This study was conducted lo determine whether naturally occurring xylem cytokinins, when supplied to leaves via the xylem at approximately endogenous concentrations, increase transpiration and delay senescence in selected monocot species (oat and wheat). The concentrations of some of the major cytokinins (zeatin, dihydrozeatin, ciszeatin and their ribosides, the O-glucosides and nucleotides) were determined in the xylem exudate of oat and wheat seedlings by radioimmunoassay. Evidence is presented that the small volume of exudate (4–5 mm3) collected per plant was xylem sap in transit at the time of shoot excision. Using the data on cytokinin levels, the individual bases and ribosides (and a base/riboside mixture), at multiples of concentrations determined in xylem sap, were tested in transpiration and senescence bioassays. The individual O-glucosides (and mixtures of the O-glucosides) were similarly tested at (i) multiples of the molar concentrations of the corresponding bases and ribosides, and/or at (ii) multiples of the endogenous concentrations. Similarly, zeatin and dihydrozeatin nucleotides were tested at multiples of the molar concentration of zeatin riboside and, in some instances, at multiples of endogenous concentrations. Our results suggest that, at least in oat and possibly in wheat, zeatin-type bases, ribosides and O-glucosides supplied to the leaf in xylem sap are likely to play a role in regulating transpiration in vivo. O-glucosides in oat xylem sap may be important regulators of leaf senescence in the intact plant. The nucleotides were present in xylem sap at lower concentrations than most of the bases, ribosides and O-glucosides. The nucleotides appear likely to play a lesser role than the bases, riboside and O-glucosidcs in controlling transpiration and senescence in the intact plant.  相似文献

The naturally occurring cytokinins, zeatin, zeatin riboside and dihydrozeatin did not promote the germination of celery (Apium graveolens L.) seeds and 6-Δ2-isopentenyladenine (2iPA) and its riboside were only moderately active. Of the synthetic cytokinins, kinetin, kinetin riboside, and the disubstituted urea, N-phenyl-N′-pyridyl urea (NC5392) were moderately active, and 6-benzyl-aminopurine (BA) and its derivatives BA riboside and 6-benzyl-amino-9(tetrahydropyran-2yl)purine (SD8339) were the most active cytokinins tested. 6-(o-hydroxybenzyl)aminopurine (hyd-BA) and its naturally occurring riboside inhibited germination under normally inductive conditions. All the cytokinins examined were more active in promoting germination of lettuce (Lactuca sativa L.) than celery seeds. BA, BA riboside and SD8339 were again the most active cytokinins. In contrast to the results with celery, zeatin and zeatin riboside were highly active. The other cytokinins also showed high activity with the exception of dihydrozeatin, hyd-BA and hyd-BA riboside which were less active. Cytokinin ribosides were less active than the corresponding free bases during the early period of the lettuce seed incubation but total germination after 90 h was similar.  相似文献

We describe a new enzymatic reaction method for the preparation of the radioisotope-labeled cytokinins isopentenyladenine (iP), trans-zeatin (tZ), and their ribosides. The method is based on the three enzyme activities of an adenylate isopentenyltransferase (IPT; EC 2.5.1.27) from Arabidopsis thaliana, an alkaline phosphatase (EC 3.1.3.1) from calf intestine, and a purine-nucleoside phosphorylase (EC 2.4.2.1) from Escherichia coli. The A. thaliana IPT, AtIPT7, utilized both dimethylallyldiphosphate and 4-hydroxy-3-methyl-2-(E)-butenyl diphosphate as isoprenoid donors. The dual specificity of the substrates enabled us to produce iP-type and tZ-type cytokinins separately in the same system simply by switching the substrates. Our method affords a much higher yield of the labeled products than the chemical reaction methods previously used. These labeled compounds will be useful tools for cytokinin research, such as receptor–ligand assays and cell metabolism studies.  相似文献

