Recent studies are emphasising the importance and putative modes of action of specific flavonoids as bioactive components of the diet in in vivo and in vitro models. Thus, it is important to have a clear idea of the major phenolic families of which fruit and vegetables are comprised and the levels contained therein. Regularly consumed fruit and vegetables of mixed varieties available on the UK market were analysed for the composition of the major individual phenolic components. The total phenolic content (applying the Folin assay) and the vitamin C levels were also determined. The antioxidant capacities of aqueous/methanolic extracts were comparatively assessed using the TEAC (Trolox Equivalent Antioxidant Capacity), the FRAP (Ferric Reducing Ability of Plasma) and ORAC (Oxygen Radical Absorbance Capacity) assays, which comprise contributions from polyphenols, simple phenols and the ascorbate component. The results were calculated in terms of 100 g fresh weight (FW) uncooked portion sizes. Fruit and vegetables rich in anthocyanins (e.g. strawberry, raspberry and red plum) demonstrated the highest antioxidant activities, followed by those rich in flavanones (e.g. orange and grapefruit) and flavonols (e.g. onion, leek, spinach and green cabbage), while the hydroxycinnamate-rich fruit (e.g. apple, tomato, pear and peach) consistently elicited the lower antioxidant activities. The TEAC, FRAP and ORAC values for each extract were relatively similar and well-correlated with the total phenolic and vitamin C contents. The antioxidant activities (TEAC) in terms of 100 g FW uncooked portion size were in the order: strawberry> raspberry = red plum > red cabbage >grapefruit = orange > spinach > broccoli > green grape approximately/= onion > green cabbage > pea > apple > cauliflower tomato approximately/= peach=leek > banana approximately/= lettuce.  相似文献

Increased consumption of vegetables or plant food has been associated with decreased risk of developing major chronic diseases, such as cancers, diabetes, cardiovascular diseases, and age-related functional decline. Ramie leaves are rich in phenolics and flavonoids, which have been suggested for human health benefits. Phenolic contents, flavonoid contents, phenolic compounds, and anti-cancer properties in six species of ramie leaves were analyzed by Folin-reagent method, sodium borohydride/chloranil-based assay (SBC), HPLC method and antiproliferation, cytoxicity, respectively. Antioxidant activities were measured through peroxyl radical scavenging capacity (PSC) method, oxygen radical absorbance capacity (ORAC) method, and cellular antioxidant activity (CAA). Research indicated that Boehmeria penduliflora contained the highest total phenolic content (2313.7±27.28 mg GAE/100 g FW), and flavonoid content (1682.4±27.70 mg CAE/100 g FW). Boehmeria tricuspis showed the highest PSC value (9574.8±117.63 µM vit. C equiv./100 g FW), while Boehmeria penduliflora indicated the highest ORAC value (330.44±16.88 µmol Trolox equiv./g FW). The antioxidant activities were correlated with phenolic contents and flavonoid contents. Boehmeria tricuspis had the highest antiproliferative capacity with the lowest EC50 (4.11±0.19 mg/mL). The results for the analyzed ramie for CAA were significantly different from each other (p<0.05), Boehmeria tricuspis had the highest CAA value (133.63±7.10 µmol QE/100 g). Benzoic acid, 4-coumaric acid, caffeic acid, and ferulic acid were the dominant phenolic ingredients in the ramie leaves according to HPLC analysis. Our research is the first report to study the phytochemical profiles and antioxidant activities in different species of ramie leaves for their health benefit.  相似文献

