We have studied the fate of electron dense microbodies in nematode-trapping organs (traps) of the fungus A. oligospora during the initial hours following nematode capture. The interaction studies were performed with isolated traps which had captured a nematode under conditions where the fungal cells had no access to external energy sources. Video enhanced contrast microscopy showed that under these conditions the number of dense bodies present in the trap cell that formed the penetration tube, rapidly decreased. During subsequent penetration and development of the infection bulb this decrease continued while at this time common cell organelles such as mitochondria and vacuoles were formed. This was confirmed by electron microscopy which also revealed that the dense bodies were degraded by means of an autophagic process. The organelles were degraded individually and finally turned into compartments which, based on ultrastructural criteria, were considered vacuoles. Fusion of such vacuoles into larger organelles frequently occurred. The degradation process was initiated early in the interaction since initial stages were already evident within 15 min after capture. Generally it took 1–2 h before the infection bulb had fully developed and trophic hyphae formation started. During this time the original trap cell, characterized by numerous dense bodies, was transformed into an active vegetative hyphal cell containing typical cell organelles such as nuclei, mitochondria, a strongly proliferated endoplasmic reticulum, vacuoles and normal microbodies but lacked dense bodies. This disappearance of dense bodies was confined to the cell that penetrated the nematode and—less frequently—its two neighbouring cells in the hyphal loop. In the other cells, constituting the trap, the dense bodies remained unaffected. As will be discussed, the present results support our current view that traps of A. oligospora contribute to the survival of the organism in its natural environment.  相似文献

This paper describes the results of an ultrastructural study on the subcellular events occurring in nematode-infecting (trophic) hyphae of the nematophagous fungus Arthrobotrys oligospora. In early stages of the infection process (30 min-4 h), the infection bulb and developing trophic hyphae are characterized by a highly proliferated endoplasmic reticulum (ER). Its membranes often appeared vesiculated and occur in close association with the cell membrane of the cells. Upon further invasion of the nematode, lipid droplets developed in the trophic hyphae; these droplets were first observed 4–5 h after the infection but were abundantly present after 24–36 h. Along with the formation of lipid droplets proliferation of microbodies was observed. These organeles were characterized by the presence of catalase and thiolase and were frequently observed in close association with the lipid droplets. Later on the lipid droplets disappeared. During this period new vegetative mycelium developed from the trap that had originally captured the nematode. Our results suggest that part of the nutrients released from the nematode are first converted into lipids by the fungus which in turn are degraded via the -oxidation pathway and further metabolized to support growth of new vegetative hyphae.  相似文献

Quantitative investigation of the factors responsible for trap formation in the nematophagous fungus Duddingtonia flagrans F-882 in submerged liquid culture was carried out. The data obtained suggest a complex program for the regulation of zootrophic nutrition in D. flagrans. Optimal concentrations of such carbon and nitrogen sources as sucrose (0.4%), ammonium ions (0.2%), and tryptone (0.2%) promote trap formation in the case of contact with the nematodes Panagrellus redivivus. Increased concentrations of these compounds, however, inhibit trap formation. The sensitivity of the mycelium to nematode excreta depends on the state of the culture and is increased under limitation by certain nutrient components or in the course of prolonged starvation. A direct correlation was found between the number of caught nematodes and the number of chlamydospores formed on the mycelium. The nutrients obtained from the nematode biomass are used for formation of additional chlamydospores (on average, about 20 chlamydospores per nematode). Environmental and evolutionary aspects of the role of zootrophic nutrition in carnivorous fungi are discussed.  相似文献

This study measured trap induction and trapping on agar disks as affected by juvenile stages (J1, J2, J3, and J4) of the nematode Caenorhabditis elegans and by species of nematode-trapping fungi. Eight species of nematode-trapping fungi belonging to the family Orbiliaceae and producing four kinds of traps were studied: adhesive network-forming Arthrobotrys oligospora, A. vermicola, and A. eudermata, constricting ring-forming Drechslerella brochopaga, and Dr. stenobrocha, adhesive column-forming Dactylellina cionopaga, and adhesive knob-forming Da. ellipsospora, and Da. drechsleri. The number of traps induced generally increased with increasing juvenile stages of C. elegans. The ability to capture the juveniles tended to be similar among isolates that produced the same kind of trap but differed among species that produced different kinds of traps. Trapping by Dr. stenobrocha and Da. cionopaga was correlated with trap number and with juvenile stage. A. oligospora and A. vermicola respectively captured more than 92 and 88% of the J1, J3, and J4 but captured a lower percentage of J2. The knob-producing isolates captured more younger than elder juveniles. Partial correlation analyses demonstrated that the trap induction of the most fungal species positively correlated with the juvenile size and motility, which was juvenile stage dependent. Overall, trap induction and trapping correlated with C. elegans juvenile stage (size and motility) in six species of trapping fungi.  相似文献

