  • 1.1. Since glucose is one of the main energetic substrates for general metabolic processes in crustaceans, analysis of carbohydrate levels can furnish information on the energy metabolism of intact animals during osmoregulation.
  • 2.2. Different groups of Chasmagnathus granulata were transferred to different salinities (0 and 40%), and the glucose and glycogen concentrations in blood, gills, muscle and hepatopancreas were determined at the beginning of the experiment and 24, 72, 168 and 360 hr after the salinity changes.
  • 3.3. Differences in tissues carbohydrate levels were observed between summer and winter, that reflected differences in reserve mobilization.
  • 4.4. In the summer, hypo- and hyperosmotic shocks induced an increase in carbohydrate levels in almost all tissues studied, indicating gluconeogenesis.
  • 5.5. In the winter, a carbohydrate mobilization occurred only in the gills and hepatopancreas after both osmotic shocks.
  • 6.6. Thus, the substrate reserve used for energy production required for osmoregulation seems to be dependent on the season and tissues.

  • 1.1. Acid and alkaline phosphatase activities of eight different snake venoms were determined quantitatively by using synthetic substrates, o-carboxyphenylphosphate and p-nitrophenylphosphate respectively.
  • 2.2. It was found that most of Elapidae venoms investigated had both acid and alkaline phosphatase activities.
  • 3.3. Three Crotalidae venoms investigated did not show any alkaline phosphatase activity.
  • 4.4. The strength of venom acid phosphatase activity is as follows: Agkistroden acutus > Naja haje > Naja naja samarensis > Naja naja atra > Naja melanoleuca.

  • 6.6. Acid phosphatase activity of all the Elapidae venoms was inhibited completely by fluoride. The alkaline phosphatase activity of Elapidae venoms was not inhibited by fluoride either using p-nitrophenylphosphate or o-carboxyphenylphosphate.
  • 7.7. The acid phosphatase of all the Elapidae venoms was not inhibited by zinc ion. However, most of the venom alkaline phosphatases were inhibited by zinc ion.
  • 8.8. Ethylenediaminetetraacetic acid (EDTA) had inhibitory action on venom phosphatase activity. However, tris-(hydroxymethyl)-aminoethane had a counter effect on the inhibitory action of EDTA.

  • 1.1. In intact (control) crabs (Carcinus maenas) and crayfish (Orconectes limosus) a significant (P < 0.01) increase in both glucose and lactate concentrations in the blood was observed after exposure to air. Such changes were not observed in either eyestalk-less or eyestalk-less and saline injected animals (P > 0.05).
  • 2.2. Injections of Crustacean hyperglycemie hormone (CHH) into eyestalk-less animals before exposure to air were able to reverse the effects of eyestalk ablation, i.e., significant increases (P < 0.01) in blood glucose and lactate could again be observed.
  • 3.3. Significant hyperglycemia (P < 0.01), but no changes in lactate concentration (P > 0.05), was observed after injection of CHH in eyestalk-less submerged animals.
  • 4.4. These results suggest that the increase in glycolysis after air exposure is facilitated by CHH, possibly by increased substrate availability due to glycogen degradation.

  • 1.1. An artificial diet, consisting of a dry aggregate of 59 chemical substances, was used to assess the requirements of the sea slater Ligia pallasii for vitamins, carbohydrates, fatty acids, cholesterol and minerals.
  • 2.2. Good growth and survival of L. pallasii was obtained on the diet, comparable to that on seaweeds and to that shown by a field population.
  • 3.3. No dietary requirements for vitamins, fatty acids or cholesterol were shown for periods of 40 weeks or more for L. pallasii.
  • 4.4. Carbohydrates were shown to be required by L. pallasii in its diet, in the order: starch, lactose > maltose, glucose > sucrose, cellulose.
  • 5.5. Dietary requirements for minerals were, in order: calcium, magnesium, phosphorus > copper, nickel, zinc > iron, manganese, sulphur > iodine, silicon.
  • 6.6. The results are discussed in relation to the role of gut bacteria in supplying required nutrients to their isopod hosts and the enhancement of this process through coprophagic behaviour.

  • 2.2. Both the anterior and posterior gills contain cAMP-dependent protein kinase and endogenous protein substrate for phosphorylation.
  • 3.3. Phosphorylation occurs in both “particulate” and “soluble” subcellular fractions but its stimulation by cAMP is restricted to the “soluble” fraction.
  • 4.4. serotonin (5-HT) and dopamine receptors are present only in the “light particulate” fraction isolated from the posterior gills.
  • 1.(a) Serotonin and dopamine have no effect on the phosphorylation observed in a subcellular fraction alone.
  • 2.(b) Activation of the phosphorylation by serotonin and dopamine is found when the soluble fraction (source of cAMP-dependent protein kinase) is added to the fraction P3 from the posterior gills.

