Pregnant sows and gilts were administered either 0, 2.5, 5, 10 or 20 mg prostaglandin F(2)alpha (PGF(2)alpha) intramuscularly on Day 112 or 113 of gestation at 0800 h in an effort to induce parturition. The average interval from PGF(2)alpha injection to farrowing was 55.1 +/- 5.7, 29.4 +/- 3.1, 32.1 +/- 4.6, 27.8 +/- 1.8 and 26.9 +/- 1.1 h for 0, 2.5, 5, 10 and 20 mg, respectively. All PGF(2)alpha treatments increased (P < 0.01) over controls the number of sows farrowing 23 to 33 h after injection. The average gestation length was significantly shorter in treated gilts; however, no detrimental effect on pig performance or pig survivability was observed. A second trial evaluated the effect of a 10-mg dose of PGF(2)alpha on the induction of parturition in sows in order to obtain a majority of sows farrowing within normal working hours (0700 to 1700 h). The interval from injection to farrowing was decreased (P < 0.05) by PGF(2)alpha treatment (66.2 +/- 5.3 vs 28.1 +/- 2.2 h). Fifty-seven percent (P < 0.05) of PGF(2)alpha-treated sows farrowed between 0700 and 1700 h as compared to 13.6% for control sows. A third trial was conducted to examine a sequential treatment of PGF(2)alpha and oxytocin to control the time of parturition more precisely. Sows receiving only 10 mg of PGF(2)alpha farrowed on an average 31.1 +/- 1.4 h after injection. The injection of 40 IU oxytocin 24 to 28 h after PGF(2)alpha decreased (P < 0.05) the interval from PGF(2)alpha to farrowing (28.1 +/- 0.9 h). The addition of oxytocin increased (P < 0.05) the number of sows farrowing within 3 h of injection (33 vs 86% for PGF(2)alpha and PGF(2)alpha + oxytocin treatments, respectively). A fourth trial was designed to determine if the addition of exogenous estradiol benzoate (EB) to a sequential treatment of PGF(2)alpha and oxytocin would improve the predictability and synchronization of the induced parturition. Sows were assigned to receive either saline, 10 mg PGF(2)alpha + 40 IU oxytocin or 10 mg PGF(2)alpha + 5 mg EB + 40 IU oxytocin. The addition of EB reduced (P < 0.01) the variance in the interval from oxytocin to farrowing and added precision to the predicted time of induced parturition.  相似文献

A total of 101 sows was used to examine postpartum progesterone levels and litter performance following administration of 15 mg prostaglandins F(2alpha) (PGF(2alpha) n = 48) given within 12 h after farrowing. Daily blood samples and rectal temperatures were taken from all sows during the first 3 d post partum. Plasma progesterone (P(4)) concentrations were determined by radioimmunoassay (RIA). Regardless of treatment, plasma P(4) levels for all sows decreased in a similar fashion over the 3 d sampled. Mean (+/- SEM) P(4) on Day 2 (0.55 +/- 0.06 ng/ml) and Day 3 (0.38 +/- 0.04 ng/ml) were lower (P<0.01) than on Day 1 (0.98 +/- 0.08 ng/ml). Rectal temperature did not differ between PGF(2alpha) treated and nontreated sows nor was it different over the days measured. Litter characteristics, including survival rates on Day 7 and at weaning, and body weight on Days 3 and 35, were not affected by treatment. It was concluded that PGF(2alpha) administration to sows within 12 h post farrowing had no affect on the rate of luteal regression, as determined by P(4) concentration, nor on subsequent litter performance.  相似文献

