1. Phytophagous insects frequently manipulate their host‐plant to improve their immediate environment. This generally implies substantial modifications of host metabolism, and sometimes an alteration of nitrogen allocation within the host‐plant. However, the outcome of plant manipulation on amino acid or protein content can be modulated by environmental factors and host‐plant traits. 2. It was investigated whether the pseudogall induced by the aphid Phloeomyzus passerinii (Signoret) (Aphididae: Phloeomyzinae) in the bark of its host‐plant affects the amino acid content in bark tissues, and whether the strength of the modification is modulated by the fertilisation regime and/or the resistance level of the host‐plant. The development of aphid colonies on a resistant and a susceptible poplar genotype, under three fertilisation regimes, was studied. After the development of colonies, the free and protein‐bound amino acid content of the infested bark were quantified. 3. Fertilisation enhanced poplar growth and increased the free amino acid content of bark tissues. Infestation also triggered accumulations of both free and protein‐bound amino acids in the feeding sites, but in the susceptible genotype only. The increase in amino acid content was more pronounced when fertilisation was low, and fertilisation did not enhance aphid development. 4. In conclusion, infestation by P. passerinii triggers an accumulation of amino acids, but the effect is influenced by both the fertilisation regime and the resistance level of the host‐plant. This suggests that P. passerinii could affect the allocation of nutrients within trees during outbreaks.  相似文献

  1. The woolly poplar aphid Phloeomyzus passerinii Signoret (Aphididae: Phloeomyzinae) is a major pest of poplar plantations. We hypothesized that recent temperature increases may have contributed to the emergence and subsequent northward expansion of outbreaks in France.
  2. We reared P. passerinii at four temperatures to estimate its thermal requirements. We used experimental data to parametrize a mechanistic and temperature-driven physiologically-based demographic model. The model was used to simulate the effect of temperature on aphid dynamics and to assess the role of climate warming on the spatiotemporal dynamics of outbreaks.
  3. The lower developmental threshold was estimated at 6.4 °C and the development degree-days at 171.
  4. Our model supports the hypothesis that recent warming may have promoted outbreaks in northern France. During recent exceptionally warm years, more than 70% of the northern poplar area was favourable for outbreaks.
  5. Our model suggests that climate warming is not the sole factor involved. The dominance of resistant poplar genotypes such as ‘Robusta’ or ‘Beaupré’ may have preserved plantations from outbreaks before 1996 in southern France and until 2000 in the central part. Other factors, including biological characteristics, biotic interactions, or precipitation should be investigated.

Abstract

The poplars (Populus spp.) are the most sensitive plants to water deficit conditions among the woody species utilized for biomass production for energetic purposes; their productivity is associated with water availability in the soil. In the Mediterranean environment, crops are mainly limited by evapotranspirative demand that is not balanced by rainfall supply. As new hybrids with high growth rates and resistance to water stress are selected, the use of poplar as an energy crop may increase in Southern regions of Mediterranean Europe. The growth dynamics and physiological characteristics of poplar hybrid genotypes have been monitored for 2 years at a site with a Mediterranean climate, Apulia region, that could be used for energy crops. Unrooted cuttings of three recently selected genotypes of poplar (‘Neva’, ‘Dvina’ and ‘Lena’) and two “traditional” genotypes (‘Luisa Avanzo’ and ‘Bellini’) were planted in the spring of 2010 at two different densities: (a) low plant density = 1,667 cuttings ha?1 (LPD); (b) high plant density = 6,667 cuttings ha?1 (HPD). The genotypes ‘Lena’ and ‘Dvina’ showed the lowest survival rates and the poorest growth among the hybrid poplar tested. The genotype ‘Bellini’ had low stomatal sensitivity to soil water content and a moderate productive performance. The genotypes ‘Luisa Avanzo’ and ‘Neva’ had a good degree of rooting and sprouting, high values of leaf relative water content (RWCl) and low values of stomatal conductance (g s) during the summer months. In “Neva”, these characteristics were associated with the best yields (4 t ha?1) in HPD.  相似文献

