The tension-strain, stress-strain and stress relaxation curves of longitudinal and circumferential strips of proximal thoracic aortas in normal and WHHL rabbits of different ages were determined using a tensile testing instrument. Wall distensibility of longitudinal and circumferential strips was the greatest in the normal aorta and decreased with advancing age in the atherosclerotic aorta. The wall thickness of the atherosclerotic aorta was positively related to age with a correlation coefficient of 0.66(p less than 0.01). The incremental elastic moduli calculated from the stress-strain curves increased with advancing age in the atherosclerotic aorta. Accordingly, the decreased distensibility of the atherosclerotic wall may be due to the increased wall thickness caused by the intimal thickening as well as to the increase in wall stiffness caused by the increased elastic modulus. The viscoelasticity of the atherosclerotic aorta was larger than that of the normal aorta. This reflects the mechanical effect of atherosclerotic changes that occurred in the thickened intima.  相似文献

Extensibility changes of calcified soft tissue strips from human aorta   总被引:1,自引:0,他引:1  
Some degree of calcification was noted in more than half of the 59 aortas of individuals aged from 15 to 88 we have examined at autopsy. The calcification, which is determined by x-raying the opened and flat aorta, is in patches. We have studied the influence of calcification on stress versus strain, breaking strength, and modulus of elasticity of strips of aorta to determine its importance in vascular disease. Strips of aortic wall 5 x 30 mm were cut with orientation parallel or perpendicular to the vessel axis. Elongation versus load was measured with an Instron tensile testing machine. The true stress and true strain were calculated for both calcified and uncalcified strips from the thoracic and abdominal regions in both orientations. From the stress-strain curve the following values were selected: strain, stress, and slope at 80 mmHg equivalent pressure (1 mmHg = 133.3 Pa); maximum stress, strain, and slope; and breaking stress, strain, and slope if the sample broke. There were statistically significant differences in 13 of the 36 categories between calcified and uncalcified strips. The breaking strength and strain is lower in the calcified strips. The stress-strain curve for the uncalcified strip was mathematically transformed by reducing the amount of elongation so that the curve coincided with that of the calcified strip for eight matched pairs from the same individuals. The calcification appears to immobilize part of the strip, probably causing the boundary of the calcified tissue to be a region of high stress where tissue breakdown can occur.  相似文献

We developed an extension-inflation experimental apparatus with a stereo vision system and a stress-strain analysis method to determine the regional mechanical properties of a blood vessel. Seven proximal descending thoracic aortas were investigated during the inflation test at a fixed longitudinal stretch ratio of 1.35 over a transmural pressure range from 1.33 to 21.33 kPa. Four circumferential regions of each aorta were designated as the anterior (A), left lateral (L), posterior (P), and right lateral (R) regions, and the inflation test was repeated for each region of the aortas. We used continuous functions to approximate the surfaces of the regional aortic wall in the reference configuration and the deformed configuration. Circumferential stretch and stress at the four circumferential regions of the aorta were computed. Circumferential stiffness, defined as the tangent of the stress-stretch curve, and physiological aortic stiffness, named pressure-strain elastic modulus, were also computed for each region. In the low pressure range, the stress increased linearly with increased stretch, but the mechanical response became progressively stiffer in the high-pressure range above a transition point. At a transmural pressure of 12.00 kPa, mean values of stiffness were 416±104 kPa (A), 523±99 kPa (L), 634±91 kPa (P), and 489±82 kPa (R). The stiffness of the posterior region was significantly higher than that of the anterior region, but no significant difference was found in pressure-strain elastic modulus.  相似文献

Some pathological conditions may affect osmolarity, which can impact cell, tissue, and organ volume. The hypothesis of this study is that changes in osmolarity affect the zero-stress state and mechanical properties of the aorta. To test this hypothesis, a segment of mouse abdominal aorta was cannulated in vivo and mechanically distended by perfusion of physiological salt (NaCl) solutions with graded osmolarities from 145 to 562 mosM. The mechanical (circumferential stress, strain, and elastic modulus) and morphological (wall thickness and wall area) parameters in the loaded state were determined. To determine the osmolarity-induced changes of zero-stress state, the opening angle was observed by immersion of the sectors of mouse, rat, and pig thoracic aorta in NaCl solution with different osmolarities. Wall volume and tissue water content of the rings were also recorded at different osmolarities. Our results show that acute aortic swelling due to low osmolarity leads to an increase in wall thickness and area, a change in the stress-strain relationship, and an increase in the elastic modulus (stiffness) in mouse aorta. The opening angle, wall volume, and water content decreased significantly with increase in osmolarity. These findings suggest that acute aortic swelling and shrinking result in immediate mechanical changes in the aorta. Osmotic pressure-induced changes in the zero-stress state may serve to regulate mechanical homeostasis.  相似文献

