A gel bait-resistant German cockroach, Blattella germanica (L.), strain Cincy was collected in Cincinnati, OH. This strain exhibited a high level of behavioral resistance to Avert (0.05% abamectin) and Maxforce FC (0.01% fipronil) gel baits. Topical application assays indicated moderate levels of physiological resistance of the Cincy strain to abamectin and fipronil. Resistance ratios (based on LD50 values from topical applications) to abamectin and fipronil were 2.5 and 8.7, respectively. The Cincy strain of had a significantly lower LD50 value to abamectin than a nonaverse field strain (Dorie) and similar LD50 values to fipronil as the Dorie strain. The aversion behavior (avoidance of gel baits) was therefore caused by food ingredients in the gel baits. The Cincy strain showed avoidance of agar containing fructose, glucose, maltose, and sucrose, which are phagostimulants to the laboratory strain. Modifications of the inert ingredients in the Maxforce FC gel bait significantly improved the efficacy against the Cincy strain. The Cincy strain produced significantly smaller oothecae and lower numbers of eggs in each egg capsule than the nonaverse Jwax and Dorie strains of cockroaches, suggesting fitness costs are associated with resistance.  相似文献

Abamectin was fed to German cockroaches, Blattella germanica (L.), in non-choice tests. LT50s and LC50s were estimated by probit analysis. The LT50s for the German cockroach ranged from 4.4 to 1.7 d for males, from 9.0 to 2.4 d for females, and from 4.4 to 1.6 d for nymphs for bait concentrations of abamectin between 0.0025 and 0.0500%. The LC50s of abamectin were 0.0110 and 0.0040% from males, 0.0240 and 0.0090% for females, and 0.0200 and 0.0080% for nymphs at 3 and 6 d, respectively. The LT50 values of 0.0550% abamectin bait were 3.4, 3.4, 2.4, 7.5, 2.9, and 4.5 d for Periplaneta americana (L.), P. fuliginosa (Serville), P. brunnea Burmeister, P. australasiae (F.), Blatta orientalis L., and Supella longipalpa (Serville). Although the bait was effective against various cockroach species, time to death for the larger species was longer than for the German cockroach. In preference tests in which male German cockroaches were allowed to feed on rat chow or abamectin bait, all died within 5 d of exposure to abamectin bait. Abamectin bait consumption was not significantly lower than that of untreated rat chow. Arena tests with 0.0550% abamectin bait resulted in 31-75% mortality of German cockroaches after 9 d, with most control being achieved by treating harborages with the bait. The hydramethylnon standard resulted in 65% mortality after 9 d.  相似文献

This study investigated the pharmacological profile of cyclodiene resistance in Drosophila melanogaster and the mode of action of a phenylpyrazole insecticide, JKU 0422. Toxicological studies were performed with a sucrose bait assay containing the synergist piperonyl butoxide. The Maryland strain of D. melanogaster was resistant to dieldrin, lindane, picrotoxinin, TBPS, p-CN-TBOB, and JKU 0422. In contrast, this strain was susceptible to cypermethrin and the avermectins MK-243, abamectin, and abamectin 8,9-oxide. Neurophysiological studies showed that both TBPS and JKU 0422 reversed the inhibitory action of GABA in central nerve preparations from susceptible D. melanogaster. However, the response to these compounds was attenuated in nerve preparations from the resistant Maryland strain, which indicated that the resistance was expressed at the level of the nerve. Topical toxicity bioassays with JKU 0422 on susceptible (CSMA) and cyclodiene-resistant (LPP) strains of German cockroach revealed a resistance ratio of 553-fold for this compound. These studies demonstrate that cyclodiene resistance in D. melanogaster confers broad cross resistance toward compounds thought to block the GABA-gated chloride channel in a manner similar to the cyclodienes. Moreover, the cross resistance extends to JKU 0422, and resistance to this compound is also present in a strain of cyclodiene-resistant German cockroach. These toxicological results, along with the neurophysiological studies, confirm that JKU 0422 has a mode of action that is similar to the cyclodienes and TBPS. These findings suggest that the introduction and use of new chloride channel antagonists as insecticides should be managed carefully in order to prevent the rapid development of resistance in the field. © 1994 Wiley-Liss, Inc.  相似文献