2,5,7,8-tetramethyl-2-(2'-carboxyethyl)-6-hydroxychroman ( &#102 -CEHC) has been identified as a major water-soluble metabolite of vitamin E, which circulates in the blood and is excreted with the urine. The aim of this study was to assess the antioxidant activity of &#102 -CEHC using several methods with different prooxidant challenges. In the Oxygen Radical Absorbance Capacity assay, a fluorescent protein acts as a marker for oxidative damage induced by peroxyl radicals. In the Trolox Equivalent Antioxidant Capacity (TEAC) assay, a stable free radical, 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid (ABTS &#148 + ) is reduced directly by antioxidants. Scavenging properties vs. reactive nitrogen species were studied measuring the effects on tyrosine nitration after reaction with peroxynitrite. Trolox, &#102 -tocopherol, ascorbic acid, and ( &#109 )-epicatechin were simultaneously tested in order to compare their antioxidant activities. In all mentioned systems, &#102 -CEHC exhibited antioxidant properties similar to those of Trolox. We conclude that &#102 -CEHC is a molecule with good antioxidant activity, having the advantage over Trolox of being a naturally occurring compound. These properties might be useful for research or industrial purposes.  相似文献

Recent studies show that edible berries may have potent chemopreventive properties. Anti-angiogenic approaches to prevent and treat cancer represent a priority area in investigative tumor biology. Vascular endothelial growth factor (VEGF) plays a crucial role for the vascularization of tumors. The vasculature in adult skin remains normally quiescent. However, skin retains the capacity for brisk initiation of angiogenesis during inflammatory skin diseases such as psoriasis and skin cancers. We sought to test the effects of multiple berry extracts on inducible VEGF expression by human HaCaT keratinocytes. Six berry extracts (wild blueberry, bilberry, cranberry, elderberry, raspberry seed, and strawberry) and a grape seed proanthocyanidin extract (GSPE) were studied. The extracts and uptake of their constituents by HaCaT were studied using a multi-channel HPLC-CoulArray approach. Antioxidant activity of the extracts was determined by ORAC. Cranberry, elderberry and raspberry seed samples were observed to possess comparable ORAC values. The antioxidant capacity of these samples was significantly lower than that of the other samples studied. The ORAC values of strawberry powder and GSPE were higher than cranberry, elderberry or raspberry seed but significantly lower than the other samples studied. Wild bilberry and blueberry extracts possessed the highest ORAC values. Each of the berry samples studied significantly inhibited both H 2 O 2 as well as TNF &#102 induced VEGF expression by the human keratinocytes. This effect was not shared by other antioxidants such as &#102 -tocopherol or GSPE but was commonly shared by pure flavonoids. Matrigel assay using human dermal microvascular endothelial cells showed that edible berries impair angiogenesis.  相似文献

Flavonoids are natural phenolic substances widely found in fruit, vegetables, grains, and wine. Most of these compounds exert health-promoting effects seem to attribute to their antioxidant activity. Metallothioneins (MT) has been suggested to protect against acute heavy metal toxicity in the liver, and the proteins of MT can be induced by various stimuli including antioxidant. Measuring the induction of MT genes may provide an efficient approach to understand the chemopreventive mechanisms of flavonoids. The antioxidant activity of eight flavonoids was determined by TEAC and ORAC assays and their effects on MT protein were also measured. HepG2 cells were employed to explore the mechanisms underlying flavonoid-induced MT induction. Statistical analysis revealed a positive correlation between the antioxidant activity of flavonoids and MT expression. Quercetin-induced MT expression may function by activating the phosphorylation of JNK, p38 and PI3K/Akt as well as by enhancing Nrf2 DNA-binding activity. Moreover, quercetin exhibited a potential protective effect on t-BHP-caused injury in hepatocytes through the induction of MT. These results suggest that quercetin is a natural antioxidant in the diet and the consumption of foods that are rich in quercetin could be beneficial for the prevention of environmental oxidant-induced liver damage.  相似文献

