Summary Strains of Penicillium cyclopium and Rhizopus arrhizus secrete two extracellular lipases and contain an intracellular lipase. As these intracellular enzymes exhibited good synthetic activities in organic solvent, we designed a loop fixed-bed reactor for the continuous synthesis of esters. In a preliminary study we optimized the yield of ester synthesis using response surface methodology. With dodecanoic dodecyl ester as a model compound, the yields of ester synthesis were higher than 90%. It has been demonstrated that the reactor designed for this study is more efficient than a stirred batch reactor and more efficient than a fixed-bed reactor without a loop current or without a second catalytic column. In application, we have shown that lipases from R. arrhizus and P. cyclopium do not esterify tertiary alcohols like many lipases. Correspondence to: L. C. Comeau  相似文献

Of 17 organic phases, some containing one of the five extractants for solvent extraction of lactic acid from Rhizopus arrhizus broth, solvents containing tertiary amines (Hostarex A327, trioctylamine) and secondary amine with isotridecanol modifier and trihexylphosphate, are from point of view of toxicity suitable for the in situ or on-line extraction. Pentaphosphinedipentylester is highly toxic. Toxicity of alcohols used as modifiers, decreases with an increase in the number of carbons in a molecule. Isotridecanol is medium toxic. The nonylphenol (NP) biosorption and desorption potential for fungal biomass used under batch conditions was investigated using kinetics and isotherm models. Fungal biomass of Rhizopus arrhizus TISTR 3610 exhibited preferential uptake of NP, an endocrine disrupting chemicals. Sporangiospores, asexual spores, were immobilised in chitosan beads. The biosorption data of NP on the moist heat inactivated R. arrhizus–chitosan beads were analyzed using four popular adsorption isotherms and, by using non-linear least-regression with the solver add-in in Microsoft Excel, correlated in order with the Fritz–Schluender > Redlich–Peterson > Freundlich > Langmuir isotherms. The pseudo first-order kinetics was found to have the best fit with the experimental data. The diffusivity of NP in the R. arrhizus–chitosan beads was calculated using the shrinking core model, and the diffusivity values were in the ranges of 2.3736 × 10−4–1.8950 × 10−4 cm2 s−1. Desorption to recover the adsorbed NP from the beads was performed in methanol and was best described using a pseudo second-order kinetic model.  相似文献

Summary The peparative synthesis of 35 short chain flavour esters by lipases fromMucor miehi, Aspergillus sp.,Candida rugosa andRhizopus arrhizus was investigated in organic media. Acetic, propionic, butyric, valeric and caproic acids, as well as methanol, ethanol, butanol, i-pentanol, hexanol, citronellol and geraniol were used as substrates. Most of the esters were synthesized in good yield by at least one of the lipase preparations tested. Different conversion yields were observed according to the lipase specificity toward the acid or the alcohol moiety of the ester. Methyl- and ethyl acetates were also produced by changing the organic solvent. Enzymatic catalysis in organic solvent is thought to be a valuable method for preparative synthesis of flavour esters.  相似文献

Selected samples of waste microbial biomass originating from various industrial fermentation processes and biological treatment plants have been screened for biosorbent properties in conjunction with uranium and thorium in aqueous solutions. Biosorption isotherms have been used for the evaluation of biosorptive uptake capacity of the biomass which was also compared to an activated carbon and the ion exchange resin currently used in uranium production processes. Determined uranium and thorium biosorption isotherms were independent of the initial U or Th solution concentration. Solution pH affected the exhibited uptake. In general, lower biosorptive uptake was exhibited at pH 2 than at pH 4. No discernible difference in uptake was observed between pH 4 and pH 5 where the optimum pH for biosorption lies. The biomass of Rhizopus arrhizus at pH 4 exhibited the highest uranium and thorium biosorptive uptake capacity (g) in excess of 180 mg/g. At an equilibrium uranium concentration of 30 mg/liter, R. arrhizus removed approximately 2.5 and 3.3 times more uranium than the ion exchange resin and activated carbon, respectively. Under the same conditions, R. arrhizus removed 20 times more thorium than the ion exchange resin and 2.3 times more than the activated carbon. R. arrhizus also exhibited higher uptake and a generally more favorable isotherm for both uranium and thorium than all other biomass types examined.  相似文献

Continuous hydrolysis of triglyceride in organic solvent systems using Rhizopus arrhizus mycelia as a source of insolubilized lipase has been studied in packed-bed and stirred-tank reactors. Typically a packed bed reactor containing 1 g of mycelia fed at 1 mL/min with a solution of 2.5% (w/v) olive oil in di-isopropyl ether gave a fatty acid yield of 45% at 30°C. The optimum water concentration was found to be 0.17% (w/v) except under conditions of high oil feed concentration and high yield where no optimum was established. No temperature optimum was observed over the range 20–55°C. Calculated activation energies of 13–20 kJ/mol, depending on temperature, were lower, while Km(app) values of 0.1–0.3M were higher than those for hydrolysis in conventional aqueous emulsion systems. No evidence of any significant diffusional limitation, which could account for these values, was obtained. The mycelia showed a loss of activity of 0.6–1.0%h at 30°C. The packed bed proved markedly superior to the stirred tank for this system.  相似文献