Experimental indoxacarb powder and gel baits were evaluated in the laboratory, and a gel bait was evaluated in subsequent field studies against the German cockroach, Blattella germanica (L.). In continuous exposure tests, LT50 values were 1.90 and 1.10 d for 0.25 and 1% indoxacarb powder baits, respectively. However, 0.25% indoxacarb gel bait had an LT50 value of 0.68 d, similar to a 0.05% abamectin gel bait formulated with the same bait base. There was no difference in toxicity between fresh and 7-d-old gel bait deposits. A pyrethroid-resistant strain of German cockroaches was significantly resistant to both abamectin and indoxacarb gel baits. Gel bait contained approximately 40% water, desiccated rapidly at 25-28 degrees C and 30-45% RH, but did not rehydrate when held at 56.7% RH for 3 d. Powder indoxacarb baits contained <1% water and did not desiccate or gain water. Indoxacarb gel bait (0.25%) was relatively nonrepellent (approximately 30%) and had positive maximum performance index values (approximately 100) in Ebeling choice box experiments. In field experiments in cockroach-infested kitchens, the 0.25% indoxacarb gel bait significantly reduced visual counts of German cockroaches approximately 74% at 3 d and >95% at 14 d. Indoxacarb baits are toxic, relatively nonrepellent, and can significantly reduce German cockroach populations.  相似文献

Strain differences in the response of German cockroaches to toxic baits and their potential to evolve resistance to them was evaluated in choice tests. Percentage bait consumption (amount of bait consumed divided by total food consumed) was used to estimate relative attractancy/repellency of the baits. One strain apparently had low level resistance to abamectin, but no evidence of resistance was found in other strains tested with either Roach Ender or a gel bait (abamectin-based baits). Incipient behavioral resistance occurred in two strains selected for three generations with Roach Ender; a stronger response in a strain selected with the gel bait is attributed to the development of behavioral resistance. Baygon bait was highly repellent. Low level resistance to propoxur caused a large decrease in mortality compared with susceptible strains. Strong behavioral resistance developed in strains selected with Stapleton's Magnetic Roach Food.  相似文献

Several insecticide bait formulations were evaluated for their attractiveness to cockroaches in olfactometer assays in the laboratory and in trapping experiments in the field. Included in the assays were bait stations, gels, pastes, and a powder that contained one of the following active ingredients: abamectin, boric acid, chlorpyrifos, or hydramethylnon. There were significant differences among the baits in their attractiveness to the German cockroach, Blattella germanica (L.). In trapping experiments, Avert powder (abamectin), Maxforce station and gel, and Siege gel (all hydramethylnon) were consistently attractive to B. germanica adults and nymphs. Laboratory olfactometer assays with adult males confirmed these results and showed that nymphs were as responsive as males whereas females were less responsive. Our bioassays also demonstrate that attractiveness of bait can be dramatically affected by the age of the bait. One week of aging significantly reduced the attractiveness of Avert powder in both laboratory and field assays. Aging, however did not diminish the attractiveness of Maxforce gel, indicating that the formulation may be critical for retention of attractiveness of baits. Baits that were most attractive to the German cockroach were also the most attractive to nymphs and adults of the brownbanded cockroach, Supella longipalpa (F.).  相似文献

Abamectin resistance was selected in the vegetable leafminer, Liriomyza sativae (Blanchard) (Diptera: Agromyzidae) under laboratory conditions, and cross‐resistance patterns and possible resistance mechanisms in the abamectin‐resistant strains (AL‐R, AF‐R) were investigated. Compared with the susceptible strain (SS), strain AL‐R displayed 39‐fold resistance to abamectin after 20 selection cycles during 25 generations, and strain AF‐R exhibited 59‐fold resistance to abamectin after 16 selection cycles during 22 generations. No cross‐resistance to cyromazine was found in both abamectin‐resistant strains. However, we failed to select for cyromazine resistance in L. sativae under laboratory conditions by conducting 17 selection cycles during 22 generations. However, moderate levels of cross‐resistance to abamectin (6–9 fold) were observed in strains which received cyromazine treatments. Biochemical analysis showed that glutathione S‐transferase (GST) activity in both abamectin‐resistant strains (AL‐R, AF‐R) was significantly higher than in the susceptible strain (SS), suggesting metabolically driven resistance to abamectinin L. sativae. Recommendations of mixtures or rotation of cyromazine and abamectin should be considered carefully, as consecutive cyromazine treatments may select for low‐level cross‐resistance to abamectin.  相似文献

