The aim of this study was to compare the nutritional value of three diets with rats submitted to protein malnutrition. After one month on a low protein diet (2% casein), the rats were refed with corn-soya-milk (60%/30%/5%, W/W/W) and rice-cotton-milk (60%/30%/5%) diets. This produced a higher growth rate than either the corn-cotton milk (60%/30%/5%) diets or a control diet containing 20% casein. The caloric intake/100 g body weight was about the same with the four rehabilitation diets. The highest apparent nitrogen digestive utilization coefficients (C. U. D.) values were obtained with the control diet, lower values with the rice-cotton-milk diet, then the corn-cotton-milk diet and lowest with the corn-soya-milk diet. Results of lipid C. U. D. were in the same order. The highest nitrogen balances were obtained with the control diet, then the corn-soya-milk diet, the rice-cotton-milk diet and lowest values with the corn-cotton-milk diet. Our data prove that the cotton flour mixed with rice may perfectly replace soya flour associated with corn in a nutritional rehabilitation diet.  相似文献

Albino rats (Sprague-Dawley) of mean weight 100 g were divided into four groups and given for 7 days a balanced diet. They were then placed in metabolic cages for fifteen days and fed diets containing different quantities of casein: 18% (D18), 36% (D36), 50% (D50) and 72% (D72). The levels of total calcium, inorganic phosphorus, alkaline phosphatase activity, total proteins and urea were determined. The urinary and fecal excretion of calcium were determined on specimens of urine and stool collected every two days. The metabolic balance of nitrogen was also estimated. The results show there is not a linear relationship between a high protein diet and plasma protein levels, but a progressive body calcium loss was observed with the increase of casein in the diet, which confirms what other workers have already suggested.  相似文献

Mounting doses of casein and wheat gluten protein (from 0% to 40% in the diet) were given to adult male rats aged 75-89 days for 14 days. The optimum physiological daily dose, determined from changes in body nitrogen, body water and body weight, was 2.76 g/d for casein protein (corresponding to a 10% casein protein diet) and 3.67 g/d for wheat gluten protein (corresponding to a 15% gluten protein diet). Determined from body weight changes, the daily maintaining dose of casein or wheat gluten protein was 1.054 and 1.511 mg/d respectively, from body nitrogen changes 970 and 1.514 mg/d and from body water changes 1.124 and 1.637 mg/d. Compared with newly weaned animals aged 35-49 days, the optimum physiological daily dietary protein doses for adult animals fall, while the maintaining doses rise.  相似文献

We previously demonstrated that feeding a diet containing a high level of amino acid mixture simulating casein (AA) induced an increase in pancreatic protease activities in rats. In the present study, this effect of dietary AA was further characterized with three separate experiments. These experiments (1) examined periodic changes in pancreatic and small intestinal trypsin activities after switching from a 20% (a normal nitrogen level) AA diet to a 60% AA (a high nitrogen level) diet; (2) measured the abundance of mRNA for four trypsinogen isozymes and for intestinal cholecystokinin (CCK) and secretin in rats fed 20% and 60% AA diets for 10 days compared with rats fed 20% and 60% casein diets; and (3) measured the abundance of mRNA for four trypsinogen isozymes after chronic administration of CCK. Trypsin activities were gradually increased in both the pancreas and the small intestinal lumen and reached maximum at 5 days after the switch to the 60% AA diet (Exp. 1). This result is evidence that the increase in the protease activity in the pancreas is due to enhancement of pancreatic trypsin production. In experiment 2, pancreatic trypsinogen isozymes I, II, III, and IV mRNA abundance were evaluated by the Northern blotting method using cDNA probes specific for each isozyme mRNA. Abundance of trypsinogen mRNA without trypsinogen I tended to increase in the rats fed the 60% casein diet but tended to decrease in the rats fed the 60% AA diet compared with the respective normal nitrogen level diet groups without significant difference. CCK mRNA abundance in the jejunal mucosa increased as a result of feeding the 60% casein diet, but not the 60% AA diet. Subcutaneous CCK injections (3.5 nmole/kg body weight/day, twice daily, at 8:30 am and 7:30 pm) for 10 days resulted in increased pancreatic trypsin activity, whereas the changes in mRNA of the four trypsinogen isozymes was similar between the 20% and 60% casein groups but differed between the 20% and 60% AA groups (Exp. 3). These results suggest that CCK is not involved in the induction of pancreatic trypsin that occurs with feeding of a high AA diet and that the mechanism of protease induction by dietary AA is different from that in the case of dietary protein.  相似文献

