Gastrointestinal strongylosis are the dominant parasitic infections of sheep and goats. The successful control of these parasites cannot be done exclusively with anthelmintics, but the first step is an integrated program for environmental prophylaxis. The correct planning of the prophylaxis program has to be preceded by the analysis of the related problems: (1) the parasitological status of farm livestock; (2) knowledge of the farm management; (3) hydrogeological, pedological and climatic-environmental aspects. The environmental control strategies can be resumed as follows: avoiding animals from different farms to share the same pasture; avoiding animals of different age classes to graze together; parcel the pastures to permit a rational rotation; rotational grazing of pastures according to the seasonal development of parasites; stocking rate; young animals grazing ahead of the older animal ones; crop management practices (draining, ploughing, harrowing, scrub clearing, fertilizing, etc.). These measures make the habitat less suitable for the free-living stages of gastrointestinal strongyles, reducing the potential of infection of the same pastures. 相似文献

Effect of dietary fiber on microbial activity and microbial gas production in various regions of the gastrointestinal tract of pigs. 总被引:5,自引:4,他引:5

The microbial activity, composition of the gas phase, and gas production rates in the gastrointestinal tract of pigs fed either a low- or a high-fiber diet were investigated. Dense populations of culturable anaerobic bacteria, high ATP concentrations, and high adenylate energy charges were found for the last third of the small intestine, indicating that substantial microbial activity takes place in that portion of the gut. The highest microbial activity (highest bacterium counts, highest ATP concentration, high adenylate energy charge, and low pH) was found in the cecum and proximal colon. Greater microbial activity was found in the stomach and all segments of the hindgut in the pigs fed the high-fiber diet than in the pigs fed the low-fiber diet. Considerable amounts of O2 were found in the stomach (around 5%), while the content of O2 in gas samples taken from all other parts of the gastrointestinal tract was < 1%. The highest concentrations and highest production rates for H2 were found in the last third of the small intestine. No methane could be detected in the stomach or the small intestine. The rate of production and concentration of methane in the cecum and the proximal colon were low, followed by a steady increase in the successive segments of the hindgut. A very good correlation between in vivo and in vitro measurements of methane production was found. The amount of CH4 produced by pigs fed the low-fiber diet was 1.4 liters/day per animal. Torulopsis pintolopesii is an indigenous yeast that colonizes the secreting epithelia in the stomachs of mice and rats. A wild-type strain of this microbe was isolated and identified. To attempt to learn characteristics of the yeast that are advantageous to it in colonizing its natural habitat in vivo, we examined some aspects of its nutrition and energy-yielding metabolism and some environmental conditions that influence its growth in vitro. The yeast appeared to be limited in the compounds it can utilize as carbon and nitrogen sources. It grew best at 37 degrees C and did not grow at 23 or 43 degrees C. It grew optimally at neutral pH but could grow aerobically at pH values as low as 2.0 and anaerobically at pH values as low as 3.4. As assessed by measurements of growth rates and yield coefficients, it grew better aerobically than anaerobically. When grown aerobically, it had a cyanide-sensitive system for taking up O(2) and tested positively for cytochrome c oxidase activity. A petite mutant strain isolated from the wild-type strain had a growth rate and yield coefficient when incubated aerobically that were essentially the same as those of the wild-type parent grown anaerobically. Likewise similar to the wild-type parent grown anaerobically, the petite strain, though incubated aerobically, did not take up O(2). Yeast-free mice associated with either the wild-type or the petite mutant strain were colonized at essentially the same rates and to similar final population levels by both strains. The yeast's capacity to respire may be of little advantage to it in its natural environment. By contrast, its abilities to grow best at 37 degrees C and to grow at low pH values are undoubtedly advantageous characteristics in this respect. Considerable variation in counts of antibiotic-resistant enterobacteria in the ovine gastrointestinal tract was observed. The occurrence of ruminal and fecal isolates resistant to ampicillin (Ap), kanamycin (Km) and tetracycline (Tc) culminated in summer months, followed by rapid decline in subsequent months. Using PCR the tem1bla (Apr), aphA1 (Kmr) and tetB (Tcr) genes were found to be predominant. Sheep and lambs from 14 farms in southern Queensland and one from central New South Wales were surveyed to determine the prevalence of Shiga toxin-producing Escherichia coli (STEC). STEC, isolated from 45% of 144 sheep faeces collected on the farms and 36% of 72 lamb faeces from abattoir yards, were tested for the presence of genes encoding virulence factors. Most (64%) of the 117 STEC isolates contained Shiga toxin 1 and 2 genes, 22% contained those encoding Shiga toxin 1, and 14% contained genes encoding Shiga toxin 2. The genes encoding the E. coli attaching and effacing factor were present in 2.6% of STEC and 26% contained the enterohaemolysin gene. The isolates that contained the E. coli attaching and effacing gene were serotype O157:H. This study has shown that STEC are widely distributed in eastern Australian sheep and lambs and are shed in their faeces prior to slaughter. Thus, there is potential for contamination of carcasses and entry of STEC into the human food chain. 相似文献

