SUMMARY.
  • 1 The invertebrate predators. Chaohorus, Leptodora, Hydra, Utricularia, and cyclopoid copepods were ineffective as inducers of high helmets in laboratory reared Daphnia retrocurva. Lako water from different sources, when used as culture medium, was also ineffective.
  • 2 High helmets were induced when clones were grown under warm and turbulent conditions. Presence or absence of illumination and varying photoperiod did not affect helmet growth in warm turbulent cultures. Different clones grown under warm turbulent conditions differed in their responsiveness to the treatment, but no relationship between helmet size in clones and helmet size in the field on the dates of clone isolation was demonstrated. Laboratory-induced phenotypes were rarely as extreme as those of field animals.

Although growth in adult copepods is frequently assumed to be similar to juvenile growth, some evidence have pointed out that under in situ conditions, it can be lower, with, as a consequence, underestimation of secondary production. In addition, under field conditions, juvenile growth in copepods is close to maximum rates estimated at food-saturated conditions. Based on previous assumptions, this study aimed to test the applicability of a new approach for copepod production estimate, derived from temperature-dependent growth models and in situ data, such as seawater temperature and copepod biomass. For this purpose, site-specific copepod juvenile growth models, defined for Acartia tonsa and A. clausi populations from a Southern European estuary (Canal de Mira, Ria de Aveiro, Portugal), were used and copepod biomass was taken from zooplankton samples collected during 2 years at six sampling stations. By comparing the obtained results with published data, the feasibility of the approach for copepod secondary production estimates and its applicability in worldwide marine ecosystems was confirmed. Future studies should combine the estimates of adult and juvenile production in order to evaluate its relative contribution and to obtain a more precise estimate of secondary production.  相似文献

A group of 20 young and another of 20 adult Lymnaea truncatula were abundantly supplied with food and kept continuously under cold conditions (5 degrees C) in the laboratory for 3 months and the effects of low temperature on their behavior, growth and reproduction were studied. The results indicate that at low temperature the activity of L. truncatula was markedly reduced but complete hibernation did not occur. The snails seem to be unaffected by the low temperature itself since none (both young and adult) died during the 3 months that they were kept at 5 degrees C. Reduced feeding, even in the presence of abundant food, during the cold conditions caused an almost total inhibiton of growth. Of even more significance was the suppression of reproduction which was connected with the metabolic rate of adult snails kept at low temperature. Young snails seem to profit by exposure to low temperature. On the return to normal laboratory temperature (16-22 degrees C) the young snails became very active, fed voraciously, grew rapidly, tended to live longer and produced more offspring than the controls. Low temperature, however, appears to have an adverse after-effect on the growth and reproduction of mature snails. Relatively, fewer eggs were deposited in this case. The results indicate that under natural field conditions in England, where temperature fluctuations during the usually mild winter months are common, the greater burden of increasing the population in overwintered snails must rest on the younger members of the community.  相似文献

This review discusses the growth and activity of vesicular-arbuscular (VA) mycorrhizal fungi in saline conditions. The review includes examination of the effects of high concentrations of salts on the occurrence of VA mycorrhizal fungi in field soils, and on spore germination, growth of hyphae, establishment of the symbiosis and production of spores in controlled conditions. Information on the growth and reproduction of VA mycorrhizal fungi under saline conditions is scarce and is often circumstantial. There is clear evidence that germination of spores and subsequent hyphal growth of some VA mycorrhizal fungi are reduced by increasing concentration of salts. However, in plant growth experiments, experimental designs and methodologies have generally not allowed the direct effects of salinity on fungal growth to be separated from plant-mediated effects. There is a need for controlled studies to investigate the responses of VA mycorrhizal fungi to soil salinity. Research is required which distinguishes between effects on different phases of the fungus lifecycle and which includes in its design the ability to separate direct effects from plant-mediated influences on fungal growth and reproduction.  相似文献

Fermentation optimization experiments are ideally performed at small scale to reduce time, cost and resource requirements. Currently microwell plates (MWPs) are under investigation for this purpose as the format is ideally suited to automated high-throughput experimentation. In order to translate an optimized small-scale fermentation process to laboratory and pilot scale stirred-tank reactors (STRs) it is necessary to characterize key engineering parameters at both scales given the differences in geometry and the mechanisms of aeration and agitation. In this study oxygen mass transfer coefficients are determined in three MWP formats and in 7.5 L and 75 L STRs. k(L)a values were determined in cell-free media using the dynamic gassing-out technique over a range of agitation conditions. Previously optimized culture conditions at the MWP scale were then scaled up to the larger STR scales on the basis of matched k(L)a values. The accurate reproduction of MWP (3 mL) E. coli BL21 (DE3) culture kinetics at the two larger scales was shown in terms of cell growth, protein expression, and substrate utilization for k(L)a values that provided effective mixing and gas-liquid distribution at each scale. This work suggests that k(L)a provides a useful initial scale-up criterion for MWP culture conditions which enabled a 15,000-fold scale translation in this particular case. This work complements our earlier studies on the application of DoE techniques to MWP fermentation optimization and in so doing provides a generic framework for the generation of large quantities of soluble protein in a rapid and cost-effective manner.  相似文献

