While a large number of studies have examined the effects of increased ultraviolet-B radiation (UV-B) on growth and physiological function of plants, UV-B effects on pollination success and fitness are poorly understood. To examine this question, we measured growth, timing of flowering, pollination success, production of pollen, ovules, flowers, fruits, and seeds, and quality of offspring produced by Brassica nigra and B. rapa in a garden experiment. A total of 313 plants of the two species were randomly divided into two treatment groups. One group received only natural ambient levels of UV-B, while the other received an artificially enhanced UV-B dose. Fitness of B. nigra declined at the higher UV-B dose while B. rapa fitness did not change. One possible cause of this result was a shift in the relative attractiveness of the two species to pollinators: visitation to B. nigra declined at the high UV-B dose while B. rapa visitation increased. Seedlings of two Mediterranean plants, the slow-growing, evergreen sclerophyll Ceratonia siliqua L. and the fast growing drought semi-deciduous Phlomis fruticosa L., were grown for one year in the field at ambient or ambient plus supplemental UV-B radiation (equivalent to a 15% ozone depletion) and two levels of applied fertilizers (NPK). The effects on growth, morphological, anatomical and physiological parameters were measured at final plant harvest. Additional nutrients increased leaf nitrogen, improved growth and reduced the root/shoot ratio in both plants, yet these effects were more pronounced in the fast growing P. fruticosa. A nutrient-induced increase in chlorophyll content was also observed in this plant. The growth responses to UV-B radiation were different for the two species. Growth in C. siliqua was not affected by UV-B radiation at both nutrient levels and the same was true for P. fruticosa at low nutrients. However, at the high nutrient level, supplemental UV-B radiation improved growth in P. fruticosa, indicating a strong interaction between the treatments. Photosystem II (PSII) photochemical efficiency, methanol-extractable UV-B absorbing capacity, total phenolics and tannins were not affected by either treatment in both plants. It is concluded that nutrient levels can strongly modify the UV-B radiation effects on growth of P. fruticosa. We presume that this may be correlated to the fast growing habit of this species. 相似文献

The evolution of dry stigmas has been accompanied by the development — in the pollen — of mechanisms for accessing water from the stigmatic epidermis. Development of self- and cross-pollen on the stigmatic surface has been examined in Brassica oleracea, focusing on the hydration of the grains. Unlike self-compatible (SC) Arabidopsis thaliana, pollen hydration of self-incompatible (SI) Brassica oleracea is preceded by a latent period of between 30–90 min, which is significantly shortened by inhibition of protein synthesis in the stigma. Physiological experiments, some with isolated pollen coatings, indicate that during the latent period signals passing from the pollen to the sigma are responsible for readying the stigmatic surface for penetration and — after self-pollination — activation of the SI system. The changes at the stigma surface include the expansion of the outer layer of the cell wall beneath the grain. This expansion does not occur following self-pollination, when coating-derived signals stimulate a stigmatic response which interrupts hydration and arrests grain development. Cell manipulation studies suggest that self grains are not inhibited metabolically, but are physiologically isolated from the subjacent stigmatic papilla. This focusing of the SI response at the pollen-stigma interface ensures that a single papilla can simultaneously accept cross-pollen and reject self-grains. The evolution of this highly efficient SI system is disussed in the perspective of pathogen-defence mechanisms known also to be located in epidermal cells. 相似文献

The effects of enhanced UV-B radiation on three examples of European shrub-dominated vegetation were studied in situ. The experiments were in High Arctic Greenland, northern Sweden and Greece, and at all sites investigated the interaction of enhanced UV-B radiation (simulating a 15% reduction in the ozone layer) with artificially increased precipitation. The Swedish experiment also involved a study of the interaction between enhanced UV-B radiation and elevated CO2 (600 ppm). These field studies were supported by an outdoor controlled environment study in the United Kingdom involving modulated enhancement of UV-B radiation in combination with elevated CO2 (700 ppm). Effects of the treatments on plant growth, morphology, phenology and physiology were measured. The effects observed were species specific, and included both positive and negative responses to the treatments. In general the negative responses to UV-B treatments of up to three growing seasons were small, but included reductions in shoot growth and premature leaf senescence. Positive responses included a marked increase in flowering in some species and a stimulation of some photosynthetic processes. UV-B treatment enhanced the drought tolerance of Pinus pinea and Pinus halepensis by increasing leaf cuticle thickness. In general, there were few interactions between the elevated CO2 and enhanced UV-B treatments. There was evidence to suggest that although the negative responses to the treatments were small, damage may be increasing with time in some long-lived woody perennials. There was also evidence in the third year of treatments for effects of UV-B on insect herbivory in Vaccinium species. The experiments point to the necessity for long-term field investigations to predict the likely ecological consequences of increasing UV-B radiation. 相似文献