The responses of luteinizing hormone (LH) and follicle stimulating hormone (FSH) secretion to acute dietary supplementation were studied in goats with different body weights. Ovariectomized Shiba goats (n = 11) were used and were maintained with a feed of 100% of their energy requirement. They were implanted subcutaneously with an oestradiol capsule and were divided into light (LBW; ≤24 kg, n = 6, mean ± S.D., 21.8 ± 2.7 kg) and heavy body weight (HBW; >24 kg, n = 5, mean ± S.D., 32.0 ± 6.3 kg) groups on the basis of their body weights at 8 days before the start of treatment. At the start of treatment (Day 1), the level of a feed changed to 250% of their energy requirement and this level was maintained for 7 days in both groups. Blood samples were collected daily from Day −7 to Day 7 for the analysis of FSH, glucose, and insulin profiles in plasma. Frequent blood samples were also collected at 10 min intervals for 6 h on Day 0, Day 3, and Day 7 for analysis of LH pulses. LH pulse frequency increased significantly on Day 3 as compared with that on Day 0 in both the HBW (7.4 ± 0.5 pulses/6 h vs. 6.2 ± 0.8 pulses/6 h, p < 0.05) and LBW (6.5 ± 0.8 pulses/6 h vs. 5.5 ± 0.5 pulses/6 h, p < 0.05) groups, whereas it decreased on Day 7 (HBW, 6.4 ± 0.9 pulses/6 h; LBW, 6.3 ± 1.6 pulses/6 h, p > 0.05 vs. Day 0). Plasma glucose and insulin concentrations increased temporarily from Day 2 to Day 4 and then decreased to the level before the start of dietary supplementation in both groups. There was no significant difference in the LH pulse frequency or daily concentrations of FSH, glucose, or insulin between the HBW and LBW groups throughout the experimental period. The present study indicated that acute dietary supplementation stimulates pulsatile LH secretion in parallel with a rise of blood glucose and insulin levels. However, the influence of body weight on these responses between light and heavy animals was not observed. 相似文献

