To find the cause of individual differences in caffeine intake and its metabolism, we investigated the effects of lifestyle and genetic polymorphisms of caffeine metabolic enzymes on coffee or black tea and urinary caffeine levels among 259 male Japanese. It was seen that cigarette smokers drank more coffee or black tea than non-smokers (p < 0.001). There was an inverse correlation between the amount of coffee or black tea consumed and age or the frequency of alcohol drinking (p < 0.05). Genetic polymorphisms of N -acetyltransferase 2 (NAT2), cytochrome P450 (CYP)1A1 and 2E1 did not significantly affect the habit of drinking coffee or black tea. The frequency of allele 1, the NAT2 allele of rapid acetylators, increased according to coffee or black tea consumption (0.05 < p < 0.1). Among lifestyle factors, two factors, i.e. smoking and the amount of coffee or black tea consumed, were related to urinary caffeine levels (p < 0.05). Geometric means of urinary caffeine levels were higher in the group who consumed higher amounts of coffee or black tea (p < 0.05) and those of smokers were lower than non-smokers- approximately 70% of non-smokers (p < 0.05). The genetic polymorphisms of NAT2, CYP1A1 and CYP2E1 were not significantly associated with the urinary caffeine levels according to each consumption level of coffee or black tea. This study suggests that smoking should be considered for the proper appreciation of individual differences in caffeine intake and urinary caffeine levels.  相似文献

A series of experiments were applied to test how leaf orientation within microcosms affect consumption rates (Experiment 1), and to discover intra-specific differences in leaf litter consumption (Experiment 2) of the common isopod species Porcellio scaber and Porcellionides pruinosus. A standardised microcosm setup was developed for feeding experiments to maintain standard conditions. A constant amount of freshly fallen black poplar litter was provided to three distinct size class (small, medium, large) of woodlice. We measured litter consumption after a fortnight. We maintained appr. constant isopod biomass for all treatments, and equal densities within each size class. We hypothesized that different size classes differ in their litter consumption, therefore such differences should occur even within populations of the species. We also hypothesized a marked difference in consumption rates for different leaf orientation within microcosms. Our results showed size-specific consumption patterns for Porcellio scaber: small adults showed the highest consumption rates (i.e. litter mass loss / isopod biomass) in high density microcosms, while medium-sized adults of lower densities ate the most litter in containers. Our objective was to determine whether the consumption of green tea, coffee, or black tea influences the incidence of dementia and mild cognitive impairment (MCI) in older people. We conducted a population-based prospective study with Japanese residents aged >60 years from Nakajima, Japan (the Nakajima Project). Participants received an evaluation of cognitive function and blood tests. The consumption of green tea, coffee, and black tea was also evaluated at baseline. Of 723 participants with normal cognitive function at a baseline survey (2007–2008), 490 completed the follow up survey in 2011–2013. The incidence of dementia during the follow-up period (mean ± SD: 4.9±0.9 years) was 5.3%, and that of MCI was 13.1%. The multiple-adjusted odds ratio for the incidence of overall cognitive decline (dementia or MCI) was 0.32 (95% CI: 0.16–0.64) among individuals who consumed green tea every day and 0.47 (95% CI: 0.25–0.86) among those who consumed green tea 1–6 days per week compared with individuals who did not consume green tea at all. The multiple-adjusted odds ratio for the incidence of dementia was 0.26 (95% CI: 0.06–1.06) among individuals who consumed green tea every day compared with those who did not consume green tea at all. No association was found between coffee or black tea consumption and the incidence of dementia or MCI. Our results indicate that green tea consumption is significantly associated with reduced risk of cognitive decline, even after adjustment for possible confounding factors.  相似文献

Wild animals substantially support crop production by providing ecosystem services, such as pollination and natural pest control. However, the strengths of synergies between ecosystem services and their dependencies on land-use management are largely unknown. Here, we took an experimental approach to test the impact of land-use intensification on both individual and combined pollination and pest control services in coffee production systems at Mount Kilimanjaro. We established a full-factorial pollinator and vertebrate exclosure experiment along a land-use gradient from traditional homegardens (agroforestry systems), shaded coffee plantations to sun coffee plantations (total sample size = 180 coffee bushes). The exclusion of vertebrates led to a reduction in fruit set of ca 9%. Pollinators did not affect fruit set, but significantly increased fruit weight of coffee by an average of 7.4%. We found no significant decline of these ecosystem services along the land-use gradient. Pest control and pollination service were thus complementary, contributing to coffee production by affecting the quantity and quality of a major tropical cash crop across different coffee production systems at Mount Kilimanjaro.  相似文献

