On average, 50 per cent of maize pollen grains can be kept viableand almost 30 per cent remain fertile for up to a year whenthe water content on a fresh weight basis is reduced to about30 per cent of the original and the pollen is stored at –76or –196 °C. Supplementing pollen suspension cultures with CO2 (3–5per cent) caused a marked increase in germination and tube growthin vitro in Brassica campestris L. cv. toria. A weakening ofself-incompatibility by increased CO2 levels from 3–5per cent was observed. The percentage of pollen tubes whichpenetrated the cuticle layer of stigmatic papilla cells in self-pollinatedpistils was high when CO2 level was 5 per cent. Phosphoenolpyruvate (PEP) carboxylase activity was greater in the pollengerminated in 4 per cent CO2 as compared to air (0.03 per cent).A possible role of CO2 for self-recognition and control of pollentube growth is proposed, proposed. The mineral, carbohydrate, protein, amino acid, lipid and energycontents of the seven main pollens gathered by honey-bees insouth-east Scotland were determined. Neither dry matter (mean73 per cent) nor ash (mean 3 per cent) were very variable. Thecalcium content of all pollens was relatively similar as werethe magnesium, potassium and sodium contents but manganese whichwas relatively high in ericaceae and phosphorus which was lowin Trifolium were more variable. Acer pollen contained mostprotein and Fagus least, and of the amino acids only serine,cystine and histidine were significantly different in the differentpollens. Over 30 per cent of all pollens was a mixture of fructoseand glucose, ericaceae containing most and Acer least. Hemicellulose(mean 7 per cent) was high in Ranunculus and low in Acer, cruciferaeand Fagus. ‘Cellulose’ (mean 0•5 per cent)was high in cruciferae and low in ericacea. Ether extract (mean5 per cent) was high in cruciferae and Fagus. The mean grossenergy of pollen was 23 kJ g–1. Fresh pollen is comparable with lean meat as a source of energyand protein, and is more like dormant plant seeds than herbagein its water, ash and protein content but stores energy as sugarrather than starch or lipid.  相似文献

Brassica oleracea pollen was applied to a basic medium of 1.5per cent agar and 15 per cent sucrose to which flavanoids wereadded at three concentrations. Two types of agar were used;with agar 1, quercetin at a concentration of 0.5 x 10–3per cent gave an increase in percentage germinating grains.With agar 2, an increase in germination occurred with kaempferoland naringin at concentrations of 0.5 x 10–3 and 0.5 x10–1 per cent respectively. Increase in pollen tube lengthoccurred with agar 2 and quercetin at a concentration of 0.5x 10–3 per cent. The stigma tissue of B. oleracea contains at least three andthe pollen at least one glycoside of quercetin. The sugars inthe glycosides were not identified. Pollen germination and pollentube extension were not stimulated exclusively by the flavanoidspresent in the stigma. The flavanoid composition of the stigmadid not vary amongst five different S-allele genotypes, indicatingthat flavanoids are probably not directly involved in the incompatibilityreaction of B. oleracea.  相似文献

Modification of the Pollen-Stigma Interaction in Brassica oleracea by Water   总被引:2,自引:0,他引:2  
The presence of a film of distilled water on the stigma surfaceof freshly opened flowers results in complete inhibition ofpollen following both incompatible and compatible pollinationsin self-incompatible (SI) genotypes of Brassica oleracea, SIgenotypes of B. campestris and one self-compatible (SC) genotypeof B. campestris. The application of water to the stigmas afterpollination also resulted in a marked reduction in pollen germinationand tube penetration. An increase in the time intervals betweenthe application of pollen onto the stigma and the water treatmentprogressively reduced this inhibition. Pollen germination wasalso completely inhibited when stigmas from freshly-opened flowersof SI B. campestris and B. oleracea genotypes were washed inwater, dried and pollinated with pollen grains of either compatibility.The ability of stigmas to induce pollen germination and tubegrowth was restored over a period, the length of which was dependenton the incompatibility (S) genotype. Stigmas of B. napus (SC)and SC mutants of SI B. campestris were found to be affectedby washing, but stigmas of a SC variety of B. campestris andthe immature stigmas from buds of B. oleracea were found tobe considerably less affected. Microscopic examination of pollenplaced on washed stigmas reveals that grains, irrespective oftheir compatibility, fail to hydrate normally. When inducedto hydrate by raising atmospheric humidity, pollen grains onwashed stigmas did germinate, but most of the tubes failed topenetrate the papillar wall and very few entered the style.It is proposed that the water treatment mobilises componentsof the pellicle which reorganize to block the activity of molecules,present in both SC and SI