The survival of Rhizobium leguminosarum biovar phaseoli on seeds of bean was tested, using the cultivar Carioca. The seeds were treated seven days before inoculation with Benlate, Vitavax, Banrot, Difolatan or Ridomil fungicides. The rhizobial strains used were: CIAT 899, CPAC 1135 and CIAT 652. Strain CIAT 899 showed greater survival on the seed with fungicide than the other strains. Two hours after the contact with fungicides strains CIAT 652 and CPAC 1135 had significantly lower numbers of rhizobia than the treatment without fungicide. The Benlate and Banrot fungicides had the greatest effect on survival of rhizobial strains. There was a drastic mortality of the two strains, CIAT 652 and CPAC 1135, on seeds treated with Benlate and Ridomil. Under field conditions, granular inoculation produced fewer nodules, but a similar total nodule weight as seed inoculation. Serological tests (ELISA) showed that seed treatment with Benlate in connection with seed inoculation reduced drastically the occurrence of inoculated strains in nodules, while the same fungicide treatment and inoculation applied in the seed furrow did not affect the survival of the inoculated strain.  相似文献

This paper examines the importance of salt tolerance of host cultivars, Bradyrhizobium strains, and host-Bradyrhizobium combinations on the symbiotic nitrogen fixation potential of soybean under NaCl and KCl salt stress. Plants were grown in a soil medium, and the experiments were conducted under controlled environment growth room conditions. Bradyrhizobium growth was examined in yeast-mannitol broth andB. japonicum strains tolerant of NaCl and KCl (80 mM) stress were identified. Soybean cultivar Williams, which was sensitive to salt stress, performed poorly both in growth and symbiotic nitrogen fixation, irrespective of whether it was matched with a tolerant or sensitive Bradyrhizobium strain. Tolerant cultivar Manchu sustained nodulation and nitrogen fixation, irrespective of whether it was matched with a tolerant or sensitive Bradyrhizobium strain. Evidence presented here suggests a need, first to select soybean cultivars that are tolerant to salt stress, and then to match them with tolerant and effective Bradyrhizobium strains.  相似文献

Common bean plants inoculated with salt-tolerant Rhizobium tropici wild-type strain CIAT899 formed a more active symbiosis than did its decreased salt-tolerance (DST) mutant derivatives (HB8, HB10, HB12 and HB13). The mutants formed partially effective (HB10, HB12) or almost ineffective (HB8, HB13) nodules (Fix(d)) under non-saline conditions. The DST mutant formed nodules that accumulated more proline than did the wild-type nodules, while soluble sugars were accumulated mainly in ineffective nodules. Under salt stress, plant growth, nitrogen fixation, and the activities of the antioxidant defense enzymes of nodules were affected in all symbioses tested. Overall, mutant nodules showed lower antioxidant enzyme activities than wild-type nodules. Levels of nodule catalase appeared to correlate with symbiotic nitrogen-fixing efficiency. Superoxide dismutase and dehydroascorbate reductase seem to function in the molecular mechanisms underlying the tolerance of nodules to salinity.  相似文献

Plasmid profile analysis is useful to characterize Rhizobium strains within the same species. Among the 16 Rhizobium strains examined, 14 had distinct plasmid profiles. The size of plasmids ranged from 40 to 650 kb, and three plasmids of 650, 510 and 390 kb were common to several strains. Plasmid analysis revealed that Rhizobium etli contained a mega-plasmid, similar in size to Rhizobium tropici. All the salt-tolerant strains examined had a plasmid of 250 kb, except for strain EBRI 29. This suggests that this plasmid may play an important adaptive role under salt stress conditions.  相似文献

Fifteen genotypes of faba bean (Vicia faba L.) were inoculated with salt-tolerant Rhizobium leguminosarum biovar. viciae strain GRA 19 in solution culture with 0 (control) and 75 mM NaCl added immediately after transplanting. Genotypes varied in their tolerance of high levels of NaCl. Physiological parameters (dry weight of shoot and root, number and dry weight of nodules) were not affected by salinity in lines VF46, VF64 and VF112. Faba bean line VF60 was sensitive to salt stress. Host tolearance appeared to be a major requisite for nodulation and N2 fixation under salt stress. Tolerant line VF112 sustained nitrogen fixation under saline conditions. Activity of the ammonium assimilation enzymes glutamine synthetase and glutamate synthase, and soluble protein content, were reduced by salinity in all genotypes tested. A cDNA fragment encoding a common bean (Phaseolus vulgaris) proliferating cell nuclear antigen (PCNA) was isolated using rapid amplification of cDNA 3' end (3' RACE) method, cloned and sequenced. The nucleotide sequence of this clone contains an open reading frame of 798 nucleotides encoding a protein of 265 amino acids. Alignment of the common bean PCNA predicted sequence shows its high degree of identity with PCNA from other plant species. Analysis of PCNA content in the germinating embryos of common bean showed a decrease in the protein level after 60h of germination. Moreover, PCNA was not detected in the tested plant organs (root, stem, leaf and flower). The presence of PCNA in the germinating seeds and its absence from mature plants suggests that this protein plays a crucial role during early stages of plant development.  相似文献