Two independent studies have shown that the cell wall of pollen tubes from tobacco and tomato species contained fucosylated xyloglucan (XyG). These findings are intriguing as many reports have shown that XyG of somatic cells of these species is not fucosylated but instead is arabinosylated. In order to produce fucosylated XyG, plants must express a functional galactoside α-2-fucosyltransferase. Here, using a bioinformatics approach, we show that several candidate genes coding for XyG fucosyltransferases are present in the genome of coffee and several Solanaceae species including tomato, tobacco, potato, eggplant and pepper. BLAST and protein alignments with the 2 well-characterized XyG fucosyltransferases from Arabidopsis thaliana and Pisum sativum revealed that at least 6 proteins from different Solanaceae species and from coffee displayed the 3 conserved motifs required for XyG fucosyltransferase activity.  相似文献

Sublingual route has been widely used to deliver small molecules into the bloodstream and to modulate the immune response at different sites. It has been shown to effectively induce humoral and cellular responses at systemic and mucosal sites, namely the lungs and urogenital tract. Sublingual vaccination can promote protection against infections at the lower and upper respiratory tract; it can also promote tolerance to allergens and ameliorate asthma symptoms. Modulation of lung’s immune response by sublingual immunotherapy (SLIT) is safer than direct administration of formulations by intranasal route because it does not require delivery of potentially harmful molecules directly into the airways. In contrast to intranasal delivery, side effects involving brain toxicity or facial paralysis are not promoted by SLIT. The immune mechanisms underlying SLIT remain elusive and its use for the treatment of acute lung infections has not yet been explored. Thus, development of appropriate animal models of SLIT is needed to further explore its potential advantages. This work shows how to perform sublingual administration of therapeutic agents in mice to evaluate their ability to protect against acute pneumococcal pneumonia. Technical aspects of mouse handling during sublingual inoculation, precise identification of sublingual mucosa, draining lymph nodes and isolation of tissues, bronchoalveolar lavage and lungs are illustrated. Protocols for single cell suspension preparation for FACS analysis are described in detail. Other downstream applications for the analysis of the immune response are discussed. Technical aspects of the preparation of Streptococcus pneumoniae inoculum and intranasal challenge of mice are also explained.SLIT is a simple technique that allows screening of candidate molecules to modulate lungs’ immune response. Parameters affecting the success of SLIT are related to molecular size, susceptibility to degradation and stability of highly concentrated formulations.  相似文献

Coffee (Coffea arabica L.) is of economic importance worldwide. Its growth in organic-rich acidic soils is influenced by aluminium such that coffee yield may be impaired. Herein we have used the Al-sensitive C. arabica suspension cell line L2 to analyse the effect of two different Al species on the phosphoinositide signal transduction pathway. Our results have shown that the association of Al with coffee cells was affected by the pH and the form of Al in media. More Al was associated with cells at pH 4.3 than 5.8, whereas when Al was present as hydroxyaluminosilicates (HAS) the association was halved at pH 4.3 and unchanged at pH 5.8. Two signal transduction elements were also evaluated; phospholipase C (PLC) activity and phosphatidic acid (PA) formation. PLC was inhibited ( approximately 50%) when cells were incubated for 2 h in the presence of either AlCl(3) or Al in the form of HAS. PA formation was tested as a short-term response to Al. By way of contrast to what was found for PLC, incubation of cells for 15 min in the presence of AlCl(3) decreased the formation of PA whereas the same concentration of Al as HAS produced no effect upon its formation. These results suggest that Al is capable to exert its effects upon signal transduction as Al((aq))(3+) acting upon a mechanism linked to the phosphoinositide signal transduction pathway.  相似文献

