6—OHDA损毁背NE束对大鼠齿状回习得性长时程增强的影响   总被引:3,自引:0,他引:3

本工作用6-OHDA损毁大鼠双侧背去甲肾上腺素束,其后在条件性饮水反应建立中检测齿状回(DG)的突触效应及条件反应的变化,以探查NE在习得性长时程增强形成中的作用。结果如下:双测背NE束注6-OHDA(12μg/4μl)后,DG的群体锋电位(PS)的峰幅值持续下降,注药后第6天下降至50±6.3%,第12天下降至28±6.5%。在第6-12天这段时间,每天给大鼠进行20次训练,经7d 140次训练,DG并无习得性LTP产生,条件性饮水反应也不能建立,这些结果提示DG的NE的正常水平对其习得性LTP的形成是必要的,并提示脑干的蓝斑核可能通过背NE束对海马DG的习得性LTP的形成起调制作用。  相似文献

人和动物形成多样的、快速可变的刺激-反应联合关系的过程被称为条件性运动学习。条件性运动学习使得人和动物具有很强的适应优势。损毁或行为电生理研究表明:运动前区背外侧部、基底神经节以及前额叶皮层腹侧部在条件性运动学习中起至关重要的作用;海马在条件性运动学习中也起着一定的作用;而杏仁核等一些结构在条件性运动学习中不起作用。  相似文献

在大鼠条件性饮水反应的建立、消退和再建立过程中,于海马 CA_3区记录电极部位微量注射 M-胆碱受体阻断剂东莨菪碱和 GABA 受体阻断剂印防已毒素,观察其对习得性长时程突触增强的影响。结果表明,东莨菪碱有明显的抑制作用,印防已毒素则有明显的易化作用,同时相应地影响条件性行为;并发现习得性长时程突触增强的发展与变化是超前于条件性行为的发展和改变的。上述结果为进一步论证习得性长时程突触增强可能是学习和记忆的神经基础之一提供了证据,并提示海马 CA_3区习得性长时程增强的产生与保持有胆碱受体与 GABA 受体参与。  相似文献

实验在25只乌拉坦(700m/kg)、氯醛糖(35mg/kg)麻醉,肌肉麻痹,人工呼吸的家兔上进行。第一组16只家兔中,单或双脉冲刺激下丘脑和中脑防御反应区,在延髓腹侧记录刺激所兴奋的单位。大部分单位分布于网状巨细胞核腹侧α部。52%的单位有自发放电活动。用阈下强度同时刺激下丘脑和中脑,97%单位有兴奋反应,提示家兔下丘脑和中脑防御反应区在延髓腹侧有聚合投射。第二组9只家兔中,在延髓腹表面单侧应用甘氨酸滤纸片或电凝损毁时,血压轻度下降,刺激下丘脑和中脑防御反应区引起的升压反应也部分被阻断。双侧应用甘氨酸或损毁,血压下降到脊动物水平,升压反应几乎完全被阻断。上述结果提示家兔延髓腹侧神经元在维持正常血压水平和在中继防御反应传出通路中起重要作用。  相似文献

目的:探讨杏仁中央核(CeA)损毁对缺钠大鼠钠欲行为启动和表达的影响。方法:将18只成年雄性SD大鼠随机分为3组(n=6):双侧Ce A损毁组、假损毁组和不损毁组,手术恢复后给予大鼠14 d低钠饲料摄食以建立缺钠大鼠模型,运用单笼双瓶选择测试方法观察缺钠大鼠在24 h内5个不同时间段对0.3 mol/L NaCl和自由饮水的摄入情况。应用免疫荧光化学染色方法观察杏仁中央核损毁与否对缺钠或正常大鼠孤束核内醛固酮敏感神经元活动的影响。结果:低钠饮食14 d后,大鼠对0.3 mol/L NaCl 24 h内饮用量和偏爱率比低钠饮食前明显增加(P<0.01);杏仁中央核损毁后缺钠大鼠对0.3 mol/L Na Cl溶液的摄入量和偏爱率显著下降(P<0.01)。杏仁中央核损毁对低钠饮食诱发的大鼠孤束核内醛固酮敏感神经元活动增加没有影响。结论:低钠饮食诱导大鼠钠欲行为表达增加;杏仁中央核损毁压抑缺钠大鼠钠欲行为的表达,而对缺钠大鼠的钠欲行为的启动没有影响。  相似文献

损毁双侧海马后,大白鼠迷宫分辨学习的保存和防御条件反应均遭到严重的破坏。大区域损毁海马,不仅影响动物原先学会的反应,而且影响重新学习的能力。同等量皮层损毁对记忆功能没有影响。海马背部与腹部在学习记忆中的作用是不同的,海马腹部在迷宫分辨学习的记忆中起主要作用,而背部对这种分辨记忆不起作用。  相似文献

