The formation of anaerobic granular sludge on a sulfate-containing waste-water was studied in up-flow anaerobic sludge blanket reactors. Three systems were examined: a sulfidogenic system, a methanogenic system and a mixed sulfidogenic/methanogenic system. No significant granulation was observed in the sulfidogenic system. For the methanogenic and the mixed methanogenic/sulfidogenic system granulation proceeded well, and no significant difference in the granule diameter could be detected. In the three systems studied, different types of sludge developed. A (mainly) methanogenic granular sludge was developed in the methanogenic system, a (more) sulfate-reducing granular sludge was developed in the mixed methanogenic/sulfidogenic system, and a flocculant sulfate-reducing sludge was developed in the sulfidogenic system. In upflow anaerobic sludge blanket (UASB) digesters treating raw sewage at low temperatures, the sludge progressively lost methanogenic activity, indicating the possibility of methanogenic activity inhibition caused by wastewater constituents. To check this fact, batch and semi-continuous methanogenic toxicity assays were carried out with raw and centrifuged sewage. Permanent methanogenic toxicity on anaerobic sludge of approximately 50% was found when the sludge exposure to wastewater was renewed in a semi-continuous way. A stronger methanogenic inhibition of about 70-100% was observed when an active anaerobic sludge was exposed to mixed liquor from the UASB digester treating municipal wastewater. Suspended solids removal from sewage slightly reduced methanogenic toxicity. Effective concentration of municipal wastewater that caused a 50% reduction in methanogenic activity was estimated to be in the range of 150-200 mg CODl(-1). As methanogenic inhibition appeared to be related to remaining COD, higher methanogenic toxicity in digesters operating with low conversion efficiency will be expected.  相似文献

After immobilization of anaerobes on polyurethane foam in a thermophilic, fixed-bed, anaerobic digester supplied with acetate, the results of real-time PCR analysis indicated that the major immobilized methanogenic archaea were Methanosarcina spp., and that the major free-living methanogenic archaea were Methanosarcina and Methanobacterium spp. 16S rRNA gene densities of Methanosarcina spp. and Methanobacterium spp. immobilized on the polyurethane foam were 7.6x10(9) and 2.6x10(8) copies/cm3, respectively. Immobilized methanogenic archaea could be concentrated 1000 times relative to those in the original anaerobically digested sludge from a completely mixed thermophilic digester supplied with cattle waste. On the other hand, immobilized bacteria could be concentrated only 10 times. The cell densities of the immobilized methanogenic archaea and bacteria were higher than those of the free-living methanogenic archaea and bacteria in the reactor. The results of clone analysis indicate that the major methanogenic archaea of the original thermophilic sludge are members of the order Methanomicrobiales, and that the major methanogenic archaea immobilized on the polyurethane foam are Methanosarcina spp., and those of the liquid phase are Methanobacterium spp. The results of the real time PCR analysis approximately agree with those of the clone analysis. These results indicate that real-time PCR analysis is useful for quantitatively describing methanogenic communities.  相似文献

Batch anaerobic codigestion of municipal household solid waste (MHSW) and digested manure in mesophilic conditions was carried out. The different waste-to-biomass ratios and intensity of mixing were studied theoretically and experimentally. The experiments showed that when organic loading was high, intensive mixing resulted in acidification and failure of the process, while low mixing intensity was crucial for successful digestion. However, when loading was low, mixing intensity had no significant effect on the process. We hypothesized that mixing was preventing establishment of methanogenic zones in the reactor space. The methanogenic zones are important to withstand inhibition due to development of acids formed during acidogenesis. The 2D distributed models of symmetrical cylinder reactor are presented based on the hypothesis of the necessity of a minimum size of methanogenic zones that can propagate and establish a good methanogenic environment. The model showed that at high organic loading rate spatial separation of the initial methanogenic centers from active acidogenic areas is the key factor for efficient conversion of solids to methane. The initial level of methanogenic biomass in the initiation centers is a critical factor for the survival of these centers. At low mixing, most of the initiation methanogenic centers survive and expand over the reactor volume. However, at vigorous mixing the initial methanogenic centers are reduced in size, averaged over the reactor volume, and finally dissipate. Using fluorescence in situ hybridization, large irregular cocci of microorganisms were observed in the case with minimal mixing, while in the case with high stirring mainly dead cells were found.  相似文献

