Experimental evidence on the protective properties of S-allylcysteine (SAC) was collected from three models exerting striatal toxicity. In the first model, SAC (120 mg kg(-1)x5) prevented lipoperoxidation (LP) and mitochondrial dysfunction (MD) in synaptosomal fractions from 1-methyl-4-phenyl-1,2,3,6-tetrahydropiridinium-treated mice (30 mg kg(-1)), but without complete restoration of dopamine levels. In the second model, SAC (300 mg kg(-1)x 3), prevented LP and MD in synaptosomes from rats infused with 6-hydroxydopamine (8 microg microl(-1)) into the substantia nigra pars compacta, but again, without total reversion of depleted dopamine levels. In the third model, SAC (100 mg kg(-1)x 1) prevented MD in synaptosomes from rats injected with 3-nitropropionic acid (10 mg kg(-1)), but in contrast to the other models, it failed to prevent LP. SAC also prevented the aberrant motor activity patterns evoked by the three toxins. Altogether, the results suggest that the antioxidant properties of SAC are responsible for partial or total preservation of neurochemical, biochemical and behavioural markers, indicating that pro-oxidant reactions underlie the neurotoxicity in these models.  相似文献

The effects of 5-HT and glutamate on dopamine synthesis and release by striatal synaptosomes were investigated and compared with the action of acetylcholine, which acts presynaptically on this system. 5-HT inhibited (28%) synthesis of [14C]dopamine from L-[U-14C]tyrosine, at 10-5M and above. This contrasts with the action of acetylcholine, which stimulated [14C]-dopamine synthesis by 24% at 10-4 M. Tissue levels of GABA were unaffected by either 5-HT or acetylcholine up to concentrations of 10-4 M. The inhibitory action of 5-HT (5 × 10?5 M and 2 × 10?4 M) on [19C]dopamine synthesis was completely abolished by methysergide (2 × 10?6 M). Higher concentrations of methysergide (10?4 M) or cyproheptadine (10?5 M) inhibited [14C]dopamine synthesis by 28% and 25%, respectively, when added alone to synaptosomes. However, only methysergide prevented the further inhibition of synthesis caused by 5-HT. At concentrations of 2 × 10?5 M and above, 5-HT stimulated [14C]dopamine release. This releasing action differed from that of acetylcholine, which occurred at lower concentrations (e.g., 10?6 M). Methysergide (up to 10?4 M) or cyproheptadine (2 × 10?4 M) did not reduce the 5-HT (5 × 10?5 M)-induced release of [14C]dopamine, but methysergide (10?4 M) showed a potentiation (49%) of this increased release. The stimulatory effects of 5-HT (2 × 10?5 M) and K+ (56 mM) on [14C]dopamine release were additive, indicating that two separate mechanisms were involved. However, when both agents were present the stimulatory effect of K+ (56 mM) on [14C]dopamine synthesis was not seen above the inhibitory effect of 5-HT. Glutamate (0.1-5 mM) did not affect [4C]dopamine release or its synthesis from L-[U-14C]tyrosine. It is concluded that 5-HT modulates the synthesis of dopamine in striatal nerve terminals through a presynaptic receptor mechanism, an action antagonised by methysergide. The releasing action of 5-HT apparently occurs through a separate mechanism which is also distinct from that involved in the response to K+ depolarisation.  相似文献

The concentrations of the 16 U.S. Environmental Protection Agency polycyclic aromatic hydrocarbons (PAHs) were measured in urban street dust with a view to understanding the potential risk to urban residents exposed to street dust, sources, and distribution of the PAHs. Gas chromatography–mass spectrometry was used to measure the concentrations of the PAHs after ultra-sonication with hexane/dichloromethane and clean up. The total concentrations of the ?16 PAHs in the urban street dust from Warri and its environs ranged from 165.1 to 1012 µg kg?1. The isomeric ratios and the PCA indicated that combustion of petroleum and biomass are the major sources of PAHs in Warri and its environs. The total BaP carcinogenic and mutagenic potency concentrations (?BaPTEQ and ?BaPMEQ) were in the range of 0.03 to 219 µg kg?1 and 0.52 to 182 µg kg?1, respectively. The incremental life cancer risk from exposure of the residents of Warri and its environs ranged from 4.07 × 10?7 to 3.11 × 10?3 and 2.13 × 10?7 to 1.49 × 10?3 for the children and adults, respectively, which were higher than the baseline value of acceptable risk of 10?6 (i.e., one case per million people).  相似文献