individuals, responsible for establishingfull contact between the pellicle and pollen grain coating. Cytoplasm of freshly-harvested, ungerminated Lilium longiflorum,cv. Ace pollen contains 0.14 per cent soluble and 0.35 per centprotein-bound proline (pro). Their metabolic fates in germinationand tube elongation are not known with certainty. Here is reportedconversion of pro to hydroxyproline (hyp)—containing constituentsas well as distribution and isolation of these constituents.Colorimetry revealed pro and hyp in wall, trichloroacetic acid(TCA)—precipitable, and TCA-soluble cytoplasmic fractions.A balance sheet summarizing quantitative changes in pro andhyp for these fractions revealed that TCA—precipitablecytoplasmic pro could be a precursor to wall-bound pro and asubstrate for hydroxylation yielding cytoplasmic and wall-boundhyp. To determine whether hyp was a component of tube and/orgrain walls, pollen was allowed to germinate 1.5 h and thentransferred to sorbitol medium which prevented further tubeelongation. Hyp was absent from walls of transferred pollen.Electron microscope autoradiography of tubes exposed to 2H-prosuggested that a pro- and/or hyp-containing constituent waslocalized in the growing tip. Light microscope autoradiographyof intact tubes labelled with 14C-pro showed that the constituentwas distributed throughout the pollen tubes. Previous studies have demonstrated that pollen of sorghum [Sorghumbicolor (L.) Moench] loses capacity to both germinate in vitroand to set seed in vivo soon after being shed. The current studyevaluates the capacity for dehydrated pollen to effect in vitrogermination, reduce tetrazolium chloride, and set seed on cytoplasmicmale sterile plants. Morphological changes during pollen germinationwere examined by scanning electron microscopy (SEM). Close to70% of the pollen germinated in 5 min, or less, when collectedat 80% relative humidity (RH) and stored in sealed glass vials.Pollen tubes elongated autotropically with atmospheric humidityapparently being a controlling factor in the process. Pollendehydrated at 50% RH and 25°C for 15-30 min neither germinatedin vitro, reduced tetrazolium chloride, nor set seed on malesterile plants. Rehydrating the pollen did not restore the capacityfor germination. SEM micrographs demonstrated that elongatingpollen tubes encircled the pollen grain and were contiguousto the surface. Previous work on chlorophyll-deficiency mutations in pea andbarley has shown that a significant increase in mutations isinduced by storing seeds under various conditions which leadto losses of viability to about 50 per cent. The work here showsthat a detectable increase in mutation frequency is also associatedwith much smaller losses of viability. Pea seeds were storedat 35 °C and 16.5 per cent moisture content for 40 and 57d when viability fell from 99 to 93 and 82 per cent, respectively.At the same time mutation frequency (percentage of seeds containingrecessive point mutations) increased from 1.62 per cent in thecontrol treatment to about 3 to 4 per cent. Barley seeds at15.5 per cent moisture content were stored at 50 °C for42 and 54 h, and at 35 °C for 28 and 39 d. During theseageing treatments viability fell from 98 to 75, 26, 93 and 48per cent respectively and the mutation frequency increased fromzero to between about 0.3 to 0.9 per cent. In both species theinduction of mutation by ageing treatments was significant butthe differences between the various ageing treatments were not.It is concluded that there is probably no safe threshold lossof viability which completely avoids mutation, and these resultssupport the view that for genetic conservation seeds shouldbe stored under conditions which minimise loss of viability. Tubulin was localized in pollen tubes of the grass Alopecuruspratensis by immunofluorescence using a rat monoclonal yeastantitubulin as a primary antibody. In tubes fixed in activegrowth with paraformaldehyde fixative and permeabilized withdetergent, the two male gametes were found to be linked by tubulin-containingcytoplasmic bridges. The connections appeared to persist throughoutthe growth of the tube, but they were highly mutable, the distancebetween the gamete pair varying greatly, the gametes sometimesactually overlapping in the tube. Systems of longitudinally disposed, discrete microtubule strandscomparable with those seen in generative cells of species withbicellular pollen grains were not observed in the gametes. However,an envelope of antitubulin-binding material was apparent atthe cell boundary in many instances, possibly to be interpretedas a sheath of closely packed microtubules seen in profile.Irregular masses, probably representing pools of unpolymerizedor partly polymerized tubulin, were also present in most ofthe gametes. Azetidine-2-carboxylic acid (AZC), which occurs naturally inLiliaceous plants, is reported to be a proline (pro) analoguePlant cell walls contain ‘extensin’, which is richin hydroxyproline (hyp). Peptidyl