Amino acids present in blood plasma may be targets for oxidation and chlorination by HOCl/OCl(-). N-Chloroamino acids have been reported to be less reactive, but more selective than HOCl/OCl(-) in their reactions; therefore, they may act as secondary mediators of HOCl/OCl(-)-induced injury. This study compared the effects of five N-chloroamino acids (AlaCl, LysCl, SerCl, AspCl and PheCl) on erythrocytes with the action of HOCl/OCl(-). The N-chloroamino acids differed in stability and reactivity. They had a weaker haemolytic action than HOCl/OCl(-); HOCl/OCl(-), AlaCl and PheCl increased osmotic fragility of erythrocytes at a concentration of 1 mm. Oxidation of glutathione, formation of protein-glutathione mixed disulphides and efflux of GSSG from erythrocytes were observed for erythrocytes treated with all the employed chloroderivatives, while increased oxidation of 2',7'-dichlorofluorescin was detected only after treatment of the cells with 1 mm HOCl/OCl(-), AlaCl and PheCl. Generally, the reactivity of at least some N-chloroamino acids may be not much lower with respect to HOCl/OCl(-).  相似文献

l-Alanine adding enzymes from Bacillus subtilis and Bacillus cereus which catalyzed l-alanine incorporation into UDPMurNAc were partially purified and the properties of the enzymes were examined. The enzyme from B. subtilis was markedly stimulated by reducing agents including 2-mercaptoethanol, dithiothreitol, glutathione and cysteine. Mn2+ and Mg2+ activated l-alanine adding activity and their optimal concentrations were 2 to 5 mm and 10 mm, respectively. The optimum pH was 9.5 and the Km for l-alanine was 1.8×10?4m. l-Alanine adding reaction was strongly inhibited by p-chloromercuribenzoate and N-ethyl-maleimide. Among glycine, l- and d-amino acids and glycine derivatives, glycine was the most effective inhibitor of the l-alanine adding reaction. The enzyme from B. cereus was more resistant to glycine than that from B. subtilis. Glycine was incorporated into UDPMurNAc in place of l-alanine, and the Ki for glycine was 4.2×l0?3m with the enzyme from B. subtilis. From these data, the growth inhibition of bacteria by glycine is discussed.  相似文献

An intermediate radical, ?H2OH, was produced in aqueous methanol solution containing nitrous oxide by γ-irradiation. Yields of ethylene glycol and formaldehyde, the major and the minor product from ?H2OH, respectively, changed on the addition of some solutes. Cysteine lowered the both product yields to zero even at a low concentration of 5 × 10?5m. Oxygen of low concentrations (2.5~7.5 × 10?5 m) changed effectively the major product from ethylene glycol to formaldehyde. k (CySH+?H2OH)/k(O2+?H2OH) was calculated as 0.5.

Ascorbic acid (5 × 10?5 m) lowered ethylene glycol yield to 48%, cystine (10?3m) to 15%, methionine (10?3m) to 31%, histidine (10?3m) to 42%, tryptophan (10?3m) 46%, tyrosine (10?3m) to 77%, phenylalanine (10?3m) to 73%, hypoxanthine (10?3m) to 37%, adenine (10?3m) to 52%, uracil (10?3m) to 20%, thymine (10?3m) to 10%, cytosine (10?3 m) to 49%, rutin (10?3m) to 23%, pyrogallol (10?3m) to 41%, and gallic acid (10?3m) to 78% of the control. These results suggest that the reactions of the secondary radicals such as ?H2OH perform an important role in material change of foods irradiated with γ rays.  相似文献

Polyamine oxidase from Penicillium chrysogenum oxidized spermine rapidly and spermidine slightly at pH 7.5. The apparent Km values for spermine and spermidine were calculated to be 2.25 × 10?5 m and 9.54 × 10?6 m, respectively. The relative maximum velocities for spermine and spermidine were 3.37 × 10?3 m (H2O2) per min per mg of protein and 2.08 × 10?4 m (H2O2) per min per mg of protein, respectively. Spermine oxidation of the enzyme was competitively inhibited by spermidine and putrescine. The apparent Ki values by spermidine and putrescine were calculated to be 3.00 × 10?5 m and 1.80 × 10?8 m, respectively. On the other hand, polyamine oxidase from Aspergillus terreus rapidly oxidized both spermidine and spermine at pH 6.5. The apparent Km values for spermidine and spermine were 1.20 × 10?8 m and 5.37 × 10?7 m, respectively. The relative maximum velocities for spermidine and spermine were 1.55 × 10?2 m (H2O2) per min per mg of protein and 6.20 × 10?3 m (H2O2) per min per mg of protein, respectively.