Exposure to benzo(a)pyrene (B(a)P) for health risk was studied in soils from the Delhi region, India. The mean and median concentrations of benzo(a)pyrene were 0.031 and 0.029 (±0.002) mg/kg, respectively. The lifetime average daily dose (LADD) for adults and children was 1.7 × 10?8 mg kg?1 d?1 and 7.5 × 10?8 mg kg?1 d?1, respectively, with incremental life time cancer risk (ILCR) of 1.2 × 10?7 and 5.5 × 10?7, respectively. The Index of Additive Cancer Risk (IACR) was 0.084. Our screening-level risk assessment shows that the observed ILCR and IACR values are much lower than the guideline values of 10?6 ? 10?4 (ILCR) and <1 (IACR), respectively, and therefore the measured B(a)P levels in soil may not portend environmental and human health risks.  相似文献

The critical value of soil Olsen-P is the point above which the probability of crop yield response to fertilizer P is small or nil. Determining this critical value is fundamental when making appropriate P fertilizer recommendations. In this study, the critical values were determined for continuous maize (Zea mays L.)-winter wheat (Triticum aestivum L.) cropping systems from a 15-year field experiment across three sites in China using linear-linear, linear-plateau and Mitscherlich models. The mean critical values for maize using the three models ranged from 12.1 to 17.3 mg P kg?1 (average 15.3 mg P kg?1) and for winter wheat from 12.5 to 19.0 mg P kg?1 (average 16.3 mg P kg?1) among study sites. The mean critical value for maize was approximately 7% lower than that for winter wheat across all sites based on the three models. Critical values identified by the Mitscherlich model were 1.4 to 2.1 times those from linear-linear and 1.3 to 1.9 times of those from linear-plateau and were crop and site dependent. There was a significant negative correlation (P?<?0.05) between the mean critical value from the three models and the observed P uptake by either maize or wheat. Our study shows that the critical values can vary with sites, crops and models used, and thus caution should be taken when selecting the most appropriate one when making P fertilizer recommendations for agronomic return and to minimize chances of negative environment impact from overfertilization.  相似文献

This study assessed the interactive effect of selenium (Se) and farmyard manure (FYM) on soil microbial activities, growth, yield, and Se accumulation by wheat grains. Preliminarily, the effect of Se (0–250 µg kg?1 soil) and FYM (0–12.5 g kg?1 soil) was assessed on soil microflora. Selenium exhibited an adverse impact on soil microflora; respiration was decreased at?≥?10 µg kg?1 soil while dehydrogenase and urease activities were decreased at?≥?125 µg kg?1 soil. At 250 µg Se kg?1 soil, respiration, dehydrogenase and urease activities were decreased by 81, 40 and 35%, respectively, on unamended soil, and by 9, 47 and 22%, respectively, on FYM-amended soil. The subsequent plant experiments were conducted with same Se and FYM rates; one was harvested 42 days after sowing and other at crop maturity. The application of 125 µg Se kg?1 and 12.5 g FYM kg?1 soil improved seedling biomass by 12.6 and 22%, respectively, while their combined use lacked synergistic effect. Similarly, at maturity Se and FYM increased grain yield while their combined effect was not synergistic. The Se-induced suppression in microbial activities was not related to yield which was improved (11% at the highest rate in unamended soil) by Se application. Selenium application increased grain Se content in a rate-dependent manner, it increased from 0 to 1025 µg kg?1 by applying 250 µg Se kg?1 soil. Moreover, FYM application decreased Se accumulation in grains. It is concluded that FYM application increased soil microbial activities and yield but reduced grain Se accumulation in wheat on Se-applied soil.

