Human peripheral blood monocyte-depleted lymphocytes, T lymphocytes, and non-T lyphocytes were studied for their locomotor activity in response to several common chemotactic stimuli. The factors used to stimulate lymphocyte locomotion were casein, C5a, and f-Met-Leu-Phe. Chemotaxis (directional locomotion) as well as chemokinesis (nondirectional locomotion) in response to each factor were delineated. Monocyte-depleted lymphocyte locomotion was stimulated significantly by all of the above factors. Separation of lymphocytes into T cells and non-T cells indicated that T-lymphocyte locomotion was stimulated by casein and C5a but not by f-Met-Leu-Phe. Non-T lymphocytes were found to respond to C5a and f-Met-Leu-Phe but responded minimally to casein. Additional experiments indicated that casein and f-Met-Leu-Phe were chemokinetic for both monocyte-depleted lymphocytes and non-T lymphocytes, while C5a was chemotactic for both monocyte-depleted lymphocyte preparations and purified T cells.  相似文献

Human blood lymphocytes were separated into T and non-T cells and cultured with pokeweed mitogen (PWM). It was found that in the absence of T cells no differentiation of B cells into immunoglobulin-containing blasts and plasma cells took place. Moreover, the cell yields and the rate of DNA synthesis and blast transformation were very low. The influence of T cells on PWM-induced B-lymphocyte differentiation was studied in mixtures of T/non-T cells at various ratios. Addition of even a few T lymphocytes caused a considerable stimulation of B cells by all parameters used. The responses of T/non-T mixtures of the original cellular composition were of the same order as those of cultures of unseparated cells. It is concluded that the differentiation of human blood B lymphocytes into cells actively synthesizing immunoglobulins, as induced by PWM, is strongly dependent upon the presence of T cells.  相似文献

The response of human newborn lymphocytes in autologous mixed lymphocyte culture was examined for specificity (by restimulation), responder cell phenotype, and responder cell frequency. When the newborn T cells were separated from non-T cells by rosetting with sheep erythrocytes (E) in fetal calf serum (FCS), approximately 1:20,000 T cells proliferated. These responders had specificity for E + FCS, were T4+, and were self-restricted. Significant numbers of responder T cells were not found when newborn T and non-T cells were separated by nylon wool. Responses in parallel autologous cultures of adult T cells showed that 1) adults had a higher frequency than newborns of E + FCS specific responders, 2) evidence for self specificity was lacking in restimulated cultures, and 3) occasional responses to antigen on the surface of monocytes could not be excluded.  相似文献

Whole leukocytes, mononuclear cells, polymorphonuclear cells (PMN), MONOCYTES, PURIFIED LYMPHOCYTES, AND T (rosette-forming cells, RFC) and non-T (nonrosette-forming cells, nonRFC) lymphocytes isolated from the human peripheral blood were stimulated by Sendai virus, respectively, and examined for interferon production in their culture fluids. High levels of interferon were produced by mononuclear cells, but not by PMN. Removal of monocytes from the mononuclear cell population did not affect at all the levels of interferon produced, although it strongly suppressed interferon induction by polyinosinic-polycytidylic acid (poly IC) and mitogenic response to phytohemagglutinin (PHA) of the lymphocytes. Purified monocytes and T lymphocytes were unresponsive to the virus. In contrast, a population of purified non-T lymphocytes produced high levels of interferon. Addition of monocytes to the interferon-producing non-T lymphocytes did not affect the levels of interferon produced. No detectable levels of interferon were produced in the mixture of T lymphocytes and monocytes. It is concluded that non-T lymphocytes may be a major target for interferon induction of human leukocytes by Sendai virus.  相似文献

The capacity of human lymphocytes to adhere to paramyxovirus-infected monolayers and their capacity to kill paramyxovirus-infected cells was investigated. A large fraction of human lymphocytes was found to adhere firmly to the paramyxovirus-infected monolayers. Predsorption of lymphocytes on mumps virus-infected cells impaired their adsorption to a second cell monolayer of the same type. The cytotoxic activity of lymphocytes against mumps virus-infected cells was also reduced after predsorption on mumps virus- or Newcastle disease virus-infected (NDV) cell monolayers. Exposure of lymphocytes to trypsin did not significantly decrease either adsorption or cytotoxicity. Pretreatment of lymphocytes with neuraminidase (NANase) partly inhibited adsorption whereas cytotoxicity was not decreased. Cell fractionation experiments after rosetting of the lymphocytes with sheep erythrocytes (E) indicated that T cells were equally or better adsorbed than "non-T" cells. Taken together with previous experiments which showed that the majority of T lymphocytes are not cytotoxic against mumps virus-infected cells these results suggest that adherence of lymphocytes to infected cells and cytotoxicity may be unrelated phenomena.  相似文献