Summary Mice were starved for 2 days and then fed on either a protein-free diet or a 25 per cent casein diet for 7 days and then sacrificed. The former group, in contrast to the latter, decreased in weight during these 7 days and showed significantly lower weight absolute and relative (g/100 g initial body weight) of kidneys, liver and spleen. In the animals deprived of protein, the ratio between total liver nucleolar volume per nucleus and the nuclear volume was larger, than in the casein fed animals. The results suggest that the liver nucleolar apparatus reacts to a protein-free diet in principally the same way in mice as in rats.The investigation was supported by a grant from the Swedish Medical Research Foundation.  相似文献

Six-month-old male rats were given diets with mounting casein and wheat gluten protein concentrations from 0% to 40% and after 14 days the optimum and minimum physiological doses were determined from changes in body nitrogen, body water, body weight and protein intake. The optimum dose for casein protein was 1.54 g/d (5% protein in the diet) and for wheat gluten protein 2.10 g/d (7.5% protein in the diet). The minimum casein and wheat gluten protein doses for 180- to 194-day-old rats, determined from body nitrogen changes, were 1499 mg/d (4.86%) and 1995 mg/d (7.12%) respectively, from body water changes 1561 mg/d (5.06%) and 2027 mg/d (7.23%) and from body weight changes 1333 mg/d (4.32%) and 1951 mg/d (6.06%). It should be noted that, unlike younger animals aged 35-49, 75-89 and 120-134 days, the optimum and minimum doses for six-month-old rats were approximately the same.  相似文献

After male rats of the Sprague Dawley strain, 5 weeks old, were fed a 20% casein diet with or without 0.5% nicotinamide for 13 days, 180 mg/kg body weight of alloxan was injected in- traperitoneally into the rats. The rats were kept for 18 days with the same diet. The level of blood glucose was increased 6-fold in the group on a 20% casein diet by the injection of alloxan, while there was only a 2-foid increase in the group on a nicotinamide-containing diet and the decreased body weight was also lower in the group on the nicotinamide diet than the group on the casein diet. The body weight was indirectly related to the concentration of blood glucose. A marked increase was observed in the activities of tryptophan oxygenase, aminocarboxymuconate-semialdehyde decarboxylase, and nicotinamide methyltransferase upon the injection of alloxan with both diets; on the other hand, the activities of kynureninase and NAD+ synthetase were decreased by the injection of alloxan. The activity of kynurenine aminotransferase increased in the group on the 20% casein diet by the injection of alloxan, while in the group on the nicotinamide-containing diet its activity was not increased by the injection. These changes in the above enzyme activities mean that the conversion ratio from tryptophan to niacin is lower in the alloxan diabetic rat than normal rat. It was found that the activities of tryptophan oxygenase, aminocarboxymuconate-semialdehyde decarboxylase, and nicotinamide methyltransferase were directly related to the concentration of blood glucose, and that the activities of kynureninase and NAD+ synthetase were inversely related. There was no difference in the activities of 3-hydroxyanthranilic acid oxygenase and nicotinamide mononucleotide adenylyltransferase upon the injection of alloxan with both diets.  相似文献

Protein-calorie malnutrition was induced by giving a 2% casein diet to a strain of post-weaning male Wistar rats from 26 to 56 days of age. They were then rehabilitated by a 15% casein diet till 86 days of age. Body and testis weights, diameter of seminiferous tubules and spermatogenetic score were compared to those of controls fed with the 15% casein diet. The unbalanced diet retarded growth by 30 days. Although rehabilitation of body and testis weights was incomplete, the spermatogenetic function equalled that of the controls at the end of the experiment. This aspect has been discussed.  相似文献

The amount of extra- and intravascular albumin was estimated in two groups of rats, i.e., those fed a 20% casein (20% protein) diet and a 3% casein (low protein or 3% protein) diet.

