The organophosphate pesticide chlorpyrifos (CP) has been used extensively since the 1960s for insect control. However, its toxic effects on mammals and persistence in environment necessitate its removal from contaminated sites, biodegradation studies of CP-degrading microbes are therefore of immense importance. Samples from a Pakistani agricultural soil with an extensive history of CP application were used to prepare enrichment cultures using CP as sole carbon source for bacterial community analysis and isolation of CP metabolizing bacteria. Bacterial community analysis (denaturing gradient gel electrophoresis) revealed that the dominant genera enriched under these conditions were Pseudomonas, Acinetobacter and Stenotrophomonas, along with lower numbers of Sphingomonas, Agrobacterium and Burkholderia. Furthermore, it revealed that members of Bacteroidetes, Firmicutes, α- and γ-Proteobacteria and Actinobacteria were present at initial steps of enrichment whereas β-Proteobacteria appeared in later steps and only Proteobacteria were selected by enrichment culturing. However, when CP-degrading strains were isolated from this enrichment culture, the most active organisms were strains of Acinetobacter calcoaceticus, Pseudomonas mendocina and Pseudomonas aeruginosa. These strains degraded 6–7.4 mg L?1 day?1 of CP when cultivated in mineral medium, while the consortium of all four strains degraded 9.2 mg L?1 day?1 of CP (100 mg L?1). Addition of glucose as an additional C source increased the degradation capacity by 8–14 %. After inoculation of contaminated soil with CP (200 mg kg?1) disappearance rates were 3.83–4.30 mg kg?1 day?1 for individual strains and 4.76 mg kg?1 day?1 for the consortium. These results indicate that these organisms are involved in the degradation of CP in soil and represent valuable candidates for in situ bioremediation of contaminated soils and waters.  相似文献

Polygonum minus has been reported to contain valuable metabolites and to date, there is no report on using cell culture technique for metabolite production in P. minus. Naphthalene acetic acid (NAA) concentrations in the range of 2–6 mg L?1 were used in a matrix of combinations with dichlorophenoxyacetic acid (2,4-D) concentrations in the range of 2–10 mg L?1 as plant growth regulators (PGRs) to induce callus cultures. Media that were supplemented with 2 mg L?1 2,4-D + 4 mg L?1 NAA, 2 mg L?1 2,4-D + 6 mg L?1 NAA and 6 mg L?1 2,4-D + 8 mg L?1 NAA were effective for callus induction (93.3 % of the explants produced callus). To establish cell culture, the best growth was obtained from medium that was supplemented with 1 mg L?1 2,4-D + 2 mg L?1 NAA. From a 1-g inoculum size, the fresh weight increases exponentially after 5–10 days of culture, and a 26.71 g maximum fresh weight was obtained after 25 days of culture. The cell culture medium was then analyzed using gas chromatography–mass spectrometry (GC–MS). Jasmonic acid (100, 50, 25 and 5 μM), salicylic acid (100, 50, 25 and 5 μM), yeast extract (500, 250 and 100 mg L?1) and glass beads were used in this research as elicitors. The cell cultures were then incubated with the different elicitors for 1, 2, 3 and 4 days. Several compounds with high peak area percentages were detected, including 2-furancarboxaldehyde, 5-hydroxymethyl, furfural, and 2-cyclopenten-1-one, 2-hydroxy. These results show the diversity of metabolites released by P. minus cell into the culture medium under control conditions.  相似文献

The bacterium with dye degrading ability was isolated from effluent disposal sites of textile industries, Tirupur and was identified as Moraxella osloensis based on the biochemical and morphological characterization as well as 16S rRNA sequencing. This organism was found to decolorize 87 % of Mordant Black 17 at 100 mg l?1 under shake culture condition compared to 92 % under stationary culture condition. Maximum degradation of the dye by M. osloensis was achieved when the mineral salt medium was supplemented with 0.5 % glucose and 0.1 % ammonium nitrate at 35 °C. Degradation of dye was found to follow first order kinetics with the k value of 0.06282 h?1 and a R2 value of 0.955. Analyses for the identification of intermediate compounds confirmed the presence of naphthalene, naphthol, naphthoquinone, salicylic acid and catechol. Based on this finding a probable pathway for the degradation of Mordant Black 17 by M. osloensis has been proposed.  相似文献

