Pathophysiological and pharmacological concentrations of tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) in the cerebrospinal fluid (CSF) induce anorexia in normal rats. Obesity in humans and rodents is associated with increased TNF-α messenger RNA and protein levels in various cell types. This suggests that obese individuals may have differential regulation of cytokine production and dissimilar responsiveness to cytokines. In the present study, we investigated the effects of the intracerebroventricular (ICV) microinfusion of TNF-α (50, 100, and 500 ng/rat), IL-1β (1.0, 4.0, and 8.0 ng), and TNF-α (100 ng) plus IL-1β (1.0 ng) on obese (fa/fa) and lean (Fa/Fa) Zucker rats. The results show that: TNF-α and IL-1β, and the concomitant administration of TNF-a and IL-ip decreased the short-term (4 hours), nighttime (12 hours), and total daily food intakes in obese and lean rats; IL-1β was more potent relative to TNF-α; obese rats showed greater responsiveness to IL-1β: 8.0 ng IL-1β, for example, decreased the 12-hour food intake by 52% in obese and 22% in lean rats. On the other hand, obese and lean rats did not exhibit a significantly different responsiveness to the anorexia induced by 50,100, or 500 ng TNF-α at the 4-hour period; and the concomitant ICV administration of TNF-α and IL-1β induced anorexia with additive (4-hour period) or synergistic (12-hour and 24-hour periods) effects in obese rats. The effect of TNF-α plus IL-1β in lean rats was greater than additive for the 12-hour and 24-hour periods. The difference in suppression of total daily food intake by TNF-α plus IL-1β in obese (-43%) versus lean (-23%) rats was significantly different (p<0.01). The results show that obese (fa/fa) and lean (Fa/Fa) Zucker rats have differential responsiveness to the ICV microinfusion of two different classes of cytokines.  相似文献

The purpose of this research is to examine the heart rate responses and the perceived exertion in college students during scootering, and to examine if scootering possibly makes heart rate increase up to the level that can contribute to maintaining or developing cardiorespiratory fitness. Five male students (20-23 yrs) participated in this research, mainly assigned to scooter on an official 400 m-tartan track. Each session of scootering was six minutes. Each subject did three sessions of scootering at different speeds, slow, ordinary, and very fast. During the scootering, heart rate was measured using a Polar Vantage XL. Immediately after each session, the subjects were questioned about their perceived exertion. To evaluate heart rate during scootering on the track, maximal heart rate was measured in advance with graded maximal tests. In each speed in the track trial, the mean heart rates and the standard deviations were 106 +/- 5.9, 129 +/- 4.2, and 179 +/- 13.7 beats/min respectively. They correspond to 54.0 +/- 4.2%, 65.8 +/- 4.2%, and 91.2 +/- 5.5% of the maximal heart rate respectively. The mean and standard deviation of perceived exertion based on Borg's scale in each scootering session were 7.2 +/- 0.45, 10.2 +/- 1.10, and 16.6 +/- 2.79 respectively. Conclusively, at ordinary speed, the heart rates of the college students on a tartan track were situated around the level of the lower boundary which the American College of Sports Medicine recommended to develop and maintain cardiorespiratory fitness for apparently healthy people. If people have places to ride a scooter briskly, their heart rate could rise above the minimum level.  相似文献

The purpose of this investigation was to evaluate the effect of passive smoke inhalation on submaximal and maximal exercise performance. Eight female subjects ran on a motor driven treadmill for 20 min at 70% VO2max followed by an incremental change in grade until maximal work capacity was obtained. Each subject completed the exercise trial with and without the presence of residual cigarette smoke. Compared to the smokeless trials, the passive inhalation of smoke significantly reduced maximal oxygen uptake by 0.25 l X min-1 and time to exhaustion by 2.1 min. The presence of sidestream smoke also elevated maximal R value (1.01 vs 0.93), maximal blood lactate (6.8 vs 5.5 mM), and ratings of perceived exertion (17.4 vs 16.5 units). Passive inhalation of smoke during submaximal exercise significantly elevated the CO2 output (1.68 vs 1.58 l X min-1), R values (0.91 vs 0.86), heart rate (178 vs 172 bts X min-1) and rating of perceived exertion (13.8 vs 11.8 units). These findings suggest that passive inhalation of sidestream smoke adversely affects exercise performance.  相似文献

