One-year rooted shoots of apple M. VII rootstock were grownfor a single season by spraying their roots continuously withnutrient solutions containing <3, 10, 45, and 135 ppm Mg.These treatments respectively produced plants with a severeMg deficiency (Mg(0)), a mild deficiency (Mg(1)), normal foliageand growth (Mg(2)), and a high level of Mg (Mg(3)). The effectof Mg nutrition on the leaves can explain most of the observedeffects on vegetative growth. Depression of growth of Mg(1))plants was not detected until late in the season although necrosis,typical of Mg deficiency, occurred on the lower leaves by mid-July.This loss of photosynthetic area of leaf was to a large extentcompensated for by increased efficiency of the remaining greenleaves; as the deficiency symptoms progressed, the loss of leaf,and therefore of carbohydrate production, predominated and totalgrowth was affected. The form of the plant was little altered. Symptoms were apparent on leaves of severely deficient Mg(0)plants by late June, and reductions in growth and modificationsin form by August. The poor root systems and thin shoots arelargely attributed to loss of leaf rather than to the low Mgconcentration in these organs. No symptoms of toxicity developed on the Mg(3) plants althoughby the end of the season the total dry weights were less thanthose of the Mg(2) controls, and the plants also produced thickershoots. Plant dry weight, and more especially Mg uptake, increasedfrom Mg(0) to Mg(2) but above this level the gradient of Mguptake in relation to supply was less steep. The excess Mg takenup at the Mg(j) level led to the relatively lower weights ofthese plants. Much of the excess Mg accumulated in the leaves,especially in the older ones. Mg moved from, or accumulatedin, the lower leaves of the shoot according to the level ofMg supplied to the plants. Thus the shoot apex tended towardsnormality both in the concentration of Mg and in its growth. In general, growth of the plant, and of individual componentparts, governed the uptake of elements other than Mg, althoughmore K, Ca, and Mn and less Fe per unit of plant dry weightaccumulated at the lower levels of Mg than at the higher levels.  相似文献

Apple rootstocks were grown with either 0.02 ppm Fe (Fe0) or5 ppm (Fe3), to give very chlorotic or dark-green plants. Toinvestigate whether iron can be supplied through leaves insteadof roots the shoots of half the plants in each treatment weredipped periodically in solutions of iron. This prevented chlorosisin Fe0 plants and increased their growth, which did not, however,equal that of Fe3 plants supplied with iron through the roots.Growth of Fe3 plants was reduced by dipping. Iron was not translocated from leaves to roots, although theconcentration in leaves was greatly increased by dipping. Dippingreduced the amount of manganese in Fe0 roots to one-quarterof that in roots of undipped Fe0 plants. Effects of treatmentson nitrogen, potassium, calcium, magnesium, and copper levelsare also described.  相似文献

Carob seedlings were grown hydroponically for 9 weeks under360 and 800 µl l-1CO2. One of two nitrogen sources, nitrateor ammonium, was added to the nutrient medium at concentrationsof 3 mol m-3. Root systems of the developing plants suppliedwith nitrate compared to those supplied with ammonium were characterizedby:(a)more biomass on the lower part of the root;(b)fewer lateralroots of first and second order;(c)longer roots;(d)higher specificroot length;(e)a smaller root diameter. The morphology of theroot systems of nitrate-fed plants changed in the presence ofelevated carbon dioxide concentrations, resembling, more closely,that of ammonium-fed plants. Total leaf area was higher in ammonium-than in nitrate-fed plants. Nitrate-fed plants had greater totalleaf area in the presence of high carbon dioxide than in normalCO2, due to an increase in epidermal cell size that led to developmentof larger leaflets with lower stomatal frequency. The observedchanges in the morphology of roots and shoots agreed with theresults observed for total