Tremendous strides have been made in recent years to broaden our understanding of reproductive processes in nondomestic felid species and further our capacity to use this basic knowledge to control and manipulate reproduction of endangered cats. Much of that progress has culminated from detailed scientific studies conducted in nontraditional laboratory settings, frequently at collaborating zoological parks but also under more primitive conditions, including in the field. A mobile laboratory approach is described, which incorporates a diverse array of disciplines and research techniques. This approach has been extremely useful, especially for conducting gamete characterization and function studies as well as reproductive surveys, and for facilitating the development of assisted reproductive technology. With continuing advances in assisted reproduction in rare felids, more procedures are being conducted primarily as service-related activities, targeted to increase effectiveness of species propagation and population management. It can be a challenge for both investigators and institutional animal care and use committees (IACUCs) to differentiate these service-based procedures from traditional research studies (that require IACUC oversight). For research with rare cat species, multi-institutional collaboration frequently is necessary to gain access to scientifically meaningful numbers of study subjects. Similarly, for service-based efforts, the ability to perform reproductive procedures across institutions under nonstandard laboratory conditions is critical to applying reproductive sciences for managing and preserving threatened cat populations. Reproductive sciences can most effectively assist population management programs (e.g., Species Survival Plans) in addressing conservation priorities if these research and service-related procedures can be conducted "on the road" at distant national and international locales. This mobile laboratory approach has applications beyond endangered species research, notably for other scientific fields (e.g., studies of hereditary disease in domestic cat models) in which bringing the laboratory to the subject is of value.  相似文献

Physiological data are needed for life history studies on krill, and as parameters for input into energy budgets and models. In conjunction with moult and growth data, these may also prove useful for assessing the fishable biomass of krill. Here, the development of physiological concepts in experimental krill research is briefly evaluated, with emphasis on the gaps to be filled. Krill growth is very flexible, as well as strongly temperature and nutrition dependent. The polar Antarctic krill Euphausia superba grows as fast as the boreal species Meganyctiphanes norvegica, at least during the first 2.5 years, and the species are comparable in terms of physiological plasticity. Accordingly, as krill appear to adjust quickly to specific laboratory conditions, short-term experiments are essential if field conditions are to be reflected as closely as possible. Furthermore, direct comparisons between laboratory experiments and swarming studies in the field are advantageous. For these, M. norvegica is particularly well-suited, as swarms can be followed over longer times and more easily than in E. superba. For example, processes of moult and reproduction were found to be highly coordinated in swarms and populations of Northern krill. For this species a conceptual model of reproduction was developed based on a combination of short-term laboratory observations coupled with field data on moult and ovary stages. In further physiological experiments krill should be studied as groups when swarming. Using proxies, that is applying physiological and/or biochemical methods side by side, is a promising way to enhance the reliability of life history data.  相似文献

A short review is given about recent observations of infections in pregnant laboratory mice with strains of Trypanosoma musculi (= T. duttoni) isolated in southern Europe. When being infected during the highly sensitive period of mbryogenesis, the female is completely unable to inhibit the reproduction of the trypanosome. Like an excessively growing benign tumor, the normally well-adapted nonpathogenic blood parasite population becomes, under these conditions, harmful to the host. The placenta acts as a “focus” of the trypanosome reproduction. Unusual, incomplete multiple fissions occur as big rosette formations in the placental blood. The possible microëcological conditions of the accelerated trypanosome growth are briefly discussed. An American strain of this trypanosome species shows the same behavioral pattern in pregnant hosts.  相似文献

Competition and metabolism should be linked. Intraspecific variation in metabolic rates and, hence, resource demands covary with competitive ability. The effects of metabolism on conspecific interactions, however, have mostly been studied under laboratory conditions. We used a trait‐specific response‐surface design to test for the effects of metabolism on pairwise interactions of the marine colonial invertebrate, Bugula neritina in the field. Specifically, we compared the performance (survival, growth, and reproduction) of focal individuals, both in the presence and absence of a neighbor colony, both of which had their metabolic phenotype characterized. Survival of focal colonies depended on the metabolic phenotype of the neighboring individual, and on the combination of both the focal and neighbor colony metabolic phenotypes that were present. Surprisingly, we found pervasive effects of neighbor metabolic phenotypes on focal colony growth and reproduction, although the sign and strength of these effects showed strong microenvironmental variability. Overall, we find that the metabolic phenotype changes the strength of competitive interactions, but these effects are highly contingent on local conditions. We suggest future studies explore how variation in metabolic rate affects organisms beyond the focal organism alone, particularly under field conditions.  相似文献