Female German cockroaches, Blattella germanica (L.) (Dictyoptera: Blattellidae) and nymphs (stages 1, 3 and 6) were used to test the attractiveness and the feeding stimulation of several commercial gel formulations of insecticide food baits in laboratory binary choice tests. The four toxic gel baits tested were: 'Avert' (0.05% abamectin=avermectin B1), 'Drax' (33.3% boric acid), 'Goliath' (0.05% fipronil) and 'Maxforce' (2.15% hydramethylnon). The attractiveness of a bait was estimated by the number of cockroaches that chose it first, and by their tendency to change to it secondarily. Feeding stimulation was estimated by the mean duration of feeding bouts and total duration of food intake. Variability of these factors was assessed in relation to age of cockroaches and to age of baits. With a few exceptions, cockroach age did not affect performance in any of these tests. Attractiveness to B. germanica ranked 'Goliath' gel higher than 'Avert', 'Drax' and 'Maxforce' gels, whether or not the active ingredient fipronil was present in 'Goliath' gel. Feeding stimulation ranked 'Avert' and 'Maxforce' gels higher than 'Goliath' gel, and 'Drax' gel was inferior. With ageing up to 3 months, the feeding stimulation power of 'Goliath' and 'Maxforce' gels increased, whereas that of 'Avert' fluctuated. Assuming that all types of bait effectively kill cockroaches once ingested, these results illustrate the importance of behavioural observations on bait efficiency and show that ranking of these different food baits varies in relation to the parameter analysed.  相似文献

Topical and oral toxicity of fipronil, compared to chlorpyrifos, was determined for the German cockroach, Blattella germanica (L.), and American cockroach, Periplaneta americana (L.). Fipronil and Combat bait matrices were evaluated for their attractancies to both species. In the topical toxicity tests, LD50's of fipronil, at 72 h after topical application, were 0.03 and 0.02 µg/g for adult B. germanica and P. americana, respectively. Fipronil was significantly more toxic than topically applied chlorpyrifos (LD50's were 0.06 and 0.16 µg/g for B. germanica and P. americana, respectively). The oral toxicity of fipronil and chlorpyrifos in Petri dish experiments, against both species, was affected by stage (for B. germanica), diet concentration, and feeding assay. Fipronil caused higher mortality of B. germanica than chlorpyrifos in two feeding assays (continuous and abbreviated). Both compounds were equally toxic to adult males of P. americana at all rates. Fipronil caused higher nymphal mortality than chlorpyrifos 48–72 h after exposure in both feeding assays. In large population chamber tests, fipronil bait was more effective and faster in killing P. americana than Raid and Combat. LT50's were 0.8, 2.4, and 7.6 d for fipronil, Raid (a.i. = chlorpyrifos), and Combat (a.i. = hydramethylnon) baits, respectively. Mortality reached 96.5, 93.4, and 84.6%, respectively, at the end of the 14 d test. In the bait attractancy tests, both strains of B. germanica were attracted similarly to fipronil and Combat bait matrices. P. americana were attracted more to fipronil than to Combat bait matrix or to other alternative foods.  相似文献