The atpA and atpB genes coding for the alpha and beta subunits, respectively, of membrane ATPase were cloned from a methanogen Methanosarcina barkeri, and the amino acid sequences of the two subunits were deduced from the nucleotide sequences. The methanogenic alpha (578 amino acid residues) and beta (459 amino acid residues) subunits were highly homologous to the large and small subunits, respectively, of vacuolar H+-ATPases; 52% of the residues of the methanogenic alpha subunit were identical with those of the large subunit of vacuolar enzyme of carrot or Neurospora crassa, respectively, and 59, 60, and 59% of the residues of the methanogenic beta subunit were identical with those of the small subunits of N. crassa, Arabidopsis thaliana, and Sacharomyces cerevisiae, respectively. The methanogenic subunits were also highly homologous to the corresponding subunits of Sulfolobus acidocaldarius ATPase. The methanogenic alpha and beta subunits showed 22 and 24% identities with the beta and the alpha subunits of Escherichia coli F1, respectively. Furthermore, important amino acid residues identified genetically in the E. coli enzyme were conserved in the methanogenic enzyme. This sequence conservation suggests that vacuolar, F1, methanogenic, and S. acidocaldarius ATPases were derived from a common ancestral enzyme.  相似文献

【目的】通过对51个产甲烷古菌基因组中成簇的规律间隔短回文重复序列(Clustered regularly interspaced short palindromic repeats,CRISPR)的组成和来源进行研究,推测产甲烷古菌与环境中其他微生物的物质交换和相互作用,在基因组水平上阐述产甲烷古菌之间的遗传差异。【方法】利用CRISPRdb和CRISPRFinder,找出产甲烷古菌基因组中所有潜在的CRISPR簇。对CRISPR簇的基本组成部分进行分析:利用BLASTCLUST对重复序列(Repeat)进行分类;分别将间隔序列(Spacer)与Refseq病毒基因组、Refseq质粒基因组和Refseq产甲烷古菌基因组进行比对,从而获得间隔序列的物种来源和功能信息的注释。【结果】在51个产甲烷古菌中共找到了196个CRISPR簇,这些CRISPR簇中包含了总共4 355条间隔序列。在这些产甲烷古菌中,CRISPR簇的分布是不均匀的,且每个物种的间隔序列数量与其CRISPR簇数量是不成正比的。在对重复序列进行分类之后,发现Mclu1是分布最广且最具代表性的一类重复序列。在4 355条间隔序列中有388条具有物种注释信息,266条具有功能注释信息。从CRISPR簇间隔序列的来源来看,产甲烷古菌曾受到来自Poxiviridae、Siphoviridae以及Myoviridae属病毒的攻击,并且产甲烷古菌之间存在比较广泛的遗传物质交换。【结论】产甲烷古菌基因组中的CRISPR簇在组成和来源上存在较大的差异,这些差异与它们的生存环境有较大的关系。从CRISPR簇的角度阐述了产甲烷古菌之间基因组序列的差异。  相似文献

In this study we investigated whether a relationship exists between the methanogenic activity and the content of specific methanogenic cofactors of granular sludges cultured on different combinations of volatile fatty acids in upflow anaerobic sludge blanket or fluidized-bed reactors. Significant correlations were measured in both cases between the contents of coenzyme F420−2 or methanopterin and the maximum specific methanogenic activities on propionate, butyrate, and hydrogen, but not acetate. For both sludges the content of sarcinapterin appeared to be correlated with methanogenic activities on propionate, butyrate, and acetate, but not hydrogen. Similar correlations were measured with regard to the total content of coenzyme F420−4 and F420−5 in sludges from fluidized-bed reactors. The results indicate that the contents of specific methanogenic cofactors measured in anaerobic granular sludges can be used to estimate the hydrogenotrophic or acetotrophic methanogenic potential of these sludges.  相似文献

