一些细菌和真菌的解磷能力及其机理初探*   总被引:3,自引:0,他引:3  
4株细菌和8株真菌培养6d后,发现培养液中有机酸含量大幅度增加,pH大幅度地下降,磷的含量大幅度增加,真菌比细菌表现出更强的溶解磷矿粉的能力。不同的微生物分泌有机酸的数量和种类差别很大,真菌分泌的有机酸种类比细菌要多。但是,培养液中有机酸总量与解磷量之间并不存在显著的相关性。  相似文献

从玉米根际和非根际土壤中分离得到的 74株溶解磷矿粉的微生物 ,发现它们的溶磷能力差异很大 ,主要决定于菌株本身的特性 ,与其来源无关 ,真菌普遍比细菌要强。真菌的溶磷能力与其培养介质的 pH之间呈显著的负相关 ,但细菌的这种关系非常弱。二者都产生多种有机酸 ,真菌主要分泌草酸、丙二酸和乳酸 ,而细菌主要分泌草酸、酒石酸、丙二酸、乳酸和乙酸 ,不同菌株分泌有机酸的数量和种类差异很大 ,但溶磷量与有机酸总量或单个有机酸浓度之间 ,没有发现显著的相关性 ,唯独柠檬酸与真菌的溶磷量之间存在显著的相关性。说明不同菌株有完全不同的溶磷机理 ,可能多种机理并存。  相似文献

在温室沙培灭菌条件下,以Al-P为磷源、枳为试材、Glomus mosseae (G.m)和G.versiforme (G.v)为菌剂,研究低磷胁迫下AM真菌对枳实生苗干物重、吸磷效应及根系分泌有机酸的影响。结果表明,接种AM真菌显著增加枳地上部、地下部干物重,增幅16.79%~135.25%;同时显著增加其吸磷量,菌丝对植株的吸磷贡献率为17.04%~71.95%(G.m>G.v),施Al-P显著提高菌丝吸磷贡献率。接种AM真菌的根系分泌的有机酸种类与对照有所不同,未接种处理枳分泌的有机酸有草酸、苹果酸、乳酸、乙酸、顺丁烯二酸和柠檬酸等6种,而接种G.m的则检测到草酸、酒石酸、苹果酸、乳酸、乙酸、柠檬酸、丁二酸等7种,G.v处理的检测到酒石酸,接种处理均未检测到顺丁烯二酸;接种丛枝菌根真菌增加了枳根系分泌有机酸的量(比未接种处理增加19.80~56.87 mg/kg,且施用AlPO4后有机酸含量显著增加(增加20.06~21.84 mg/kg);未接种植株根系仅分泌少量有机酸;接种植株根系分泌的有机酸以苹果酸(42.87%)、柠檬酸(39.22%)和草酸(12.06%)为主。  相似文献

以土壤为钾源,通过液体培养试验研究了8株菜豆根瘤菌对土壤钾的活化作用。结果表明,菜豆根瘤菌能释放大量的氢离子,使液体培养基的pH值大幅度降低,氢离子的浓度至少提高22倍以上。根瘤菌分泌有机酸的种类与数量因菌株不同而异,这些有机酸包括甲酸、乙酸、草酸、丁二酸、柠檬酸、苹果酸和乳酸等,其中全部菌株均能分泌草酸和苹果酸,大部分菌株能分泌乙酸。在接种根瘤菌的液体培养基中,可溶性钾含量显著高于不接种的液体培养基,土壤矿物结构钾则显著降低。由于土壤是培养基钾的唯一来源,故根瘤菌可促进土壤无效钾的溶解。相关分析表明,土壤矿物结构钾与有机酸分泌总量呈极显著负相关(r=-0.878**,n=9),与培养液pH值呈极显著正相关(r=0.863**,n=9),说明根瘤菌分泌的有机酸和氢离子可能溶解土壤无效钾。考虑到根瘤菌草酸分泌量大,络合钙、镁、铁、铝的能力强,且与有机酸分泌总量呈极显著正相关(r=0.870**,n=9),推测草酸分泌在活化土壤无效钾的过程中起重要作用。此外,根瘤菌分泌的有机酸电离产生的氢离子仅占培养液氢离子的4.15%—27.56%,推测根瘤菌直接分泌的氢离子可能是造成培养液pH值降低的主要原因之一。  相似文献