Behavioral and endocrine changes in the sow following injection with prostaglandin F2 alpha (PGF2 alpha) or its analogue, cloprostenol (CLO), were monitored to identify endocrine correlates of prepartum activity (nest-building). On Day 112 postcoitum, within 15 min after injection with 10 mg PGF2 alpha, sows offered straw in pens engaged in intense prepartum activity, but few behavioral changes occurred during the first 2 h following administration of 175 micrograms CLO. The temporal pattern of prepartum activity, however, was affected by both prostaglandins. In control sows, most prepartum activity came during Hours 16-0 before delivery of first piglet (delivery). After CLO, sows engaged in nest-building more during Hours 32-17 and less during Hours 16-0. In another experiment, sows in farrowing crates were injected with saline, 175 micrograms CLO, or 10 mg PGF2 alpha on Day 112 and blood was collected 0, 15, 30, 60, and 90 min later. Another sample was collected when spontaneous prepartum activity was first observed. For approximately 90 min after PGF2 alpha treatment, sows rooted, pawed, and bit and rubbed faces on crate bars; after saline and CLO, this behavior was rarely observed. After prostaglandin treatment, plasma progesterone tended to decline, a 10-fold rise in relaxin came within 15 min, but estrone did not change. Plasma prolactin rose 10-fold within 30 min after PGF2 alpha treatment, and rose more gradually after CLO treatment. The objective of this study was to assess the effects of the addition of prostaglandin F2alpha (PGF2alpha) to semen, to increase reproductive performance in sows. Sows in 11 large production units, were inseminated (AI) either with PGF2alpha enriched fresh semen (group 1, n=1258) or with untreated fresh semen (group 2, n=1101). Conception rate, regular return to estrus, farrowing rate, subsequent total and live-born litter size, subsequent weaning to estrus intervals, pigs born and pigs weaned per sow per year were evaluated. Conception and farrowing rates, as well as regular returns to estrus were altered beneficially (P<0.001) with PGF2alpha supplemented semen. Subsequent total-born (P<0.06) and subsequent live-born (P<0.15) litter size, as well as subsequent weaning to estrus intervals (P<0.22), were not affected to the same extent. Total pigs born (P<0.05) and pigs weaned (P<0.04) per sow per year were increased by using PGF2alpha supplemented semen.  相似文献

In order to determine if PGF(2)alpha could improve fertility in repeat breeder females when added to semen used for artificial insemination (AI) the following trial was performed. In a large indoor Hungarian production unit of 2000 sows, 667 repeat breeding females were assigned to two groups and were treated as follows: Group 1 (n=322), received PGF(2)alpha, added to the semen immediately before AI; Group 2 (n=345), received AI with untreated semen. Conception rate, farrowing rate, subsequent total and live born litter size and subsequent weaning to estrus intervals were evaluated. Conception and farrowing rates revealed highly significant differences between the PGF(2)alpha-treated and nontreated animals (P<0.001). Subsequent total born (P<0.07), and live born litter size (P<0.13), and subsequent weaning to estrus intervals (P<0.23) showed no significant differences. It is reasonable to suggest that exogenous PGF(2)alpha added to AI semen improves conception and farrowing rates.  相似文献

Eighty crossbred, multiparous sows, weighing between 190 and 320 kg, were randomly assigned to the following four treatment groups of 20 sows each: 1) saline-saline, 2) cloprostenol-saline, 3) saline-xylazine and 4) cloprostenol-xylazine. The mean gestation length of each multiparous sow was calculated. Cloprostenol (250 ug/sow, i.m.) or saline was given 3 d prior to the calculated due date at 11:30 a.m. Xylazine (2 mg/kg, i.m.) or saline was given 20 h after either the cloprostenol or previous saline treatment. Cloprostenol-xylazine treated sows had the shortest mean farrowing interval (1.5 +/- 0.3 h) when compared with the rest of the treatment groups (saline-saline:66.0 +/- 8.1, cloprostenol-saline:10.5 +/- 1.9, saline-xylazine:60.6 +/- 5.6 h). Farrowing time, percentage of stillbirths, average birth weight, d-5 and d-21 postbirth weights, number of pigs born, number of pigs born alive, and number of pigs surviving at 5 and 21 d afterbirth were not significantly different among the four groups. This study demonstrated that cloprostenol-xylazine treatment decreases the time to onset of farrowing with less variation than cloprostenol or xylazine alone. Therefore, the use of a cloprostenol-xylazine combination is suggested as an alternative method for inducing farrowing.  相似文献