本工作采用了行为和脑内注射相结合的方法研究了大鼠尾壳核的 GABA 能传递在条件性行为调控中的作用。在分辨学习的基础上训练大鼠完成条件性回避任务,以比较药物对分辨学习和条件性回避的不同效应。实验结果表明,于大鼠双侧尾壳核内分别注入 γ-氨基丁酸(GABA)(每侧100μg/μl)和 GABA 受体激动剂蝇蕈醇(Muscimol)(每侧0.1μg/μl)后可暂时抑制条件性回避反应的出现,但分辨学习无明显影响。作为对照,于尾壳核内注入等量的生理盐水则既不影响条件性回避反应,也不影响分辨学习。在条件性回避反应被 Muscimol抑制后于尾壳核内再注入 GABA 受体阻断剂印防己毒素(PTX)(每侧0.1μg/μl)则可拮抗Muscimol 的行为抑制效应,即条件反应的出现率可恢复到或接近注射前水平。实验结果表明,大鼠尾壳核的 GABA 能传递在条件性行为调控中的重要作用。  相似文献

我们的工作表明,大鼠在明暗辨别学习过程中海马齿状回有习得性长时程增强(Long-term potentiation,LTP)现象,又CA_3区在大鼠学习和记忆过程有重要作用。本实验观察大白鼠海马CA_3区锥体细胞在条件性饮水反应的建立、巩固和消退过程中其突触效应的变化规律,以进一步探讨习得性LTP的特性,及从突触水平探讨海马CA_3区在学习记忆功能中的作用。  相似文献

电刺激缰核(Hb)可使大鼠血压明显升高,但心率变化不明显。电刺激Hb血压升高的同时,延髓腹侧结构(VM)中46.5%的神经元自发放电频率增加,这些神经元分布在VM的升压神经元群中,41.9%的神经元自发放电频率降低,这些神经元分布在VM的降压神经元群中,11.6%的神经元自发放电频率不变,升压和降压神经元群中均有这种神经元。单侧损毁VM的网状旁巨细胞核后,动物血压基本不变,也不影响电刺激Hb引起的升压反应。双侧损毁VM的网状旁巨细胞核后,动物血压从98.4±11.2mmHg降至45.2±10.3mmHg,阻断了电刺激Hb引起的升压反应。单侧或双恻损毁疑核及部分延髓网状核腹侧后,动物血压基本不变,且不影响电刺激Hb引起的升压反应。 上述结果表明,Hb主要通过VM中升压神经元群的网状旁巨细胞核参与心血管活动的调节。  相似文献

In one experiment using conditioned taste aversion and the unconditioned stimulus (US) preexposure procedure, one group of rats was given LiCl exposure for 3 days, whereas two other groups received saline. Following this phase, all groups were given a novel flavour (saccharine) to drink following either LiCl (group preexposed and one of the control groups) or saline injections (the remaining control group) and the consumption of the flavour was assessed. After this neophobia test, the acquired saccharine aversion was evaluated. The results show that three LiCl injections are enough to produce a US preexposure effect on backward excitatory taste aversion conditioning, whereas this number of injections procedure does not produce habituation of the increment in neophobia, an unconditioned response to the LiCl. The results are discussed taking into account different mechanisms involved in US preexposure effect.  相似文献

Rats with mammillary electrolytic lesions show a strong polydipsia and polyuria. This over-consumption may be primary or secondary to the polyuric effect. In this regard, mammillary lesioned rats excrete a greater amount of urine compared with control animals when matched in daily water consumption (partial water deprivation). Moreover, this abnormal water intake is significantly reversed by treatment with Pitressin, a vasopressin analogue. These results suggest that the polydipsia may be determined by the urinary water loss. However, when subjected to the bilateral ureter ligation, the experimental animals still outdrink the control ones, thus also suggesting a primary component of the polydipsia under study. The possible explanation of these components in relation to the mammillary polydipsia is discussed.  相似文献

本实验应用慢性埋植电极技术以电生理学结合行为学的方法,观察大鼠条件性饮水反应的建立、消退和再建立过程中,其海马CA_3区突触效应的变化规律。以刺激内嗅区的穿通纤维(PP)诱发的单突触的群体锋电位(PS)及群体兴奋性突触后电位(EPSPs)为指标,经叠加处理分析,发现随着条件反应的建立,海马CA_3锥体细胞出现突触效应的长时程增强(LTP),它随行为反应的实验性消退而消退,而在随后再次建立条件反应时,又重新出现;且无论此LTP达最高水平还是它的完全消退均超前于条件性行为反应的水平。又在一个实验日训练作业结束时PS并未立即随之增大,在24h内它随时间而发展,但到第4小时已达最高水平,且条件反应率是与PS的水平相应的,对PS与EPSPs的斜率进行相关分析表明,PS的变化主要是突触传递功效的变化。上述结果表明,海马CA_3区随着行为训练有习得性LTP产生。从其发神变化特点及其与条件性行为的关系,提示此习得性LTP极其可能是本实验中学习和记忆的展经基础。  相似文献

