Summary A physiologically based steady-state model of whole leaf photosynthesis (WHOLEPHOT) is used to describe net photosynthesis daily time courses in Prunus armeniaca. Net photosynthesis rates are calculated in response to incident light intensity, leaf temperature, air carbon dioxide concentration, and leaf diffusion resistance measured at five minute intervals. The steady-state calculations closely approximate the observed net photosynthesis rates for a broad range of weather conditions and leaf stomatal behavior.  相似文献

Summary A physiologically based steady-state model of whole leaf photosynthesis (WHOLEPHOT) is detailed which describes the functional dependence of net photosynthesis in C 3 leaves on [CO2], [O2], incident radiant flux (PhAR), and leaf temperature. The model simulates among other phenomena a) observed [CO2], [O2], and temperature effects on the initial slope of light response curves, b) a C 3 type temperature response curve of net photosynthesis, c) a shift of the optimum temperature of net photosynthesis to higher temperatures with increasing light intensity, and d) observed temperature and [O2] effects on the CO2 compensation point. Model parameters are derived from published response data of several C 3 species. Simulations also demonstrate that parameter changes based on literature data result in acclimation-like changes in net photosynthesis response with respect to light intensity and temperature. The advantages of this model are that the number of parameters is minimized in order to focus on environmental effects and that all parameters can be determined from measured net photosynthesis responses.  相似文献

A mathematical model for leaf photosynthesis is constructed.The model relates the net rate of carbon dioxide exchange perunit leaf area to the ambient carbon dioxide and oxygen concentrationsand the light-flux density incident on the leaf surface. The behaviour of the model under steady-state conditions isexamined and discussed. Simulated carbon dioxide and light-responsecurves for the net carbon dioxide exchange rate accord wellwith experimental observation. The model describes an inhibitionof the net exchange rate by increasing oxygen concentrationswhich is similar to the commonly observed inhibition.  相似文献

Large underestimates of the limitation to photosynthesis imposed by stomata can occur because of an error in the standard method of calculating average substomatal pressures of carbon dioxide when heterogeneity of those pressures occurs across a leaf surface. Most gas exchange data supposedly indicating nonstomatal inhibition of photosynthesis by water stress could have this error. However, if no stomatal closure occurs, any reduction in photosynthesis must be due to nonstomatal inhibition of photosynthesis. Net carbon dioxide exchange rates and conductances to water vapor were measured under field conditions in upper canopy leaves of tomato plants during two summers in Beltsville, Maryland, USA. Comparisons were made near midday at high irradiance between leaflets in air with the ambient water vapor content and in air with a higher water content. The higher water content, which lowered the leaf to air water vapor pressure difference (VPD), was imposed either one half hour or several hours before measurements of gas exchange. In both seasons, and irrespective of the timing of the imposition of different VPDs, net photosynthesis increased 60% after decreasing the VPD from 3 to 1 kPa. There were no differences in leaf conductance between leaves at different VPDs, thus transpiration rates were threefold higher at 3 than at 1 kPa VPD. It is concluded that nonstomatal inhibition of photosynthesis did occur in these leaves at high transpiration rate.  相似文献