In this article, we describe the mechanisms by which morphogens in the Xenopus embryo exert their long-range effects. Our results are consistent with the idea that signalling molecules such as activin and the nodal-related proteins traverse responding tissue not by transcytosis or by cytonemes but by movement through the extracellular space. We suggest, however, that additional experiments, involving real-time imaging of morphogens, are required for a real understanding of what influences signalling range and the shape of a morphogen gradient.  相似文献

For bacteria, the structural integrity of its cell wall is of utmost importance for survival, and to this end, a rigid scaffold called peptidoglycan, comprised of sugar molecules and peptides, is synthesized and located outside the cytoplasmic membrane of the cell. Disruption of this peptidoglycan layer has for many years been a prime target for effective antibiotics, namely the penicillins and cephalosporins. Because this rigid layer is synthesized by a multi-step pathway numerous additional targets also exist that have no counterpart in the animal cell. Central to this pathway are four similar ligase enzymes, which add peptide groups to the sugar molecules, and interrupting these steps would ultimately prove fatal to the bacterial cell. The mechanisms of these ligases are well understood and the structures of all four of these ligases are now known. A detailed comparison of these four enzymes shows that considerable conformational changes are possible and that these changes, along with the recruitment of two different N-terminal binding domains, allows these enzymes to bind a substrate which at one end is identical and at the other has the growing polypeptide tail. Some insights into the structure-function relationships in these enzymes is presented.  相似文献

Background, aim and scope

The evaluation of packaging’s environmental performance usually concentrates on a comparison of different packaging materials or designs. Another important aspect in life cycle assessment (LCA) studies on packaging is the recycling or treatment of packaging wastes. LCA studies of packed food include the packaging with specific focus on the contribution of the packaging to the total results. The consumption behaviour is often assessed only roughly. Packaging is facilitating the distribution of goods to the society. Broader approaches, which focus on the life cycle of packed goods, including the entire supply system and the consumption of goods, are necessary to get an environmental footprint of the system with respect to sustainable production and consumption.

Materials and methods

A full LCA study has been conducted for two food products: coffee and butter packed in flexible packaging systems. The aim was to investigate the environmental performance of packaging with respect to its function within the life cycle of goods. The study looks at the environmental relevance of stages and interdependencies within the life cycle of goods whilst taking consumers’ behaviour and portion sizes into consideration. The impact assessment is based on the following impact categories: non-renewable cumulative energy demand (CED), climate change, ozone layer depletion (ODP), acidification, and eutrophication.

Results

The study shows that the most relevant environmental aspects for a cup of coffee are brewing (i.e. the heating of water) and coffee production. Transport and retail packaging are of minor importance. Brewing and coffee production have an impact share between 40% (ODP, white instant coffee) and 99% (eutrophication, black coffee). Milk added for white coffee is relevant for this type of preparation. The instant coffee in the one-portion stick-pack needs more packaging material per cup of coffee and is prepared by a kettle with lower energy demand, such as a coffee machine, thus leading to higher shares of the retail packaging in all indicators. A one-portion stick-pack can prevent wastage and resources related to coffee production can be saved. The most relevant aspect regarding the life cycle of butter is butter production, dominated by the provision of milk. Over 80% of the burdens in butter production stem from the provision of milk for all indicators discussed. Regarding climate change, methane and dinitrogen monoxide, emissions of milk cows and fodder production are most relevant. Fertilisation during livestock husbandry is responsible for most burdens regarding acidification and eutrophication. The distribution and selling stage influences the indicators CED and ODP distinctly. The reasons are, on the one hand, the relatively energy-intensive storage in supermarkets and, on the other hand, the use of refrigerants for chilled storage and transportation. The storage of butter in a refrigerator for 30 days is responsible for about 10% of the CED.

Discussion

Several aspects have been modelled in a sensitivity analysis. The influence of coffee packaging disposal is very small due to the general low influence of packaging. In contrast, the brewing behaviour is highly relevant for the environmental impact of a cup of coffee. That applies similarly to the type of heating device—i.e. using a kettle or an automatic coffee machine. Wastage leads to a significant increase of all indicators. Under the wastage scenario, the coffee from one-portion stick-packs has a considerable better environmental performance concerning all indicators because, in case of instant coffee wastage of hot water and in case of ground coffee wastage of prepared coffee, has been predicted. Regardless of urban or countryside distances, grocery shopping has a low impact. The storage time of butter is relevant for the results in the indicator non-renewable CED. This is mainly the case when butter is stored as stock in the freezer. The end of life treatment of the packaging system has practically no influence on the results. Grocery shopping is of limited importance no matter which means of transport are used or which distances are regarded. Spoilage or wastage is of great importance: a spoilage/wastage of one third results in about 49% increased impacts compared to the standard case for all indicators calculated.

