The present work investigates some probiotic properties of four different microorganisms (Bifidobacterium animalis var. lactis BB-12, Escherichia coli EMO, Lactobacillus casei and Saccharomyces boulardii). In vitro and in vivo tests were carried out to compare cell wall hydrophobicity, production of antagonistic substances, survival capacity in the gastrointestinal tract of germ-free mice without pathological consequence, and immune modulation by stimulation of Küpffer cells, intestinal sIgA and IL-10 levels. In vitro antagonism against pathogenic bacteria and yeast was only observed for the probiotic bacteria B. animalis and L. casei. The hydrophobic property of the cell wall was higher for B. animalis and E. coli EMO, and this property could be responsible for a better ability to colonize the gastrointestinal tract of germ-free mice. Higher levels of sIgA were observed mainly for S. boulardii, followed by E. coli EMO and B. animalis, and only S. boulardii induced a significant higher level of IL-10. In conclusion, for a probiotic use, S. boulardii presented better characteristics in terms of immunomodulation, and B. animalis and L. casei for antagonistic substance production. The knowledge of the different probiotic properties could be used to choice the better microorganism depending on the therapeutic or prophylactic application.  相似文献

For comparative study of carbohydrate chains of N-glycoproteins, method of "oligosaccharide maps" has been developed. It consists in fractionation of reduced oligosaccharide fragments by gel-chromatography and HPLC on reverse phase and amino columns. Using two HPLC retention time values for each oligosaccharide, two-dimensional maps for both variants of H1 hemagglutinin were constructed. The monosaccharide composition of the majority of oligosaccharides isolated was also elucidated. The carbohydrate chain's patterns for the H1 hemagglutinin variants were found to be similar but to differ considerably from those for H3 hemagglutinin. The data obtained show that the glycosylation pattern depends on virus strain, i.e. on the structure of the polypeptide chain of hemagglutinin.  相似文献

The ATP/ADP exchange is shown to be a partial reaction of the (H+ + K+)-ATPase by the absence of measurable nucleoside diphosphokinase activity and the insensitivity of the reaction to P1, P5 -di(adenosine-5′) pentaphosphate, a myokinase inhibitor. The exchange demonstrates an absolute requirement for Mg2+ and is optimal at an ADP/ATP ratio of 2. The high ATP concentration (K0.5 = 116 μM) required for maximal exchange is interpreted as evidence for the involvement of a low affinity form of nucleotide site. The ATP/ADP exchange is regarded as evidence for an ADP-sensitive form of the phosphoenzyme. In native enzyme, pre-steady state kinetics show that the formation of the phosphoenzyme is partially sensitive to ADP while modification of the enzyme by pretreatment with 5,5′-dithiobis(2-nitrobenzoic acid) (DTNB) in the absence of Mg2+ results in a steady-state phosphoenzyme population, a component of which is ADP sensitive. The ATP/ADP exchange reaction can be either stimulated or inhibited by the presence of K+ as a function of pH and Mg2+.  相似文献

Abstract: The termination of neurotransmission is achieved by rapid uptake of the released neurotransmitter by specific high-affinity neurotransmitter transporters. Most of these transporters are encoded by a family of genes (Na+/Cl transporters) having a similar membrane topography of 12 transmembrane helices. An evolutionary tree revealed five distinct subfamilies: γ-aminobutyric acid transporters, monoamine transporters, amino acid transporters, "orphan" transporters, and the recently discovered bacterial transporters. The bacterial transporters that belong to this family may help to develop heterologous expression systems with the aim of solving the three-dimensional structure of these membrane proteins. Some of the neurotransmitter transporters have been implicated as important sites for drug action. Monoamine transporters, for example, are targeted by major classes of antidepressants, psychostimulants, and antihypertensive drugs. Localization of individual transporters in specific cells and brain areas is pertinent to understanding their contribution to neurotransmission and their potential as targets for drugs. The most important questions in the field include resolving the mechanism of neurotransmitter transport, the structure of the transporters, and the interaction of each transporter in complex neurological activities.  相似文献