A total of 11 cycling does weighing between 24 and 50 kg were injected with varying dosages of prostaglandin F(2)alpha (PGF(2)alpha) between 7 and 10 days into each estrous cycle. Five injections each of 1.25, 2.5, 5.0, or 7.5 mg PGF(2)alpha were alternated with five injections of 1.0 ml saline. Saline treated does served as controls. All does were teased twice daily with a buck and observed for signs of estrus for 5 days post-injection. Daily systemic concentrations of progesterone (P(4)) were determined by radioimmunoassay. The mean (+/- S.E.) hours from injection to estrus was 47 +/- 3.3, 42 +/- 4.3, 44 +/- 8.5, and 43 +/- 5.5 for does receiving 1.25, 2.5, 5.0, and 7.5 mg PGF(2)alpha, respectively. None of the does receiving saline exhibited estrus in the 5-day post-injection observation period. Mean (+/- S.E.) concentrations of systemic P(4) in all does on the day of injection was 4.22 +/- 0.45 ng/ml. Concentrations 24 hours post-injection were 0.21 +/- 0.02, 0.15 +/- 0.05, 0.17 +/- 0.04, 0.16 +/- 0.04, and 4.5 +/- 1.36 ng/ml for does receiving 1.25, 2.5, 5.0, and 7.5 mg PGF(2)alpha, and 1.0 ml saline, respectively. The results suggested that 1.25 mg PGF(2)alpha was effective for induction of estrus in the cycling goat.  相似文献

A 4000 sow farm in the US using early weaning and a computerized record system was recruited. Farrowed sows were assigned into two experimental treatments: prostaglandin F2alpha injection or control. Sows were assigned by a farm worker to obtain even parity distributions between two groups in each farrowing group. A single i.m. injection of 2 ml of prostaglandin F2alpha between 24 and 48 h after farrowing was administered in the muscle immediately lateral to the vulva. Control sows received no treatment. Of 3562 farrowed sows, 1592 were administered with prostaglandin F2alpha. Parity distributions were not different between control and treatment groups. Parity was categorized into two groups: parity 1-2 or > or = 3. Mean lactation length was 18 days and there was no difference between the control and treatment groups. No main effects of prostaglandin F2alpha administration were found in either parity group on adjusted 21-day litter weight, weaning-to-first-mating interval or weaning-to-conception interval. In parity > or = 3 sows, a two-way interaction between the association of lactation length and treatment with pigs born alive at subsequent farrowing was found (P = 0.044), while no such interaction was found in parity 1-2 sows (P = 0.14). The prediction line for subsequent pigs born alive indicates that prostaglandin F2alpha administration alters the relationship between lactation length and subsequent litter size on mid- or old-parity sows.  相似文献