Cancer prevention and treatment using traditional Chinese medicines have attracted increasing interest. This study characterizes antioxidant activity and phenolic compounds of traditional Chinese medicinal plants associated with anticancer, comprising 112 species from 50 plant families. The improved ABTS(*+) method was used to systematically assess the total antioxidant capacity (Trolox equivalent antioxidant capacity, TEAC) of the medicinal extracts. The TEAC values and total phenolic content for methanolic extracts of herbs ranged from 46.7 to 17,323 micromol Trolox equivalent/100 g dry weight (DW), and from 0.22 to 50.3 g of gallic acid equivalent/100 g DW, respectively. A positive, significant linear relationship between antioxidant activity and total phenolic content (all R(2) values>/=0.95) showed that phenolic compounds were the dominant antioxidant components in the tested medicinal herbs. Major types of phenolic compounds from most of the tested herbs were preliminarily identified and analyzed, and mainly included phenolic acids, flavonoids, tannins, coumarins, lignans, quinones, stilbenes, and curcuminoids. These medicinal herbs exhibited far stronger antioxidant activity and contained significantly higher levels of phenolics than common vegetables and fruits. Traditional Chinese medicinal plants associated with anticancer might be potential sources of potent natural antioxidants and beneficial chemopreventive agents.  相似文献

Antioxidant capacity and phenolic compounds (phenolic acids and anthocyanins) of four berry fruits (strawberry, Saskatoon berry, raspberry and wild blueberry), chokecherry and seabuckthorn were compared in the present study. Total phenolic content and total anthocyanin content ranged from 22.83 to 131.88 g/kg and 3.51 to 13.13 g/kg, respectively. 2,2-Diphenyl-1-picryhydrazyl free radical scavenging activity ranged from 29.97 to 78.86%. Chokecherry had the highest antioxidant capacity when compared with berry fruits and seabuckthorn. The highest caffeic acid, gallic acid and trans-cinnamic acid levels were found in chokecherry (6455 mg/kg), raspberry (1129 mg/kg) and strawberry (566 mg/kg), respectively. Caffeic acid was also the major phenolic acid in Saskatoon berry (2088 mg/kg) and wild blueberry (1473 mg/kg). The findings that chokecherry has very high antioxidant capacity and caffeic acid levels, are useful for developing novel value-added antioxidant products and also provide evidence essential for breeding novel cultivars of fruit plants with strong natural antioxidants.  相似文献

In this study, we investigated the phenolic composition of the crude extract (MeOH 80 %) of Alnus cordata (Loisel .) Duby stem bark (ACE) and its antioxidant and skin whitening properties. RP‐LC‐DAD analysis showed a high content of hydroxycinnamic acids (47.64 %), flavanones (26.74 %) and diarylheptanoids (17.69 %). Furthermore, ACE exhibited a dose‐dependent antioxidant and free‐radical scavenging activity, expressed as half‐maximal inhibitory concentration (IC50): Oxygen radical absorbance capacity (ORAC, IC50 1.78 μg mL?1)>Trolox equivalent antioxidant capacity (TEAC, IC50 3.47 μg mL?1)>2,2‐Diphenyl‐1‐picrylhydrazyl (DPPH, IC50 5.83 μg mL?1)>β‐carotene bleaching (IC50 11.58 μg mL?1)>Ferric reducing antioxidant power (FRAP, IC50 17.28 μg mL?1). Moreover, ACE was able to inhibit in vitro tyrosinase activity (IC50 77.44 μg mL?1), l ‐DOPA auto‐oxidation (IC50 39.58 μg mL?1) and in an in vivo model it exhibited bleaching effects on the pigmentation of zebrafish embryos (72 h post fertilization) without affecting their development and survival. In conclusion, results show that A. cordata stem bark may be considered a potential source of agents for the treatment of skin disorders due to its bleaching properties and favorable safety profiles, associated to a good antioxidant power.  相似文献