biomass production. Nitrate-fed plantsincreased their biomass production by 100% in the presence ofelevated CO2compared to 15% in ammonium-fed plants, indicatingthat the response of carob to high CO2concentrations is verydependent on the nitrogen source. Under elevated CO2, nitrate-grownplants had a larger content of sucrose in both roots and shoots,while no significant difference was observed in the contentof sucrose in ammonium-grown plants, whether in ambient or enrichedcarbon dioxide. The growth of garden orache, A triplex hortensis was studiedunder conditions of mild NaCl or Na2SO4 salinity. Growth, drymatter production and leaf size were substantially stimulatedat 10 mM and 50 mM Na+ salts. Increased growth, however, appearedto be due to a K+-sparing effect of Na+ rather than to salinityper se. The distribution of K+ and Na+ in the plant revealeda remarkable preference for K+ in the roots and the hypocotyl.In the shoot the K/Na ratio decreased strongly with leaf age.However, the inverse changes in K+ and Na+ content with leafage were dependent on the presence of bladder hairs, which removedalmost all of the Na+ from the young leaf lamina. Measurementsof net fluxes of K+ and Na+ into roots and shoots of growingAtriplex plants showed a higher K/Na selectivity of the netion flux to the root compared to the shoot. With increasingsalinity the selectivity ratio SK, Na* of net ion fluxes tothe roots and to the shoots was increased. The data suggestthat recirculation of K+ from leaves to roots is an importantlink in establishing the K/Na selectivity in A. hortensis plants.The importance of K+ recirculation and phloem transport forsalt tolerance is discussed. The effects of salinity on growth, water relations, glycinebetainecontent, and ion accumulation in the perennial halophyte Atriplexgriffithii var. stocksii were determined. The following questionswere addressed: (1) What effect does salinity have on growthresponses at different ages? (2) Is A. griffithii an ion accumulator?(3) Does A. griffithii accumulate glycinebetaine in responseto salinity? Atriplex griffithii plants were grown in pots at0, 90, 180 and 360  m M NaCl in sand culture in a plantgrowth chamber and plants were harvested after 30, 60 and 90d. Plant total dry weight was significantly inhibited at 360m M NaCl. Root growth showed a substantial promotion at 90 mM NaCl. The water potential and osmotic potential of shootsbecame more negative with increasing salinity and time of growth.The Na+and Cl-content in both shoots and roots increased withincreases in salinity. Increased treatment levels of NaCl induceddecreases in Ca+, K+and Mg2+in plants. Atriplex griffithii accumulateda large quantity of ions, with the ash content reaching 39%of the dry weight in leaves. Inorganic ion accumulation is significantin osmotic adjustment and facilitates water uptake along a soil-plantgradient. Glycinebetaine concentration was low in roots, andin stems it increased with increases in salinity. Little is known about transport of Zn from leaves to other plantorgans. The present study tested a range of Zn forms appliedfoliarly for their suitability to provide adequate Zn nutritionto wheat (Triticum aestivum L.). Transport of65Zn applied eitherto leaves or to one side of the root system was also studied.Inorganic (ZnO, ZnSO4) and chelated sources of Zn (ZnEDTA, glycine-chelatedBiomin Zn) applied foliarly provided sufficient Zn for vigorousgrowth. Zinc concentrations in roots and shoots were in thesufficiency range, except in the -Zn control. Foliar treatmentswith ZnSO4and chelated Zn forms resulted in shoot Zn concentrationsin 7-week-old plants being about two-fold greater than thosein plants supplied with Zn in the root environment or via foliarspray of ZnO. Adding surfactant to foliar sprays containingchelated forms of Zn did not cause negative growth effects,but surfactant added to ZnO or ZnSO4foliar sprays decreasedshoot growth. Adding urea to the ZnO foliar spray had no effecton shoot growth. Foliarly-applied65Zn was translocated to