Mangrove forest sediments produce significant amounts of methane, but the diversity of methanogenic archaea is not well known at present. Therefore, 16S rRNA gene libraries were made using archaea-specific primers and DNA extracted directly from Tanzanian mangrove sediment samples as a template. Analysis of sequence data showed phylotypes closely related to cultivated methylotrophic methanogenic archaea from the marine environment, or distantly related to acetoclastic and hydrogenotrophic methanogenic archaea. In an attempt to isolate relevant methanogenic archaea, we succeeded in obtaining a new mesophilic methylotrophic methanogenic archaeon (strain MM1) capable of utilizing methanol and methylated amines as the only substrates. Under optimum conditions, the cells of strain MM1 exhibited a high specific growth rate (μ) of 0.21±0.03 (i.e. doubling time of 3.2 h) on both methanol and trimethylamine. The 16S rRNA gene sequence of strain MM1 clustered with five environmental clones, indicating that MM1 is an important methanogenic methylotroph in mangrove sediments. Based on physiological and phylogenetic analyses, strain MM1 is proposed to be included in the species of Methanococcoides methylutens .  相似文献

甲烷是重要的温室气体,低温湿地是大气甲烷的重要来源,因为湿地土壤中生活着大量的微生物包括甲烷古菌,它们将有机物降解转化为甲烷.本文总结了近年来低温湿地甲烷古菌群落组成、甲烷产生途径及其与环境的关系.研究显示,乙酸是低温湿地中主要的产甲烷物质,氢产甲烷过程主要发生在中温地区或酸性泥炭土中,而在盐碱水域中甲醇、甲胺是甲烷的重要底物.位于我国青藏高原的若尔盖湿地具有高海拔但低纬度的地理特征,我们的前期研究却显示甲醇在该湿地的甲烷排放中具有重要贡献.相应地,低温湿地中的甲烷古菌主要是利用甲基类化合物/乙酸的甲烷八叠球菌目和氢营养型的甲烷微球菌目.然而不同类型湿地甲烷排放途径及甲烷古菌的差异主要与环境的土壤类型、pH及植被类型相关,如刚毛荸荠生长的若尔盖湿地土壤中来源于甲醇的甲烷占全部甲烷的l7%;而木里苔草土壤中乙酸是产甲烷的主要前体物质.尽管已知冷适应的甲烷古菌在低温湿地的甲烷排放中发挥重要作用,但目前获得培养的嗜冷甲烷古菌却很少.冷响应的组学研究显示甲烷古菌的冷适应涉及到全局性生物学过程.  相似文献

青藏高原三个盐碱湖的产甲烷菌群和产甲烷代谢途径分析   总被引:1,自引:0,他引:1  
【目的】分析青藏高原不同类型盐碱湖中的优势产甲烷菌群和优势产甲烷代谢途径。【方法】以不同盐度和植被类型的公珠错、昆仲错和无植被的兹格塘错的沉积物为研究对象,通过高通量测序和q PCR定量古菌16S r RNA多样性分析优势古菌类群;模拟原位盐浓度及p H,比较不同产甲烷底物(甲醇、三甲胺、乙酸和H_2/CO_2)富集沉积物的产甲烷速率,分析其优势产甲烷菌代谢类型。通过添加产甲烷抑制剂(2-溴乙烷磺酸盐),检测沉积物中产甲烷底物积累,确定不同盐碱湖中主要的产甲烷途径。【结果】昆仲错的优势菌群包括甲基/乙酸型的甲烷八叠球菌科(Methanosarcinaceae,11%),乙酸型的甲烷鬃菌科(Methanosaetaceae,7.9%)和氢型甲烷菌甲烷杆菌目(Methanomicrobiales,7.4%);公珠错和兹格塘错的优势菌群为甲烷鬃菌科(Methanosaetaceae)分别占15%和15.3%,及甲烷杆菌属(Methanobacterium)和甲基型的甲烷叶菌属(Methanolobus)。公珠错和昆仲错分别以乙酸和甲醇产甲烷速率最高,而兹格塘错从不同底物产甲烷速率无差异。抑制甲烷产生后,公珠错主要积累乙酸,昆仲错主要积累甲醇;兹格塘错不仅甲烷排放低,也无产甲烷物质显著积累。【结论】昆仲错沉积物中的甲烷主要来自甲醇,公珠错中的甲烷主要来自乙酸,而兹格塘错产甲烷和底物积累不活跃。因而推测高原盐碱湖主要的产甲烷途径和菌群可能与周围植被类型的相关性更高,而与盐度的直接相关性较低。  相似文献

