Summary Two types of polysaccharides were separated from the extracellular polysaccharide produced by Anabaena flos-aquae A-37 by ion exchange chromatography. The neutral polysaccharide is composed of mainly glucose with minor amounts of xylose in a molar ratio of 8:1. Glucose is believed to constitute the polysaccharide core to which xylose is attached. The acidic polysaccharide is composed of glucose and uronic acid as the major monomers with equal amounts of xylose and ribose as the minor constituents. The molar ratio of the monomers found in the acidic polymer is 6:1:1:10 as glucose: xylose: ribose: uronic acid. Chemical analyses showed that the extracellular polysaccharide consists of more neutral polymer (62%) than the acidic polymer (38%).  相似文献

The main component of the mucilage in the bulbs of Suisen (Narcissus tazetta L., var. chinensis Roem) has been shown to be a glucomannan composed of D-glucose and D-mannose in the ratio of 2:3 and having a relatively low degree of branching. Acetolysis of the polysaccharide led to the isolation of β-(1→4)-β-(1→3)-linked oligosaccharides composed of D-mannose and/or D-glucose residues. The average chain length (c.1.) of the polysaccharide was determined by methylation analysis to be about 22.  相似文献

Summary A polysaccharide-producing Gram-negative bacterium was isolated from a sample of hay. It grew best on nitrate-containing media with sucrose as carbon source; the colony form was highly unusual. No polysaccharide was formed on glucose. A spontaneous mutant producing the same polysaccharide on both sucrose- and glucose-containing media was isolated. The polysaccharide has been characterized as an acid heteropolymer containing D-galacturonic acid, D-glucose, D-galactose and D-mannose in the approximate molar ratio 1:1:3:1, together with about 2 mol of acetate. The polysaccharide in aqueous solution was highly viscous with pseudoplastic characteristics.  相似文献

Rhodococcus erythropolis PR4 is a marine bacterium that can degrade various alkanes including pristane, a C(19) branched alkane. This strain produces a large quantity of extracellular polysaccharides, which are assumed to play an important role in the hydrocarbon tolerance of this bacterium. The strain produced two acidic extracellular polysaccharides, FR1 and FR2, and the latter showed emulsifying activity toward clove oil, whereas the former did not. FR2 was composed of D-galactose, D-glucose, D-mannose, D-glucuronic acid, and pyruvic acid at a molar ratio of 1:1:1:1:1, and contained 2.9% (w/w) stearic acid and 4.3% (w/w) palmitic acid attached via ester bonds. Therefore, we designated FR2 as a PR4 fatty acid-containing extracellular polysaccharide or FACEPS. The chemical structure of the PR4 FACEPS polysaccharide chain was determined by 1D (1)H and (13)C NMR spectroscopies as well as by 2D DQF-COSY, TOCSY, HMQC, HMBC, and NOESY experiments. The extracellular polysaccharide produced by Lactobacillus rhamnosus strain C83 was found to be composed of D-glucose and D-galactose in a molar ratio of 2:3. A water-soluble polysaccharide, (Fr. I) isolated from the aqueous extract of an edible mushroom, Volvariella diplasia, is composed of D-glucose, D-mannose, and D-galactose in a molar ratio 3:1:1. Burkholderia caribensis strain MWAP71 was isolated from rhizosphere soil microaggregates in Martinique. The extracellular polysaccharide produced by this strain was found to be composed of D-glucose (D-Glc), 6-deoxy-L-talose (L-6dTal), 3-deoxy-D-manno-oct-2-ulosonic acid (Kdo), and an O-acetyl group in a molar ratio of 2:1:1:1. Rhodococcus sp. 33 can tolerate and efficiently degrade various concentrations of benzene, one of the most toxic and prevailing environmental pollutants. This strain produces a large quantity of extracellular polysaccharide (33 EPS), which plays an important role in the benzene tolerance in Rhodococcus sp. 33, especially by helping the cells to survive an initial challenge with benzene. This EPS has been reported to be composed of D-galactose, D-glucose, D-mannose, D-glucuronic acid, and pyruvic acid at a molar ratio of 1:1:1:1:1. To understand the protective effect of 33 EPS, we determined its chemical structure by using 1H and 13C NMR spectroscopy including 2D DQF-COSY, TOCSY, HMQC, HMBC, and NOESY experiments. The seeds of Anthocephalus indicus contain a water-soluble polysaccharide composed of D-xylose, D-mannose, and D-glucose in the molar ratios 1:3:5. Methylation analysis afforded 2,3,4-tri-O-methyl-D-xylose, 2,3,6,-tri-O-methyl-D-mannose, 2,3,6-tri-O-methyl-D-glucose, 2,3-di-O-methyl-D-glucose, and 2,3,4,6-tetra-O-methyl-D-glucose in the molar ratios 7:21:12:15:8. Periodate oxidation and methylation data indicated 22.5% and 21.9% of end groups, respectively. The above findings, together with the results of partial hydrolysis with acid, indicate the polysaccharide to consist of a linear chain of (1→4)-linked β-D-mannosyl and β-D-glucosyl residues to which α-D-xylosyl and β-D-glucosyl groups are attached by (1→6)-linkages.  相似文献