Screening of Rhizobium leguminosarum bv. phaseoli strains showed some that were able to nodulate common beans (Phaseolus vulgaris L.) at high temperatures (35 and 38°C/8 h/day). The nodulation ability was not related to the capability to grow or produce melanin-like pigment in culture media at high temperatures. However, nodules formed at high temperatures were ineffective and plants did not accumulate N in shoots. Two thermal shocks of 40°C/8 h/day at flowering time drastically decreased nitrogenase activity and nodule relative efficiency of plants otherwise grown at 28°C. Recovery of nitrogenase activity began only after seven days, when new nodules formed; total incorporation of N in tops did not recover for 2 weeks. Non-inoculated beans receiving mineral N were not affected by the thermal shock, and when growing continuously at 35 or 38°C had total N accumulated in shoots reduced by only 18%.  相似文献

Similar ranges of gibberellins (GAs) were detected by high-performance liquid chromatography (HPLC)-immunoassay procedures in ten cultures of wild-type and mutant strains of Rhizobium phaseoli. The major GAs excreted into the culture medium were GA1 and GA4. These identifications were confirmed by combined gas chromatographymass spectrometry. The HPLC-immunoassays also detected smaller amounts of GA9- as well as GA20-like compounds, the latter being present in some but not all cultures. In addition to GAs, all strains excreted indole-3-acetic acid (IAA) but there was no obvious relationship between the amounts of GA and IAA that accumulated. The Rhizobium strains studied included nod and fix mutants, making it unlikely that the IAA- and GA-biosynthesis genes are closely linked to the genes for nodulation and nitrogen fixation.The HPLC-immunoassay analyses showed also that nodules and non-nodulated roots of Phaseolus vulgaris L. contained similar spectra of GAs to R. phaseoli culture media. The presented experiments evaluated the symbiotic performance of soybean genotypes with contrasting salt stress tolerance to arbuscular mycorrhizal fungi (AMF) inoculation. In addition, the physiological stress tolerance mechanisms in plants derived from mutualistic interactions between AMF and the host plants were evaluated. Plant growth, nodulation, nitrogenase activity and levels of endogenous growth hormones, such as indole acetic acid and indole butyric acid, of salt-tolerant and salt-sensitive soybean genotypes significantly decreased at 200 mM NaCl. The inoculation of soybean with AMF improved the symbiotic performance of both soybean genotypes by improving nodule formation, leghemoglobin content, nitrogenase activity and auxin synthesis. AMF colonization also protected soybean genotypes from salt-induced membrane damage and reduced the production of hydrogen peroxide, subsequently reducing the production of TBARS and reducing lipid peroxidation. In conclusion, the results of the present investigation indicate that AMF improve the symbiotic performance of soybean genotypes regardless of their salt stress tolerance ability by mitigating the negative effect of salt stress and stimulating endogenous level of auxins that contribute to an improved root system and nutrient acquisition under salt stress.  相似文献