hyp arises through hydroxylationof peptidyl pro followed by glycosylation (arabinose attachment)of hyp Because AZC replaces peptidyl prolyl residues, it maybe a useful tool for evaluating the significance of hyp-o-arabinoselinkages in cell elongation. Therefore, we determined the effectof AZC on [14C]pro uptake, incorporation and conversion to wall-bound[14C]hyp in relation to elongation of lily pollen tubes whosewalls consist, in part, of hyp-containing glycopeptides TheAZC suppressed pollen germination 9–42 per cent (1–10mM) and subsequent tube elongation 40–54 per cent (0·1–1mM without affecting respiration In contrast, similar hyp concentrationswere without effect on tube elongation Whereas uptake of [14C]prowas 16·5–6·2 per cent of the control at0·1–1 mM AZC, [14C]leucine uptake was 85–25per cent of the control. Light microscope radioautography revealedfewer silver grains over tubes elongated in 0·1–1mM AZC than in its absence. Incorporation of [14C]pro into tnchloroaceticacid (TCA)-precipitable cytoplasm was reduced by only 10 percent at 0·01–1 mM but 43 per cent at 10 mM AZCGel filtration of cytoplasm from pollen germinated without AZCbut with [14C]pro resulted in labelled void volume (V) and threeretarded peaks (RI–III) Incorporation into V and RI wasinhibited at both 0·01 and 1 mM AZC These AZC concentrationsreduced conversion of [14C]pro to wall-bound hyp by 20 percent However, total incorporation of [14C]pro into salt-water-purifiedwall fractions was suppressed 47–53 per cent (0·1–1mM AZC). 应用石蜡制片、活体压片、光学显微镜及扫描电子显微镜观察等方法,研究了四倍体河南云台山野菊(Dendranthema indicum)与栽培菊花'钟山金山'(D.grandiflorum 'Zhongshanjinshan')种间杂交中父本花粉活力、花粉在柱头萌发、花粉管生长及胚胎发育情况等.结果发现,父本云台山野菊的花粉活力在授粉时为12%左右.人工授粉后的不同时间,在柱头上都观察到正常萌发的花粉粒,且花粉管都能进入柱头,其中,在授粉后0.5 h时,平均每柱头有5.9粒花粉萌发;12 h时,为59.9粒;而24和48 h时,则分别降为47.1和35.7粒.此外,在授粉后8、10、12和15 d时,分别在49.1%、40.8%、39.7%和38.5%子房内观察到正常发育的胚胎,最终杂交结实率为44.8%,而母本自然开放结实率为52.3%.研究表明,授粉前其多数母本雌蕊发育良好、授粉后多数花粉能在柱头正常萌发和花粉管正常生长,在受精后大部分胚胎发育正常是野菊与栽培菊种间杂交较高结实率的重要保证,而授粉前父本较低的花粉活力对杂交结实率影响不大.  相似文献

Mixed fluorescence/bright field microscopy of Rhododendron pollentubes in the first 72 h after germination reveals a lens-shapedgenerative cell which divides to give two associated spermswithin the original cell boundary. The generative cell is closelyassociated with the vegetative nucleus which precedes it in92 per cent of pollen tubes. Three-dimensional reconstruction from serial thin sections ofa pollen tube fixed 24 h after germination shows that the associationbetween the generative cell and vegetative nucleus is extremelycomplex. Elongated tails of the generative cell physically enfoldthe vegetative nucleus and penetrate into enclaves within it.The association has been clarified by use of the periodic acid-phosphotungsticacid-chromic acid technique to enhance electron contrast ofthe plasma membranes surrounding the generative cell. In thisbicellular system, the male germ unit association is apparentlyinitiated after pollen maturity but prior to generative celldivision. It has been reported that in species of Plumbaginaceae, Chenopodiaceae,Cruciferae and Amaryllidaceae a ‘male germ unit’is formed in which the two male gametes remain inter-connected,with one of the pair linked intimately to the vegetative nucleus.In two species the unit has been shown to remain intact in thepollen tube, and some accounts imply that it is polarized inits movement, the vegetative nucleus leading in the tube. Evidence given in this paper indicates that such a unit is unlikelyto be present in Helleborus foetidus L. (Ranunculaceae). Applicationof an optical sectioning technique has shown that at no timeis there a persistent linkage between the generative cell andthe vegetative nucleus in unhydrated, hydrated and germinatingpollen, nor is one present in the early pollen tube. Furthermore,no inter-connections between the two entities were seen in protoplastsfrom living, hydrated and incipiently germinating grains isolatedmechanically in an osmotically balancing medium. Following germination,the vegetative nucleus leaves the grain in advance of the generativecell in most instances, but in the samples examined the generativecell led in about 30 per cent of the tubes. Assembling a polarisedmale germ unit in these circumstances would require (a) theformation of an inter-connection between the vegetative nucleusand the generative cell or one of the gametes derived from itduring passage through the tube, and (b) where the generativecell initially leads in the tube, an exchange in relative positions.It is considered improbable that these conditions could consistentlybe met. Mature, incipiently germinating pollen of H. foetidus releasesa fibrillar component when extruded into suitable media. Websor clusters of fibrils are commonly seen to be associated withboth the vegetative nucleus and the generative cell. Maize plants, subjected to 0, 80, 120 and 160 meq l–1salinity using NaCl, showed adverse effects on viability, germinationand tube growth of pollen, besides