Concentrations of aluminum (Al) were determined in leaves of native terrestrial plants, macrophytes and fruit parts (watermelon and tomato) using inductively coupled plasma mass spectrometry. Al concentrations in water and soil were determined by inductively coupled plasma optical emission spectrometry. Potamogeton thunbergii (macrophyte) and Cynodon aethiopicus (terrestrial grass) had the highest leaf Al concentrations (2 and 1 g kg?1 dw, respectively). Transfer factors (mg kg?1 dw plants/mg kg?1 dw soil) based on total Al concentrations in soil varied from 2 × 10?3 to 0.05 and from 1.9 to 78 based on mobile Al concentrations determined after sequential extraction. Bioconcentration factors (mg kg?1 dw plants/mg L?1 water) varied from 19 to 9.5 × 103 L kg?1 dw. Plants can accumulate high concentrations of Al when growing in neutral pH soils and slightly alkaline lakes in the Ethiopian Rift Valley. Controlled experiments showed that C. aethiopicus can accumulate high levels of Al both in root and shoot. Compared to Arabidopsis thaliana, C. aethiopicus was more tolerant to Al exposure as ≥400 μM AlCl3 was needed to inhibit root growth compared to 200 μM in A. thaliana. After exposing C. aethiopicus and A. thaliana in 800 μM AlCl3, alkaline comet assay indicates significant DNA (deoxyribonucleic acid) damage in A. thaliana while C. aethiopicus was unaffected. No significant induction of reactive oxygen species (ROS), in terms of leaf H2O2 levels, could be observed in C. aethiopicus. C. aethiopicus has mechanisms to suppress both Al-induced ROS and DNA damage, thereby increasing tolerance of the species to high Al concentrations.  相似文献

The present study shows the feasibility of a newly isolated strain Acinetobacter sp. B9 for concurrent removal of phenol and Cr (VI) from wastewater. The experiments were conducted in a batch reactor under aerobic conditions. Initially, when mineral salt solution was used as the culture medium, the strain was found to utilize phenol as sole carbon and energy source while no Cr (VI) removal was observed. However, the addition of glucose as co-carbon source resulted in the removal of both toxicants. This co-removal efficiency of the strain was further improved with nutrient-rich media (NB). Optimum co-removal was determined at 188 mg L?1 of phenol and 3.5 mg L?1 of Cr (VI) concentrations at pH 7.0. Strain B9 followed the orthometabolic pathway for phenol degradation. Transmission electron microscopy (TEM) and Fourier transform infrared spectroscopy (FT-IR) studies showed sorption of chromium as one of the major mechanisms for Cr (VI) removal by B9 cells. Acinetobacter sp. B9 was later on checked for bioremediation of real tannery wastewater. After 96 h of batch treatment of tannery effluent containing an initial 47 mg L?1 phenol and 16 mg L?1 Cr (VI), complete removal of phenol and 87 % reduction of Cr (VI) were attained, showing high efficiency of the bacterial strain for potential application in industrial pollution control.  相似文献

Polycyclic aromatic hydrocarbons (PAHs), which are hard to degrade, are the main pollutants in the environment. Degradation of PAHs in the environment is becoming more necessary and urgent. In the current study, strain PL1 with degradation capability of pyrene (PYR) and benzo[a]pyrene (BaP) was isolated from soil and identified as Klebsiella pneumoniae by morphological and physiological characteristics as well as 16S rDNA sequence. With the presence of 20 mg L?1 PYR and 10 mg L?1 BaP in solution, the strain PL1 could degrade 63.4 % of PYR and 55.8 % of BaP in 10 days, respectively. The order of biodegradation of strain PL1 was pH 7.0?>?pH 8.0?>?pH 10.0?>?pH 6.0?>?pH 5.0. Strain PL1 degradation ability varied in different soil. The half-life of PYR in soil was respectively 16.9, 24.9, and 88.9 days in paddy soil, red soil, and fluvo-aquic soil by PL1 degradation; however, the half-lives of BaP were respectively 9.5, 9.5, and 34.0 days in paddy soil, red soil, and fluvo-aquic soil by PL1 degradation. The results demonstrate that the degradation capability on PYR and BaP by PL1 in paddy soil was relatively good, and K. pneumoniae PL1 was the new degradation bacterium of PYR and BaP. K. pneumoniae PL1 has potential application in PAH bioremediation.  相似文献