The genus Populus represents one of the most economically important groups of forest trees. It is composed by approximately 30 species used for wood and non-wood products, phytoremediation and biomass. Poplar is subjected to several biological and environmental threats although, compared to annual crops, we know far less about the genetic bases of biotic stress resistance. Woolly poplar aphid (Phloeomyzus passerinii) is considered a main pest of cultivated poplars in European and American countries. In this work we present two high density linkage maps in poplar obtained by a genotyping by sequencing (GBS) approach and the identification of QTLs involved in Ph. passerinii resistance. A total of 5,667 polymorphic markers (5,606 SNPs and 61 SSRs) identified on expressed sequences have been used to genotype 131 plants of an F1 population P ×canadensis obtained by an interspecific mate between Populus deltoides (resistant to woolly poplar aphid) and Populus nigra (susceptible to woolly poplar aphid). The two linkage maps, obtained following the two-way pseudo-testcross mapping strategy, have been used to investigate the genetic bases of woolly poplar aphid resistance. One major QTL and two QTLs with minor effects (mapped on LGV, LGXVI and LG XIX) explaining the 65.8% of the genetic variance observed in the progeny in response to Ph. passerinii attack were found. The high density coverage of functional markers allowed the identification of three genes belonging to disease resistance pathway as putative candidates for P. deltoides resistance to woolly poplar aphid. This work is the first report on genetic of woolly poplar aphid genetic resistance and the resistant loci associated markers identified represent a valuable tool in resistance poplar breeding programs.  相似文献

Abstract

Metal-based defense against biotic stress is an attractive hypothesis on evolutionary advantages of plant metal hyperaccumulation. Metals may compensate for a defect in biotic stress signaling in hyperaccumulators (metal-therapy) by either or both direct toxicity to pathogens and by metal-induced alternative signaling pathways. Jasmonic acid (JA) and salicylic acid (SA) are well-established components of stress signaling pathways. However, few studies evaluate the influence of metals on endogenous concentrations of these defense-related hormones. Even less data are available for metal hyperaccumulators. To further test the metal-therapy hypothesis we analyzed endogenous SA and JA concentrations in Noccaea praecox, a cadmium (Cd) hyperaccumulator. Plants treated or not with Cd, were exposed to mechanical wounding, expected to enhance JA signaling, and/or to infection by biotrophic fungus Erysiphe cruciferarum for triggering SA. JA and SA were analyzed in leaf extracts using LC–ESI(?)–MS/MS. Plants without Cd were more susceptible to fungal attack than plants receiving Cd. Cadmium alone tended to increase leaf SA but not JA. Either or both fungal attack and mechanical wounding decreased SA levels and enhanced JA in the Cd-rich leaves of plants exposed to Cd. High leaf Cd in N. praecox seems to hamper biotic-stress-induced SA, while triggering JA signaling in response to fungal attack and wounding. To the best of our knowledge, this is the first report on the endogenous JA and SA levels in a Cd-hyperaccumulator exposed to different biotic and abiotic stresses. Our results support the view of a defect in SA stress signaling in Cd hyperaccumulating N. praecox.  相似文献

Abstract

Populus species are valued for their fast growth and are cultivated widely. Many of the commonly used species and hybrids are, however, regarded as drought sensitive, which poses a problem for large-scale cultivation particularly in light of climate change-induced drought spells in areas of poplar growth. While many hundreds of drought-induced genes have been identified in Populus species, very little is known about the genes and the signaling process that leads to a drought response in these species. Based on sequence similarity, the poplar G059200 gene is a potential ortholog of AtPP2CA, an inhibitor of drought and abscisic acid (ABA) responses in Arabidopsis thaliana. To test if G059200 has a similar function, we generated transgenic A. thaliana plants overexpressing this gene. These transgenic lines exhibited reduced responses to exogenous ABA and reduced tolerance to osmotic stress. Finally, drought tolerance of plants was also significantly reduced. Taken together, these data provide evidences that G059200 acts as a negative regulator of ABA responses. The ability to negatively regulate drought stress responses suggests that G059200 may be targeted for drought tolerance breeding, for example, by identification of individuals harboring natural or induced loss-of-function alleles, or by RNA interference technology, to generate poplar plants with reduced activity of G059200.  相似文献