We evaluated the effect of different types of sprint interval sessions on the balance between anabolic and catabolic hormones and circulating inflammatory cytokines. Twelve healthy elite junior handball players (17-25 years) participated in the study. Exercise consisted of increasing distance (100 m, 200 m, 300 m, 400 m) and decreasing distance (400 m, 300 m, 200 m, 100 m) sprint interval runs on a treadmill (at random order), at a constant work rate of 80% of the personal maximal speed (calculated from the maximal speed of a 100 m run). The total rest period between the runs in the different interval sessions were similar. Blood samples were collected before, after each run, and after 1-hour recovery. Both types of sprint interval trainings led to a significant (p < 0.05) increase in lactate and the anabolic factors growth hormone, insulin-like growth factor-I (IGF-I), IGF binding protein-3 (IGFBP-3), and testosterone levels. Both types of sprint interval sessions led to a significant (p < 0.05) increase in the circulating pro- and anti-inflammatory mediators IL-1, IL-6, and IL1ra. IL-6 remained elevated in both sessions after 1-hour recovery. Area under the curve was significantly greater (p < 0.05) for lactate and growth hormone (GH) in the decreasing distance session. In contrast, rate of perceived exertion was higher in the increasing distance session, but this difference was not statistically significant (p = 0.07). Changes in anabolic-catabolic hormones and inflammatory mediators can be used to gauge the training intensity of anaerobic-type exercise. Changes in the GH-IGF-I axis and testosterone level suggest exercise-related anabolic adaptations. Increases in inflammatory mediators may indicate their important role in muscle tissue repair after anaerobic exercise. The decreasing distance interval was associated with a greater metabolic (lactate) and anabolic (GH) response but not with a higher rate of perceived exertion. Coaches and athletes should be aware of these differences, and as a result, of a need for specific recovery adaptations after different interval training protocols.  相似文献

目的:探讨慢性阻塞性肺疾病(COPD)螺旋CT吸气相和呼气相扫描体积变化与血清细胞因子水平的内在关系。方法:记录46例符合纳入标准的COPD患者的临床资料、缓解期的肺功能(FEV1%),采用西门子16排螺旋CT对患者进行吸气相和呼气相扫描,计算吸气前后肺体积变化百分比。采用酶联免疫吸附法分别测定急性发作期和缓解期血浆可溶性细胞间粘附分子-1((plasma soluble intercellular adhesion molecule- 1, slCAM- 1)和TNF-α(tumornecrosisfactor-α)变化水平。对比分析肺功能、肺体积变化与细胞因手的相关性。结果:双相CT扫描肺体积变化百分比为24.27±9.82%,FEVl%平均值为53.50±12.21%,血清slCAM。1和TNF-α表达水平为166.58±25.15ng/ml、22.05±4.36pg/ml。COPD肺功能、体积变化百分比与细胞因子表达存在负相关性.肺功能与肺体积变化存在正相关性(P〈O.05)。结论:螺旋CT双相扫描能准确反映COPD肺体积的顺应性变化,与slCAM-1和TNF-α表迭水平都可作为判断COPD肺功能及疗效的重要指标。  相似文献