Escherichia coli type 1 fimbriae contain in association with the major structural protein a lectin-like adhesin moiety that mediates attachment of E. coli to mannose-containing receptors on the surface of host cells. We have investigated the lymphocyte mitogenic activity of this mannose-specific adhesin by comparing the ability of purified wild type type 1 fimbriae containing the adhesin and mutant type 1 fimbriae lacking the adhesin to stimulate proliferation in human lymphocytes. Both fimbriae stimulated a peak of proliferation at 8 days whereas only the wild type fimbriae stimulated an additional peak of proliferation occurring at 3 days. Proliferation at 3 days but not at 8 days could be blocked by the addition of alpha-methyl-D-mannoside. Neonatal lymphocytes from umbilical cord blood responded to both wild type and mutant fimbriae in a fashion similar to adult cells. Stimulation of separated T and non-T cell populations indicated that the proliferation seen at 3 days was solely due to non-T cells whereas the 8-day response was due to T cell proliferation. The addition of gamma-irradiated T cells did not appear to enhance the 3-day response of the non-T cells. However, the 8-day response by T cells was dependent on the presence of gamma-irradiated non-T cells. In cultures of unseparated cells, wild type fimbriae stimulated more than 75% of the B cells to enter the S and G2 phase at 3 days whereas at 8 days cycling T cells were present in both wild type and mutant fimbriae-stimulated cultures. Taken together, our observations suggest that the adhesin molecule stimulates a polyclonal mitogenic response in B cells that peaks at 3 days, and other structural components of the fimbriae are responsible for evoking an 8-day (probably immune) response in T cells.  相似文献

Cord blood T cells did not produce interleukin 2 (IL-2) nor acquire responsiveness to it in autologous mixed-lymphocyte reaction (AMLR) as they do when activated by phytohemagglutinin (PHA). The ability of the cells to respond to IL-2 was restored either by the addition of recombinant IL-2 to the AMLR culture or by the preculture of non-T stimulator cells with recombinant interferon-gamma (IFN-gamma). IL-2 production was also induced when the T cells were added with recombinant IL-2 at the initiation of the AMLR culture, preceded by the treatment of non-T cells with recombinant IFN-gamma. IL-2-producing cells of cord blood induced in the above-mentioned condition were defined to be OKT4+ T cells, because the deletion of OKT4+ T cells from T-cell population abrogated the reaction, while that of OKT8+ T cells did not. Acquisition of IL-2 responsiveness and IL-2 production of T cells seemed to be mediated by HLA-DR and HLA-DQ molecules of non-T cells because these reactions were blocked by the treatment of non-T cells either with monoclonal anti-HLA-DR or with anti-HLA-DQ antibody. The HLA-DR and HLA-DQ densities of cord blood non-T cells were low as compared with those of adult, but the expression of HLA-DQ was remarkably improved by IFN-gamma treatment. In regard to IL-2, both IFN-gamma and IL-2 were needed to enable the lymphocytes to produce. This may suggest that some functional maturation by IL-2 of responder T cells is further required. These combined data suggested that cord blood non-T cells are defective as a stimulator in AMLR and this could be corrected by enhancing the expression of HLA-DQ antigen.  相似文献

In 21 patients with chronic lymphocytic leukemia (CLL) and in 8 hematologically normal persons the number of DNA-synthesizing peripheral blood lymphocytes was investigated by autoradiographic techniques. The lymphocytes were differentiated by EN-rosette tests into T and non-T lymphoid cells. The results show a normal number of proliferating T lymphoid cells and an increased number of proliferating non-T lymphoid cells in clinical stages O-I. Stages III-IV demonstrate a significant increase of the proliferation rate of both T and non-T lymphoid cells. The possible pathogenetic factors and the prognostic value of these results are discussed.  相似文献

Natural killing (NK) in human donors was determined by the ability of peripheral blood subpopulations to lyse the myeloid target, K562, in a 2 to 4 hr 51Cr release assay. The most active cell was a non-T cell which passed through nylon columns (representing 10 to 25% of column passed cells). A second column passed cell population, with characteristics of T lymphocytes (75 to 90% of column passed cells), was also capable of mediating natural killing. Non-T cells which were retained by the nylon columns (45 to 55% of adherent cells) lacked NK activity. However, nylon adherent T cells (45 to 55% of adherent cells) were consistently active in NK assays, illustrating an important subset of NK effector cell often overlooked. Both column passed and adherent T cells were further separated according to their ability to bind IgG or IgM immune complexes, showing that those mediating NK have receptors for IgG (Tγ+) but not for IgM (Tμ+).  相似文献

