Studies were conducted to evaluate the toxicity of insecticides to adult Diadegma insulare (Cresson) and its host the diamondback moth, Plutella xylostella (L.). Leaf-dip and direct-dip bioassays for diamondback moth larvae and residual bioassays for adults of diamondback moth and D. insulare were used to assess mortalities. Larval mortalities at field rates were significantly higher with carbaryl, permethrin, spinosad, and tebufenozide when compared with Bacillus thuringiensis, or imidacloprid in the larval-dip bioassay 72 h after treatment. In the leaf-dip and residual bioassays, both permethrin and spinosad caused 100% mortalities to diamondback moth larvae and adults, respectively, 72 h after treatment. Of all the materials tested, only B. thuringiensis and tebufenozide were not toxic to D. insulare 24 h after treatment. Spinosad was not toxic to D. insulare 30 min after treatment. However, 100% mortality was observed 8 h after treatment.  相似文献

Parasitism of Plutella xylostella (L.) third and fourth instars was evaluated in a cabbage field in Geneva, NY, in 1999. Over the entire season, average parasitism was 33.6% for third instars and 53.6% for fourth instars, and the main parasitoids were Diadegma insulare (Cresson) and Microplitis plutellae Muesbeck. In the early season, total parasitism was low, and mainly caused by D. insulare. However, later in the season, parasitism reached >80% for the fouth instars and 50% for the third instars. Our survey indicated that M. plutellae heavily parasitized P. xylostella, and provided higher parasitism rates than D. insulare in the late season. Comparison of these two species in laboratory bioassays indicated there were no significant differences in susceptibility to four insecticides commonly used in crucifer fields. For both parasitoids, an experience with P. xylostella on a damaged leaf increased their host-searching efficacy. Compared with M. plutellae, D. insulare was a better host-searcher both for the naive and the experienced adults. Although both parasitoids can cause high mortality rates of P. xylostella, D. insulare may be more suitable to be released in fields to enhance natural control against P. xylostella.  相似文献

Phenotypic variation in behavioral response and physiological tolerance to permethrin was measured and compared between two populations of diamondback moth, Plutella xylostella (L.). A field population from Celeryville, OH, and a laboratory population originally collected from Wooster, OH, were compared. In laboratory choice bioassays, females from both populations were less likely to oviposit on cabbage leaf disks and seedlings treated with permethrin. The oviposition-deterrent effect was positively related to permethrin concentration. The laboratory population was significantly more behaviorally responsive to the insecticide and showed a significantly greater avoidance than the field population of the highest concentration of permethrin tested (1.50 g [AI] /liter). The physiological response of each population was measured by feeding bioassays, and the laboratory population was physiologically more susceptible to the permethrin. Larvae from the laboratory population that were fed permethrin-treated leaves had significantly lower growth rate, higher mortality, and lower adult fecundity compared with larvae from the field population. These data suggest that female moths that are more behaviorally responsive to permethrin produce offspring that tend to be more susceptible to the same insecticide, at least for the populations tested. The relationship between adult behavioral response and larval physiological tolerance to permethrin is discussed.  相似文献

In the laboratory and in cages in the greenhouse, we evaluated the toxicity of two insecticides (lambda-cyhalothrin and spinosad) on the parasitoid, Diadegma insulare (Cresson), and the predator, Coleomegilla maculate (DeGeer), both natural enemies of the diamondback moth, Plutella xylostella (L.). Lambda-cyhalothrin was very toxic to both natural enemies. Spinosad was less toxic to C. maculata adults and larvae, and slightly toxic to D. insulare. Both natural enemies suppressed P. xylostella populations in cages with 80% spinosad-treated and 20% nontreated plants; such suppression was not seen when lambda-cyhalothrin was used. Using broccoli, Brassica oleracea L. variety italica, a common host for P. xylostella, we also studied direct and indirect effects of both natural enemies in the presence and absence of the two insecticides and to different P. xylostella genotypes: resistant to the insecticide, susceptible, or heterozygous. Neither natural enemy could distinguish host genotype if P. xylostella were feeding on nontreated plants. They could also not distinguish between larvae feeding on spinosad-treated plants and nontreated plants, but D. insulare could distinguish between larvae feeding on lambda-cyhalothrin treated and nontreated plants. Our studies suggest that lambda-cyhalothrin has direct toxicity to these two natural enemies, can affect their host foraging and acceptance of P. xylostella and consequently would not be compatible in conserving these natural enemies in a program for suppression of P. xylostella. In contrast, our studies suggest that treatment with spinosad has much less effect on these natural enemies and would allow them to help suppress populations of P. xylostella. These findings are discussed in relation to the evolution of insecticide resistance and suppression of the pest populations.  相似文献