【目的】探究白蜡虫Ericerus pela(Chavannes)雄若虫分泌的蜡花中微生物的多样性,比较不同厚度蜡花(随雄若虫发育蜡质层厚度增加)中细菌及真菌的种类和变化。【方法】采用Mi Seq高通量测序方法对不同发育阶段白蜡虫雄若虫分泌的蜡花中细菌16S rRNA和真菌ITS进行测序,利用Mothur软件计算蜡花中细菌和真菌的操作分类单元(operational taxonomic unit,OTU)及其丰富度、Alpha多样性分析。【结果】细菌3个样品共获470个OTU,真菌2个样品共获264个OTU。细菌共鉴定出18门30纲92科173属;真菌共鉴定出3门15纲43科68属。其中丛毛单胞菌属Comamonas细菌在蜡花中为优势菌,分别占前、中、后期雄若虫分泌的蜡花中细菌总量的79.79%,41.61%和46.86%;枝胞属Cladosporium在前期雄若虫分泌的蜡花中占真菌总量的17.01%,隐球菌属Cryptococcus在后期雄若虫分泌的蜡花中占真菌总量的60.84%,是蜡花中真菌重要组成部分。【结论】不同发育阶段雄若虫分泌的蜡花中细菌及真菌群落存在差异,随着蜡花厚度增加,细菌多样性先增加后减少,而真菌多样性降低。  相似文献

【目的】外生菌根真菌是森林生态系统中的重要组成成分,与木本植物的根系形成菌根,参与树木养分吸收。【方法】试验在液培条件下,以土壤为钾源,利用我国西南地区分离的牛肝菌(Boletnus sp.,Bo 07)、松乳菇(Lactarius delicious,Ld 03)、彩色豆马勃(Pisolithus tinctorius,Pt 715)和内蒙古大青山分离的土生空团菌(Cenococcum geophilum,Cg 04)为材料,研究了它们的生长、钾吸收、氢离子和有机酸分泌,以及土壤钾的变化。【结果】结果表明,Bo 07、Ld 03和Pt 715的生物量、含钾量和吸收量显著高于Cg 04,说明在长期缺钾的环境中,外生菌根真菌经过"物竞天择"可能进化出适应低钾和较强的吸钾能力。以土壤为钾源培养菌根真菌,培养液中的钾浓度显著提高,故外生菌根真菌可促进土壤钾的溶解。在外生菌根真菌的培养液中,分别检测到苹果酸、丁二酸和柠檬酸,均检测到草酸和乙酸。Bo 07、Ld 03和Pt 715显著提高土壤交换性钾含量,Bo 07和Ld 03还显著降低矿物结构钾含量,说明外生菌根真菌不同程度地活化了土壤无效钾,分离自南方的菌株Bo 07、Ld 03和Pt 715总体上大于分离自北方的菌株Cg 04。此外,土壤矿物结构钾分别与总有机酸和草酸分泌量呈极显著负相关(r有机酸=-0.989**,r草酸=-0.950*,n=5),与培养液pH值呈显著正相关(r=0.916*,n=5),故外生菌根真菌分泌的氢离子和有机酸尤其是草酸可能活化土壤无效钾。【结论】供试外生菌根真菌能不同程度的活化土壤无效钾,其活化能力可能与分泌氢离子和有机酸尤其是草酸密切相关。  相似文献

