Fourteen yeast isolates belonging to the Metschnikowia clade were isolated from the digestive tracts of lacewings (Neuroptera: Chrysopidae), soldier beetles and leaf beetles (Coleoptera: Cantharidae and Chrysomelidae), and a caddisfly (Trichoptera: Hydropsychidae). The insect hosts were associated with sugary substances of plants, a typical habitat for yeasts in this clade. Based on DNA sequence comparisons and phenetic characters, the yeasts were identified as Candida picachoensis, Candida pimensis, and four undescribed taxa. Among the undescribed taxa, three yeasts were distinguished from one another and from other described taxa by nucleotide differences in the ribosomal DNA repeat, which were sufficient to consider them as new species. Two of the novel yeast species are described as Metschnikowia noctiluminum (NRRL Y-27753T) and M. corniflorae spp. nov. (NRRL Y-27750T) based in part on production of needle-shaped ascospores, which are found in most Metschnikowia species. Sexual reproduction was not observed in the third new yeast, Candida chrysomelidarum sp. nov. (NRRL Y-27749T). A fourth isolate, NRRL Y-27752, was not significantly distinct from Metschnikowia viticola and Candida kofuensis to be described as a new species. Phylogenetic analysis of the D1/D2 loop sequences placed M. noctiluminum within the M. viticola clade, while C. chrysomelidarum was a sister taxon of Candida rancensis. Metschnikowia corniflorae was phylogenetically distinct from other new species and fell outside of the large-spored Metschnikowia group.  相似文献

Changes in symbiont assemblages can affect the success and impact of invasive species, and may provide knowledge regarding the invasion histories of their vectors. Bark beetle symbioses are ideal systems to study changes in symbiont assemblages resulting from invasions. The red turpentine beetle (Dendroctonus valens) is a bark beetle species that recently invaded China from its native range in North America. It is associated with ophiostomatalean fungi in both locations, although the fungi have previously been well-surveyed only in China. We surveyed the ophiostomatalean fungi associated with D. valens in eastern and western North America, and identified the fungal species using multi-gene phylogenies. From the 307 collected isolates (147 in eastern North America and 160 in western North America), we identified 20 species: 11 in eastern North America and 13 in western North America. Four species were shared between eastern North America and western North America, one species (Ophiostoma floccosum) was shared between western North America and China, and three species (Grosmannia koreana, Leptographium procerum, and Ophiostoma abietinum) were shared between eastern North America and China. Ophiostoma floccosum and O. abietinum have worldwide distributions, and were rarely isolated from D. valens. However, G. koreana and L. procerum are primarily limited to Asia and North America respectively. Leptographium procerum, which is thought to be native to North America, represented >45% of the symbionts of D. valens in eastern North America and China, suggesting D. valens may have been introduced to China from eastern North America. These results are surprising, as previous population genetics studies on D. valens based on the cytochrome oxidase I gene have suggested that the insect was introduced into China from western North America.  相似文献

It was found that plant storage tissues (fleshy sugar-containing fruits, subsurface metamorphically altered plant organs (storage roots, tubers, etc.), and starch-containing seed lobes) nearly always contain yeasts that are able to actively reproduce in these tissues causing no visible damage. Within storage tissues, yeast cells were detected both in the intercellular space and inside plant cells. In the tissues of fleshy fruits, endophytic yeasts are represented by the same species as epiphytic ones; cryptococci of the order Filobasidiales and ascomycetes belonging to the genera Hanseniaspora and Metschnikowia are predominant. In subsurface plant organs, red pigmented basidiomycetous yeasts of the genus Rhodotorula prevail. Selective growth of representatives of one species, Candida railenensis, is typical of starch-containing storage tissues of seeds. The results obtained change the established notion of the distributional patterns of yeast fungi in natural habitats and suggest that internal storage tissues of plants can be considered as a new interesting model for studies of coevolving plant-microbial associations.  相似文献

The yeast Pseudozyma graminicola isolated from plants inhibited growth of almost all ascomycetes and basidiomycetes tested (over 270 species of ca. 100 genera) including pathogenic species. This yeast secreted a fungicidal agent, which was identified as a glycolipid composed of cellobiose residue with two O-substituents (acetyl and 3-hydroxycaproic acid) and 2,15,16-trihydroxypalmitic acid. The release of ATP from the glycolipid-treated cells indicated that this glycolipid impaired the permeability of the cytoplasmic membrane. Basidiomycetes were more sensitive to the cellobiose lipid than ascomycetes.  相似文献