Gastrointestinal (GI) parasitism are the most serious problem affecting sheep and goats worldwide. Economic losses caused by GI strongyles are related to decreased production, costs for treatment and prophylaxis and animal death. Effective control of internal parasites in small ruminants is one of the most difficult challenges encountered by veterinary in practice. For control of helminth parasites in grazing animals it is important knowledge of epidemiology of the parasite as it interacts with the host in a specific climatic, management and production environment. Sheep and goats are infected by many of the same GI nematodes, but feeding and management practices alter levels of parasitism between the two ruminants. The most commonly anthelmintics used for helminth control in small ruminants include three chemical groups: Imidazothiazoles/Pyrimidines (Im/Pm), Benzimidazoles/Probenzimidazoles (Bz/Pbz) and Macrocyclic Lactones (ML). The control of gastrointestinal strongyles infections in goats shows specific patterns. The use of anthelmintics in goats is based upon anthelmintics goats-suited dose rates taking into account the specific pharmacokinetics features of some anthelmintics in this species. Scrapie, an invariably fatal disease of sheep and goats, is a transmissible spongiform encephalopathy (TSE). The putative infectious agent is the host-encoded prion protein, PrP. The development of scrapie is closely linked to polymorphisms in the host PrP gene. The pathogenesis of most TSEs involves conversion of normal, cellular PrP into a protease-resistant, pathogenic isoform called PrPSc. The conversion to PrPSc involves change in secondary structure; it is impacts on these structural changes that may link polymorphisms to disease. Within the structured C-terminal part of PrP polymorphisms have been reported at 15 and 10 codons of the sheep and goat PrP genes respectively. Three polymorphisms in sheep are acutely linked to the occurrence of scrapie: A136V, R154H and Q171R/H. These generate five commonly observed alleles: ARQ, ARR, AHQ, ARH and VRQ. ARR and AHQ are associated with resistance; ARQ, ARH and VRQ are associated with susceptibility. There are subtle effects of specific allele pairings (genotypes). Generally, more susceptible genotypes have younger ages at death from scrapie. Different strains of scrapie occur which may attack genotypes differently. Different sheep breeds vary in the assortment of the five alleles that they predominantly encode. The reason for this variation is not known. Furthermore, certain genotypes may be susceptible to scrapie in some breeds and resistant in others. The explanation is not known, but may relate to different scrapie strains circulating in different breeds, or there may be effects of other genes which modulate the effect of PrP. 相似文献

The incidence of haemonchosis in sheep and goats in Sierra Leone showed a seasonal variation with a high peak in the dry season (October to January) and a low one from March to May. Mean relative densities were significantly higher in young hosts and showed two peaks, a high one from August to December and a low one from April to June while the mean relative densities of old hosts were low and exhibited an irregular seasonal pattern with no defined peaks. The peak seasons in young hosts coincided approximately with the dry and rainy seasons. Male hosts showed an overall higher but not a significant mean relative density than females but for most of the months mean relative densities of infection of female hosts were not significantly higher than those of males. The roles of climatic conditions and arrested development (hypobiosis) and self-cure and host age immunity on Haemonchus contortus worm burdens are discussed. 相似文献

We tested for cross‐species amplification of microsatellite loci located throughout the domestic sheep (Ovis aries) genome in two north American mountain ungulates (bighorn sheep, Ovis canadensis, and mountain goats, Oreamnos americanus). We identified 247 new polymorphic markers in bighorn sheep (≥ 3 alleles in one of two study populations) and 149 in mountain goats (≥ 2 alleles in a single study population) using 648 and 576 primer pairs, respectively. Our efforts increased the number of available polymorphic microsatellite markers to 327 for bighorn sheep and 180 for mountain goats. The average distance between successive polymorphic bighorn sheep and mountain goat markers inferred from the Australian domestic sheep genome linkage map (mean ± 1 SD) was 11.9 ± 9.2 and 15.8 ± 13.8 centimorgans, respectively. The development of genomic resources in these wildlife species enables future studies of the genetic architecture of trait variation. 相似文献

During the past decade it became evident that anaerobic cultivation-based approaches provides an incomplete picture of the microbial diversity in the GI tract, since at present only a minority of microbes can be obtained in culture. The application of molecular, mainly 16S ribosomal RNA (rRNA)-based approaches enables researchers to bypass the cultivation step and has proven its usefulness in studying the microbial composition in a variety of ecosystems, including the gastrointestinal (GI) tract. This critical review summarizes the impact of these culture-independent approaches on our knowledge of the ecology of the GI tract and provides directions for future studies which should emphasize function of specific strains, species and groups of microbes. 相似文献