In the present study, the kinetics of the prostaglandin F2alpha (PGF2alpha)-metabolite 15-keto-13,14-dihydro-PGF2alpha after a single intramuscular application of various doses of the natural PGF2alpha dinoprost at Day 7 of the cycle in the mare were investigated. Effects of low doses on estrous cycle length and life span of corpus luteum were examined, because release of PGF2alpha is still under discussion to have detrimental influence on success rates of transcervical transfer of equine embryos. Eight Shetland pony mares were each randomly assigned to each of four treatments: (a) 0.8 mg/100 kg (group T1), (b) 0.4 mg/100 kg (group T2), (c) 0.2 mg/100 kg BM dinoprost i.m. (group T3), and (d) 1 ml physiological saline i.m. (group CO). Treatments were administered as single doses on Day 7 of the estrous cycle. Administration of dinoprost caused dose-dependent rises of plasma concentrations of PGF2alpha-metabolite, although values of individual mares showed great variation within groups. Prostaglandin treatments resulted in a distinct decrease of plasma progesterone concentrations to values between 1.6 and 7.9 ng/ml within 24 h. Treatment groups had significantly lower progesterone area under the curve (AUC: T1 942.8+/-175.9, T2 1050+/-181.2 and T3 1117+/-179.8 ng/ml/h) when compared with controls (CO 1601.9+/-227.6; t-test, P<0.05 ). There was a small, but significant negative correlation between AUC of progesterone and of PGF2alpha-metabolite ( R=-0.4; P=0.05 ). Administration of PGF2alpha caused secretion of oxytocin in three (T1, T2) and two (T3) mares out of eight ranging from 19.3 to 63.1 pg/ml. The AUC of oxytocin was positively correlated with AUC of PGF2alpha-metabolite ( R=0.4, P<0.05) and negatively correlated with AUC of progesterone ( R=-0.4, P<0.05). Administration of dinoprost yielded significantly shorter intervals from treatment to estrus and ovulation (values in parentheses), respectively, when compared with controls: T1 3.9+/-0.7 days ( 12.1+/-0.7 days), T2 4.5+/-0.6 ( 12.3+/-0.6 ), T3 4.9+/-0.5 ( 12.3+/-0.6 ), and CO 8.9+/-0.6 days ( 16.5+/-0.8 days) (t-test, P<0.01 ) (Fig. 2). Different doses of PGF2alpha caused similar effects. Data suggest that progesterone concentrations at applications influence efficacy of treatments more than doses administered, as demonstrated by their high correlation with estrous cycle patterns. It is important to note that differences we achieved are gradual and that all mares responded to treatment by luteolysis and premature estrus, regardless of doses applied.  相似文献

Domestic pigs build a maternal nest in the day preceding parturition. We have shown that prostaglandin F(2alpha) (PGF(2alpha)) induces nest building behaviour in non-pregnant pigs. The aim of this experiment was to examine the effects of different environmental temperatures on PGF(2alpha)-induced nest building. Data were collected from 9 Large White (LW) and 10 Large Black (LB) 8-9-month-old nulliparous sows (gilts). The pigs were housed in social groups between experiments and tested individually in pens (1.8mx1.8m) containing straw, within an environmentally controlled chamber. Pigs were habituated to the testing pens (maintained at 17 degrees C) and tested once at each of three temperatures (low, 5 degrees C; moderate, 17 degrees C; high, 30 degrees C). During testing the temperature of the chamber was adjusted at 09.00h and had reached set point by10.00h. The pigs were injected intramuscularly with 3ml saline at 10.30h and 0.1mg/kg PGF(2alpha) (Lutalyse, Upjohn) at 11.30h. Behaviour was scored for 1h after treatment with saline and 1h after treatment with PGF(2alpha) using one/zero sampling from video recordings. Nest building behaviour (rooting, pawing and gathering straw) was induced by PGF(2alpha) at all temperatures in both LW and LB breeds. There was a significant increase in rooting behaviours with decreasing temperature. No significant effects of temperature were found on the scores for gather or paw. The pigs spent more time lying down at the high compared to the low temperature after both saline and PGF(2alpha) treatment. Other behaviours unrelated to nest building but induced by PGF(2alpha), such as scratching, were unaffected by temperature. The results show that the nest building behaviour of non-pregnant pigs can be induced by exogenous PGF(2alpha) treatment, and that some, but not all, aspects of PGF(2alpha)-induced nest building (rooting but not pawing or gathering) are altered by environmental temperature.  相似文献