Eryngium planum L. cell and organ cultures were maintained on Murashige and Skoog media (MS), supplemented with exogenous hormones of different types and various concentrations for high biomass growth. The callus and cell suspension cultures were treated with increased sucrose concentration and/or elicited by methyl jasmonate for the enhancement of selected phenolic acids accumulation. Three phenolic acids, rosmarinic acid (RA), chlorogenic acid (CGA) and caffeic acid (CA), were detected by HPLC-DAD in those cultures. The sum of their content in the dry material was found to be higher in the shoot culture (3.95 mg g?1), root culture (7.05 mg g?1), callus (6.20 mg g?1) and cell suspension (2.04 mg g?1) than in the leaves (1.87 mg g?1) and roots (0.76 mg g?1) of intact plants. The major compound of in vitro cultures was always rosmarinic acid. The content of RA could be increased approximately threefold (16.24 mg g?1) in the callus culture and approximately twofold (3.91 mg g?1) in the cell suspension culture by elicitation with 100 μM methyl jasmonate (MeJA). The higher concentration of sucrose (S) in the medium (5, 6 %) led to over a twofold increase of CGA content in the callus culture (2.54 mg g?1). The three mentioned phenolic acids have been found in E. planum undifferentiated and differentiated in vitro cultures for the first time.  相似文献

The present study investigated growth and biodegradation of 4-bromophenol (4-BP) by Arthrobacter chlorophenolicus A6 in batch shake flasks as well as in a continuously operated packed bed reactor (PBR). Batch growth kinetics of A. chlorophenolicus A6 in presence of 4-BP followed substrate inhibition kinetics with the estimated biokinetic parameters value of μ max = 0.246 h?1, K i = 111 mg L?1, K s  = 30.77 mg L?1 and K = 100 mg L?1. In addition, variations in the observed and theoretical biomass yield coefficient and maintenance energy of the culture were investigated at different initial 4-BP concentration. Results indicates that the toxicity tolerance and the biomass yield of A. chlorophenolicus A6 towards 4-BP was found to be poor as the organism utilized the substrate mainly for its metabolic maintenance energy. Further, 4-BP biodegradation performance by the microorganism was evaluated in a continuously operated PBR by varying the influent concentration and hydraulic retention time in the ranges 400–1,200 mg L?1 and 24–7.5 h, respectively. Complete removal of 4-BP was achieved in the PBR up to a loading rate of 2,276 mg L?1 day?1.  相似文献

The influence of cytokinins and culture conditions including medium volume, harvest time and elicitation with abiotic elicitors (SA/MeJ) have been studied for the optimal production of biomass and withanolides in the multiple shoot culture of Withania somnifera. Elicitation of shoot inoculum mass (2 g l?l FW) with SA at 100 μM in the presence of 0.6 mg l?l BA and 20 mg l?l spermidine for 4 h exposure time at the 4th week in 20 ml liquid medium recorded higher withanolides production (withanolides A [8.48 mg g?l DW], withanolides B [15.47 mg g?l DW], withaferin A [29.55 mg g?l DW] and withanone [23.44 mg g?l DW]), which were 1.14 to 1.18-fold higher than elicitation with MeJ at 100 μM after 5 weeks of culture. SA-elicited cultures did not exhibit much variation in biomass accumulation when compared to control. This cytokinin induces and SA-elicited multiple shoot culture protocol provides a potential alternative for the optimal production of biomass and withanolides utilizing liquid culture.  相似文献