We report a case of eczema-like cutaneous mucormycosis caused by Rhizopus arrhizus. A 4-year-old child was presented to our hospital with a history of gradually enlarging papule and plaque in the periumbilical area for nearly 4 years since 2 weeks after his birth, and it has been misdiagnosed as eczema for nearly 3 years. Based on histopathology examination, the fungus culture test and DNA sequencing, it was revealed that R. arrhizus should be the responsible fungus for skin infection. The patient was successfully cured by combination of intravenous drip and percutaneous injection amphotericin B for nearly 3 months, and no recrudescence was seen during a follow-up of 6-month observation.

Summary Oxygen consumption of Rhizopus arrhizus cultures was studied in order to understand why anaerobic-type metabolism takes place during growth, with high lactic acid synthesis. Rather than insufficient oxygen supply, pellet-type morphology was found to be responsible. Cultures with carriers and on thin liquid films were investigated with very good results for the second method. Not only was biomass production improved, but also carbon consumption was higher and lactic acid synthesis less compared with traditional submerged flask cultures. Offprint requests to: J. C. The co-culture of Bacillus subtilis IFO 3335 with Rhizopus peka P8 or Rhizopus oligosporus P12 in liquid medium was found to increase production of antibiotic activity and to alter the spectrum of activity relative to the pure cultures. However, a mixed culture of Rhizopus arrhizus P7 and Rhizopus oryzae P17 did not produce antibiotic activity. The concentration, ratio, and time of addition of B. subtilis to the R. peka culture was found to influence antibiotic yields. Solid-state fermentations using mixed cultures of R. peka and B. subtilis were investigated. The growth of Escherichia coli IFO 3792 as a target bacterium was inhibited by the mixed culture. These results suggest the possibility of biopreservation of fermented foods by novel co-culture systems.  相似文献

To discover the differences and modes of saccharifying ability among Rhizopus arrhizus strains isolated from various habitats, we grew strains in glutinous rice media to monitor the production of glucose from starch using spectrophotometry at regular time intervals. The saccharifying capacity of R. arrhizus var. delemar was found to be weaker than that of var. arrhizus and var. tonkinensis. These data provide additional phenotypic support for recognization of this genetically distinct lineage (var. delemar) as a separate species. Clinical isolates were generally low in saccharifying abilities, while all strains from southwest China had an excellent diastatic power.  相似文献

The asymmetric resolution of various 1-aryl-3-buten-1-ols via microbial hydrolysis of the corresponding acetates has been investigated using different Rhizopus species. The chosen species, R. arrhizus (wild type), efficiently hydrolyzed 1-phenyl- and 1-para-substituted phenyl-3-buten-1-ol acetates, producing the enantiomerically pure (R)-alcohols with 53–65% yields. Although the antipode acetates were obtained with 9–52% enantiomeric excess, the (S)-alcohols were amenable in > 99% enantiomeric excess via a R. arrhizus mediated asymmetric reduction of the corresponding ketones.  相似文献

Zn biosorption by Rhizopus arrhizus and other fungi   总被引:1,自引:0,他引:1  
Biosorption of zinc ions by inactivated fungal mycelia was studied. Of the six fungal species, Rhizopus arrhizus, Mucor racemosus, Mycotypha africana, Aspergillus nidulans, Aspergillus niger and Schizosaccharomyces pombe, R. arrhizus exhibited the highest capacity (Q max = 213 μmol g−1 dry weight). Further experiments with different cellular fractions of R. arrhizus showed that Zn was predominantly bound to cell-wall chitin and chitosan (Q max = 312 μmol g−1 dry weight). Adsorption data were best modelled by the Langmuir isotherm, although they can be modelled by the Freundlich equation as well at relatively low aqueous concentrations. Biosorption generally decreased with increase in biosorbent particle size and its concentration. Low pH reduced Zn sorption, because of the strong competition from hydrogen ions for binding sites on fungi. The presence of ligands reduced metal uptake, chiefly by forming metal complexes of a less biosorbable nature. An efficient and convenient strategy for synthesis of enantiomerically pure S-2-(1-hydroxy-3-butenyl)-5-methylfuran was for the first time described utilizing a lipase-mediated asymmetric acylation in organic solvents. Rhizopus arrhizus lipase was chosen as the biocatalyst, and different immobilization methods including sol–gel encapsulation and covalent attachment were adopted to improve its catalytic characteristics. Various influential factors of the reaction were also investigated. Finally, the results showed that the lipase covalently attached onto epoxy resin exhibited the highest enantioselectivity and operational stability. Under optimized reaction conditions, i.e., n-hexane as the solvent, 5/1 (mol/mol) of vinyl acetate to 2-(1-hydroxy-3-butenyl)-5-methylfuran and 30 °C, the ee value of S-1 reached up to above 98% at 52% conversion with an E value of 99.  相似文献