A cell-wall preparation from the cells of Elsinoe leucospila, which produces elsinan extracellularly when grown on sucrose or glucose-potato extract medium, was fractionated systematically. The heteropolysaccharide that was released by treatment with Actinase E digestion, comprised D-mannose, D-galactose, and D-glucose (molar ratio, 1.5:1.0:0.1). Methylation, mild acid hydrolysis, and 13C-NMR studies suggested that the polysaccharide contains a backbone of alpha-(1----6)-linked D-mannose residues having two kinds of side chains, one attached at the O-4 with single or short beta-(1----6)-linked D-galactofuranosyl residues, and the other attached at O-2 with short side chains, most probably, of alpha-(1----3)-linked D-mannopyranosyl residues. A moderately branched D-glucan fraction, obtained from the cold alkali extract, was fractionated to give an antitumor-active purified beta-(1----3)-glucan having branches of single beta-D-glucosyl groups, one out of eight D-glucose residues being substituted at the O-6.  相似文献

The extracellular, acidic heteropolysaccharide from Xanthomonas S19 consists of D-glucuronic acid, D-glucose, D-galactose, and D-mannose residues in the approximate molar ratios of 1.6:3:1:1, plus acetyl groups liked to C-2 and/or C-3 of a large proportion of the glucose residues. Methylation studies showed that the glucose is present as non-reducing end-group also as 1,2- and 1,4-linked units, the galactose residues are solely 1,3-linked, a major proportion of the mannose residues are 1,2,4-linked and the rest 1,2-linked. A high proportion of the glucuronic acid units are 1,4-linked. Periodate oxidation confirmed the presence of these linkages. The disaccharides D-Glc-(1→4)-D-Glc,D-Glc-(1→2)-D-Man, D-Glc-(1→3)-D-Gal, D-Gal-(1→2)-D-Glc, D-GlcA-(1→4)-D-GlcA, and β-D-GlcA-(1→4)-D-Man were isolated from a partial hydrolysate of the polysaccharide, and characterised. The similarities and differences between this polysaccharide and those from other Xanthomonas species are discussed.  相似文献

长白楤木是一种具有药用价值的植物。本实验用热水浸提、醇析、Severge法除蛋白,水相透析得到粗多糖。粗多糖经过DEAE-纤维素柱层析洗脱,再经SephadexG-200葡聚糖凝胶柱层析后,得到4种纯多糖,命名为根(gNaCl)茎(JH2O)茎(JNaCl)叶(YNaCl)。红外光谱和其完全水解的高效液相色谱分析表明,4种多糖不含硫酸基团和乙酰基,具备β-糖苷键以及糖类特征吸收峰。其中,根的NaCl洗脱多糖组分为D-半乳糖醛酸、D-果糖、D-葡萄糖,茎的水洗脱液多糖组分为D-甘露糖、D-葡萄糖,茎的NaCl洗脱多糖组分D-葡萄糖、D-半乳糖,叶的洗脱多糖组分为D-半乳糖醛酸和D-山梨糖。  相似文献