Using a cost-benefit model, the leaf nitrogen concentrationand root : shoot ratio that maximize whole-plant relative growthrate are determined as a function of the above-ground environment(integrated daily photon flux density and the concentrationof carbon dioxide at the site of fixation within the leaf).The major advantage of this approach is that it determines theadaptive significance of leaf physiology by considering thefunctional integration of leaves and roots. The predicted responseto increasing daily photon flux densities is an increase inoptimal leaf N concentration (Nopt) and a concomitant increasein root: shoot ratio. Increased carbon dioxide concentrations,on the other hand, reduce Nopt and only slightly change root:shoot ratio. The observed increase in leaf nitrogen concentrationfound in plants growing at high altitudes (low CO2 partial pressure)is also predicted. Since these responses to light and CO2 maximizethe whole-plant relative growth rate, the observed adjustmentsthat plants make to light and carbon dioxide concentration appearto be adaptive. We show that the relationship between photosynthesis and leafnitrogen concentration is complex and depends on the light andCO2 levels at which photosynthesis is measured. The shape ofthis function is important in determining Nopt and the oppositeresponse of leaf nitrogen to light and carbon dioxide is shownto be the result of the different effects of light and CO2 onthe photosynthesis-leaf nitrogen curve. Current-year shoots of Sitka spruce ( Picea sitchensis (Bong.) Carr.) were removed from the forest canopy. After steady-state rates of net photosynthesis were obtained in a leaf chamber, the shoots were excised in air and removed at different times to establish a relationship between net photosynthesis and xylem water potential. The experiment was repeated at five ambient carbon dioxide concentrations.
Net photosynthesis remained constant over a wide range of xylem water potential and increased linearly with ambient carbon dioxide concentration between 20 and 300 cm3 m−3. At low water potential net photosynthesis declined at each ambient carbon dioxide concentration and there was little difference in the potential (±0.05 MPa) at which zero photosynthesis was observed.
There was a small increase in the CO2 compensation concentration at low xylem water potentials, but calculated mesophyll conductance still declined at low water potential after correction for this change in compensation concentration. Mesophyll conductance reached zero within the same range of water potential as net photosynthesis. The results suggested that the non-stomatal contribution to the decline of photosynthesis was approximately 30% until almost complete stomatal closure occurred.  相似文献

Bot. 36: 1240–1248. The effect of salinity upon net photosynthesis and transpirationby individual leaves of rice has been investigated by gas exchangemeasurements in seedlings at the five to six leaf stage. Salinitydid not, initially, reduce net photosynthesis in the whole plantbut only in the older leaves in which sodium accumulated. Analysisof the course of events in leaf four following salinizationof the medium showed that net photosynthesis was inversely correlatedwith the sodium concentration in the leaf tissue. There wasno evidence of a threshold effect; net photosynthesis declinedlinearly with increasing leaf sodium concentration and was reducedby 50% at only 05 mmol sodium per gram dry weight. The relationshipbetween transpiration rate and leaf sodium concentration closelyparalleled that for photosynthesis; there was no effect of leafsodium concentration on the carbon dioxide concentration inthe intercellular spaces, showing that sodium accumulation inthe leaf affected stomatal aperture and carbon dioxide fixationsimultaneously. Photosynthesis was reduced by half at a sodiumconcentration in the leaf which did not reduce the concentrationof chlorophyll. The nature of the effect of salinity upon leafgas exchange is discussed. The process of photosynthesis is of such importance that most courses of biochemistry and physiology include some reference to it. This paper describes a simple experimental system, which uses radioactive carbon dioxide to study whole leaf photosynthesis under a variety of conditions.

An apparatus of very simple construction, in which the experiments can be safely carried out, is described. The experiments basically consist of the exposure of leaves to 14CO2 and the subsequent determination of radioactivity distribution and intensity by counting of leaf discs in a Geiger-Muller system and/or by autoradiography.

Experiments to demonstrate photosynthesis in the presence and absence of light, in variegated leaves of different types and to illustrate the importance of stomata are described.  相似文献

A method for measuring whole plant photosynthesis in Arabidopsis thaliana   总被引:5,自引:0,他引:5  
Measurement of photosynthesis of intact leaves of Arabidopsis thaliana has been prohibitive due to the small leaf size and prostrate growth habit. Because of the widespread use of Arabidopsis for plant science research it is important to have a procedure for accurate, nondestructive measurement of its photosynthesis. We developed and tested a method for analysis of photosynthesis in whole plants of Arabidopsis. Net carbon assimilation and stomatal conductance were measured with an open gas exchange system and photosynthetic oxygen evolution was determined from chlorophyll fluorescence parameters. Individual plants were grown in 50 cubic centimeter tubes that were attached with an air tight seal to an enclosed gas exchange chamber for measurement of carbon dioxide and water exchange by the whole plant. Chlorophyll fluorescence from intact leaves was simultaneously measured with a pulse modulated fluorometer. Photosynthetic CO2 assimilation and stomatal conductance rates were calculated with established gas exchange procedures and O2 evolution was determined from chlorophyll fluorescence measurement of Photosystem II yield. Carbon assimilation and oxygen evolution in response to light intensity and ambient CO2 concentration was measured and is presented here to demonstrate the potential use of this method for investigation of photosynthesis of Arabidopsis plants in controlled environment conditions.  相似文献