We investigated the genetic basis of adult behavioral response and larval physiological tolerance to permethrin within two diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), populations from Wooster and Celeryville, OH, with different average levels of larval tolerance. The adult behavioral response was measured as oviposition site preference and was investigated using full-sib design and parent-offspring regression. Additive genetic variance (0.134 +/- 0.02) and the heritability (h2 = 0.31 +/- 0.08) for the behavioral response was significant for the Celeryville population, suggesting that in this population, a high proportion of phenotypic variation for adult behavioral response to permethrin was heritable genetic variation. The larval physiological response was measured with a topical application bioassay and was investigated using a half-sib design. Significant additive genetic variances and heritabilities for physiological tolerance to permethrin were detected in both populations. The genetic correlation between adult behavioral response and larval physiological tolerance to permethrin were negative, but significant only in the Celeryville population; indicating that adults from this population that are more behaviorally responsive produced offspring that are more susceptible to permethrin. Our findings have implications for the evolution and management of insecticide resistance in the diamondback moth. The adult behavioral response can lower the exposure of larvae to the insecticide, lowering selection pressure for physiological resistance in larvae. Furthermore, to the extent that the adult behavioral response increases fitness, it can indirectly select for larval susceptibility because of the negative correlation between the two traits.  相似文献

During an outbreak of the diamondback moth, Plutella xylostella (L.), in California in 1997, nine populations were collected from the major broccoli areas throughout the state. Populations were assayed for their susceptibility to currently used materials (Bacillus thuringiensis subsp. kurstaki, permethrin, and methomyl) and to newer materials that had not yet been commercially used in California (spinosad, emamectin benzoate, and chlorfenapyr). For the currently used insecticides, elevated levels of resistance were seen only with permethrin and seven of the nine populations had tolerance ratios (TR) of > 100. With the newer chemistries, TR values were all < 15. To compare potential cross-tolerance, TR values of the currently used insecticides were compared with TR values of the newer insecticides. There were significant relationships found between: methomyl and emamectin benzoate, methomyl and spinosad, and permethrin and spinosad. Further biochemical studies are needed to confirm the actual mechanisms that lead to these relationships and field tests are needed to determine what impact, if any, such TR levels would have on control in the field. These data indicate that resistance to at least one of the commonly used insecticides (permethrin) may have played a role in the outbreak during 1997. However, other factors may have been at least equally important. The winter of 1996-1997 was warmer than normal, and during the period from February through August of 1997 the amount of rainfall was < 50% of normal. Hot and dry conditions are known to be conducive to outbreaks of P. xylostella. These data add to an overall knowledge about the geographic variation of resistance in P. xylostella populations within the United States. They also serve as a baseline for monitoring changes in susceptibility to these newer insecticides and can also help explain the occurrence of outbreaks caused by factors other than insecticide resistance.  相似文献