Conclusions

The most important factors concerning the environmental impact from the whole supply chain of a cup of coffee are the brewing of coffee, its cultivation and production and the milk production in case of white coffee. The study highlights consumer behaviour- and packaging-related measures to reduce the environmental impact of a cup of coffee. The most relevant measures reducing the environmental impacts of butter consumption are the optimisation of the milk and butter production. Another important factor is the consumers’ behaviour, i.e. the reduction of leftovers. The consumer can influence impacts of domestic storage using efficient and size-adequate appliances. The impacts of packaging in the life cycle of butter are not of primary importance.

Recommendations and perspectives

This study shows that, in the case of packaging industry, a reduction of relevant environmental impacts can only be achieved if aspects indirectly influenced by the packaging are also taken into account. Thus, the packaging industry should not only aim to improve the production process of their packages, but also provide packages whose functionality helps to reduce other more relevant environmental impacts in the life cycle such as, for example, losses. Depending on the product, tailor-made packaging may also help to increase overall resource efficiency.

Among the lactic acid bacteria, L. plantarum attracted many researchers because of its wide applications in the medical field with antioxidant, anticancer, anti-inflammatory, antiproliferative, anti-obesity and antidiabetic properties. The present study aimed to investigate the in vitro importance of L. plantarum toward medical applications. Moreover, this report short listed various reports related to the applications of this promising strain. The effects of temperature on embryogenesis and postinfection development in Meloidogyne konaensis were examined. Embryogenesis was evaluated at 5, 8, 10, 13, 16, 20, 22, 24, 26, 28, 30, 35, and 40 C. No embryonic development occurred at 5 C. Some development, although incomplete, occurred at 8 and 10 C. The rate of embryogenesis was linear from 13 to 30 C, but decreased at 35 C. The lowest egg mortality occurred at 24 C, whereas all eggs died within 24 hours at 40 C. Postinfection development was determined on coffee and tomato in a greenhouse at an average temperature of 30 C and in a growth chamber with a constant temperature of 26 C. Development of M. konaensis J2 to mature female required 38 and 48 days on coffee at 30 and 26 C, respectively. This process took 20 and 26 days, respectively, on tomato.  相似文献

In plants, PPO has been related to defense mechanism against pathogens and insects and this role was investigated in coffee trees regarding resistance against a leaf miner and coffee leaf rust disease. PPO activity was evaluated in different genotypes and in relation to methyl-jasmonate (Meja) treatment and mechanical damage. Evaluations were also performed using compatible and incompatible interactions of coffee with the fungus Hemileia vastatrix (causal agent of the leaf orange rust disease) and the insect Leucoptera coffeella (coffee leaf miner). The constitutive level of PPO activity observed for the 15 genotypes ranged from 3.8 to 88 units of activity/mg protein. However, no direct relationship was found with resistance of coffee to the fungus or insect. Chlorogenic acid (5-caffeoylquinic acid), the best substrate for coffee leaf PPO, was not related to resistance, suggesting that oxidation of other phenolics by PPO might play a role, as indicated by HPLC profiles. Mechanical damage, Meja treatment, H. vastatrix fungus inoculation and L. coffeella infestation caused different responses in PPO activity. These results suggest that coffee resistance may be related to the oxidative potential of the tissue regarding the phenolic composition rather than simply to a higher PPO activity.  相似文献