This work aims to determine the phytochemical characterization of the pericarp of Chamaedorea radicalis Mart. fruit as a non-timber product with potential to obtain phytochemicals with potential applications in the industry. Fruit from C. radicalis were grouped in four ripening stages named as S1, S2, S3 and S4, according to maturity; S1 the most unripe stage and S4 the completely ripe stage. Determinations of total phenolic compounds, free radical scavenging activities and total flavonoid contents using spectrophotometric methods were done. Also, the tentative identification of phytochemicals during fruit ripening was done using UPLC-MS-MS. Total phenolic compound (TPC) content ranged from 7.24 to 12.53 mg gallic acid equivalents per gram of fresh weight (mg GAE/ g FW). Total flavonoids (TF) contents ranged from 0.163 to 0.23 mg of quercetin equivalents per g FW (mg QE/g FW). Free radical scavenging activity against DPPH and ABTS radicals varied from 40.80 to 53.68 and from 22.29 to 37.76 mmol Trolox equivalents g FW (mmol TE/g FW), respectively. Antioxidant assay in vitro by FRAP (ferric reducing antioxidant power) method showed that S3 was the highest level with antioxidant power while S4 was the lowest with Red ripeness stage showed the lowest contents for all determinations. Mass spectrometry allowed detection of 26 compounds, including phenolics, alkaloids and saponins. Afzelin, Kaempferol 3-neohesperidoside and the four saponins identified were present in all ripeness stages. Preliminary phytochemical identi- fication and the spectrophotometric determinations showed that the pericarp of C. radicalis presented antioxidants and compounds related to alkaloids, phenolics and saponins. The presence and abundance of each phytochemical regarding each ripeness stage should be considered.  相似文献

This study aimed to determine the phenolic compounds profile, antioxidant potential and cytotoxicity of extracts and fractions of Caesalpinia palmeri. Methanolic extracts were generated from C. palmeri berries, stems and flowers. The latter was subjected to liquid-liquid partition, obtaining hexane, ethyl acetate and residues fractions. Results showed that the flower extract and ethyl acetate fraction had a larger concentration of phenolic compounds (148.9 and 307.9 mg GAE/g, respectively), being ellagic acid (6233.57 and 19550.08 μg/g, respectively), quercetin-3-β-glycoside (3023.85 and 8952.55 μg/g, respectively) and gallic acid (2212.98 and 8422.34 μg/g, respectively) the most abundant compounds. Flower extract and ethyl acetate fraction also presented the highest antioxidant capacity on all tested methods (DPPH, ABTS, ORAC and FRAP) and low cytotoxicity against ARPE-19 cells (IC50 >170 μg/mL). C. palmeri possessed high antioxidant potential, associated with the presence of phenolic compounds and low cytotoxicity, suggesting that they could represent an option to counter oxidative stress.  相似文献

The antioxidant activity of eight edible species of Malaysian North Borneo seaweeds obtained from Sabah waters (Kudat, Tanjung Aru and Semporna) consisting of three red seaweeds (Eucheuma cottonii, E. spinosum and Halymenia durvillaei), two green seaweeds (Caulerpa lentillifera and C. racemosa) and three brown seaweeds (Dictyota dichotoma, Sargassum polycystum and Padina sp.) were determined. Methanol and diethyl ether were used as extraction solvent. The antioxidant activities were determined by two methods, TEAC (trolox equivalent antioxidant capacity) and FRAP (ferric reducing antioxidant power) assays. The total phenolic content of the extract was determined according to the Folin–Ciocalteu method and results were expressed as phloroglucinol equivalents. The methanolic extracts of green seaweeds, C. lentillifera and C. racemosa, and the brown seaweed, S. polycystum showed better radical-scavenging and reducing power ability, and higher phenolic content than the other seaweeds. The TEAC and FRAP assays showed positive and significantly high correlation (R 2 = 0.89). There was a strong correlation (R 2 = 0.96) between the reducing power and the total phenolic content of the seaweeds methanolic dry extracts. These seaweeds could be potential rich sources of natural antioxidants.  相似文献