The influence of water temperature, salinity, and pH on the multiplication of toxigenic Vibrio cholerae serovar O1 cells and their attachment to live planktonic crustaceans, i.e., copepods, was investigated by using laboratory microcosms. By increasing water temperatures up to 30 degrees C, a pronounced effect on the multiplication of V. cholerae was demonstrated, as was attachment of the cells to live copepods. These were measured by culturable counts on agar plates and direct observation by scanning electron microscopy, respectively. Of the three salinities examined (5, 10, and 15%), maximum growth of V. cholerae and attachment to copepods occurred at 15%. An alkaline pH (8.5) was optimal both for attachment and multiplication of V. cholerae, as compared with pH 6.5 and 7.5. It is concluded that conditions affecting attachment of V. cholerae serovar O1 to live copepods observed under laboratory conditions may also occur in the natural estuarine environment and, thereby, are significant in the epidemiology of cholera.  相似文献

Reproduction is costly and life‐history theory predicts that current parental investment will result in lower survival or decreased future reproduction. The physiological mechanisms mediating the link between reproduction and survival are still under debate and elevated oxidative damage during reproduction has been proposed as a plausible candidate. Previous studies of oxidative stress during reproduction in animals under natural conditions have been restricted to analyses of blood. Herein, we measured the level of oxidative damage to lipids (tiobarbituric‐acid‐reactive substances) and proteins (carbonyls) in the liver, kidneys, heart and skeletal muscles in free‐living bank vole females from spring and autumn generations, before and after reproduction. Antioxidant defense in the liver and kidneys was also determined. We expected oxidative damage to tissues and hypothesized that the damage would be more uniform between tissues in wild animals compared to those breeding under laboratory conditions. Considering all combinations of markers/tissues/generations, oxidative damage in females did not differ before and after reproduction in 12 comparisons, was lower after reproduction in three comparisons, and was higher after breeding in one comparison. The total glutathione was significantly increased after reproduction only in the liver of the autumn generation and there was no change in catalase activity. Our results confirm—for the first time in the field—previous observations from laboratory studies that there is no simple link between oxidative stress and reproduction and that patterns depend on the tissue and marker being studied. Overall, however, our study does not support the hypothesis that the cost of reproduction in bank voles is mediated by oxidative stress in these tissues.  相似文献

Xenopus laevis, commonly known as the South African Clawed frog, is a hardy adaptable species that is relatively easy to maintain as a laboratory animal. Gametogenesis in wild Xenopus laevis is continuous and under ideal conditions, reproduction can occur year round. This unique aspect of amphibian reproduction offers an advantage over mammalian model systems: the eggs and oocytes collected from laboratory maintained Xenopus laevis provide an abundant and readily obtainable supply of material for cellular and biological research. However, many investigators report that laboratory Xenopus laevis go through periods of unexplained inefficient or complete failure of oocyte production or the production of poor quality oocytes. This results in experimental delays, inability to reproduce data, and ultimately the use of more animals. There is a lack of evidenced based information regarding the housing conditions that are necessary to optimize the health and fecundity of this species in captivity, but studies of wild Xenopus laevis have shown that temperature, age of the female, and nutrition are of key importance. The objective of this report is to review oogenesis with a special emphasis on these factors as they pertain to laboratory Xenopus laevis maintained for the purpose of providing a steady supply of eggs and oocytes. Harvesting methods and other experimental techniques that affect the quality of eggs and oocytes are also discussed.  相似文献

The purpose of this investigation was to determine the long-term effect of enterically administered indomethacin (a prostaglandin synthetase inhibitor) on reproduction in Peromyscus maniculatus (deermice) under both laboratory and field conditions. Indomethacin-coated wheat (1 mg/kg wheat) was provided to the experimental animals in the laboratory and under field conditions ad libitum. Rat chow was also provided to the animals in the laboratory. The laboratory data indicate that the number and size of the litters from the control breeding pairs were larger (number of litters=22; average number of pups/litter=4.9) than the experimental breeding pairs (number of litters=2; average number of pups/litter=2.5) over a 100-day experimental period. Field data indicate that the number of pregnant animals, the total number of animals, and mean juvenile males and females live-trapped decreased in the experimental group with respect to the control group during the experiment. The data suggest that deermice will consume indomethacin under both laboratory and field conditions, and that it effectively inhibits reproduction on a long-term basis. It is, therefore, possible that prostaglandin synthetase inhibitors may be used to control populations of rodent pests. Additionally, the data raise questions about the effect of long-term use of nonsteroidal analgesic compounds similar to indomethacin on human reproduction.  相似文献