Summary A technique for cultivating isolated flowers of Brassica napus has been developed. Flowers were harvested at anthesis, the surface of their peduncles was then sterilized and they were cultivated in a hormonefree medium. We used an MS medium supplemented with 3% sucrose as a source of organic carbon. From our experiments, it was concluded that no exogenous growth regulator is required to ensure normal growth and development in vitro. The flowers, and thereafter the pods, can be kept in culture until seed maturity. After 30 days, seed development resulted in three types of seeds: (1) normal, (2) milky and (3) aborted. The results show that the number of seeds per pod was not dependent on the order of flowers on the raceme (except the first 10 flowers and flowers above row 50). Our study supports the validity of this model as an easy tool for studying pollination and early seed development. 相似文献

Genetically fixed differences in the demands for optimal pollen germination among species were found. The experiments were designed to define optimal content of mineral salts, sugar, and PEG for every investigated species or hybrids. The morphologically diverse flowers in the genusSatyrium reflect adaptations to a wide range of pollinators. Several recently discovered pollination systems inSatyrium are described and illustrated here; these include pollination by solitary bees, carrion flies, butterflies and moths. Two basic types of floral mechanism are recognised inSatyrium: (1) Species pollinated by lepidoptera and birds have long floral spurs and plate-like viscidia seated in lateral rostellum notches; these viscidia become attached to the proboscis or bill of the pollinator. We report a long-term experiment on the photosynthetic response of natural vegetation of Deschampsia antarctica (Poaceae) and Turgidosculum complicatulum (Lichenes) to altered UV-B levels on Léonie Island, Antarctica.UV-B above the vegetation was reduced by filter screens during two seasons. Half of the screens were transparent to UV-A and UV-B (ambient treatment) or absorbing UV-B and part of the UV-A (below-ambient treatment). Half of the wedge- shaped filters had side walls leading to an enhancement of the daily mean temperature in summer by 2–4 °C, simulating rising mean air temperature on the Antarctic Peninsula. The other half of the filters were without side walls resulting in close-to-ambient temperature underneath. Plots without filters served as controls.UV-B supplementation of an extra 1.3 kJ UV-BBE was achieved using UV-mini-lamp systems during 15 days in the second season.We found no evidence that altered incident UV-B levels and temperature had an effect on maximum photosystem II efficiency (F
v/F
m) and effective photosystem II efficiency (F/F
m) in both species. UV-B reduction did not influence contents of chlorophyll, carotenoids and methanol-soluble UV absorbing compounds in D. antarctica.Flowering shoot length of D. antarctica was not affected by UV-B reduction. Temperature enhancement tended to result in longer inflorescence axes. Results of two austral summer seasons of UV- reduction in natural stands of D. antarctica and T. complicatulum suggest that current ambient levels of UV-B do not have a direct effect on the photosynthetic performance and pigment contents of these species. The effects of enhanced UV-B radiation on hormone changes in vegetative and reproductive tissues of tomato (Lycopersicon esculentum Mill.) and their relationships with reproductive characteristics were studied. Two cultivars, TongHui (TH) and XiaGuang (XG), were grown in the field for one growing season under ambient (Control), ambient plus 2.54 kJ m–2 d–1 (T1) or ambient plus 4.25 kJ m–2 d–1 (T2) of supplemental ultraviolet-B (280–320 nm). The number of open flowers increased significantly in the TH cultivar under T2 while it declined in the XG cultivar under T1. Although pollen germination from both cultivars was inhibited by UV-B treatment, fruit number was enhanced in the TH cultivar at both UV-B doses and in the XG cultivar at the low dose. On the other hand, seed size (dry weight) was reduced in the XG cultivar by both UV-B doses and in the TH cultivar at the low UV-B dose. The final germination rates of seeds from control and UV-B treated plants of both cultivars showed no significant differences (p > 0.05), while germination was delayed in the TH cultivar at both doses of UV-B and in the XG cultivar only for T2. To determine the mechanism of UV-B's effects on developmental processes, hormone concentrations in leaves, pistils and seeds were analyzed using ELISA on partially purified extracts. The results suggested that enhanced UV-B radiation induced hormone changes in both vegetative and reproductive tissues. The alteration of flower number may be associated with the changes of ZR in leaves under enhanced UV-B radiation and the delayed germination may due to the changes in seed ABA and GAs. 相似文献