A chromosomal gene, required for nodule development on Phaseolus bean, was characterized from Rhizobium etli strain TAL182. MLC640 is a Tn5 insertion mutant of TAL182 which shows decreased motility in soft TY agar and is defective in nodule development. The site of Tn5 insertion in MLC640 mapped to a 3.6-kb EcoRI chromosomal fragment. The 3.6-kb fragment was subcloned from the cosmid pUHR80 which complemented MLC640. Further subcloning and site-directed Tn5 mutagenesis localized the gene for nodule development to a 1.7-kb region within the 3.6-kb EcoRI fragment. Phaseolus vulgaris (cv. Hawkesbury Wonder) was grown over a range of NaCl concentrations (0–150 mM), and the effects on growth, ion relations and photosynthetic performance were examined. Dry and fresh weight decreased with increasing external NaCl concentration while the root/shoot ratio increased. The Cl- concentration of leaf tissue increased linearly with increasing external NaCl concentration, as did K+ concentration, although to a lesser degree. Increases in leaf Na+ concentration occurred only at the higher external NaCl concentrations (100 mM). Increases in leaf Cl- were primarily balanced by increases in K+ and Na+. X-ray microanalysis of leaf cells from salinized plants showed that Cl- concentration was high in both the cell vacuole and chloroplast-cytoplasm (250–300 mM in both compartments for the most stressed plants), indicating a lack of effective intracellular ion compartmentation in this species. Salinity had little effect on the total nitrogen and ribulose-1,5-bisphosphate (RuBP) carboxylase (EC 4.1.1.39) content per unit leaf area. Chlorophyll per unit leaf area was reduced considerably by salt stress, however. Stomatal conductance declined substantially with salt stress such that the intercellular CO2 concentration (C i) was reduced by up to 30%. Salinization of plants was found to alter the 13C value of leaves of Phaseolus by up to 5 and this change agreed quantitatively with that predicted by the theory relating carbon-isotope fractionation to the corresponding measured intercellular CO2 concentration. Salt stress also brought about a reduction in photosynthetic CO2 fixation independent of altered diffusional limitations. The initial slope of the photosynthesis versus C i response declined with salinity stress, indicating that the apparent in-vivo activity of RuBP carboxylase was decreased by up to 40% at high leaf Cl- concentrations. Two cultivars of Phaseolus vulgaris L., one responsive to colonization with microsymbionts (Mexico 309) and one less-responsive (Rio Tibagi) were grown in Leonard jars containing sand/vermiculite under greenhouse conditions. Bean plants were either left non-inoculated (controls) or were inoculated with the vesicular-arbuscular mycorrhizal (VAM) fungus Glomus etunicatum or a strain of Rhizobium leguminosarum bv. phaseoli (UMR-1899). Plants from the Mexico 309 cultivar maintained a higher growth rate, supported proportionately more nodules and mycorrhizae, and assimilated relatively more N or P when colonized by Rhizobium or Glomus, respectively, than did plants of the Rio Tibagi cultivar. Estimated specific nodule activity for Mexico 309 beans was more than twice that of Rio Tibagi, whereas the specific phosphorus uptake rate (SPUR) was 35% greater in the non-inoculated roots of Rio Tibagi compared to Mexico 309. Colonization by G. etunicatum more than doubled the SPUR for each cultivar compared to control roots. New acid phosphatase isozymes appeared in VAM-colonized roots of both cultivars compared to controls. Acid and alkaline phosphatase activities were significantly higher in G. etunicatum-colonized Mexico 309 roots, but not in Rio Tibagi mycorrhizae, compared to uninfected roots. Polyphosphate hydrolase activity was elevated in mycorrhizae of both cultivars compared to control roots. These results indicate that the dependence of a host on a specific endophyte increases when there are limitations to the supply of a nutrient that the endophyte can provide. The greater the increase in absorption or utilization capacity following colonization by the microsymbiont, the greater the dependence by the host. More importantly, identification of enzymatic activities that influence these plant-microbe associations opens the possibility that the specific genes that code for these enzymes could be targeted for future manipulation.  相似文献

pIJ1008, a Rhizobium leguminosarum plasmid which determines hydrogen uptake ability and symbiotic functions in pea was transferable to three of seven natural isolates of R. meliloti tested. In these three strains, pIJ1008 was maintained stably with the respective sym megaplasmid indigenous to each R. meliloti strain. These strains carrying both plasmids nodulated alfalfa but not pea. By reisolation and examination of the strains from alfalfa nodule tissue, it was shown that pIJ1008 continued to be maintained but that pea-nodulation ability was suppressed.In one strain of R. meliloti which carries a 200 kb cryptic plasmid (in addition to a megaplasmid), the transfer and selection for pIJ1008 resulted in the loss of the cryptic plasmid.In three separate plant growth experiments, alfalfa nodules induced by each of the R. meliloti strain carrying both sym plasmids were assayed for hydrogen uptake activity. The average activity was 40-, 3.5-and 2-fold higher than with the respective pIJ1008-free strains. The objective of this study was to determine the role of 1-aminocyclopropane-1-carboxylate (ACC) deaminase of symbionts in nodulation and growth of Leucaena leucocephala. The acdS genes encoding ACC deaminase were cloned from Rhizobium sp. strain TAL1145 and Sinorhizobium sp. BL3 in multicopy plasmids, and transferred to TAL1145. The BL3-acdS gene greatly enhanced ACC deaminase activity in TAL1145 compared to the native acdS gene. The transconjugants of TAL1145 containing the native or BL3 acdS gene could grow in minimal media containing 1.5mM ACC, whereas BL3 could tolerate up to 3mM ACC. The TAL1145 acdS gene was inducible by mimosine and not by ACC, while the BL3 acdS gene was highly inducible by ACC and not by mimosine. The transconjugants of TAL1145 containing the native- and BL3-acdS genes formed nodules with greater number and sizes, and produced higher root mass on L. leucocephala than by TAL1145. This study shows that the introduction of multiple copies of the acdS gene increased ACC deaminase activities of TAL1145 and enhanced its symbiotic efficiency on L. leucocephala.  相似文献

