Photoinhibition is a state of physiological stress that occurs in all oxygen evolving photosynthetic organisms exposed to light. The primary damage occurs within the reaction center of Photosystem II (PS II). While irreversible photoinduced damage to PS II occurs at all light intensities, the efficiency of photosynthetic electron transfer decreases markedly only when the rate of damage exceeds the rate of its repair, which requires de novo PS II protein synthesis. Photoinhibition has been studied for over a century using a large variety of biochemical, biophysical and genetic methodologies. The discovery of the light induced turnover of a protein, encoded by the plastid psbA gene (the D1 protein), later identified as one of the photochemical reaction center II proteins, has led to the elucidation of the underlying mechanism of photoinhibition and to a deeper understanding of the PS II `life cycle.' This brief historical development of the biology of the mycoplasmas begins with their discovery in 1898 to the present. Mycoplasmas are the smallest free-living microorganisms and for years were thought to be viruses because they passed through the usual bacterial filters. They lack a cell wall, are widespread in nature and many are animal, plantand human pathogens. The extensive use of cell cultures in the last fifty years and their frequent contamination with mycoplasmas, together with their possession of the smallest genome of any free-living organism, has drawn enormous attention to these organisms and has revealed considerably more about their biology.  相似文献

This paper asks a simple question: why did Western and other European politicians become so alarmed and, in some cases, downright apocalyptic at the rise of asylum seekers in 2014–16, especially compared to the previous refugee crisis in the 1990s? This paper argues that in 2014/2015, a “perfect storm” developed, bringing together factors that in the past had been largely unrelated and then converged with new ones. Peeling the onion of societal discontent with migrants and refugees has revealed five necessary and sufficient conditions: (1) discomfort with immigration and integration of colonial and labour migrants from North Africa and Turkey (1970–80s); (2) growing social inequality and widespread pessimism about globalization (1980s–); (3) A growing discomfort with Islam (1990s–); (4) Islamist terrorism (2000s–) and (5) the rise of radical right populist parties (2000s).  相似文献

 One hundred years ago, the developmental origin of endosperm from double fertilization was discovered independently by Navashin and Guignard. For much of the twentieth century, specific events related to the evolutionary origin of the endosperm of flowering plants remained a mystery. However, during the past 20 years, advances in phylogenetic reconstruction of seed plants, genetic theory associated with kin selection, and comparative study of the reproductive biology of the closest living relatives of angiosperms (Gnetales) have advanced our understanding of the evolutionary events associated with the origin of double fertilization and endosperm. Recent developmental analyses of Ephedra and Gnetum (members of Gnetales) indicate that these nonflowering seed plants undergo a regular process of double fertilization that yields two diploid zygotes. Use of explicit genetic and developmental criteria for analysis of evolutionary homology demonstrates congruence with the hypothesis that double fertilization processes in Gnetales and angiosperms were inherited from a common ancestor of the two lineages. In its rudimentary form, the second fertilization event in the ancestors of flowering plants yielded a supernumerary diploid embryo that was genetically identical to the normal embryo, a process most similar to what occurs in extant Ephedra. Subsequent to the divergence of the angiosperm stem lineage, the supernumerary embryo derived from double fertilization was developmentally modified into an embryo-nourishing structure, endosperm, that now characterizes angiosperms. The meaning of the word homology has changed. From being a comparative concept in pre-Darwinian times, it became a historical concept, strictly signifying a common evolutionary origin for either anatomical structures or genes. This historical understanding of homology is not useful in classification; therefore I propose a return to its pre-Darwinian meaning.  相似文献

The “aquatic phycomycetes” constitute an ecologically and economically important assemblage of eukaryotic microorganisms, because they share many morphological traits and important ecological functions and they interact with each other in aquatic ecosystems. The last two decades of research have provided both molecular and structural evidence that the “aquatic phycomycetes” are a diverse, polyphyletic grouping and therefore not a valid taxonomic entity. Very little research has been conducted for many years with the “aquatic phycomycetes”, possibly because in general these microorganisms are often hard to isolate and maintain in most laboratory facilities, little background data has been available to identify the species, the studies were time consuming, the state of the art technology in many cases did not permit studies on these groups and they were thought to be economically and ecologically unimportant. However, this perception has changed recently. For example, some of these species (1) are now known to play important roles in biomass conversion and sequestration of CO2, (2) are parasites of many fungal, plant and animal species, (3) may harbor genes of important enzymes for industrial applications and (4) can serve as indicator species for eco-tox monitoring. This review discusses the assemblages of microorganisms which Sparrow placed into the aquatic phycomycetes, their history in brief and their current phylogenetic placement.  相似文献