Biodegradation of pyridine by a novel bacterial strain, Rhizobium sp. NJUST18, was studied in batch experiments over a wide concentration range (from 100 to 1,000 mg l?1). Pyridine inhibited both growth of Rhizobium sp. NJUST18 and biodegradation of pyridine. The Haldane model could be fitted to the growth kinetics data well with the kinetic constants μ* = 0.1473 h?1, K s = 793.97 mg l?1, K i = 268.60 mg l?1 and S m = 461.80 mg l?1. The true μ max, calculated from μ*, was found to be 0.0332 h?1. Yield coefficient Y X/S depended on S i and reached a maximum of 0.51 g g?1 at S i of 600 mg l?1. V max was calculated by fitting the pyridine consumption data with the Gompertz model. V max increased with initial pyridine concentration up to 14.809 mg l?1 h?1. The q S values, calculated from $V_{ \hbox{max} }$ , were fitted with the Haldane equation, yielding q Smax = 0.1212 g g?1 h?1 and q* = 0.3874 g g?1 h?1 at S m′ = 507.83 mg l?1, K s′ = 558.03 mg l?1, and K i′ = 462.15 mg l?1. Inhibition constants for growth and degradation rate value were in the same range. Compared with other pyridine degraders, μ max and S m obtained for Rhizobium sp. NJUST18 were relatively high. High K i and K i′ values and extremely high K s and K s′ values indicated that NJUST18 was able to grow on pyridine within a wide concentration range, especially at relatively high concentrations.  相似文献

A Gram-negative, rod-shaped bacterium was isolated from a mixed culture that degraded tert-butyl alcohol (TBA) in a granular-activated carbon (GAC) sample from a Biological-GAC reactor. Strain YZ2T was assigned to the Betaproteobacteria within the family Comamonadaceae based on 16S rRNA gene similarities. The nearest phylogenetic relative (95.0 % similarity) with a valid name was Hydrogenophaga taeniospiralis. The DNA G+C content was 66.4 mol%. DNA:DNA hybridization indicated that the level of relatedness to members of the genus Hydrogenophaga ranged from 1.1 to 10.8 %. The dominant cellular fatty acids were: 18:1 w7c (75 %), 16:0 (4.9 %), 17:0 (3.85 %), 18:0 (2.93 %), 11 methyl 18:1 w7c (2.69 %), Summed Feature 2 (2.27 %), and 18:0 3OH (1.35 %). The primary substrate used was TBA, which is a fuel oxygenate and groundwater contaminant. YZ2T was non-motile, without apparent flagella. It is a psychrotolerant, facultative aerobe that grew between pH 6.5 and 9.5, and 4 and 30 °C. The culture grew on and mineralized TBA at 4 °C, which is the first report of psychrotolerant TBA degradation. Hydrogen was used as an alternative electron donor. The culture also grew well in defined freshwater medium with ethanol, butanol, hydroxy isobutyric acid, acetate, pyruvate, citrate, lactate, isopropanol, and benzoic acid as electron donors. Nitrate was reduced with hydrogen as the sole electron donor. On the basis of morphological, physiological, and chemotaxonomic data, a new species, Hydrogenophaga carboriunda is proposed, with YZ2T as the type strain.  相似文献

Cultivation of seaweeds on a commercial scale requires a large number of propagules with desirable phenotypic traits which include high growth rates and resistance to diseases. Seaweed micropropagation can be considered as one of the best methods to provide a large amount of seedlings for commercial cultivation. This study was carried out to optimize the parameters known to affect the growth of Kappaphycus alvarezii in vitro and subsequently improve the production of seedlings through micropropagation. Suitability of media, concentration of phytoregulators, types and concentration of fertilizers, culture density, light intensity, interval of aeration activity, salinity, and pH were found to be critical factors for the growth of K. alvarezii. The optimum condition for direct regeneration of K. alvarezii in a culture vessel was found to be cultivation of explants in Provasoli's enriched seawater (PES) media supplemented with 2.5 mg L?1 6-benzylaminopurine (BAP), 1.0 mg L?1 indole-3-acetic acid (IAA), and 3.0 mg L?1 natural seaweed extract (NSE) with culture density of 0.4 %?w/v, under light intensity of 75 μmol photons m?2 s?1, continuous aeration of 30.0 L h?1, salinity of 30.0 ppt, and pH 7.5. An airlift photobioreactor was constructed for the mass propagation of K. alvarezii explants with optimum culture conditions obtained from the study. The optimum growth rates of the K. alvarezii explants in culture vessels (5.5 % day?1) and photobioreactor (6.5 % day?1) were found to be higher than the growth rate observed in field trials in the open sea (3.5 % day?1). The information compiled during the course of this study will be of utility to commercial seaweed cultivators.  相似文献