leavesabove and below the treated leaf as well as to the root tips.Stem girdling confirmed that65Zn transport toward lower leavesand roots was via the phloem. Split-root experiments showedintensive accumulation of65Zn in the stem and transport to allleaves as well as to the root tips in the non-labelled sideof the root system. Foliar application of Zn in inorganic ororganic form is equally suitable for providing adequate Zn nutritionto wheat. The growth (fresh weight), morphogenesis (leaves, roots and shoots) and essential oil composition of mint (Mentha sp. L.) and thyme (Thymus vulgaris L.) plants were determined after 8 weeks under 350, 1,500, 3,000, 10,000 and 30,000 µmol mol-1 CO2. Plants were grown in vitro on basal medium (BM) consisting of Murashige and Skoog salts and 0.8% agar that contained either 0 or 3% sucrose under a 16-h (day)/8-h (night) photoperiod at a light intensity of 180 µmol s-1 m-2 or in soil in a greenhouse under conditions of natural sunlight. Ultra-high CO2 levels (i.e. ́,000 µmol mol-1 CO2) substantially increased fresh weights, leaves, shoots and roots for all plants compared to plants grown under ambient air (350 µmol mol-1 CO2) both in vivo and in vitro. For both species, 10,000 µmol mol-1 CO2 was the optimum concentration to obtain the largest growth and morphogenesis responses under in vitro conditions, while the 3,000- to 10,000-µmol mol-1 CO2 range provided the largest yields for soil-grown plants. Essential oil composition (i.e. monoterpenes, piperitonone oxide and limonene from mint and aromatic phenol and thymol from thyme) from the shoot portion of plants grown at all CO2 levels was analyzed in CH2Cl2 extracts via gas chromatography. Higher levels of secondary compounds occurred in vitro when cultures were grown under ultra-high CO2 levels than in ambient air. The concentration of thymol, a major secondary compound in thyme plants grown on BM containing sucrose, was 317-fold higher at 10,000 µmol mol-1 CO2 than in plants grown under ambient air conditions with the same BM. The levels of secondary compound in in-vitro-grown plantlets exposed to ultra-high CO2 concentrations exceeded those occurring in plants grown in the greenhouse under the same CO2 levels. Substantially higher levels of secondary compound occurred in plants under ultra-high CO2 levels on BM containing sucrose than on BM lacking sucrose or in soil. Thymol levels in thyme plants grown on BM containing sucrose were 3.9-fold higher at 10,000 µmol mol-1 CO2 than in shoots grown on BM without sucrose under the same CO2 levels. High positive correlations occurred between thymol concentrations and CO2 levels, fresh weights, shoots, roots and leaves when thyme shoots were grown on BM with sucrose. High positive correlations for thyme shoots grown on BM without sucrose only occurred between thymol concentrations and CO2 levels, fresh weights, shoots and leaves. No positive correlations between thymol concentrations and CO2 levels or any growth or morphogenesis responses occurred for thyme shoots when grown in soil.  相似文献

The effects of three ranges of CO2 concentration on growth,carbon distribution and loss of carbon from the roots of maizegrown for 14 d and 28 d with shoots in constant specific activity14CO2 are described. Increasing concentrations of CO2 led toenhancement of plant growth with the relative growth rate (RGR)of the roots affected more than the RGR of the shoots. Between16% and 21% of total net fixed carbon (defined as 14C retainedin the plant plus 14C lost from the root) was lost from theroots at all CO2 concentrations at all times but the amountsof carbon lost per unit weight of plant decreased with time.Possible mechanisms to account for these observations are discussed. Ten-day old kidney bean plants (Phaseolus vulgaris L. cv. Shin-edogawa)were exposed to 2.0 and 4–0 parts 10–6 NO2, and0.1, 0.2, and 0.4 parts 10–6 O3 alone or in combinationfor 2, 4, and 7 d. The effects of these air pollutants wereexamined with respect to the growth, partitioning of assimilates,nitrogen