In this study, HPLC-MS and ICP-MS methods were used for the determination of histidine and nickel in Morus L., Robinia pseudoacacia L., and Populus nigra L. leaves taken from industrial areas including Gaziantep and Bursa cities. In the determination of histidine by HPLC-MS, all of the system parameters such as flow rate of mobile phase, fragmentor potential, injection volume and column temperature were optimized and found to be 0.2 mL min?1, 70 V, 15 µL, and 20°C, respectively. Under the optimum conditions, histidine was extracted from plant sample by distilled water at 90°C for 30 min. Concentrations of histidine as mg kg?1 were found to be between 2–9 for Morus L., 6–13 for Robinia pseudoacacia L., and 2–10 for Populus nigra L. Concentrations of nickel were in the ranges of 5–10 mg kg?1 for Morus L., 3–10 mg kg?1 for Robinia pseudoacacia L., and 0.6–4 mg kg?1 for Populus nigra L. A significant linear correlation (r = 0.78) between histidine and Ni was observed for Populus nigra L., whereas insignificant linear correlation for Robinia pseudoacacia L. (r = 0.22) were seen. Limits of detection (LOD) and quantitation (LOQ) were found to be 0.025 mg Ni L?1 and 0.075 mg Ni L?1, respectively.  相似文献

The experiment was organized in a 3×2 factorial arrangement with three dietary fat blends and a basal (20 mg kg?1 diet) or supplemented (220 mg kg?1) level of α-tocopheryl acetate. Dietary vitamin E and monounsaturated to polyunsaturated fatty acid ratio (dietary MUFA/PUFA) affected muscle α-tocopherol concentration (α-tocopherol [log μg g?1]=0.18 (±0.105)+0.0034 (±0.0003)·dietary α-tocopherol [mg kg?1 diet] (P<0.0001)+0.39 (±0.122)·dietary MUFA/PUFA (P<0.0036)). An interaction between dietary α-tocopherol and dietary MUFA/PUFA exists for microsome α-tocopherol concentration (α-tocopherol [log μg g?1]=1.14 (±0.169) (P<0.0001)+0.0056 (±0.00099)·dietary α-tocopherol [mg kg?1 diet] (P<0.0001)+0.54 (±0.206)·dietary MUFA/PUFA (P<0.0131)?0.0033 (±0.0011)·dietary α-tocopherol [mg kg?1)]×dietary MUFA/PUFA (P<0.0067)), and hexanal concentration in meat (hexanal [ng·g?1]=14807.9 (±1489.8)?28.8 (±10.6) dietary α-tocopherol [mg·kg?1] (P<0.01)?8436.6 (±1701.6)·dietary MUFA/PUFA (P<0.001)+24.0 (±11.22)·dietary α-tocopherol·dietary MUFA/PUFA (P<0.0416)). It is concluded that partial substitution of dietary PUFA with MUFA lead to an increase in the concentration of α-tocopherol in muscle and microsome extracts. An interaction between dietary α-tocopherol and fatty acids exists, in which at low level of dietary vitamin E inclusion, a low MUFA/PUFA ratio leads to a reduction in the concentration of α-tocopherol in microsome extracts and a concentration of hexanal in meat above the expected values.  相似文献

The concentrations of Pb and Cd, and trace elements (Cu and Zn) in the urban topsoil, rook (Corvus frugilegus) feces and feathers and human scalp hair were analyzed to examine the potential ecological risk posed by Pb and Cd on local residents of Qiqihar City, northeastern China. Results revealed that the Cd concentrations in the topsoil were ranged from 0.14 to 3.55 mg kg?1 dry weight (dw). The maximal geoaccumulation indices [a value from logarithmic (a measured metal content/1.5 × background content of the metal in this region), introduced by Muller] of Cd exceeded 3.5, which suggested that this region was seriously contaminated by Cd. The corresponding average detectable concentrations in C. frugilegus feathers and feces were 1.38 and 3.97 mg kg?1 dw for Pb and 1.04 and 0.69 mg kg?1 dw for Cd. High Pb and Cd concentrations, respectively, ranging from 7.46 to 24.9 mg kg?1 dw and from 0.35 to 0.92 mg kg?1 dw were also detected in the human scalp hair samples. These high Pb and Cd concentrations in C. frugilegus and local people were possibly associated with local industrial wastes and vehicle exhausts. The external tissues (feces and feather) of the rook species can be considered as an indicator of potential Cd toxic risk in this species; however, the human scalp hair is not a reliable biomarker for risk of Pb and Cd in the human being. Effective measures should be established to reduce the inputs of Pb and Cd into the urban environment and to protect the health of local people.  相似文献