enhancing the bursting ofpollen. The endogenous levels of various metabolites in pollenwere also affected. Pollen grains from salinized plants hadmore soluble carbohydrates, free amino acids, especially proline,phenols and DNA and less starch, protein and RNA compared tothe non-saline controls. Salinity also resulted in the accumulationof ions such as Na+, K+ and Cl while it caused a reductionin the boron content of pollen. These metabolic disturbancespossibly lead to decreased viability, germination and tube growthof pollen thereby resulting into a reduction in reproductivecapacity of the plants under salt stress. Summary The number of pollen grains placed upon a stigma influence both the development of pollen tubes and subsequently the progeny which result from fertilizations by gametes from these pollen tubes. The first influence is demonstrated by reduced pollen tube growth rates when pollen grains are few in number. This may indicate direct effects of pollen tubes upon the stylar tissues or perhaps more complex interactions between pollen and style. The second and potentially more important influence of limited pollination is upon the progeny. This was demonstrated with studies on three species. In each case, variation among the resultant plants was greater when pollen was limited than when normal, that is excessive, pollen was used. The mechanism of this phenomenon is not certain, but our data indicate that it is not simply an artefact of variation in seed size.  相似文献

Using fluorescence microscopy, deposition of pollen on stigmas and pollen tube growth in the gynoecium of Sagittaria potamogetifolia Merr., a monoecious species with an apocarpous gynoecium, were observed. The maximum rate of pollination averaged 83.9 +/- 4.7 %, and the number of pollen grains per stigma ranged from zero to 30. Pollen tubes grew through one stigma to the base of the ovary at almost the same speed, but generally only one of the pollen tubes then turned towards the ovule and finally entered the nucellus through the micropyle. The other pollen tubes grew through the ovary base and the receptacle tissue into ovules of adjacent carpels whose stigmas were not pollinated or which had been pollinated later. This phenomenon is termed pollen tube 'reallocation' by the authors. To verify the direct effect of the phenomenon on fruit set, artificial pollination experiments were conducted in which two or more pollen grains were placed onto only one stigma in each gynoecium; frequently more than one fruitlet was obtained from each flower treated. The reallocation of pollen tubes among pistils in the gynoecium could effect fertilization of ovules of unpollinated pistils and lead to an increase in sexual reproduction efficiency. It would, to some extent, also increase pollen tube competition among pistils of the whole gynoecium.  相似文献

We studied the functional relationship between pollination intensity and fruit survival as well as the number of seeds per pod in the tropical tree Theobroma cacao L. on a Forastero Upper-Amazon clone (UPA 409) in Ivory Coast. Cutting the style 24 h after pollination allowed for counting the number of pollen grains deposited on a stigma without affecting fruit set and seed development. Forty-three pollen grains were necessary to reach 50% of maximum fruit set 28 days after pollination. Above 115 pollen grains, the proportion of developing ovaries reached a maximum of 88% 28 days after pollination and 75% at maturity. With fewer than 238 pollen grains per stigma, there was a close relationship between pollination intensity and number of seeds per pod; the pollenseed ratio increased from 1.61 to 3.81 for PI increasing from 30 to 238 pollen grains. For higher pollination intensities, the average number of seeds per pod reached a maximum of 58. The relationship between pollination intensity and seed content was modelled. Results are consistent with the hypothesis that ovules attracted pollen tubes in a similar way regardless of whether or not they had already been reached by another pollen tube.  相似文献

 We examined the influence of pollen competitive environment on pollen performance in Mirabilis jalapa. We used the number of pollen grains and the number of pollen tubes per pistil as measures of pollen competition. Pollen germination, pollen tube penetration into the style, and pollen tube growth rates were used as measures of pollen performance. All three measures of pollen performance were affected by the competitive environment. Pollen germination was greatest at intermediate pollen load sizes. The percentage of germinated pollen grains that penetrated the stigma and grew into the style decreased with pollen load size. Pollen tube growth rate in the style was greater and more variable with larger numbers of pollen tubes in the style. Controlling for the degree of selection at the stigma indicated that pollen-pollen or pollen-style interactions were the likely causes of increased growth rates.