In this paper we review the current literature on antioxidants from fruit of red raspberry (Rubus idaeus) and place these in context concerning what is known from other food species. The review concentrates on the methods of antioxidant testing, the diversity of antioxidants in raspberry, effects of ripeness, cultivar, storage and processing techniques, and the bioavailability of raspberry antioxidants in humans after eating the fruit. It is clear that raspberry, like several other fruits and vegetables such as tomato, strawberry, kiwi and broccoli, represents a valuable contrasting source of potentially healthy compounds and can represent an important component of a balanced diet.  相似文献

Epidemiology studies have shown that consumption of fruits and vegetables is associated with the prevention of chronic diseases such as cancer and cardiovascular disease. Induction of cellular phase II detoxifying enzymes is associated with cancer preventive potential. Phenolsulfotransferases (PSTs) are traditionally known as phase II drug-metabolizing or detoxifying enzymes that facilitate the removal of drugs and other xenobiotic compounds. Phenolic acids are known to increase the activities of PSTs. In the present study, human HepG2 cells were used as model to investigate the influence of twenty vegetables on human PST activity and to evaluate the relationships to their antioxidant activity and total phenolics content. The result showed that PST-P activity was significantly (p < 0.01) induced by asparagus, broccoli, cauliflower, celery and eggplant, whereas PST-M activity was induced by asparagus, broccoli, carrot, eggplant and potato at a concentration of 100 microg/ml. The vegetable extracts that induced both forms of PSTs activities were found to have higher antioxidant capacities and total phenolic content in the oxygen radical absorbance capacity (ORAC) and Folin-Ciocalteu assay. The major polyphenols in broccoli, the most potential inducer in both forms of PSTs activities, was antioxidant phenolic acids. HPLC retention times and standard spiked indicated the presence of gallic acid, p-hydroxybenzoic acid, p-coumaric acid, gentisic acid and ferulic acid in broccoli. The overall effect of vegetables tested on the activity of PST-P was well correlated to their ORAC value and total phenolics content (r= 0.82, p < 0.05 and r = 0.78, p < 0.05). These results imply that vegetables have a capability of inducing PST activity, and the PST induction may be possibly ascribed to antioxidant phenolic acids in vegetable extracts.  相似文献

The phenolic constituents of Mauritian endemic plants from the Rubiaceae and Myrtaceae family were assessed and correlated with their potential antioxidant activities in vitro. The antioxidant activities of the plant extracts ranged from 0.27 to 1.49mmol Trolox equivalent/g FW and from 0.20 to 1.39mmol Fe(II) equivalent/g FW in the TEAC and FAP assays, respectively, with Syzygium commersonii showing the highest activity in these two systems. Eugenia orbiculata and all the Syzygium species were effective scavengers of hypochlorous acid while Monimiastrum acutisepalum was the most potent inhibitor of deoxyribose degradation. The plant extracts inhibited microsomal lipid peroxidation with low IC(50)s ranging from 0.02 to 1.75mgFW/mL when reaction was initiated with Fe(3+)/ascorbate and from 0.093 to 1.55mgFW/mL in the AAPH-dependent lipid peroxidation. The potential prooxidant nature of the plant extracts was compared with ascorbate (250microM) using copper-phenanthroline assay. The plant extracts at concentrations up to 5gFW/L were not prooxidant. However, Myonima nitens, Syzygium commersonii, Syzygium glomeratum and Syzygium mauritianum at concentrations of 10gFW/L had potency approaching 50% of the prooxidant activity of ascorbic acid in vitro, suggesting relative safeties. The total phenolics influenced the antioxidant activities in the TEAC, FRAP and HOCl scavenging assays whereas a negative correlation was observed with the deoxyribose assay. The high levels of polyphenolic compounds and the significant antioxidant activities of these Rubiaceae and Myrtaceae plant family make them suitable candidates as prophylactic agent.  相似文献