An exotic invasive pest of pines, the red turpentine beetle, Dendroctonus valens LeConte (Scolytidae) (RTB), was first detected in Shanxi Province, northern China, in 1998 and started causing widespread tree mortality there in 1999. This outbreak continues and has spread to three adjacent provinces, causing unprecedented tree mortality. Although it is considered a minor pest of pines in North America, RTB has proven to be an aggressive and destructive pest of Pinus tabuliformis, China’s most widely planted pine species. The bionomics and occurrence, distribution, response to host volatiles, and host preference of this pine beetle in China are compared with what is known of the beetle in its native range in North America. Factors likely contributing to D. valens success in China and control of the beetle outbreak are discussed. (−)-β-pinene was shown to be the most attractive host volatile for D. valens from the Sierra Nevada of California, whereas 3-(+)-carene is the most attractive host volatile for beetles in China. Monocultures of Pinus tabuliformis, several consecutive years of drought conditions and warm winters have apparently factored D. valens invasion and establishment in China.  相似文献

Genes involved in mating type determination and recognition were examined in Metschnikowia and related species, to gather insights on factors affecting mating compatibility patterns among haplontic, heterothallic yeast species of the genus. We confirmed the universality of the special mating locus organisation found in Clavispora lusitaniae across and exclusive to the family Metschnikowiaceae (i.e., Metschnikowia and Clavispora). Timing of the divergence between idiomorphs was confirmed to coincide with the origin of the larger (CUG-ser) clade comprising the Debaryomycetaceae and the Metschnikowiaceae, exclusive of Cephaloascus fragrans. The sequence of the a mating pheromone is highly conserved within the large-spored Metschnikowia species, including Metschnikowia orientalis and Metschnikowia hawaiiana, but not Metschnikowia drosophilae or Metschnikowia torresii, which have a pattern of their own, as do other clades in the genus. In contrast, variation in α pheromones shows a more continuous, although imperfect correlation with phylogenetic distance as well as with in vivo mating compatibility.  相似文献

A new yeast species, Candida gelsemii, is described to accommodate three isolates recovered in Georgia, USA, from the toxic nectar of the Carolina jessamine (Gelsemium sempervirens). The species resembles other members of the Metschnikowiaceae clade that have been recovered from nectar, but differs in a number of morphological and physiological characteristics. Analysis of rDNA sequences places the new species well into the clade, but in a basal position with respect to a group of Metschnikowia and Candida species known to occur in association with nectars and bees, as well as marine invertebrates. Exotic forest insects and their symbionts pose an increasing threat to forest health. This is apparently true for the red turpentine beetle (Dendroctonus valens), which was unintentionally introduced to China, where the beetle has killed millions of healthy native pine trees. Previous population genetics studies that used cytochrome oxidase I as a marker concluded that the source of D. valens in China was western North America. In contrast, surveys of fungi associated with D. valens demonstrated that more fungal species are shared between China and eastern North America than between China and western North America, suggesting that the source population of D. valens could be eastern North America. In this study, we used microsatellite markers to determine population structure of D. valens in North America as well as the source population of the beetle in China. The analyses revealed that four genetically distinct populations (herein named the West, Central, Northeast and Mexico) represent the native range of D. valens. Clustering analyses and a simulation‐based approximate Bayesian computation (ABC) approach supported the hypothesis that western North America is the source of the invasive D. valens population. This study provides a demonstration of non‐congruence between patterns inferred by studies on population genetics and symbiont assemblages in an invasive bark beetle.  相似文献