The activity of pure Escherichia coli murein (peptidoglycan) amidase (N-acetylmuramoyl-l-alanine amidase, EC 3.5.1.28) was measured after preincubation with E. coli phosphatidylglycerol microdispersions in final concentration ranging over micro- and millimolarities. The enzyme activity was increased up to 160% of the control for phosphatidylglycerol concentrations increasing from 2 to 50 μM. After a plateau extending from 0.05 to 0.3 mM, higher phosphatidylglycerol concentrations inactivated the enzyme down to 15% of initial activity for concentrations of 2 mM. Positive kinetic cooperativity was observed for the activation as well as for the inactivation processes. Cardiolipin (or diphosphatidylglycerol) from the same origin and under same conditions had no significant effect. Molecular sieving experiments have shown that, when inactivated, the enzyme remained firmly bound to the phosphatidylglycerol vesicles, whereas the activated phosphatidylglycerol-enzyme complex was totally dissociable by dilution. Activated phosphatidylglycerol complexes were recovered by gel exclusion chromatography at equilibrium in 40 μM phosphatidylglycerol. Possible physiological meaning of the results is briefly discussed in the context of our work and that done previously by others.  相似文献

The Enzymes II of the PEP:carbohydrate phosphotransferase system (PTS) specific for N-acetylglucosamine (IINag) and beta-glucosides (IIBgl) contain C-terminal domains that show homology with Enzyme IIIGlc of the PTS. We investigated whether one or both of the Enzymes II could substitute functionally for IIIGlc. The following results were obtained: (i) Enzyme IINag, synthesized from either a chromosomal or a plasmid-encoded nagE+ gene could replace IIIGlc in glucose, methyl alpha-glucoside and sucrose transport via the corresponding Enzymes II. An Enzyme IINag with a large deletion in the N-terminal domain but with an intact C-terminal domain could also replace IIIGlc in IIGlc-dependent glucose transport. (ii) After decryptification of the Escherichia coli bgl operon, Enzyme IIBgl could substitute for IIIGlc. (iii) Phospho-HPr-dependent phosphorylation of methyl alpha-glucoside via IINag/IIGlc is inhibited by antiserum against IIIGlc as is N-acetylglucosamine phosphorylation via IINag. (iv) In strains that contained the plasmid which coded for IINag, a protein band with a molecular weight of 62,000 D could be detected with antiserum against IIIGlc. We conclude from these results that the IIIGlc-like domain of Enzyme IINag and IIBgl can replace IIIGlc in IIIGlc-dependent carbohydrate transport and phosphorylation.  相似文献

Robust expansion of human hepatocytes in Fah-/-/Rag2-/-/Il2rg-/- mice   总被引:2,自引:0,他引:2  
Mice that could be highly repopulated with human hepatocytes would have many potential uses in drug development and research applications. The best available model of liver humanization, the uroplasminogen-activator transgenic model, has major practical limitations. To provide a broadly useful hepatic xenorepopulation system, we generated severely immunodeficient, fumarylacetoacetate hydrolase (Fah)-deficient mice. After pretreatment with a urokinase-expressing adenovirus, these animals could be highly engrafted (up to 90%) with human hepatocytes from multiple sources, including liver biopsies. Furthermore, human cells could be serially transplanted from primary donors and repopulate the liver for at least four sequential rounds. The expanded cells displayed typical human drug metabolism. This system provides a robust platform to produce high-quality human hepatocytes for tissue culture. It may also be useful for testing the toxicity of drug metabolites and for evaluating pathogens dependent on human liver cells for replication.  相似文献