  • 4.(d) Cyproheptadine, a serotonin receptor antagonist, inhibits the 5-HT dependent increase in phosphorylation.
  • 5.(e) The dopamine receptor antagonist, chlorpromazine, inhibits dopamine-stimulated phosphorylation.
  • 6.5. Ouabain mimics the effect of cyproheptadine on the serotonin-stimulated phosphorylation found in the posterior gills.

  • 1.1. Semaphore crabs (Heloecius cordiformis) are active in air at low tide. Their branchial chambers are lined with a vascular epithelium and are expanded above the gills (five pairs) to form air cavities which could function as lungs. Water is continuously circulated over the gills.
  • 2.2. The relative contribution made by the gills and lungs to gas exchange in semaphore crabs active in air and circulating branchial water, was determined by measuring oxygen consumption (at 25°C) in crabs with and without branchial water, and in crabs with their lungs subsequently occluded.
  • 3.3. Activity levels and VO2 were unaffected by the absence of branchial water.
  • 4.4. With their lungs occluded, VO2 dropped (on average) by 61% in crabs with branchial water (i.e. gills still functional) and by 81% in crabs without branchial water (gill function impaired).
  • 5.5. It is concluded that semaphore crabs are obligate air breathers while active on land, despite carrying water within their branchial chambers. Lung development and gill reduction in land crabs is discussed briefly in relation to “terrestriality”.

  • 1.1. Lipid, glucose and glycogen concentrations were measured in different tissues of the crab Chasmagnathus granulata during emersion.
  • 2.2. After 6 hr of emersion no reduction in the total amount of carbohydrates was found to occur, suggesting that a general metabolic arrest was taking place.
  • 3.3. A transitory increase in haemolymphatic glucose and lipid levels was observed. Possible causes are therefore discussed in relation to changes in the flux of substrates for energy production.

  • 1.1. The purpose of this study was to determine whether biochemical changes of skeletal muscle that occur as a result of exercise in young rats persist into adulthood.
  • 2.2. Littermates (10 days old) were assigned to a 3, 6 and 12 week control or training group. In addition, a rest-exercise group (R-E) and exercise-rest (E-R) group were included.
  • 3.3. The rest-exercise and exercise-rest rats were maintained for the 12 weeks with the first 6 weeks being either rest or exercise and the condition reversed during the last 6 weeks of the experiment.
  • 4.4. Myofibril ATPase activity of rat plantaris increased from the 10d to 12 week animals (P < 0.05). As anticipated, training resulted in a lowered activity at 6 and 12 weeks compared to controls.
  • 5.5. The Ca2+ uptake and Ca2+-ATPase activity of the sarcoplasmic reticulum followed a similar pattern.
  • 6.6. With regard to the exercise-rest rats, the myofibril and SR ATPase activities at 12 weeks were comparable to the 12 weeks control rats.
  • 7.7. The rest-exercise group approximated the 12 week training group with regard to myofibril and SR ATPase activities (P > 0.05).
  • 8.8. The results suggest that the training adaptations that occur during development of skeletal muscle return to normal, when training ceases in the adult rat.
  • 9.9. Furthermore, animals that started to train prior to puberty do not have a greater capacity to adapt than animals which initiated training during adulthood.

  • 1.1. The levels of adenylates, arginine phosphate, arginine and various other metabolites related to anaerobic metabolism were determined in foot and gills of Cardium edule during anoxia (incubation in oxygen-free sea water) up to 48 hr and after electrical stimulation of the foot muscle.
  • 2.2. In the foot the phosphagen was depleted quite rapidly in the first hours of anoxia. Free arginine levels rose while there was little octopine accumulation. d-Lactate appeared to be the predominant end product. Alanine and succinate were also produced and the concentration of aspartate decreased, suggesting this metabolite as precursor for succinate. In the gills, where no arginine phosphate was present, succinate production was higher than in the foot and exceeded lactate formation.
  • 3.3. Only a small decline in ATP occurred during anoxia and consequently, only moderate changes in the energy charge were observed in both tissues.
  • 4.4. Electrical stimulation of the foot also resulted in the breakdown of arginine phosphate and formation of mainly lactate and not octopine.
  • 5.5. Total ATP consumption during anoxia has been calculated to be more rapid during the first hours. The ATP consumption rate was reduced by a factor of 7.8 during 48 hr of anoxia.
  • 6.6. During the first 4 hr of anoxia about 60% of the total consumed energy (as ATP equivalents) was provided by the phosphagen, while later glycolysis contributed about 80%. During electrical stimulation about 75% of the energy supplied was derived from the phosphagen.
  • 7.7. A comparison of the C. edule results with the data during anoxia and exercise of the jumping cockle is made.