In this study, random amplified polymorphic DNA polymerase chain reaction (RAPD-PCR) was used to identify polymorphic genomic DNA that would discriminate among cyromazine-resistant, abamectin-resistant, and susceptible Liriomyza trifolii (Burgess) (Diptera: Agromyzidae) leafminers. Using a reference strain that was susceptible to both cyromazine and abamectin, and a cyromazine-resistant strain and an abamectin-resistant strain, 400 oligonucleotides were assayed using RAPD-PCR. We found that two oligonucleotides, B10 and G16, amplified unique bands in the cyromazine-resistant strain but not in the reference or abamectin-resistant strains. Three oligonucleotides, K04, J13, and I02, showed polymorphisms unique to the abamectin-resistant strain but not in the reference or cyromazine-resistant strain. Leaf dip bioassays and RAPD-PCR were performed on two additional reference strains, seven strains from commercial ornamental production greenhouses, and one field strain. The two reference strains were negative for the resistance-correlated oligonucleotides. Of the seven strains from ornamental greenhouses, leaf dip bioassays showed that five had some level of resistance to both abamectin and cyromazine, whereas two were susceptible. The field strain was susceptible to both cyromazine and abamectin. In RAPD-DNA analyses, the five strains with abamectin resistance were positive for the three abamectin resistance-correlated oligonucleotides K04, J13, and I02. In the cases of cyromazine resistance, the five strains with cyromazine resistance were positive for the two cyromazine resistance-correlated oligonucleotides B10 and G16. The field strain and two greenhouse strains that were susceptible in leaf dip bioassays were negative for all three abamectin resistance-correlated oligonucleotides. The field strain and one greenhouse strain were negative for the two cyromazine resistance-correlated oligonucleotides; however, one greenhouse strain that was susceptible to cyromazine in leaf dip bioassay tested positive for one of the cyromazine resistance-correlated oligonucleotides. This method can be used to quickly identify cyromazine resistance, abamectin resistance, or both in leafminers, enabling a grower to choose an effective insecticide for leafminer control in a timely manner.  相似文献

Ovicidal activity of the benzoylphenylurea noviflumuron was evaluated in the laboratory on three adult groups (virgin females, virgin males, and fertilized, nongravid females) of the German cockroach, Blattella germanica (L.), through ingestion of treated bait. Novifumuron caused significant ovicidal effects at concentrations ranging from 10 to 5000 ppm after 5-d feeding exposure to virgin and fertilized females. Untreated females produced little or no viable oothecae when mated with virgin males that had previously ingested bait (5-d exposure) with 1000 ppm or 5000 ppm noviflumuron. The highest tested concentration of noviflumuron (5000 ppm) caused 100% ovicidal activity through two ovarian cycles for all three adult groups. Noviflumuron seems to have broader ovicidal activity against B. germanica than reported for other benzoylphenylurea insecticides and can potentially impact cockroach populations through a combination of nymphal mortality and ovicidal activity.  相似文献

Consumption was measured of three commercially available cockroach gel baits (0.01 and 0.05% fipronil and 0.6% indoxacarb) by two pest blattellid (German cockroach, Blattella germanica L., and brownbanded cockroach, Supella longipalpa Fabricius) and three pest blattid [oriental cockroach, Blatta orientalis L., American cockroach, Periplaneta americana L., and smokybrown cockroach, Periplaneta fuliginosa (Serville)] species (Dictyoptera), and direct and secondary effects were quantified. All three baits were greatly preferred for consumption over dog food; however, virtually all consumption (ca. 98%) by pest blattids was gel baits containing sugar feeding stimulants and water. Pest blattid greater preference for gel baits was probably due to their greater need for nutrients in baits due to their greater cuticular water permeability and higher metabolism than the pest blattellids. Brownbanded cockroaches had lowest percentage gel bait selection. Pest blattellids consumed greater amounts of bait per g body weight than pest blattids. Cockroaches consumed more active ingredient than needed to cause mortality; however, based on bait consumption, a 30‐g tube of gel bait potentially killed from 394 to 6 966 adult cockroaches, depending on species. Mortality for all cockroach species was faster for adults (≥3 days) than for nymphs (≥7 days); however, most brownbanded cockroaches exposed to indoxacarb survived despite consuming 1.5‐ to >3‐fold more than other baits, suggesting low enzyme production by brownbanded cockroaches and consequently lower conversion of indoxacarb into its toxic form. Besides direct mortality, German cockroaches died from indirect effects: exposure to debris from other cockroaches that had direct access to the gel baits or bait contact without ingestion. Although maximization of bait consumption is important, factors that enhance secondary mortality and contact toxicity should also be considered.  相似文献