试验研究了在铝胁迫条件下,6种(株)外生菌根真菌(ECMF)的生长、草酸分泌,以及磷、钾、铝的吸收状况。结果表明,铝对抗(耐)型菌种Pt715、HrSp、CgSIV的生长无抑制作用,但显著抑制敏感型菌株LbS238N、LbS238A和Lb270的生长,说明ECMF对铝胁迫的生长反应可能是筛选抗(耐)铝的指标之一。在铝胁迫条件下,无论是抗(耐)型还是敏感型菌种(株),都会发生一系列有益于抗(耐)铝的生化反应,包括草酸分泌量、菌丝磷和钾含量增加,H+分泌改变等。在培养液中,草酸电离产生的H+仅占H+总浓度的少数,说明溶液中H+的主要来源不是ECMF所分泌的草酸,而是菌丝细胞为保持吸收阳离子的电荷平衡排出的H+或分泌的其它有机酸。  相似文献

采用根系分泌有机酸原位收集方法及高效液相色谱技术分析了供磷及缺磷后不同时间白羽扇豆(LupinusalbusL .)非排根区根尖和排根分泌有机酸的种类和数量 ,以及相应的根尖、排根组织 ,茎木质部、韧皮部汁液中有机酸含量的变化。结果表明 :(1)缺磷能够诱导白羽扇豆根系产生大量排根 ,根系的有机酸分泌量也明显增加。 (2 )无论在供磷或缺磷条件下 ,排根与非排根区根尖组织中的有机酸种类相同 ,但排根主要分泌柠檬酸和苹果酸 ,而非排根区根尖主要分泌苹果酸和乙酸。 (3)缺磷后非排根区根尖分泌苹果酸的量增加 ,至第 17天达到高峰 ;排根开始分泌柠檬酸的时间相对较晚。缺磷后排根分泌柠檬酸的量随缺磷时间的延长不断增加。 (4 )在缺磷的排根与非排根区根尖组织和茎木质部伤流液中含有大量柠檬酸和苹果酸 ,但在茎韧皮部汁液中则几乎检测不到这两种有机酸。上述结果表明 ,尽管排根和非排根区根尖组织中的有机酸种类相同 ,但它们向外分泌的有机酸种类不同。缺磷后排根及非排根区根尖增加向外分泌的有机酸主要在根中合成  相似文献

以南北方不同生境下的10株外生菌根真菌为研究对象,采用液体培养的方法,研究了铝对不同菌根真菌的生物量、有机酸分泌及养分含量的影响,以期筛选出抗铝性强的优良菌株,并探讨其抗铝机理。结果表明:外生菌根真菌Sl 08抗铝性最强;Pt 715、Ld 03、Bo 11、Sl 01、Bo 15也具有不同程度的耐铝性;Sl 14、Gc 99、Cg 04抗铝性较差;Sg 11抗铝性最差。来自南方酸性森林土壤的菌株总体抗铝性强于来自北方石灰性土壤的菌株,这表明外生菌根真菌的铝耐受能力与其原始生境有着密切的联系。外生菌根真菌能分泌多种有机酸,且不同菌株分泌的有机酸种类不同。其中,受铝胁迫分泌量增加最多的是草酸。研究中,铝胁迫能增加大多数铝抗性菌株的草酸分泌量,其中铝抗性最强的Sl 08表现最为明显。但铝胁迫并没有促进具备一定铝抗性的Bo 11和Sl 01草酸的分泌量,同时在铝敏感的菌株中均观察到了草酸分泌量的增加。这表明分泌草酸可能并不是外生菌根真菌抵抗铝毒的唯一途径。对各菌株铝胁迫下对氮,磷及钾的吸收研究表明,除铝敏感菌株Sl 14外,铝胁迫均能促进各供试菌株对氮,磷或钾的吸收。综上,在一定铝浓度下,一些外生菌根真菌可通过增加草酸分泌来抵御铝毒。此外,铝胁迫下外生菌根真菌还可通过调控氮、磷、钾等营养元素的吸收来抵抗铝毒,即通过增加对营养元素的吸收来增强其在铝胁迫下的生存能力,这可能是其抵御铝胁迫的应激反应之一。  相似文献