Described are a method and apparatus that allowin situ measurement of predation on zooplankton byMysis relicta. The method, which can be generalized to other predators, involves lowering paired large-volume (30-1) plankton traps to the depth of interest, with subsequent trapping of the ambient zooplankton assemblage in each trap and release of predators into one of the traps. The statistical adequacy of the method was shown by error propagation theory to depend on the percentage of available prey consumed, on the number of prey captured by the traps, and on the distribution of zooplankton within the volume of water captured by the traps. Repeated casts of the apparatus showed that, in contrast to other studies of zooplankton distribution, various zooplankton categories were statistically underdispersed (evenly dispersed in space) or at least not more statistically dispersed (clumped) than was a random distribution at a space scale of 1 m. An error analysis of many replicated feeding experiments showed that the errors obtained were reasonably small and that they conformed with or were less than those predicted by error propagation theory that assumed random distribution of zooplankton. Thus, these results supported the practical application of the method and corroborated the conclusion of random dispersion or underdispersion drawn from the experiment of repeated casts of the apparatus.  相似文献

Sticky traps can provide large numbers of spatially referenced samples for use in molecular ecological studies of insects. However, the adhesives used on these traps, and the methods used to clean adhesive off trapped individuals, could potentially interfere with downstream molecular analyses. Specimens captured on sticky traps have been successfully used to analyse mitochondrial or multiple‐copy ribosomal DNA markers, but not single‐copy nuclear markers. Furthermore, the effects of trap adhesive and cleaning protocol on the success of molecular analyses have not been explored. Here, we examine the effects of trap adhesive, sample cleaning method, and sample storage condition on DNA concentration and purity, and on the ability to amplify single‐copy, nuclear microsatellite loci, using specimens of the western cherry fruit fly, Rhagoletis indifferens (Diptera: Tephritidae) captured on sticky traps in an orchard. We could extract DNA of high purity, and amplify microsatellite loci in multi‐plex polymerase chain reaction (PCR), under all combinations of treatments. However, DNA yield, DNA purity and the yield of PCR products were affected by treatment, with complex interactions among trap adhesive, sample cleaning method, and storage condition. Samples that were cleaned with acetone and stored dry had the highest DNA concentration. With respect to PCR amplification, samples cleaned with Histo‐clear produced much less product than those cleaned with acetone or not cleaned at all, whereas samples that were stored dry produced more PCR product than samples stored in ethanol. Insects captured on sticky traps can thus provide genetic data appropriate for molecular ecological analyses under a wide range of treatment conditions. However, potential interactions among adhesives, cleaning protocols and storage conditions suggest that any novel combination for treatment of samples from sticky traps should be tested on a small scale prior to collecting large numbers of samples for genetic studies.  相似文献

Development of effective trapping tools for forest pests and evaluating the key components of these tools is necessary to locate early‐stage infestations and develop management responses to them. Agrilus planipennis Fairmaire (emerald ash borer) is an introduced pest of ash (Fraxinus spp. L.) in North America. The effectiveness of different trap and lure combinations were tested in areas with low and high density populations of A. planipennis. At low density sites, purple prism traps outperformed green traps and girdled ash trap trees in capture rates (adults per day) and rates of detection of A. planipennis. Also, manuka oil lures, used as a standard lure in a national survey programme, captured higher rates of A. planipennis than did previous standards of girdled ash trap trees. There was no logistic relationship between the detection of A. planipennis on a trap and the diameter of the ash tree from which the trap was suspended, possibly because of the use of artificial lures with these traps. There was also no difference in the mean number of A. lanipennis captured per day between ash species and between vigour rating of ash associated with the traps. However, traps placed in open grown and dominant trees captured more beetles than traps placed in lower canopy class trees. At sites defined as low and high density, there was no difference in the larval density per cm3 of phloem. This suggests that exposure time to A. planipennis has been shorter at those low density sites. By exploiting the trap and tree characteristics that improve A. planipennis capture rates and detection efficacy, there can be future improvement in management of this pest. If detection can occur before infested ash trees exhibit signs and symptoms, there may be a potential for reducing the mortality of ash within stands.  相似文献