Abstract:  To understand the risk of resistance and the possible mechanisms of resistance to abamectin in B-type Bemisia tabaci (Gennadius) better, a resistant strain of B. tabaci was selected in the laboratory and cross-resistance pattern and resistance mechanisms to abamectin were investigated. The NJ-Abm strain of B. tabaci was derived from a field population (NJ) collected in Nanjing, China in 2002 with 18 generations of selection with abamectin in the laboratory. Compared with the unselected NJ strain, the selected NJ-Abm strain developed 14.5-fold resistance to abamectin and showed significant cross-resistance to emamectin benzoate (4.4-fold) and imidacloprid (3.4-fold), but no cross-resistance to fipronil. The oxidase inhibitor piperonyl butoxide (PBO) and glutathione S -transferase inhibitor diethyl maleate (DEM) produced significant synergism on abamectin in the NJ-Abm strain (with synergistic ratios of 3.9- and 4.1-fold respectively); however, the esterase inhibitor triphenyl phosphate (TPP) did not act synergistically with abamectin. Biochemical analysis confirmed that P450 monooxygenase activity and glutathione S -transferase activity of the NJ-Abm strain were elevated to 2.1- and 2.0-fold, respectively, compared with that of the NJ strain. This indicated that enhanced metabolism mediated by P450 monooxygenase and glutathione S -transferase is likely to be involved in abamectin resistance and cross-resistance to imidacloprid and emamectin benzoate in the NJ-Abm strain.  相似文献

The two-spotted spider mite, Tetranychus urticae, is one of the most destructive pests of various orchard trees and garden plants. Biochemical mechanisms of abamectin resistance in two T. urticae strains (PTF, 239-fold resistance; AbaR, ca. 4753-fold resistance) were investigated. The involvement of both esterase (Est) and mixed function oxidase (MFO) in abamectin resistance was suggested by synergistic bioassays, in which median lethal time (LT50) values were significantly reduced by pretreatment with triphenyl phosphate (TPP) and piperonyl butoxide, respectively. Detoxification enzyme assays confirmed that Est and MFO were related to abamectin resistance as metabolic factors. Moreover, some Est bands on a native isoelectric focusing gel were specifically inhibited by TPP, implying their association with resistance. Pretreatment with verapamil in synergistic bioassays did not reduce the LT50 to abamectin, suggesting that the ABC transporter is not likely involved in resistance. However, enhanced MFO and Est activities in the AbaR strain were not enough to account for the extremely high level of abamectin resistance, which suggests the involvement of additional resistance mechanisms, such as target site insensitivity.  相似文献

Three populations of the leafminer, Liriomyza trifolii (Burgess), were collected from commercial ornamental production greenhouses in the United States and tested for susceptibility to three commercial insecticides. A leaf dip bioassay of leaves containing young (1-2-d-old) larvae was used. Based on larval mortality and compared with a susceptible laboratory reference colony, the three strains varied in spectrum and level of resistance to the insecticides. CA-1, collected from Gerbera daisy, was moderately resistant to cyromazine (18.1-fold) and abamectin (22.0-fold), but highly resistant to spinosad (> 188-fold). CA-2, collected from chrysanthemums, was not resistant to abamectin, had a low level of resistance to cyromazine (8.2-fold), but was extremely resistant to spinosad (1,192-fold). GA-1, collected from chrysanthemums, had very low levels of resistance to cyromazine (5.4-fold) and spinosad (1.9-fold) but was moderately resistant to abamectin (30.6-fold). When reared in the absence of insecticide selection pressure, all three strains reverted to approximately the level of the reference strain. The CA-1 strain reverted in nine generations to cyromazine; however, the lowest levels of abamectin and spinosad resistance reverted to was 3.1-fold at F8 and 3.2 at the F10, respectively. The CA-2 strain reverted in five generations to both cyromazine and spinosad. GA-1 reverted in five generations to abamectin. Based on the results, resistance to these three insecticides was unstable. Additionally, there was no cross-resistance among these three insecticides.  相似文献

Fipronil, a phenylpyrazole insecticide, was made available in 1999 in bait formulations for use against the German cockroach, Blattella germanica (L.). We have investigated resistance to fipronil in the descendants of cockroaches collected just before, or contemporaneously with, the introduction of fipronil baits. Cockroaches were obtained in two types of settings: homes that either had or had not been serviced by a pest management professional while occupied by their current residents. Thorough inspections by us turned up no evidence that fipronil had been used in any of the homes, and in addition, no residents claimed to have used baits containing fipronil. Resistance to fipronil was detected by topically dosing adult males with the LC99 of fipronil, the value of which was determined in a dose-response assay with males of an insecticide-susceptible strain. Fewer than 99 of 100 males of all field-collected strains died within 72 h of being treated. Moreover, substantial numbers of males survived doses three and 10-fold greater than the LC99. Regression analysis showed that 67% of the variation in the percentage of males that died after being treated with fipronil was explained by a linear relationship with the percentage that died after being treated with dieldrin. Therefore, it appears that resistance to fipronil in German cockroaches--whose ancestors had never been exposed to it--is attributable to enduring resistance to the cyclodienes, which were formerly used for cockroach control and have a similar mode of action as fipronil. Lastly, we found that insects resistant to topically administered fipronil were likewise resistant, and to a similar degree, to ingested fipronil.  相似文献