AIMS: To investigate the in vitro production of metal-chelating compounds by ectomycorrhizal fungi collected from pine plantations in southern Chile. METHODS AND RESULTS: Scleroderma verrucosum, Suillus luteus and two isolates of Rhizopogon luteolus were grown in solid and liquid modified Melin-Norkans (MMN) media with and without iron addition and the production of iron-chelating compounds was determined by Chrome Azurol S (CAS) assay. The presence of hydroxamate and catecholate-type compounds and organic acids was also investigated in liquid medium. All isolates produced iron-chelating compounds as detected by CAS assay, and catecholates, hydroxamates as well as oxalic, citric and succinic acids were also detected in all fungal cultures. Scleroderma verrucosum produced the greatest amounts of catecholates and hydroxamates whereas the highest amounts of organic acids were detected in S. luteus. Nevertheless, the highest catecholate, hydroxamate and organic acid concentrations did not correlate with the highest CAS reaction which was observed in R. luteolus (Yum isolate). CONCLUSIONS: Ectomycorrhizal fungi produced a variety of metal-chelating compounds when grown in liquid MMN medium. However, the addition of iron to all fungi cultures reduced the CAS reaction, hydroxamate and organic acid concentrations. Catecholate production was affected differently by iron, depending on the fungal isolate. SIGNIFICANCE AND IMPACT OF THE STUDY: The ectomycorrhizal fungi described in this study have never been reported to produce metal-chelating compound production. Moreover, apart from some wood-rotting fungi, this is the first evidence of the presence of catecholates in R. luteolus, S. luteus and S. verrucosum cultures.  相似文献

太子参为福建道地中药材,连作效应严重限制其产业发展。休耕可以在一定程度上缓解连作效应,然而休耕对于缓解连作效应的机制尚不清楚。本文通过高通量测序分析不同休耕年限对太子参土壤微生物丰度及群落多样性的影响,并探索土壤理化性质、酚酸类物质与土壤微生物群落之间的关联性。结果表明:与对照太子参土壤相比,休耕土壤真菌多样性降低,细菌多样性增多。在细菌优势菌门中,酸杆菌门相对丰度显著增加,变形菌门与放线菌门相对丰度显著降低;真菌优势菌门相对丰度无显著变化。土壤酸性减弱,有机质含量随休耕年限增加呈现递减模式,酚酸类物质中苯甲酸和水杨酸含量显著降低,而对香豆酸等逐渐积累。综上,太子参土壤休耕改善了土壤微生物的菌群结构、土壤性质,从而缓解了连作效应。  相似文献

SUMMARY: Over a hundred isolates which produced haloes around their colonies on dilution plates containing calcium carbonate or dicalcium phosphate have been obtained in pure culture from the root region of the oat plant. Of these, more than 50% were pleomorphic, and this group had the highest proportion of isolates which could produce clear zones on agar media containing either calcium carbonate, dicalcium phosphate, tricalcium phosphate, freshly precipitated hydroxyapatite or basic slag. None of the isolates showed dissolving ability on agar media containing gafsa rock phosphate, variscite, strengite or taranakite. However, when an analytical method was used, 82% of the isolates tested proved able to release phosphate from gafsa rock phosphate, though to a much lesser extent than from dicalcium phosphate. None of the isolates tested by this method released phosphate from variscite, strengite or taranakite.
The nature of the organic acids produced from glucose by 26 of the isolates was also investigated. The majority produced mainly lactic acid, but a few also gave an acid with chromatographic properties similar to those of 2-keto-gluconic acid.  相似文献