It has been reported that the gum resin of Boswellia serrata (BS), which has been shown to have antiinflammatory properties, might also have anticancer effects. This study examined the potential of BS as an anticancer agent. The BS extract induces apoptosis in HeLa human cervical carcinoma cells, as confirmed by two apoptosis analyses, Hoechst staining and Annexin V/PI assay. Among the apoptosis pathways, the ER stress-associated mechanism was examined to determine its role in BS-induced apoptosis. The expression of GRP78 and CHOP, which are representatives of the ER stress proteins, and the calcium-binding protein-calpain were determined. The results showed significantly higher levels of both GRP78 and CHOP, and stronger calpain activity in the BS-treated cells than in the control cells. This shows that there is a correlation between ER stress signaling and apoptosis, which suggests the possibility of the BS-ER stress initiator as an anticancer therapeutic agent in human cervical carcinoma.  相似文献

Background and Aims Diaspores of heteromorphic species may germinate at different times due to distinct dormancy-breaking and germination requirements, and this difference can influence life history traits. The primary aim of this study was to determine the effect of germination time of the two seed morphs of Suaeda corniculata subsp. mongolica on life history traits of the offspring.Methods Germinated brown and black seeds were sown on the 20th of each month from April to September in a simulated but near-natural habitat of the species. Phenological and vegetative traits of the maternal plants, and number, size and germination percentage of the offspring were determined.Key Results Germinated seeds sown late in the year produced smaller plants that had a higher proportion of non-dormant brown than dormant black seeds, and these brown seeds were larger than those produced by germinated seeds sown early in the year. The length of the seedling stage for brown seeds was shorter than that for black seeds, and the root/shoot ratio and reproductive allocation of plants from brown seeds were more variable than they were for plants from black seeds. Late-germinating brown seeds produced larger plants than late-germinating black seeds.Conclusions Altering the proportion of the two seed types in response to germination timing can help alleviate the adverse effects of delayed germination. The flexible strategy of a species, such as S. corniculata, that produces different proportions of dimorphic seeds in response to variation in germination timing may favour the maintenance and regeneration of the population in its unpredictable environment.  相似文献

A cup of instant coffee (150 ml) of normal strength (15 mg/ml) was found to contain about 500 and 750 micrograms of hydrogen peroxide soon after its preparation at 37 degrees C and 80 degrees C, respectively, but the concentration of hydrogen peroxide in the coffee increased with time for up to 24 h after its preparation. Thus coffee contains a hydrogen peroxide generating system. As extracts of green coffee beans were found to have very low capacity to generate hydrogen peroxide, this generating system is produced by roasting coffee beans. Hydrogen peroxide itself was only weakly mutagenic to Salmonella typhimurium TA100, but in the presence of methylglyoxal, which is also present as a mutagenic component in coffee, hydrogen peroxide showed strong mutagenicity. Hydrogen peroxide and methylglyoxal seem to be responsible for most of the mutagenicity of instant coffee.  相似文献

Background

Isodon rugosus is used traditionally in the management of hypertension, rheumatism, tooth-ache and pyrexia. Present study was arranged to investigate I. rugosus for phytoconstituents, phytotoxic and cytotoxic activities to explore its toxicological, pharmacological potentials and to rationalize its ethnomedicinal uses. Briefly, qualitative phytochemical analysis of the plant extracts were carried out for the existence of alkaloids, flavonoids, saponins, oils, glycosides, anthraquinones, terpenoids, sterols and tannins. Plant crude methanolic extract (Ir.Cr), its subsequent fractions; n-hexane (Ir.Hex), chloroform (Ir.Chf), ethyl acetate (Ir.EtAc), aqueous (Ir.Aq) and saponins (Ir.Sp) in different concentrations were tested for phytotoxic and cytotoxic activities using radish seeds and brine shrimps (Artemia salina) respectively. The phytotoxic activity was determined by percent root length inhibition (RLI) and percent seeds germination inhibition (SGI) while the cytotoxicity was obtained with percent lethality of the brine shrimps.

Results

Ir.Cr was tested positive for the presence of alkaloids, glycosides, flavonoids, oils, terpenoids, saponins, tannins and anthraquinones. Among different fractions Ir.Sp, Ir.Chf, Ir.EtAc, and Ir.Cr were most effective causing 93.55, 89.32, 81.32 and 58.68% inhibition of seeds in phytotoxicity assay, with IC50 values of 0.1, 0.1, 0.1 and 52 μg/ml respectively. Similarly, among all the tested samples, Ir.Sp exhibited the highest phytotoxic effect causing 91.33% root length inhibition with IC50 of 0.1 μg/ml. Ir.Sp and Ir.Chf were most effective against brine shrimps showing 92.23 and 76.67% lethality with LC50 values of 10 and 12 μg/ml respectively.