Differential determination of spermine and spermidine was carried out using the two enzymes. The initial rate was assayed with Penicillium enzyme and the end point was measured afte addition of Aspergillus enzyme. Small amounts of polyamines (25 to 200 nmol of spermine and 25 to 250 nmol of spermidine) were assayed by solving two simultaneous equations obtained from the rate assay method and the end point assay method. The calculated values were in close agreement with those obtained by an amino-acid analyzer.  相似文献

The properties of the tyrosinase from Pseudomonas melanogenum was investigated with the crude enzyme preparation. Optimum temperature and pH of the enzyme were 23°C and 6.8, respectively. l-Tyrosine, d-tyrosine, m-tyrosine, N-acetyl-l-tyrosine and l-DOPA were utilized as a substrate by the enzyme. The value for Km obtained were as follows: l-tyrosine 6.90 × 10?4 m, d-tyrosine 1.43 ×10?3 m and l-DOPA 9.90 × 10?4 m. The enzyme was inhibited by chelating agents of Cu2+ l-cysteine, l-homocysteine, thiourea and diethyl-dithiocarbamate and the inhibition was completely reversed by the addition of excess Cu2+ From these results it is concluded that the enzyme is a copper-containing oxidase.  相似文献

The crystalline d-mannitol dehyrogenase (d-mannitol:NAD oxidoreductase, EC 1.1.1.67) catalyzed the reversible reduction of d-fructose to d-mannitol. d-Sorbitol was oxidized only at the rate of 4% of the activity for d-mannitol. The enzyme was inactive for all of four pentitols and their corresponding 2-ketopentoses. The apparent optimal pH for the reduction of d-fructose or the oxidation of d-mannitol was 5.35 or 8.6, respectively. The Michaelis constants were 0.035 m for d-fructose and 0.020 m for d-mannitol. The enzyme was also found to be specific for NAD. The Michaelis constans were 1 × 10?5 m for NADH2 and 2.7 × 10?4 m for NAD.  相似文献

d-Glucose-isomerizing enzyme has been extracted in high yield from d-xylose-grown cells of Bacillus coagulans, strain HN-68, by treating with lysozyme, and purified approximately 60-fold by manganese sulfate treatment, fractionation with ammonium sulfate and chromatography on DEAE-Sephadex column. The purified d-glucose-isomerizing enzyme was homogeneous in polyacrylamide gel electrophoresis and ultracentrifugation and was free from d-glucose-6-phosphate isomerase. Optimum pH and temperature for activity were found to be pH 7.0 and 75°C, respectively. The enzyme required specifically Co++ with suitable concentration for maximal activity being 10?3 m. In the presence of Co++, enzyme activity was inhibited strongly by Cu++, Zn++, Ni++, Mn++ or Ca++. At reaction equilibrium, the ratio of d-fructose to d-glucose was approximately 1.0. The enzyme catalyzed the isomerization of d-glucose, d-xylose and d-ribose. Apparent Michaelis constants for d-glucose and d-xylose were 9×10?2 m and 7.7×10?2 m, respectively.  相似文献

3-Methylthiopropylamine (MTPA) formation from l-methionine in Streptomyces sp. K37 was studied in detail. The reaction was confirmed to be catalyzed by the decarboxylase of l-methionine. The properties of the enzyme were studied in detail using acetone dried cells or cell-free extract. The enzyme was specific for l-methionine. Pyridoxal phosphate stimulated the reaction and protected the enzyme against heat inactivation. The optimum pH for the reaction was 6.0~8.0 and the optimum temperature was about 40°C. Carbonyl reagents (10?2~10?3 m) inhibited the reaction completely, and silver nitrate and mercuric chloride (10?3~10?4 m) markedly inhibited the reaction. Km value for the reaction was 1.21 × 10?5 m. l-Methionine assay using the decarboxylase was attempted and was found to be applicable to practical use.  相似文献

1. Several bacteria were isolated from soil which grew on both d- and l-aminolactam and whose cells had an activity to racemize them. They were identified as Achromobacter obae nov. sp., Achr. cycloclastes, Alcaligenes faecalis and Flavobacterium arborescens.