Many physicochemical and biotic aspects of the soil environment determine the community composition of bacteria. In this study, we examined the effects of arbuscular mycorrhizal fungi, common symbionts of higher plants, on the composition of bacterial communities after long-term (7-8 years) enrichment culture in the presence of a plant host. We showed that the phylogeny of arbuscular mycorrhizal fungal isolates was a highly significant predictor of bacterial community composition, as assessed by cluster analysis, redundancy analysis and linear discriminant analysis of phospholipid fatty acid patterns. Numerous phospholipid fatty acids differed between the phylogenetic groupings; this pattern also held for fungal-origin phospholipid fatty acids and in a combined bacterial/fungal analysis, suggesting that categorizing phospholipid fatty acids into predominantly bacterial and fungal origin did not affect the overall outcome. The mechanisms underlying this observation could include substrate quality (and quantity) effects, interactions mediated by the host plant (e.g. rhizodeposition) and direct biotic interactions between arbuscular mycorrhizal fungi and bacterial populations. Our results suggest that aspects of arbuscular mycorrhizal fungal functions may be partially explained by the symbiosis-accompanying bacterial communities, a possibility that should be explicitly considered in studies examining the roles of arbuscular mycorrhizal fungal species diversity in soil and ecosystem processes.  相似文献

磷矿粉用量明显地影响节杆菌1TCRi7菌株对其溶解,随着磷矿粉用量的增加,溶磷率急剧下降,超过5 g/L,溶磷率不到0.1%,培养液的pH也随磷矿粉用量增加而升高,但菌体生长繁殖几乎不受磷矿粉用量的影响。振荡培养时,第1 d菌体繁殖量就达到高峰,pH也降至最低,同时表现出强烈的溶磷活力,但菌株的溶磷量并不与其生长量吻合,溶磷量第9 d才达到高峰,以后逐渐下降。培养过程中有机酸分泌发生交替变化,主要分泌乙酸和丙二酸。第1 d、第9 d和第15 d出现3个峰值,说明微生物利用了所分泌出来的有机酸,并发生了有机酸代谢的变化。  相似文献

The purpose of the present work was to determine if selected fungal strains belonging to wood-rotting Basidiomycetes are able to grow on and to solubilize different insoluble oxides in solid media. Twenty-eight strains of white rot fungi were checked for their growth on oxide-amended media (ZnO, CaO, Cu2O). All strains displayed growth on Zn-amended plates but to a different extent, and Cu2O-amended plates turned out to be the most toxic oxide. Most of the tested strains solubilized oxalates and produced noticeable clear zones under the mycelium. These clear zones were tested for the presence of organic acids, the level of which was clearly elevated upon exposure of fungal strains to insoluble oxides. We determined the presence of oxalic, malic, and formic acids, with oxalic acid the predominant one.  相似文献

Capacity of the fungi isolated from the surface of stone monuments for acid formation was studied in cultures under various carbon sources and cultivation conditions. The composition of organic nutrients was adjusted according to the results of investigation of the surface layers from the monuments in urban environment. The primary soil formed at the surface of the stone monuments under urban conditions was shown to contain a variety of carbon and nitrogen sources and is a rich substrate for fungal growth. Oxalic acid was produced by fungi grown on media with various concentrations of sugars, sugar alcohols, and organic acids. Malic, citric, fumaric, and succinic acids were identified only at elevated carbohydrate concentrations, mostly in liquid cultures. Oxalic acid was the dominant among the acids produced by Aspergillus niger at all experimental setups. Unlike A. niger, the relative content of oxalic acid produced by Penicillium citrinum decreased at high carbohydrate concentrations.  相似文献