The fractional turnover rate of whole body plasma albumin was also measured in the two groups of rats, employing the constant infusion method of Waterlow et al. At the same time, the fractional turnover rate of the whole body protein was measured.

When the diet was changed from the 20% protein to the 3% protein diet, the amount of albumin mass in both extra- and intravascular spaces decreased significantly. During 7 days on the diet, the extra- and intravascular albumin mass per 100 g of body weight did not change significantly in the rats fed the 20% protein diet. On the other hand, rats fed the 3% protein diet lost almost 30% of the extra- and intravascular albumin per lOOg body weight.

The fractional turnover rates of whole body albumin were estimated to be 31.7 and 19.8%/day in the 20% protein and the 3% protein diet-fed rats, respectively. The fractional turnover rates of whole body protein were 16.1 and 10.6%/day in the 20% protein and the 3% protein diet-fed rats, respectively.

The leucine fluxes to albumin synthesis and whole body protein synthesis were calculated to be 5.9 and 83 μmol/hr, respectively, in the 20% protein diet-fed rats. The leucine fluxes in the 3% protein diet-fed rats were 2.5 and 54μmol/hr for the albumin synthesis and for the whole body protein synthesis, respectively.

These results demonstrate the quantitative significance of albumin metabolism in the whole body protein turnover in rats fed on two levels of protein intake.  相似文献

The metabolic and cardiovascular adjustments of the whole body and skeletal muscle were studied during moderate and severe acute anemia. In 15 anesthetized dogs, venous outflow from the gastrocnemius-plantaris muscle group was isolated. Cardiac output (QT) muscle blood flow (QM), total body and muscle oxygen uptake (VO2) were determined during a control period, and at 30 and 60 min of either (i) moderate anemia (n = 8) in which the mean hematocrit (Hct) was 25% or (ii) progressive anemia (n = 7) in which the mean Hct values were 25% at 30 min and 16% at 60 min of anemia. Muscle VO2, QT, and QM were increased in both groups at 30 min of anemia. By 60 min, QT and QM declined to preanemic control values in the moderate anemia group; whole body VO2 was maintained at the control level. Arterial oxygen transport was the same in the two groups at both 30 and 60 min of anemia despite the difference in Hct at 60 min. Muscle VO2 showed a further and similar rise in both groups between 30 and 60 min of anemia. These data show that the rise in muscle VO2 during acute anemia was not directly proportional to the degree of the hematocrit reduction. Further, the findings suggest that the muscle VO2 response was related to the decrease in arterial oxygen transport.  相似文献

Abstract— From the third day of pregnancy rats were fed a diet containing either 7% casein (experimental) or 24% casein (control). During lactation the control dams were fed the 24% casein diet and the experimental dams a 12% casein diet. From 25 to 50 days of age the experimental and control progeny were fed diets containing 7 and 24% casein, respectively. Between 50 and 120 days both groups were fed a diet containing 24% crude protein. Several indications of brain maturation in two brain areas were examined at various stages of development. In addition to retardation of brain growth, protein restriction led to myelin of an immature composition at 25 and 50 days of age. The immature composition was indicated by a low plasmalogen content at 25 days and by a high phospholipid and low galactolipid and plasmalogen contents at 50 days of age. The activity of the myelin marker enzyme, 2′3′-cyclic nucleotide 3′-phosphohydrolase (CNP), was significantly lower in the brains (excluding the cerebella) of malnourished rats at 21, 30 and 50 days. At all ages except at 50 days the activity of CNP in the cerebellum was higher in protein-deprived animals than in controls. The activity of glutamic acid decarboxylase (GAD) in the brains (excluding the cerebella) of protein-deprived rats was significantly lower at 21, 25 and 30 days but not at 50 and 65 days of age. As indicated by brain/body ratios, myelin composition and GAD activity, nutritional rehabilitation led to almost complete recovery of brain maturity, but the activity of CNP remained lower in the experimental group after rehabilitation.  相似文献

