Studies on the development of cell populations are often based on results of the theory of stochastic birth- and death-processes (continuous or discrete (seee.g. references inVogel, Niewisch &Matioli (1969), in some cases, death may be interpreted not as actual death of the cell bute.g. as a recruitment of the cell considered into another cell compartment, etc.). It is usually assumed that the conditions for the development are homogeneous,i.e. that the probabilities of births and deaths are independent on the time. However, in most situations, this assumption is not fulfilled (owing to the maturation and differentiation of cells, changes of the microenvironment, inducing factors, etc.). Then it is necessary to study the development of cell populations under non-homogeneous conditions. In this paper, some properties of appropriate birth- and death-processes under nonhomogeneous conditions are studied; formulae given in section 4 permit calculation of some characteristics describing the cell population size distribution at individual discrete epochs (at individual generations) during the development of the cell population considered.  相似文献

The carbon flux distribution in the central metabolism of Corynebacterium glutamicum was studied in batch cultures using [1-13C]- and [6-13C]glucose as substrate during exponential growth as well as during overproduction of l-lysine and l-glutamate. Using the 13C NMR data in conjunction with stoichiometric metabolite balances, molar fluxes were quantified and normalised to the glucose uptake rate, which was set to 100. The normalised molar flux via the hexose monophosphate pathway was 40 during exponential growth, whereas it was only 17 during l-glutamate production. During l-lysine production, the normalised hexose monophosphate pathway flux was elevated to 47. Thus, the carbon flux via this pathway correlated with the NADPH demand for bacterial growth and l-lysine overproduction. The normalised molar flux in the tricarboxylic acid cycle at the level of 2-oxoglutarate dehydrogenase was 100 during exponential growth and 103 during l-lysine secretion. During l-glutamate formation, the normalised flux through the tricarboxylic acid cycle was reduced to 60. In contrast to earlier NMR studies with C. glutamicum, no significant activity of the glyoxylate pathway could be detected. All experiments indicated a strong in vivo flux from oxaloacetate back to phosphoenolpyruvate and/or pyruvate, which might be due to phosphoenolpyruvate carboxykinase activity in C. glutamicum.  相似文献

Multiple births are very rare among gibbons. The birth of siamang twins at the Zürich Zoo in 1992 therefore presented a valuable opportunity to observe the development of the twins and to contrast it with a survey of previous reports on the development of single offspring of siamang and gibbons of thelar group. Furthermore, the hypothesis that the presence of twins among siamang may facilitate the occurrence of helping behaviour (defined as the care of offspring by individuals other than their parents) was re-examined (Dielentheis et al., 1991). The Zürich twins (one male and one female) were observed for a total of 74 hr during their first year of life. The results show that: (1) The twins exhibited more rapid behavioural development than that reported for single offspring. (2) A clear difference between the twins was observed: the female twin developed more rapidly than the male. (3) Neither the twins' father nor their older sisterRama was ever observed carrying the twins. The hypothesis ofDielentheis et al. (1991) is hence not supported by the present study, although it is possible that the older sisterRama did not carry the twins because she was younger than the juvenile in that study. (4) Siamangs may have a longer maturation period than gibbons of thelar group.  相似文献

A pollen diagram was derived from a 150 cm core taken from the shallow hypersaline Lake Maharlou in the south-eastern part of the Zagros Mountains, SW Iran. The pollen record shows that Quercus brantii woodland and Pistacia–Amygdalus scrub dominated the area during the late Holocene. The record starts at around 5700 cal b.p. with a dry period during which both Pistacia–Amygdalus scrub and Quercus brantii woodland were at their minimum extent. This period was followed by the expansion of Pistacia–Amygdalus scrub in the area and the spread of Quercus brantii woodlands at higher altitudes. An important occupation phase, characterized by the appearance of several cultivated tree species such as Juglans, Olea, Vitis and Platanus, started at ca. 4300 cal b.p., coinciding with the onset of the Bronze Age civilization of Jiroft in Central Iran. Human activities become very clear after 3700 cal b.p. Around 2700 cal b.p., extensive stands of Pistacia–Amygdalus scrub became profoundly degraded, presumably under strong human pressure coinciding with the beginning of the Persian Empires. The maximum expansion of the Quercus brantii woodland occurred about 2100 to 1700 cal b.p. This woodland remained relatively stable until the end of the diagram at 400 cal b.p.  相似文献

