McNemar test is commonly used to test for the marginal homogeneity in 2 × 2 contingency tables. McNemar test is an asymptotic test based either on standard normal distribution or on the chi‐square distribution. When the total sample size is small, an exact version of McNemar test is available based on the binomial probabilities. The example in the paper came from a clinical study to investigate the effect of epidermal growth factor for children who had microvillus inclusion diseases. There were only six observations available. The test results differ between the exact test and the asymptotic test. It is a common belief that with this small sample size the exact test be used. However, we claim that McNemar test performs better than the exact test even when the sample size is small. In order to investigate the performances of McNemar test and the exact test, we identify the parameters that affect the test results and then perform sensitivity analysis. In addition, through Monte Carlo simulation studies we compare the empirical sizes and powers of these tests as well as other asymptotic tests such as Wald test and the likelihood ratio test.  相似文献

The Cochran-Armitage test has commonly been used for a trend test in binomial proportions. The quasi-likelihood method provides a simple approach to model extra-binomial proportions. Two versions of the score and Wald tests using different parameterizations for the extra-binomial variance were investigated: one in terms of intercluster correlation, and another in terms of variance. The Monte Carlo simulation was used to evaluate the performance of the each version of the score test and the Wald test, and the Cochran-Armitage test. The simulation shows that the Cochran-Armitage test has the proper size only for the binomial sample data, and the test is no longer valid when applied to the extra-binomial data. The Wald test is more likely to exceed the nominal level than the score test under either intercluster correlation model or variance model. Both score tests performed very well even with the binomial data; the tests control the type I error and in the meantime maintain the power of detecting the dose effects. Based on the design considered in this paper, the two scores test are comparable. The score test based on the intercluster correlations model seems better controlling the Type I error but appears less powerful than that based on the variance model. Summary We present an adaptive percentile modified Wilcoxon rank sum test for the two‐sample problem. The test is basically a Wilcoxon rank sum test applied on a fraction of the sample observations, and the fraction is adaptively determined by the sample observations. Most of the theory is developed under a location‐shift model, but we demonstrate that the test is also meaningful for testing against more general alternatives. The test may be particularly useful for the analysis of massive datasets in which quasi‐automatic hypothesis testing is required. We investigate the power characteristics of the new test in a simulation study, and we apply the test to a microarray experiment on colorectal cancer. These empirical studies demonstrate that the new test has good overall power and that it succeeds better in finding differentially expressed genes as compared to other popular tests. We conclude that the new nonparametric test is widely applicable and that its power is comparable to the power of the Baumgartner‐Weiß‐Schindler test.  相似文献

目的:根据计数培养基适用性检查和计数方法适用性试验结果,建立维生素K1微生物限度测定方法。方法:以聚山梨酯80作为乳化剂,使维生素K1在p H 7.0无菌氯化钠-蛋白胨缓冲液中充分乳化。取1:20供试液1 m L,按平皿法分别用营养琼脂培养基和玫瑰红钠琼脂培养基培养,以计数细菌、霉菌和酵母菌。结果:验证所用培养基的菌落平均数均大于对照培养基上的70%,且菌落形态大小一致。稀释液对照组和试验组培养基上菌落平均数的回收率均大于70%,且菌落形态大小一致。三批维生素K1及其加速和长期稳定性样品中均未见菌落。结论:所选培养基适宜大肠埃希菌、金黄色葡萄球菌和白色念珠菌生长。且含聚山梨酯80的p H 7.0无菌氯化钠-蛋白胨缓冲液和维生素K1对所含大肠埃希菌、金黄色葡萄球菌和白色念珠菌无抑菌性。  相似文献

Constants of allometric growth are commonly estimated by the first eigenvector of the covariance matrix of log measurements. Hills (1982, in Encyclopedia of Statistical Sciences, 48-54) defines a model of allometric extension for two related species by the conditions that (a) the constants of allometric growth are identical for both species and (b) the vector of mean differences is proportional to the common first eigenvector of both covariance matrices. We give a test for allometric extensionand discuss estimation of the parameters of the allometric extension model, including standard errors.  相似文献