The purpose of this study was to determine the effects of varying intensities of exercise in normoxic and hypoxic environments on selected immune regulation and metabolic responses. Using a within-subjects design, subjects performed maximal tests on a cycle ergometer in both normoxic (PiO2 = 20.94%) and hypoxic (PiO2 = 14.65%) environments to determine [latin capital V with dot above]O2max. On separate occasions, subjects then performed four randomly assigned, 1-hour exercise bouts on a cycle ergometer (two each in normoxic and hypoxic environments). The hypoxic environment was created by reducing the O2 concentration of inspired air using a commercially available hypoxic chamber. The intensities for the exercise bouts were predetermined as 40 and 60% of their normoxic [latin capital V with dot above]O2max for the normoxic exercise bouts and as 40 and 60% of their hypoxic [latin capital V with dot above]O2max for the hypoxic exercise bouts. Blood samples were collected preexercise, postexercise, 15 minutes postexercise, 2 hours postexercise, and 24 hours postexercise for the determination of interleukin-1 (IL-1), tumor necrosis factor-[alpha] (TNF-[alpha]), glucose, glycerol, free fatty acids, epinephrine, norepinephrine, and cortisol. There were no significant differences (p < 0.05) between condition or intensity for IL-1 or TNF-[alpha]. Significant differences (p < 0.05) between intensities were demonstrated for epinephrine, norepinephrine, and cortisol (p < 0.05). A significant difference was identified between normoxic and hypoxic environments with respect to nonesterifed fatty acids (0.45 +/- 0.37 vs. 0.58 +/- 0.31 mEq x L-1, respectively; p = 0.012). During prolonged exercise at 40 and 60% of their respective [latin capital V with dot above]O2max values, hypoxia did not seem to dramatically alter the response of the selected immune system or metabolic markers. Exercise training that uses acute hypoxic environments does not adversely affect immune regulation system status and may be beneficial for those individuals looking to increase endurance performance.  相似文献

The aim of this study was to compare selected acute cardiorespiratory and metabolic effects of exercise on a Fitness Flyer (FF) aerobic rider to those of treadmill (TM) running. Fourteen women, aged 23-35 years, performed incremental exercise tests to exhaustion on the TM and FF. Ratings of perceived exertion (RPE), heart rate (HR), minute ventilation (VE), VO2, and ventilatory equivalent (VEq) were compared in each subject during each phase of the exercise protocols, and blood lactate concentrations were measured before and 2-3 minutes after the exercise tests on the 2 modalities. Peak VO2 was higher (p < 0.05) on the TM than on the FF. Mean submaximal HR and VEq at a given VO2 was, however, higher on the FF than on the TM (p < 0.05). Maximum mean energy expenditure on the FF corresponded with mean energy expenditure on the TM at 8 km.h(-1) at an 18% gradient. Posttest blood lactate concentrations and RPE were higher on the FF than on the TM (p < 0.05). The results indicate that although exercising on an FF elicits less maximal cardiorespiratory response than does TM running, the FF may be better suited to developing local muscle endurance in the thigh muscles.  相似文献

目的:探讨芪蝎活血通络汤对局灶性脑缺血再灌注损伤模型大鼠神经功能、氧化应激及炎症因子的影响。方法:50只SD大鼠随机选取40只采用线栓法构建脑缺血再灌注模型,其中36只成功,4只死亡。随机数字表法将36只脑缺血再灌注模型大鼠分为模型组、低剂量组(芪蝎活血通络汤2.0 mg/kg灌胃处理)、中剂量组(芪蝎活血通络汤4.0 mg/kg灌胃处理)和高剂量组(芪蝎活血通络汤8.0 mg/kg灌胃处理),每组9只,剩余10只大鼠仅切开皮肤分离和夹闭血管(对照组)。建模后芪蝎活血通络汤各剂量组给予相应剂量灌胃,对照组和模型组给予同剂量生理盐水灌胃,持续4周。用药4周后测评各组大鼠神经功能缺损程度、双侧贴纸去除时间、平衡木过杆时间,并测定各组大鼠脑组织中含水量、脑梗死面积以及氧化应激[过氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、丙二醛(MDA)]和炎症因子[白细胞介素-1β(IL-1β),白细胞介素-6(IL-6),肿瘤坏死因子-α(TNF-α)]指标。结果:与对照组比较,模型组大鼠m NSS评分、脑组织含水量、MDA含量、IL-6、IL-1β、TNF-α水平升高(P<0.05),双侧贴纸去除时间、平衡木过杆时间延长(P<0.05),脑梗死面积增大(P<0.05),SOD、GSH-Px、CAT活性降低(P<0.05);与模型组比较,芪蝎活血通络汤各剂量组大鼠m NSS评分、脑组织含水量、MDA含量、IL-6、IL-1β、TNF-α水平降低(P<0.05),双侧贴纸去除时间、平衡木过杆时间缩短(P<0.05),脑梗死面积缩小(P<0.05),SOD、GSH-Px、CAT活性升高(P<0.05);与低剂量组比较,中、高剂量组大鼠m NSS评分降低(P<0.05),双侧贴纸去除时间、平衡木过杆时间缩短(P<0.05);高剂量组大鼠脑梗死面积、脑组织MDA、IL-6、IL-1β、TNF-α低于低剂量组(P<0.05),SOD、GSH-Px、CAT高于低剂量组(P<0.05)。结论:芪蝎活血通络汤可降低大鼠脑缺血再灌注损伤,改善神经功能,其治疗作用可能与抗氧化,抗炎作用有关,8.0 mg/kg剂量效果最显著。  相似文献