Two sets of experiments to determine the effect of mycorrhiza on soybean (Glycine max) growth under saline conditions and to investigate the salt acclimation of mycorrhizal fungi were conducted. In the first experiment, the effect of an arbuscular mycorrhizal (AM) fungus Glomus etunicatum on mineral nutrient, proline and carbohydrate concentrations and growth of soybean. Under different NaCl concentrations (0, 50, 100, 150 and 200mM) was evaluated. Salinity decreased AM colonization. In both the M and nonAM plants shoot and root proline and shoot Na and Zn concentrations were increased under salinity. Soybean plants inoculated with the AM fungus had significantly higher fresh and dry weight, root proline, P, K and Zn but lower shoot proline and Na concentrations compared to the non inoculated plants. In the second experiment, the AM fungus was pre-treated with NaCl (salt acclimation) then was used as inoculum for soybean plants subjected to 100mM NaCl. Root colonization, fresh and dry weight, root proline, P, K and Zn concentrations were greater in soybean plants inoculated with the salt pre-treated fungus, compared to those inoculated with the nonsalt pre-treated fungus. However, for Na, the situation was the opposite. Based on these results, the AM inoculation helps the growth of soybean plants grown in saline conditions. When the AM fungus was pre-treated with NaCl with a gradual increase of concentration, and then exposed to a sudden salt stress, their efficiency was increased. This may be due to the acclimation of the AM fungus to salinity.  相似文献

A field experiment under rainfed conditions was conducted in Durango, México, to assess N2-fixation of three cultivars of common bean (Phaseolus vulgaris L.) using 15N-methodology. In addition, diversity of rhizobial isolates obtained from nodules of the different plant genotypes was evaluated by intrinsic antibiotic resistance (IAR), PCR using enterobacterial repetitive intergenic consensus (ERIC) primers, PCR-RFLP analysis of the 16S rRNA gene and multilocus enzyme electrophoresis (MLEE). Selected isolates were used to determine acetylene reduction and competitive ability under greenhouse conditions. The three cultivars tested did not show high variation in N2-fixation, the %Ndfa values ranged from 19 to 26%. Variability in N2-fixation efficiency among various native rhizobial isolates was very high and our results indicate that differences in competitive abilitiy exist also. PCR-RFLP of the 16S rRNA gene and MLEE revealed that most of the isolates belong to the species Rhizobium etli. Intrinsic antibiotic resistance analysis and ERIC-PCR showed high diversity among isolates. In contrast, our results using MLEE show low genetic diversity (H = 0.105).  相似文献

To analyse nodular antioxidant enzyme expression in response to salt stress, Phaseolus vulgaris genotype BAT477 was inoculated with reference strain CIAT899, and treated with 50 mM NaCl. Plant growth, nodulation and nitrogen fixing activity were analysed. Results showed that: (1) all parameters, particularly in nodules, were affected by salt treatments, and (2) confirmed preferential growth allocation to roots. The ARA was significantly decreased by salt treatments. Protein dosage confirmed that nodules were more affected by salt treatment than were roots. We analysed superoxide dismutase, catalase, ascorbate peroxidase and peroxidase in nodules, roots and a free rhizobial strain. Our results indicated that SOD and CAT nodular isozymes had bacterial and root origins. The SOD expressed the same CuZn, Fe and Mn SOD isoforms in nodules and roots, whereas in free rhizobia we found only one Fe and Mn SOD. APX and POX nodule and root profiles had only root origins, as no rhizobial band was detected. Under salt stress, plant growth, nitrogen fixation and activities of antioxidant defense enzymes in nodules were affected. Thus, these enzymes appear to preserve symbiosis from stress turned out that NaCl salinity lead to a differential regulation of distinct SOD and POX isoenzyme. So their levels in nodules appeared to be consistent with a symbiotic nitrogen fixing efficiency hypothesis, and they seem to function as the molecular mechanisms underlying the nodule response to salinity.  相似文献

Two plasmids, pAgK84::Tn5-Mob from Agrobacterium radiobacter carrying genes for the production of agrocin 84, and RP4-4 from E. coli were inserted either separately or together into a strain of Rhizobium meliloti. Each of these plasmid-containing R. meliloti transconjugants was less effective than the wild type strain in their ability to fix nitrogen in Medicago tornata. The pAgK84::Tn5-Mob-containing transconjugant was significantly less effective than that containing RP4-4. The transconjugant strains were inferior to the wild type strain in their ability to nodulate seedlings and to compete for nodulation.  相似文献