The Michaelis-Menten parameters, JM and Km of the initial 1-min fluxes of uptake of l-phenylalanine and of α-aminoisobutyric acid were determined for extracellular concentrations of Na+ ranging from 0.5 to 110 mequiv/l for Ehrlich ascites tumor cells. The maximal initial flux, JM, decreased with decrease in extracellular Na+ for both α-aminoisobutyric acid and phenylalanine but the Km for α-aminoisobutyric acid increased markedly as the Na+ concentration fell whereas the Km for phenylalanine decreased. Cycloleucine behaved like phenylalanine.The data provides strong evidence that the Na+-independent flux of phenylalanine is an exchange diffusion flux that can be varied by changing the intracellular level of amino acids such as phenylalanine. For phenylalanine, cyclolcucine, and methionine this exchange diffusion flux appears to be additive with the Na+-dependent initial flux. α-Aminoisobutyric acid also has an exchange diffusion that is Na+-independent but it has a high Km and is not additive with the Na+-dependent flux.  相似文献

Balb/c mice were immunized with aluminium hydroxide [alum, Al(OH)3]-adjuvanted hepatitis B (HB) vaccines of subtypesadr,ayworadw. Spleen cells from the immune animals were fused with SP2/O cells. Eight hybridoma clones producing antibodies specific or HB surface antigen (HBsAg) were selected. Monoclonal antibodies (mAbs) of four clones were specific for group-specific antigen/a, and the other of four clones were specific for subtype antigen/d,y,r, orw. The anti-HBs/amAbs were classified into three non-competitive groups.Quantitation of group-specific determinantaof HBsAg (HBsAg/a) was performed by sandwich enzyme-linked immunosorbent assay (ELISA), in which a solid phase of anti-HBs guinea-pig polyclonal antibodies (pAb), the HBsAg for testing, anti-HBs/amouse mAb and horseradish peroxidase (HRP)-conjugated anti-mouse IgG were used.The unadsorbed HBsAg was used to establish the standard curve HBsAg/a. The lower detection limits were 0·5 to 1 ng/ml of HBsAg. Methods of solubilization of alum were investigated to quantify HBsAg/ain adsorbed HB vaccines. The recovery rate was more than 60% if vaccines were prediluted. The recovery of HBsAg/ain HB vaccines produced by the same manufacturer showed the similar recovery rate, and the contents of HBsAg/ain adsorbed HB vaccines could be estimated by the recovery rate determined for adsorbed HB vaccines.  相似文献

The nitrogen requirement of plants is predominantly supplied by NH4+ and/or NO3? from the soil solution, but the energetic cost of uptake and assimilation is generally higher for NO3? than for NH4+. We found that CO2 enrichment of the atmosphere enhanced the root uptake capacity for NO3?, but not for NH4+, in field-grown loblolly pine saplings. Increased preference for NO3? at the elevated CO2 concentration was accompanied by increased carbohydrate levels in roots. The results have important implications for the potential consequences of global climate change on plant-and ecosystem-level processes in many temperate forest ecosystems.  相似文献

The effect of Dermatophiluscongolensis on reproductive performance and milk production of dairy cattle was investigated using a prospective cohort study design. The following results were obtained: the calving to conception interval was 122 vs 104 days (P<0.05) for cows with lesions (cases) and those without lesions (non-cases), respectively; the 180-day pregnancy rate was 70.3% vs 84.3% (P<0.02) for cases and non-cases, respectively; the interval to first service was 69 days for cases and 64 days for non-cases (P<0.05) and the interval from first to second service was 32.7 days for cases and 49.4 days (P<0.01) for non-cases. The prevalence of cows with lesions was 33% (66200). Lesions were most commonly found on the skin covering the coccygeal and sacral vertebrae and the gluteal muscles. White skin had a greater prevalence of lesions than did black skin (P<0.05).  相似文献

NMR titration curves have been recorded for all the 13C resonances of cis and transN-acetyl-dl-proline in 2H2O. the measured pK2H values are 3.4 ± 0.8 and 4.13 ± 0.08 respectively; the free energy of ionization for the trans isomer being (3.8 kJ/mole) greater than for the cis. The ionization shifts of the two isomers differ significantly only at the acetyl carbonyl and Cγ positions. It is suggested that these are related to conformational changes which stabilize the trans form at low p2H.  相似文献