The present study aimed to assess antioxidant activities of three organs (flower, fruit, and stem) extracts of Tunisian Ferula (F.) communis. Various experimental models were used to characterize the antioxidant activities in vitro as well as on ROS-induced fluorescence using dichlorofluorescein technique from phorbol myristate acetate (PMA)-stimulated human myeloid cell line HL-60. Results showed that the antioxidant activities varied considerably with organs. Thus, flower exhibited higher DPPH-scavenging ability, reducing and chelating power than stem and fruit. Also, antioxidant capacities using ORAC method and a cell-based assay showed that fruit and stem exhibited statistically similar antioxidant activities. Moreover, F. communis contains high amounts of flavonoids with various health benefits attributed to their antioxidant potential. Likewise, to obtain biologically relevant information, the antioxidant activities of the extracts were evaluated on cellular models implicating the antioxidant activities; this test generally showed that F. communis flower extracts have the highest antioxidant capacities correlated to the highest total phenolic content. The identification of phenolic compounds in F. communis extracts using RP-HPLC revealed that resorcinol, ferulic, and syringic acids together with coumarin were the major molecules.  相似文献

Fruits and vegetables are rich sources of antioxidants in human diets and their intake is associated with chronic disease prevention. Lettuce (Lactuca sativa L.) is a common vegetable in diets worldwide, but its nutritional content is relatively low. To elucidate the genetic basis of antioxidant content in lettuce, we measured the oxygen radical absorbance capacity (ORAC) and chlorophyll (Chl) content as a proxy of β-carotene in an F8 recombinant inbred line (RIL) in multiple production cycles at two different production sites. Plants were phenotyped at the open-leaf stage to measure genetic potential (GP) or at market maturity (MM) to measure the influence of head architecture (‘head’ or ‘open’). Main effect quantitative trait loci (QTL) were identified at MM (three Chl and one ORAC QTL) and GP (two ORAC QTL). No main effect QTL for Chl was detected at GP, but epistatic interaction was identified in one pair of marker intervals for each trait at GP. Interactions with environment were also detected for both main and epistatic effects (two for main effect, and one for epistatic effect). Main effect QTL for plant architecture and nutritional traits at MM colocated to a single genomic region. Chlorophyll contents and ORAC values at MM were significantly higher and Chl a to Chl b ratios were lower in ‘open’ types compared to ‘head’ types. The nutritional traits assessed for GP showed a significant association with plant architecture suggesting pleiotropic effects or closely linked genes. Taken together, the antioxidant and chlorophyll content of lettuce is controlled by complex mechanisms and participating alleles change depending on growth stage and production environment.  相似文献

Antioxidants are compounds that scavenge the free radicals produced in living organisms. The antioxidant potential of eight Arctic lichen species was evaluated in vitro using free radical scavenging activity (FRS), inhibition of lipid peroxidation (ILP), and Trolox equivalent antioxidant capacity assay (TEAC). FRS activities of lichen species in various organic solvents such as methanol, ethanol, acetone, and dimethyl sulphoxide (DMSO) were in the range 9.6–51.77%, while ILP activities in these solvents ranged from 32.5 to 82.43%. Pseudophebe pubescens showed the highest ILP (82.43%) and FRS (51.77%) activities as compared to other lichen species and the standard antioxidants butylated hydroxyanisole (BHA) and butylated hydroxytoluene (BHT). The TEAC value was also found to be higher in all species compared to the standard water soluble vitamin E analog Trolox (3.9 mM). The order of antioxidative activities in lichen species was Pseudophebe pubescens > Cladonia amaurocraea > Cladonia mediterranea > Physcia caesia > Flavocetraria nivalis > Cetraria fastigata > Xanthoria elegans > Umbilicaria hyperborea. This is the first report of the measurement of antioxidant potential in Arctic lichens.  相似文献