Conclusions

It may be inferred from the current investigations that I. rugosus contains different secondary metabolites and is a potential source for the isolation of natural anticancer and herbicidal drug molecules. Different fractions exhibited phytotoxic and cytotoxic activities, thus providing pharmacological basis for ethnomedicinal uses of this plant.  相似文献

Products of convenience are playing an increasingly large role in today's society. These products provide a competitive advantage over their conventional counterparts by requiring less time and effort to produce a similar service or experience. At the same time, these products are often also more materials intensive to produce and create a greater amount of waste. A comparative midpoint life cycle assessment of different coffee brewing systems is presented in order to explore the comparative impact of three different systems: drip filter, french press, and pod style (a product of convenience). Utilizing a comparative functional unit, the drip filter system method was found to have the greatest environmental impact in all impact categories, whereas the pod style had the least in six of the impact categories (with the french press having the least in two of the impact categories, and a tie between pod style and french press in a single impact category). This suggests that contrary to popular belief, the pod style coffee may be the more environmentally friendly option. The two most significant contributors to environmental impact in all of the categories considered was the amount of dry coffee utilized and the energy needed to brew the coffee, although in some categories considered transportation was also significant. There is the potential for the environmental impact of coffee brewing to shift if coffee wastage occurs (likely in the case of the drip filter and french press system) or if substantial changes in materials or energy consumption were to occur (in the case of the pod‐style brewing system). From the perspective of industrial ecology, this analysis suggests that, in regard to products of consumer convenience, the convenient alternative may not have a significantly greater environmental impact than its conventional counterpart, and that it may be time to question that often‐held assumption.  相似文献

Anticancer drugs exert their effects on cancer cells by deregulating many pathways linked to cell cycle, apoptosis, etc. but cancer cells gradually become resistive against anticancer drugs, thereby necessitating the development of newer generation anticancer molecules. N-end rule pathway has been shown to be involved in the degradation of many cell cycle and apoptosis-related proteins. However, the involvements of this pathway in cancer are not well established. Recently, we developed a non-peptide-based N-end rule pathway inhibitor, RF-C11 for type 1 and 2 recognition domains of E3 ubiquitin ligases. The inhibitor significantly increased the half-life of potential N-degrons leading to significant physiological changes in vivo. We hypothesized RF-C11 may be used to decipher the N-end rule pathway's role in cancer towards the development of anticancer therapeutics. In this study, we showed that RF-C11, barring noncancer cells, significantly sensitizes cancer cells towards different anticancer agents tested. We further find that the profound cellular sensitization to anticancer drugs was affected by (a) downregulation of X-linked inhibitor of apoptosis protein, an antiapoptotic protein and (b) by stabilization of RAD21, and thereby inhibiting metaphase to anaphase promotion. The study shows that RF-C11 or its analogs may be used as a novel additive in combination therapy against cancer.  相似文献

Whitefly, Bemisia tabaci (Gennadius) (Hemiptera: Aleryrodidae), is a serious pest of black gram, (Vigna mungo (L.) Hepper), an important legume pulse crop grown in north India. This research investigated the potential role of selected plant oxidative enzymes in resistance/susceptibility to whitefly in nine black gram genotypes. Oxidative enzyme activity was estimated spectrophotometrically from leaf samples collected at 30 and 50 d after sowing (DAS) from whitefly infested and uninfested plants. The enzymes showed different activity levels at different times after the infestation. The results indicated that in general, whitefly infestation increased the activities of peroxidase and decreased the catalase activity. Resistant genotypes NDU 5-7 and KU 99-20 recorded higher peroxidase and catalase activities at 30 and 50 DAS under whitefly-stress conditions as compared with non-stressed plants. The results suggest that the enhanced activities of the enzymes may contribute to bioprotection of black gram plants against B. tabaci infestation. The potential mechanisms to explain the correlation of resistance to whitefly in black gram genotypes with higher activities of oxidative enzymes are also discussed.  相似文献