The influence of plant growth regulators on biomass growth and the accumulation of medicinally-relevant isoflavone phytoestrogens, derivatives of genistein and daidzein (8 compounds including aglycones, glucosides and glucoside esters) in callus cultures of Genista tinctoria (Fabaceae) was examined. The experiments included 10 auxins [2,4-dichlorophenoxyacetic acid (2,4-D), p-chlorophenoxyacetic acid, indole-3-acetic acid, indole-3-butyric acid, indole-3-propionic acid, 1-naphthaleneacetic acid, β-naphthoxyacetic acid, picloram, 2,3,5-triiodobenzoic acid (TIBA), 2,4,5-trichlorophenoxyacetic acid (2,4,5-T)] and 7 cytokinins [6-benzylaminopurine, forchlorfenuron, 1,3-diphenylurea, 2-isopentenyladenine, kinetin (KIN), thidiazuron, zeatin] applied at 0.5 and 5.0 mg l?1, jointly with 5.0 or 0.5 mg l?1 KIN or 2,4-D (for auxins and cytokinins, respectively—36 phytohormone combinations in total). Statistical analysis of the relationships between callus growth [expressed as growth index (Gi)] and the accumulation of isoflavones showed positive correlation in the cytokinin group (rxy values from 0.13 to 0.61) and negative correlation within auxins (rxy values from ?0.31 to ?0.39). Among the cytokinins tested, the highest isoflavone content (6,436.26 mg/100 g dry weight) and the fastest biomass growth (Gi = 892.46 %) were obtained for 0.5 mg l?1 KIN used jointly with 5.0 mg l?1 2,4-D. In the group of auxins, the combination of 0.5 mg l?1 TIBA and 5.0 mg l?1 KIN provided the fastest culture growth (Gi = 983.07 %) and the isoflavone concentration of 10,474.23 mg/100 g dry weight, which is so far the highest amount of these metabolites achieved in callus cultures of higher plants.  相似文献

The filamentous Cyanobacterium Arthrospira is commercially produced and is a functional, high-value, health food. We identified 5 low temperature and low light intensity tolerant strains of Arthrospira sp. (GMPA1, GMPA7, GMPB1, GMPC1, and GMPC3) using ethyl methanesulfonate mutagenesis and low temperature screening. The 5 Arthrospira strains grew rapidly below 14?°C, 43.75 μmol photons m?2 s?1 and performed breed conservation at 2.5?°C, 8.75 μmol photons m?2 s?1. We used morphological identification and molecular genetic analysis to identify GMPA1, GMPA7, GMPB1 and GMPC1 as Arthrospira platensis, while GMPC3 was identified as Arthrospira maxima. Growth at different culture temperatures was determined at regular intervals using dry biomass. At 16?°C and 43.75 μmol photons m?2 s?1, the maximum dry biomass production and the mean dry biomass productivity of GMPA1, GMPB1, and GMPC1 were 2057?±?80 mg l?1, 68.7?±?2.5 mg l?1 day?1, 1839?±?44 mg l?1, 60.6?±?1.8 mg l?1 day?1, and 2113?±?64 mg l?1, 77.7?±?2.5 mg l?1 day?1 respectively. GMPB1 was chosen for additional low temperature tolerance studies and growth temperature preference. In winter, GMPB1 grew well at mean temperatures <10?°C, achieving 3258 mg dry biomass from a starting 68 mg. In summer, GMPB1 grew rapidly at mean temperatures more than 28?°C, achieving 1140 mg l?1 dry biomass from a starting 240 mg. Phytonutrient analysis of GMPB1 showed high levels of C-phycocyanin and carotenoids. Arthrospira metabolism relates to terpenoids, and the methyl-d-erythritol 4-phosphate pathway is the only terpenoid biosynthetic pathway in Cyanobacteria. The 1-deoxy-d-xylulose 5-phosphate reductoisomerase (DXR) gene from GMPB1 was cloned and phylogenetic analysis showed that GMPB1 is closest to the Cyanobacterium Oscillatoria nigro-viridis PCC711. Low temperature tolerant Arthrospira strains could broaden the areas suitable for cultivation, extend the seasonal cultivation time, and lower production costs.  相似文献