The winter annual species Anthemis arvensis L. (Asteraceae) was grown for 3.5 months in the field under ambient or ambient plus supplemental UV-B radiation, simulating a 15% ozone depletion over Patras (38.3° N, 29.1° E). Enhanced UV-B radiation had no effect on the methanol extractable UV-B absorbing capacity of leaves, phenological and morphometric parameters of anthesis (flowering time, anthesis duration, head life span, number of heads per plant, number of tubular and ligulate florets per head, area per ligulate floret). Concerning the optical properties of heads, enhanced UV-B radiation had no significant effect on the extractable absorbance of both floret types nor on the spectral reflectance of the tubular florets. However, under UV-B supplementation the white ligulate florets exhibited a slight, statistically significant decrease of reflectance in the visible region of the spectrum. This may be due to structural changes of the floret surface, since microscopic examination under SEM revealed the papillae of the adaxial epidermal cells to be swollen. The above ground dry mass measured at plant harvest was not affected but a significant increase in root biomass (and accordingly in root/shoot ratio) was observed. We conclude that Anthemis arvensis is resistant against UV-B radiation damage. The possible consequences of UV-B induced structural changes on floret epidermis are discussed. 相似文献

The surface structure and composition of surface lipids were examined in leaves of barley, bean, and cucumber seedlings grown in a growth chamber under white light and low levels of ultraviolet (UV-B; 280–320 nm) radiation. The cuticular wax of cucumber cotyledons and bean leaves appeared as a thin homogeneous layer, whereas on barley leaves crystal-like structures could be observed under these irradiation conditions. Principally, the amount of cuticular wax found in barley leaves was five times greater than in bean or cucumber leaves. The prediominant wax components were primary alcohols in barley, primary alcohols and monoesters in bean, and alkanes in cucumber cotyledons. Irradiation with enhanced UV-B levels caused an increase of total wax by about 25% in all plant species investigated. Aldehydes, detected as a minor constituent of cucumber and barley wax, increased twofold. Distribution patterns of the homologs within some wax classes were different at low and enhanced UV-B levels. In general, the distribution of the homologs was shifted to shorter acyl chain lengths in wax of leaves exposed to enhanced UV-B levels. This was most apparent in cucumber wax, less in bean or barley wax. The UV-B-caused effects upon cucumber wax were mainly due to a response by the adaxial surface of the leaf.Abbreviation UV-B

Summary A structural study of pollination in the dimorphic flowers ofCollomia grandiflora, a cleistogamous species, reveals significant differences in stigma behavior during pollination, stylar structure, the timing of generative cell division, and pollen tube growth rate patterns. The cleistogamous flower shows a loss of protandry and the stigma is receptive only after reflexing and closing of its lobes. In contrast, the chasmogamous stigma is receptive when reflexed and closes when pollen has been deposited on the lobes. Pollen tube penetration of the dry stigma papillae and entry into the style is similar in the two morphs. The chasmogamous style is solid and the cleistogamous style partly hollow. The matrix of secretion produced by the transmitting tract cells is mainly carbohydrate with a trace of lipids. It is fibrillar in nature and appears to be partly comprised of wall material from the transmitting tract cells. In the chasmogamous pollen, the generative cell enters the tube before division, which occurs between 30 and 60 min after pollination. This division correlates with an increased growth rate for the pollen tube. In the cleistogamous pollen, contact with the stigma triggers generative cell division inside the hydrated pollen grain before germination. The two resulting sperm cells exit the grain 15–30 min after pollination when the pollen tube is in the stigma lobes. The cleistogamous pollen tube shows only one phase of growth which occurs at a rate similar to that of the slow, first phase of the chasmogamous pollen.Abbreviations CH