Enzyme spectrophotometric assays based on acetylcholinesterase (AChE) inhibition were used in combination with Artificial Neural Network (ANN) chemometric analysis for the resolution of pesticides mixtures of chlorpyriphos, dichlorvos and carbofuran. Electric eel (EE) AChE and the recombinant B394-AChE from Drosophila melanogaster were selected due to their different sensitivities to insecticides. These enzymes were used in association with phosphotriesterase (PTE), an enzyme allowing to discriminate between organophosphate and carbamate insecticides. The combined response of three enzymes systems composed of EE-AChE, EE-AChE + PTE, and B394-AChE + PTE was modelled by means of ANN. Specifically, an ANN was constructed where the structure providing the best modelling was a single hidden layer containing four neurons. To prove the concept, a study to resolve pesticide mixtures was done with spectrophotometric measurements. Finally the developed system was successfully applied to the determination of carbofuran, CPO and dichlorvos pesticides in real water samples.  相似文献

The elastic properties of the abdominal aorta at regions of junctions were studied using strips from 17 dogs. Strips of tissue cut longitudinally and circumferentially at the celiac, mesenteric, and renal branches were used to compare the properties of the proximal and distal junctions, as well as the aorta and artery regions adjoining. The tissues were stored for at least 24 h to ensure that no active component of the smooth muscle remained. The elastic properties measured here are due to elastin and collagen, with a small contribution from dead smooth muscle cells. The tissue strips were tested at 20 degrees C while immersed in saline using an Instron tensile testing machine. Elongation of the three regions was measured from photographs taken as the tissue was stretched. Stress values went to 200 kN/m2 as the strain increased to approximately 0.8. (The physiological range for the dog was calculated as 40-85 kN/m2.) The distal junctional region was found to be the most extensible for both longitudinally and circumferentially oriented strips. These results have important implications for flow models as they imply that the shape of the junctional region probably changes between diastole and systole.  相似文献

Strips of human thoracic aortic wall taken at autopsy from 23 individuals aged 15-81 years have been tested in two ways: in uniaxial loading and by digestion in 0.1 M NaOH at 75 degrees C under a load of 50 g. The circumferentially oriented strips were more extensible in loading and took a longer time to fail while being digested under load than the longitudinal strips from the same location. The stress versus strain curve was fitted to an exponential equation of the form, stress = A[exp(B strain)]. For circumferentially oriented strips from 19 subjects, parameter A was independent of age. Parameter B increased by a factor of 2.75 from 19 to 81 years. For longitudinally oriented strips for 14 subjects, A was also independent of age and B increased by 2.4 times from 25 to 81. With digestion under load for 11 matched pairs of strips aged from 32 to 75 the circumferential strips failed in 117 +/- 23 min (standard error), while the longitudinal ones failed in 20.3 +/- 3.2 min (standard error) (p less than 0.003). The results have important implications for vessel attachments to the aorta in heart transplantation and in vascular surgery.  相似文献