This article continues the series of Surveys and Summaries on restriction endonucleases (REases) begun this year in Nucleic Acids Research. Here we discuss ‘Type II’ REases, the kind used for DNA analysis and cloning. We focus on their biochemistry: what they are, what they do, and how they do it. Type II REases are produced by prokaryotes to combat bacteriophages. With extreme accuracy, each recognizes a particular sequence in double-stranded DNA and cleaves at a fixed position within or nearby. The discoveries of these enzymes in the 1970s, and of the uses to which they could be put, have since impacted every corner of the life sciences. They became the enabling tools of molecular biology, genetics and biotechnology, and made analysis at the most fundamental levels routine. Hundreds of different REases have been discovered and are available commercially. Their genes have been cloned, sequenced and overexpressed. Most have been characterized to some extent, but few have been studied in depth. Here, we describe the original discoveries in this field, and the properties of the first Type II REases investigated. We discuss the mechanisms of sequence recognition and catalysis, and the varied oligomeric modes in which Type II REases act. We describe the surprising heterogeneity revealed by comparisons of their sequences and structures.  相似文献

Epiascidiate leaves are those foliar organs whose adaxial (ventral) side is the inside of a tube. Such tubular leaves are found in Nepenthaceae, Sarraceniaceae, Cephalotaceae, and Lentibulariaceae. Throughout botanical history these leaves have received considerable attention because of their bizarre morphology and problems of interpretation. This paper documents the attempts of the last 150 years to correctly understand their organographic nature. All epiascidiate foliar organs are structurally similar in their early ontogeny, each forms a distinctive adaxial outgrowth (Querzone), and the diverse morphologies of the mature organs seem to be modifications upon a similar primordial ground plan. Typologically these leaves are directly related to peltate leaves and phyllodes (non-petiolar,sensu Boke). Except for certain, highly speculative,de novo theories, such as the “foliar runner” theory of Croizat, all misinterpretations of the nature of epiascidiate leaves are directly attribuable to earlier errors in ascertaining the organography of typologically related leaf forms. Accordingly, the sympodial tubular leaf (Roth) is rejected, as is the petiolar nature of tubular leaves (de Candolle). The typological relationships of these leaves to unifacial foliar organs (Troll) seems well substantiated from both an organographic and a histogenetic viewpoint. The peltate carpel theory (?elakovský; Troll) is, in reality, an epiascidiate carpel theory. The idea of a fundamentally tubular carpel seems correct from both a typological and phylogenetic standpoint. To comprehend the various contradictory interpretations which have been used to explain morphologically complex problems, as well as to understand the genesis of morphological theories, it is necessary to acquire an accurate historical perspective of the subject.  相似文献

In 2017, a Special Issue of Biophysical Reviews was devoted to “Titin and Its Binding Partners. The issue contained a review: “An historical perspective of the discovery of titin filaments” by dos Remedios and Gilmour that was intended to be a history of the discovery of the giant protein titin, previously named connectin. The review took readers back to the earliest discovery of the so-called third filament component of skeletal and cardiac muscle sarcomeres and ended in 1969. Recently, my colleague Shin’ichi Ishiwata gently reminded me of two papers published in 1990 and 1993 that were unwittingly omitted from the original historical perspective. In the first paper (J Cell Biol 110:53–62, 1990), Funatsu et al. examined the elastic filaments in skeletal muscle using a combination of light and electron microscopy, but they also measured resting as well as passive stiffness mechanical measurements to establish that connectin (titin) is responsible for both stiffness and fiber tension. In the second paper (J Cell Biol 120:711–724, 1993), Funatsu et al. used permeabilised cardiac muscle myocytes (from rabbit papillary muscles) and focussed on filament ultrastructure using either freeze-substitution or deep-etched replica methods to visualise connectin/titin filaments in fibers with and without actin and myosin filaments.  相似文献

Background

Current treatment regimens for pulmonary tuberculosis require at least 6 months of therapy. Immune adjuvant therapy with recombinant interferon-γ1b (rIFN-γb) may reduce pulmonary inflammation and reduce the period of infectivity by promoting earlier sputum clearance.

Methodology/Principal Findings

We performed a randomized, controlled clinical trial of directly observed therapy (DOTS) versus DOTS supplemented with nebulized or subcutaneously administered rIFN-γ1b over 4 months to 89 patients with cavitary pulmonary tuberculosis. Bronchoalveolar lavage (BAL) and blood were sampled at 0 and 4 months. There was a significant decline in levels of inflammatory cytokines IL-1β, IL-6, IL-8, and IL-10 in 24-hour BAL supernatants only in the nebulized rIFN-γ1b group from baseline to week 16. Both rIFN-γ1b groups showed significant 3-fold increases in CD4+ lymphocyte response to PPD at 4 weeks. There was a significant (p = 0.03) difference in the rate of clearance of Mtb from the sputum smear at 4 weeks for the nebulized rIFN-γ1b adjuvant group compared to DOTS or DOTS with subcutaneous rIFN-γ1b. In addition, there was significant reduction in the prevalence of fever, wheeze, and night sweats at 4 weeks among patients receiving rFN-γ1b versus DOTS alone.