The effects of dominance rank and group size on female lifetime reproductive success in wild long-tailed macaques,Macaca fascicularis 总被引:5,自引:0,他引:5
Demographic changes were recorded throughout a 12-year period for three social groups ofMacaca fascicularis in a natural population at Ketambe (Sumatra, Indonesia). We examined the prediction that females' lifetime reproductive success
depended on dominance rank and group size. Average birth rate was 0.53 (184 infants born during 349 female years). For mature
females (aged 8–20 yr) birth rate reflected physical condition, being higher in years with high food availability and lower
in the year following the production of a surviving infant. High-ranking females were significantly more likely than low-ranking
ones to give birth again when they did have a surviving offspring born the year before (0.50 vs 0.26), especially in years
with relatively low food availability (0.37 vs 0.10). Controlled comparisons of groups at different sizes indicate a decline
in birth rate with rroup size only once a group has exceeded a certain size. The dominance effect on birth rate tended to
be strongest in large groups.
Survival of infants was rank-dependent, but the survival of juveniles was not. There was a trend for offspring survival to
be lower in large groups than in mid-sized or small groups. However, rank and group size interacted, in that rank effects
on offspring survival were strongest in large groups. High-ranking females were less likely to die themselves during their
top-reproductive years, and thus on average had longer reproductive careers.
We estimated female lifetime reproductive success based on calculated age-specific birth rates and survival rates. The effects
of rank and group size (contest and scramble) on birth rate, offspring survival, age of first reproduction for daughters,
and length of reproductive career, while not each consistently statistically significant, added up to substantial effects
on estimated lifetime reproductive success. The group size effects explain why large groups tend to split permanently.
Since females are philopatric in this species, and daughters achieve dominance rank positions similar to their mother, a close
correlation is suggested between the lifetime reproductive success of mothers and daughters. For sons, too, maternal dominance
affected their reproductive success: high-born males were more likely to become top-dominant (in another group). These data
support the idea that natural selection has favored the evolution of a nepotistic rank system in this species, even if the
annual benefits of dominance are small. 相似文献

Oleosins, which are structural proteins on the surface of intracellular oil bodies, have been found in the sporophytic seeds of angiosperms. Here, we report an oleosin from the female gametophyte of gymnosperm Pinus ponderosa Laws, seed and another oleosin from the male gametophyte of Brassica napus L. With the pine seed gametophyte, we identified two putative oleosins of 15 and 10 kDa, which are similar to the oleosins in angiosperm seeds in terms of their presence in the oil bodies in massive quantity. The complete sequence of the cDNA encoding the gametophytic 15-kDa oleosin was obtained, and it has a predicted amino-acid sequence similar to those of oleosins in angiosperm sporophytic seeds. A Brassica napus pollen cDNA sequence, which was reported earlier, would encode an amino-acid sequence somewhat similar to those of seed oleosins. We tested if the dissimilarity signifies a substantially different oleosin in the Brassica male gametophyte or an analytic error. By direct sequencing of a polymerase chain reaction (PCR)-amplified fragment of genomic DNA, we obtained evidence showing that this reported dissimilarity is likely to have arisen from a sequencing error. Our predicted sequence of the Brassica pollen oleosin has all the structural characteristics of seed oleosins. A phylogenic tree of 20 oleosins, including those from sporophytic and gametophytic tissues of angiosperm and gymnosperm, was constructed based on their amino-acid sequences. We discuss the evolution of oleosins, and conclude that oleosins are ancient proteins with multiple lineages whose root cannot be determined at this time.Abbreviations PCR

Identification of differentially expressed genes in seeds of two near-isogenic Brassica napus lines with different oil content 总被引:5,自引:0,他引:5
The regulation of seed oil synthesis in rapeseed is largely unknown. In this study, we compared the gene expression during seed development between two lines of Brassica napus with a 10% difference in oil content. We isolated the immature seeds 15 and 25 days after flowering at periods preceding and including the major accumulation of storage oils and proteins. The differentially expressed gene clones between the two rape lines were isolated by subtractive suppression hybridization (SSH). All SSH clones were arrayed and screened by dot blot hybridization, followed by RT-PCR analysis for selected clones. A total of 217 cDNA clones corresponding to 30 genes were found to have a high expression in seeds with high oil content. Six genes were highly expressed in seeds with low oil content. Northern blot and enzyme activity analysis demonstrated a change in expression pattern of several genes. The results provide information on gene-encoding factors responsible for the regulation of oil synthesis. The possible role of these genes in seeds is discussed. The genes in this study may be suitable as novel targets for genetic improvement of seed oil content and may also provide molecular markers for studies of rape breeding. 相似文献