Cytokinin-induced bud formation in moss protonemata is specific for cytokinin bases, their ribosides being relatively inactive. Binding of [3H]benzyladenine (BA) to a 13,000–80,000 x g subcellular fraction from extracts of Funaria hygrometrica (L.) Sibth. was measured by a centrifugation assay. Increasing concentrations of non-radioactive BA decreased the binding proportionally to the logarithm of the BA concentration between 3×10-8 and 10-4M. [3H]Zeatin also bound to these fractions, although the extent of binding was not as great as with [3H]BA. Biologically active cytokinins, including BA, zeatin, 6-(3-methyl-2-enylamino)purine (IPA) and kinetin, competed for the binding of [3H]BA, whereas the ribosides of BA, zeatin and IPA competed poorly. Other biologically inactive compounds, such as adenine and 9-methyl-BA, were also ineffective as competitors. The ability to bind BA by the 13,000–80,000 x g fraction was greatly reduced by treatment with 1% Triton X-100, and heat treatment eliminated more than one-half of the binding activity. Competitive binding appeared to be pH-dependent, with maximal activity between pH 6.0 and 6.5. After fractionation by differential centrifugation, the ability to bind cytokinins was not correlated with the RNA content of the fraction and thus probably did not represent binding to ribosomes which has been reported in other plant tissues. Cytokinins also exhibited competitive binding to non-biological materials, e.g., talc. The detailed characteristics of the binding of BA to talc were different from those to the biological fractions. Summary The levels of formyltetrahydrofolate synthetase and cyclohydrolase in M. aerogenes were enhanced 3-to 10-fold by growth in media containing formate of histidine. This induced synthesis was decreased by the simultaneous addition of ribosides or ribotides. Histidine, but not formate, also induced the synthesis of formimino transferase and/or cyclodeaminase. The specific activities of N10-formyltetrahydrofolate deacylase, serine hydroxymethylase and N5, N10-methylenetetrahydrofolate dehydrogenase were not affected by formate or histidine. These observations have been discussed with respect to the known mechanisms of regulation of tetrahydrofolate linked enzymes.Dedicated to Prof. C. The endogenous pool of cytokinin metabolites during sexual differentiation of Mercurialis annua L. was studied with a computerized gas-chromatography-mass spectrometry system. Certain metabolites were common to both sexes: ribosides (isopentenyl-adenosine, ribosylzeatin) and the nucleotide of I6-Ade. Zeatin could be detected only in females while its nucleotide was present in males. The results were obtained with differentiating apices and whole plants. The high Z concentration and the low level of its nucleotide are related to the absence of two dominant complementary genes, determining maleness. The natural occurrence of 6-benzylaminopurine, 6-(2-hydroxybenzylamino)purine (ortho-topolin), 6-(3-hydroxybenzylamino)purine (meta-topolin), their ribosides and 9-glucosides is reported using specific antibodies to these groups of compounds in high performance liquid chromatography/enzyme-linked immunosorbent assay (HPLC/ELISA). Compounds were identified by their retention times and differential cross-reactivities with six antisera in analyses carried out in two laboratories using different HPLC gradient systems. Identities were confirmed by immunoaffinity purification followed by HPLC with on-line UV spectrum analysis. Further confirmation of the occurrence of ortho-topolin riboside and isopentenyladenine-9-glucoside was obtained from gas chromatography-mass spectrometry analysis of permethylated HPLC fractions of an extract of oil palm tissues. The aromatic cytokinins, and in particular, ortho-topolin riboside, were found in a variety of oil palm tissues at concentrations exceeding those of the isoprenoid cytokinins, zeatin, isopentenyladenine, dihydrozeatin, and their ribosides. The 9-glucosides of isopentenyladenine and zeatin were more abundant than those of the aromatic types. Cytokinins are naturally occurring substances that act as plant growth regulators promoting plant growth and development, including shoot initiation and branching, and also affecting apical dominance and leaf senescence. Aromatic cytokinin 6-benzylaminopurine (BAP) has been widely used in micropropagation systems and biotechnology. However, its 9-glucoside (BAP9G) accumulates in explants, causing root inhibition and growth heterogenity. To overcome BAP disadvantages, a series of ring-substituted 2′-deoxy-9-(β)-d-ribofuranosylpurine derivatives was prepared and examined in different classical cytokinin bioassays. Amaranthus, senescence and tobacco callus bioassays were employed to provide details of cytokinin activity of 2′-deoxy-9-(β)-d-ribosides compared to their respective free bases and ribosides. The prepared derivatives were also tested for their recognition by cytokinin receptors of Arabidopsis thaliana AHK3 and CRE1/AHK4. The ability of aromatic N6-substituted adenine-2′-deoxy-9-(β)-d-ribosides to promote plant growth and delay senescence was increased considerably and, in contrast to BAP, no loss of cytokinin activity at higher concentrations was observed. The presence of a 2′-deoxyribosyl moiety at the N9-position led to an increase in cytokinin activities in comparison to the free bases and ribosides. The antioxidant capacity, cytotoxicity and effect on the MHV-68 gammaherpesvirus strain were also examined.  相似文献