The proton magnetic resonance spectra of the mannose-containing polysaccharides of 48Candida species were determined. Two other species formed heteropolymers for which p.m.r. spectra could not be obtained. The species were grouped as follows: 17 species formed mannose-containing polysaccharides with spectra like those ofHansenula anomala, Pichia farinosa, Pichia membranaefaciens orPichia robertsii mannans; 12, like those ofMetschnikowia andDebaryomyces mannans; 6, like those of mannans ofSaccharomyces species; 4, like that ofTorulopsis bombicola mannan; and 4, like those ofCandida obtusa andCandida tepae polysaccharides. Yeast predation was studied with respect to the range of its distribution among ascomycetous yeasts, the range of yeast species that can be affected, and nutritional aspects of the phenomenon. The yeasts identified as predators belong to the Saccharomycopsis clade as defined on the basis of rDNA sequence relatedness. The 11 recognized species in the clade, plus three undescribed but related Candida species, were shown to be incapable of utilizing sulfate as sole source of sulfur, and all but two (Saccharomycopsis capsularis and Saccharomycopsis vini) were observed to penetrate and kill other yeasts under some conditions. Other unrelated sulfate transport-deficient yeasts (strains in the genera Pichia and Candida and the two known species of Starmera) are not predacious. The predacious species vary considerably as to the optimal environmental conditions that favour predation. Some are inhibited by the presence of rich nitrogenous nutrients, organic sulfur compounds, or higher concentrations of ammonium nitrogen, whereas other species may be stimulated under the same conditions. An attempt was made to correlate prey susceptibility to the excretion of substances that stimulate the growth of predators, but no correlation was detected between the two phenomena. The range of susceptible prey covers both ascomycetes and basidiomycetes, and includes Schizosaccharomyces pombe, which was previously thought to be immune. The achlorophyllous alga Prototheca zopfii is not killed by predacious yeasts, but the initial steps of penetration have been observed in some cases. Predacious species attack other predacious species, and in some cases, young cultures may penetrate older cultures of the same strain.  相似文献

One hundred and fifty-six yeast strains were obtained by the enrichment technique from the phylloplanes of 85 rice leaf samples collected from seven provinces in Thailand. On the basis of the D1/D2 domain of the large subunit rRNA gene sequence analysis, 156 strains were identified as 34 known species in 18 genera consisting of 25 species in 13 genera of the phylum Ascomycota and nine species in five genera of the phylum Basidiomycota. The species in the phylum Ascomycota comprised 24 species in 12 genera of the order Saccharomycetales and one species viz. Yarrowia lipolytica in Saccharomycetales incertae sedis. The 24 species viz. Candida glabrata in the Nakaseomyces clade of Saccharomycetaceae, Candida jaroonii, Candida membranifaciens and Candida terebra in the Yamadazyma clade of Debaryomycetaceae, Candida pseudolambica in the Pichia clade of Pichiaceae, Candida ruelliae in the Metschnikowia clade of Metschnikowiaceae, and three unaffiliated clade Candida species (Candida catenulata, Candida rugosa and Candida tropicalis); Clavispora lusitaniae, Kodamaea ohmeri, Metschnikowia koreensis and Metschnikowia lopburiensis in Metschnikowiaceae; Cyberlindnera fabianii, Cyberlindnera rhodanensis and Wickerhamomyces ciferrii in Wickerhamomycetaceae; Debaryomyces nepalensis, Meyerozyma caribbica, Meyerozyma guilliermondii, Millerozyma koratensis, and Yamadazyma mexicanum in Debaryomycetaceae; Pichia kudriavzevii in Pichiaceae; and Lachancea thermotolerans in Saccharomycetaceae. The species in Basidiomycota viz. Cryptococcus flavescens, Cryptococcus laurentii, Cryptococcus aff. laurentii and Cryptococcus rajasthanensis in the Tremellales lineage, Bulleromyces clade, Tremellales, Tremellomycetes, Agaricomycotina; Pseudozyma antarctica and Pseudozyma aphidis in Ustilaginales, Ustilaginomycetes, Ustilaginomycotina; Rhodotorula taiwanensis and Sporobolomyces blumeae in Sporidiobolales, Microbotryomycetes, Pucciniomycotina; and Trichosporon asahii in Trichosporonales, Tremellomycetes, Agaricomycotina. The most prevalent species was R. taiwanensis with a 23 % frequency of occurrence followed by Candida tropicalis (16 %) and Cryptococcus fabianii (12 %).  相似文献