We used ion-sensitive, double-barrel microelectrodes to measure changes in hepatocyte transmembrane potential (V m), intracellular K+, Cl-, and Na+ activities (a i k, a Cl i and a Na i ), and water volume during l-alanine uptake. Mouse liver slices were superfused with control and experimental Krebs physiological salt solutions. The experimental solution contained 20 m l-alanine, and the control solution was adjusted to the same osmolality (305 mOsm) with added sucrose. Hepatocytes also were loaded with 50 mm tetramethylammonium ion (TMA+) for 10 min. Changes in cell water volume during l-alanine uptake were determined by changes in intracellular, steady-state TMA+ activity measured with the K+ electrode. Hepatocyte control V m was -33±1 mV. l-alanine uptake first depolarized V m by 2±0.2 mV and then hyperpolarized V m by 5 mV to-38±1 mV (n = 16) over 6 to 13 min. During this hyperpolarization, a Na i increased by 30% from 19±2 to 25±3 mm (P < 0.01), and a K i did not change significantly from 83±3 mm. However, with added ouabain (1 mm) l-alanine caused only a 2-mV increase in V m, but now a K i decreased from 61±3 to 54±5 mm (P < 0.05). Hyperpolarization of V m by l-alanine uptake also resulted in a 38% decrease of a Cl i from 20±2 to 12±3 mm (P < 0.001). Changes in V m and V ClV m voltage traces were parallel during the time of l-alanine hyperpolarization, which is consistent with passive distribution of intracellular Cl with the V m in hepatocytes. Added Ba2+ abolished the l-alanineinduced hyperpolarization, and a Cl i remained unchanged. Hepatocyte water volume during l-alanine uptake increased by 12±3%. This swelling did not account for any changes in ion activities following l-alanine uptake. We conclude that hepatocyte a K i is regulated by increased Na+-K+ pump activity during l-alanine uptake in spite of cell swelling and increased V m due to increased K+ conductance. The hyperpolarization of V m during l-alanine uptake provides electromotive force to decrease a Cl i . Escherichia coli W was genetically engineered to produce l-alanine as the primary fermentation product from sugars by replacing the native d-lactate dehydrogenase of E. coli SZ194 with alanine dehydrogenase from Geobacillus stearothermophilus. As a result, the heterologous alanine dehydrogenase gene was integrated under the regulation of the native d-lactate dehydrogenase (ldhA) promoter. This homologous promoter is growth-regulated and provides high levels of expression during anaerobic fermentation. Strain XZ111 accumulated alanine as the primary product during glucose fermentation. The methylglyoxal synthase gene (mgsA) was deleted to eliminate low levels of lactate and improve growth, and the catabolic alanine racemase gene (dadX) was deleted to minimize conversion of l-alanine to d-alanine. In these strains, reduced nicotinamide adenine dinucleotide oxidation during alanine biosynthesis is obligately linked to adenosine triphosphate production and cell growth. This linkage provided a basis for metabolic evolution where selection for improvements in growth coselected for increased glycolytic flux and alanine production. The resulting strain, XZ132, produced 1,279 mmol alanine from 120 g l−1 glucose within 48 h during batch fermentation in the mineral salts medium. The alanine yield was 95% on a weight basis (g g−1 glucose) with a chiral purity greater than 99.5% l-alanine. The weekend effect hypothesis proposes that captive primates are more likely to give birth during times of low disturbance and reduced staff activity. The hypothesis specifically predicts that laboratory‐housed primates will be more likely to give birth during the weekend than weekdays when staff activity is reduced. To date, support for the weekend effect hypothesis has been mixed and based on studies with relatively few subjects. To further examine the hypothesis, we analyzed the birthing patterns of three genera of laboratory‐housed primates: squirrel monkeys (Saimiri species, N = 2,090 births), owl monkeys (Aotus species, N = 479 births), and rhesus macaques (Macaca mulatta, N = 2,047 births). Contrary to predictions derived from the weekend effect hypothesis, the frequencies of births during weekends for all taxa were not significantly different from rates that would be expected by chance. However, while there was no variance across days of the week, all three taxa gave birth at nighttime, when staff was absent. This parallels reports of births in wild and captive monkeys, both diurnal and nocturnal, which are more likely to give birth during the night; plausibly a time when the environmental and social disturbance is lowest and the mother is safest to bond with her newborn infant. As all births occurred at night, we also explored the relationship between the lunar cycle and the timing of births timing. While the diurnal primates (i.e., Saimiri and Macaca) were no more likely to give birth on “bright” nights than “dark” nights, owl monkeys (Aotus) had a much higher frequency of births on bright nights than darker ones, and at rates that deviated from chance. Our data provide a more detailed understanding on how the environment may influence captive monkey births but do not support the oft‐cited weekend effect hypothesis.  相似文献