uptake, soluble sugar content, and root respiration. Decreased dry matter production was significant with all treatmentsexcept 2.0 parts 10–6 NO2 and 0.1 parts 10–6 O3.Exposure to mixtures of the gases produced more severe suppressionof growth than exposure to the single gases. Root/shoot ratiowas significantly lowered at 7 d by the gas treatments otherthan 2.0 parts 10–6 NO2 and 0.1 parts 10–6 O3. Thetotal nitrogen content of plants was increased by all treatments;the higher percent of nitrogen found with O3 exposure will resultfrom the growth retardation which increases the concentrationof nitrogen in the plants because the absorption of nitrogenby roots was unaffected. The combination of O3 with NO2 significantlydecreased the assimilation of NO2 by the plants. The concentration of soluble sugars in roots was decreased bythe gas treatments. There was a strong positive correlationbetween soluble sugar content and dry weight of the roots harvestedat 7 d. Root respiration was relatively unchanged until 5 dand then decreased significantly at 7 d by 2.0 parts 10–6NO2 and 0–2 parts 10–6 O3. Retarded growth of theroots and the decreased root respiration may be due to diminishedtranslocation of sugars from leaves to roots caused by exposureto air pollutants. Respiratory oxygen consumption by roots was 1·4- and1·6-fold larger in NH+4-fed than in NO-3-fed wheat (Triticumaestivum L.) and maize (Zea mays L.) plants respectively. Higherroot oxygen consumption in NH+4-fed plants than in NO-3-fedplants was associated with higher total nitrogen contents inNH+4-fed plants. Root oxygen consumption was, however, not correlatedwith growth rates or shoot:root ratios. Carbon dioxide releasewas 1·4- and 1·2-fold larger in NO+3-fed thanin NH+4-fed wheat and maize plants respectively. Differencesin oxygen and carbon dioxide gas exchange rates resulted inthe gas exchange quotients of NH-4-fed plants (wheat, 0·5;maize, 0·6) being greatly reduced compared with thoseof NO-3-fed plants (wheat, 1·0; maize, 1·1). Measuredrates of HCO-3 assimilation by PEPc in roots were considerablylarger in 4 mM NH+4-fed than in 4 NO-3 plants (wheat, 2·6-fold;maize, 8·3-fold). These differences were, however, insufficientto account for the observed differences in root carbon dioxideflux and it is probable that HCO-3 uptake is also importantin determining carbon dioxide fluxes. Two varieties (Nihonbare and Koshihikari) of rice plants (Oryzasativa L.) were grown hydro-ponically with two levels (20 and100 mg N liter –1) of ammonia. Variations in levels ofnatural abundance of 15N (15N) were analyzed in the ammoniaand organic nitrogen of shoots and roots, as well as in theammonia in the culture solution. There was substantial fractionationof nitrogen isotopes during the uptake of ammonia. When plantsabsorbed a large proportion of ammonia from a solution witha low concentration, less negative 15N values in plants andhigh positive 15N values in the ammonia remaining in solutionwere observed. The reverse was found when a smaller fractionof ammonia was absorbed from a solution with a higher concentrationof ammonia. The l5N values of ammonia in shoots and roots werehigher than in the respective constituent organic nitrogen,suggesting the fractionation of nitrogen isotopes during theassimilation of ammonia. Wild-type and mutant cells of the cyanobacterium(blue-green alga) Synechococcus PCC 7942 were grown in nitrate-or ammonia-containing medium as the source of nitrogen. Fractionationof nitrogen isotopes during the uptake of nitrate was limited,whereas that during the uptake of ammonia was considerable. 