2. Racemization of d- and l-aminolactam was investigated using the lyophilized cells of Achr. obae nov. sp. The optimum pH value of the reaction was about 8.0. The racemizing activity was completely inhibited by 10?4 m hydroxylamine, and the inhibition was removed by 10?4 m pyridoxal phosphate. Five percent d- and l-aminolactam solutions were completely racemized with a concomitant slight formation of l-lysine.  相似文献

Mutants resistant to α-amino-β-hydroxyvaleri0c acid (AHV) were derived from various bacteria which belong to Corynebacterium, Brevibacterium, Arthrobacter, Microbacterium, or Bacillus by mutational treatment with N-methyl-N′-nitro-N-nitrosoguanidine(NTG), and screened for their ability to produce l-threonine. A number of l-threonine producers were obtained from each group of bacteria. Among them, the mutants derived from C. glutamicum KY9159(Met?) were further mutagenized with NTG to derive thialysine(S-Lys)-resistant mutants. An AHV-resistant mutant, KY10484 was proved to be much more sensitive to the growth inhibition by thialysine than the parent strain, KY9159. From KY10484, a number of AHV- and thialysine-resistant mutants were derived. Approximately a half of these mutants were found to produce more l-threonine than KY10484. Among these mutants, KY10440 (Met?, AHVR, s-LysR) was used to investigate the cultural conditions for l-threonine production. The growth of KY10440 decreased largely with addition of l-homoserine, a threonine precursor. l-Asparagine, l-cystine, l-glutamine or l-arginine partially reversed the inhibitory effect of l-homoserine. Addition of these amino acids at low level led to increase l-threonine production. The amount of l-threonine accumulation reached to a level of 14mg/ml with a medium containing 10% glucose and to a level of 10 mg/ml with a medium containing 5% molasses (as glucose).

Another AHV- and thialysine-resistant mutant, KY10251 which was also derived from KY9159 was found to produce both 9 mg/ml of l-threonine and 5.5 mg/ml of l-lysine in a culture broth.  相似文献

The substrate specificity and the mode of action of the protease from Streptomyces cellulosae were investigated, using many kinds of peptides and proteins as substrates. The protease hydrolyzed peptides consisting of hydrophobic amino acids such as L-Phe-L-Leu-NH2, L-Pro-L-Phe-NH2, l-Leu-L-Met, L-Leu-L-Leu, Gly-L-Ile, L-Phe-L-Phe, L-Pro-L-Leu-Gly-NH2, etc. The protease hydrolyzed zein best among the proteins tested, but weakly hydrolyzed gelatin, myoglobin, bovine serum albumin, γ-globulin, and collagen. The protease mainly hydrolyzed Ser12-Leu13, Leu13-Tyr14, and Tyr14-Gln15 bonds in the oxidized A-chain of insulin and at least the Leu15-Tyr16 bond in the oxidized B-chain of insulin.  相似文献

2-Ketogluconate reductase (2KGR) from the cell free extract of Gluconobacter liquefaciens (IFO 12388) was purified about 1000-fold by a procedure involving ammonium sulfate fractionation and column chromatographies using DEAE-cellulose, hydroxylapatite, and Sephadex gel The purified enzyme gave a single band on polyacrymamide gel electrophoresis. NADP was specifically required for the oxidation reaction of gluconic acid. Using gel filtration a molecular weight of about 110,000 was estimated for the enzyme. The pH optimum for the oxidation of gluconic acid (GA) to 2-ketogluconic acid (2KGA) by the enzyme was 10.5 and for the reduction of 2KGA was 6.5. The optimum temperature of the enzyme was 50 C for both reactions of oxidation and reduction. The enzyme was stable at pH between 5.0 and 11.0 and at temperature under 50°C, The enzyme activity was strongly inhibited with p-chloromercuribenzoate and mercury ions, but remarkably stimulated by manganese ions (1×10?3 m). Km value of the enzyme for GA was 1.3×10?2 m and for 2KGA was 6.6×10?3 m. Km values for NADP and NADPH2 were 1.25×10?5 and 1.52×10?5 m respectively.  相似文献