To determine if photoperiod and food intake influenced age and body composition at vaginal opening (VO), 80 mice were exposed to either 18L:6D (LL) or 6L:18D (SL) and fed either 3.64 g/mouse per day (HIGH) or 2.73 g/mouse per day (LOW) of Wayne Lab-Blox. Treatments began at weaning (21 to 23 days of age). Mice were killed at VO and analyzed for fat, water and protein content. Mice fed the HIGH ration were younger (P less than 0.01; 35.9 vs. 41.1 days) and heavier (P less than 0.001; 17.8 vs. 15.4 g) at VO than mice fed the LOW ration. Neither age nor weight at VO were affected by photoperiod. Fat content at VO was greater (P less than 0.001) for HIGH than LOW. Differences in fat percentage at VO between diet groups were greater for LL (9.7 vs. 7.0%) than for SL (9.1 vs. 8.6%), resulting in a photoperiod X ration interaction (P less than 0.01). Percent water and protein at VO were not significantly affected by photoperiod or diet. Within-group correlations between body weight and age at VO were not significant. Body water/body weight was inversely proportional to age at VO. This resulted from increased fat deposition in older animals of the LL-LOW group and increased percent protein in older animals of other groups. We concluded that dietary intake did not influence age at VO by affecting age at which a critical body weight or fat content were attained. A photoperiod of 18L:6D did not affect age at VO relative to 6L:18D.  相似文献

Over a 14-day period, male SPF Wistar rats with an initial weight of 60, 200 and 250 g (ages 30, 75 and 90 days) were given diets with a constant protein content (10% casein) and a mounting fat content (10, 15, 20, 25, 30, 40 and 50% margarine, at the expense of the saccharides in the standard diet). The utilization parameter of protein biological value (NPU) for the various diets was determined from protein intake and the body nitrogen values. The reciprocal relation of the intake of the two nutrients was determined on the basis of linearity between th growth parameter NPR and protein and fat intake and, after substituting the optimum protein intake, was used to compute the optimum fat content of the diet. From the aspect of the maximum NPU value the optimum fat contents were 30% for 30-day-old, 15% for 75-day-old and 10% for 90-day-old animals. In all three age groups (according to the growth curve for the standard Larsen diet animals with an equal growth rate of 3 g/day), protein utilization under optimum nutritional conditions was the same. The optimum fat content of the diet computed from the reciprocal relationship of fat and protein intake is in agreement with the value obtained by the biological method - 29.4% (49.5 energy %) for 30-day-old animals, 15,8% (30.4% of the total energy value of the diet) for 75-day-old animals and 11.83% (23.7% of total energy) for 90-day-old rats. By using a biological and a mathematical method, this study contributes to the determination of optimum physiological doses of nutrients.  相似文献

Growing rats were examined for the influence of a buckwheat protein diet on muscle weight and protein. In experiment 1, the rats were fed on a diet containing either casein or a buckwheat protein extract (BWPE) as the protein source (10%, 20% or 30%) for 5 wk. The relative weights (g per kg of body wt) of the gastrocnemius, plantaris and soleus muscles were higher in the BWPE-fed animals than in the casein-fed ones, but were unaffected by the dietary level of protein. These differences were not associated with growth. In experiment 2, the rats were fed on either a casein or BWPE diet at the 20% protein level for 5 wk. BWPE intake significantly elevated the gastrocnemius muscle weight, carcass protein and water, and reduced carcass fat. These results demonstrate that BWPE consumption causes muscle hypertrophy, elevates carcass protein and water, and reduces body fat.  相似文献