An efficient transformation protocol was developed for vanilla (Vanilla planifolia) using protocorm-like bodies (PLBs) derived from shoot tips as explants. Of the ten media tested, Murashige and Skoog (MS) medium containing 0.45 μM thidiazuron (TDZ) produced maximum PLBs per shoot tip. Genetic fidelity of PLB-derived plantlets was confirmed by random amplified polymorphic DNA (RAPD) using 23 random primers. PLBs were co-cultured with Agrobacterium tumefaciens strain EHA105 harbouring the binary vector pBI121 containing the β-glucuronidase (gusA) and neomycin phosphotransferase II (npt II) genes for 3 days in MS medium supplemented with acetosyringone and transferred to selective regeneration medium containing 4.43 μM benzyladenine (BA), 2.68 μM naphthalene acetic acid (NAA) supplemented with 50 mg l?1kanamycin and 250 mg l?1 cefotaxime. After 15 days of culture, the surviving explants were transferred to the same regeneration medium but with a higher concentration of kanamycin (75 mg l?1). Finally, explants surviving after 30 days were subjected to more stringent selection in the regeneration medium supplemented with 100 mg l?1 kanamycin. Strong β glucuronidase activity was detected in the transformed plantlets by histochemical assay. Integration of T-DNA into the nuclear genome of transgenic plants was confirmed by polymerase chain reaction and Southern hybridization, while expression of transgene was confirmed by northern hybridization. This protocol allows effective and high frequency transformation of vanilla.  相似文献

Pistachio is a tree of the arid and semi-arid regions where salinity and boron (B) toxicity can be major environmental stresses. In this study, individual and combined effects of different concentrations of NaCl (0, 800, 1600, 2400 and 3200 mg kg?1 soil) and B (0, 2.5, 5.0, 10.0 and 20.0 mg kg?1 soil) were studied on growth, gas-exchange and mineral composition of pistachio seedlings for a duration of 120 days. Excess amounts of salinity (> 1600 mg NaCl kg?1 soil) and B (20.0 mg kg?1 soil) significantly reduced the plant growth and CO2 assimilation, which was associated with accumulation of Na, Cl and B in leaves. There was also a decline in cell membrane stability index (MSI). Reduced stomatal conductance (g s) was the primary cause of inhibition of photosynthesis rate (P N) under mild to moderate salinity. However, under severe salt stress and B toxicity, non-stomatal effects contributed to the inhibition of CO2 assimilation in addition to the decline in g s value. Application of 5.0–10.0 mg B kg?1 soil significantly improved the plant growth and P N and also recovered the MSI as countermeasures against salt stress. These observations were related to the role of B in cell membrane structure and functioning which reduced the concentration of toxic ions in the leaves. However, cell membrane damages and chlorophyll loss in plants affected by salt were observed to be exacerbated when excess amounts of B were present. In conclusion, the results revealed that optimizing the B nutrition can improve the performance of pistachio seedlings under salt stress, and NaCl also showed a mitigating effect on B toxicity if its concentration in the soil is kept under the plant salt tolerance threshold.  相似文献

In this study, adventitious roots of Panax quinquefolium L. have been successfully established. The highest induction rate of roots was obtained in MS medium containing 3 mg L?1 IBA after 4 weeks of culture. The culture conditions of adventitious root were optimized and evaluated with response surface methodology. The best culture conditions for root growth seemed to be 0.75 salt strength MS medium, 4.70 g L?1 inoculum size and 40 days of culture. The active component contents of adventitious roots were compared with those of native roots. The total saponins content was found to be 16.28 mg g?1 in native root and 4.64 mg g?1 in adventitious root. The polysaccharide content of the adventitious root was 1.5 times higher than that in the native P. quinquefolium (30.54 vs. 20.28 mg g?1).  相似文献

