American lobsters Homarus americanus were inoculated with a field isolate of the Gram-positive bacterium Aerococcus viridans var. homari, causative agent of gaffkemia, at 1 x 10(6), 1 x 10(4) or 1 x 10(2) colony forming units (CFU) kg(-1) or with sterile 3% NaCl and maintained at 10 or 15 degrees C until they died or were euthanised. Progression of disease in individual animals was monitored daily by total haemocyte count (THC) and haemolymph culture. Post-mortem examinations were performed on all lobsters. Effects of both ambient temperature and infective dose on survival time were observed. Marked bacteraemia occurred in all mortalities. Haemocytopenia (THC < 10 x 10(9) cells l(-1)) preceded death in most, but not all, mortalities. 相似文献

Virulent and avirulent strains of Aerococcus viridans (var.) homari were used to extend previous studies to determine and confirm differences between the 2 types. Virulent strains possessed polysaccharide capsules and were not agglutinated by lobster hemolymph serum; avirulent strains did not have capsules, were agglutinated by the lobster hemolymph serum, and most did not grow well in lobster hemolymph serum. Growth of the avirulent strains in sterile lobster hemolymph serum induced the production of capsules (which reached a maximum after 5 to 7 d incubation), eliminated susceptibility of the strains to the lobster serum agglutinin, and restored their virulence against lobsters. The factor(s) in lobster hemolymph serum inducing the long-lasting phenotypic response of virulence was (were) heat labile. 相似文献

Hemocytes of the American lobster (Homarus americanus H. Milne Edwards) were classified after examination of Wright-Giemsa stained cytocentrifuge preparations by brightfield light microscopy. Eleven hemocyte types were identified using morphologic criteria. The classification system was then used to monitor changes in the differential hemocyte count (DHC) of lobsters infected with the Gram positive coccus Aerococcus viridans var. homari, etiologic agent of gaffkemia. The appearance of less mature hemocytes in the DHCs of lobsters in the late stages of infection was similar to the 'left shift' of vertebrate inflammation. Results from this study suggest that DHCs can be used to assess and characterize inflammation in H. americanus and possibly other crustaceans. 相似文献

The Canadian lobster industry holds lobsters Homarus americanus in captivity for various periods to supply markets with live product year-round. Mortality during holding results in considerable losses, estimated at 10 to 15 % yr(-1) by the industry. This study examined the prevalence of Anophryoides haemophila and Aerococcus viridans, causative agents of 'bumper car' disease and gaffkemia, respectively, in lobsters freshly captured in the waters of Prince Edward Island during the spring and fall fishing seasons of 1997. A total of 116 lobsters were sampled in the spring, and 138 in the fall. A. haemophila was not detected in the spring, while the prevalence was 0.72 % in the fall with a 95% confidence interval (CI) of 0.02 to 3.97% and an overall prevalence of 0.39% (95% CI: 0.01 to 2.17%). The prevalence of A. viridans was estimated at 6.9% (95% CI: 3.0 to 13.14%) in the spring, 5.8% in the fall (95% CI: 2.54 to 11.10%), and 6.30% overall (95% CI: 3.64 to 10.03%). Because of the reduced interest in food of diseased lobsters, and compromised metabolism in the case of gaffkemia, these prevalence estimates are likely underestimates of the true prevalence of gaffkemia and 'bumper car' disease in the wild populations of lobster around Prince Edward Island. 相似文献

A combination of 16S rRNA sequencing and random amplified polymorphic DNA (RAPD) analysis was used to evaluate the genetic diversity within Aerococcus viridans var. homari, the causative agent of gaffkemia in lobsters. A collection of 7 A. viridans var. homari strains and 2 avirulent A. viridans-like cocci isolated from homarid lobsters harvested from different regions on the Atlantic Coast of North America were analyzed. The isolates are separated geographically and temporally between the years 1947 and 2000. Sequencing of 16S rRNA genes confirmed the inclusion of all 9 isolates in the monophyletic A. viridans clade (99.8 to 100% similarity). RAPD analysis revealed that the 9 A. viridans var. homari isolates could be separated into 2 distinct subtypes. Subtype 1 included the 7 pathogenic lobster isolates and constituted a homogeneous group regardless of their geographical, temporal or virulence differences. Subtype 2 contained the 2 avirulent A. viridans-like cocci that had distinct RAPD patterns and clustered separately with the non-marine A. viridans. RAPD analysis represented a useful method for determining molecular subtyping for the intraspecific classification and epidemiological investigations of A. viridans var. homari. 相似文献

