrogenases have a strong reaction in the cytoplasm; in addition, these enzymes may be localized in the kinetics involved in the neoformation of the oral concrescence. Nuclear reorganization is attended by an increased frequency of peripheral dehydrogenase activity in the late stages of regeneration. These observations are compared with catalytic systems known to function in protozoan metabolism and in vertebrate regeneration.  相似文献

The activities of NADP-linked malic enzyme, hexose monophosphate shunt dehydrogenases and NADP-linked isocitrate dehydrogenase were studied during development of skeletal muscle and compared with those in the liver. The variation patterns of malic enzyme activity in the liver and in the skeletal muscle were very similar, however the amplitude of the changes was different. The enzyme activity increased approx 16-fold in the liver and about 2-fold in skeletal muscle at the same stage of development. In skeletal muscle the increase of the malic enzyme activity was only slightly higher than of lactic dehydrogenase and citrate synthase. Studies on the intracellular distribution of malic enzyme in skeletal muscle showed that both mitochondrial and extramitochondrial enzymes increased between 20th and 37th day of life, the increase of the extramitochondrial enzyme being more pronounced. The hexose monophosphate shunt dehydrogenases activity showed an increase in the liver but no change was observed in the skeletal muscle at the weaning time. Changes in the activity of the liver and skeletal muscle isocitrate dehydrogenase were not significant between 10th and 80th day of life. The results suggest that the malic enzyme in the liver is playing a different physiological role than in the skeletal muscle.  相似文献

In comparative study of respiratory metabolism, it was established that the relative proportions of respiratory end-products (succinic, acetic and lactic acids) differed consistently in two strains of Hymenolepis diminuta (Toronto and ANU). The ANU strain produced more lactic acid and less succinic acid under aerobic and anaerobic conditions. In the shift from aerobic to anaerobic conditions both strains compensated by increasing their outputs of succinic acid. The ANU strain possessed significantly higher activities of hexokinase, pyruvate kinase, lactate dehydrogenase, cytosolic and mitochondrial malic enzyme and cytosolic α-glycerophosphate dehy drogenase. The Toronto strain had significantly higher activities of fumarase, succinate dehydrogenase, and fumarate reductase. There were no significant differences in the activities of phosphoenolpyruvate carboxykinase and malic dehydrogenase between strains. The fumarase activity in the Toronto strain was 16 times that of the ANU strain, its Km (malate) was 0.8mM, as opposed to 2.5 mM, and it was less sensitive to inhibition by NAD or ATP. These observations are consistent with the patterns of end-product formation in the two strains. Ratios of end-products and calculations of approximate redox balance suggest that the Toronto strain may have a greater capacity for aerobic metabolism.  相似文献

Feeding a basal diet free of vitamins E and C to weanling male rats for 8 months resulted in biochemical changes characteristic of vitamin E deficiency. These included increased liver thiobarbituric acid values; decreased blood GSH levels, plasma vitamin E levels, and glutathione peroxidase activities; and increased activities of plasma pyruvate kinase, glutamic-oxaloacetic transaminase, creatine kinase, lactic dehydrogenase, and malic dehydrogenase. Tube-feeding vitamin C for 21 days resulted in partial reversal effects on the above parameters except activities of glutathione peroxidase, lactic dehydrogenase, and malic dehydrogenase. The results suggest that vitamin C may spare in part the metabolism of vitamin E through its antioxidant property.  相似文献

Summary A histochemical observation was made of various dehydrogenase activities in oral squamous epithelia. The localization of dehydrogenases showed a relatively similiarity except for the intensity of the dehydrogenase activity. Succinic dehydrogenase activity was generally confined to the basal cell layer and adjacent cell layers; superficial layers did not show any enzymatic activity. Lactic, and malic dehydrogenase activities were localized in the basal cells to st. granulosum, and the activity of lactic dehydrogenase was the highest. -Glycerophosphate, glutamic, glucose-6-phosphate and TPN-isocitric dehydrogenase activities were observed in all the epithelial cells with the exception for the hornified layer, and they were found generally low. -Hydroxybutyric dehydrogenase was low and contained in both of st. germinativum and st. granulosum, the keratohyalin in st. granulosum being occasionally found reactive to this enzymatic activity.In connective tissue cells and collagen bundels, activities of lactic, and malic dehydrogenase were intense, while other dehydrogenases were low or trace amount.In the oral squamous epithelium under normal conditions, the dehydrogenase localization concerning the glucose metabolism and TCA cycle member and other close pathways was not similar. Nor were their activities found likewise. Adult male rats of Wistar strain were subcutaneously injected with a single dose of 0.025 mM CdCl2/kg body weight. The autopsies were performed 2 and 12 weeks post cadmium injection. Experimental animals showed a normal gain of body weight while a marked lowering of the testes weight was observed. The lowering of testes weight of cadmium treated rats occured mainly due to the necrosis of seminiferous tubules, volume of which is markedly lower if compared to intact control rats. After 2 weeks of experiment a marked enlargement of the interstitial gland of the testis occured while after 12 weeks the gland is markedly lower if compared to control rats and to rats studied 2 weeks post cadmium injection. Histochemical reactions for lactic, succinic, alpha-glycerophosphate and 3beta-hydroxy steroid dehydrogenases, NADH tetrazolium reductase, acid phosphatase and nonspecific esterases showed for irreversible necrotic changes of seminiferous tubules of cadmium treated rats while the damage to interstitial gland is only temporary.  相似文献