Conclusion

Recombinant interferon-γ1b adjuvant therapy plus DOTS in cavitary pulmonary tuberculosis can reduce inflammatory cytokines at the site of disease, improve clearance of Mtb from the sputum, and improve constitutional symptoms.

This article explores the history of forensic science in terms of ideologies and institutions rather than developing technique. It presents an analytical framework for characterising forensic institutions and practices, past and present. That framework highlights the distinct issues of means of witness, accredited testimony, and the reaching of juridical decisions. The article applies the framework by comparing four forensic ‘formations,’ (or ‘cultures’) which have been prominent at various times and places in the western world from the early modern period onward: these are the central European heritage of the Caroline code, a eugenically-oriented forensic enterprise of late nineteenth-century America, the forensic perspective in nineteenth-century British India, and the representation of forensic certainty in contemporary American popular culture. The article concludes with a critique of what seems an increasingly common expectation: that forensic science evolves independently of legal institutions, and can ultimately displace them.  相似文献

Prostate cancer is the second most common cancer in men and a major cause of cancer deaths worldwide. Ionizing radiation has played a substantial role in the curative treatment of this disease. The historical evolution of radiotherapy techniques through 3D-conformal radiotherapy (3D-CRT), intensity-modulated radiotherapy (IMRT), and image-guided radiotherapy (IGRT) has allowed more accurate and precise treatments toward significant improvements in the therapeutic ratio. The addition of androgen deprivation therapy has significantly improved overall survival becoming the standard therapy for intermediate- and high-risk disease. Many randomized controlled trials have shown improved local control with dose escalation, and hypofractionated RT has been consolidated with proven efficacy and safe clinical results. However, several questions remain open in the radiotherapeutic management of prostate cancer patients and hopefully ongoing studies will shed light on these uncertainties. More individualized approaches are essential through better prognostic and novel predictive biomarkers of prostate radiotherapy response. Clinicians should critically interpret the evolving technologies in prostate cancer radiotherapy with important optimism but balancing the costs and the actual magnitude of clinical benefit. This article provides an overview of the basic aspects of radiotherapy treatment in localized prostate cancer from a physician’s perspective.  相似文献

Food packaging serves purposes of food product safety and easy handling and transport by preventing chemical contamination and enhancing shelf life, which provides convenience for consumers. Various types of materials, including plastics, glass, metals, and papers and their composites, have been used for food packaging. However, owing to consumers’ increased health awareness, the significance of transferring harmful materials from packaging materials into foods is of greater concern. This review highlights the interactions of food with packaging materials and elaborates the mechanism, types, and contributing factors of migration of chemical substances from the packaging to foods. Also, various types of chemical migrants from different packaging materials with their possible impacts on food safety and human health are discussed. Ireland has a temperate climate influenced by the Atlantic Gulf Stream. This, combined with fertile soils and adequate rainfall, provides an environment ideally suited for grass. Grass is an inexpensive, nutritious feed for livestock, giving Ireland a competitive advantage in this sector. Tillage farming is also an important sector for Ireland, and evidence of the Great Irish Famine of the mid-eighteen hundreds is still visible in the landscape. In this paper, we examine aspects of meteorological involvement in Irish agriculture, from the perspective of the Irish Meteorological Service. We examine the rise of agrometeorology in Ireland and take a look at the development of related services, including potato blight warnings. Two influential characters of Irish agrometeorological history, Austin Bourke and Tom Keane, are examined, as well as organisations such as the AGMET Group. The paper concludes with a brief snapshot of current Irish agrometeorological activities along with future ambitions.  相似文献

Difficulties are encountered in the thermodynamic characterization of interactions between a protein ligand and a linear acceptor, such as a polynucleotide or a polysaccharide, because of the involvement of more than one unit of the polymer chain in each attachment of a protein molecule. Complications arise from the fact that random attachment of ligand to the polymer chain, each unit of which is a potential binding site, initially leads to suboptimal location of protein molecules along the polymer chain—a situation that has to be rectified before the attainment of thermodynamic equilibrium can be realized. Kinetic as well as thermodynamic consequences of such nonspecific binding, termed the parking problem, therefore need to be considered in any quantitative characterization of the interaction between a large ligand and a linear polymer acceptor chain. Results for the thrombin–heparin interaction have been used to illustrate a thermodynamic characterization of nonspecific binding that takes into account these consequences of the parking problem.  相似文献