We examined the survival potential of methanogenic archaea exposed to different environmental stress conditions such as low temperature (down to −78.5°C), high salinity (up to 6 M NaCl), starvation (up to 3 months), long-term freezing (up to 2 years), desiccation (up to 25 days) and oxygen exposure (up to 72 h). The experiments were conducted with methanogenic archaea from Siberian permafrost and were complemented by experiments on well-studied methanogens from nonpermafrost habitats. Our results indicate a high survival potential of a methanogenic archaeon from Siberian permafrost when exposed to the extreme conditions tested. In contrast, these stress conditions were lethal for methanogenic archaea isolated from nonpermafrost habitats. A better adaptation to stress was observed at a low temperature (4°C) compared with a higher one (28°C). Given the unique metabolism of methanogenic archaea in general and the long-term survival and high tolerance to extreme conditions of the methanogens investigated in this study, methanogenic archaea from permafrost should be considered as primary candidates for possible subsurface Martian life.  相似文献

Anaerobic heterotrichous ciliates (Armophoridae and Clevelandellidae) possess hydrogenosomes that generate molecular hydrogen and ATP. This intracellular source of hydrogen provides the basis for a stable endosymbiotic association with methanogenic archaea. We analyzed the SSU rRNA genes of 18 heterotrichous anaerobic ciliates and their methanogenic endosymbionts in order to unravel the evolution of this mutualistic association. Here, we show that the anaerobic heterotrichous ciliates constitute at least three evolutionary lines. One group consists predominantly of gut-dwelling ciliates, and two to three, potentially four, additional clades comprise ciliates that thrive in freshwater sediments. Their methanogenic endosymbionts belong to only two different taxa that are closely related to free-living methanogenic archaea from the particular ecological niches. The close phylogenetic relationships between the endosymbionts and free-living methanogenic archaea argue for multiple acquisitions from environmental sources, notwithstanding the strictly vertical transmission of the endosymbionts. Since phylogenetic analysis of the small-subunit (SSU) rRNA genes of the hydrogenosomes of these ciliates indicates a descent from the mitochondria of aerobic ciliates, it is likely that anaerobic heterotrichous ciliates hosted endosymbiotic methanogens prior to their radiation. Therefore, our data strongly suggest multiple acquisitions and replacements of endosymbiotic methanogenic archaea during their host's adaptation to the various ecological niches.  相似文献

Polyclonal antisera have been used for the development of a novel serotaxonomic system for methanogenic bacteria by quantifying immunological cross-reactions of the antisera with heterologous strains. This system corresponds well to phylogenetic relationships among methanogenic bacteria as analysed by other authors, and is in good agreement with the existing immunological fingerprint for immunotyping of methanogenic bacteria.Offprint requests to: W. The potential for degradation of four nitrogen-heterocyclic compounds was investigated in fresh-water sediment slurries maintained under denitrifying, sulfate-reducing, and methanogenic conditions. Pyridine (10 mg/l) was rapidly transformed within 4 weeks under denitrifying conditions but persisted for up to 3 months under sulfate-reducing and methanogenic conditions. No intermediate biotransformation products of pyridine metabolism were detected under denitrifying conditions. Quinoline (10 mg/l) was completely transformed without a lag phase under methanogenic and sulfate-reducing conditions after incubation for 23 and 45 days, respectively. 2-Hydroxyquinoline was produced concomitantly with quinoline transformation under methanogenic and sulfate-reducing conditions. Under denitrifying conditions, less than 23% of the initial concentration of quinoline was transformed after anaerobic incubation for 83 days. Indole, however, was completely removed from sediment slurries under denitrifying, sulfate-reducing, and methanogenic conditions after anaerobic incubation for 18, 27, and 17 days, respectively. Only low amounts of oxindole (2–4 mg/l) accumulated during indole metabolism under methanogenic and denitrifying conditions, but under sulfate-reducing conditions, oxindole accumulation was stoichiometric with indole transformation. No evidence for biotransformation of carbazole was noted for all anaerobic conditions tested.  相似文献