枯草芽胞杆菌菌肥对有机冬瓜根区土壤微生态的影响   总被引:4,自引:0,他引:4  
【背景】微生物肥料已广泛应用于我国有机作物的种植,其对有机种植土壤微生态的影响尚需科学评测。【目的】高通量测序技术可用于精确分析土壤微生物群落,从细菌、真菌群落结构和多样性的角度阐释枯草芽胞杆菌菌肥对有机农田根区土壤微生物群落的影响。【方法】在有机农田轮作种植条件下,施用枯草芽胞杆菌菌肥后提取冬瓜根区土壤基因组DNA,通过PCR扩增建立文库,利用IlluminaMiSeq高通量测序技术,并结合相关生物信息学方法分析土壤细菌16SrRNA基因V3-V4区和真菌ITS1区的多样性指数及群落结构;测定根区土壤化学性质及酶活性,分析有机冬瓜果实品质,并作相关分析。【结果】从6个有机冬瓜根区土壤样本中获得14199个细菌操作分类单元(OTU)和3378个真菌OTU,细菌和真菌文库测序覆盖率分别在98%、99%以上。枯草芽胞杆菌菌肥会在一定程度上提高土壤细菌种群多样性而降低真菌种群多样性,丰富了细菌群落结构,但显著降低了真菌群落丰富度(P0.05);并减少了根区土壤特有细菌和真菌物种。变形菌门、厚壁菌门和放线菌门是优势细菌,子囊菌门是优势真菌;枯草芽胞杆菌菌肥会提高绿弯菌门和子囊菌门的相对丰度,比例分别为46.23%、10.01%;降低变形菌门和担子菌门的相对丰度,比例分别为11.14%、74.72%。枯草芽胞杆菌菌肥显著降低了土壤pH,显著提高了有机冬瓜果实总氨基酸、可溶性固形物等营养成分含量(P0.05)。【结论】施用枯草芽胞杆菌菌肥改变有机冬瓜根区土壤细菌和真菌的丰富度和多样性,降低了土壤pH,提高了有机冬瓜果实品质。  相似文献

The aim of this study was to test the potential of four isolates of ectomycorrhizal (EM) fungi to utilize organic nitrogen (N) at two different substrate pHs. The organic N source (15N labelled lyophilised fungal mycelium) was mixed with either untreated peat/sand mixture (pH 4.9) or peat/sand mixture limed to a pH of 5.9 and put in cylindrical containers added to each pot. The content of the containers was separated from the roots of Pinus sylvestris seedlings by a nylon mesh and a 2 mm air gap to reduce diffusion of labelled N to the roots. The mycorrhizal plants (except those colonized by Suillus variegatus 2) took up significantly more 15N from the labelled mycelium than uncolonized seedlings. Liming significantly reduced the uptake of 15N by one of the EM fungi (unidentified) but not the other tested species (Paxillus involutus and two isolates of S. variegatus). The EM fungal isolates differed in their influence on the bacterial activity of the soil. This was reduced with P. involutus at both pH levels and increased with one of the two S. variegatus isolates at the high pH and with the other S. variegatus isolate at the low pH level. Liming the soil generally increased bacterial activity. The influence of liming on the proportion of organic N uptake in relation to inorganic N uptake by ectomycorrhizal trees is discussed.  相似文献

Fatty acid variation among culture collection strains and 40 new isolates of Isochrysis galbana Parke was analyzed by quantitative genetic methods. Fatty acid variation among strains and among isolates was highly significant for major fatty acids showing the existence of a genetic component in the determination of differences in fatty acid content. The heritabilities for the major fatty acids ranged between 0.68 and 0.99 among collection strains and between 0.31 and 0.43 among isolates. Eicosapentaenoic acid (EPA) had the highest heritability in I. galbana, but the majority of remaining fatty acids also showed high heritability values. A similar experiment with five UTEX strains of Phaeodactylum tricornutum also showed the presence of a genetic component in four out of seven major fatty acids. Nevertheless, the UTEX strains did not differ significantly in EPA content, although they showed a heritability of 0.40 for this fatty acid. An additional experiment culturing the same isolates of I. galbana in larger volumes of media showed that there was a high significant positive linear relation between EPA content in different volumes. Therefore, EPA content in small volume cultures was an unbiased indicator of EPA content in larger volume cultures. Our results provide support for the genetic determination of fatty acid content in microalgae and suggest that selection, and mutation and selection, are likely to improve EPA content in I. galbana and probably in many other microalgae. Such a selection program can be carried out in small-volume cultures with high confidence.  相似文献