The bronchial mucosa contributes to elastic properties of the airway wall and may influence the degree of airway expansion during lung inflation. In the deflated lung, folds in the epithelium and associated basement membrane progressively unfold on inflation. Whether the epithelium and basement membrane also distend on lung inflation at physiological pressures is uncertain. We assessed mucosal distensibility from strain-stress curves in mucosal strips and related this to epithelial length and folding. Mucosal strips were prepared from pig bronchi and cycled stepwise from a strain of 0 (their in situ length at 0 transmural pressure) to a strain of 0.5 (50% increase in length). Mucosal stress and epithelial length in situ were calculated from morphometric data in bronchial segments fixed at 5 and 25 cmH(2)O luminal pressure. Mucosal strips showed nonlinear strain-stress properties, but regions at high and low stress were close to linear. Stresses calculated in bronchial segments at 5 and 25 cmH(2)O fell in the low-stress region of the strain-stress curve. The epithelium of mucosal strips was deeply folded at low strains (0-0.15), which in bronchial segments equated to < or =10 cmH(2)O transmural pressure. Morphometric measurements in mucosal strips at greater strains (0.3-0.4) indicated that epithelial length increased by approximately 10%. Measurements in bronchial segments indicated that epithelial length increased approximately 25% between 5 and 25 cmH(2)O. Our findings suggest that, at airway pressures <10 cmH(2)O, airway expansion is due primarily to epithelial unfolding but at higher pressures the epithelium also distends.  相似文献

Successful management of the bed bug, Cimex lectularius L. (Hemiptera: Cimicidae), is difficult because of its pesticide resistance, which can allow a reduction in population, but not elimination. We evaluated the effect of heat and/or air circulation on the efficacy of dichlorvos resin strips in the control of bed bugs. Treatments were performed in unoccupied dormitory rooms and consisted of dichlorvos resin strips containing 18.6% active ingredient, the same strips + fan, and strips + fan + heat. The mortality of recently fed bed bugs and weight loss of the dichlorvos strips were evaluated over 7 days. Dichlorvos resin strips killed bed bugs and eggs in just over 7 days. The addition of a fan or a fan + heat decreased time to 100% mortality to 3 days and 36 h, respectively. Eggs located in treated rooms did not hatch. Resin strips in the strips + fan treatment and the strips + fan + heat treatment volatilized 10 and 70 times, respectively, faster than strips in the strips-only treatment. The addition of heat in treatments with dichlorvos resin strips enhances the overall efficacy of the volatile insecticide and reduces the time required to eliminate live bed bugs and eggs.  相似文献

The static and dynamic viscoelastic properties were studied of longitudinal and circumferential strips excised from various large veins of dogs. The mechanical behavior in longitudinal direction could be regarded as elastic, while that in circumferential direction was highly viscoelastic. No distinct regionality was observed in either of the longitudinal and the circumferential groups. Noradrenaline and papaverine did not alter the elastic behavior of the longitudinal strips. In circumferential strips, however, noradrenaline caused a considerable decrease in stress relaxation and some steepening in the slope of the upper limb of hysteresis loop. Papaverine did not affect the circumferential characteristics. These findings suggest the dominant contribution of smooth muscle tone to the circumferential characteristics of venous walls. Pretreatment with formic acid abolished the occurrence of stress relaxation in circumferential direction but produced no change in the longitudinal behavior. This indicates that elastin fibers may be a principal determinant of the elastic behavior in longitudinal direction and that a residual tension observed in stress-relaxation tests of circumferential strips may be due to stretched elastin fibers. The elastic moduli of elastase pretreated venous walls were in the order of 10(8) dynes/cm2, about 1000 times higher than those of the control. Accordingly, collagen fibers seemed not to play any appreciable role in the rheological behavior of venous walls under physiological conditions. This inference was supported by histological observations of venous walls under unstretched and stretched states. Models were proposed in regard to the architecture of the fibrous elements in the venous walls.  相似文献

In vitro PGI2 synthesis by aortic strips obtained from thoracic aorta of rabbits fed a high cholesterol diet was examined and compared with that of control rabbits fed a normal diet. In this report, the amounts of PGI2 produced were shown as 6-keto-PGF1 alpha per microgram of aortic tissue DNA instead of per mg wet weight. We also investigated PGI2 synthesis by cultured smooth muscle cells (SMC) obtained from atherosclerotic intima. Basal PGI2 production by aortic strips from atherosclerotic rabbit aorta was significantly augmented compared with that of controls. Arachidonic acid (AA)-induced PGI2 production by atherosclerotic aorta was also significantly higher than that of controls. PGI2 producing capacities of intimal and medial layers, separated from atherosclerotic aorta, were examined and the intimal layer was found to elicit a significantly greater PGI2 production than the medial layer. Furthermore, cultured intimal SMC obtained from atherosclerotic rabbit aorta produced a greater amount of PGI2 than medial SMC from normal rabbit aorta at various cultured conditions. These results suggest that the possibility of enhanced PGI2 production by atherosclerotic aorta may well be considered as a defence mechanism of the vessel wall against damaging stimuli.  相似文献