Effects of the substrate and the coenzyme on the crystalline yeast phosphoglyceric acid mutase activity have been investigated. Lineweaver-Burk plots at different concentrations of the substrate (d-3-phosphoglyceric acid: 3×10?7 to 8×10?3m) and the coenzyme (d-2, 3-diphosphoglyceric acid: 8×10?7 to 10?5m) change in such a way to indicate the involvement of an enzyme-substrate-coenzyme ternary complex as an active intermediate in the enzymic reaction process. It is concluded that the reaction catalyzed by the yeast enzyme follows the sequential pathway and that a phosphorylated enzyme does not participate as an obligatory intermediate in the reaction mechanism, if it occurs. Kinetic studies indicate Km values of 6×10?4m for d-3-phosphoglyceric acid and 8×10?7m for d-2, 3-diphosphoglyceric acid. The substrate is a competitive inhibitor of the coenzyme with a Ksi (inhibition constant) of 3.2×10?3m. The coenzyme inhibition is not observed at concentration tested. A kinetic treatment to determine the mechanism of the enzyme reaction from the experimental data which are obtaind in the range of inhibitory substrate concentrations is presented.  相似文献

Regulatory properties of chorismate mutase from Corynebacterium glutamicum were studied using the dialyzed cell-free extract. The enzyme activity was strongly feedback inhibited by l-phenylalanine (90% inhibition at 0.1~1 mm) and almost completely by a pair of l-tyrosine and l-phenylalanine (each at 0.1~1 mm). The enzyme from phenylalanine auxotrophs was scarcely inhibited by l-tyrosine alone but the enzyme from a wild-type strain or a tyrosine auxotroph was weakly inhibited by l-tyrosine alone (40~50% inhibition, l-tyrosine at 1 mm). The enzyme activity was stimulated by l-tryptophan and the inhibition by l-phenylalanine alone or in the simultaneous presence of l-tyrosine was reversed by l-tryptophan. The Km value of the reaction for chorismate was 2.9 } 10?3 m. Formation of chorismate mutase was repressed by l-phenylalanine. A phenylalanine auxotrophic l-tyrosine producer, C. glutamicum 98–Tx–71, which is resistant to 3-amino-tyrosine, p-aminophenylanaine, p-fluorophenylalanine and tyrosine hydroxamate had chorismate mutase derepressed to two-fold level of the parent KY 10233. The enzyme in C. glutamicum seems to have two physiological roles; one is the control of the metabolic flow to l-phenylalanine and l-tyrosine biosynthesis and the other is the balanced partition of chorismate between l-phenylalanine-l-tyrosine biosynthesis and l-tryptophan biosynthesis.  相似文献

d-Gluconate dehydrogenase catalyzing the oxidation of d-gluconate to 2-keto-d-gluconate was solubilized with Triton X-100 from the membrane of Gluconobacter dioxyacetonicus IFO 3271 and purified to an almost homogeneous state by chromatographies on DEAE-cellulose and CM-Toyopearl in the presence of 0.1% Triton X-100. The enzyme had three subunits with molecular weights of 64,000, 45,000 and 21,000, and contained approximately 2 mol of heme per mol of the enzyme. The prosthetic group of the dehydrogenase was found to be a flavin covalently bound to the enzyme protein. The substrate specificity of the purified enzyme was very strict for d-gluconate and the apparent Michaelis constant for d-gluconate was 2.2 mm. The optimum pH and temperature of the purified enzyme were 6.0 and 40°C, respectively.  相似文献

The phenolsulphatase of Aerobacter aerogenes was found to be formed in cell suspensions containing tyramine and casein hydrolyzate under aerobic conditions in a few hours.

Factors affecting phenolsulphatase synthesis in cell suspensions were studied.