In order to know the genetic differences of de- and redifferentiation capacities, seven Brassica species (B. campestris, B. nigra, B. oleracea, B. hirta, B. carinata, B. juncea and B. napus) were cultured in vitro, and their response to the medium supplemented with various combinations of auxin and cytokinin hormones was compared. Important factors for callus initiation were shown to be auxin and species. Calli were induced most frequently in Murashige and Skoog (MS) medium with 1.0 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D), whereas -naphthaleneacetic acid (NAA) induced preferentially roots at a concentration of 2 to 5 mg/l. Callus-, root- and shoot-forming capacities from explanted cotyledon tissues were significantly different among the seven Brassica species. Calli derived from cotyledons and hypocotyls of seven species were transferred to MS media with 20g/l sucrose, 0 to 0.1 mg/l NAA and 0 to 4 mg/l kinetin to compare their regeneration capacities. Among the seven species tested, B. napus (2n=4x=38, genome AACC) had the highest shoot forming capacity (20.0%). Other amphiploid species, B. carinata (2n=4x=34, BBCC) and B. juncea (2n=4x=36, AABB) formed shoots at low frequencies (2.8% and 1.2%, respectively). A diploid species, B. oleracea (2n=2x 18, CC) also showed high shoot formation (10.2% on average). This result suggests that the gene(s) controlling shoot formation may be localized in the C genome. Differences were also found among varieties and cultivars within a species. One of the cultivars, Siberian kale (B. oleracea var. acephala) gave about 50% shoot formation. This kale was shown cytologically to be an autotetraploid (2n=4x=36, CCCC), thus probably possessing a double set of the shoot-forming gene(s). 相似文献

Barley (Hordeum vulgare), corn (Zea mays), bean (Phaseolus vulgaris), and radish (Raphanus sativus) seedlings were continuously irradiated under a lighting device for 5–10 d at an increased ultraviolet (UV)-B fluence rate. In their growth parameters, composition, and leaf surface, these four species responded differently to the increased UV-B exposure. Bean seedlings suffered the most serious effects, radish and barley less, and corn was hardly influenced at all. In all plant species, the fresh weight, the leaf area, the amounts of chlorophylls, carotenoids and the galactolipids of the chloroplasts were reduced. The lipid content of the corn and bean seedlings also diminished. But all the irradiated plants showed a rise in their protein content compared to the control plants. The content of flavonoids increased in barley and radish seedlings by about 50%. The effects on growth parameters and composition were more extensive with increasing UV-B fluence rates, at least as shown in the case of barley seedlings. The fresh weights fell proportionally with the chlorophylls and carotenoids. In contrast, the flavonoid content of barley leaves rose parallel to the increasing UV-B fluence rates and reached 180% of the value in the control plants with the highest UV-B fluence rate. Scorching appeared regularly in the form of bronze leaf discoloration at the highest UV-B fluence rates. Leaves exposed to above-ambient fluxes of ultraviolet-B (UV-B) radiation commonly contain increased concentrations of phenolic
compounds which may influence herbivores. However, the hypothesis that elevated UV-B modifies herbivory, whether mediated
by phenolics or other plant constituents, has rarely been studied experimentally. We investigated the responses of the mothAutographa gamma L. (Lepidoptera: Noctuidae) to pea (Pisum sativum L.) grown at a range of plant-effective UV-B fluxes. Although total phenolics did increase significantly with increasing
UV-B, this change had little deleterious effect on the 5th instar larvae ofA. gamma. However, tissue nitrogen also increased with increasing UV-B. Increased nitrogen was correlated with an increase in the
efficiency with which larvae utilized their food and in larval growth rate, but in a reduction in the amount of plant material
consumed. The apparently major role of nitrogen in determining herbivore responses to changing UV-B demonstrates the risks