An experimental program has been carried out in order to investigate the mechanical behavior of porcine corneas. We report the results of inflation tests on the whole cornea and uniaxial tests on excised corneal strips, performed on 51 fresh porcine eyes. Uniaxial tests have been performed on specimens cut from previously inflated corneas. The cornea behavior is characterized by means of elastic stiffness, measured on both average pressure-apex displacement and average uniaxial stress-strain curves; and by means of transversal contraction coefficient, peak stress, and failure stress measured on uniaxial stress-strain curves. Uniaxial tests performed on excised strips allowed to measure the anisotropy in the corneal stiffness and to compare the stiffness of the cornea with the one of the sclera. Viscous properties of the cornea have been obtained through uniaxial relaxation curves on excised corneal strips. The relevant geometrical parameters have been measured and, with the aid of the elastic thin shell theory, a stress-strain curve has been derived from the average inflation test data and compared with similar data available in the literature. The experimental system has been developed in view of future applications to the mechanical testing of both porcine and human corneas.  相似文献

The passive anisotropic elastic properties of rat's aorta were studied in vitro by subjecting cylindrical segments of thoracic and abdominal aorta to a wide range of deformations. Using data on pressure, axial stretch, outer diameter, axial force and wall thickness, incremental moduli of elasticity in the circumferential, axial and radial directions were computed. Results indicate that while the elastic behavior of the aortic wall is globally anisotropic, there exists a state of deformation at which the vessel displays incremental isotropy. This state of deformation corresponds approximately to the loading conditions to which the aorta is exposed in situ. Values of the moduli, analyzed as a function of transmural pressure, show that the stiffness of the aortic wall is fairly constant at low pressures but raises steeply for pressures higher than physiological. For axial stretches as occurring in situ, the magnitudes of the circumferential and radial moduli do not differ significantly for the thoracic aorta; hence this vessel can be regarded as transversely isotropic over a wide range of pressures. The same observation is valid also for the abdominal aorta when pressures equal or smaller than physiological are considered. For both the thoracic and abdominal segments of the aorta, the circumferential and radial moduli are smaller than the axial modulus at low pressures, while the reverse is true for large pressures.  相似文献

We have recently shown that estrogen causes vessel dilation through receptor-mediated stimulation of nitric oxide (NO) production. Here, we hypothesize that estrogen modulates the mechanical homeostasis in the blood vessel wall through NO production. The mechanical properties of female ovariectomized (ovx) mice, female mice lacking the gene for endothelial NO synthase (eNOS(-/-)), and control female and male mice were studied to test the hypothesis. The femoral and carotid arteries and aorta were cannulated in situ and mechanically distended. The stress, strain, elastic modulus, and wall thickness of vessels in ovx and eNOS(-/-) mice, as well as intact female and male mice, were determined. Western blot and immunohistochemistry were used to assess eNOS protein expression in the aorta. Moreover, NO by-products of the femoral and carotid artery were determined by measuring the levels of nitrite and nitrate. Our results show that ovariectomy and eNOS(-/-) significantly decrease the strain in all arteries. Furthermore, the eNOS protein was significantly reduced in ovx mice. Finally, the NO metabolites were significantly decreased both in ovx and eNOS(-/-) mice. We found statistically significant correlations between the structural (wall thickness), mechanical (stress, strain, and elastic modulus), and biochemical parameters (NO by-products). These novel results connect NO to the structural and mechanical properties of the vessel wall. Hence, the effect of endogenous estrogen on the arterial mechanical properties is mediated by the regulation of NO derived from eNOS.  相似文献