1 In this report, the term ammonia refers indiscriminately toboth NH3 or NH4+. This work aimed to study the impacts of acquisition and assimilationof various nitrogen sources, i.e. NO3, NH4+ or NH4NO3,in combination with gaseous NH3 on plant growth and acid-basebalance in higher plants. Plants of C3 Triticum aestivum L.and C4 Zea mays L. grown with shoots in ambient air in hydroponicculture solutions with 2 mol m–3 of nitrogen source asNO3, NH4+ or NH4NO3 for 21 d and 18 d, respectively,had their shoots exposed either to 320 µg m–3 NH3or to ambient air for 7 d. Variations in plant growth (leaves,stubble and roots), and OH and H+ extrusions as wellas the relative increases in nitrogen, carbon and carboxylatewere determined. These data were computed as H+/N, H+/C, (C-A)/N,and (C-A)/C to analyse influences of different nitrogen sourceson acid-base balance in C3 Triticum aestivum and C4 Zea maysplants. Root growth in dry weight gain was significantly reduced bytreatment with 320 µg m–3 NH3 in Triticum aestivumand Zea mays growing with different N-forms, whereas leaf growthwas not significantly affected by NH3. In comparison with C3Triticum aestivum, non-fumigated C4 Zea mays had low ratiosof OH/N in NO3–3-grown plants and of H+/N in NH4+- and NH 4NO3-grown plants. Utilization of NH3 from the atmospherereduced both the OHN ratios in NO3 -grown plantsand the H+/N ratio in NH4+ - and NH4NO3 -grown plants of bothspecies. Furthermore, Zea mays had higher ratios of (C-A)/Nin NH4+ - and NH4NO3-grown plants than Triticum aestivum. Thismeans that C4 Zea mays had synthesized more organic anion perunit increase in organic N than C3 Triticum aestivum plants.Within both species, different nitrogen sources altered theratios of (C-A)/N in the order: NH4NO3>NH4+>NO3.Fumigation with NH3 increased organic acid synthesis in NO3- and NH4+ - grown plants of Triticum aestivum, whereas it decreasedorganic acid synthesis in Zea mays plants under the same conditions.Furthermore, these differences in acid-base regulation betweenC3 Triticum aestivum and C4 Zea mays plants growing with differentnitrogen sources are discussed. Kouchi, H. and Higuchi, T. 1988. Carbon flow from nodulatedroots to the shoots of soybean {Glycine max L. Merr.) plants:An estimation of the contribution of current photosynthate toureides in the xylem stream.–J. exp. Bot. 39: 1015–1023. Well-nodulated, water-cultured soybean plants were allowed toassimilate 13CO2 at a constant specific activity for 10 h andthe 13C-labelling of total carbon and ureides in xylem sap wasinvestigated. Labelled carbon appeared very rapidly in the xylem stream. Percentageof labelled carbon (relative specific activity, RSA) in xylemsap was 18% at 2 h after the start of 13CO2 assimilation andreached 53% at the end of the 10 h assimilation. The amountof labelled carbon exported from nodulated roots to the shootsvia the xylem during the 10 h labelling period accounted for33% of total labelled carbon imported into the nodulated roots.Ureides (allantoin and allantoic acid) in xylem sap were stronglydependent on currently assimilated carbon. The RSA of ureidesin xylem sap had reached 83% at the end of the assimilationperiod. Labelled carbon in ureides accounted for 51% of totallabelled carbon returned from nodulated roots to the shootsvia the xylem during the 10 h assimilation period. A treatmentwith 20 mol m–3 nitrate in the culture medium for 2 ddecreased the ureide concentration in the xylem sap slightly,but greatly decreased the RSA of ureides. By comparing the data with the results of analysis of the xylemsap of nodule-detached plants, it was concluded that the majorityof labelled carbon exported to the xylem stream from noduleswas in ureide form. A considerable amount of carbon was alsoreturned from roots to shoots via the xylem stream but it wasmore dependent on (non-labelled) carbon reserved in the roottissues. We studied the responses of Xanthium occidentale (Bertol.) (cockleburor Noogoora burr), a noxious weed, to atmospheric CO2 enrichmentand nitrate-N concentrations in the root zone ranging from 0.5to 25 mM. CO2 enrichment (1500 cm3 m–3) increased dry-matterproduction to about the same extent (18 per cent) at all levelsof supplied N: most of the increment in dry matter was distributedequally between leaves and roots so that there was little effecton shoot-to-root dry-weight ratios. Growth was stimulated greatlyby N and plateaued at 12 mM supplied N. Shoot-to-root dry-weightand total N ratios increased