The time-course of changes in total amount of proteins of sarcoplasmic, myofibrillar and stromal fractions in muscle of the rats fed a protein-free diet for 8, 16, 24 and 32 days, together with the referential data of those changes in the rats fed a protein-free diet up to time of death and a 60% casein diet for 12 days was determined respectively. The results were as follows: (1) The sarcoplasmic and the myofibrillar fractions decreased much more than the stromal fraction in the earlier stages of protein depletion following the same pattern as seen in reserve proteins. (2) The sarcoplasmic fraction decreased slightly more than the myofibrillar fraction as early as 8 days of the depletion, but the relative proportion between these two fractions was thereafter almost the same as that of the standard diet group. (3) In rats fed a 60% casein diet, the sarcoplasmic fraction increased markedly than the others.  相似文献

Effects of protein malnutrition on adipose tissue development were studied in weanling male Sprague-Dawley rats fed isocaloric diets ad libitum containing either 22% (controls) or 8% (protein-malnourished rats) casein, and in rats pair-fed to the protein-malnourished rats with the 22% casein diet. After 32 days on the diet, protein-malnourished rats were 37% and pair-fed 67% the weight of the controls, while torso length was 37% and 73% of controls, respectively. Food consumption relative to body weight was greatest in protein-malnourished rats. Compared to control rats, the distal epididymal adipocyte number in the protein-malnourished rats was decreased in proportion to the decrease in body size and was more closely related to the protein intake than to the total calories consumed. After 32 days on diet, mean adipocyte number per 2 distal pads was 11.7 x 10(6) in controls and 4.3 x 10(6) in protein-malnourished rats. In pair-fed rats, cell number lagged behind controls at 4 and 11 days, but was normal at 32 days (11.4 x 10(6) cells). The distal epididymal pad adipocyte size and percent lipid were similar in all groups during the first 25 days of dietary treatment. Adipocyte size was increased significantly in controls at day 32 compared to the other two groups. At each time studied through day 25 on diet, epididymal pad weight was related to the adipose cell number rather than the cell size. It is concluded that severe restriction of dietary protein during the postweaning period of growth in rats results in decreased epididymal adipocyte proliferation and/or differentiation concomitant with generalized growth retardation, whereas isocaloric feeding of a diet of normal protein content is associated with only a transient delay in adipose tissue development.  相似文献

Role of muscle loss in the age-associated reduction in VO2 max   总被引:6,自引:0,他引:6  
A progressive decline in maximal O2 consumption (VO2max) expressed traditionally as per kilogram body weight generally occurs with advancing age. To investigate the extent to which this decline could be attributable to the age-associated loss of metabolically active tissue, i.e., muscle, we measured 24-h urinary creatinine excretion, an index of muscle mass, in 184 healthy nonobese volunteers, ages 22-87 yr, from the Baltimore Longitudinal Study of Aging who had achieved a true VO2max during graded treadmill exercise. A positive correlation was found between VO2max and creatinine excretion in both men (r = 0.64, P less than 0.001) and women (r = 0.47, P less than 0.001). As anticipated, VO2max showed a strong negative linear relationship with age in both men and women. Creatinine excretion also declined with age in men and women. When VO2max was normalized for creatinine excretion, the variance in the VO2max decline attributable to age declined from 60 to 14% in men and from 50 to 8% in women. Thus comparing the standard age regression of VO2max per kilogram body weight with that in which VO2max is normalized per milligram creatinine excretion, the decline in VO2max between a hypothetical 30 yr old and a 70 yr old was reduced from 39 to 18% in men and from 30 to 14% in women. We conclude that in both sexes, a large portion of the age-associated decline in VO2max in non-endurance-trained individuals is explicable by the loss of muscle mass, which is observed with advancing age.  相似文献