The understanding of the dynamics and regulation of follicle development in the goat has increased in recent years due to the use of ultrasonography. Previous findings observed in other ruminants extend to this species such as: the wavelike pattern of follicular growth with waves emerging every 5-7 days; the presence of follicular dominance, particularly during wave 1 and the ovulatory wave; and the role of progesterone concentrations on follicular wave turnover. However, traditional hormone treatments for oestrus synchronisation and superovulatory programs remain essentially unmodified and without taking into account this new information. The use of long progestogen priming is largely unjustified. Short progestogen priming can be used successfully in both anoestrous and cycling goats (in this case associated with PGF2alpha), and also linked with timed artificial insemination. On the other hand, recent attempts to overcome the deleterious effect of the dominant follicle on superovulatory response using the emergence of the wave 1 have been successful. Overall, using ultrasonography a new field has been opened and surely successful output will be forthcoming in the next years to improve reproductive management of the goat, the fastest growing ruminant population in the world. 相似文献

The effects of different sources of dietary fibre on gastrointestinal tract (GIT) development and digesta pH were studied in 25-day-old broilers. There were four diets: a control diet low in fibre content (37.3 g neutral detergent fibre and 16.0 g acid detergent fibre/kg) and three additional diets in which 30 g/kg of cellulose (CEL), sugar beet pulp (SBP) or oat hulls (OH) were included at the expenses of sepiolite (a complex magnesium silicate clay). The relative weight (RW, %BW) of the GIT was greater with the SBP – than with the CEL – and the control diets. Also, gizzard RW was higher with the diet containing OH than with the diet containing CEL or the control diets, with the SBP diet being intermediate (P≤0.001). Fibre inclusion modified the pH of the GIT (P≤0.05) but the effects depended on the type of fibre. Crop pH was higher and proventriculus pH lower with SBP than with the other diets. However, gizzard pH was reduced with SBP and OH but not with CEL (P≤0.05). The pH of the digesta of the duodenum was similar for all diets but that of the jejunum, Meckel's diverticulum and ileum increased with SBP and decreased with CEL inclusion (P≤0.05). We concluded that dietary fibre affects the development and pH value of the different segments of GIT and that the effects differ according to the physico-chemical characteristics of the source of fibre used. 相似文献

The present study deals with the effect of hypergravity (2xg) on the pH and on the proteolytic activity in the digesta of the gastrointestinal tract of Japanese quails during intense growth. The birds were raised on a semisynthetic diet containing free amino acids (A) and a commercial diet (B). During days 35 till 40 post-hatching the quails were exposed to hypergravity (2xg) using a specially designed centrifuge. On days 40 (experimental group, 2xg) and 41 (control group, 1xg) the animals were sacrificed. The pH of the digesta in various segments of the gastrointestinal tract was measured by means of a semi-microelectrode. Total proteolytic activity was determined by means of azo-dye-modified proteins serving as general proteolytic substrates. Hypergravity leads in general to an alkalization of digesta in various parts of the gastrointestinal tract. In case of the gizzard and duodenum (diet A) and also in the distal jejunum (diet B) the differences are significant. With both diets, hypergravity leads to a considerable decrease in the total proteolytic activity. The reduction is most expressed in the duodenum and jejunum. Changes in the pH of digesta compensate for the decrease in the proteolytic activity. This may explain why hypergravity per se does not seem to impair growth of the Japanese quails. 相似文献

Oxidation-reduction potentials (Eh), pH and rH in the gastrointestinal tract were measured in six goats and two sheep fed on ground barley and hay, with or without the addition of urea. Each ration was supplied for three weeks. The animals were slaughtered after morning feeding, the contents of relevant parts of the gastrointestinal tract were sampled, pH and Eh values were measured and rH values calculated. The range of the oxidation-reduction potential was rather extensive, from -300 to +186 mV. The variability of rH values was smaller, being between 4.6 and 12.9, except for three values. The following linear relationship holds for findings of Eh and pH: Eh (mV) = 292-69.9 pH with a correlation coefficient of r = 0.84. In those parts of the gastrointestinal tract, where the fermentation process occurs (the rumen, caecum and colon), the Eh and rH values are lower than those in the abomasum and duodenum. Urea addition has no effect on the oxidation-reduction equilibrium. 相似文献