Enzymes of polyol metabolism were studied in basidiospore germination of Schizophyllum commune during periods of in vivo arabitol and mannitol pool depletion (growth on glucose-asparagine) and during their subsequent synthesis (growth on acetate-NH 4 + ). Optimal conditions for assays were established and specific activities of enzymes employing d-arabitol, d-mannitol, d-ribulose, d-fructose and d-xylulose as substrates were traced. Inquiries into the products formed during these reactions showed that d-ribulose generated arabitol while d-fructose produced mannitol with d-xylulose giving rise to xylitol. The dehydrogenase reactions were further investigated using polyacrylamide disc gel electrophoresis. Here was revealed the existence of at least two separate enzymatic activities pertaining to the catabolism of arabitol and mannitol. Also noted were the electrophoretic patterns when d-sorbitol, ribitol, xylitol and ethanol were used as substrates.  相似文献

Summary Pitfall-traps are used to study the phenology and activity phenomena of four surface dwelling Collembola e. g. Entomobrya nivalis (Linné), Lepidocyrtus cyaneus Tullberg, Orchesella cincta (Linné) and Tomocerus minor (Lubbock).It is concluded that Entomobrya is active during the day, whereas Orchesella and Tomocerus are active during the night. As to Lepidocyrtus no definite conclusion can be drawn.A digging-in effect is demonstrated for Tomocerus minor. A negative effect of the roofs appears during the day for Entomobrya nivalis and Lepidocyrtus cyaneus.Weather conditions exert an influence upon these surface dwelling Collembola.
Zusammenfassung Die Fallenfangmethode wurde angewandt zum Studium der Phänologie und der Aktivität von vier Collembolen-Arten der Bodenoberfläche, nämlich Entomobrya nivalis (Linné), Lepidocyrtus cyaneus Tullberg, Orchesella cincta (Linné) und Tomocerus minor (Lubbock). Entomobrya erwies sich als tagaktive Art, Orchesella und Tomocerus sind nachtaktive Tiere. Für Lepidocyrtus waren die Resultate nicht eindeutig.Eine Reaktion auf das Eingraben der Fallen wurde festgestellt für Tomocerus minor.Fallendächer haben einen negativen Effekt auf den Fang von Entomobrya nivalis und Lepidocyrtus cyaneus während des Tages.Die Aktivität dieser Collembolen der Oberfläche wird durch Witterungsverhältnisse beeinflußt.

Holocene environmental and climatic changes are reconstructed using analyses of biological proxies in lake sediments from Vuolep Njakajaure, a lake located near the altitudinal treeline in northern Sweden (68°20′ N, 18°47′ E). We analysed biological proxy indicators from both aquatic and terrestrial ecosystems, including diatoms, pollen and chironomid head capsules, in order to reconstruct regional Holocene climate and the development of the lake and its catchment. During the early Holocene and after 2500 cal b.p., Fragilaria taxa dominated the diatom assemblages, whereas planktonic Cyclotella taxa prevailed during the major part of the Holocene (7800–2300 cal b.p.), indicating the importance of the pelagic habitat for diatom assemblage composition. The planktonic diatoms appeared at the same time as Alnus became established in the catchment, probably altering nutrient availability and catchment stability. The pollen record is dominated by mountain birch (Betula pubescens ssp. tortuosa) pollen throughout the Holocene, but high percentage abundances of Scots pine (Pinus sylvestris) pollen suggest the presence of a mixed pine-birch forest during the mid-Holocene (6800–2300 cal b.p.). Head capsules of Tanytarsini and Psectrocladius dominated the chironomid assemblage composition throughout the Holocene, in combination with Corynocera ambigua after 2300 cal b.p. A quantitative, diatom-based reconstruction of mean July air temperature indicated a relatively cold temperature during the early Holocene (9000–8000 cal b.p.) and after ca. 2300 cal b.p., whereas the mid-Holocene period is characterised by stable and warm temperatures. The overall patterns of Holocene climate and environmental conditions are similarly described by all biological proxy-indicators, suggesting relatively warm conditions during the mid-Holocene (ca. 7800–2300 cal b.p.), with a subsequent colder climate after 2300 cal b.p. However, the onset and magnitude of the inferred changes differ slightly among the proxies, illustrating different responses to lake development phases, land-uplift, and climate forcing (e.g., insolation patterns) during the Holocene in northern Sweden.  相似文献

