The effects of the temperature-moisture factors on the phylloplane yeast communities inhabiting Sphagnum mosses were studied along the transition from a boreal forest to a swamp biotope at the Central Forest State Biosphere Reserve (Tver region, Russia). We tested the hypothesis that microclimatic parameters affect yeast community composition and structure even on a rather small spatial scale. Using a conventional plating technique we isolated and identified by molecular methods a total of 15 species of yeasts. Total yeast counts and species richness values did not depend on environmental factors, although yeast community composition and structure did. On average, Sphagnum in the swamp biotope supported a more evenly structured yeast community. Relative abundance of ascomycetous yeasts was significantly higher on swamp moss. Rhodotorula mucilaginosa dominated in the spruce forest and Cryptococcus magnus was more abundant in the swamp. Our study confirmed the low occurrence of tremellaceous yeasts in the Sphagnum phyllosphere. Of the few isolated ascomycetous yeast and yeast-like species, some were differentiated from hitherto known species in physiological tests and phylogenetic analyses. We describe one of them as Candida sphagnicola and designate KBP Y-3887T (=CBS 11774T = VKPM Y-3566T = MUCL 53590T) as the type strain. Anastrepha fruit flies (Diptera: Tephritidae) are important pests of several crops in the Americas. Alkaline hydrolysis of proteins represents a potential source of novel attractants. Previous studies revealed a higher response by Anastrepha obliqua (Macquart) to alkaline hydrolyzed torula yeast compared with other attractants. Under field conditions, captures of Anastrepha fraterculus (Wiedemann), Anastrepha ludens (Loew), and Anastrepha serpentina (Wiedemann) were significantly higher in traps baited with alkaline hydrolyzed torula yeast than in traps baited with acid hydrolyzed protein (Captor) + borax. Fly attraction to alkaline hydrolyzed torula yeast varied with species but was higher in the 1st week and decreased after 2 or 3 weeks of use in the field. The release of ammonia gas from alkaline hydrolyzed torula yeast and CeraTrap was reduced after 3 weeks, but CeraTrap remained effective in the capture of flies whereas alkaline hydrolyzed torula yeast did not. No improvement in fly captures was observed for aged alkaline hydrolyzed torula yeast when 1% ammonium acetate was added to traps. The addition of propylene glycol or benzalkonium chloride did not improve trap captures over time. Although CeraTrap has advantages in attractiveness and durability, the attraction, simplicity, and low cost make alkaline hydrolyzed torula yeast a promising attractant when compared with the standard Captor + borax in short-term monitoring programs targeted at Anastrepha pests.  相似文献

A strain of yeast isolated from insect frass collected in Thailand was found to represent a hitherto undescribed species of a basidiomycetous anamorphic genus Trichosporon. It is described as Trichosporon siamense. In the phylogenetic tree based on the D1/D2 region sequences of 26S rDNA, this yeast constitutes a cluster with several Q-9 having species of Trichosporon including T. otae and T. brassicae but is clearly differentiated from these species by 1.8% or more base substitutions. In the internal transcribed spacer region (ITS1 and ITS2), this species differs from T. scarabaeorum, the nearest species, by 6.5% base substitution.  相似文献

The effect of different yeast variants of the yeast Saccharomyces cerevisiae on the outcome of competition between Drosophila simulans and D. melanogaster was investigated. In all experiments differential birth rate was the effective mode of species competition. Addition of live yeast and especially mixtures of yeast strains improved the competition situation of a species, but could not prevent the extinction.  相似文献

Summary A new budding yeast species isolated from soil is described. Its outstanding features are, firstly, the formation of asci containing up to sixteen long oval to reniform ascospores and, secondly, a fermentative as well as oxidative metabolism. The assimilation of nitrate is absent and no pseudomycelium is formed. The taxonomic position of the yeast is discussed and it is pointed out that, due to its exceptional ascospore number (1–16), it cannot be classified in any of the existing fermentative genera of theEndomycetaceae (in sensu Lodder et Kreger-van Rij). The reniform shape of its ascospores indicates, however, its close relationship with the multispored genusKluyveromyces, on the one hand, and the newly proposed one to four-spored genusDekkeromyces on the other. The species is provisionally classified as aKluyveromyces species,Kluyveromyces africanus nov. spec., until further information regarding its sexual characteristics becomes available. By virtue of its more or less intermediate ascospore number, it establishes the direct derivation of the genusDekkeromyces fromDipodascus uninucleatus via the multispored yeast genusKluyveromyces.  相似文献