The effect of an in vivo prostaglandin F2 alpha (PGF2 alpha) challenge in pregnant and cyclic sows was compared to determine whether PGF2 alpha-induced release of relaxin (RLX) from the corpus luteum (CL) in late pregnancy is also effective during the cycle. Ovarian venous RLX and progesterone were monitored by radioimmunoassay and RLX localized in the CL by immunohistochemistry. In Day 108 pregnant sows, infusion of PGF2 alpha (100 micrograms) into the ovarian artery resulted in an immediate and sustained rise in ovarian venous RLX with an initial decline in progesterone levels by 30 min which then returned to pretreatment levels. In Day 13 or 15 cyclic sows with functional corpora lutea (i.e., elevated progesterone), RLX was undetectable in ovarian venous blood after 100 micrograms of PGF2 alpha. Administration of PGF2 alpha via either the jugular vein or intramuscular route was also ineffective in releasing RLX from the CL of the cycle. The intensity of RLX immunostaining of the CL was similar in saline and PGF2 alpha-treated sows. These studies indicate that the control of RLX release from the sow CL differs in the estrous cycle and pregnancy.  相似文献

The objective of this study was to determine the dose of alfaprostol (18, 19, 20-trinor-17-cyclohexyl-13, 14 didehydro-PGF(2)alpha-methylester) and the day of administration most effective in inducing sows to farrow during normal working hours (0700 to 1700). One-hundred forty multiparous crossbred sows, taken from a herd whose mean gestation length was 114.3 days, were assigned to one of five treatment groups: 1) control vehicle-propylene glycol, 2) 0.5 mg alfaprostol (AP). 3) 1 mg AP, 4) 2 mg AP and 5) 3 mg AP. Sows received an intramuscular injection of AP between 0800 and 0830 on either day 111, 112 or 113 of gestation. Parameters studied included interval from injection to birth of first pig, farrowing interval, total number of pigs born, number born alive, average birth weight, percent stillborn, interval from weaning to next estrus and number born alive next litter. The mean intervals from injection to the birth of the first pig were 55.2 +/- 7.1; 41.1 +/- 5.1; 29.6 +/- 4.0; 24.3 +/- 1.1; 24.8 +/- 0.9 h for groups 1, 2, 3, 4 and 5, respectively (P<0.05). The percentage of sows that farrowed during normal working hours (23 to 33 h after injection) for groups 1, 2, 3, 4 and 5 were 15, 53, 60, 77 and 70%, respectively. Average birth weights (kg) for treatment groups 1, 2, 3, 4 and 5 were 1.54 +/- 0.05; 1.65 +/- 0.05; 1.59 +/- 0.05; 1.54 +/- 0.05 and 1.42 +/- 0.05, respectively (P<0.05). Mean differences in total number of pigs born and number born alive were also statistically significant (P<0.05) but stillbirth rate was not different (P>0.05) among treatment groups. These results indicate that a single injection of 1, 2 or 3 mg of alfaprostol will successfully induce parturition the following day in a majority of the treated sows.  相似文献

The objective of this experiment was to determine the effect of mifepristone, a progesterone receptor antagonist, on pregnancy and secretion of steroids, pregnancy-specific protein B (PSPB) and prostaglandins at mid-pregnancy in ewes. Ninety-day pregnant ewes were ovariectomized (OVX) and treatments were initiated 72 h post-OVX. Ewes received (1) vehicle, (2) prostaglandin F2alpha (PGF2alpha, 8 mg/58 kg/bw, i.m.) 84 h post-OVX, (3) mifepristone (50 mg intrajugular at 72, 84, 96, and 108 h post-OVX), (4) mifepristone (50mg) + PGF2alpha, (5) mifepristone (100 mg intrajugular at 72, 84, 96, and 108 h), and (6) mifepristone (100 mg) + PGF2alpha. Ewes treated with vehicle or PGF2alpha alone did not abort (P > or = 0.05). But, 60, 80, 60, and 100% of ewes treated with mifepristone (50 mg), mifepristone (50 mg) + PGF2alpha, mifepristone (100 mg), and mifepristone (100 mg) + PGF2alpha, respectively, aborted (P < or = 0.05). Profiles of progesterone, estradiol-17beta, prostaglandin E (PGE), or PSPB did not differ (P > or = 0.05) among treatment groups. Profiles of PGF2alpha of treatment groups receiving mifepristone with or without PGF2alpha differed (P < 0.05) from vehicle or PGF2alpha alone-treated ewes. It is concluded that progesterone actions are necessary to suppress uterine/placental secretion of PGF2alpha and that maintenance of critical progesterone: estradiol-17beta and PGE:PGF2alpha ratios are necessary for maintenance of pregnancy.  相似文献