1. The use of floral resource subsidies to improve herbivore suppression by parasitoids requires certain trophic interactions and physiological changes to occur. While the longevity and fecundity of parasitoids are positively affected by nectar subsidies in laboratory studies, the impacts of floral subsidies on the fecundity and longevity of freely foraging parasitoids have not been studied. 2. We studied the longevity and per capita fecundity of naturally occurring Diadegma insulare foraging in cabbage plots with and without borders of flowering buckwheat, Fagopyrum esculentum, as well as relationships between longevity, fecundity, sugar feeding and parasitism rates on larvae of the diamondback moth, Plutella xylostella. 3. Relative longevity was estimated by counting broken setae on the fringe of the forewing. Floral borders increased the longevity of males and females in adjacent cabbage plots. 4. The egg maturation rate of D. insulare was estimated by comparing egg loads of females collected early in the day with egg loads of females held without hosts in field cages throughout the day. Females in buckwheat cages matured 2.7 eggs per hour while females in control cages resorbed 0.27 eggs over the same time period. 5. The fecundity of females collected in the afternoon was estimated by comparing their actual egg load to the estimated egg load in the absence of oviposition for females in a given plot. Females foraging in buckwheat plots had marginally fewer eggs remaining in their ovaries, and laid marginally more eggs than females in control plots. Females from both treatments carried 30-60 eggs by the afternoon and therefore were time-limited rather than egg-limited. 6. Plots where a greater proportion of females had fed on sugar had longer-lived females. This suggests that feeding enhanced longevity of D. insulare. However, plots with longer-lived and more fecund females did not exhibit higher parasitism rates, although the power of these tests were low.  相似文献

The use of different commercial Bacillus thuringiensis (Bt) products in the Bajio guanajuatense area in Mexico began 12 yr ago, and resistance to Bt in this area has been reported for Plutella xylostella (L.) The current study provides a baseline response and resistance potential to Bt in field and laboratory strains of Bajio Trichoplusia ni (Hübner). Differences in susceptibility to Bt among T. ni populations were observed. T. ni neonates collected in Romita, Guanajuato, were more susceptible to Bt than those collected in Salvatierra or San Luis de la Paz, Guanajuato. After five generations of exposure to XenTari in the laboratory, decreased susceptibility was found only in the Salvatierra insects, with an LC50 that was 2.1-fold greater than that of a Mexican laboratory strain. The XenTari-selected San Luis de la Paz strain was from 16- to 87-fold more resistant to CrylA protoxins than U.S. (US) and Mexican laboratory strains. Although CrylAb is not a component of XenTari, this strain also was significantly less susceptible to CrylAb toxin compared with a US strain, with a resistance ratio of 40.4. The larval weights and lengths, pupal lengths, and percentage of pupation were significantly lower for the Salvatierra strain than for all other strains. The relationship of T. ni susceptibilities to Bt Cry toxins and protoxins after several generations of exposure to XenTari and its similarity to P. xylostella behavior.  相似文献

Abstract.  1. To investigate how floral resources impact feeding and nutrient dynamics of parasitoids, the ichneumonid Diadegma insulare (Cresson) was monitored in cabbage fields with and without buckwheat Fagopyrum esculentum Moench borders. Biochemical analyses of field-collected wasps determined whether sugar feeding occurred, and whether it enhanced nutrient levels and increased parasitism of diamondback larvae Plutella xylostella (L.) from 2000 to 2003. The effects of experimental spatial scale on sugar feeding dynamics were also evaluated in 2001.
2. The majority of D. insulare foraging on cabbage had fed upon sugar, but floral borders did not consistently increase the proportion of sugar-fed wasps or their levels of glycogen and lipid over all 4 years. Floral borders increased sugar storage by females in 2001. More females fed on sugar when experimental plots were separated by at least 800 m compared with 67 m.
3. Feeding on buckwheat nectar vs. honeydew produced by the soybean aphid Aphis glycines Matsumura in surrounding fields was distinguished using the ratio of fructose to total sugars, which was higher in nectar- than in honeydew-fed parasitoids. Floral borders increased the probability that females collected from adjacent crops were nectar-fed in 2002, demonstrating that floral resources were utilised by foraging parasitoids. Diadegma insulare that fed upon any sugar source had higher levels of sugar and glycogen than unfed wasps, and feeding on honeydew appeared to enhance nutrient storage.
4. The prevalence of sugar or nectar feeding by female D. insulare was not correlated with parasitism rates over the 4 years from all plots, but nectar feeding was positively correlated with parasitism rates within floral plots.  相似文献