Lichens are well-known examples of complex symbiotic associations between organisms from different Kingdoms. Microfungi in particular, establish diverse associations with the hosting lichen thallus, as species-specific parasites or transient co-inhabitants. The whole community of lichen-associated fungi constitute the ‘lichen mycobiome’ comprising both ascomycetes and basidiomycetes, including filamentous and yeast taxa. Metabarcoding results and microscopy analyses show that in some thalli, basidiomycetes are frequent lichen-associated fungi but still only a few species could be axenically isolated and morphologically characterized. Within a broad project aiming at characterizing the mycobiome diversity by culture-dependent and independent approaches in two lichen species selected as reference models – Rhizoplaca melanophthalma and Tephromela atra, we succeed in isolating and culturing 76 new strains of basidiomycetous yeasts. The lichen thalli were collected in different mountain regions worldwide and at relatively high elevation. The yeast strains were isolated on different growth media and were studied for their morphological and genetic diversity. Nuclear internal transcribed spacer (ITS) and ribosomal large subunit (LSU) sequence analyses identified them to belong to ten families within the orders Agaricostilbomycetes, Cystobasidiomycetes, Microbotryomycetes, Tremellomycetes and Ustilaginomycetes. The yeasts here detected showed patterns of host-preference in a few cases and they are potentially related to the ecological conditions.  相似文献

Immune defense imposes fitness costs as well as benefits, so organisms should optimize, not maximize, their immune function through their life cycle. We investigated this issue in the red turpentine beetle, Dendroctonus valens LeConte (Coleoptera: Curculionidae, Scolytinae), which is a pine-killing invasive beetle in China, though it is usually considered as a secondary pest in its native range of North America. We hypothesized that pathogen pressure may affect these beetles differently throughout their life history. We measured the insect's immunocompetence throughout life, determining encapsulation ability and phenoloxidase activity in larval stages, pupae and adults. Pupae had the highest encapsulation ability, but encapsulation was not different between final instar larvae and adults. Phenoloxidase (PO) activity was highest in final instar larvae and pupae, followed by the second instar larvae and adults. Total phenoloxidase activity increased significantly from the second instar larval stage to pupae, and then decreased in adults. Although the second instar larvae had the lowest phenoloxidase activity, more than 90% of total PO existed in the hemolymph in the form of the active enzyme, as compared with pupae, in which over 60% of PO occurred as a proenzyme. Both active PO and total PO were much higher in females than in males, though no significant differences were detected between the encapsulation ability of male and female adults. This result suggests the existence of a sexual dimorphism of immunocompetence in D. valens adults. Variations in immunocompetence across developmental stages suggest that D. valens adopts diverse investment strategies in immunocompetence during different stages. Potential reasons for variation in immunocompetence among developmental stages and between the sexes of D. valens are discussed.  相似文献

A literature review is given on growth of yeasts on benzene compounds and on the catabolic pathways involved. Additionally, a yeast collection was screened for assimilation of phenol and 3-hydroxybenzoic acid. Fifteen ascomycetous and thirteen basidiomycetous yeast species were selected and were tested for growth on 84 benzene compounds. It appeared that 63 of these compounds supported growth of one or more yeast species. The black yeastExophiala jeanselmei assimilated 54 of these compounds.The catechol branch of the 3-oxoadipate pathway and its hydroxyhydroquinone variant were involved in phenol and resorcinol catabolism of ascomycetes as well as of basidiomycetes. However, these two groups of yeasts showed characteristic differences in hydroxybenzoate catabolism. In the yeastlike fungusE. jeanselmei and in basidiomycetes of the generaCryptococcus, Leucosporidium andRhodotorula, the protocatechuate branch of the 3-oxoadipate pathway was induced by growth on 3- and 4-hydroxybenzoic acids. In threeTrichosporon species and in all ascomycetous yeasts tested, 4-hydroxybenzoic acid was catabolyzed via protocatechuate and hydroxyhydroquinone. These yeasts were unable to cleave protocatechuate. 3-Hydroxybenzoic and 3-hydroxycinnamic acids were catabolized in ascomycetous yeasts via the gentisate pathway, but in basidiomycetes via protocatechuate.Incomplete oxidation of phenol, some chlorophenols, cresols and xylenols was observed in cultures ofCandida parapsilosis growing on hydroquinone. Most compounds transformed by the growing culture were also converted by the phenol monooxygenase present in cell-free extracts of this yeast. They did not support growth.The relationship between the ability of ascomycetous yeasts to assimilate n-alkanes, amines and benzene compounds, and the presence of Coenzyme Q9 is discussed.  相似文献