Firing and resetting of traps in aquatic Utricularia species are associated with water flow and trap volume changes. In this study, trap thickness was used as a measure of water flow and was monitored automatically using an electronic position sensor. The basic characteristics of mechanically stimulated firing and resetting were measured in isolated traps from 13 aquatic Utricularia species and in two trap size categories in Utricularia reflexa. This allowed to study the relationship between these trap functions and trap thickness and length (criteria of trap size). Additionally, the characteristics of spontaneous firings (without any mechanical stimulation) were compared for U. reflexa traps fed or denied prey during a 1-day period. On the absolute scale, the 13 Utricularia species differed considerably in their firing and resetting rates. Significant interspecific differences were also found in the magnitude of firing (in total 3.7–4.2 times) and resetting rates (10–24 times) per unit trap thickness or length. Overall, traps of Utricularia australis, Utricularia stellaris and Utricularia inflata showed the greatest firing and resetting rates. The relative magnitude of firing per unit trap thickness or length showed a highly significant negative correlation with both trap thickness and length and the same also held for the relative resetting rates. Smaller and narrower traps are thus relatively more effective at trap firing and resetting than larger traps. Neither firing nor resetting characteristics were significantly different between unfed and prey-fed traps of U. reflexa and this was also true for the occurrence of spontaneous firings. A strict linear resetting rate, without any lag-period, was found during the first 3 min after trap firing in U. reflexa. This suggests that water is pumped out of the trap continuously and probably recirculates. Given the concepts of spontaneous firing and water recirculation, an ecological model based on the literature data has been devised to quantify the daily N and P gain from the ambient water by Utricularia traps devoid of animal prey. The model shows that the total N and P content estimated in the trap fluid is too high to be accumulated from only the ambient water. This implies that the prey-free traps do not take up N or P from the trap fluid but rather exude a quantity of these nutrients to the fluid to support the microbial community. Therefore, the trap microorganisms behave more as parasites than commensals and represent an additional ecological cost for trap maintenance.  相似文献

Abstract A trap for the collection of bedbugs, Cimex lectularius Linnaeus (Hemiptera: Cimicidae), is described. The trap was baited with CO2 (50–400 mL/min), heat (37.2–42.2 °C) and a chemical lure comprised of 33.0 μg proprionic acid, 0.33 μg butyric acid, 0.33 μg valeric acid, 100 μg octenol and 100 μg L‐lactic acid, impregnated into a gel. Laboratory studies, conducted in a square arena measuring 183 cm on each side, showed that traps with and without baits captured adult bedbugs, but traps with CO2 emissions of 50–400 mL/min caught significantly (P < 0.05) more bedbugs than traps without CO2. In an infested unoccupied apartment, traps with heat and with or without the chemical lure were tested without CO2 on 29 trap‐days and with CO2 on 9 trap‐days. The numbers of bedbugs captured were 656 and 5898 in traps without and with CO2, respectively. The numbers of bedbugs of all development stages captured were significantly greater in traps with CO22 = 15 942, d.f. = 1, P < 10?9). A non‐parametric two‐way analysis of variance evaluation of six different traps with or without CO2, heat or a chemical lure monitored over 19 trap‐days in an infested apartment showed that trap type was highly significant (n = 2833 bedbugs collected) (P < 10?7). The trap with CO2, heat and a chemical lure captured more bedbugs than the other traps, but only caught significantly more fourth and fifth instar nymphs than all other traps. Otherwise, the catches in this trap did not differ significantly from those caught by traps that contained CO2 and heat only. The total numbers of bedbugs collected for each trapping date (pooling all six traps) followed an exponential decline over the trapping period. This type of trap, which caught bedbugs in unoccupied apartments with and without furniture, and in an occupied apartment, may have utility in studying the ecology of bedbugs, in detecting bedbug infestations and in reducing numbers of bites by trapping host‐seeking bedbugs.  相似文献