The purpose of this study was to investigate the covariance between perceived exertion (recorded using Borg's category-ratio scale CR-10) and the relative oxygen uptake, and lactate and ammonia concentrations in blood from a peripheral vein. Ratings of perceived exertion (RPE) at 25%, 50%, 75% and 90% maximal oxygen uptake and lactate and ammonia concentrations were compared in well-trained women distance runners (n = 22) and untrained women (n = 10). Ammonia concentrations in peripheral venous blood were significantly correlated with RPE (P less than 0.05), both in the trained and untrained women. Differences between the trained and untrained subjects occurred when the ammonia concentration increased to 148 mumol.l-1 in both groups investigated; similarly, the mean RPE correlated significantly with the lactate concentration (P less than 0.05), both in the trained and untrained women and there was a difference in RPE between groups when lactate concentration in the blood had risen to 4.4 mmol.l-1. It would seem that the correlation of blood ammonia and lactate concentrations with RPE during exercise could be a useful indicator of the development of fatigue.  相似文献

目的探讨膝关节炎患者关节置换术后细菌感染严重程度与IL-1β、TNF-α、IL-6水平的相关性。方法选取2016年8月至2018年8月于我院进行治疗的70例膝关节炎关节置换术后细菌感染患者作为试验组,参照Michel Lequesen推荐的膝关节炎严重性判断标准将膝关节炎关节置换术后细菌感染患者分为极严重组、非常严重组、严重组、中度组和轻度组。选取同期来我院体检中心进行体检的健康者50例为对照组。采用全自动生化免疫分析仪测定血清IL-1β、IL-6及TNF-α水平。结果试验组患者血清IL-1β、TNF-α、IL-6水平均明显高于对照组(均P<0.05)。极严重组患者血清IL-1β、TNF-α、IL-6水平显著高于非常严重组、严重组、中度组和轻度组(均P<0.05)。非常严重组患者血清IL-1β、TNF-α、IL-6水平显著高于严重组、中度组和轻度组(均P<0.05)。严重组患者血清IL-1β、TNF-α、IL-6水平显著高于中度组和轻度组(均P<0.05)。IL-1β、TNF-α、IL-6水平与膝关节炎关节置换术后细菌感染严重程度呈显著正相关(均P<0.05)。结论膝关节炎关节置换术后细菌感染患者血清IL-1β、TNF-α、IL-6水平明显升高,并且病情越严重,IL-1β、TNF-α、IL-6水平越高。  相似文献

The present study investigated the effect of chronic neck muscle pain (defined as trapezius myalgia) on neck/shoulder muscle function during concentric, eccentric and static contraction. Forty-two female office workers with trapezius myalgia (MYA) and 20 healthy matched controls (CON) participated. Isokinetic (-60, 60 and 180 degrees s(-1)) and static maximal voluntary shoulder abductions were performed in a Biodex dynamometer, and electromyography (EMG) obtained in the trapezius and deltoideus muscles. Muscle thickness in the trapezius was measured with ultrasound. Pain and perceived exertion were registered before and after the dynamometer test. The main findings were that shoulder abduction torque (at -60 and 60 degrees s(-1)) and trapezius EMG amplitude (at -60, 0 and 60 degrees s(-1)) were significantly lower in MYA compared with CON (p<0.001-0.05). Deltoideus EMG and trapezius muscle thickness were not significantly different between the groups. While perceived exertion increased in both groups in response to the test (p<0.0001), pain increased in MYA only (p<0.0001). In conclusion, having trapezius myalgia was associated with decreased strength capacity and lowered activity of the painful trapezius muscle. The most consistent differences-in terms of both torque and EMG-were found during slow concentric and eccentric contractions. Activity of the synergistic pain free deltoideus muscle was not significantly lower, indicating specific inhibitory feedback of the painful trapezius muscle only. Parallel increase in pain and perceived exertion among MYA were observed in response to the maximal contractions, emphasizing that heavy physical exertion provokes pain increase only in conditions of myalgia.  相似文献