In survivorship modelling using the proportional hazards model of Cox (1972, Journal of the Royal Statistical Society, Series B, 34, 187–220), it is often desired to test a subset of the vector of unknown regression parameters β in the expression for the hazard rate at time t. The likelihood ratio test statistic is well behaved in most situations but may be expensive to calculate. The Wald (1943, Transactions of the American Mathematical Society 54, 426–482) test statistic is easier to calculate, but has some drawbacks. In testing a single parameter in a binomial logit model, Hauck and Donner (1977, Journal of the American Statistical Association 72, 851–853) show that the Wald statistic decreases to zero the further the parameter estimate is from the null and that the asymptotic power of the test decreases to the significance level. The Wald statistic is extensively used in statistical software packages for survivorship modelling and it is therefore important to understand its behavior. The present work examines empirically the behavior of the Wald statistic under various departures from the null hypothesis and under the presence of Type I censoring and covariates in the model. It is shown via examples that the Wald statistic's behavior is not as aberrant as found for the logistic model. For the single parameter case, the asymptotic non-null distribution of the Wald statistic is examined.  相似文献

When analyzing mortality data due to rare diseases in small areas, it is common to find several health zones with no mortality cases. In these circumstances, the classical homogeneous model based on the Poisson distribution used to estimate the relative risks within each area may encounter lack of fit due to a disproportionately large frequency of zeros. To cope with these zeros, the zero inflated Poisson model can be used. In this paper, we propose a test for detecting zero inflation in the context of disease mapping which is based on bootstrap techniques. The test is illustrated using male mortality data due to brain cancer in Navarra, Spain. In addition, comparisons with other tests for Poisson zero inflation such as the score test and the likelihood ratio test are carried out in terms of empirical power and size using the brain cancer scenario. Overdispersion is a common phenomenon in Poisson modeling, and the negative binomial (NB) model is frequently used to account for overdispersion. Testing approaches (Wald test, likelihood ratio test (LRT), and score test) for overdispersion in the Poisson regression versus the NB model are available. Because the generalized Poisson (GP) model is similar to the NB model, we consider the former as an alternate model for overdispersed count data. The score test has an advantage over the LRT and the Wald test in that the score test only requires that the parameter of interest be estimated under the null hypothesis. This paper proposes a score test for overdispersion based on the GP model and compares the power of the test with the LRT and Wald tests. A simulation study indicates the score test based on asymptotic standard Normal distribution is more appropriate in practical application for higher empirical power, however, it underestimates the nominal significance level, especially in small sample situations, and examples illustrate the results of comparing the candidate tests between the Poisson and GP models. A bootstrap test is also proposed to adjust the underestimation of nominal level in the score statistic when the sample size is small. The simulation study indicates the bootstrap test has significance level closer to nominal size and has uniformly greater power than the score test based on asymptotic standard Normal distribution. From a practical perspective, we suggest that, if the score test gives even a weak indication that the Poisson model is inappropriate, say at the 0.10 significance level, we advise the more accurate bootstrap procedure as a better test for comparing whether the GP model is more appropriate than Poisson model. Finally, the Vuong test is illustrated to choose between GP and NB2 models for the same dataset.  相似文献

This article focuses on conducting global testing for association between a binary trait and a set of rare variants (RVs), although its application can be much broader to other types of traits, common variants (CVs), and gene set or pathway analysis. We show that many of the existing tests have deteriorating performance in the presence of many nonassociated RVs: their power can dramatically drop as the proportion of nonassociated RVs in the group to be tested increases. We propose a class of so-called sum of powered score (SPU) tests, each of which is based on the score vector from a general regression model and hence can deal with different types of traits and adjust for covariates, e.g., principal components accounting for population stratification. The SPU tests generalize the sum test, a representative burden test based on pooling or collapsing genotypes of RVs, and a sum of squared score (SSU) test that is closely related to several other powerful variance component tests; a previous study (Basu and Pan 2011) has demonstrated good performance of one, but not both, of the Sum and SSU tests in many situations. The SPU tests are versatile in the sense that one of them is often powerful, although its identity varies with the unknown true association parameters. We propose an adaptive SPU (aSPU) test to approximate the most powerful SPU test for a given scenario, consequently maintaining high power and being highly adaptive across various scenarios. We conducted extensive simulations to show superior performance of the aSPU test over several state-of-the-art association tests in the presence of many nonassociated RVs. Finally we applied the SPU and aSPU tests to the GAW17 mini-exome sequence data to compare its practical performance with some existing tests, demonstrating their potential usefulness.  相似文献