The susceptibility of larvae of the diamondback moth, Plutella xylostella Linnaeus to purified crystal proteins and spore-crystal preparations of Bacillus thuringiensis was investigated for 13 populations from seven states in India. The LC50 (microg ml(-1), 48 h) values of Cry proteins for different populations of P. xylostella ranged from 0.14-3.74 (Cry1Aa), 0.007-1.25 (Cry1Ab), 0.18-2.47 (Cry1Ac) and 0.12-3.0 (Cry1C). The LC50 (mg (ai) l(-1), 48 h) of spore-crystal preparations ranged from 0.02-0.98 (HD-1) and 0.06-2.14 (HD-73). Significantly higher LC50 values for all tested toxins and strains were obtained with populations collected from Iruttupallam and Ottanchathiram in the southern state of Tamil Nadu, whereas some of the populations collected from the northern part of India were more susceptible than the susceptible IARI 17-65 population. The high levels of resistance in the Iruttupallam and Ottanchathiram populations to Cry1Ab suggested selection pressure by Cry1Ab, which is the predominant toxin in B. thuringiensis formulations used in India. Cry1Ab was found to be more toxic than the other toxins. The population from Iruttupallam showed increased resistance following selection with Cry1Ab in the laboratory (LC50 from 1.25 to 4.31 microg ml(-1) over two generations) and also showed cross resistance to CrylAa and CrylAc. The resistance to Biobit in the field population from Iruttupallam declined slowly; requiring c. 33 generations for an overall 10-fold decline in LC50 when the insects were reared in the laboratory without exposure to B. thuringensis.  相似文献

Red mite field populations from seven naturally infested Italian caged laying poultry farms were investigated for their susceptibility to acaricide formulations available on the market, containing amitraz, carbaryl and permethrin. A minimum of 3,000 mites of all stages were collected from each farm and were tested with five acaricide concentrations (5, 10, 20, 50, 100%) plus an untreated control (0%). Field red mite populations were found to be tolerant even with the highest concentrations with carbaryl and permethrin for six (86%) and three (42%) of the investigated farms, respectively (< 0.05). Furthermore, six (86%) of the investigated farms showed a red mite population susceptible to amitraz at any concentration. Out of the seven field populations tested with amitraz, one population is becoming less tolerant whereas another was the most tolerant to carbaryl and permethrin at any concentration. Data show that the lack of effectiveness of some acaricides is spreading in Europe and call for the adoption of alternative management strategies to avoid development of resistance.  相似文献

Head lice, Pediculus capitis , were collected from children aged 3–12 years in Maale Adumin, a town near Jerusalem, after reports of control failure with the pyrethroid insecticide permethrin. A total of 1516 children were examined: living lice and eggs were found on 12.1% of the children; or another 22.8% of the children only nits were found. Twice as many girls as boys (8.1% v 4%) were infested with lice and or nits. Head lice collected from infested children were exposed to permethrin impregnated filter-papers. Log time probit mortality (ltp) regression lines were calculated for mortality data and compared to ltp lines for a similar collection of head lice made in 1989. The regression lines for the two years were significantly different, with a 4-fold decrease in susceptibility at the LT50 level between 1989 and 1994. The slopes of the lines also suggested that the 1994 population was more heterogenous in its response to permethrin than the 1989 population. In contrast, a laboratory population of body lice (Pediculus humanus) tested with the same batch of permethrin-impregnated papers showed a slight but non-significant increase in susceptibility between 1989 and 1994. The results suggest that resistance to pyrethroids has developed rapidly among head lice since permethrin was introduced in 1991 as a pediculicide in Israel.  相似文献

The permethrin tolerance (PT) of a population of the mosquito Anopheles gambiae (Diptera: Culicidae) increased following the introduction of permethrin-impregnated nets for malaria control in certain villages near Kisumu, western Kenya. Using a biochemical test that indirectly measures oxidases associated with permethrin resistance, we found that this population had higher oxidase levels than a comparison population from villages without impregnated nets. Mosquitoes from a colony of An. gambiae selected for PT, the RSP (reduced susceptibility to permethrin) strain, were exposed to permethrin with or without the oxidase inhibitor piperonyl butoxide (PB). Significantly higher mortality rates occurred when permethrin was synergized by PB, presumably by suppression of oxidases responsible for PT. An unselected (UNS) colony of An. gambiae that was more susceptible than RSP in a permethrin-susceptibility bioassay (i.e. LT50 22 min for UNS, vs. 42min for RSP) was compared with the RSP colony for levels of oxidases and esterases. The levels of both enzymes were very significantly higher in the RSP strain (P<0.0001). We speculate that use of impregnated nets selected for higher oxidase and esterase levels in An. gambiae to metabolize permethrin acquired from the nets. Both oxidase and esterase mechanisms could confer cross-resistance to other pyrethroids.  相似文献