Heat stroke is a life threatening illness characterized by a core body temperature of >40 °C, delirium and convulsions, and often results in multi-organ dysfunction, due to the release of endotoxin through the intestinal wall into the circulation. While playing a major role in the gastrointestinal tract permeability changes seen in Crohn's disease, it is not clear whether tumor necrosis factor alpha (TNF-α) mediates the increase in intestinal permeability and the release of endotoxin into the circulation in heat stroke. The aim of the present study was to determine the acute effects of a single dose of TNF-α antibody on gut permeability in rats during heat stress. Fifty-five Sprague-Dawley rats (28 male and 27 female) were treated with either saline or infliximab (a monoclonal antibody to TNF-α), anesthetized with pentobarbitone (50 mg kg−1) and then exposed to either normothermic conditions or an ambient temperature of between 41 and 42 °C for 70 min. Fluorescent isothiocyanate labeled dextrans (FITC-dextrans) were administered intragastrically as a marker of intestinal permeability. Liver enzymes, endotoxin and TNF-α were analyzed in the blood. Exposure to a heat stress significantly increased intestinal permeability to FITC-dextrans compared to the controls (P<0.05). Infliximab did not have an effect on the intestinal permeability to the FITC-dextrans. Heat stress had no significant effect on liver enzymes or endotoxin concentration versus controls (P>0.05). TNF-α was not detectable in any of the samples. TNF-α did not mediate the release of endotoxin into the circulation after an acute bout of heat stroke.  相似文献

目的探讨酪酸梭菌活菌散辅助治疗手足口病(HFMD)对患儿血清超敏C反应蛋白(hs—CRP)和肿瘤坏死因子(TNF)-α水平的影响。方法选取的HFMD患儿70例,随机将患儿分为观察组和对照组,每组35例。两组患儿均予以利巴韦林、重组人干扰素ctlb及退热、营养支持等常规治疗。观察组患儿在此基础上加用酪酸梭菌活菌散,其中≤1岁,0.5g/次,2次/d;〉1岁,1.0g/次,2次/d,温开水冲服。对照组患儿除不使用酪酸梭菌活菌散外余治疗同观察组。观察两组患儿治疗前和治疗3d后血清hs—CRP和TNF—α水平的变化,进行临床疗效的观察。结果治疗3d后,两组患儿血清hs-CRP和TNF—α水平均较前明显下降(P〈0.05或P〈0.01),且观察组下降的幅度较对照组更明显(P〈0.05)。观察组患儿治疗后的总有效率(94.29%)明显高于对照组(77.14%)(x^2=4.20,P〈0.05)。结论酪酸梭菌活菌散辅助治疗对HFMD患儿的效果较好,能降低血清hs—CRP和TNF—α水平,从而抑制患儿的免疫性炎症反应过程,达到治疗的目的。  相似文献