To evaluate the effect of laboratory passaging of influenza virus A/USSR/90/77 (H1N1) on the pattern of vRNA synthesis regulation in course of the one step infection cycle, we have used the viral variants adapted to growth in the continuous cell line MDCK or to the reproduction in the mice lungs in vivo. Enhancement of regulation was registered in the adapted variants as compared to the original virus strain. The results are discussed in connection with possible significance of the vRNA synthesis regulation for the efficiency of viral reproduction under natural conditions or in laboratory passaging.  相似文献

The effects of acibenzolar-S-methyl (ASM) and four combinations of plant growth-promoting rhizobacteria (PGPR) on the reproduction of a tobacco cyst nematode, Globodera tabacum solanacearum, and growth of Nicotiana tabacum (cv. K326 and Xanthi) were tested under greenhouse and field conditions. The PGPR included combinations of Bacillus subtilis A13 with B. pumilis INR7, B. pumilis SE34, B. licheniformis IN937b, or B. amyloliquefaciens IN937a, respectively. Among the four rhizobacterial combinations, IN937a + A13 exhibited the most consistent reduction in G. t. solanacearum cysts under greenhouse and field conditions. No undesirable effects of IN937a + A13 were observed on tobacco growth under greenhouse and field conditions. Use of INR7 + A13 reduced G. t. solanacearum reproduction on flue-cured tobacco cv. K326 but not on oriental tobacco cv. Xanthi. Application of ASM reduced final numbers of G. t. solanacearum cysts, but also resulted in phytotoxicity mainly under the greenhouse conditions. When oriental tobacco seedlings were pre-grown in a IN937a + A13-treated soil-less medium, a single application of ASM at 200 mg/L one week after transplanting significantly reduced G. t. solanacearum reproduction in the field.  相似文献

This study investigates how the presence of symbiotic crabs (Pinnixa chaetopterana or Polyonyx gibbesi) in the tubes of the polychaete Chaetopterus variopedatus affects the worms' pumping activity and growth rates under laboratory and field conditions. In the field, worms whose tubes are inhabited by Pinnixa beat their fan segments significantly more frequently than do worms hosting Polyonyx, but other measures of pumping activity do not differ according to symbiont species. In the lab, worms tend to move water through their tubes at higher rates when crabs are present. In 7-month laboratory experiments, growth rates of worms hosting either species of crab did not differ from growth rates of worms without crab symbionts. Although worms hosting Polyonyx are, on average, significantly larger than worms hosting Pinnixa, this appears to be due to competition between the crab species for hosts and not due to differential effects on host growth. Unlike the crabs in this study, pea crab species inhabiting bivalves are known to have strong deleterious effects on host growth and reproduction, suggesting that the evolution of virulence in symbiotic interactions is dependent upon specific ecological context.  相似文献

A reliable marking technique was needed for a mark–release–recapture experiment with adults of Diabrotica speciosa (Germar). Four marking techniques, acrylic paint (spattered or brushed on the surface of the insect); and fluorescent pigments (dusted on surfaces or mixed with diet to produce an ingested marker), were tested. Fluorescent pigment durability for the dusting and ingested techniques was evaluated for laboratory conditions and under simulated field conditions. The impact of the techniques on beetle survival was also assessed. Both acrylic paint techniques caused mobility problems in the beetles, and neither technique lasted for more than 48 h. Both fluorescent pigment techniques were more reliable, but the dusting technique showed a significantly higher mortality than the control, and duration variations between laboratory and field conditions. Use of fluorescent pigments added to the diet was the most reliable technique. This technique allowed the manipulation of the marking period, and provided reliable timing of marker persistence in the field.  相似文献

Summary Models incorporating the energetics of individual daphnids (Cladocera) have been developed to predict the effect of environmental variables, particularly food availability, on population dynamics. One of them, that of Kooijman (1986), assumes that all assimilated energy enters a storage compartment prior to use in production and metabolism, and that under starvation the stores are used to support maintenance, reproduction and somatic growth, in that order of priority. This predicts that, under starvation, reproduction and growth will continue for a time, and that after they cease death will be immediate. Another model, that of McCauley et al. (1990), assumes that assimilated energy is used directly for maintenance and production, and that stores are accumulated to support maintenance metabolism under starvation. This predicts that growth and reproduction should cease immediately upon starvation and that death will not be immediate. We have carried out laboratory experiments, manipulating starvation time, on Daphnia magna to distinguish between these two models. The results support features of both models in that reproduction, but not growth, ceases upon starvation. We therefore developed a third model in which both maintenance and growth are supported from stores under starvation, with maintenance taking priority over growth under these conditions.  相似文献