There are indications that the cytokinin content in transgenic tissues expressing the cytokinin biosynthetic ipt gene is under metabolic control, which prevents the accumulation of cytokinins to lethal levels. The objective of this study was to investigate the relationships between the content of endogenous cytokinins and the activity of cytokinin oxidase (which is believed to be a copper-containing amine oxidase, EC 1.4.3.6.) in ipt transgenic tobacco callus. In addition, the effect of exogenously applied N-benzyladenine (BA) on this relationship was examined. Endogenous cytokinin concentrations were measured in callus of Nicotiana tabacum L. cv. Petit Havana SRI transformed with the ipt of Agrobacterium tumefaciens under the control of a light-inducible promoter and in non-transformed tissue using LC-tandem mass spectrometry. The activity of cytokinin oxidase was estimated by measuring the conversion of [2,8-3H]N6-(Δ2-isopentenyl)adenine to [3H]adenine by enzyme preparations in vitro. The 14-day-old ipt-transformed callus contained a 25-fold higher amount of cytokinins as compared to the non-transformed tissue. Mainly zeatin- and dihydrozeatin-types of cytokinins (free bases, ribosides, nucleotides and O-glucosides) accumulated in the ipt transgenic tissue. The cytokinin pool of both ipt-transformed and non-transformed tissues consisted predominantly of cytokinins that are either resistant to cytokinin oxidase attack (nucleotides and O-glucosides of cytokinins and cytokinins bearing N6-saturated side chain) or have a low affinity for the enzyme (zeatin and its riboside). The former represented 71.6 and 74.8% and the latter 27.7 and 24.4% of the pool of endogenous cytokinins in ipt-transformed and non-transformed tissues, respectively. Enzyme preparations from ipt-transformed tissue exhibited 1.5-fold higher cytokinin oxidase activity compared with that observed in control tissues. Application of exogenous BA affected the total levels of cytokinins of the two tissue lines in different ways. The cytokinin content increased by 1.7- and 1.5-fold in ipt-transformed tissues 6 and 12 h after BA application, respectively, while it declined in the non-transformed control by 1.6- to 2.0-fold between 3 and 12 h after BA application. The increase in cytokinin content in the ipt callus is due to an increase of zeatin- and dihydrozeatin-type cytokinins (nucleotides, ribosides and free bases) leading to an enhanced accumulation of O-glucosides after 12 h. Following BA treatment, the cytokinin oxidase activity increased up to 1.8-fold in ipt-transformed and 1.6-fold in non-transformed tissues. The levels of isopentenyl-type cytokinins were near the detection limit; however, the enhancement of cytokinin oxidase activity after BA treatment in both tissue lines was correlated with the content of preferred substrate of the enzyme, N6-(Δ2-isopentenyl)adenosine.  相似文献

以野生百合渥丹、山丹和传统食用的兰州百合为研究对象,对其鳞茎中多酚类物质、11种单体酚的含量及抗氧化活性(ABTS自由基、超氧阴离子、羟自由基的清除能力,铜离子还原能力以及抑制脂质过氧化活性)进行了分析。结果表明:两种野生百合鳞茎中的多酚类物质含量及抗氧化活性均显著高于兰州百合。3种百合鳞茎中单体酚的种类也有所不同,但均含有没食子酸、矢车菊素-3-芸香糖苷、儿茶素、表儿茶素、杨梅酮、芦丁、对香豆酸、山奈酚。相关性分析显示,除对羟自由基的清除力外,各酚类物质总量与不同抗氧化指标之间呈显著正相关关系。试验结果认为,野生百合鳞茎可作为天然抗氧化资源应用于食品和医药业,具有一定的开发应用前景。  相似文献