Ozone pollution may reduce net carbon gain in forests, yet data from mature trees are rare and the effects of irradiance on the response of photosynthesis to ozone remain untested. We used an open-air system to expose 10 branches within the upper canopy of an 18-m-tall stand of sugar maple (Acer saccharum Marsh.) to twice-ambient concentrations of ozone (95nmol mol?1, 0900 to 1700, 1 h mean) relative to 10 paired, untreated controls (45nmol mol?1) over 3 months. The branch pairs were selected along a gradient from relatively high irradiance (PPFD 14.5 mol m?2 d?1) to deep shade (0.7mol m?2 d?1). Ozone reduced light-saturated rates of net photosynthesis (Asat) and increased dark respiration by as much as 56 and 40%, respectively. Compared to sun leaves, shade leaves exhibited greater proportional reductions in Asat and had lower chlorophyll concentrations, quantum efficiencies, and leaf absorptances when treated with ozone relative to controls. With increasing ozone dose over time, Asat became uncoupled from stomatal conductance as ratios of internal to external concentrations of carbon dioxide increased, reducing water-use efficiency. Ozone reduced net photosynthesis and impaired stomatal function, with these effects depending on the irradiance environment of the canopy leaves. Increased ozone sensitivity of shade leaves compared to sun leaves has consequences for net carbon gain in canopies.  相似文献

The response of net photosynthesis and apparent light respiration to changes in [O2], light intensity, and drought stress was determined by analysis of net photosynthetic CO2 response curves. Low [O2] treatment resulted in a large reduction in the rate of photorespiratory CO2 evolution. Lightintensity levels influenced the maximum net photosynthetic rate at saturating [CO2]. These results indicate that [CO2], [O2] and light intensity affect the levels of substrates involved in the enzymatic reactions of photosynthesis and photorespiration. Intracellular resistance to CO2 uptake decreased in low [O2] and increased at low leaf water potentials. This response reflects changes in the efficiency with which photosynthetic and photorespiratory substrates are formed and utilized. Water stress had no effect on the CO2 compensation point or the [CO2] at which net photosynthesis began to saturate at high light intensity. Summary The protein synthesis inhibitors chloramphenicol and terramycin, and light of low intensity were used to retard the rate of chlorophyll formation in illuminated dark grown pea leaves. In the control leaves the onset of photosynthesis, as measured by carbon dioxide exchange of the whole leaves, and reduction of ferricyanide and metmyoglobin and photo-oxidation of ascorbate in isolated chloroplasts, was observed after 2–4 hours illumination. The photosynthetic activity of the treated leaves did not commence until 10–12 hours illumination had elapsed. In both the control and treated leaves the onset of photosynthesis occurred when the total chlorophyll content was 0.04 mg/g fresh weight. The precise point of photosynthetic inception was apparently more related to the attainment of a specific total chlorophyl content than to the ratio of chlorophyll a to chlorophyll b. A marked increase in the evolution of carbon dioxide in the light was observed in the treated leaves during the first 10 hours of greening. This observation could not be ascribed to photorespiration since the leaves did not possess an active photosystem. 为探讨甜瓜光响应变化特性与环境因子的关系,选择光响应曲线适宜测定的时段,以甜瓜幼苗为试材,将1 d分为3个时段:10:00-12:00、12:00-15:00和15:00-17:00,每个叶位叶片测定1 d,并采用直角双曲线修正模型拟合光响应曲线,研究不同时段下甜瓜叶片光响应曲线、光响应参数的变化趋势和不同叶位叶片光响应参数特性。结果表明:当环境中光合有效辐射增强,叶面温度(Tl)升高,空气相对湿度(RH)降低;当环境中光合有效辐射减弱,Tl降低,RH升高。10:00-12:00光响应曲线和12:00-15:00的第1-4叶光响应曲线呈双曲线,在强光下趋向饱和状况,12:00-15:00的第5叶光合速率和15:00-17:00光合速率在强光下出现明显的光抑制现象。1 d的不同时段均表现为10:00-12:00最大净光合速率(Pmax)和光饱和点(LSP)最高,12:00-17:00降低;12:00-15:00光补偿点(LCP)和暗呼吸速率(Rd)较高,其它两个时段较低,10:00-17:00光补偿点量子效率(φc)、气孔导度(Gs)和胞间CO2浓度(Ci)总体呈降低趋势,气孔限制值(Ls)升高。10:00-15:00相同时段测得的不同叶位叶片光响应参数,以第4-5叶光合性能较好,15:00-17:00以第3叶Pmax最高,第5叶次之;10:00-17:00 GsCi以第5叶较低,第1叶较高,Ls以第5叶较高,第1叶较低。RH为影响Pmax的主要决策因子,测定时段、叶面饱和蒸汽压亏缺(Vpdl)和Tl为主要限制因子。10:00-12:00适宜进行光响应曲线测定,气孔限制为不同时段光合作用不同的主要因素,非气孔限制为影响不同叶位叶片光合作用的主要因素。  相似文献