with increasing N supply. CO2 enrichmenthad no effect on the total amount of N accumulated by plants,but increased the N-use efficiency of leaves. Enriched plantshad lower concentrations and quantities of N in their leavesthan controls, and therefore lower shoot-to-root total N ratios.Little free NO3 accumulated in organs of control or enrichedplants. NO3 was the major form of N in xylem sap fromdetopped plants at low supplied NO3-N, but amino N was equalin importance at high supplied NO3-N in control and enrichedplants. Concentrations of NO3 were lower in the xylemsap of CO2 enriched plants. It was concluded that the betterN-use efficiency of CO2 enriched plants could result in increasedgrowth of X. occidentale in regions of marginal soil fertilityas atmospheric levels of CO2 increase. Growth and N-accumulation rates in leaves, stolons and rootsof individual white clover plants were studied in three experimentsusing two methods. In a growth chamber experiment, the relativedifferences between tissues were found to be almost constantfor a wide range of clover plant sizes. The stolon dry matter(DM) production was 56% and the root DM production 40% of theDM production in leaves. The N yield of stolons was 30% whileN yield in roots was 34% of N yield in leaves. The effect ofN application on these relations was investigated in a glasshouseexperiment. Application of N reduced the root:shoot N ratiofrom 0.50 to 0.28, whereas the stolon+root:leaf N ratio (i.e.for abovevs.below cutting-height tissues) was only reduced from0.97 to 0.80. In a field trial with two contrasting N regimes,growth and N accumulation were measured on individual cloverplants. Dinitrogen fixation was estimated by15N isotope dilutionbased on analysis of leaves-only or by including stolons. Usingleaves-only did not affect the calculation of percentage ofclover N derived from N2fixation (% Ndfa) since the15N enrichmentwas found to be uniform in all parts of the clover. Single, seed-grown plants of ryegrass (Lolium perenne L. cv.Melle) were grown for 49 d from the early seedling stage ingrowth cabinets at a day/night temperature of 20/15 C, witha 12 h photoperiod, and a CO2 concentration of either 340 or680µI 1–1 CO2. Following complete acclimation tothe environmental regimes, leaf and whole plant CO2 effluxesand influxes were measured using infra-red gas analysis techniques.Elevated CO2 increased rates of photosynthesis of young, fullyexpanded leaves by 35–46% and of whole plants by morethan 50%. For both leaves and whole plants acclimation to 680µI–1 CO2 reduced rates of photosynthesis in bothCO2 regimes, compared with plants acclimated to 340µll–1. There was no significant effect of CO2 regime onrespiration rates of either leaves or whole plants, althoughleaves developed in elevated CO2 exhibited generally lower ratesthan those developed in 340µI I–1 CO2. Initially the seedling plants in elevated CO2 grew faster thantheir counterparts in 340µI I–1 CO2, but this effectquickly petered out and final plant weights differed by onlyc. 10%. Since the total area of expanded and unexpanded laminaewas unaffected by CO2 regime, specific leaf area was persistently13–40% lower in elevated CO2 while, similarly, root/shootratio was also reduced throughout the experiment. Elevated CO2reduced tissue nitrogen contents of expanded leaves, but hadno effect on the nitrogen contents of unexpanded leaves, sheathsor roots. The lack of a pronounced effect of elevated CO2 on plant growthwas primarily due to the fact that CO2 concentration did notinfluence tiller (branch) numbers. In the absence of an effecton tiller numbers, any possible weight increment was restrictedto the c. 2.5 leaves of each tiller. The reason for the lackof an effect on tillering is not known. Bot. 36: 1835-1841. White clover was grown at 20 and100 Pa p(CO2). The CO2 response of net photosynthesis and differentialuptake of 14CO2 and 12CO2 by leaves were measured at varioustemperatures and at various O2 and CO2 partial pressures andcompared with predictions from ribulose