Four ascosporulating strains of an undescribed methanol-assimilating yeast species were isolated from forest habitats in Hungary. Three were recovered from rotten wood and one from leaves of a sessile oak (Quercus petraea). An additional isolate of the undescribed species sharing similar phenotypic characters with the above-noted strains was recovered from the gut of an unidentified beetle collected from under the bark of a coniferous tree in Bulgaria. A closely related, but somewhat divergent strain was recovered from insect frass in a Ponderosa pine (Pinus ponderosa) collected in New Mexico, USA. Analysis of the D1/D2 sequences of the LSU rRNA gene placed the new species in the Ogataea clade. The ITS and the D1/D2 LSU sequences of the rRNA gene repeats were compared for the above-noted strains and that of the type strain of Ogataea zsoltii, the closest neighbour among currently recognized Ogataea species. Their relatedness was investigated by parsimony network analysis as well. As a result of the sequence analysis, it was concluded that the six strains isolated from tree associated habitats represent a single new yeast species. Ogataea saltuana sp. nov. is proposed to accommodate these strains. The type strain NCAIM Y.01833T (CBS 10795T, NRRL Y-48448T) was recovered from rotten wood of Scotch pine (Pinus silvestris) in Hungary. The GenBank accession number for the D1/D2 domain nuclear large subunit rRNA gene sequence of strain NCAIM Y.01833T (CBS 10795T, NRRL Y-48448T) is EU327033. Background

Aside from the ecological impacts, invasive species fascinate ecologists because of the unique opportunities that invasives offer in the study of community ecology. Some hypotheses have been proposed to illustrate the mechanisms that allow exotics to become invasive. However, positive interactions between exotic and native insects are rarely utilized to explain invasiveness of pests.

Methodology/Principal Findings

Here, we present information on a recently formed association between a native and an exotic bark beetle on their shared host, Pinus tabuliformis, in China. In field examinations, we found that 35–40% of P. tabuliformis attacked by an exotic bark beetle, Dendroctonus valens, were also attacked by a native pine bark beetle, Hylastes parallelus. In the laboratory, we found that the antennal and walking responses of H. parallelus to host- and beetle-produced compounds were similar to those of the exotic D. valens in China. In addition, D. valens was attracted to volatiles produced by the native H. parallelus.

Conclusions/Significance

We report, for the first time, facilitation between an exotic and a native bark beetle seems to involve overlap in the use of host attractants and pheromones, which is cross-attraction. The concept of this interspecific facilitation could be explored as a novel invasive mechanism which helps explain invasiveness of not only exotic bark beetles but also other introduced pests in principle. The results reported here also have particularly important implications for risk assessments and management strategies for invasive species.  相似文献

Identification of yeasts isolated from apple juices of two cider houses (one located in a plain area and one in an alpine area) was carried out by culture-based method. Wallerstein Laboratory Nutrient Agar was used as medium for isolation and preliminary yeasts identification. A total of 20 species of yeasts belonging to ten different genera were identified using both BLAST algorithm for pairwise sequence comparison and phylogenetic approaches. A wide variety of non-Saccharomyces species was found. Interestingly, Candida railenensis, Candida cylindracea, Hanseniaspora meyeri, Hanseniaspora pseudoguilliermondii, and Metschnikowia sinensis were recovered for the first time in the yeast community of an apple environment. Phylogenetic analysis revealed a better resolution in identifying Metschnikowia and Moesziomyces isolates than comparative analysis using the GenBank or YeastIP gene databases. This study provides important data on yeast microbiota of apple juice and evidenced differences between two geographical cider production areas in terms of species composition.  相似文献

Four yeast strains were isolated from rotting wood samples collected from two sites in the Baotianman Nature Reserve and the Laojieling Nature Reserve in China. DNA sequence comparison and other taxonomic characteristics identified the strains as a single novel species of the genus Metschnikowia. The name Metschnikowia henanensis sp. nov. is proposed to accommodate these highly divergent organisms with the type strain BY-97T (= CICC 1982T = CBS 12677T). The novel species produced chlamydospores, but it did not exhibit ascospore formation in sporulation media for 4 weeks. Molecular phylogeny from the D1/D2 domains of the large subunit (LSU) rRNA gene sequences placed this new species in a basal position to the Metschnikowia viticola/Candida kofuensis/Metschnikowia noctiluminum subclade, and an undescribed Candida species namely strains IMB-EMP4 and IMB-EMP5 was a close sister to M. henanensis.  相似文献