Aims:  To screen a pair of primers suitable for denaturing gradient gel electrophoretic (DGGE) analysis of ruminal methanogenic Archaea and to detect the archaeal communities in the rumen of goat.
Methods and Results:  Nine primer pairs for 16S rDNA of methanogenic Archaea , including six for directed polymerase chain reaction (PCR) and three for nested PCR were first evaluated by PCR amplification of the total DNA from rumen fluids and bacteria. The DGGE analysis of rumen fluids was then conducted with three primer sets (344fGC/915r, 1106fGC/1378r and 519f/915rGC) of the nine pairs tested. Good separation and quality of patterns were obtained in DGGE analysis with primer pairs 1106fGC/1378r and 519f/915rGC. A total of 40 DNA fragments were excised from the DGGE gels and their sequences were determined. All fragments belonged to methanogenic Archaea while primer pair 519f/915rGC had better amplification ranges than the other two primer pairs.
Conclusions:  The procedure of DGGE analysis with primer pair 519f/915rGC was more suitable for investigating methanogenic archaeal community in the rumen. The dominant methanogenic Archaea in the rumen of goat was Methanobrevibacter sp. and an unidentified methanogenic Archaea .
Significance and Impact of the Study:  One pair of primers suitable for DGGE analysis of ruminal methanogenic Archaea was obtained and the molecular diversity of ruminal methanogenic Archaea in goat was investigated by PCR-DGGE.  相似文献

A regime shift between a macrophyte-dominated clear state and a phytoplankton-dominated turbid state can have considerable impact on ecosystem structure and function of shallow lakes. However, very little is known about the response of the methanogenic archaeal community in the sediment during this regime shift. We investigated the methanogenic archaeal community at two sites in the large, shallow, eutrophic Taihu Lake over the course of one year. One site is located in Meiliang Bay and is dominated by Microcystis blooms, and the other site is located in East Taihu Bay and is dominated by aquatic macrophytes. Terminal restriction fragment length polymorphism (T-RFLP) and phylogenetic analyses of archaeal 16S rRNA genes were used to analyze the methanogenic community. Higher ratio of methanogens in Archaea was observed in East Taihu Bay than in Meiliang Bay. The methanogenic archaeal community was dominated by the Methanobacteriales and the LDS cluster in macrophytes-dominated East Taihu Bay, while it was dominated by the Methanosarcinaceae, Methanobacteriales, and the LDS cluster in Microcystis-dominated Meiliang Bay. Clustering analysis of all of the samples revealed differences in the composition of the methanogenic archaeal communities between the two sites that were independent of seasonal variations. Further statistical analysis indicated that the chlorophyll a (Chla) concentration had a profound impact on the composition of the methanogenic archaeal community in Meiliang Bay, whereas it was primarily influenced by total organic carbon (TOC) levels in East Taihu Bay. Overall, this investigation demonstrates that intra-habitat differences in the composition of methanogenic archaeal communities are likely driven by changes in the available organic materials.  相似文献

Summary The relationship between the coenzyme F420 content and the activity of methanogenic microorganisms was investigated under different cultivation conditions in anaerobic reactors. The coenzyme F420 concentration depends on the substrate used and the cultivation conditions. Coenzyme F420 appears not to be a measure of the total methanogenic activity but rather a measure of the amount of methanogenic microorganisms in mixed anaerobic cultures.  相似文献