The American lobster fishery is a significant economic driver in coastal communities of North America. Increasingly, the impacts of infectious disease are recognized as important components and factors in the population ecology and subsequent management of the lobster fishery. Both environmental and anthropogenic factors impact marine diseases. The review herein highlights aspects of several important bacterial, fungal and protistan diseases, including gaffkemia, shell disease, vibriosis, disease caused by species of Lagenidium, Haliphthoros and Fusarium, paramoebiasis and Bumper Car disease. As the global environment continues to change, these diseases could more severely affect both wild caught and impounded lobsters. 相似文献

Larval and postlarval lobsters, injected with human somatotropin (STH, or growth hormone), grew at a more rapid rate than untreated control animals over succeeding molts. Their mean carapace length was significantly longer than that of controls and they were heavier, although the moisture content of treated and control animals was similar. Injected STH increased the growth rate of individual animals by 10 to 20%. This is the first evidence for a growth enhancing effect of human somatotropin on a Crustacean. 相似文献

This study provides a detailed account of the development of the digestive system of larval lobsters (Homarus americanus H. Milne Edwards, 1837) and the morphological changes that occur at metamorphosis. The most dramatic of these changes involves the gastric mill of the cardiac stomach. First-and second-stage lobsters lack the medial and lateral teeth characteristic of the grinding stomach of adult lobsters. Clearly recognizable, heavily cuticularized teeth first appear in the third stage, and accessory lateral teeth do not appear until the fourth stage. In place of the teeth of the gastric mill, first- and second-stage stomachs have a series of pads and ridges which are the apparent rudiments of the teeth. The development of the gastric mill during the larval stages enables lobsters to deal successfully with the more substantial food they encounter in the benthic environment, and corresponds to the drastic change of habitat and diet which occurs at metamorphosis. Confusion about the extent of the midgut and hindgut in larval lobsters has been clarified. The results of this study have shown that the larvae have a long midgut, which lacks a cuticle, and a short hindgut with a cuticular lining, just as in adult lobsters. The junction between midgut and hindgut lies in the sixth abdominal segment in all of the first four stages, as well as in the adult. 相似文献

Three experiments were conducted to determine (1) the pharmacodynamics of 5-hydroxytryptamine in juvenile lobsters; (2) the
effects of 5-hydroxytryptamine, using a range of dosages, on a motor behavior used to escape an aversive situation; and (3)
the effect of doses that did and did not inhibit this motor behavior on measures of dominance and shelter competition. The
fate of 5-hydroxytryptamine in hemolymph over a 60-min post-injection period showed that the concentration fell rapidly to
a low plateau that was maintained for at least 1 h. Low doses of 5-hydroxytryptamine did not affect locomotor behavior, but
higher doses inhibited it. Dominance and subsequent possession of a shelter were unaffected by a low dose of 5-hydroxytryptamine
but a higher dose that inhibited locomotion resulted in lobsters that lost fights and did not secure or retain possession
of the shelter. In the context of dominance and shelter competition, we were unable to demonstrate any advantage of the low
dose of exogenous 5-hydroxytryptamine and a severe disadvantage with the higher dose. Previous reports of transient increases
in aggression in 5-hydroxytryptamine-treated subordinate lobsters did not take into account motor inhibition as a possible
critical variable in aggression.