The patterns of supravital staining with euchrysine, a fluorescent stain thought to bind selectively to lysosomal membranes, were evaluated in resting human lymphocytes separated on the basis of their ability to form spontaneous rosettes with sheep red blood cells in thymus-dependent (T) and thymus-independent (non-T) subpopulations. Two basic staining patterns were found in unseparated lymphocyte populations: type I, small fluorescent granules in a conglomerate form, usually located in a single spot close to the cell membrane; type II, discrete fluorescent granules dispersed over the entire cytoplasm. The overwhelming majority of non-T lymphocytes displayed the type II pattern. Within the T-cell subpopulations both type I and type II patterns were found in proportions which were subject to donor-to-donor variability.  相似文献

Concanavalin A-stimulated human peripheral blood mononuclear cells release a lymphocyte chemotactic factor. This lymphocyte chemotactic factor is produced optimally after 24 to 48 hr of culture and is not found before 3 hr of culture, which suggests that the factor is synthesized de novo and is not preformed and secreted after Con A stimulation. This is further supported by experiments showing that the protein synthesis inhibitors cycloheximide and puromycin totally prevent the production of the chemotactic factor. Experiments using cultured and uncultured T lymphocytes as responding cells show that cultured T cells respond more efficiently than uncultured T cells to this factor. Furthermore, the lymphocyte chemotactic factor preferentially stimulates T lymphocyte locomotion as compared to peripheral blood non-T lymphocyte migration. Fractionation of mononuclear cells into glass nonadherent lymphocytes, monocyte-enriched preparations, T lymphocytes, and non-T lymphocytes shows that lymphocyte chemotactic factor is produced by Con A-stimulated, glass nonadherent lymphocytes and T cells but not by monocytes or non-T lymphocytes. Further fractionation of T lymphocytes into Leu-2 and Leu-3 T cell subpopulations shows that the production of T lymphocyte chemotactic factor can be attributed to the Leu-2 suppressor/cytotoxic T cell subset. The generation of a T lymphocyte chemotactic factor by Leu-2 T cells may represent a means of recruiting other T cells to the site of its release.  相似文献

Mitogenic substances on human peripheral blood mononuclear leukocytes were screened from culture filtrates of microorganisms newly isolated from soil and sea water by measuring [3H]- thymidine incorporation into the cells. Strong mitogenic activity was found in marine bacteria, particularly in marine vibrios. These mitogen samples exhibited neither hemagglutinating activity nor leukoagglutinating activity. They could scarcely stimulate murine lymphocytes.

Cell-cell interaction among leukocyte subsets in response to a bacterial mitogen was investigated using the most powerfully mitogenic sample (culture filtrate of strain H 52–2). A slight decrease in the mitogen response was observed on depletion of plastic surface adherent cells. Separation of T and non-T cells from each other by erythrocyte-rosette sedimentation resulted in a markedly diminished mitogen response. Considerable restoration of the mitogen response was obtained when T cells were mixed with mitomycin C-treated adherent cells or mitomycin C-treated non-T lymphocytes, or when non-T lymphocytes were mixed with mitomycin C-treated T cells.  相似文献

Recently, receptors for IgA were demonstrated on subpopulations of human T lymphocytes. In this report, TNP-modified ox erythrocytes coated with the IgA myeloma MOPC-315 were used to detect IgA receptor-bearing lymphocytes within the human non T cell lymphocyte population. A mean of 5.3% (range 2.9 to 12.4%) of E-rosette negative human lymphocytes bound IgA-coated indicator cells. Blocking studies with soluble IgA, IgG, and IgM demonstrated that the IgA receptors on the non-T cell populations were separate and distinct from the Fc-receptors for IgG and IgM. Fractionation of the non-T lymphocytes on anti-human (Fab)2 columns into sIg+ and sIg- populations or by rosetting with EAC to provide CRL+ and CRL- populations demonstrated that Fc-IgA receptors were present on a subpopulation of sIg+, CRL+ lymphocytes, and also on sIg- (non-T, non-B) lymphocytes.  相似文献

An analysis of killer cells infiltrating “sponge-matrix” allografts during rejection has been performed by preparative fractionation by density centrifugation, velocity sedimentation, and free flow cell electrophoresis and by the use of heterologous anti-T-cell sera. At the peak of rejection, 7 to 8 days after transplantation, the allograft is infiltrated by several classes of killer cells, most notably by non-T lymphocytes, monocytes/macrophages, and T lymphocytes. The predominant cell types capable of performing in vitro lysis of relevant target cells appeared to be monocytes and non-T lymphocytes. T lymphocytes formed only a minority of the killer cells at this stage of the response. In contrast, as also documented earlier, the predominant killer cells in the regional lymph nodes and the spleen of the graft recipient mice were T lymphocytes (blasts).  相似文献