  • 1.1. Partially purified rat liver ornithine decarboxylase is inhibited by several diamines including putrescine, 1,3-diaminopropane, cadaverine and p-phenylenediamine.
  • 2.2. The inhibition is dependent on pH, being strong at pH above 8 and negligible below pH 6.5.
  • 3.3. The kinetic study of the inhibition showed that while the aromatic diamine behaved as a simple competitive inhibitor, the aliphatic diamines presented a more complex pattern of inhibition in which two molecules of inhibitor might bind to the enzyme active site.
  • 4.4. The KI values for the different inhibitors were calculated and the degree of affinity for the enzyme was p-phenylenediamine > putrescine > cadaverine > 1,3-diaminopropane.
  • 5.5. A molecular mechanism explaining how one or two molecules of inhibitor can bind to the enzyme is proposed.

  • 1.1. We used protein gel-electrophoresis to investigate genetic heterogeneity at 33 protein coding loci in a total of 46 blue wildebeest (C. taurinus) kept under different management regimes.
  • 2.2. Average heterozygosity ranged from 2.14 to 4.3% and within-population differences accounted for 97.2% of total relative gene diversity.
  • 3.3. Comparatively little divergence was found between animals sampled from populations with very diverse population sizes and management histories, with the largest genetic distance estimated between any two populations being only 0.0021.
  • 4.4. We discuss our results with particular emphasis on the influence of management history on genetic diversity and divergence in C. taurinus.

  • 1.1. Metabolic rates (ml O2/mg/hr) of three geographically separated populations of the carabid beetle Calathus melanocephalus L. (Finse and Je 10y, Norway and Drenthe, The Netherlands) were measured and compared by ANCOVA.
  • 2.2. No significant relationship (P > 0.05) between metabolic rates and body weight or sex of the animals were found.
  • 3.3. Individuals mostly acclimated to low temperatures by increased metabolic rates and in the opposite direction to higher temperatures. Individuals collected in early summer also showed higher metabolic rates than those caught later in the autumn.
  • 4.4. Contradicting the theory of metabolic cold adaptation, beetles from The Netherlands had the highest metabolic rates, beetles from Finse intermediate rates and beetles from Jeløy the lowest rates.

  • 1.1. Specific Dynamic Action (SDA) effects of diet were investigated in the supralittoral isopod, Ligia pallasii, using defined chemical diets.
  • 2.2. “Apparent SDA”, or the total rise in metabolic rate following a meal, was resolved in animals eating a nutritionally complete chemical diet into three components: 8% mechanical costs of moving food through the gut, 40% “excitement costs” due to investigator disturbance and presence of food, and 52% SDA.
  • 3.3. Excitement costs in animals exposed to food but which chose not to eat showed non-significant variation between diets containing different levels of chemical nutrients, but were significantly less on a diet containing only cellulose and agar.
  • 4.4. SDA increased with increasing concentration of amino acids in the diet.
  • 5.5. Substitution of whole-protein casein for free amino acids in the diet had no significant SDA effect, while substitution of free amino acids in the ratio found in casein more than doubled the SDA effect.
  • 6.6. Deletion of alanine from the diet caused no significant effect on SDA, while deletion of phenylalanine caused a highly significant elevation in SDA.

  • 1.1. Five adult, female alligators (Alligator mississippiensis) were captured at night during the breeding season, and a blood sample taken within 5 min of capture.
  • 2.2. The alligators were physically restrained (tied to boards) and additional blood samples taken at 4, 8, 12, 16, 22, 28, 38, and 48 hr after capture. After the last blood sample was collected the animals were released.
  • 3.3. Plasma estradiol-17β and corticosterone were measured by radioimmunoassay. Estradiol declined significantly from initial values by 22 hr post capture, but remained unchanged for 48 hr.
  • 4.4. Plasma corticosterone rose from a mean of 0.8 ng/ml at capture to 12.6 ng/ml after 4 hr. Corticosterone continued to rise up to 16 hr then declined after 22 hr. From 28 until 48 hr corticosterone again increased significantly.
  • 5.5. These results demonstrate that acute stress in female alligators causes significant suppression of plasma estradiol and a biphasic pattern of corticosterone secretion.