Epidermal growth factor(EGF) has been shown to improve piglet intestinal morphology and epithelial recovery. In an attempt to further understand the mechanisms behind these improvements, this study tested the hypothesis that dietary EGF may affect intestinal morphology by stimulating the proliferation and differentiation of enterocytes in weaning piglets. In piglets receiving200 μg kg–1 EGF, crypt depth and villus height increased(P0.05). Adding 400 μg kg–1 EGF increased villus height-to-crypt depth ratio(P0.05), but reduced crypt depth(P0.05). Dietary supplementation with 200 μg kg–1 EGF significantly increased the number of Ki67-positive cells(P0.01) and tended to increase the mRNA level of proliferating cell nuclear antigen(P0.10).However, this supplementation decreased the expression level of intestinal fatty acid-binding protein(P0.05). Piglets fed with400 μg kg–1 EGF had an increased mRNA level of intestinal alkaline phosphatase(P0.05). The phosphorylation of m TOR(mammalian target of rapamycin) was observed in the 200 μg kg–1 EGF group. These results suggest that dietary supplementation with a low level of EGF improved piglet intestinal morphology through stimulating the proliferation and differentiation of enterocytes, and the mTOR signaling pathway may partly be involved in this process.  相似文献

Kinetic parameters for high affinity [HA] uptake in vitro in synaptosomes from different mouse brain regions were investigated. Vmax was highest in the striatum [200 pmol.· mg protein?1 · 4 min?1], followed by the cortex [111 pmol · mg protein?1 · 4 min?1], hippocampus [63 pmol · mg protein?1 · 4 min?1], midbrain [21 pmol · mg protein?1 · 4 min?1] and, lowest, medulla oblongata [5 pmol · mg protein?1 · 4 min?1]. Km was about the same in all brain regions [0.9–1.4 μM]. No sign of HA uptake was detected in synaptosomes from the cerebellum. A clear relationship between Vmax for synaptosomal HA uptake of Ch in vitro and apparent turnover of ACh in vivo was found between the brain regions. Administration of oxotremorine [1 mg·kg?1 i.p.] decreased Vmax for HA uptake of Ch by 60% in the cortex and hippocampus, by 50% in the striatum and by 20% in the midbrain. This effect is in accordance with the previously observed marked decrease in turnover of ACh in these brain regions following oxotremorine treatment.  相似文献

A nitrogen-based model of maintenance respiration (Rm) would link Rm with nitrogen-based photosynthesis models and enable simpler estimation of dark respiration flux from forest canopies. To test whether an N-based model of Rm would apply generally to foliage of boreal and subalpine woody plants, I measured Rm (CO2 efflux at night from fully expanded foliage) for foliage of seven species of trees and shrubs in the northern boreal forest (near Thompson, Manitoba, Canada) and seven species in the subalpine montane forest (near Fraser, Colorado, USA). At 10°C, average Rm for boreal foliage ranged from 0.94 to 6.8μmol kg?1 s?1 (0.18–0.58 μmol m?2 s?1) and for subalpine foliage it ranged from 0.99 to 7.6 μmol kg?1 s?1 (0.28–0.64μmol m?2 s?1). CO2 efflux at 10°C for the samples was only weakly correlated with sample weight (r = 0.11) and leaf area (r = 0.58). However, CO2 efflux per unit foliage weight was highly correlated with foliage N concentration [r = 0.83, CO2 flux at 10°C (mol kg?1 s?1) = 2.62 × foliage N (mol kg?1)J, and slopes were statistically similar for the boreal and subalpine sites (P=0.28). CO2 efflux per unit of foliar N was 1.8 times that reported for a variety of crop and wildland species growing in warmer climates.  相似文献