Results of two experiments are described. In the first experiment, forty-one mixed-breed goats (does) with unknown gestation lengths were given 10 mg prostaglandin F(2alpha) (PGF(2alpha)) (2 doses of 5 mg each, 24 h apart) i.m. Blood samples were obtained before each treatment with PGF(2alpha) by jugular venipuncture, and plasma progesterone (P(4)) concentrations were determined by a nonextraction solid-phase radioimmunoassay. P(4) concentrations (ng/ml) were significantly decreased (15.47 vs 1.55, P<0.005) 24 h after the first injection of PGF(2alpha). A total of 63 fetuses was collected within 46.5 h following the first injection. Mean (+/- SE) crownrump lengths and body weights of 62 fetuses were 21.46 +/- 0.29 centimeters (cm) and 575.00 +/- 20.60 g, respectively. Based on these findings, the mean gestation length of these does was estimated to be 86.96 +/- 0.74 d. Thirty-one does retained their placenta for 12 to 72 h and were treated with a single injection of 5 mg PGF(2alpha) and 800 mg oxytetracycline i.m. Placental expulsion in all does occurred within 24 h posttreatment. The results of this study suggest that two doses of 5 mg PGF(2alpha) intramuscularly (i.m.) 24 h apart is an effective abortifacient at about 3 mo of pregnancy in does. In the second experiment, 38 does from the first experiment were divided in two groups of 19 each on Day 13 postabortion. Group A (treated) was given 50 ug GnRH i.m. while Group B (control) received 1 ml 0.9% saline i.m. Blood samples were obtained prior to treatment and on Day 23 postabortion and assayed for P(4) concentrations. There was no significant difference (P>0.10) in P(4) concentrations of samples obtained pre- and post-GnRH treatment. However, 14 of 19 and 12 of 19 in Groups A and B, respectively, exhibited estrus within 52 days following abortion. Twenty-six does were bred naturally and 17 became pregnant.  相似文献

In seminatural environments, prepartum sows leave the herd and construct a maternal nest (a dug out hollow lined with vegetation) prior to the birth of their piglets. The endocrine drives motivating this behavior are not understood, but may involve prostaglandin (PG) F2alpha. This study examined the effect of PGF2alpha treatment on the behavior of pseudopregnant gifts housed in a large enclosure. Pseudopregnancy was induced using 5 mg/ml estradiol valerate/day im from days 11 to 15 of the estrous cycle (first day of estrus = day 0). The gifts' behavior was recorded on a control day, during which no treatment was given, and a test day (= 45.9 +/- 0.42 days of pseudopregnancy) when gilts received either 15 mg PGF2alpha (dinoprost: Lutalyse, Upjohn, Crawley, UK, n = 11) or 0.9% saline (n = 10) im at 11.00 h. PGF2alpha-treated gilts traveled further and were more frequently >10 m from the nearest pig than saline-treated animals. In the hour following injection, PGF2alpha-treated animals also showed increased frequencies of rooting and pawing the ground and stood for longer than saline-treated animals. However, gathering and carrying nest materials were not increased. These results suggest that PGF2alpha, given as a single dose to extensively housed gilts, initiated many, but not all, of the behaviors characteristic of prepartum nest building. The dose and duration of PGF2alpha treatment may have limited the observed behaviors. In addition, environmental feedback is likely to affect the degree to which some nest building behaviors are expressed.  相似文献