A water-soluble polysaccharide was isolated from an alkaline extract of the fruits of the ectomycorrhizal fungus, Astraeus hygrometricus. A possibility has been suggested of applying the EPS produced by Rhodococcus rhodochrous strain S-2 (S-2 EPS) to the bioremediation of oil-contaminated environments, because its addition, together with minerals, to oil-contaminated seawater resulted in emulsification of the oil, increased the degradation of polyaromatic hydrocarbons (PAH) of the oil, and led to the dominance of PAH-degrading marine bacteria. To understand the underlying principles of these phenomena, we determined the chemical structure of the sugar chain of S-2 EPS. The EPS was found to be composed of D-galactose, D-mannose, D-glucose, and D-glucuronic acid, in a molar ratio of 1:1:1:1. In addition, 0.8% (w/w) of octadecanoic acid and 2.7% (w/w) of hexadecanoic acid were also contained in its structure. Structural studies were carried out on an O-antigenic polysaccharide moiety derived from Porphyromonas circumdentaria NCTC 12469, a reference strain of Porphyromonas species. The polysaccharide chain was composed of D-glucose, D-galactose, N-acetyl-D-glucosamine, and N-acetyl-D-galactosamine in a molar ratio of 1:2:1:1. On the basis of results from 1H- and 13C-NMR spectroscopic analyses including COSY, TOCSY, and HMQC experiments together with results of Smith degradation, methylation analysis, and partial acid hydrolysis, it is concluded that the polysaccharide chain has a pentasaccharide repeating unit of -->6)-beta-D-Glcp-(1-->6)-beta-D-Galp-(1-->3)-beta-D-GlcpNAc-(1-->3)-beta-D-GalpNAc-(1-->. The immunoreaction between P. circumdentaria LPS and the corresponding antiserum was strongly inhibited by the pentasaccharide fragment (Glc-Gal-Gal-GlcNAc-GalNAc) isolated from partial acid hydrolysis of the above polysaccharide, suggestive of O-antigen specific antibodies in the used antiserum.  相似文献

短裙竹荪子实体经2%Na2CO3溶液提取,用蛋白酶法和Sevag法相结合除去蛋白,乙醇分级沉淀,级分3经DEAE-SephadexA-25柱层析纯化得到短裙竹蒸蒸荪多糖DdS3P.经测定该多糖为均一组分,分子量约为3.8×105,红外光谱呈现出典型的多糖吸收峰,含有α-型糖苷连接键,紫外扫描无核酸和蛋白质的特征吸收峰.纸层析和气相层析分析得知Dd-S3P含有D-葡萄糖(D-Glc)、D-甘露糖(D-Man)和D-木糖(D-Xyl),其摩尔比为1.83:1.00:1.21.经动物试验表明此多糖对小鼠S-180有一定的抑制作用,抑瘤率约31.3%.  相似文献

We investigated the structures of the exopolysaccharides (EPSs) produced by Streptococcus thermophilus SFi39 and SFi12. Both polymers were found to have molecular masses of greater than 2 x 10(6) Da. The SFi39 EPS consisted of D-glucose and D-galactose in a molar ratio of 1:1, whereas the SFi12 EPS was composed of D-galactose, L-rhamnose, and D-glucose in a molar ratio of 3:2:1. Methylation analysis of and nuclear magnetic resonance spectra recorded from the native polysaccharide, as well as oligosaccharides released by partial acid hydrolysis, allowed the complete structural determination of the SFi39 EPS, which consists of the following tetrasaccharide repeating unit: [formula: see text] Similar spectra recorded only from the native polysaccharide were sufficient to allow the structural determination of the SFi12 EPS, which consists of the following hexasaccharide repeating unit: [formula: see text] This study shows that the texturizing properties of different S. thermophilus ropy strains are based on the production of EPSs exhibiting chemical similarities but structural differences.  相似文献

A bacterium which was isolated from pond water and identified as Enterobacter cloacae produced a viscous extracellular polysaccharide when it was grown aerobically in a medium containing sucrose as a sole source of carbon. The maximum molecular weight of the polysaccharide was about 9.0 x 10(5). The polysaccharide was composed of fucose, galactose, glucose, and glucuronic acid in a molar ratio of 2:3:2:1, but the molecular weight and the molar ratio of the sugar component were different from those of the polysaccharide produced by the same species reported elsewhere.  相似文献

The specific capsular polysaccharide produced by Rhodococcus equi serotype 2 is a high-molecular-weight acidic polymer composed of D-glucose, D-mannose, D-glucuronic acid and 3-O-[(S)-1-carboxyethyl]-L-rhamnose in equimolar proportions. Structural analysis, employing a combination of chemical and n.m.r. techniques, established that the polysaccharide is composed of linear repeating tetrasaccharide units. (formula; see text) in which the beta-D-mannose residues carry O-acetyl groups at O-2 and O-3 to the extent of 1.7 mol equivalents. Unequivocal determination of the absolute chirality of the 3-O-[(S)-1-carboxyethyl]-alpha-L-rhamnose residues was achieved by chemical correlation with an authentic synthetic sample. The 1H and 13C-n.m.r. resonances of the native and O-deacetylated serotype 2 polysaccharides were fully assigned by homo- and heteronuclear chemical-shift correlation methods.  相似文献