Photosynthetic carbon gain in rapidly fluctuating light is controlled by stomatal conductance, activation of ribulose-1,5-bisphosphate carboxylase-oxygenase, a fast induction step in the regeneration of ribulose-1,5-bisphosphate, and the build-up of pools of photosynthetic intermediates that allow post-illumination CO2 fixation. Experimental work over recent years has identified and characterised these factors. A physiologically-based dynamic model is described here that incorporates these factors and allows the simulation of carbon gain in response to any arbitrary sequence of light levels. The model output is found to conform well to previously reported plant responses of Alocasia macrorrhiza (L.) G. Don. observed under widely differing conditions. The model shows (i) responses of net assimilation rate and stomatal conductance to constant light levels and different CO2 concentrations that are consistent with experimental observations and predictions of a steady-state model; (ii) carbon gain to continue after the end of lightflecks, especially in uninduced leaves; (iii) carbon gain to be only marginally reduced during low-light periods of up to 2 s; (iv) a fast-inducing component in the regeneration of ribulose-1,5-bisphosphate to be limiting for up to 60 s after an increase in light in uninduced leaves: the duration of this limitation lengthens with increasing CO2 concentration and is absent at low CO2 concentration; (v) oxygen evolution to exceed CO2 fixation during the first few seconds of a lightfleck, but CO2 fixation to continue after the end of the lightfleck whereas oxygen evolution decreases to low-light rates immediately. The model is thus able to reproduce published responses of leaves to a variety of perturbations. This provides good evidence that the present formulation of the model includes the essential rate-determining factors of photosynthesis under fluctuating light conditions. Light gradients within tree canopies play a major role in the distribution of plant resources that define the photosynthetic capacity of sun and shade leaves. However, the biochemical and diffusional constraints on gas exchange in sun and shade leaves in response to light remain poorly quantified, but critical for predicting canopy carbon and water exchange. To investigate the CO2 diffusion pathway of sun and shade leaves, leaf gas exchange was coupled with concurrent measurements of carbon isotope discrimination to measure net leaf photosynthesis (An), stomatal conductance (gs) and mesophyll conductance (gm) in Eucalyptus tereticornis trees grown in climate controlled whole‐tree chambers. Compared to sun leaves, shade leaves had lower An, gm, leaf nitrogen and photosynthetic capacity (Amax) but gs was similar. When light intensity was temporarily increased for shade leaves to match that of sun leaves, both gs and gm increased, and An increased to values greater than sun leaves. We show that dynamic physiological responses of shade leaves to altered light environments have implications for up‐scaling leaf level measurements and predicting whole canopy carbon gain. Despite exhibiting reduced photosynthetic capacity, the rapid up‐regulation of gm with increased light enables shade leaves to respond quickly to sunflecks.  相似文献