Spinal ganlia of a 9-day chick embryo were cultivated by the method of "floating rafts" in common medium (control) and in the medium containing amizyl (100 microgram/ml) or a neuregrowth factor (50 microgram/ml). With the action of amizyl there proved to be an increase in the number of surviving neurons; the majority of these neurons contained monoaminoxidase; there was a rise of NAD-diaphorase activity, and, to a lesser extent, of lactic dehydrogenase and isocitric dehydrogenase activities. The neurogrowth factor caused an increase in the number of nerve cells with acetylcholinesterase; there was an elevation of NAD-diaphorase and some rise of malic dehydrogenase activities; the activity of lactic dehydrogenase became maximal; as to succinic dehydrogenase--its activity was somewhat suppressed.  相似文献

Feeding the thermogenic steroid, 5-androsten-3 beta-ol-17-one (dehydroepiandrosterone, DHEA) in the diet of rats induced the synthesis of liver mitochondrial sn-glycerol 3-phosphate dehydrogenase to levels three to five times that of control rats within 7 days. The previously reported enhancement of liver cytosolic malic enzyme was confirmed. The induction of both enzymes was detectable at 0.01% DHEA in the diet, reached plateau stimulation at 0.1 to 0.2%, and was completely blocked by simultaneous treatment with actinomycin D. Feeding DHEA caused smaller, but statistically significant increases of liver cytosolic lactate, sn-glycerol 3-phosphate, and isocitrate (NADP(+)-linked) dehydrogenases but not of malate or glucose 6-phosphate dehydrogenases. The capability of DHEA to enhance mitochondrial glycerophosphate dehydrogenase and malic enzyme was influenced by the thyroid status of the rats; was smallest in thyroidectomized rats and highest in rats treated with triiodothyronine. 5-Androsten-3 beta,17 beta-diol and 5-androsten-3 beta-ol-7,17-dione were as effective as DHEA in enhancing the liver mitochondrial glycerophosphate dehydrogenase and malic enzyme. Administering compounds that induce the formation of cytochrome P450 enzymes enhanced liver malic enzyme activity but not that of mitochondrial glycerophosphate dehydrogenase. Arochlor 1254 and 3-methylcholanthrene also increased the response of malic enzyme to DHEA feeding.  相似文献

A method of obtaining an extract of soluble enzymes from peaseedling mitochondria is described. Evidence is presented thatthe mitochondrial extract contains the following enzymes: Diphosphopyridinenucleotide (DPN) and triphosphopy-ridine nucleotide (TPN) specificwocitric dehydrogenases, alcohol dehydrogenase, formic dehydrogenase,aldehyde dehydrogenase, glutamic dehydrogenase, malic enzyme,lactic dehydrogenase, fumarase, aconitase, DPN and TPN cytochrome-creductases, adenylate kinase, phosphopyridine nucleotide transhydrogenaseand oxaloacetic carboxylase. Investigations on the effect of reserpine on the histochemical reactions to some dehydrogenases (succinic, lactic, glucose-6-phosphate dehydrogenase) as well as to NADH and NADPH tetrazole reductases in the seminiferous epithelium of white rats have been performed. A weakening of the dehydrogenase activities has been mainly observed in the young cells of seminal epithelium i, e. in spermatogone and spermatocytes. The changes in histoenzymatical reactions caused by reserpine may be the evidence of the inhibition of oxidation-reduction processes in the studied reproductive cells.  相似文献