Summary This experiment investigated the effect of parental nutrient shortage on the allocation of five nutrients to seeds and rhizomes in Sorghum halepense, a perennial, noxious weed, and to seeds in Sorghum bicolor, an annual, cultivated species. Plants from both species were grown from seeds and supplied with fertilizer at three concentrations. The allocation of biomass and nutrients (N, P, K, Ca and Mg) to reproductive and vegetative parts was determined. Relative biomass allocation to reproduction (either sexual or vegetative) remained constant in S. halepense in spite of large differences in total plant weight. In S. bicolor, however, biomass allocation to sexual reproductive structures decreased significantly with decreasing nutrient supply. Individual seed weight was not modified by parental nutrient supply in S. halepense, but it increased with decreasing nutrient availability in S. bicolor. Important differences in mineral allocation to seeds were found between the two species. While S. bicolor seeds were largely buffered from the differences in parental nutrient status, concentration of nutrients in S. halepense seeds decreased significantly with decreasing supply for all the nutrients analyzed except Ca. However, mineral nutrient concentration in S. halepense rhizomes remained remarkably constant despite differences in the external supply, evincing the priority given to vegetative reproduction at the expense of sexual reproduction. Overall, the pattern of nutrient allocation in S. bicolor seeds under different nutrient supply resembled the pattern observed in S. halepense rhizomes, but it had little resemblance to the pattern of nutrient allocation in S. halepense seeds. The results are discussed in terms of differences and similarities in the reproductive strategy of these two species.  相似文献

Different methods to inoculate seedlings of Pinus pinaster and P. sylvestris with edible Lactarius species under standard greenhouse conditions were evaluated. Fungal inoculations were performed both under pure culture synthesis in vitro, followed by transplantation of acclimatized seedlings, and directly in the greenhouse using different techniques for inocula production (mycelial slurries, vegetative inoculum grown in peat-vermiculite and alginate-entrapped mycelium). In vitro inoculations with L. deliciosus produced thoroughly colonized seedlings. However, a sharp decrease in mycorrhizal colonization was detected on transplanted seedlings after 4 month's growth in the greenhouse. On the other hand, all the inocula applied directly in the greenhouse, except the alginate-entrapped mycelium, produced a variable number of mycorrhizal seedlings and colonization rates after the first growing season, depending on the plant-fungal combination and the inoculation method. Inoculations with vegetative inocula of the strain 178 of L. deliciosus were the most effective in producing mycorrhizal seedlings. All the seedlings inoculated with this strain were colonized although the colonization rates were relatively low. The commercial feasibility of the different inoculation methods for the production of seedlings colonized with edible Lactarius species is discussed.  相似文献

Delta, Covered funnel and Uni-traps containing pheromone used for monitoring male Ephestia cautella (Walker) (Lepidoptera: Phycitidae) moths were compared in a wind tunnel in moving and still air. The shape and design of the traps and the form of pheromone plume emitted from them determined the moth's latency time, numbers orientating, alighting on the traps and captured. To improve trap design, the visual responses of the moths to rectangles of different colours and sizes and to brown and white stripes, showed that more moths were attracted to darker coloured 6×50 cm vertical rectangles on a white background and to 7.5 mm brown and white stripes. A cylindrical trap containing pheromone incorporating these features captured 90% and 80% of the moths released in the wind tunnel in moving and still air respectively, compared with 70% and 35% for the best commercial traps.  相似文献

The pine sawyer beetle Monochamus galloprovincialis, a secondary pest of pines in Europe and North Africa, has become important as it was identified as the vector in Europe of Bursaphelenchus xylophilus, the causal agent of pine wilt disease (PWD). An effective trapping system is needed, not only for monitoring the insect vector but also for direct control of its population. Trapping may also provide key information on the nematode load carried by the beetles, allowing early detection of infections, provided that captured beetles remain alive within the trap. Highly effective attractants have been developed in recent years that are commonly used in combination with diverse standard trap designs. In this study, several trap designs were developed and compared to commercial standard models in order to determine which designs maximized the number of attracted insects actually caught and the proportion of them remaining alive. In total, 12 trap designs were evaluated in five field experiments carried out in France, Spain and Portugal. Teflon coating applied to the whole trap and extended, ventilated collecting cups resulted in a significant improvement of trap performance. These modifications led to significant increases of pine sawyer catches, up to 275%, when applied to multiple‐funnel or black cross‐vane traps, compared to standard designs. Furthermore, a significant proportion of the captured beetles remained alive within the trap. These findings have been used to develop new commercial traps (Econex Multifunnel‐12® and Crosstrap®; Econex, Murcia, Spain) available to forest managers. A model for insect survival within the trap was also fitted. Elapsed time between consecutive samplings, mean relative humidity and maximum radiation were the three most significant variables. Thus, traps should provide a suitable sample of live insects if sun exposure of the trap is minimized and a reasonable sampling schedule is implemented.  相似文献