The biochemical kinetic of direct fermentation for lactic acid production by fungal species of Rhizopus arrhizus 3,6017 and Rhizopus oryzae 2,062 was studied with respect to growth pH, temperature and substrate. The direct fermentation was characterized by starch hydrolysis, accumulation of reducing sugar, and production of lactic acid and fungal biomass. Starch hydrolysis, reducing sugar accumulation, biomass formation and lactic acid production were affected with the variations in pH, temperature, and starch source and concentration. A growth condition with starch concentration approximately 20 g/l at pH 6.0 and 30°C was favourable for both starch saccharification and lactic acid fermentation, resulting in lactic acid yield of 0.87–0.97 g/g starch associated with 1.5–2.0 g/l fungal biomass produced in 36 h fermentation. R. arrhizus 3,6017 had a higher capacity to produce lactic acid, while R. oryzae 2,062 produced more fungal biomass under similar conditions.  相似文献

The binding characteristics and the inhibitory power of atrazine and DCMU towards uncoupled electron flow activity were studied in acyl lipid-depleted thylakoid membranes from atrazine-susceptible and-resistant biotypes of Solanum nigrum L. For this purpose, phospholipase A2 from Vipera russelli and the lipase from Rhizopus arrhizus were used to obtain a selective lipid class (phospholipids or galactolipids) depletion which was restricted to the outer monolayer. Neither phospholipid nor galactolipid removal affected the dissociation constant and the number of binding sites of atrazine. In contrast, the dissociation constant of DCMU was increased in phospholipid-depleted thylakoid membranes but remained unchanged after galactolipid depletion. The number of DCMU binding sites decreased significantly after both lipase treatments, but only in the resistant biotype. The inhibitory effectiveness of the herbicide was either decreased or increased (to different extents) depending on the lipid class which was removed from the membrane and on the biotype considered. The specific activities of enzymes such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPOX) and glutathione reductase (GR), which are involved in protection against toxic species of oxygen, were determined in mycelia extracts of pentachloronitrobenzene (PCNB)-tolerant and susceptible soil fungi. The organisms assayed were the highly PCNB-sensitive Rhizoctonia solani and Rhizopus arrhizus; Sclerotium rolfsii and Trichoderma harzianum, which are moderately susceptible to PCNB, and the fungicide-tolerant Fusarium oxysporum f. sp. melonis and Pythium aphanidermatum. No GPOX activity was detected in the six examined fungi. Significant differences in the specific activities of the other enzyme systems among the fungi were evident. Remarkably low levels of CAT activities were measured in R. solani. Except for T. harzianum, no meaningful differences regarding SOD, CAT and GR activities with age of the fungi cultures were observed. The electrophoretic patterns of SOD and CAT displayed dissimilarities among the fungi under study. P. aphanidermatum is more polymorphic with respect to both SOD and CAT enzyme systems as compared to the other fungi. The SOD of F. oxysporum f. sp. melonis, R. arrhizus and T. harzianum is a cuprozinc enzyme, while the mangano-SOD species was detected in S. rolfsii, R. solani and T. harzianum.  相似文献

Biomasses from eight different Rhizopus species were tested for the sorption of americium from nitric acid medium. Rhizopus arrhizus NCIM 997 showed maximum sorption at pH 2. Laboratory scale experiments were carried out using this biomass in packed columns for the sorption of -activity from an americium spiked low level waste stream of PUREX process. The biomass was found to be an excellent sorbent for remediation of low level waste streams on once through basis.  相似文献

This study proposed a novel waste utilization bioprocess for production of lactic acid and fungal biomass from waste streams by fungal species of Rhizopus arrhizus 36017 and R. oryzae 2062. The lactic acid and fungal biomass were produced in a single-stage simultaneous saccharification and fermentation process using potato, corn, wheat and pineapple waste streams as production media. R. arrhizus 36017 gave a high lactic acid yield up to 0.94–0.97 g/g of starch or sugars associated with 4–5 g/l of fungal biomass produced, while 17–19 g/l fungal biomass with a lactic acid yield of 0.65–0.76 g/g was produced by the R. oryzae 2062 in 36–48 h fermentation. Supplementation of 2 g/l of ammonium sulfate, yeast extract and peptone stimulated an increase in 8–15% lactic acid yield and 10–20% fungal biomass.