bisphosphate carboxylase/oxygenasekinetics. Discrepancies between the observed gas exchange characteristicsfor the leaves and those predicted from the enzyme kineticswere interpreted as being due to a CO2 concentrating mechanism.Plants grown at 20 Pa p(CO2) showed a higher affinity for CO2than plants grown at 100 Pa p(CO2) when measured at 10 ?C. Nodifference in affinity was found at 30 ?C. The postulated CO2concentrating effect was greater in plants grown at low CO2than in plants grown at high CO2 concentration and occurredonly at low temperature and low CO2 partial pressure. It issuggested that plants grown at the lower CO2 partial pressurehave a higher affinity for CO2 due to a more efficient CO2 concentratingsystem than plants grown at the higher CO2 partial pressure. The effects of pruning shoot tops on the synthesis and degradationof ribulose 1,5–Wsphosphate carboxylase (RuBPCase) inleaves on remaining shoots were investigated in mulberry trees.Leucine labelled with 14C was fed to leaf discs from field-grownmulberry trees and 14C incorporation into RuBPCase was examined.Proportion of 14C in RuBPCase to leucine–14C absorbedby leaf discs was remarkably lowered by top-pruning, thoughoccasionally a slight increase was observed soon after pruning.Yet RuBPCase content in leaves on top-pruned shoots became progressivelyhigher than that in leaves on intact shoots. Changes in 14Cin Ru1BPCase in leaves of mulberry saplings previously fed 14CO2were followed. Following 14CO2 feeding, the attainment of themaximal level of 14C in RuBPCase was retarded by top-pruning.The highest level of 14C in RuBPCase was maintained in leaveson top-pruned shoots but decreased in leaves on intact shoots.Specific radioactivity in RuBPCase continued to increase inleaves on top-pruned shoots even after attaining a maximum levelin the control leaves. These facts suggest that the increasein RuBPCase by top-pruning results from a cessation of its degradationfor the remobilization of nitrogen for newly developing leaveson shoot tops. We investigated the role of intracellular Mg2+(Mgi2+) on the ATP regulation ofNa+/Ca2+ exchanger in squid axons and bovineheart. In squid axons and nerve vesicles, the ATP-upregulated exchangerremains activated after removal of cytoplasmic Mg2+, evenin the absence of ATP. Rapid and complete deactivation of theATP-stimulated exchange occurs upon readmission ofMgi2+. At constant ATP concentration, the effectof intracellular Mg2+ concentration([Mg2+]i) on the ATP regulation of exchangeris biphasic: activation at low [Mg2+]i,followed by deactivation as [Mg2+]i isincreased. No correlation was found between the above results and thelevels of phosphatidylinositol 4-phosphate and phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P2] measured innerve membrane vesicles. Incorporation ofPtdIns(4,5)P2 into membrane vesicles activates Na+/Ca2+ exchange in mammalian heart but not insquid nerve. Moreover, an exogenous phosphatase prevents MgATPactivation in squid nerves but not in mammalian heart. It is concludedthat 1) Mgi2+ is an essentialcofactor for the deactivation part of ATP regulation of the exchangerand 2) the metabolic pathway of ATP upregulation of theNa+/Ca2+ exchanger is different in mammalianheart and squid nerves.

Cucumber (Cucumis sativus L.) seedlings were grafted onto cucumber-(CG) or figleaf gourd- (FG, Cucurbita ficifolia Bouché)seedlings in order to determine the effect of solution temperature(12, 22, and 32°C) on the mineral composition of xylem sapand the plasma membrane K+-Mg++-ATPase activities of the roots.Low solution temperature (12°C) lowered the concentrationof NO3 and H2PO4 in xylem sap of CG plants butnot of FG plants. Concentrations of K+, Ca++ and Mg++ in xylemsap were less affected than anions by solution temperature.The plasma membrane of FG plants grown in 12°C solutiontemperature showed the highest K+- Mg++-ATPase activity at allATP concentrations up to 3 mM and at low reaction temperatureup to 12°C, indicating resistance of figleaf gourd to lowroot temperature.