Abstract

The soybean aphid [Aphis glycines Matsumura (Hemiptera: Aphididae)] is one of the most important insect pests of soybean [Glycine max (L.) Merr] in the northern USA and southern Canada, and four resistance loci (Rag1rag4) have been discovered since the pest was identified in the USA in 2000. The objective of this research was to determine whether resistance expression in recently identified soybean aphid-resistant plant introductions (PIs) was associated with the four Rag loci using a collection of 21 F2 populations. The F2 populations were phenotyped with soybean aphid biotype 1, which is avirulent on plants having any of the currently identified Rag genes, using choice tests in the greenhouse and were tested with genetic markers linked to the four Rag loci. The phenotyping results indicate that soybean aphid resistance is controlled by a single dominant gene in 14 PIs, by two genes in three PIs, and four PIs had no clear Mendelian inheritance patterns. Genetic markers flanking Rag2 were significantly associated with aphid resistance in 20 PIs, the Rag1 region was significantly identified in five PIs, and the Rag3 region was identified in one PI. These results show that single dominant gene action at the Rag2 region may be a major source for aphid resistance in the USDA soybean germplasm collection.  相似文献

Background and aims

The quantification of root dynamics remains a major challenge in ecological research because root sampling is laborious and prone to error due to unavoidable disturbance of the delicate soil-root interface. The objective of the present study was to quantify the distribution of the biomass and turnover of roots of poplars (Populus) and associated understory vegetation during the second growing season of a high-density short rotation coppice culture.

Methods

Roots were manually picked from soil samples collected with a soil core from narrow (75 cm apart) and wide rows (150 cm apart) of the double-row planting system from two genetically contrasting poplar genotypes. Several methods of estimating root production and turnover were compared.

Results

Poplar fine root biomass was higher in the narrow rows than in the wide rows. In spite of genetic differences in above-ground biomass, annual fine root productivity was similar for both genotypes (ca. 44 g DM m?2 year?1). Weed root biomass was equally distributed over the ground surface, and root productivity was more than two times higher compared to poplar fine roots (ca. 109 g DM m?2 year?1).

Conclusions

Early in SRC plantation development, weeds result in significant root competition to the crop tree poplars, but may confer certain ecosystem services such as carbon input to soil and retention of available soil N until the trees fully occupy the site.  相似文献

This is the first report on genetic analysis and genome mapping of major dominant genes for near non-host resistance to barley crown rust ( Puccinia coronata var. hordei ) in common wheat.

Abstract

Barley crown rust, caused by Puccinia coronata var. hordei, primarily occurs on barley (Hordeum vulgare L.) in the Great Plain regions of the United States. However, a few genotypes of common wheat (Triticum aestivum L.) were susceptible to this pathogen among 750 wheat accessions evaluated. To investigate the genetics of crown rust resistance in wheat, a susceptible winter wheat accession PI 350005 was used in crosses with two resistant wheat varieties, Chinese Spring and Chris. Analysis of F1 plants and F2 populations from these two crosses indicated that crown rust resistance is controlled by one and two dominant genes in Chris and Chinese Spring, respectively. To determine the chromosome location of the resistance gene Cr1 in Chris, a set of 21 monosomic lines derived from Chris was used as female parents to cross with a susceptible spring type selection (SSTS35) derived from the PI 350005/Chris cross. Monosomic analysis indicated that Cr1 is located on chromosome 5D in Chris and one of the crown rust resistance genes is located on chromosome 2D in Chinese Spring. The other gene in Chinese Spring is not on 5D and thus is different from Cr1. Molecular linkage analysis and QTL mapping using a population of 136 doubled haploid lines derived from Chris/PI 350005 further positioned Cr1 between SSR markers Xwmc41-2 and Xgdm63 located on the long arm of chromosome 5D. Our study suggests that near non-host resistance to crown rust in these different common wheat genotypes is simply inherited.  相似文献