Here we describe a random effects threshold dose-response model for clustered binary-response data from developmental toxicity studies. For our model we assume that a hormetic effect occurs in addition to a threshold effect. Therefore, the dose-response curve is based on two components: relationships below the threshold (hormetic u-shaped model) and those above the threshold (logistic model). In the absence of hormesis and threshold effects, the estimation procedure is straightforward. We introduce score tests that are derived from a random effects hormetic-threshold dose-response model. The model and tests are applied to clustered binary data from developmental toxicity studies of animals to test for hormesis and threshold effects. We also compare the score test and likelihood ratio test to test for hormesis and threshold effects in a simulated study.  相似文献

O'Brien (1984, Biometrics 40, 1079-1087) introduced a simple nonparametric test procedure for testing whether multiple outcomes in one treatment group have consistently larger values than outcomes in the other treatment group. We first explore the theoretical properties of O'Brien's test. We then extend it to the general nonparametric Behrens-Fisher hypothesis problem when no assumption is made regarding the shape of the distributions. We provide conditions when O'Brien's test controls its error probability asymptotically and when it fails. We also provide adjusted tests when the conditions do not hold. Throughout this article, we do not assume that all outcomes are continuous. Simulations are performed to compare the adjusted tests to O'Brien's test. The difference is also illustrated using data from a Parkinson's disease clinical trial.  相似文献

The mechanical strength of a plant stem (a load-bearing organ) helps the plant resist drooping, buckling and fracturing. We previously proposed a method for quickly evaluating the stiffness of an inflorescence stem in the model plant Arabidopsis thaliana based on measuring its natural frequency in a free-vibration test. However, the relationship between the stiffness and flexural rigidity of inflorescence stems was unclear. Here, we compared our previously described free-vibration test with the three-point bending test, the most popular method for calculating the flexural rigidity of A. thaliana stems, and examined the extent to which the results were correlated. Finally, to expand the application range, we present an example of a modified free-vibration test. Our results provide a reference for improving estimates of the flexural rigidity of A. thaliana inflorescence stems.  相似文献

Summary .   Due to competition among individual trees and other exogenous factors that change the growth environment, each tree grows following its own growth trend with some structural changes in growth over time. In the present article, a new method is proposed to detect a structural change in the growth process. We formulate the method as a simple statistical test for signal detection without constructing any specific model for the structural change. To evaluate the p -value of the test, the tube method is developed because the regular distribution theory is insufficient. Using two sets of tree diameter growth data sampled from planted forest stands of Cryptomeria japonica in Japan, we conduct an analysis of identifying the effect of thinning on the growth process as a structural change. Our results demonstrate that the proposed method is useful to identify the structural change caused by thinning. We also provide the properties of the method in terms of the size and power of the test.  相似文献

The sibship disequilibrium test (SDT) is designed to detect both linkage in the presence of association and association in the presence of linkage (linkage disequilibrium). The test does not require parental data but requires discordant sibships with at least one affected and one unaffected sibling. The SDT has many desirable properties: it uses all the siblings in the sibship; it remains valid if there are misclassifications of the affectation status; it does not detect spurious associations due to population stratification; asymptotically it has a chi2 distribution under the null hypothesis; and exact P values can be easily computed for a biallelic marker. We show how to extend the SDT to markers with multiple alleles and how to combine families with parents and data from discordant sibships. We discuss the power of the test by presenting sample-size calculations involving a complex disease model, and we present formulas for the asymptotic relative efficiency (which is approximately the ratio of sample sizes) between SDT and the transmission/disequilibrium test (TDT) for special family structures. For sib pairs, we compare the SDT to a test proposed both by Curtis and, independently, by Spielman and Ewens. We show that, for discordant sib pairs, the SDT has good power for testing linkage disequilibrium relative both to Curtis''s tests and to the TDT using trios comprising an affected sib and its parents. With additional sibs, we show that the SDT can be more powerful than the TDT for testing linkage disequilibrium, especially for disease prevalence >.3.  相似文献