Two feeding experiments were carried out with castrated male pigs weighing between 10 and 30 kg to study acute and persisting dietary effects on growth and on protein and energy metabolism in growing pigs. Pigs were fed semi-synthetic isoenergetic and isonitrogenous diets at 50% protein requirement with either soy protein isolate (SPI) or casein (CAS) as sole protein source. Intake of protein and ME amounted to 9% w/w and 1800 kJ x kg BW (-0.62) x d(-1) in Exp. 1, respectively, and 9% w/w and 1430 kJ x kg BW(-0.62) x d(-1) in Exp. 2. The CAS diet was supplemented by Lys, Met, Thr and Trp. In Exp. 1 (acute effects), 18 pigs received the CAS diet for 24 days (period 1); 9 pigs were then switched to a SPI diet whereas 9 pigs continued on the CAS diet for another 31 days (period 2). In Exp. 2, a third period of 31 days was added in which the SPI group was switched back to CAS diet. The control group was fed on the CAS diet throughout Exp. 2 (86 days). Altogether the majority of parameters were not affected neither comparing SPI with CAS in Exp. 1 nor inspecting possible persistence of effects in Exp. 2. In detail, in Exp. 1 SPI compared to CAS feeding resulted in a lower efficiency of protein utilisation and lower protein retention. Attendant upon the lower protein retention an increased energy retention as fat was only observed in tendency. SPI feeding caused a decreased body weight, thyroid weight and increased hepatic carbohydrate content that persisted after the diet was changed back to CAS (Exp. 2).  相似文献

Changes in plasma lipid and apolipoprotein profiles were evaluated in 12 healthy, unfit subjects (VO(2peak) 39.1+/-2.8 ml.kg(-1).min(-1); 5 women, 7 men) at baseline and following endurance exercise training. The exercise protocol consisted of a 6-week endurance exercise training program (4-5 days week(-1); 60 min.session(-1); > or =65% HR(max)). Subjects were randomly assigned to consume an egg- (n=6; 12 eggs.week(-1)) or no-egg (n=6; 0 eggs.week(-1))-based, eucaloric, standardized diet for 8 weeks. Both diets were macronutrient balanced [60% carbohydrate, 30% fat, 10% protein (0.8 g.kg(-1).day(-1))] and individually designed for weight maintenance. Plasma lipids were measured twice within the same week at baseline and following exercise training. At baseline, subjects were normolipidemic with values of 163.9+/-41.8, 84.8+/-36.7, 60.6+/-15.4 and 93.1+/-52 mg dl(-1) for total cholesterol, LDL cholesterol and HDL cholesterol and triglyceride concentrations, respectively. A two-way ANOVA was used to analyze diet and exercise effects and interactions. In both groups, endurance exercise training resulted in a significant 10% increase in HDL-C (P<.05), a 19% decrease in Apo B concentrations (P<.05) and reductions in plasma CETP activity (P<.05). Plasma LDL-C decreased by 21% (P=.06). No main effects of diet or interactions with plasma lipids or Apo B concentrations were observed. These data demonstrate that endurance training improved the plasma lipid profiles of previously unfit, normolipidemic subjects independent of dietary cholesterol intake from eggs.  相似文献

A study of exercise performance was carried out in 17 obese girls and young adults. During submaximal steady-state bicycle exercise oxygen intake (Vo2) for a given work output (W) was raised in obese subjects but minute ventilation at a fixed carbon dioxide output, gas exchange, blood gases, and cardiac output at a given VO2 were similar to the values previously found for normals. In obese subjects high levels of VO2 for fixed W were also obtained on the treadmill but when these were standardized for body weight (unlike the bicycle test) it was shown that the obese girls and women exercised within the normal (expected) range of aerobic energy expenditure. During maximal performance the absolute VO2 max was the same in obese and nonobese subjects but for a given body weight, lean body mass, and leg muscle (plus) bone volume, VO2max was reduced by 23.8, 16.3, and 24.5% respectively, in the former group. It was concluded that obesity though having minimal affect on responses to submaximal exercise is nevertheless associated with a marked reduction in physiological performance at or near maximal effort.  相似文献
