The interaction between human neutrophils and the Gram negative gastrointestinal pathogen Yersinia pseudotuberculosis was investigated in vitro. Despite the wealth of data describing how Yersinia can affect the function of neutrophils, there are no published studies describing if neutrophil cells can affect the viability of Y. pseudotuberculosis. The wild-type IP32953 strain of Y. pseudotuberculosis was found to be resistant to killing by human neutrophils. Confocal examination and flow-cytometric analysis of this interaction revealed that bacteria were taken up.  相似文献

Ultrathin sectioning and cryofracture of fibrouskerite, sampled from 1.8–1.75 billion year old Volynsediments (Ukraine), revealed in bacteria-like bodies thepresence of structures similar to sheath, cell wall,periplasm, cytoplasm, septum, membranes, intramembraneparticles, poly--hydroxybutyrate inclusions. The techniques of sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and immunoblotting were evaluated for the serodiagnosis of human infections with Yersinia enterocolitica and Yersinia pseudotuberculosis. Lipopolysaccharide (LPS) was prepared from strains comprising four serogroups of Y. enterocolitica and five serogroups of Y. pseudotuberculosis, tested against 200 sera submitted to the Laboratory of Enteric Pathogens for routine serodiagnosis, and shown to contain antibodies to Yersinia LPS by agglutination. Forty four sera were found to contain antibodies that bound to one of the LPS preparations used in the immunoassay. Thirty five of the sera contained antibodies to the LPS of Y. enterocolitica O3, whilst three contained antibodies to the LPS of Y. enterocolitica O5, 27 and Y. enterocolitica O9 LPS respectively. Two sera had antibodies to the LPS of Y. pseudotuberculosis II and a single serum contained antibodies to Y. pseudotuberculosis IV. The SDS-PAGE-immunoblotting procedure described proved to be a reliable procedure for the serodiagnosis of infections with Y. enterocolitica and Y. pseudotuberculosis.  相似文献

The study of the antigenic relationship between Y. pseudotuberculosis in red blood cells and different highly vulnerable tissues of the human body (in the liver, the spleen, the kidneys, the lymph nodes, the intestine) has been carried out; as a result, the presence of heterogeneous substances has been shown. Y. pseudotuberculosis heterogeneous antigens play an important role in the formation of severe forms of the disease and determine the frequency of relapses; they also determine the specific action of diagnostic and therapeutic preparations.  相似文献

The transmission of Yersinia pseudotuberculosis in the pork production chain was followed from farm to slaughterhouse by studying the same 364 pigs from different production systems at farm and slaughterhouse levels. In all, 1,785 samples were collected, and the isolated Y. pseudotuberculosis strains were analyzed by pulsed-field gel electrophoresis. The results of microbial sampling were combined with data from an on-farm observation and questionnaire study to elucidate the associations between farm factors and the prevalence of Y. pseudotuberculosis. Following the same pigs in the production chain from farm to slaughterhouse, we were able to show similar Y. pseudotuberculosis genotypes in live animals, pluck sets (containing tongue, tonsils, esophagus, trachea, heart, lungs, diaphragm, liver, and kidneys), and carcasses and to conclude that Y. pseudotuberculosis contamination originates from the farms, is transported to slaughterhouses with pigs, and transfers to pluck sets and carcasses in the slaughter process. The study also showed that the high prevalence of Y. pseudotuberculosis in live pigs predisposes carcasses and pluck sets to contamination. When production types and capacities were compared, the prevalence of Y. pseudotuberculosis was higher in organic production than in conventional production and on conventional farms with high rather than low production capacity. We were also able to associate specific farm factors with the prevalence of Y. pseudotuberculosis by using a questionnaire and on-farm observations. On farms, contact with pest animals and the outside environment and a rise in the number of pigs on the farm appear to increase the prevalence of Y. pseudotuberculosis.  相似文献

When observations began the population of Watch Lane Flash consisted of Gammarus duebeni and Sigara lateralis. Gammarus tigrinus was then introduced; probably during a transfer of water from a nearby canal. G. tigrinus rapidly increased in numbers and at the same time G. duebeni and S. lateralis decreased to zero. There followed a partial recolonisation by corixids but the species present were Sigara concinna and Sigara dorsalis. Throughout the study period corixid populations were stable in two immediately adjacent water bodies, one of which also contained G. duebeni. Thus the changes described were associated with the presence of G. tigrinus. The reasons for the success of G. tigrinus and its effects on other gammarids and corixids are discussed.  相似文献

The cytotoxic activity of L-asparaginases from Yersinia pseudotuberculosis and from Erwinia carotovora were investigated in vitro using human T-lymphoblastic leukemia (Jurkat and Molt-4) and also solid tumor cell lines MCF-7 (human breast adenocarcinoma), LnCap (human prostate carcinoma), NGUK1 (rat Gasser node neurinoma). E.coli L-asparaginase produced by Medak (Germany) was used as a reference preparation. The data obtained indicate that Y. pseudotuberculosis L-asparaginase significantly inhibits growth of leukemic and solid tumor cells. Its antitumor activity is comparable to that of the reference preparation of L-asparaginase (Medak). These results suggest that the recombinant L-asparaginase can be used for the development of new preparations for the therapy of different types of tumors.  相似文献

Strains (105) of Yersinia pseudotuberculosis isolated in Brazil between 1982 and 1990 were bio-serotyped. They were also studied for plasmid profile, autoagglutination and calcium dependence at 37 °C, Congo red uptake, pyrazinamidase activity, esculin hydrolysis, salicin fermentation and drug sensitivity: 95·24% were biotype 2, serogroup O:3; 2·86% were biotype 1, serogroup O:1; and 1·90% were biotype 2, non-agglutinable. Plasmids were found in 77·14% of the strains (one in each strain). There was total correlation between the presence of the virulence plasmid and autoagglutination, calcium dependence at 37 °C and Congo red uptake. The esculin, salicin and pyrazinamidase tests were not efficient in differentiating pathogenic from non-pathogenic Y. pseudotuberculosis isolates. All strains were highly sensitive to the drugs used. These results indicate that Y. pseudotuberculosis is a potential pathogen for humans in Brazil, especially because the bio-serogroups detected among animals are those most frequently associated with human diseases.  相似文献

It has been suggested that the causative agent of human acute non-bacterial gastro-enteritis, based on morphology, buoyant density in caesium choloride, resistance to acid, and reactions to ether and heat, may be a parvovirus. Using conditions optimal for parvovirus multiplication, attempts were made to isolate a parvovirus from such specimens. Results were negative indicating that the agent is not a parvovirus.  相似文献