The behavioral responses exhibited between four permethrin-resistant field-derived populations and a long-established, permethrin susceptible colony population of Culex quinquefasciatus mosquitoes was assessed for behavioral responses to permethrin in an excito-repellency (ER) test system. After a 30-min exposure to two different insecticide concentrations (200 and 500 mg/m2) in paired contact and noncontact test designs, the initial knockdown and escape (exit) responses were recorded followed by mortality after 24-h. All five test populations rapidly escaped from the chambers treated with permethrin regardless of background insecticide susceptibility status or chemical concentration. The greatest contact escape response was seen from the colonized population with 88 % of specimens escaping the chamber within 30-min. Contact irritancy was the predominant response compared to relatively weak noncontact spatial repellency in all test populations at both low and high concentrations. At the lower concentration, the contact escape responses did not appear influenced by background susceptibility status, whereas at the higher concentration the permethrin-resistant field populations exhibited significantly reduced avoidance behavior compared to the laboratory susceptible strain.  相似文献

The frequency of insecticide-resistance alleles for two genes (Vssc1 and CYP6D1) was studied in field collected populations of house flies from two different climates. While the frequency of these resistance alleles in flies at dairies from four states has recently been reported, there is no information on the relative change of these allele frequencies over time. House flies were collected during the 2003-2004 season from New York and Florida before the first application of permethrin, during the middle of the field season, after the final application, and again the following spring (following months without permethrin use). Bioassay results indicated that homozygous susceptible and extremely resistant flies were rare, while moderately and highly resistant individuals were relatively common at all times in both states. The frequency of resistance alleles at the New York dairy rose during the season and declined over the winter, suggesting an overwintering fitness cost associated with these alleles. The super-kdr allele was detected for the first time in North America at the end of 2003. In Florida the frequency of the resistance alleles did not increase during the spray season or decrease during the winter, suggesting there is substantial immigration of susceptible alleles to the Florida dairy and no overwintering fitness cost associated with resistance alleles in this climate. Resistance to permethrin correlated well with the frequency of the Vssc1 and CYP6D1 resistance alleles in flies from New York, but not as well in the population from Florida. This suggests there may be a new resistance mechanism or allele evolving in Florida.  相似文献

Anopheles subpictus s.l. Grassi (Diptera: Culicidae) is a malaria vector in South Asia, where insecticides are the mainstay for vector control interventions. Information on any variation in metabolic enzyme levels in mosquitoes is helpful with respect to adapting alternative strategies for vector control. The scarce data on the biochemical basis of insecticide resistance in malaria vectors of Pakistan limit the available information for vector control interventions within the country. The insecticide susceptibility status and its biochemical basis against dichlorodiphenyltrichloroethane (DDT) (4%), deltamethrin (0.05%) and permethrin (0.75%) in An. subpictus s.l. collected from all Tehsils of district Kasur were evaluated. For this purpose, a World Health Organization susceptibility bioassay was performed followed by the detection of altered metabolic enzyme activity using biochemical assays. Similarly, a significant difference in knock‐down effect was observed among field collected and susceptible strain against all insecticides 24 h post exposure. The overall mean mortality rates of DDT, deltamethrin and permethrin were 27.86% [95% confidence interval (CI) = 29.65–26.06], 44.89% (95% CI = 46.23–43.54) and 78.82% (95% CI = 80.16–77.47), respectively. The biochemical assays revealed an elevated level of metabolic enzymes in the field population. The results provide evidence of resistance against organochlorine and pyrethroid groups in a field population of An. subpictus s.l. from district Kasur mediated by multiple metabolic mechanisms, including acetylcholinesterases, esterases, cytochrome P450 and glutathione S‐transferases.  相似文献