Among various nutritional mutants with weak 5′-nucleotidase derived from Bacillus subtilis IAM 1145, the adenine-requiring mutants could convert exogenously added hypo- xanthine, guanine, xanthine and their ribosides to 5′-inosinic acid (IMP) and accumulate it in the medium. Synthesis of IMP from purine derivatives was observed predominantly in an early stage of the cultivation. The conversion was stimulated by Fe2+ or Mn2+, and markedly depressed by an excess amount of adenine in the production-medium.  相似文献

The effect of glycerol, glucose, and starch as carbon sources on the antioxidant defense system such as superoxide dismutase (SOD) and catalase (CAT) activities, pyruvate levels, and membrane lipid peroxidation (LPO) levels of Streptomyces sp. M4018, after isolation from the rhizosphere samples of Colutea arborescens and identification as a strain of S. hiroshimensis based on phenotypic and genotypic characteristics, were investigated. As an antioxidant defense enzyme, SOD activities increased up to 20 g/L of glycerol and 15 g/L of starch, while they showed negative correlation with glucose concentration. CAT activity variations of glycerol- and glucose-supplemented mediums showed significant positive correlations with the trend of SOD activities. However, CAT activity, in contrast to SOD, in Streptomyces sp. M4018 tended to decrease as the starch concentration increased. The production of pyruvate increased with respect to glycerol and starch up to 15 g/L, while it was positively correlated with glucose concentration. The highest pyruvate production was seen at 20 g/L glucose. Membrane LPO levels were negatively correlated with the activities of SOD and CAT enzymes, and the minimum LPO level was determined at 5 g/L of glucose, where SOD and CAT activities reached their maximum levels. Nevertheless, the higher SOD and CAT activities in a wider range of incubation period compared to the beginning by resulting in insignificant increases in membrane LPO levels showed the unusual antioxidant response capacities of the in Streptomyces sp. M4018 against the potentially deleterious effects of reactive oxygen species (ROS) for glycerol, glucose, and starch as carbon sources.  相似文献

The superoxide-scavenging and the tyrosinase-inhibitory activities of 28 kinds of plants used as Chinese medicines were evaluated. Methanol/water extracts were used for the screening tests, and for those which represented high activities, other kinds of extracts were also studied. The extracts of Mallotus japonicus Muell. Arg. scavenged superoxide strongly; the half-inhibiting concentration (IC50) of its 50% methanol/water extract was 10.57 μg of dried material in 1 ml of reaction mixture. The extracts of Fntillaria thunbergii Miq., Carthamus tinctorius L., and Prunus persica (L.) Batsch had strong tyrosinase-inhibitory activities, and the extracts of Scutellariu baicalensis Georgi represented both kinds of activities. These facts suggested that Chinese medicines may be a treasure house of chemical compounds that have the superoxide-scavenging and the tyrosinase-inhibitory activities.  相似文献

The aim of this study is to enhance 3-hydroxyhexanoate (3HHx) fractions of poly(3-hydroxybutyrate-co-3-hydroxyhexanoate), abbreviated as PHBHHx, through site-directed mutagenesis of Aeromonas hydrophila enoyl Coenzyme A hydratase (PhaJAh). Two amino acids (Leu-65 and Val-130) were selected as a substitutional site based on the structural information of PhaJAh. The purified proteins from the wild-type enzyme and mutants were used to determine hydratase activities. Hydratase activities of four single-mutation enzymes were similar to those of the wild type PhaJAh, while hydratase activities of two double-mutation enzymes were much lower. In addition, the mutated phaJ Ah was individually co-transformed into E. coli BL21 (DE3) together with pFH21, which carried the PHA synthase (PhaCAh) gene from A. hydrophila. The recombinant E. coli harboring plasmid pETJ1 (L65A), pETJ2 (L65V) or plasmid pETJ3 (V130A) synthesized the enhanced 3HHx fractions of PHBHHx from dodecanoate, indicating that Leu-65 and Val-130 of PhaJAh play an important role in determining the acyl chain length substrate specificity. The mutated PhaJAh (L65A, L65V, or V130A) provided higher 3HHx precursors for PHA synthase, resulting in the enhanced 3HHx fractions of PHBHHx. It is possible to change the acyl chain length substrate specificity of PhaJ through site-directed mutagenesis and produce PHBHHx with a wider range of alterable monomer composition.  相似文献