Trap type may influence captures of individuals in different age-sex categories in small mammal studies, resulting in biased population and demographic information. We deployed 4 live trap types at burrow systems of the rodent, Octodon degus Molina, 1782, in central Chile to determine trap efficacy in capturing individuals of 6 demographic categories. We captured 2672 individuals in 17 709 trap days (15.1% trapping success). Tomahawks were the most efficient trap capturing half of individuals during both years, followed by mesh Sherman traps, large Sherman traps, and medium Sherman traps in 2005. All trap types equally sampled sexes. Large and medium Sherman traps provided similar demographic structure, where half of the individuals captured were pups; Tomahawk traps sampled more adults than pups. Relative captures of pups were similar across different trap types, suggesting that pups are equally sampled by each of the deployed trap types. Relative captures of adults were lower in Sherman traps, suggesting that this age class avoided solid-walled traps. For Octodon degus, the sole use of Tomahawk traps may produce sufficient, unbiased demographic data. Only 4 trap mortalities occurred (0.15%). Researchers may minimize trap mortality without compromising sufficient demographic sampling by trapping during peak animal activity.  相似文献

The citrus leafminer, Phyllocnistis citrella Stainton (Lepidoptera: Gracillariidae), is an important world‐wide pest of citrus. Larval mining within leaf flush impacts yield and predisposes trees to infection by citrus canker, Xanthomonas axonopodis pv. citri. The present series of studies sought to identify factors affecting male P. citrella catch in pheromone‐baited traps with the intent of developing effective monitoring. A commercially available pheromone lure (Citralure, ISCA Technologies, Riverside, CA, USA) was highly effective in attracting male P. citrella to traps. Pherocon VI Delta (Trécé Inc., Adair, OK, USA) traps baited with a Citralure captured more male P. citrella than identically baited Pherocon IC Wing traps (Trécé Inc.). The superiority of the Delta‐style trap was found to be due to a 3 cm long closing latch that likely prevents males from flying directly through the trap without capture. Within canopies of mature citrus trees (approximately 3.5 m high), traps at mid‐canopy height (2.0 m) captured more males than traps placed higher (3.5 m) or lower (0.6 m). On the canopy perimeter and in between canopies, traps near ground level (0.6 m height) captured more males than traps at 2.0 and 3.5 m heights. Male catch was greater within the tree canopy or on the canopy perimeter than 2.0 away from the canopy. Traps deployed in trees on the edge of groves captured more males than traps placed 120 and 240 m away from the grove edge and within the grove interior. In non‐pheromone‐treated grove plots, the optimal dosage for catching males was between 0.1 and 1.0 mg of the 3 : 1 blend of (Z,Z,E)‐7,11,13‐hexadecatrienal and (Z,Z)‐7,11‐hexadecadienal; however, in pheromone‐treated plots a higher 10.0 mg dosage lure was most effective. Male catch in pheromone‐baited traps exhibited a diel rhythm with most males captured during scotophase (22:00–23:00 h) and no males captured during photophase.  相似文献

Drosophila suzukii (Matsumura) (Diptera: Drosophilidae) were trapped in the field using colored plastic sphere traps coated with insect Tangle‐trap. Red and black spheres captured significantly more D. suzukii than white spheres. Translucent deli‐cup traps deployed in cherry orchards and baited with yeast, the Alpha Scents lure, or the Scentry lure captured significantly more flies than the Trécé lure and Suzukii bait; all attractants had poor selectivity for D. suzukii. No‐choice evaluations of attractants conducted in field cages corroborated the cherry orchard field study, though translucent deli‐cup traps provisioned with the yeast bait captured significantly more flies than those baited with the Alpha Scents lure. Red sphere traps baited with the Scentry lure captured 3–6× more flies than the deli‐cup trap baited with the same lure, and 3–4× more flies than the deli‐cup trap baited with yeast bait, demonstrating that a trap integrating both visual and olfactory cues is a superior tool for monitoring D. suzukii. Moreover, this simple sticky, dry trap design requires far less labor and maintenance than does a liquid‐based deli‐cup trap.  相似文献