The in vivo binding of the radiobrominated neuroleptic brombenperidol in rat brain was studied. The accumulation of the radiolabeled neuroleptic was high in the striatum and relatively low in the cerebellum, cortex, and blood. Striatal binding of brombenperidol was saturable and displaced by subsequent administration of benperidol. The rationale for the development of 75Br-brombenperidol as a radiopharmaceutical for the non-invasive imaging of cerebral dopamine receptor areas is presented.  相似文献

The incompatibility of acid gelatin/iota-carrageenan mixtures has been studied. Both these biopolymers undergo a conformational coil-helix transition under suitable conditions of temperature and salt. The aim of this work was to study the concentration at which mixtures are incompatible and the influence of pH, salt and temperature on the phase diagram. Incompatibility occurred over a wide range of concentrations for mixtures prepared in deionized water. Compatibility was increased by increasing the pH or the salt concentration. Temperature did not greatly influence the size of the incompatible region. This is in agreement with the hypothesis that attractive electrostatic interactions lead to associative phase separation (traditionally called complex coacervation).  相似文献

(1) A flow-tube apparatus suited for measurement of rapid efflux of sugars from adipocytes is described. (2) Due to heterogeneity of fat cell populations, a conventional analysis of the time-course of net efflux of 3-O-methylglucose based on the integrated rate equation can produce gross errors in estimates of kinetic parameters. (3) The half-saturation constant and maximum transport capacity for 3-O-methylglucose transport were found to be about 3-fold higher for net efflux than for equilibrium exchange flux, both in insulin-stimulated and non-stimulated adipocytes. This suggests asymmetric kinetic parameters for 3-O-methylglucose transport.  相似文献

A cell-free particulate enzyme preparation of Mycobacterium smegmatis ATCC 607 catalyzed the transfer of labeled mannose from GDP[14C]mannose to methyl-α-mannopyranoside (an exogenously added acceptor) to form a product that was characterized to be 2-O-α-d-[14C]mannopyranosyl-methyl-α-D-mannopyranoside. This tranmannosylase activity was specific for both the sugar nucleotide donor and methyl monosaccharide acceptor. The reaction was stimulated by the addition of various metal ions and had a pH optimum of 6.0. The apparent Km of this transmannosylase reaction for methyl-α-d-mannopyranoside was 35 mM.The possible relationship between this “artificial” mannosyl-transfer system and the “natural” system which leads to the formation of the oligomannosides and glycoproteins is discussed.  相似文献

The Km(app) of ADP for photophosphorylation in lettuce chloroplasts was measured both at various light intensities and in the presence of various uncoupler (nigericin + K+) concentrations. Lowering the light intensity results in both, a decrease in the rate of phosphorylation and a several fold decrease in the Km(app) of ADP for the reaction. However, when increasing concentrations of the uncoupler nigericin + K+ are employed, the rate of photophosphorylation is decreased but a several-fold increase in the Km(app) of ADP for the reaction is observed. The results are discussed in terms of the chemiosmotic hypothesis. It is suggested that these effects might indicate the existence of a mechanism controlling the rate of ATP formation which is different than the formation of the electrochemical gradient.  相似文献

应用RT-PCR, 从真骨总目(Teleostei)5目15种鱼类中首次克隆了核糖体大亚基蛋白L15 (RPL15, ribosomal protein L15) 的完整cDNA序列。以海鲢形亚组(Elopomorpha)的鳗鲡作为外类群, 对这些真骨鱼类的核糖体蛋白L15 cDNA序列进行了系统发育分析, 结果表明: (1)RPL15基因在真骨鱼类等许多真核生物进化中高度保守; (2)系统树中各物种之间的关系与形态分类一致。RPL15编码区适合于真骨鱼类目以上分类阶元的分子系统学研究。   相似文献