The relationship between the rate of enzyme activity formed and the level of tyramine concentration in 10?4 m to 10?3 m is linear. The optimum concentration of tyramine for its utilization and phenolsulphatase induction is in both cases 5 × 10?3 m. The opt. concentration of casamino acids for phenolsulphatase induction is 0.2 per cent and the opt. pH is 7.2. Agents such as 2,4-dinitrophenol, sodium azide, arsenate and potassium cyanide inhibit the induction of this enzyme. The external inducer of phenolsulphatase should be highly specific. On addition of tyramine, hydroxytyramine and noradrenalin are effective, although the effect of noradrenalin is very weak.  相似文献

A tetrathionate-decomposing enzyme that catalyzes the decomposition of tetrathionate into thiosulfate and sulfate was purified to homogeneity from tetrathionate-grown Thiobacillus thiooxidans. The enzyme had an apparent molecular weight of 104,000, and was composed of two identical subunits (MW = 58,000) as estimated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The enzyme had an isoelectric point at 9.6 and was most active at pH 3.0–3.5 and 40°C. Enzyme activity was increased approximately 100-fold in the presence of 400 mm sulfate ion. The Michaelis constant of this enzyme for tetrathionate in the presence of 20, 50, and 200 mm of sulfate ion was 2.4 mm. Mercuric and ferric ions completely inhibited the enzyme activity at 1 mm. Though cupric ion up to 0.01 mm markedly stimulated the activity in the presence of 20 mm sulfate ion, a higher concentration (1 mm) rather strongly inhibited the activity. Ethylenediaminetetraacetic acid (EDTA) strongly inhibited the activity, but this inhibiton was completely restored by cupric ion.  相似文献

Candida species YK 11 and YK 92 and Geotrichum candidum YK 57, which were isolated as nitrite-resistants, converted nitrite in the culture medium to nitrate stoichiometrically during growth. The nitrite-oxidizing reaction was confirmed under aerobic conditions in the intact cell system with 15 mm nitrite, 150 mm glucose, and 100mm Tris-HCl buffer (pH 7.0). Glucose or other carbohydrate which supported the microbial growth was indispensable for the reaction. The rate of oxidation (0.9 ~ 1.3 × 105 μg-N/g of YK 92 cells·day) and the maximum amounts of nitrate formed in the culture medium (200 mm, 2800 μg-N/ml) were much larger than those of other heterotrophic nitrifiers and almost the same as those of Nitrobacter.

The nitrite-oxidizing activity was demonstrated in many types of yeast species.  相似文献

l-Fucose (l-galactose) dehydrogenase was isolated to homogeneity from a cell-free extract of Pseudomonas sp. No 1143 and purified about 380-fold with a yield of 23 %. The purification procedures were: treatment with polyethyleneimine, ammonium sulfate fractionation, chromatographies on phenyl-Sepharose and DEAE-Sephadex, preparative polyacrylamide gel electrophoresis, and gel filtration on Sephadex G-100. The enzyme had a molecular weight of about 34,000. The optimum pH was at 9 — 10.5 and the isoelectric point was at pH 5.1. l-Fucose and l-galactose were effective substrates for the enzyme reaction, but d-arabinose was not so much. The anomeric requirement of the enzyme to l-fucose was the β-pyranose form, and the reaction product from l-fucose was l-fucono- lactone. The hydrogen acceptor for the enzyme reaction wasNADP+, and NAD + could be substituted for it to a very small degree. Km values were 1.9mm, 19mm, 0.016mm, and 5.6mm for l-fucose, l- galactose, NADP+, and NAD+, respectively. The enzyme activity was strongly inhibited by Hg2 +, Cd2 +, and PCMB, but metal-chelating reagents had almost no effect. In a preliminary experiment, it was indicated that the enzyme may be usable for the measurement of l-fucose.  相似文献

The effects of phytate on the hydrolysis of lactose with β-galactosidases from bovine liver and Escherichia coli were investigated. The activities of both β-galactosidases were decreased to the same extent by increased concentrations of phytate. The rates of inhibition of β-galactosidase activity from E. coli in a reaction mixture containing 10 mm of phytate were 78.9% and 64.4%, respectively, in the absence of and with 4 mm of Mg2 +. Therefore, it was found that the stimulatory effect of Mg2+ was hardly affected by the presence of phytate in the range from 2 to 10 mm. The β-galactosidase activity was also not influenced by preincubating β-galactosidase or lactose with phytate. Kinetic studies showed that the inhibition of β-galactosidase activity by phytate was of an uncompetitive type with a Ki value of 3.46 mm. Therefore, it is considered that phytate may interact with a complex of ß-galactosidase and lactose.  相似文献