Abstract

Leghold traps continue to be an essential tool for managing brushtail possums (Trichosurus vulpecula) in New Zealand. Concerns about the injuries such traps cause have given rise to efforts to make these traps more humane. One trap modification that had potential for reducing injuries was the placement of an appropriately sized spring between two links of a trap chain to provide a cushioning effect when the chain is extended. Such springs were added to Lanes‐Ace and Victor No. 1½ traps and the injuries caused to captured possums were compared with those to possums captured in the same traps without springs or in Victor No. 1 traps. The addition of chain‐springs reduced the severity of injuries significantly, but injury scores were still significantly higher than for possums captured in Victor No. 1 traps. Consequently, although chain‐springs improved the welfare performance of the Lanes‐Ace and Victor No. 1½ traps, they did not do so to such an extent that those traps could be considered as humane as the currently acceptable Victor No. 1 traps.  相似文献

Monochamus (Coleoptera: Cerambycidae) species are longhorn pine sawyers that serve as insect vectors of the pinewood nematode Bursaphelenchus xylophilus (Nematoda: Parasitaphelenchidae), which are responsible for debilitating pine wilt disease. An aggregation pheromone, 2‐(1‐undecyloxy)‐1‐ethanol (hereafter referred to as monochamol), was shown to be effective at attracting Monochamus species. However, attraction of the pine sawyers to aggregation pheromones varied depending on semiochemicals, including host plant volatiles and kairomones. In this study, we investigated the abilities of monochamol and the host‐plant volatiles α‐pinene and ethanol to attract M. saltuarius in a pine forest in Cheongsong, Gyeongsangbuk‐do, Korea. A total of 91 M. saltuarius (28 males and 63 females) were captured. The combination of monochamol (700 mg) with α‐pinene and ethanol exhibited a synergistic effect on attracting M. saltuarius (11.0 beetles per trap), whereas monochamol alone and a mixture of α‐pinene and ethanol resulted in the capture of 3.2 beetles and 3.6 beetles per trap, respectively. Our results suggest that multi‐funnel traps baited with a blend of monochamol, α‐pinene and ethanol are highly effective for monitoring M. saltuarius and M. alternatus in pine forests.  相似文献

调查了内蒙古草原化荒漠区3种锦鸡儿[垫状锦鸡儿(Caragana tibetica)、荒漠锦鸡儿(C.roborovskyi)和狭叶锦鸡儿(C.stenophylla)]灌丛内、外土壤线虫群落多样性、组成和代谢足迹,以及相关的土壤理化性质和植物群落特征,旨在探讨锦鸡儿属灌丛对土壤线虫群落的影响,并分析这种影响是否具有灌木种间差异,同时从线虫功能团水平上探究灌丛如何通过非生物因素和生物因素影响线虫群落组成。结果表明:锦鸡儿属灌丛对土壤线虫多度、丰富度和多样性无显著影响,但却显著影响土壤线虫群落组成。原因是灌丛对土壤线虫群落具有物种选择性;灌丛可能主要是通过根系分泌物、凋落物质量等因素,而不是通过土壤理化性质和林下植物影响线虫群落组成。土壤线虫组成的变化引起线虫代谢足迹发生明显变化。灌丛内土壤线虫功能代谢足迹大于灌丛外(除狭叶锦鸡儿外),说明灌丛内土壤线虫群落对碳的利用率更高。3种锦鸡儿属灌木中,狭叶锦鸡儿的结构代谢足迹最大,表明狭叶锦鸡儿对捕食杂食类线虫代谢活性的促进作用更强。土壤线虫组成的变化通过线虫代谢足迹导致土壤食物网结构发生显著性变化。狭叶锦鸡儿灌丛土壤干扰程度低,营养富集状况好...  相似文献