Chlorantraniliprole is the first commercial insecticide from a new class of chemistry, the anthranilic diamides. Chlorantraniliprole provides an effective alternative insecticide for control of Plutella xylostella (L.) populations resistant to other insecticides. Baseline susceptibility to chlorantraniliprole for P. xylostella was surveyed previously from 16 geographical populations sampled from China during 2008-2009, and the median lethal concentrations (LC50s) varied among populations from 1.8- to 8.9-fold higher than the LC50 of a susceptible strain (Roth). In the present work, 20 field populations of P. xylostella sampled in 2010-2011 from China were tested with laboratory bioassays to determine if resistance to chlorantraniliprole had evolved in the field. The LC50s of the 14 populations from northern China ranged from 1.7- to 5.4-fold compared with the LC50 of Roth, which indicates these populations remain reasonably susceptible to chlorantraniliprole. However, the LC50s of the six populations from southern China (Guangdong Province) were 2.6-, 12-, 18-, 81-, 140-, and 2,000-fold higher than the LC50 of Roth. The results showed that high levels of resistance to chlorantraniliprole had evolved in field populations from Guangdong Province of southern China. Intensive use and misuse of chlorantraniliprole may be responsible for the rapid evolution of high-level resistance in P. xylostella in this region. The implementation of resistance monitoring plans and resistance management strategies is urgently needed in China to preserve susceptibility to chlorantraniliprole in P. xylostella.  相似文献

Abstract:  To monitor the resistance of field populations of the diamondback moth Plutella xylostella in China to the insecticidal protein Cry1Ac, Cry1Ba and commercial formulation Bacillus thuringiensis var. kurstaki (Btk), six representative populations of the diamondback moth were collected from Shanghai, Shandong, Hubei, Hunan, Zhejiang and Guangdong provinces of China where crucifer crop plants are intensively planted. Bioassay results showed that the populations of the diamondback moth from different locations exhibited different levels of resistance, compared with a susceptible laboratory population. The Guangdong field population was 56.15- and 21.90-fold resistant to Cry1Ac and Btk, respectively. Shanghai, Hunan, Shandong and Zhejiang populations were 37.85-, 17.24-, 10.24- and 9.41-fold resistant to Cry1Ac, respectively, but were not resistant to Btk. The Hubei population did not show resistance to Cry1Ac and Btk. Almost all tested populations were susceptible to Cry1Ba, but the Guangdong population showed some tolerance to Cry1Ba with a LC50 of 0.69  μ g/ml which was 6.17-fold higher than that of the susceptible population. The results suggested that the complex resistance patterns of field populations of P. xylostella need to be considered for expression of Bt toxin genes in genetically-engineered crop plants and commercial formulations.

1 Control failures of insecticides used against the tomato leafminer Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) in Brazil led to the investigation of the possible occurrence of resistance of this insect pest to abamectin, cartap, methamidophos and permethrin. 2 The insect populations were collected from seven sites in the states of Minas Gerais, Rio de Janeiro, and São Paulo. These populations were subjected to concentration–mortality bioassays using insecticide‐impregnated filter papers. 3 We were unable to obtain a single population which provided a susceptibility standard for all insecticides tested. Therefore, the resistance levels were estimated in relation to the most susceptible population to each insecticide. Resistance to abamectin and cartap were observed in all populations when compared with the susceptible standard population, with resistance ratios ranging from 5.2‐ to 9.4‐fold and from 2.2‐ to 21.9‐fold for abamectin and cartap, respectively. Resistance to permethrin was observed in five populations with resistance ratios ranging from 1.9‐ to 6.6‐fold, whereas resistance to methamidophos was observed in four populations with resistance ratios ranging from 2.6‐ to 4.2‐fold. 4 The long period and high frequency of use of these insecticides against this insect pest suggest that the evolution of insecticide resistance on them has been relatively slow. Alternatively, the phenomenon might be widespread among Brazilian populations of T. absoluta making the finding of suitable standard susceptible populations difficult and leading to an underestimation of the insecticide resistance levels in this pest. 5 Higher levels of resistance to abamectin, cartap and permethrin are correlated with greater use of these compounds by growers. This finding suggests that local variation in insecticide use was an important cause of variation in susceptibility.  相似文献