Generalization of defensive conditioned reflexes elaborated to electrical stimulation of the lateral hypothalamus (LH) was studied in four dogs with electrodes implanted in various structures of the limbic system. Electrocutaneous stimulation was switched off when the dog lifted the foreleg to a definite level. Generalization of the conditioned reflex was manifested in different degrees when testing different formations of the limbic system, or testing one and the same structure, but at different stages of conditioning. Two types of generalization were found: the first one--a well pronounced motor reaction, by its latency, level and duration of lifting the foreleg similar to movements appearing in response to the conditioned stimulus; and the second one--low amplitude or short-term movements differing from conditioned ones. The first type of generalization was observed in response to stimulation of LH, contralateral to the point of signal stimulation, of the ventromedial hypothalamic nucleus, the mammillary bodies and the basal, lateral and to a lesser degree, the central nucleus of the amygdaloid complex; the second type--in response to stimulation of the ventral hippocampus, the medial and lateral septum nuclei.  相似文献

The induction of excessive grooming by intraventricular administration of ACTH1–24 was studied in rats with lesions in midbrain-limbic structures. Such areas have been reported to be implicated in mediating ACTH-induced effects on avoidance behavior, sexual excitement or stretching and yawning. Electrolytic lesions in the septal complex, the anterior hypothalamic/preoptic area, the mammillary bodies, the amygdala, the posterior thalamus and dorsal or ventral hippocampus did not interfere with ACTH-induced excessive grooming. Lesioning of the hippocampal complex by aspiration led to an inhibition of excessive grooming depending on the degree of hippocampal damage. Amygdala and hippocampal lesions enhanced the display of stretching and yawning activity after treatment with the peptide. The data indicate differences in the neural substrates mediating the effect of ACTH on extinction of conditioned avoidance behavior, excessive grooming, sexual excitement and stretching and yawning.  相似文献

An extinguished conditioned response can sometimes be restored. Previous research has shown that this renewal effect depends on the context in which conditioning versus extinction takes place. Here we provide evidence that the dorsal hippocampus is critically involved in the representation of context that underscores the renewal effect. We performed electrolytic lesions in dorsal hippocampus, before or after extinction, in a conditioned taste aversion paradigm with rats. Rats that underwent all conditioning, extinction and testing procedures in the same experimental context showed no renewal during testing in the original context. In contrast, rats that underwent extinction procedures in a different experimental context than the one in which they had acquired the conditioned response, showed a reliable renewal effect during testing in the original context. When electrolytic lesion was performed prior to extinction, the context-dependent renewal effect was disrupted. When electrolytic lesion was undertaken after extinction, we observed a complex pattern of data including the blockage of the conventional renewal effect, and the appearance of an unconventional renewal effect. The implications of these results are discussed with respect to current views on the role of the dorsal hippocampus in processing context information.  相似文献

The antipsychotic profile of 5-[2-[4-(6-fluoro-1H-indole-3-yl)piperidin-1-yl]ethyl]-4-(4-fluorophenyl)thiazole-2-carboxylic acid amide (NRA0562) was investigated using the conditioned avoidance test in rats. NRA0562 is a putative "atypical" antipsychotic agent with moderate to high affinities for dopamine D(1), D(2), D(4), 5-hydroxytryptamine(2A) receptors and alpha(1) adrenoceptor. NRA0562 (1 and 3 mg/kg, p.o.) dose-dependently and significantly impaired the conditioned avoidance response. Likewise other atypical antipsychotics such as risperidone (1 and 3 mg/kg, p.o.) and clozapine (100 mg/kg, p.o.) dose-dependently and significantly impaired the conditioned avoidance response in rats. In addition, typical antipsychotics, haloperidol (1 and 3 mg/kg, p.o.) potently impaired the conditioned avoidance response.These results suggest that antipsychotic profile of NRA0562 is consistent with profiles of clozapine or risperidone and may be considered an atypical antipsychotic agent.  相似文献

Injection of rats with cyclophosphamide (CY) after their consumption of a novel saccharin-flavored drinking solution results in a conditioned aversion to saccharin and a conditioned suppression of immune responses. In this study, female Lewis X Brown Norwegian F1 rats were conditioned by pairing saccharin with 50 mg/kg CY. Seven weeks later (day 0), a graft-vs-host response (GvHR) was induced in these animals by injecting splenic leukocytes from Lewis donors into a rear footpad. At this time, some conditioned animals were reexposed to saccharin, the conditioned stimulus. During the 7-wk interval between conditioning and immunization, subgroups of conditioned rats were given 0, 4, 9, or 18 extinction trials (saccharin followed by saline injections). Animals receiving 4, 9, or 18 extinction trials showed a greater preference for saccharin on day 0 than did animals receiving no extinction trials, but these groups did not differ among themselves; all conditioned groups showed a lower preference for saccharin than placebo-treated animals. There was a clear effect of number of extinction trials on the GvHR. Animals receiving 9 or 18 extinction trials did not differ from controls, whereas animals receiving 0 or 4 trials had a milder GvHR than did conditioned rats that were not reexposed to saccharin at the time of immunization. These results confirm a previous report of conditioned suppression of a GvHR, demonstrate that conditioned immunopharmacologic responses are subject to experimental extinction, and indicate that conditioned immunosuppression can be dissociated from conditioned taste aversion.  相似文献