Summary Catalase activity of a loamy sand under a 3-year crop rotation in the southeastern U.S.A. was monitored. Corn (Zea mays L.), cotton (Gossypium hirsutum L.), and soybean [Glycine max (L.) Merr.] were the summer crops in the rotation. Winter wheat (Triticum aestivum L.) was planted after corn, and soybean was followed by a winter fallow period. Cotton was followed by a mixture of common vetch (Vicia sativa L.) and crimson clover (Trifolium incarnatum Gibelli & Belli) which was eventually plow-incorporated as a green manure. Highest mean catalase activities were recorded in soil under the wheat, soybean, and winter legume crops; lowest activities were found in soil bearing corn and cotton, and during the winter fallow period. The fertilization regime influenced soil catalase activity independently of the crop. Soil deficient in any of the major elements showed low enzyme activity. Highest activity was found in soil fertilized with P and K, and with N supplied by a winter legume crop. Addition of supplementary mineral nitrogen to this regime reduced catalase activity. Elimination of the winter legume crop from an otherwise complete fertilization regime resulted in a drastic reduction in enzyme activity. In soil receiving a complete fertilization regime there was a close correlation between soil catalase and xylanase activities. A similar correlation between these two enzymes was not found in soil receiving incomplete fertilization.  相似文献

In this study, a new phthalonitrile derivative namely 4-[(2,4-difluorophenyl)ethynyl]phthalonitrile ( 1 ) and its metal phthalocyanines ( 2 and 3 ) were synthesized. The resultant compounds were conjugated to silver nanoparticles and characterized using transmission electron microscopy (TEM) images. The biological properties of compounds ( 1 – 3 ), their nanoconjugates ( 4 – 6 ), and silver nanoparticles ( 7 ) were examined for the first time in this study. The antioxidant activities of biological candidates ( 1 – 7 ) were studied by applying the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. The highest antioxidant activity was obtained 97.47 % for 200 mg/L manganese phthalocyanine-silver nanoconjugates ( 6 ). The antimicrobial and antimicrobial photodynamic therapy (APDT) activities of biological candidates ( 1 – 7 ) were examined using a micro-dilution assay. The highest MIC value was obtained 8 mg/L for nanoconjugate 6 against E. hirae. The studied compounds and their silver nanoconjugates exhibited high APDT activities against all the studied microorganisms. The most effective APDT activities were obtained 4 mg/L for nanoconjugates ( 5 and 6 ) against L. pneumophila and E. hirae, respectively. All the studied biological candidates displayed high cell viability inhibition activities against E. coli cell growth. The biofilm inhibition activities of the tested biological candidates were also investigated against S. aureus and P. Aeruginosa. Biological candidates ( 1 – 6 ) can be considered efficient metal nanoparticle-based materials for multi-disciplinary biological applications.  相似文献

The antisenescent activity of naturally occurring cytokinins (bases and ribosides) has been evaluated by measuring chlorophyll retention in detached wheat (Triticum vulgare) leaf segments. 6-(3-Methyl-2-butenylamino)-2-methylthiopurine (ms2ip) was the most active cytokinin followed by 6-(4-hydroxy-3-methyl-trans-2-butenylamino)purine (tZ). 6-(4-Hydroxy-3-methyl-cis-2-butenylamino)-9-β-D-ribofuranosylpurine (cZR), 6-(4-hydroxy-3-methyl-trans-2-butenylamino)-2-methylthio-9β-D-ribofuranosylpurine (MstZR), and 6-(4-hydroxy-3-methyl-cis-2-butenylamino)-2-methylthio-9-β-D-ribofuroanosylpurine (mscZR) were essentially inactive. 9-Ribosyl substitution did not affect the activity of tZ, (±)-6-(4-hydroxy-3-methylbutylamino)purine (DHZ), or 6-(3-methyl-2-butenylamino)purine (2ip), but lowered the activity of 6-(o-hydroxybenzylamino)purine (OHBA) and 6-(4-hydroxy-3-methyl-cis-2-butenylamino)purine (cZ). 2-Methylthio substitution increased the activity of 2ip and DHZ, decreased the activity of tZ, and had no effect on the activity of cZ. The activities of the simultaneously substituted 2-methylthio-9-ribosyl compounds are lower than those of their corresponding unsubstituted or 2-methylthio substituted bases with the exception of DHZ. Structure-activity relationships for chlorophyll retention did not parallel many of the relationships found for callus tissue growth stimulation.  相似文献