Summary In South-West Madagascar, the area from which outbreaks ofLocusta migratoria capito (Sauss.) andNomadacris septemfasciata (Serv.) proceed, a seasonal pluviometric inversion (an unusual occurence) will mean successful and simultaneous hatches due to edaphic and biotic effects. Edaphic effect: it maintains ground-moisture to optimum conditions for the eggs, which thereby are saved the over-dryness during the cool season and the over-moisture during the hot season in normal years. Rare and light rains in the hot season mean simultaneous hatches. Biotic effect: dryness at an undue time of the year disturbs the biological status, by jeopardizing the main enemy of the eggs:Caloglyphus berlesei Mich. (Acarina).   相似文献

The late-glacial vegetation development in northern Norway in response to climate changes during the Aller?d, Younger Dryas (YD), and the transition to the Holocene is poorly known. Here we present a high-resolution record of floral and vegetation changes at lake Lusvatnet, south-west And?ya, between 13500 and 8000 cal b.p. Plant macrofossil and pollen analyses were done on the same sediment core and the proxy records follow each other very closely. The core has also been analyzed using an ITRAX XRF scanner in order to check the sediment sequence for disturbances or hiatuses. The core has a good radiocarbon-based chronology. The Saksunarvatn tephra fits very well chronostratigraphically. During both the Aller?d and the Younger Dryas time-periods arctic vegetation prevailed, dominated by Salix polaris associated with many typically arctic herbs such as Saxifraga cespitosa, Saxifraga rivularis and Oxyria digyna. Both periods were cold and dry. Between 12450 and 12250 cal b.p. during the Younger Dryas chronozone, the assemblage changed, particularly in the increased abundance of Papaver sect. Scapiflora and other high-Arctic herbs, suggesting the development of polar desert vegetation mainly as a response to increased aridity. After 11520 cal b.p. a gradually warmer and more oceanic climate initiated a succession to dwarf-shrub vegetation and the establishment of Betula woodland after 1,000 years at c. 10520 cal b.p. The overall late-glacial aridity contrasts with oceanic conditions in southern Norway and is probably related to sea-ice extent.  相似文献

The metabolism ofl-proline toN-acetyl-d-glucosamine (GlcNAc) during germ tube formation ofCandida albicans (C. albicans) ATCC 1002 was studied. In uptake experiments, 6.9 nmol ofl-[14C]proline were taken up by 1×106 cells during 3 h of incubation at 37°C. The percentage of germ tube formation was 94 under the same condition. The presence of GlcNAc reduced the uptake ofl-proline to 3.0 nmol. The percentage of germ tube formation was 95 in the presence and absence of GlcNAc. The [3H]GlcNAc uptake was 3.0 nmol and was constant whetherl-proline was present or not. After the preparation of a chitin fraction from germ tubes that were labeled withl-[14C]proline, the radioactivity froml-proline was detected in the glucosamine (GlcN) fraction by thin-layer chromatography (TLC). The metabolism ofl-proline to GlcNAc in chitin during germ tube formation was confirmed in this experiment.  相似文献

From a monthly survey over a ten-year period (1983–1992) of the Ishitegawa Reservoir, Japan, two multiple regression equations describing the mean chlorophylla (Chl-a) concentration at 0.5 m depth during July, August and September (C S ) and the maximum Chl-a concentration in the photic zone, including its subsurface maximum during this period (C M ), were obtained. The conductivity at 0.5 m depth in May and the mean air temperature during May or during May and June were used as independent variables. These equations were calculated using seven years of data (1983–1988 and 1992). From 1989 to 1991 two impermeable plastic-coated sheet fences were installed across the upper end of the reservoir along the top 5 m of water column. The equations were used to determine the effectiveness of this flow regulation on the summer Chl-a concentration. In 1989 and 1991, when the fences were in place from June to August, no effects were found on the observed C S and C M values. In 1990, when the fences had been in place since October 1989, the observed values were lower than the predicted values.  相似文献

Cell growth limitation is known to be an important condition that enhances l-valine synthesis in Corynebacterium glutamicum recombinant strains with l-isoleucine auxotrophy. To identify whether it is the limited availability of l-isoleucine itself or the l-isoleucine limitation-induced rel-dependent ppGpp-mediated stringent response that is essential for the enhancement of l-valine synthesis in growth-limited C. glutamicum cells, we deleted the rel gene, thereby constructing a relaxed (rel ) C. glutamicum ΔilvA ΔpanB Δrel ilvNM13 (pECKAilvBNC) strain. Variations in enzyme activity and l-valine synthesis in rel + and rel strains under conditions of l-isoleucine excess and limitation were investigated. A sharp increase in acetohydroxy acid synthase (AHAS) activity, a slight increase in acetohydroxyacid isomeroreductase (AHAIR) activity, and a dramatic increase in l-valine synthesis were observed in both rel + and rel cells exposed to l-isoleucine limitation. Although the positive effect of induction of the stringent response on AHAS and AHAIR upregulation in cells was not confirmed, we found the stringent response to be beneficial for maintaining increased AHAS, dihydroxyacid dehydratase, and transaminase B activity and l-valine synthesis in cells during the stationary growth phase.  相似文献