Summary Fragaria vesca, the woodland strawberry, was grown under a series of controlled environments including variations in light intensity, average temperatures, and temperature amplitude around a constant mean. Observations on CO2 exchange capacities, leaf anatomy, and cell ultrastructure were made for each treatment to determine relationships between these variables. With increasing light intensity, leaf thickness, leaf density, and mesophyll cell surface area and volume per leaf surface area increased. Net photosynthesis (NPS) per leaf weight decreased with increasing light pretreatment while NPS per area increased from low to medium intensity, then decreased at the highest intensity. Depression of photosynthesis at the highest light pretreatment may have been due to massive starch accumulation in the chloroplasts associated with the sodium vapor lamps used. Correlation of all anatomical variables was highly significant with dark respiration and NPS per dry weight but insignificant for NPS per leaf area. In the variable temperature treatments, photosynthetic acclimation occurred with a shift in optimum temperature for NPS in the direction of prevailing growth temperature. Absolute rates were highest at moderate pretreatment temperatures and were reduced by extreme growth temperatures. Thick leaves with low density mesophyll became thinner and more dense with increasing growth temperature corresponding to an increase in maximum net photosynthetic rates. Leaves became thicker and more dense at the highest temperatures, but with an increase in cell damage and indications of changes in metabolic pathways. Highest correlations for gas exchange rates were with specific leaf weight (weight per area). Correlation with other anatomical variables were scattered or insignificant. It was concluded that adaptation to a range of environmental conditions cannot be consistently attributed to changes in mesophyll cell volume or surface area.  相似文献

Temperate forests are characterised by variable light quality (i.e. spectral composition of light) at or near the forest floor. These understory environments have a high concentration of green light, as red and blue light are preferentially absorbed by upper canopy leaves. Understory species may be well-adapted for using green light to drive photosynthesis. Angiosperms have been shown to use green light for photosynthesis, but this ability has not been demonstrated in shade-dwelling bryophytes. In this study, net photosynthetic rate (PN) of three temperate understory species of moss (Dichodontium pellucidum (Hedw.) Schimp., Leucobryum albidum (Brid. ex P.Beauv) Lindb. and Amblystegium serpens (Hedw.) Schimp.) was measured under green, red?+?blue, and red?+?blue?+?green light to assess green light use efficiency. All three species were capable of photosynthesising beyond their respiratory demands using solely green light, with higher green light use efficiency measured in plants collected from areas with greater canopy cover, suggesting growth in a green light concentrated environment increases green light use efficiency. Each species was also collected from sites differing in their degree of canopy cover and grown under three light treatments (high light, low light, and green light). Photosynthetic efficiency (chlorophyll fluorescence), tissue nitrogen and carbon isotope concentrations were assessed after a short growth period. Growth conditions had little effect on leaf chemistry and monochromatic green light did not significantly degrade photosynthetic efficiency. This study provides the first evidence to date of positive net ‘green light photosynthesis’ in mosses.  相似文献

Summary A model predicting net photosynthesis of individual plant leaves for a variety of environmental conditions has been developed. It is based on an electrical analogue describing gas diffusion from the free atmosphere to the sites of CO2 fixation and a Michaelis-Menten equation describing CO2 fixation. The model is presented in two versions, a simplified form without respiration and a more complex form including respiration. Both versions include terms for light and temperature dependence of CO2 fixation and light control of stomatal resistance. The second version also includes terms for temperature, light, and oxygen dependence of respiration and O2 dependence of CO2 fixation.The model is illustrated with curves based on representative values of the various environmental and biological parameters. These curves relate net photosynthesis to light intensity, [CO2], [O2], temperature, and resistances to CO2 uptake. The shape of the [CO2]-net photosynthesis curves depends on the total diffusion resistance to CO2 uptake and the Michaelis constant for CO2 uptake. The curves range from typical Michaelis-Menten to Blackman types.The model is combined with a model of leaf energy exchange permitting simultaneous estimation of net photosynthesis and transpiration. The combined model is illustrated with curves relating transpiration to photosynthesis under a wide variety of environmental conditions. Environmental regimes yielding maximum efficiency of water use are identified for the given assumptions and biological parameters.  相似文献

Excised leaves of silver maple (Acer saccharinum L.) exposed to 0, 0.045, 0.090, or 0.180 mM Cd24 exhibited reduced net photosynthesis and transpiration, and increased dark respiration. Rates of net photosynthesis and transpiration diminished with time and were strongly correlated with solution concentration and tissue content of Cd24, Net photosynthesis and transpiration were reduced to 18 and 21%, respectively, of the untreated controls after 64 h. Dark respiration increased as much as 193% of the untreated controls but was poorly correlated with solution concentration or tissue content of Cd24, Diffusive resistances of leaves to carbon dioxide and water vapor transfer increased with both increasing Cd24 concentration and time.