The timing of PGF(2alpha) release and the timing and extent of the rise in endometrial oxytocin receptors was determined in relation to the timing of the progesterone fall during luteolysis in cycling cows. In cows undergoing luteolysis (n = 6), measurement of PGF(2alpha) metabolite in hourly plasma samples collected during daily 10 h sampling periods identified a total of 2.2+/-0.5 PGF(2alpha) release episodes per animal, each of 4.0+/-0.4 h duration. In cows in which luteolysis was not observed (n = 4) no PGF(2alpha) release episodes were identified. In a further three cows in which additional repeated uterine biopsies were collected on days 15, 17, 19, 21 and 23, endometrial oxytocin receptors were initially undetectable (<15 fmol/mg protein) but had increased to 120+/-19 fmol/mg protein prior to the initiation of PGF(2alpha) release episodes. Receptor concentrations then continued to increase reaching peak concentrations of 651+/-142 after luteolysis had been completed.  相似文献

In most cyclic females, prostaglandin F(2alpha) (PGF(2alpha)) triggers a uterine motility response resembling that of oxytocin (OT). To determine if PGF(2alpha) is a uterokinetic substance in the cycling mare, uterine motility was measured by intrauterine balloon technique in 12 conscious, normally cyclic mares. After 60 min of saline infusion, continuous intravenous (i.v.) infusion with OT (1 i.u./min) was followed by PGF(2alpha) (200 mug/min) for 60 min each. The experiment was repeated 3 wk later except with PGF(2alpha) preceeding OT. A second group of mares was administered OT (60 i.u.) either i.v., intramuscularly (i.m.), or intrauterinely (i.u.). Plasma samples were studied for progesterone concentration. Control uterine motility for the first group of mares was (mean +/- SEM) 545.83 +/- 45.10 mm(2). Significant (P<0.05) elevation in uterine motility was recorded for OT (1118.60 +/- 70.56 mm(2)) regardless if PGF(2alpha) preceded OT infusion or vice-versa. No significant difference (P>0.05) was seen in motility after PGF(2alpha) (423.33 +/- 31.12 mm(2)) infusion. The uterokinetic effect of OT was greatest when OT was administered i.v. (1696.50 +/- 195.46 mm(2)) followed by i.m. (819.82 +/- 39.96 mm(2)), and it was least effective when administered i.u. (607.83 +/- 21.56 mm(2)) as compared to control uterine motility (279.78 +/- 22.33 mm(2)). Skin electrical resistance values rose from 0 to 2000 ohms with PGF(2alpha) infusion (but not with OT), indicating that PGF(2alpha) was bioactive. It was concluded that PGF(2alpha) was not a uterokinetic substance in the cyclic mare.  相似文献

There is evidence that the dose of PGF(2)alpha generally used to synchronize estrus (25 mg) is higher than required to induce luteolysis in cattle. To investigate this, 98 Holstein cows from three farms were assigned at random within farm to be treated with a single dose of 25 mg (n=33), 17.5 mg (n=33) or 10 mg (n=32) of PGF(2)alpha on Day 10+/-0.5 (mean +/- SEM) of the estrous cycle. Statistical analyses were conducted using analyses of variance and Chisquare test. Only 59.3% of the cows treated with 10 mg of PGF(2)alpha were detected in estrus compared with 72.7 and 78.7% of the cows treated with 17.5 and 25.0 mg doses, respectively (P>0.05). There were no differences (P>0.05) in pregnancy rates at the first service (40.0, 66.6 and 50.0% for 25, 17.5 and 10 mg, respectively). Concentrations of progesterone in blood were different (P<0.05) for cows treated with 10 mg compared with those of cows treated with 17.5 or 25 mg of PGF(2)alpha. The pattern of changes in progesterone concentrations between the last two groups was not different, and progesterone concentrations of less than 1 ng/ml of serum were observed within the first 36 h post PGF(2)alpha administration. In cows treated with 10-mg dose of PGF(2)alpha, concentrations of progesterone declined during the first 24 h, however, by the end of the experimental period, they were not different to pretreatment concentrations (treatment x time; P<0.05). It is suggested that reducing the dose of PGF(2)alpha from 25 to 17.5 mg do not affect estrus response or pregnancy rate in Holstein cows.  相似文献