Automated cell image analysis of light and electron microscopic pictures was used for differentiation of nonlabeled lymphocytes in blood smears and in smears of purified lymphocyte suspensions. The percentages of T and B lymphocytes were determined by a two-step rosette assay with sheep red blood cells (T cells) and an immunofluorescence assay with FITC-labeled antihuman globulin (B cells). Images from 1,400 Feulgen-stained and 12,000 Pappenheim-stained cells were analyzed. Various classification methods allowed two lymphocyte subpopulations to be discriminated at the light and electron microscopic levels on the basis of different visual and subvisual morphologic features. As found by immunologic methods, morphologically determined subpopulations corresponded to T and non-T cells, with no further differentiation of non-T cells into B or null cells possible. The results allow the conclusion that there are morphologic differences between human T and non-T cells, with the differences distinguishable from individual variations as well as from alterations induced by sample preparation.  相似文献

The circulating mononuclear cells of normal adult human volunteers were fractionated into T and non-T lymphocytes. The T cells were then fractionated into TM and non-TM cells and the non-T cells were further fractionated, by both positive and negative isolation, into B cells and null cells. Culture of non-T cells, TM cells, and PWM for 7 to 8 days resulted in cytoplasmic Ig (cIg) synthesis by all B cells, as detected by immunofluorescence. However, culture of either purified B or null cells with TM cells and PWM resulted in a marked reduction (75%) of cIg-synthesizing cells. The addition of monocytes did not enhance cIg synthesis. Culture of reconstituted B and null cells with TM cells and PWM restored the capacity to synthesize cIg to the cells. It is concluded that null cells are required, as well as TM cells and PWM, for cIg synthesis by B cells.  相似文献

Human peripheral blood lymphocytes were mixed with erythrocyte-antibody (EA) complexes and separated into EA-rosette forming cell (EA-RFC)-enriched and EA-RFC-depleted suspensions. Thymidine incorporation of EA-RFC-enriched population in the presence of T cell mitogens (PHA, Con A, PWM) was about half of that of EA-RFC-depleted or of unseparated cells. The dose-response curves and kinetics of proliferation were found to be very similar in the three populations. Proliferative response of EA-RFC-enriched lymphocytes was strictly T cell dependent, although non-T cells were later recruited to incorporate thymidine. The interaction of T lymphocytes bearing surface receptors for IgG (TG) with insoluble complexes followed by a post-binding temperature sensitive event, resulted in the modulation of Fc receptors associated with an impaired proliferative response to PHA, Con A, and PWM, without significant change in metabolic cell activity as shown by cell viability, sponaneous leucine incorporation, or β2 microglobulin release.  相似文献

The ability of purified monocytes to regulate in vitro immunoglobulin (Ig) production was examined in 12 patients after HLA-identical marrow grafting. Five patients were studied less than 3 mo after grafting and seven more than 1 yr after grafting. One of the former had acute graft-vs-host disease and five of the latter had chronic graft-vs-host disease. Ficoll-Hypaque-separated peripheral blood mononuclear cells from patients, normal marrow donors, or healthy unrelated individuals were separated into T and non-T cells by sheep erythrocyte rosetting. Highly enriched monocyte and B cell subpopulations were obtained by placing the non-T cells over discontinuous Percoll gradients. Co-cultures of patient or normal monocyte populations with either normal or patient T and B cells with pokeweed mitogen were performed. A hemolytic plaque assay was used to assess Ig secretion after 6 days of culture. Co-culture of T and non-T cells from 10 of 12 patients failed to produce Ig. Monocyte-enriched fractions from all patients provided normal accessory cell functions when co-cultured with normal T and B cells. Two of five patients with chronic graft-vs-host disease had monocytes that suppressed Ig synthesis at high ratios of monocytes to normal T and B cells. Normal monocyte-enriched fractions did not restore Ig production to T and B cells of patients whose T and non-T cells failed to produce Ig. These data indicate that the observed defects in pokeweed mitogen-driven Ig secretion after marrow grafting are due primarily to defective T and B cell functions and that the monocyte accessory function is intact in most patients studied.  相似文献

We have earlier demonstrated that a mixed population of immunologically specific killer cells, including cytotoxic T lymphocytes, non-T (“B”) lymphocytes and monocytes, infiltrate “sponge matrix” allografts at the peak of rejection on Day 8 after transplantation. We have now performed a sequential study covering both early and late stages of the rejection response. We demonstrate that the early infiltrating killer cells are sensitive to anti-Ø and anti-T cell serum plus complement treatment but the late killer cells are not. This finding indicates that the first cytotoxic host cells infiltrating the allograft are predominantly T lymphocytes, whereas as the rejection process proceeds also cytotoxic non-T (“B”) lymphocytes and monocytes are recruited to the site of inflammation.  相似文献