In meta-analysis, hypothesis testing is one of the commonly used approaches for assessing whether heterogeneity exists in effects between studies. The literature concluded that the Q-statistic is clearly the best choice and criticized the performance of the likelihood ratio test in terms of the type I error control and power. However, all the criticism for the likelihood ratio test is based on the use of a mixture of two chi-square distributions with 0 and 1 degrees of freedom, which is justified only asymptotically. In this study, we develop a novel method to derive the finite sample distribution of the likelihood ratio test and restricted likelihood ratio test statistics for testing the zero variance component in the random effects model for meta-analysis. We also extend this result to the heterogeneity test when metaregression is applied. A numerical study shows that the proposed statistics have superior performance to the Q-statistic, especially when the number of studies collected for meta-analysis is small to moderate.  相似文献

The proportional odds model provides a powerful tool for analysing ordered categorical data and setting sample size, although for many clinical trials its validity is questionable. The purpose of this paper is to present a new class of constrained odds models which includes the proportional odds model. The efficient score and Fisher's information are derived from the profile likelihood for the constrained odds model. These results are new even for the special case of proportional odds where the resulting statistics define the Mann‐Whitney test. A strategy is described involving selecting one of these models in advance, requiring assumptions as strong as those underlying proportional odds, but allowing a choice of such models. The accuracy of the new procedure and its power are evaluated.  相似文献

为筛选一株产黑色素能力强的菌株并优化其培养条件。通过ITS测序鉴定11株供试菌株,以菌丝生长速度、平板L值等指标筛选出一株产黑能力强的香灰菌,并对其生长所需碳源、氮源、pH等培养条件进行优化。研究表明,11株供试菌株均为香灰菌(Hypoxylon sp.),其中Hp.sp0006菌丝生长速度较快、菌球大且均匀、L值最低,并且发酵液黑色素含量最高。该菌株最优培养条件是,以葡萄糖为碳源、牛肉浸膏为氮源、碳氮比20∶1并添加10 mg维生素B1,黑色素含量可达(1.21±0.17)g/L。香灰菌Hp.sp0006是一株产黑色素较高的菌株,优化后的培养基更有利于黑色素的合成,为香灰菌黑色素的开发利用奠定基础。  相似文献

Fisher's exact test for comparing response proportions in a randomized experiment can be overly conservative when the group sizes are small or when the response proportions are close to zero or one. This is primarily because the null distribution of the test statistic becomes too discrete, a partial consequence of the inference being conditional on the total number of responders. Accordingly, exact unconditional procedures have gained in popularity, on the premise that power will increase because the null distribution of the test statistic will presumably be less discrete. However, we caution researchers that a poor choice of test statistic for exact unconditional inference can actually result in a substantially less powerful analysis than Fisher's conditional test. To illustrate, we study a real example and provide exact test size and power results for several competing tests, for both balanced and unbalanced designs. Our results reveal that Fisher's test generally outperforms exact unconditional tests based on using as the test statistic either the observed difference in proportions, or the observed difference divided by its estimated standard error under the alternative hypothesis, the latter for unbalanced designs only. On the other hand, the exact unconditional test based on the observed difference divided by its estimated standard error under the null hypothesis (score statistic) outperforms Fisher's test, and is recommended. Boschloo's test, in which the p-value from Fisher's test is used as the test statistic in an exact unconditional test, is uniformly more powerful than Fisher's test, and is also recommended.  相似文献

In constructed wetlands for the treatment of industrial effluents and in contaminated waterlogged soils, wetland plants (helophytes) are exposed to toxic chemicals. Hence, the plants' resistance to contaminants is an important prerequisite for applying phytoremediation to solve these environmental problems. For toxicity tests on the germination and growth of various helophytes (Phalaris arundinaceae and Phragmites australis), phenol, phenanthrene, and a mixture of both were used as examples of chemicals from the petrol- and coal-processing industries. The germination rate, shoot length, root length, and influence on leaves, young shoots, and dry weight were studied. Although an increase in contaminant concentration decreased plant growth (dry weight, shoot length); interestingly, the number of young shoots rose. Low contaminant concentration (about 50 mg/l in case of phenol) stimulated the plant growth. The cress seed germination test was less susceptible compared with plantlet exposure in the case of phenol and phenanthrene. Due to its low bioavailability, solid phenanthrene (without solutizer) did not significantly affect plant growth.  相似文献