This study aimed to investigate the effects of the 5-HT7 receptor agonist (LP44) and antagonist (SB269970) on LPS-induced in vivo tissue damage and cell culture by molecular methods. This study was conducted in two steps. For in vivo studies, 24 female rats were divided into four groups. Group I: healthy; II (2nd h): LPS 5 mg/kg administered intraperitoneally (i.p.); III (4th h): LPS 5 mg/kg administered i.p.; IV (8th h): LPS 5 mg/kg administered i.p. For in vitro studies, we used the A549 cell line. Groups: I control (healthy) (2–4 h); II LPS: 1 µg/ml E. Coli O55:B5 strain (2–4 h); III agonist (LP44) 10?9 M (2–4 h); IV antagonist (SB269970) 10?9 M (2–4 h); V LPS+agonist 10?9 M (LP44 1 µg/ml) (2–4 h); VI LPS+antagonist 10?9 M (2–4 h). In molecular analyses, we determined increased TNF-α, IL-1β, NF-κB, and 5-HT7 mRNA expressions in rat lung tissues and increased TNF-α, iNOS, and 5-HT7 mRNA expressions in the A549 cell line. In in vitro parameters, LP44 agonist administration-related decrease was observed. Our study showed that lung 5-HT7 receptor expression is increased in LPS-induced endotoxemia. All this data suggest that 5-HT7 receptor overexpression is an important protective mechanism during LPS-induced sepsis-related cell damage.  相似文献

Phytoextraction of Risk Elements by Willow and Poplar Trees   总被引:1,自引:0,他引:1  
To characterize the phytoextraction efficiency of two clones of willow trees (Salix x smithiana Willd., Salix rubens) and two clones of poplar trees (Populus nigra x maximowiczii, Populus nigra Wolterson) were planted in contaminated soil (0.4–2.0 mg Cd.kg?1, 78–313 mg Zn.kg?1, 21.3–118 mg Cu.kg?1). Field experiment was carried out in Czech Republic. The study investigated their ability to accumulate heavy metals (Cd, Zn, and Cu) in harvestable plant parts. The poplars produced higher amount of biomass than willows. Both Salix clones accumulated higher amount of Cd, Zn and Cu in their biomass (maximum 6.8 mg Cd.kg?1, 909 mg Zn.kg?1, and 17.7 mg Cu.kg?1) compared to Populus clones (maximum 2.06 mg Cd.kg?1, 463 mg Zn.kg?1, and 11.8 mg Cu.kg?1). There were no significant differences between clones of individual species. BCs for Cd and Zn were greater than 1 (the highest in willow leaves). BCs values of Cu were very low. These results indicate that Salix is more suitable plant for phytoextraction of Cd and Zn than Populus. The Cu phytoextraction potential of Salix and Populus trees was not confirmed in this experiment due to low soil availability of this element.  相似文献

Concentrations of four metals (Cu, Zn, Pb, and Cd) in the sediments of the Anzali Lagoon in the northern part of Iran were determined to evaluate the level of contamination and spatial distribution. The sediments were collected from 21 locations in the lagoon. At each lagoon site a core, 60 cm long, was taken. The ranges of the measured concentrations in the sediments are as follows: 17–140 mg kg?1 for Cu, 20–113 mg kg?1 for Zn, 1–37 mg kg?1 for Pb and 0.1–3.5 mg kg?1 for Cd in surficial (0-20 cm) and 16–87 mg kg?1 for Cu, 28.5–118 mg kg?1 for Zn, 3–20 mg kg?1 for Pb and 0.1–3.5 mg kg?1 for Cd in deep (40–60 cm) sediments. The results of the geoaccumulation index (Igeo) show that Cd causes moderate to heavy pollution in most of the study area. Environmental risk evaluation showed that the pollution in the Anzali Lagoon is moderate to considerable and the ranking of the contaminants followed the order: Cd > Cu > Pb > Zn. Some locations present severe pollution by metals depending on the sources, of which sewage outlets and phosphate fertilizers are the main sources of contaminants to the area.  相似文献