Cocultivation of Pleurotus ostreatus with eight yeast species were investigated on water agar. Special mycelial structures contacting with yeast cells were found in such cultures: nipple-like appendages and coralioid hyphae. Three out of eight species, Hanseniaspora uvarum, Rhodotorula minuta, and Saccharomyces cerevisiae were identified as trophic preferendum for P. ostreatus. These three yeast species were used for mushroom cultivation on sunflower seed peel. The biomass of fruiting bodies increased by 52.8–75.7% with the H. uvarum and S. cerevisiae suspension presence in the substrate.  相似文献

Abstract

In this study, an antagonistic yeast isolate, Wickerhamiella versatilis was considered as a promising biocontrol agent against Pectobacterium carotovorum subsp. Carotovorum (Pcc) the causal agent of soft rot disease of potato. Antagonistic yeast inhibited the growth of Pcc in vitro, and reducing the soft rot severity of infected potato tubers (cv. Diamant) under greenhouse conditions. Consequently, cellulase and pectinase hydrolytic activities in infected potato tubers with yeast?+?Pcc were decreased compared with infected tubers with Pcc. The histological characterization of treated potato tubers with antagonistic yeast W. versatilis using scanning electron microscope showed the accumulation of extracellular substances that may induce plant resistant and protects potato tubers from hydrolysis and damages. This study may introduce the possibility of using the antagonistic yeast isolate, as a biocontrol agent against soft rot of potato tubers.  相似文献

Two strains of a basidiomycetous yeast were derived from an insect trypanosomatid culture isolated from the intestine of a plant bug, Collaria oleosa (Heteroptera: Miridae), collected in Costa Rica. The yeast did not form ballistoconidia but reproduced only by budding. Teliospores were not observed in individual and crossed cultures of each strain. Morphological and other taxonomic characteristics of the yeast were similar to those of the species in the polyphyletic genus Rhodotorula. However, molecular phylogeny inferred from the internal transcribed spacers and D1/D2 region of the large subunit rRNA gene showed that the strains represent a new species placed among the smut fungi in the family Ustilentylomataceae, which includes Aurantiosporium subnitens, Fulvisporium restifaciens, Ustilentyloma fluitans, and Rhodotorula hordea. Given the well distinguished phylogenetic position of this novel species within the Ustilentylomataceae, we propose Microbotryozyma collariae gen. nov., sp. nov. to accommodate the yeast isolated from C. oleosa, with strain American Type Culture Collection MYA-4666T (= PRA303-1S = CBS 12537) designated as the type strain.  相似文献

A bacterium growing inside yeast cytoplasm was observed by light microscope without staining. The bacterium was separately stained from yeast cell by a fluorescent dye, 4′,6-diamidino-2-phenylindole (DAPI). The bacterium actively moved inside yeast cytoplasm and propagated in company with the yeast growth. The bacterium was separated from the yeast cytoplasm by selective disruption of yeast cells and the yeast without the intracellular bacterium (YWOB) was obtained by selective inactivation of bacterial cells. The yeast and the intracellular bacterium were identified as Candida tropicalis and Microbacterium sp., respectively. The length of Microbacterium sp. and C. tropicalis measured with SEM image was smaller than 0.5 μm and was larger than 5 μm, respectively. The yeast with the intracellular bacterium (YWIB) grew in a starch-based medium but the YWOB was not C. tropicalis has neither extracellular nor intracellular saccharification enzyme. Glucose was produced from starch by the extracellular crude enzyme (culture fluid) of Microbacterium sp. YWIB produced significantly more ethanol from glucose than YWOB but did not from starch. Conclusively, C. tropicalis is thought to catabolize starch dependent upon Microbacterium sp. growing in its cytoplasm and furnish stable habitat for the Microbacterium sp.  相似文献