Although Tumor Necrosis Factor alpha (TNFα) is used as a preconditioning mimetic in vitro, its role in ischaemic preconditioning (IPC) has not been clearly defined. Here, we propose to use an in vivo model (that takes into account the activation of leukocytes which may affect levels of TNFα) to demonstrate that i) TNFα acts as a trigger in IPC and ii) the dose-dependent nature of this cardioprotective effect of TNFα. Male Wistar rats were subjected to 30 min of left coronary artery occlusion (index ischaemia), followed by 24 h reperfusion. In the presence or absence of a soluble TNFα receptor (sTNFα-R), preconditioning was induced by 3 cycles of ischaemia (3 min)/reperfusion (5 min) (IPC) or various doses (0.05-4 μg/kg) of exogenous TNFα. Following 24 h reperfusion, infarct size (IS, expressed as % of the area at risk (AAR)) was assessed. Tissue levels of TNFα from the AAR, following IPC and TNFα stimulus were determined using Western Blot. IPC caused decrease in IS (4.5 ± 1.3% vs 30.8 ± 4.3% in ischaemic rats; P < 0.001) and increase of TNFα levels following the IPC stimulus. The protective effect of IPC was abrogated in the presence of the sTNFα-R. In addition, exogenous TNFα dose-dependently reduced IS with maximal protection at a dose of 0.1 μg/kg (IS = 12.6%, P < 0.01 vs ischaemic). In conclusion our data provide strong evidence for a role of TNFα during the trigger phase of IPC. In addition, exogenous TNFα mimics IPC by providing a dose-dependent cardioprotective effect against ischaemia-reperfusion injury in vivo.  相似文献

The etiology of colon cancer is a complex phenomenon that involves both genetic and environmental factors. However, only about 20% have a familial basis with the largest fraction being attributed to environmental causes that can lead to chronic inflammation. While the link between inflammation and colon cancer is well established, the temporal sequence of the inflammatory response in relation to tumorigenesis has not been characterized. We examined the timing and magnitude of the intestinal inflammatory cytokine response in relation to tumorigenesis in the ApcMin/+ mouse. ApcMin/+ mice and wildtype mice were sacrificed at one of 4 time-points: 8, 12, 16, and 20 weeks of age. Intestinal tissue was analyzed for polyp burden (sections 1, 4 and 5) and mRNA expression and protein concentration of MCP-1, IL-1β, IL-6 and TNF-α (sections 2 and 3). The results show that polyp burden was increased at 12, 16 and 20 weeks compared to 8 weeks (P < 0.05). Gene expression (mRNA) of MCP-1, IL-1β, IL-6 and TNF-α was increased in sections 2 and 3 starting at week 12 (P < 0.05), with further increases in MCP-1, IL-1β and IL-6 at 16 weeks (P < 0.05). Protein concentration for these cytokines followed a similar pattern in section 3. Similarly, circulating MCP-1 was increased at 12 weeks (P < 0.05) and then again at 20 weeks (P < 0.05). In general, overall polyp number and abundance of large polyps were significantly correlated with the inflammatory cytokine response providing further support for a relationship between polyp progression and these markers. These data confirm the association between intestinal cytokines and tumorigenesis in the ApcMin/+ mouse and provide new information on the timing and magnitude of this response in relation to polyp development. These findings may lead to the development of inflammatory mediators as important biomarkers for colon cancer progression. Further, these data may be relevant in the design of future investigations of therapeutic interventions to effectively target inflammatory processes in rodent models.  相似文献

Repeated ethanol exposure and withdrawal in mice increases voluntary drinking and represents an animal model of physical dependence. We examined time- and brain region-dependent changes in gene coexpression networks in amygdala (AMY), nucleus accumbens (NAC), prefrontal cortex (PFC), and liver after four weekly cycles of chronic intermittent ethanol (CIE) vapor exposure in C57BL/6J mice. Microarrays were used to compare gene expression profiles at 0-, 8-, and 120-hours following the last ethanol exposure. Each brain region exhibited a large number of differentially expressed genes (2,000-3,000) at the 0- and 8-hour time points, but fewer changes were detected at the 120-hour time point (400-600). Within each region, there was little gene overlap across time (~20%). All brain regions were significantly enriched with differentially expressed immune-related genes at the 8-hour time point. Weighted gene correlation network analysis identified modules that were highly enriched with differentially expressed genes at the 0- and 8-hour time points with virtually no enrichment at 120 hours. Modules enriched for both ethanol-responsive and cell-specific genes were identified in each brain region. These results indicate that chronic alcohol exposure causes global ‘rewiring‘ of coexpression systems involving glial and immune signaling as well as neuronal genes.  相似文献