We recently proposed a metabolic engineering strategy for l-ornithine production based on the hypothesis that an increased intracellular supply of N-acetylglutamate may further enhance l-ornithine production in a well-defined recombinant strain of Corynebacterium glutamicum. In this work, an argJ-deficient arginine auxotrophic mutant of C. glutamicum is suppressed by a different locus of C. glutamicum ATCC13032. Overexpression of the NCgl1469 open reading frame (ORF), exhibiting N-acetylglutamate synthase (NAGS) activity, was able to complement the C. glutamicum arginine-auxotrophic argJ strain and showed increased NAGS activity from 0.03 to 0.17 units mg−1 protein. Additionally, overexpression of the NCgl1469 ORF resulted in a 39% increase in excreted l-ornithine. These results indicate that the intracellular supply of N-acetylglutamate is a rate-limiting step during l-ornithine production in C. glutamicum.  相似文献

Diastereoselectivity-enhanced mutants of l-threonine aldolase (l-TA) for l-threo-3,4-dihydroxyphenylserine (l-threo-DOPS) synthesis were isolated by error-prone PCR followed by a high-throughput screening. The most improved mutant was achieved from the mutant T3-3mm2, showing a 4-fold increase over the wild-type l-TA. When aldol condensation activity was examined using whole cells of T3-3mm2, its de was constantly maintained at 55% during the batch reactions for 80 h, yielding 3.8 mg l-threo-DOPS/ml.  相似文献

Bacillus subtilis has various cell wall hydrolases, however, the functions and hydrolase activities of some enzymes are still unknown. B. subtilis CwlK (YcdD) exhibits high sequence similarity with the peptidoglycan hydrolytic l,d-endopeptidase (PLY500) of Listeria monocytogenes phage and CwlK has the VanY motif which is a d-alanyl-d-alanine carboxypeptidase (Pfam: http://www.sanger.ac.uk/Software/Pfam/). The β-galactosidase activity observed on cwlK-lacZ fusion indicated that the cwlK gene was expressed during the vegetative growth phase, and Western blotting suggested that CwlK seems to be localized in the membrane. Truncated CwlK fused with a histidine-tag (h-ΔCwlK) was produced in Escherichia coli and purified on a nickel column. The h-ΔCwlK protein hydrolyzed the peptidoglycan of B. subtilis, and the optimal pH, temperature and NaCl concentration for h-ΔCwlK were pH 6.5, 37°C, and 0 M, respectively. Interestingly, h-ΔCwlK could hydrolyze the linkage of l-alanine-d-glutamic acid in the stem of the peptidoglycan, however, this enzyme could not hydrolyze the linkage of d-alanine-d-alanine, suggesting that CwlK is an l,d-endopeptidase not a d,d-carboxypeptidase. CwlK could not hydrolyze polyglutamate from B. natto or peptidoglycan of Staphylococcus aureus. This is the first report describing the characterization of an l,d-endopeptidase in B. subtilis and also the first report in bacteria of the characterization of a PLY500 family protein encoded in chromosomal DNA. Summary We examined the ability of stingless bees to recruit nest mates to a food source (i) in group foraging species laying pheromone trails from the food to the nest (Trigona recursa Smith, T. hypogea Silvestri, Scaptotrigona depilis Moure), (ii) in solitary foraging species with possible but still doubtful communication of food location inside the nest (Melipona seminigra Friese, M. favosa orbignyi Guérin), and (iii) in species with a less precise (Nannotrigona testaceicornis Lep., Tetragona clavipes Fab.) or no communication (Frieseomelitta varia Lep.). The bees were allowed to collect food (sugar solution or liver in the necrophageous species) ad libitum and the forager number to accumulate, as it would do under normal unrestrained conditions. The median number of bees collecting differed considerably among the species (1.0–1436.5). It was highest in the species employing scent trails. The time course of recruitment was characteristic for most of the species and largely independent of the number of foragers involved. The two Melipona species recruited other bees significantly faster than T. recursa, S. depilis, and N. testaceicornis during the first 10 to 30 minutes of an experiment. In species laying a scent trail to guide nestmates to a food source the first recruits appeared with a delay of several minutes followed by a quick increase in forager number. The median time required to recruit all foragers available differed among the species between 95.0 and 240.0 min. These differences can at least partly be explained by differences in the recruitment mechanisms and do not simply follow from differences in colony biomass.  相似文献