The total polyphenol, oxygen radical absorbance capacity (ORAC), ferric reducing antioxidant power (FRAP), radical cation scavenging ability, flavonol and flavanone contents were measured in the leaves, bulbs, roots, flowers and fruit (dry weight) of two natural populations of Gethyllis multifolia (Kukumakranka) and Gethyllis villosa. The flowers and fruit of G. multifolia and G. villosa showed higher, and in some cases significantly (P < 0.05) higher antioxidant activities when compared to the leaves, bulbs and roots. This, however, was not true for the flavanone content in both species. The total polyphenol content in the fruits of G. multifolia (21.54 mg GAE/g) and G. villosa (27.64 mg GAE/g) were found to be in agreement with those of raisins (28.30 mg GAE/g), blueberries (24 mg GAE/g) and strawberries (15.40 mg GAE/g). The FRAP values of G. multifolia flowers (76.66 μmole AAE/g) and fruit (91.51 μmole AAE/g) were found to be significantly (P < 0.05) higher than those of the other plant parts (16.76 to 39.08 μmol AAE/g). On the other hand, the flowers (590.23 μmol TE/g) and fruit (741.16 μmol TE/g) of G. villosa revealed a significantly (P < 0.05) higher ORAC when compared to the other plant parts (251.25 to 410.60 μmol TE/g). A strong correlation was evident in the fruit of both species between the total polyphenols and FRAP (r = 0.95), ORAC (r = 0.95) and flavonol content (r = 0.79).  相似文献

Two solvent extraction procedures were used to investigate the extraction efficiency in terms of total antioxidant capacity and total phenols in apricot fruit. Samples were either sequentially extracted with aqueous ethanol (ethanol/water 80% v/v) and tetrahydrofuran or directly extracted with tetrahydrofuran. Each extract was analyzed for total antioxidant capacity by the Trolox Equivalent Antioxidant Capacity (TEAC) assay and total phenols by the Folin-Ciocalteu assay. The results showed that using sequential solvent extraction, the majority (85%) of the total antioxidant capacity and total phenols was due to hydrophilic compounds. In tetrahydrofuran direct extractions, the total antioxidant capacity and total phenols were higher than values obtained with aqueous ethanol and the sum of results obtained from sequential extracts for either total antioxidant capacity or total phenols was similar to the tetrahydrofuran-extract antioxidant values. A linear correlation between total antioxidant capacity and total phenols was found and was independent of the solvent extraction method. In conclusion, the choice of solvent is related to the antioxidant potential of fruit and depends on the food hydrophilic/lipophilic composition.  相似文献

The hypoglycemic and hypolipidemic effects of Lycium barbarum fruit water decoction, crude polysaccharide extracts (crude LBP), and purified polysaccharide fractions (LBP-X) in alloxan-induced diabetic or hyperlipidemic rabbits were investigated through designed sequential trials and by measuring blood glucose and serum lipid parameters. Total antioxidant capacity was also assessed using trolox equivalent antioxidant capacity (TEAC) and oxygen radical absorbance capacity (ORAC) assay. It was found that the three Lycium barbarum fruit extracts/fractions could significantly reduce blood glucose levels and serum total cholesterol (TC) and triglyceride (TG) concentrations and at same time markedly increase high density lipoprotein cholesterol (HDL-c) levels after 10 days treatment in tested rabbits, indicating that there were substantial hypoglycemic and hypolipidemic effects. Hypoglycemic effect of LBP-X was more significant than those of water decoction and crude LBP, but its hypolipidemic effect seemed to be weaker. Total antioxidant capacity assay showed that all three Lycium barbarum extracts/fractions possessed antioxidant activity. However, water and methanolc fruit extracts and crude polysaccharide extracts exhibited stronger antioxidant activity than purified polysaccharide fractions because crude extracts were identified to be rich in antioxidants (e.g., carotenoids, riboflavin, ascorbic acid, thiamine, nicotinic acid). Lycium barbarum polysaccharides (glycocojugates), containing several monosaccharides and 17 amino acids, were major bioactive constituents of hypoglycemic effect. Both polysaccharides and vitamin antioxidants from Lycium barbarum fruits were possible active principles of hypolipidemic effect.  相似文献