A three-dimensional and pulsatile blood flow in a human aortic arch and its three major branches has been studied numerically for a peak Reynolds number of 2500 and a frequency (or Womersley) parameter of 10. The simulation geometry was derived from the three-dimensional reconstruction of a series of two-dimensional slices obtained in vivo using CAT scan imaging on a human aorta. The numerical simulations were obtained using a projection method, and a finite-volume formulation of the Navier-Stokes equations was used on a system of overset grids. Our results demonstrate that the primary flow velocity is skewed towards the inner aortic wall in the ascending aorta, but this skewness shifts to the outer wall in the descending thoracic aorta. Within the arch branches, the flow velocities were skewed to the distal walls with flow reversal along the proximal walls. Extensive secondary flow motion was observed in the aorta, and the structure of these secondary flows was influenced considerably by the presence of the branches. Within the aorta, wall shear stresses were highly dynamic, but were generally high along the outer wall in the vicinity of the branches and low along the inner wall, particularly in the descending thoracic aorta. Within the branches, the shear stresses were considerably higher along the distal walls than along the proximal walls. Wall pressure was low along the inner aortic wall and high around the branches and along the outer wall in the ascending thoracic aorta. Comparison of our numerical results with the localization of early atherosclerotic lesions broadly suggests preferential development of these lesions in regions of extrema (either maxima or minima) in wall shear stress and pressure.  相似文献

The presence and localization of fibronectin in normal and mechanically injured aorta in rabbits was studied using an indirect immunoperoxidase technique on tissue specimens fixed in formaldehyde, embedded in paraffin and pretreated with pepsin. The effect on staining quality of treatment with testicular hyaluronidase prior to immunoperoxidase staining was also examined. In the intima from normal aorta fibronectin was present in the subendothelial basal layer, along the internal and external elastic laminae, around and between the smooth muscle cells of the media and along the collagen and elastic fibres in the adventitia. Sixteen days after a single mechanical dilatation of the descending thoracic aorta all animals developed gross atherosclerotic-like changes. Microscopic examination revealed prominent neo-intimal hyperplasia with subendothelial, cushion-like thickenings but no medial or adventitial alterations. Fibronectin, in increased amounts, was found between and around the endothelial cells and in the subendothelial thickenings between the proliferating smooth muscle cells in relation to the fine, thin elastic and argyrophilic fibres. In the media and adventitia the amount and distribution of fibronectin was indistinguishable from uninjured control aortas. Treatment with testicular hyaluronidase before immunoperoxidase staining resulted in a higher staining resolution in normal and injured aorta. The conspicuous observation in the present study is that fibronectin exclusively accumulates in areas of tissue repair. The origins and functions of fibronectin during tissue injury and repair are discussed.  相似文献

目的:探讨长期四氢生物喋呤(BH4)治疗对自发性高血压大鼠(SHR)血管形态及血管力学性质的影响;方法:选用4周龄雄性SHR36只,随机分为实验组和对照组,每组18只。实验组每周2次腹腔注射BH420mg/kg,对照组注射等容量生理盐水,于实验第4、16和26周龄时各取6只测量动脉收缩压(SBP),并使用计算机图像分析的方法分别测量主动脉血管零应力状态张开角、压力-直径关系及肠系膜动脉血管的壁/腔比值。结果:至BH4治疗后的第16和26周龄,SHR的SBP明显降低(P〈0.01);实验组SHR胸主动脉张开角显著减小(P〈0.01),压力-直径(P-D)关系曲线上移;实验组肠系膜动脉三级分支血管壁/腔(W/L)值减小(P〈0.05)。结论:BH4可以减弱由于长期高血压所导致的血管肥厚和管腔狭窄,恢复血管弹性。  相似文献

The results of this study dealing with the human thoracic foetal aorta testify that even in the middle of the fifth month of development the internal elastic membrane is not yet completely continuous. Furthermore they show that elastogenesis in the tunica media of the human thoracic aorta does not begin directly below the internal elastic membrane, as it does in the foetal aorta of the laboratory rat, but, as it can be seen in our material, somewhat deeper in the developing tunica media. A thin layer of less differentiated tunica media cells persists for a long time in the vicinity of the internal elastic membrane. In the middle of the fifth month, the fusing elastic membrane segments in the tunica media still consist of very immature elastic tissue with a large proportion of the microfibrillar component. The collagen fibrils in the intercellular spaces in the whole depth of the wall of the developing aorta do not become a part of the elastic membranes. Their bundles merely accompany all the elastic membranes in the wall of the thoracic aorta, including the internal elastic membrane.  相似文献