The objective of this research was to examine the role and type of behavioural mechanisms that function in house fly, Musca domestica L. (Diptera: Muscidae), resistance to an imidacloprid‐containing commercial fly bait, QuickBayt®, using an insecticide‐susceptible and an imidacloprid‐resistant strain. Mortality and feeding behaviour were observed through choice bioassays of three post‐imidacloprid selected house fly generations to determine whether flies would consume the bait in the presence of an alternative food source. Mortality rates in choice containers progressively decreased in post‐selection flies as QuickBayt® no‐choice selections proceeded. There were no differences between the proportions of flies observed contacting QuickBayt® and sugar, respectively, a finding that eliminates repellency as a mechanism of stimulus‐dependent behavioural resistance. However, differences in QuickBayt® consumption and subsequent mortality between choice and no‐choice containers provided strong support for the evolution of consumption irritancy‐ or taste aversion‐related behavioural resistance. The results of this study support the responsible rotation of insecticide bait formulations for house fly control.  相似文献

Blattella germanica (L.) that were fed hydramethylnon bait produced residues that were toxic to exposed conspecifics. Insecticidal activity was traced to the feces of treated insects by feeding radiolabeled material, where approximately 50% of the recovered radioactivity was unmetabolized parent compound. Ingestion of toxicant-laden feces by all life stages was evident, but the effect of this behavior was greatest on early instar nymphs. Baits containing toxicants with delayed activity, such as hydramethylnon, probably affect cockroach field populations indirectly through coprophagy.  相似文献

Diamondback moth (DBM), Plutella xylostella (L.) (Lepidoptera: Plutellidae), is one of the most destructive pests in Brassicaceae crops, such as Chinese cabbage (Brassica rapa L.). It is rapidly developing resistance to abamectin, the dominant insecticide utilized in controlling P. xylostella in China and other southeastern Asian countries. The target of abamectin, the alpha subunit of glutamate‐gated chloride channel (GluClα), is thought to be involved in the development of abamectin resistance in nematodes and insects. This study investigated variants of GluClα in both abamectin‐susceptible and resistant strains of P. xylostella. A comparison of the PxGluClα sequences revealed three variants, including a 63‐bp substitution, a 36‐bp deletion, and a 65‐bp insertion. The frequency of the 36‐bp deletion was much higher in the abamectin‐resistant strain compared to the susceptible strain, whereas the 63‐bp substitution and 65‐bp insertion showed no significant difference between the resistant and susceptible strains. The in vitro expression of PxGluClα (with or without the 36‐bp deletion) in Xenopus laevis (Daudin) oocytes indicated that PxGluClα with the 36‐bp deletion was less sensitive to both glutamate and abamectin compared to the wild‐type PxGluClα. These findings suggest that the variant 36‐bp deletion in PxGluClα may confer abamectin resistance in P. xylostella after continuous abamectin selection, providing new insights into the management of this pest and contributing to the development of new reagents for pest control.  相似文献

The death rates of three toxic gel baits (fipronil bait, 0.05%; hydramethylnon, bait 2.15%; and abamectin B1 bait, 0.05%) were estimated in Blattella germanica (L.). After ingestion, all three baits killed similar proportions of cockroaches, but they died more rapidly after ingestion of fipronil bait than after ingestion of abamectin B1 bait or of hydramethylnon bait. Laboratory experiments evaluated the impact of secondary transmission of fipronil bait and hydramethylnon bait. Mortality rates caused by secondary transmission were higher for fipronil bait than for hydramethylnon bait. Under controlled laboratory conditions, secondary transmission occurred mainly through direct contact with, or ingestion of, traces of baits dispersed in the environment by contaminated cockroaches. These traces were either deposited by "trampling" in the environment or on dead contaminated cockroaches. Social interactions and cannibalism played a minor role in secondary transmission of these two baits.  相似文献