In many disparate taxa, including crayfish, a freshwater decapod crustacean, the presence of one's offspring has been shown to be an important variable in the level of maternal aggression. Ovigerous American lobsters (Homarus americanus) show a territorial advantage against nonmaternal females, even though no posthatch care is provided. The eggs are attached to the pleopods (swimmerets) throughout embryogenesis. We evaluated the effect of stripping ovigerous H. americanus females of their eggs on maternal territoriality. In one treatment group, maternal females were stripped of all eggs. Twelve days later, including being individually isolated for an additional 48 h in a test tank, each resident was serially intruded upon by 4 non-maternal conspecific females. An identically treated control group of ovigerous female residents was left intact prior to the serial intrusions. Various biochemical parameters of the stripped and unstripped animals were measured before and after the experimental treatment (or control). The behavioral modulators measured were the biogenic amines, octopamine and serotonin, and the stress indicators were a heat-shock protein (HSP70), methyl farnesoate, and crustacean hyperglycemic hormone. The intact ovigerous residents showed a significant territorial advantage over the intruders, whereas the contest outcomes of stripped residents were reduced to chance. The presence of eggs in maternal H. americanus is therefore necessary for the maintenance of the shelter-related territorial advantage. However, no significant differences in any of the biochemical parameters were observed between or within treatment conditions. It appears that these amines are not prominently involved in the mechanism of maternal aggression, and that these changes in territorial defense are not simply due to changes in general stress as a result of stripping the females of their eggs. 相似文献

Changes in the heart rates of lobsters (Homarus americanus) were used as an indicator that the animals were capable of sensing a reduction in the salinity of the ambient seawater. The typical response to a gradual (1 to 2 ppt/min) reduction in salinity consisted of a rapid increase in heart rate at a mean threshold of 26.6 +/- 0.7 ppt, followed by a reduction in heart rate when the salinity reached 22.1 +/- 0.5 ppt. Animals with lesioned cardioregulatory nerves did not exhibit a cardiac response to changes in salinity. A cardiac response was elicited from lobsters exposed to isotonic chloride-free salines but not to isotonic sodium-, magnesium- or calcium-free salines. There was little change in the blood osmolarity of lobsters when bradycardia occurred, suggesting that the receptors involved are external. Furthermore, lobsters without antennae, antennules, or legs showed typical cardiac responses to low salinity, indicating the receptors are not located in these areas. Lobsters exposed to reductions in the salinity of the ambient seawater while both branchial chambers were perfused with full-strength seawater did not display a cardiac response until the external salinity reached 21.6 +/- 1.8 ppt. In contrast, when their branchial chambers were exposed to reductions in salinity while the external salinity was maintained at normal levels, changes in heart rate were rapidly elicited in response to very small reductions in salinity (down to 29.5 +/- 0.9 ppt in the branchial chamber and 31.5 +/- 0.3 ppt externally). We conclude that the primary receptors responsible for detecting reductions in salinity in H. americanus are located within or near the branchial chambers and are primarily sensitive to chloride ions. 相似文献

Oxygen consumption and ammonia excretion rates of juvenile lobsters spanning the size range from shelter-restricted to vagile (7-60 mm CL, carapace length) were measured. There are no significant size-dependent break points in the slope of oxygen consumption or nitrogen excretion rates versus weight in freshly caught wild or laboratory fed lobsters. This suggests that there are no inherent changes in energy metabolism in juvenile lobster in the 7-60 mm CL range (stage V to adolescent lobsters), despite major behavioral shifts related to foraging and shelter use. In addition, the O : N ratio of freshly caught wild lobsters (15.7+/-7.4, mean+/-S.D.) is not significantly different from that of lobsters fed brown algae (13.4+/-4.9) or a mixed carbohydrate/protein diet (13.7+/-6.8) in lab. The O : N ratio of wild lobsters is significantly higher than lobsters fed a high protein diet (7.5+/-2.5) in the lab. This suggests that carbohydrates, particularly algal-derived carbohydrates, make up a significant portion of the metabolic fuel of wild juvenile lobsters from settlement through adolescence. 相似文献

The lobster mandibular organ is well vascularized and its polygonal cells are arranged loosely around blood vessels and blood sinuses. Numerous mitochondria and microbodies (peroxisomes) give the acidophilic cytoplasm a finely granular appearance, but there is no evidence of secretory granules. The abundant endoplasmic reticulum is almost entirely agranular and occurs in two morphologically distinct forms: tubular and cisternal. The tubular reticulum is randomly distributed and may represent the site of synthesis and transport of the mandibular organ product. The cisternal reticulum is frequently associated with microbodies. Both forms of endoplasmic reticulum proliferate during mid to late premolt. Mandibular organ ultrastructure closely resembles that of cells known to synthesize steroids or lipids, which suggests that this organ may have a similar function. There is no functional evidence of involvement in molt control in Homarus, but ultrastructural and other evidence suggests an analogy with insect corpus allatum. 相似文献