Abstract

In the present study, uptake, susceptibility and metabolism of SO2 were analyzed in the deciduous tree species poplar (Populus × canescens). A particular focus was on the significance of sulfite oxidase (SO) for sulfite detoxification, as SO has been characterized as a safety valve for SO2 detoxification in herbaceous plants. For this purpose, poplar plants were exposed to different levels of SO2 (0.65, 0.8, 1.0, 1.2 μL L?1) and were characterized by visible injuries and at the physiological level. Gas exchange parameters (stomatal conductance for water vapor, CO2 assimilation, SO2 uptake) of the shoots were compared with metabolite levels (sulfate, thiols) and enzyme activities [SO, adenosine 5′-phosphosulfate reductase (APR)] in expanding leaves (80–90 % expanded). The critical dosage of SO2 that confers injury to the leaves was 1.2 μL L?1 SO2. The observed increase in sulfur containing compounds (sulfate and thiols) in the expanding leaves strongly correlated with total SO2 uptake of the plant shoot, whereas SO2 uptake rate was strongly correlated with stomatal conductance for water vapor. Furthermore, exposure to high concentration of SO2 revealed channeling of sulfite through assimilatory sulfate reduction that contributes in addition to SO-mediated sulfite oxidation to sulfite detoxification in expanding leaves of this woody plant species.  相似文献

Abstract

Energy allocation between growth and defense is elegantly balanced to achieve optimal development in plants. Brassinosteroids (BRs), steroidal hormones essential for plant growth, are regulated by other plant hormones, including auxin and jasmonates (JA); auxin stimulates the expression of a key brassinosteroid (BR) biosynthetic gene, DWARF4 (DWF4), whereas JA represses it. To better understand the interaction mechanisms between growth and defense, we isolated a fast-growing mutant, gulliver3-D (gul3-D), that resulted from the activation tagging of DWF4, and examined the response of this mutant to defense signals, including JA, Pseudomonas syringae pv. tomato (Pst DC3000) infection, and wounding. The degree of root growth inhibition following MeJA treatment was significantly decreased in gul3-1D/dwf4-5D relative to the wild type, suggesting that JA signaling is partially desensitized in gul3-1D. Quantitative RT-PCR analysis of the genes involved in JA and salicylic acid (SA) responses, including MYC2, PDF1.2, CORI3, PR1, and PR2, revealed that JA signaling was preferentially activated in gul3-1D, whereas SA signaling was suppressed. As a result, gul3-1D was more susceptible to a biotrophic pathogen, Pst DC3000. Based on our results, we propose a model in which BR and JA cooperate to balance energy allocation between growth and defense responses. In ambient conditions, BRs promote plant growth; however, when stresses trigger JA signaling, JA compromises BR signaling by downregulating DWF4 expression.  相似文献

Abstract

Non-host interactions between Puccinia striiformis f. sp. tritici (Pst) and two rice subspecies were characterized using 23 rice varieties, including 11 japonica and 12 indica. Results showed that the infected fungal structures were easily produced in the leaves of indica, whereas only several substomatal vesicles and primary infection hyphae were observed in the leaves of japonica. This result indicated that indica is less resistant or more susceptible to Pst than japonica. Hydrogen peroxide accumulated in the initial phase of japonicaPst interaction but not in indicaPst interaction. A set of reactive oxygen species (ROS)-related genes was also induced in response to Pst infection, suggesting that ROS activation is one of the major mechanisms of non-host resistance of rice to Pst.  相似文献

Abstract

Protoplast regeneration is essential for somatic hybridizations. In this study, a standard method for plantlet regeneration from Cichorium protoplasts was developed. We evaluated the effect of the low melting point agarose (LMPA) bead technique on the regeneration capacity of protoplasts of seven C. intybus and four C. endivia genotypes. The LMPA bead technique was more efficient than culture in liquid or solid medium and allowed us to obtain plating efficiencies up to 4.9?% in C. intybus genotypes and efficiencies of up to 0.7?% in C. endivia genotypes. Moreover, the LMPA bead technique offers great advantages over liquid and solid culture systems: the media can be readily refreshed, protoplasts can be monitored separately, and microcalli can easily be removed from the beads. This increased efficiency was observed for all of the 11 Cichorium genotypes tested. Shoot formation was induced more efficiently when using 0.5?mg?l?1 indole-3-acetic acid-enriched medium (up to 87.5?% of the protoplast-derived calli started shoot development) compared to 1-naphthaleneacetic acid-enriched medium. The LMPA bead technique optimized in this study enabled for the first time the full plantlet regeneration from protoplasts of C. endivia genotypes and increased the protoplast regenerating ability in other Cichorium species. This fine-tuned LMPA bead technique can therefore be applied for protoplast regeneration after protoplast fusions of the genus Cichorium.  相似文献