The decapitated primary root of 3-day-old Alaska pea seedlings has been used as a test system to determine the activities on lateral root formation of six auxins, six cytokinins and several other naturally-occurring compounds. Their effects were assessed on (1) the initiation of lateral root primordia, (2) the emergence of visible lateral roots, and (3) the elongation of these laterals. All the auxins, at the optimum concentration of 10-4M, promoted the initiation of lateral root primordia, and all except 3-indolylpropionic acid inhibited the elongation of the resulting lateral roots. Their effects on the emergence of laterals were small and varied. All the cytokinins, at 10-6M and above, inhibited both the initiation and the emergence of lateral roots, zeatin being the most powerful inhibitor. The emergence process was about twice as sensitive as the initiation of primordia to the presence of cytokinins. The cytokinin ribosides were generally less active than the free bases. Abscisic acid and xanthoxin inhibited both emergence and elongation, the concentration for 50% decrease of emergence being about 10-4M. Gibberellic acid had little clear effect on any of the three criteria. Nicotinic acid and thiamine at 10-3M promoted both the initiation of primordia and their emergence: pyridoxal phosphate stimulated both emergence and elongation but did not influence the initiation of primordia. Adenine and guanine had little effect but decreased root elongation some 25%. The strong inhibiting effect of the cytokinins may well be the basis for the marked inhibition exerted by the root-tip on lateral root formation, while the promoting effects of auxins may explain the previously observed promotion of lateral root formation by the young shoot and cotyledons.  相似文献

Partially purified amylases produced by Lactobacillus amylovorus and L. amylophilus were compared and they differed in several properties. The maximum amylase activity of L. amylovorus was higher than that of L. amylophilus. As estimated by sodium dodecyl sulphate-polyacrylamide gel electrophoresis, the molecular mass of the enzymes was 140 kDa for L. amylovorus amylase and 100 kDa for L. amylophilus amylase. Maximum enzymatic activities were obtained when the strains were grown in the presence of CaCO3, on maltose with L. amylovorus and on sucrose with L. amylophilus. Optimal activities were obtained at pH values between 5.0 and 6.0 for both amylases. The L. amylovorus amylase was stable at a higher temperature (50°C) than the L. amylophilus amylase (40°C). Of six substrates examined, greatest activity was obtained by both enzymes on soluble starch. Neither enzymes hydrolysed pullulan or - and \-cyclodextrins. With the exception of Hg2+, which partially inhibited both enzymes, various metal ions, such as 1 mm Ca2+ and Ba2+, stimulated L. amylophilus amylase activity whereas they inhibited L. amylovorus amylase activity. 该研究采用水培试验,分析了俄罗斯杨(Populus russkii)对短期硼胁迫(1、5、10mmol/L硼酸)的生长和生理响应特征,探明其对硼胁迫的耐受范围,初步揭示其耐硼的生理基础。结果显示:(1)与对照相比,1mmol/L硼酸处理俄罗斯杨水培苗的生长情况更优,叶片光合色素含量和净光合速率显著上升,SOD和APX活性及抗坏血酸含量显著上调。(2)5mmol/L硼酸处理幼苗有轻微胁迫受害症状,叶片褪绿变黄,叶绿素b含量比对照显著上升,而Chla/Chlb比值显著下降,其SOD和APX活性显著上调。(3)10mmol/L硼酸处理幼苗表现出严重毒害症状,叶片发黄、茎尖和根尖生长受抑制;叶片PSⅡ原初光能转换效率(F_v/F_m)和Chla/Chlb比值比对照显著下降;其叶片MDA含量、POD活性、抗坏血酸含量以及游离脯氨酸和可溶性糖含量均比对照显著上升。研究表明,在不同浓度硼酸短期胁迫下,俄罗斯杨水培苗能通过上调抗氧化酶活性和抗氧化物质、渗透调节物质含量来有效清除体内胁迫产生的活性氧,减轻膜脂过氧化伤害,使其在低于5mmol/L硼酸胁迫环境下正常生长,表现出较强的耐硼性。  相似文献