The effectiveness of three hormone treatments commonly used for artificial reproduction of tench was evaluated under three thermal regimes, with focus on the need for dopamine antagonist. Mature females were divided into nine groups (n?=?8) and gradually exposed, over a 24?h period, to three temperature regimes: cold (18.1?±?0.02?°C), optimal (22.05?±?0.03?°C), and warm (26.3?±?0.01?°C). Each temperature regime comprised three experimental groups injected with one of three hormone treatments: carp pituitary extract (CPE; 3?mg?kg?1); [D-Arg6, Pro9, NEt]-sGnRH (10???g?kg?1); and [D-Arg6, Pro9, NEt]-sGnRH (10???g?kg?1)?+?metoclopramide (20?mg?kg?1) (combined treatment). No differences were found between ovulation induction (ovulation rate????75?%) with sGnRHa alone and with the combined treatment; whereas CPE at cold and warm water temperatures was significantly less effective (P?<?0.05) than above mentioned treatments. Administration of sGnRHa alone induced a gradual increase in luteinizing hormone (LH) levels with LH peaks close to ovulation, in contrast to the immediate LH surge with high LH levels throughout the entire study observed with the combined treatment under all thermal regimes. LH levels induced by GnRHa alone were significantly lower (P?<?0.05) at all temperatures compared to the combined treatment, with the exception of the final sample at 26?°C, when no difference was recorded. Based on these results we recommend the application of sGnRHa without the addition of dopamine antagonist as a reliable method for inducing ovulation in tench under suboptimal temperature conditions.  相似文献

Dichlorodiphenyltrichloroethane (DDT), hexachlorocyclohexane (HCH), and their isomers’ levels in residential soils were determined for the assessment of health risk in Korba, India. Observed concentrations of total HCH and total DDT in soils were more or less comparable with other parts of India and the world. ΣHCH and ΣDDT concentrations ranged between 0.9–20 μg kg?1 and 2–315 μg kg?1, respectively, which were lower than recommended soil quality guidelines indicating low ecotoxicological risk. Carcinogenic and non-carcinogenic impacts of HCH and DDT on human populations through soil ingestion were evaluated and presented. The incremental lifetime cancer risk (ILCR) for adults and children ranged between 7.8 × 10?10–1.6 × 10?7 and 4.1 × 10?9–8.2 × 10?7, respectively. Non-cancer health hazard quotient (HQ) ranged between 5.9 × 10?7–1.8 × 10?3 and 3.1 × 10?6–9.4 × 10?3, respectively, for adults and children. The estimated ILCR and HQ were within the safe acceptable limits of 10?6–10?4 and ≤1.0, respectively, indicating low risk to human populations from exposure to organochlorine pesticides (HCH and DDT) in the study area.  相似文献