Ethyl tert-butyl ether (ETBE) enrichment was obtained by adding contaminated groundwater to a mineral medium containing ETBE as the sole carbon and energy source. ETBE was completely degraded to biomass and CO2 with a transient production of tert-butanol (TBA) and a final biomass yield of 0.37?±?0.08 mg biomass (dry weight).mg?1 ETBE. Two bacterial strains, IFP 2042 and IFP 2049, were isolated from the enrichment, and their 16S rRNA genes (rrs) were similar to Rhodococcus sp. (99 % similarity to Rhodococcus erythropolis) and Bradyrhizobium sp. (99 % similarity to Bradyrhizobium japonicum), respectively. Rhodococcus sp. IFP 2042 degraded ETBE to TBA, and Bradyrhizobium sp. IFP 2049 degraded TBA to biomass and CO2. A mixed culture of IFP 2042 and IFP 2049 degraded ETBE to CO2 with a biomass yield similar to the original ETBE enrichment (0.31?±?0.02 mg?biomass.mg?1 ETBE). Among the genes previously described to be involved in ETBE, MTBE, and TBA degradation, only alkB was detected in Rhodococcus sp. IFP 2042 by PCR, and none were detected in Bradyrhizobium sp. Phosphorus (P) loss from land can impair surface water quality. Losses can occur from soil and plant components. While it is known that P losses increase with soil P concentration, it is not known how losses from pasture plants vary with soil P concentration or between different forages. We examined total P and filterable reactive P (FRP) in water extracts of plant shoots, used as a measure of potential P loss to surface runoff, in different forage species relative to soil P concentration in field trials and a glasshouse experiment. The mean total P concentration of 16 forage species in grazed field plots was greater (P?<?0.01; LSD05?=?117 mg kg?1) in legumes (3,480 mg kg?1) than for grasses (3,210 mg kg?1). Total plant P concentrations of grasses and legumes increased with soil Mehlich-3 P concentrations in both glasshouse and field trials with concentrations close to 6,000 mg kg?1 in arrowleaf clover at 680 mg kg?1 Mehlich-3 soil P. FRP in water extracts of plant shoots increased relative to plant total P as soil Mehlich-3 P increased, with the greatest concentrations shown by crimson clover and arrowleaf clover. Analysis of water extracts of ryegrass and clover herbage from a field trial showed that while FRP was increasing, phytase-available-P decreased significantly from about 70% of filterable unreactive P at the lowest Mehlich-3 P concentrations, to close to zero at 200 mg kg?1 Mehlich-3 P. The wide variation, and enrichment of FRP in water extracts and total P with increasing Mehlich-3 P among species, indicates that cultivar and site selection and sward management provide a potential option to mitigate P loss to surface waters.  相似文献

Background and aims

Variations in responses to soil N between a non-N-fixing shrub, Baccharis halimifolia L., and a N-fixing shrub, Morella cerifera (L.) Small, were tested over 12 weeks to determine whether N availability is the sole cause of persistent dominance of M. cerifera on barrier islands.

Methods

Plants were supplied increasing levels of soil N up to 200 mg kg?1. Measurements included gas exchange and chlorophyll fluorescence parameters across treatments, species, and time. Tissues were analyzed for differences in biomass and nutrients.

Results

Baccharis halimifolia had reduced physiological responses across all treatment levels, but M. cerifera had comparatively few variations. Across all treatments B. halimifolia photosynthesis and stomatal conductance were reduced by 62 and 76 %, respectively,by week 12. Increasing foliar δ15N values across treatments for M. cerifera indicated a shift from utilizing fixed N to available soil N. Biomass was highest at 200 mg kg?1 N for both species. Baccharis halimifolia showed indications of stress response and resource limitation based on physiological responses, nutrient contents, and isotope effects.

Conclusions

Baccharis halimifolia showed signs of co-limitation of both N and P whereas M. cerifera was limited by neither, suggesting that dominance of M. cerifera is only partially explained by actinorhizal symbiosis and N availability.  相似文献