Acute renal failure was induced in male rats by the subcutaneous injectioon of 4 mg HgC12 per kg body weight. Enzyme activities of the proximal tubule were studied histochemically at six time intervals from 15 min to 24 h. The enzyme studied were alkaline phosphatase, 5'-nucleotidase, acid phosphatase, alpha-glycerophosphate dehydrogenase (NAD-independent), malic dehydrogenase, succinic dehydrogenase, latic dehydrogenase, glucose-6-phosphate dehydrogenase and glucose-6-phosphatase. Decreases in activity were observed for alkaline phosphatase and 5'-nucleotidase after 15 min. Acid phosphatase was decreased after 30 min. These three enzymes returned to control levels after 3 h, but malic dehydrogenase and alpha-glycerophosphate dehydrogenase were decreased at this time interval. Succinic dehydrogenase was first decreased after 6 h. The earliest morphological changes detectable by light microscopy were observed in pars recta tubules in the medullary rays after 6 h, a time when all enzymes studied showed widespread decreased activity throughout the proximal tubule. After 24 h, the pars convoluta appeared morphologically normal but the pars recta was necrotic and exhibited calcification, whereas enzyme activity was decreased (absent in some cases) in both pars convoluta and pars recta. These results support the hypothesis that Hg++, when given in a sublethal dose, is associated with early histochemical changes in the brush border of the proximal tubule, which may be related to early changes in sodium reabsorption and to the subsequent development of acute renal failure. The observation that changes in plasma membrane-associated enzymes occur early and prior to alterations in enzymes of mitochondria and the endoplasmic reticulum suggests that Hg++ interacts initially with the plasma membrane.  相似文献

Cercariae of Plagiorchis elegans Rudolphi 1802 collected from experimentally infected snails, Lymnaea palustris, were subjected to various histochemical tests for dehydrogenase systems. A high degree of activity was demonstrated for succinic dehydrogenase (EC 1.3.99.1), malic dehydrogenase (EC 1.1.1.37), isocitric dehydrogenase (EC 1.1.1.41), α-glycerophosphate dehydrogenase (EC 1.1.1.8), and glucose 6-phosphate dehydrogenase (EC 1.1.1.49). These enzymes were present in the tegument, tail, caudal pocket, excretory bladder, acetabulum, and oral sucker, particularly in the muscles around the stylet. Only moderate activity was obtained for lactic dehydrogenase (EC 1.1.1.27) and 6-phosphogluconate dehydrogenase (EC 1.1.1.44) at these sites, glutamic dehydrogenase (EC 1.4.1.2) was localized only in the tails of the cercariae and tests for alcohol dehydrogenase (EC 1.1.1.1) were completely negative. The cerebral ganglia and its commissures stained intensely in the tests for succinic, isocitric, α-glycerophosphate, and glucose 6-phosphate dehydrogenase systems. The results indicate the possibility that several energy-producing sequences may be available to these cercariae.  相似文献

The rates of activity of the dehydrogenase systems in Tetrahymena, which are concerned with carbohydrate oxidation, in descending order of activity are: lactic > isocitric > succinic = glucose > glucose-6-phosphate = 6-phosphogluconic = malic > glutamic = cytochrome linked α-glycerophosphate dehydrogenase. No evidence was obtained to indicate the presence of DPN linked α-glycerophosphate dehydrogenase.  相似文献

The tubulin content and biochemical components were determined in the cerebrum, cerebellum and hypothalamus from intact and T3-treated male and female rats during early life. T3-treatment between 0 and 9 days of age increased soluble protein, RNA DNA and tubulin content (mg per g tissue) in the 10-day-old male cerebellum but not in the cerebrum and hypothalamus except for soluble protein and tubulin (mg per g tissue), respectively. Intracellular tubulin content (mg per mg DNA) was increased by the T3-treatment in the 10-day-old male hypothalamus but not the other regions. When T3 was administered between 10 and 19 days, there was little effect of the treatment; increased tubulin (mg per g tissue) in the cerebrum and decreased RNA (mg per g tissue) and a ratio of tubulin to protein in the cerebellum from 20-day-old males. Less response to T3-treatment was observed in female cerebrum and hypothalamus but not in the cerebellum, compared with the male. These results suggest that the effect of T3-treatment on brain is modified by several factors such as tissue specificity, age-dependency and sexual differences. Modification by these factors might depend, at least in part, on changes in the number of T3-receptors due to the hormone treatment.  相似文献