Samples of water, sediments and aquatic weeds were collected from 26 sites in the Nyando River catchment of the Lake Victoria basin in 2005–2006. The objective was to investigate levels of organochlorine pesticides that have either been banned or are restricted for use in Kenya. The pesticides investigated were lindane, aldrin, endosulfan, endrin, dieldrin, DDT, heptachlor and methoxychlor. These pesticides had previously found wide applications in public health and agriculture in Kenya for control of disease vectors and crop pests respectively. Results showed that mean concentrations were highest for methoxychlor (8.817 ± 0.020?µg l?1) in water, sediments (92.893 ± 3.039 µg kg?1), and weeds (39.641 ± 3.045?µg kg?1), the weeds also tended to accumulate aldrin (15.519 ± 3.756?µg kg?1). The results show that the pesticides are still in use and are detected in the catchment. Stringent management and public awareness measures are required to enforce the ban on the organochlorine pesticides in order to safeguard the environment and ecosystems of Lake Victoria.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) in the surface urban soils of Shenyang in Northeastern China were investigated. The total concentration of the PAHs ranged from 0.09 to 8.35 mg kg?1, with an average value of 1.51 ± 1.64 mg kg?1. 3–5-ring PAHs accounted for 90% of total PAHs. The functional areas, such as the industrial regions (4.95 mg kg?1) and main roads (1.56 mg kg?1), as well as the administrative divisions, including the districts of Shenhe (1.49 mg kg?1), Heping (2.08 mg kg?1), and Tiexi (2.14 mg kg?1), were heavily polluted by PAHs. The diagnostic ratios and principal component analysis (PCA) for PAHs indicate that the pollutants probably originated primarily from coal combustion and petroleum sources. The Nemerow composite index, used to assess environmental quality, shows that the soil samples were heavily polluted with PAHs, and although 52.8% of the soil sampling sites were safe, 47.2% of the soil sampling sites registered different grades of PAH pollution. Synovial fluid (SF) contains lubricant macromolecules, hyaluronan (HA), and proteoglycan 4 (PRG4). The synovium not only contributes lubricants to SF through secretion by synoviocyte lining cells, but also concentrates lubricants in SF due to its semi‐permeable nature. A membrane that recapitulates these synovium functions may be useful in a bioreactor system for generating a bioengineered fluid (BF) similar to native SF. The objectives were to analyze expanded polytetrafluoroethylene membranes with pore sizes of 50 nm, 90 nm, 170 nm, and 3 µm in terms of (1) HA and PRG4 secretion rates by adherent synoviocytes, and (2) the extent of HA and PRG4 retention with or without synoviocytes adherent on the membrane. Experiment 1: Synoviocytes were cultured on tissue culture (TC) plastic or membranes ± IL‐1β + TGF‐β1 + TNF‐α, a cytokine combination that stimulates lubricant synthesis. HA and PRG4 secretion rates were assessed by analysis of medium. Experiment 2: Bioreactors were fabricated to provide a BF compartment enclosed by membranes ± adherent synoviocytes, and an external compartment of nutrient fluid (NF). A solution with HA (1 mg/mL, MW ranging from 30 to 4,000 kDa) or PRG4 (50 µg/mL) was added to the BF compartment, and HA and PRG4 loss into the NF compartment after 2, 8, and 24 h was determined. Lubricant loss kinetics were analyzed to estimate membrane permeability. Experiment 1: Cytokine‐regulated HA and PRG4 secretion rates on membranes were comparable to those on TC plastic. Experiment 2: Transport of HA and PRG4 across membranes was lowest with 50 nm membranes and highest with 3 µm membranes, and transport of high MW HA was decreased by adherent synoviocytes (for 50 and 90 nm membranes). The permeability to HA mixtures for 50 nm membranes was ~20 × 10?8 cm/s (? cells) and ~5 × 10?8 cm/s (+ cells), for 90 nm membranes was ~35 × 10?8 cm/s (? cells) and ~19 × 10?8 cm/s (+ cells), for 170 nm membranes was ~74 × 10?8 cm/s (± cells), and for 3 µm membranes was ~139 × 10?8 cm/s (± cells). The permeability of 450 kDa HA was ~40× lower than that of 30 kDa HA for 50 nm membranes, but only ~2.5× lower for 3 µm membranes. The permeability of 4,000 kDa HA was ~250× lower than that of 30 kDa HA for 50 nm membranes, but only ~4× lower for 3 µm membranes. The permeability for PRG4 was ~4 × 10?8 cm/s for 50 nm membranes, ~48 × 10?8 cm/s for 90 nm membranes, ~144 × 10?8 cm/s for 170 nm membranes, and ~336 × 10?8 cm/s for 3 µm membranes. The associated loss across membranes after 24 h ranged from 3% to 92% for HA, and from 3% to 93% for PRG4. These results suggest that semi‐permeable membranes may be used in a bioreactor system to modulate lubricant retention in a bioengineered SF, and that synoviocytes adherent on the membranes may serve as both a lubricant source and a barrier for lubricant transport.