In the ewe, a rise in circulating concentrations of FSH preceding follicular wave emergence begins in the presence of growing follicles from a previous wave. We hypothesized that prostaglandin F(2alpha) (PGF(2alpha)) given at the time of an endogenous FSH peak in cyclic ewes would result in synchronous ovulation of follicles from two consecutive waves, increasing ovulation rate. Twelve Western White Face (WWF) ewes received a single i.m. injection of PGF(2alpha) (15 mg/ewe) at the expected time of a peak in FSH secretion, from Days 9 to 12 after ovulation. The mean ovulation rate after PGF(2alpha) treatment (2.3+/-0.3) did not differ (P>0.05) from the pre-treatment ovulation rate (1.7+/-0.1). Five ewes ovulated follicles from follicular waves emerging before and after PGF(2alpha) injection (3.0+/-0.6 ovulations/ewe) and seven ewes ovulated follicles only from a wave(s) emerging before PGF(2alpha) treatment (2.0+/-0.3 ovulations/ewe; P>0.05). The mean interval from PGF(2alpha) to emergence of the next follicular wave (1.0+/-0.4 and 4.0+/-0.0 d, respectively; P<0.001) and the interval from PGF(2alpha) treatment to the next FSH peak (0 and 3.5+/-0.4d, respectively; P<0.05) differed between the two groups. Six ewes ovulated after the onset of behavioral estrus, with a mean ovulation rate of 1.7+/-0.2, and six ewes ovulated both before and after the onset of estrus (3.0+/-0.5 ovulations/ewe; P<0.05). None of the ovulations that occurred before estrus resulted in corpora lutea (CL) with a full life span. At 24h before ovulation, follicles ovulating before or after the onset of estrus differed in size (4.1+/-0.3 or 5.5+/-0.4mm, respectively; P<0.05) and had distinctive echotextural characteristics. In conclusion, the administration of PGF(2alpha) at the expected time of an FSH peak at mid-cycle in ewes may alter the endogenous rhythm of FSH secretion and was not consistently followed by ovulation of follicles from two follicular waves. In non-prolific WWF ewes, PGF(2alpha)-induced luteolysis disrupted the normal distribution of the source of ovulatory follicles and may be associated with untimely follicular rupture and luteal inadequacy.  相似文献

Addition of prostaglandin F2alpha (PGF2alpha) to extended boar semen has been shown to slightly increase reproductive parameters in sows such as the conception rate and the total number of piglets born alive. The mechanisms by which PGF2alpha affect these parameters have not yet been elucidated, but it is possible that the sperm transport after insemination is increased. This study investigated whether the sperm motility from 20 Piétrain boars improved when PGF2alpha (Dinolytic; 5 mg PGF2alpha/ml) was added to diluted semen. Different amounts of PGF2alpha (0, 0.5, 1 and 2 ml/100 ml) were tested and the motility was evaluated immediately after addition of PGF2alpha, after 30 min, 2 h, and 24 h. Two computer-assisted semen analysis (CASA) systems, namely the Sperm Quality Analyzer (SQA-IIC) and the Hamilton Thorne (HTR Ceros 12.1) were used to assess the motility parameters. With the SQA-IIC, sperm motility index values of the treated groups were only slightly higher (P>0.05) compared to the negative control group. The different motility parameters measured with the HTR Ceros 12.1 were similar between the treatment groups, except for beat cross frequency, which was higher in the control group (1.5-5%; P<0.001). This study documented that the addition of 2.5, 5 or 10 mg PGF2alpha to 100 ml diluted boar sperm does not increase any sperm motility parameter. Further research is necessary to elucidate mechanisms by which PGF2alpha in diluted semen may improve the reproductive performance in swine farms.  相似文献