Phylogenetic relationships among those yeast species that form saturn-shaped ascospores and which are assigned to the generaWilliopsis andPichia were estimated from their extent of nucleotide sequence divergence in three regions of ribosomal RNA. ThePichia species (P. dispora, P. saitoi, P. zaruensis andP. sp. nov.) are a closely clustered group only distantly related toWilliopsis, and it is proposed that they be reassigned toSaturnospora gen. nov. Aims: To characterize and identify a novel contaminant of aviation fuel. Methods and Results: Micro‐organisms (yeasts and bacteria) were isolated from samples of aviation fuel. A yeast that proved to have been unrecorded previously was isolated from more than one fuel sample. This novel yeast proved to be a new species of Candida and is described here. Ribosomal RNA gene sequence analyses of internal transcribed spacer (ITS) regions (including 5·8S subunit) plus the 26S D1/D2 domains showed the strains to cluster within the Candida membranifaciens clade nearest to, but distinct from, Candida tumulicola. Phenotypic tests were identical for both isolates. Physiological and biochemical tests supported their position as a separate taxon. The yeast was assessed for its effect on the main constituent hydrocarbons of aviation fuel. Conclusions: Two strains (IMI 395605T and IMI 395606) belonging to the novel yeast species, Candida keroseneae, were isolated from samples of aircraft fuel (kerosene), characterized and described herein with reference to their potential as contaminants of aviation fuel. Significance and Impact of the Study: As a result of isolating a novel yeast from aviation fuel, the implications for microbial contamination of such fuel should be considered more widely than previously thought.  相似文献

Sporisorium reilianum f. sp. zeae is the causal agent of head smut of maize. Although the main symptom of this disease is the formation of a black fungal sorus on the reproductive parts of the maize, the infection always occurs via the roots. Early infection stages are characterised by a hyphal proliferation of the fungus around the roots. In this paper, we describe effects of a fraction extracted from maize root exudates on growth of S. reilianum f. sp. zeae. The fungus grew as a yeast form on artificial medium, but in presence of these fractions, some yeasts switched to a hyphal form. In addition, an increased proliferation of the yeast form was also observed with exudates from a variety of maize susceptible to head smut. In the presence of exudates obtained from a tolerant variety of maize, proliferation of the yeast form was inhibited, whereas the induction of yeast-hypha transition was always observed. These results indicated that some molecules in root exudates could play a role in the pre-infectious stage between maize and S. reilianum f. sp. zeae.  相似文献

Summary A new haploid, homothallic yeast isolated from the larval feed ofXylocopa caffra is described. A study of its properties shows that the evolution of its sexual and biochemical characteristics has not taken place at the same rate as in the genusHansenula and the proposed genusDekkeromyces. This led the author to the conclusion that in the yeast domain phylogenetic lines may exist in which a unilateral evolution of biochemical characteristics has taken place. At least thre groups of yeasts exist where such discrepancies exist between the evolution of their sexual and biochemical characteristics. The inclusion of the new species in these groups is discussed. The new species has provisionally been classified in the genusPichia (in sensu Phaff). The role of chemotherapy, X-irradiation and a combination of both on the phenomenon of adherence of yeast to buccal epithelial cells (BEC) was investigated in vitro. Growth of three Candida spp. in the presence of eight of eleven antineoplastic agents led to reduction of adherence of the isolates tested (reduction between 30% and 61% of the control value), and this effect was observed whether exponential or stationary phase Candida cells were used. Exposure of C. albicans to various doses of radiation also led to a reduction in adherence of this yeast to BEC between 31% and 53% of the control value. This reduction was shown to be dose related. Similar results were obtained when BEC were exposed to radiation, and the effect of radiation treatment was accentuated when both yeast and BEC were irradiated simultaneously. Furthermore, treating C albicans with a combination of chemotherapy and radiation led to the greatest reduction in adherence of yeast to BEC compared to when the yeast was treated with either chemotherapy or radiation alone (reduction between 63% to 74% as compared with control). The possible mechanism/s involved in reduction of adherence of yeast to BEC are discussed.  相似文献
